INTRODUCTION The rapid growth of e-cigarette usage among youth and young people has emerged as a significant public health concern. It is imperative to initiate effective vaping prevention campaigns and undertake relevant research to address this pressing issue. This research seeks to identify effective video advertisements to deter young people from starting to use e-cigarettes. It aims to offer evidence-based insights and recommendations for creating communication materials and designing messages for youth e-cigarette prevention efforts. METHODS College students aged 18-24 years (n=40) participated in focus groups within this qualitative study. After viewing four stimulus videos, participants discussed what they perceived as effective and ineffective video characteristics, as well as suggestions for future videos. RESULTS Effective video characteristics included the use of real-life testimonials, displaying specific health hazards, revealing harmful chemical ingredients and the deceptive nature of flavors, and positively perceived effectiveness. Participants generally found that videos with strong visual impact and graphics were more engaging and that approaches using fear and emotion were more effective. Ineffective characteristics included complex and exaggerated information, lack of empathy and irrelevance, insufficiently specific information, extreme and death-themed content, industry messages, as well as preachy tones, animations, metaphors, dull formats, excessive length, and scenes of e-cigarette use. CONCLUSIONS Developing anti-e-cigarette campaign materials for youth necessitates target audience-focused qualitative research. This helps in deeply exploring and identifying effective themes and messages, as well as video characteristics and details while avoiding ineffective or even misleading messages and themes from young people's perspectives outside the United States. Future development of e-cigarette prevention videos for Chinese college students may consider incorporating localized real-life testimonial cases to convey specific harms, including self-efficacy information, and utilizing fear and emotional appeals. 'General requirements for the production of extracellular vesicles derived from human stem cells' is the first guideline for stem cells derived extracellular vesicles in China, jointly drafted and agreed upon by experts from the Chinese Society for Stem Cell Research. This standard specifies the general requirements, process requirements, packaging and labelling requirements and storage requirements for preparing extracellular vesicles derived from human stem cells, which is applicable to the research and production of extracellular vesicles derived from stem cells. It was originally released by the China Society for Cell Biology on 30 August 2022. We hope that the publication of this guideline will promote institutional establishment, acceptance and execution of proper protocols, and accelerate the international standardisation of extracellular vesicles derived from human stem cells. Context: Yunpi Heluo (YPHL) decoction is a Chinese herbal formula with particular advantages for treating type 2 diabetes. Yet, its exact mechanism of action is not fully understood. Objective: To examine the therapeutic effect of YPHL on ectopic lipid deposition (EDL) in Zucker diabetic fatty (ZDF) rats and the underlying mechanism. Materials and methods: The ZDF Rats were randomized into five groups, including model, YPHL (200mg/kg/d for 10 weeks), SIRT1-overexpression (injected with HBAAV2/9-r-SIRT1-30-flag-GFP), NC (injected with HBAAV2/9-CMV-GFP as blank control) and control group. Pancreatic b-cells obtained from high-lipid-high-glucose fed rats were treated with YPHL (10mg/mL) for 48 h. Lipid deposition and autophagosomes were analyzed by transmission electron microscopy. Intracellular H2O2 and ROS concentrations were measured by flow cytometry. SIRT1, FOXO1, LC3 and P62 mRNA and protein levels were analyzed using qRT-PCR and Western blots. Results: Compared with the model group, blood glucose levels in YPHL and si-SIRT1 groups were reduced by 19.3% and 27.9%, respectively. In high-lipid-high-glucose cells treated with YPHL, lipid droplets were reduced and decrease in apoptosis rate (38.6%), H2O2 (31.2%) and ROS (44.5%) levels were observed. After YPHL intervention or SIRT1 overexpression, LC3 and p62 expression increased. Protein expression of SIRT1 and LC3 in model, si-SIRT1, si-NC and si-SIRT1þYPHL groups was lower than those in control group, while FoxO1 expression was increased. All of these protein level alterations were reversed in the si-NCþYPHL group. Discussion and conclusions: YPHL reduced EDL by regulating the SIRT1-FoxO1 autophagy pathway in diabetic rats, which could lead to future perspectives for the treatment of diabetes. The relationship of Porphyromonas gingivalis and oral squamous cell carcinoma (OSCC) has been studied for several years. Previous studies have focused on the direct effect of P. gingivalis on the activities of primary epithelial cells and OSCC cells. However, the immune system is responsible for mediating cancer development, whether P. gingivalis can affect oral cancer immunity has seldom been explored to date. In this study, we investigated the role of P. gingivalis in the immunoevasion of OSCC. We evaluated the effect of P. gingivalis on the phagocytosis of Cal-27 cells (OSCC cell line) by bone marrow-derived macrophages in vitro and studied the effect of P. gingivalis on the growth of OSCC and the polarization of tumor-associated macrophages in vivo. We found that P. gingivalis was able to inhibit the phagocytosis of Cal-27 cells by macrophages, and membrane-component molecules of P. gingivalis, such as proteins, were speculated to be the effector components. In addition, sustained infection with antibiotics-inactivated P. gingivalis promoted OSCC growth in mice and induced the polarization of macrophages into M2 tumor-associated macrophages, which mainly display protumor properties. Transcriptome analysis and quantitative RT-PCR revealed that P. gingivalis infection upregulated the expression of genes encoding protumor molecules in Cal-27 cells (suprabasin, IL-1R2, and CD47) and in macrophages (IL-1a, CCL-3, and CCL-5). Our in vitro and in vivo data suggest that P. gingivalis can promote immunoevasion of oral cancer by protecting cancer from macrophage attack. This paper investigates the effect of the addition of natural gas (NG) and engine load on the cylinder pressure, combustion process, brake thermal efficiency, and methane combustion efficiency of a heavy-duty NG-diesel dual fuel engine. Significantly increased peak cylinder pressure (PCP) was only observed with the addition of NG at 100% load. The addition of a relatively large amount NG at high load slightly retarded the premixed combustion, significantly increased the peak heat release rate (PHRR) of the diffusion combustion, decreased the combustion duration, and advanced combustion phasing. The accelerated combustion process and increased heat release rate (HRR) at high load were supported by the increased NOx emissions with the addition of over 3% NG (vol.). By comparison, when operated at low load, the addition of a large amount of NG decreased the PHRR of the premixed combustion and slightly increased the PHRR during the late diffusion combustion. Improved brake thermal efficiency was only observed with the addition of a relatively large amount of NG at high load. The improved thermal efficiency was due to a decrease in combustion duration and the shifting of the combustion phasing toward the optimal phasing. The overall combustion efficiency of the dual fuel operation was always lower than diesel-only operation as indicated by the excess emissions of the unburned methane and carbon monoxide from dual fuel engine. This deteriorated the potential of dual fuel engine in further improving the brake thermal efficiency although the combustion duration of dual fuel engine at high load was much shorter than diesel only operation. The addition of NG at low load should be avoided due to the low combustion efficiency of NG and the decreased thermal efficiency. Considering the growing importance of Corporate Social responsibility (CSR) among various stakeholders, the present study investigates the association between corporate social responsibility (CSR) and CEO (Chief Executive Officer) dismissals. Based on a sample of Chinese listed firms from 2009 to 2019, this research study used a random effect logistic model. The findings of the study indicate that total CSR is positively associated with CEO dismissals. In further analysis, we decompose CSR and show that average CSR is negatively linked with CEO dismissals, indicating that CEOs mitigate career concerns by maintaining an optimal range of CSR investment. Moreover, above-average CSR is positively linked with CEO dismissals, indicating that when CSR investments deviate from their optimal range, CEOs' risk of dismissal increases. Background: Tissue-engineering strategies based on mesenchymal stem cells (MSCs) and cell sheets have been widely used for periodontal tissue regeneration. However, given the complexity in periodontal structure, the regeneration methods using a single species of MSC could not fulfill the requirement for periodontal regeneration. Methods: We researched the interaction between the periodontal ligament stem cells (PDLSCs) and jaw bone marrow-derived mesenchymal stem cells (JBMMSCs), and constructed a composite cell sheet comprising both of the above MSCs to regenerate complex periodontium-like structures in nude mice. Results: Our results show that by co-culturing PDLSCs and JBMMSCs, the expressions of bone and extracellular matrix (ECM)-related genes and proteins were significantly improved in both MSCs. Further investigations showed that, compared to the cell sheet using PDLSCs or JBMMSCs, the composite stem cell sheet (CSCS), which comprises these two MSCs, expressed higher levels of bone- and ECM-related genes and proteins, and generated a composite structure more similar to the native periodontal tissue physiologically in vivo. Conclusions: In conclusion, our results demonstrate that the crosstalk between PDLSCs and JBMMSCs in cell sheets facilitate regeneration of complex periodontium-like structures, providing a promising new strategy for physiological and functional regeneration of periodontal tissue. Introduction: Cell-based therapy represents a new frontier in the treatment of a wide variety of human diseases traditionally associated with morbidity outcomes, including those involving inflammation, autoimmunity, tissue damage, and cancer. However, the use of mesenchymal stem cells (MSCs) to treat multiple myeloma (MM) bone disease has raised concerns. Specifically, evidence has shown that infused MSCs might support tumor growth and metastasis. Methods: In this study, we used a standard disseminated MM model in mice to identify the in vivo effects of intravenous MSC infusion. In addition, a series of in vitro co-culture assays were preformed to explore whether Fas/Fas ligand (Fas-L) is involved in the inhibitory effects of MSCs on MM cells. Results: In the MM mouse model, treatment of MSCs with highly expressed Fas ligand (Fas-Lhigh MSCs) showed remarkable inhibitory effects on MM indenization in terms of extending the mouse survival rate and inhibiting tumor growth, bone resorption in the lumbus and collum femoris, and MM cell metastasis in the lungs and kidneys. In addition, reduced proliferation and increased apoptosis of MM cells was observed when co-cultured with Fas-Lhigh MSCs in vitro. Furthermore, mechanistically, the binding between Fas and Fas-L significantly induced apoptosis in MM cells, as evidenced through an increase in the expression of apoptosis marker and Fas in MM cells. In contrast, Fas-Lnull MSCs promote MM growth. Conclusions: These data suggest that Fas/Fas-L-induced MM apoptosis plays a crucial role in the MSC-based inhibition of MM growth. Although whether MSCs inhibit or promote cancer growth remains controversial, the levels of Fas-L expression in MSCs determine, at least partially, the effects of MSCs on MM cell growth. Simulation of flow and contaminant transport through the vadose zone requires accurate parameterization of the soil hydraulic properties. This requirement is particularly important for soils with a complex structure. In the present study, a physically based closed-form expression for the bimodal unsaturated hydraulic conductivity function is proposed for soils with bimodal pore-size distribution. It combines the bimodal representation of the soil-water characteristic curve (SWCC) function of Liu with the conductivity representation model of Mualem. The proposed equations are defined by parameters that have physical significance, which can be related to the properties of the materials. Experimental data for the representation of bimodal SWCCs and corresponding hydraulic conductivity curves were used to demonstrate the applicability of the proposed functions. The proposed approaches resulted in good agreement with experimental data. In the last decades several approaches have been developed to describe bimodal or multimodal soil-water characteristic curves (SWCCs). Unfortunately, most of these models were derived empirically. In the presented study, physically based bimodal and multimodal SWCC functions have been developed for structural soils. The model involved two or more continual pore series; the probability density functions for each pore series were assumed to be lognormal distribution and can be superposed to obtain the overall probability density function of the structural soils. The proposed functions were capable of simulating bimodal or multimodal SWCCs using parameters which can be related to physical properties of the structural soils. The experimental SWCC data were used to verify the proposed method. The fitting results showed that the proposed approaches resulted in good agreement between measurement and simulation. Powdery mildew, caused by Blumeria graminis f. sp. tritici (Bgt), is a devastating disease that seriously threatens wheat yield and quality. To control this disease, host resistance is the most effective measure. Compared with the resistance genes from common wheat, alien resistance genes can better withstand infection of this highly variable pathogen. Development of elite alien germplasm resources with powdery mildew resistance and other key breeding traits is an attractive strategy in wheat breeding. In this study, three wheat-rye germplasm lines YT4-1, YT4-2, and YT4-3 were developed through hybridization between octoploid triticale and common wheat, out of which the lines YT4-1 and YT4-2 conferred adult-plant resistance (APR) to powdery mildew while the line YT4-3 was susceptible to powdery mildew during all of its growth stages. Using genomic in situ hybridization, multi-color fluorescence in situ hybridization, multi-color GISH, and molecular marker analysis, YT4-1, YT4-2, and YT4-3 were shown to be cytogenetically stable wheat-rye 6R addition and T1RS(1BL translocation line, 6RL ditelosomic addition and T1RS(1BL translocation line, and T1RS(1BL translocation line, respectively. Compared with previously reported wheat-rye derivative lines carrying chromosome 6R, YT4-1 and YT4-2 showed stable APR without undesirable pleiotropic effects on agronomic traits. The follicle is the basic structural and functional unit of the ovary in female mammals. The excessive depletion of follicles will lead to diminished ovarian reserve or even premature ovarian failure, resulting in diminished ovarian oogenesis and endocrine function. Excessive follicular depletion is mainly due to loss of primordial follicles. Our analysis of published human ovarian single-cell sequencing results by others revealed a significant increase in rho-associated protein kinase 1 (ROCK1) expression during primordial follicle development. However, the role of ROCK1 in primordial follicle development and maintenance is not clear. This study revealed a gradual increase in ROCK1 expression during primordial follicle activation. Inhibition of ROCK1 resulted in reduced primordial follicle activation, decreased follicular reserve, and delayed development of growing follicles. This effect may be achieved through the HIPPO pathway. The present study indicates that ROCK1 is a key molecule for primordial follicular reserve and follicular development. Objective: To report an extremely rare case of bladder cancer patient with cervical lymph nodes, abdominal lymph nodes, and bone metastases at the same time. Methods and results: The case was investigated by follow-up and immunohistochemistry was used in the pathological part. Result: The patient was diagnosed with bladder cancer (high-grade urothelial metastatic epithelial cell carcinoma) by pathology and immunohistochemistry after transurethral resection of bladder tumor (TURBT) and metastatic bladder cancer by pathology and immunohistochemistry after cervical lymph node aspiration due to neck lymph node enlargement 1 year later, and a CT of the chest and abdomen suggested that the patient also had abdominal lymph node and bone metastases. At the 2.5-year regular chemotherapy follow-up, the patient showed that the abdominal lymph node metastasis disappeared, the cervical lymph node fusion shrank, and the bone metastasis still existed. Conclusion: 1. Regular postoperative review is particularly important; 2. For patients with UCB who undergo TURBT, a effective regular perfusion program should be performed throughout the postoperative period; 3. For patients with postoperative metastatic symptoms of UCB, Complex treatment has a positive effect on patient prognosis; 4. Porcine epidemic diarrhea virus (PEDV) is a highly pathogenic swine coronavirus causing severe diarrhea and high mortality to piglets. PEDV strain AH2012/12 isolated from a diarrheal piglet has been passaged in vitro for over 102 passages. Viral infection assay revealed that PEDV AH2012/12-P102 (the 102nd passage of AH2012/12) showed an enhanced fusogenicity than the wild-type AH2012/12. Animal experiments demonstrated that AH2012/12-P102 is an attenuated PEDV strain as shown by the evidence of no mortality, extremely low virus shedding, and no sign of diarrhea in the AH2012/12-P102 challenged piglets. Compared with AH2012/12, AH2012/12-P102 had two obvious deletions in the genome, one deletion is in the S1 gene and the second deletion contains the carboxy-terminus of the S2 gene and the start codon of ORF3. Using the reverse genetic system of PEDV, we generated a series of recombinant PEDVs with deletions based on the deletion in the genome of AH2012/12-P102. Viral infection assays indicated that the second deletion could enhance the fusogenicity of PEDV AH2012/12. Animal experiments showed that the first deletion could reduce the virulence but not fully attenuate AH2012/12, but the second deletion could attenuate PEDV AH2012/12 in vivo. Further animal experiments indicated that the recombinant PEDV with deleted carboxy-terminus of S gene induced higher IgG, IgA, neutralization antibodies, and protection effects against virus challenge than the killed vaccine. Collectively, our data demonstrated two genetic features associated with the virulence of PEDV AH2012/12 and provided a promising method for the development of attenuated vaccine candidates for PEDV. IMPORTANCE Porcine epidemic diarrhea (PED) caused by PED virus (PEDV) remains a big threat to the swine industry worldwide. Vaccination with live attenuated vaccine is a promising method to prevent and control PED, because it can elicit a more protective immunity than the killed vaccine, subunit vaccine, and so on. In this study, we found two obvious deletions in the genome of a high passage of AH2012/12. We further confirmed the second deletion which contains seven amino acids at the carboxy-terminus of the S2 gene and the start codon of ORF3 can reduce its pathogenicity in vivo. Animal experiments indicated that the recombinant PEDV with deleted carboxy-terminus of S gene showed higher IgG, IgA, neutralization antibodies, and protection effects against virus challenge than the killed vaccine. Background: Apoptotic vesicles are extracellular vesicles generated by apoptotic cells that were previously regarded as containing waste or harmful substances but are now thought to play an important role in signal transduction and homeostasis regulation. Methods: In the present review, we reviewed many articles published over the past decades on the subtypes and formation of apoptotic vesicles and the existing applications of these vesicles. Results: Apoptotic bodies were once regarded as vesicles released by apoptotic cells, however, apoptotic vesicles are now regarded to include apoptotic bodies, apoptotic microvesicles and apoptotic exosomes, which exhibit variation in terms of biogenesis, sizes and properties. Applications of apoptotic vesicles were first reported long ago, but such reports have been rarer than those of other extracellular vesicles. At present, apoptotic vesicles have been utilized mainly in four aspects, including in direct therapeutic applications, in their engineering as carriers, in their construction as vaccines and in their utilization in diagnosis. Conclusion: Building on a deeper understanding of their composition and characteristics, some studies have utilized apoptotic vesicles to treat diseases in more novel ways. However, their limitations for clinical translation, such as heterogeneity, have also emerged. This research investigated the combustion process of an AVL Model LEF/Volvo 5312 single cylinder engine configured to simulate the operation of a heavy-duty spark ignition (SI) natural gas (NG) engine operated on stoichiometric mixture. The factors affecting the combustion process that were examined include intake pressure, spark timing (ST), and the addition of diluents including nitrogen (N2) and carbon dioxide (CO2) to the NG to simulate low British thermal unit (BTU) gases. The mixing of diluents with NG is able to slow down the flame propagation speed, suppress the onset of knock, and allow the engine to operate on higher boost pressure for higher power output. The addition of CO2 was more effective than N2 in suppressing the onset of knock and slowing down the flame propagation speed due to its high heat capacity. Boosting intake pressure significantly increased the heat release rate (HRR) evaluated on J/°CA basis which represents the rate of mass of fuel burning. However, its impact on the normalized HRR evaluated on %/°CA basis, representing the flame propagation rate, was relatively mild. Boosting the intake pressure from 1.0 to 1.8 bar without adding diluents increased the peak HRR to 1.96 times of that observed at 1.0 bar. The increase was due to the burning of more fuel (about 1.8 times), and the 12.9% increase in the normalized HRR. The latter was due to the shortened combustion duration from 23.6 to 18.2 °CA, a 22.9% reduction. The presence of 40% CO2 or N2 in their mixture with NG increased the peak cylinder pressure (PCP) limited brake mean effective pressure (BMEP) from 17.2 to about 20.2 bar. The combustion process of a turbocharged SI NG engine can be approximated by referring to the HRR measured under a naturally aspirated condition. This makes it convenient for researchers to numerically simulate the combustion process and the onset of knock of turbocharged SI NG engines using combustion process data measured under naturally aspirated conditions as a reference. Bacterial infection is one of serious problems hindering tissue regeneration, but strategies able to perform proper antibiosis with spatiotemporal controllability to manipulate the local inflammation and immunity for effective healing of infected wound is still a challenge. Herein, we fabricated a smart hydrogel (CNMs@HA-PDA-PL) with multiple responsiveness to near infrared radiation, visible light, hyaluronidase and hydrogen peroxide for promoted healing of the infected wound via controllable anti-bacteria at designed locations and time periods as well as the on-demand regulation of local inflammation and immunity. The hydrogel consists of hyaluronic acid (HA) modified graphitic carbon nitride (g-C3N4) microsphere (CNMs), antimicrobial peptides (e-Poly-L-lysine, e-PL) and polydopamine (PDA). The PDA framework and CNMs respectively serve as photothermal agent and photodynamic agent to generate heat and radical for instant damage of major bacteria on/surrounding the hydrogel, while the e-PL is in charge of the sustained antibiosis in long term to eliminate the deep bacteria away from the hydrogel via biochemical process. Once the hydrogel was located at infected area, the over-expressed hyaluronidase and hydrogen peroxide would sequentially spread CNMs and e-PL to start photodynamic therapy and biochemical therapy in designed range after initial photothermal therapy, resulting in synergistic antibiosis with spatiotemporal controllability. Natural forests in southern China have been severely logged due to high human demand for timber, food, and fuels during the past century, but are recovering in the past decade. The objective of this study was to investigate how vegetation cover changes in composition and structure affected the water budgets of a 9.6-km2 Dakeng watershed located in a humid subtropical mountainous region in southern China. We analyzed 27 years (i.e., 1967-1993) of streamflow and climate data and associated vegetation cover change in the watershed. Land use/land cover census and Normalized Difference of Vegetation Index (NDVI) data derived from remote sensing were used to construct historic land cover change patterns. We found that over the period of record, annual streamflow ( Q) and runoff/precipitation ratio did not change significantly, nor did the climatic variables, including air temperature, Hamon’s potential evapotranspiration (ET), pan evaporation, sunshine hours, and radiation. However, annual ET estimated as the differences between P and Q showed a statistically significant increasing trend. Overall, the NDVI of the watershed had a significant increasing trend in the peak spring growing season. This study concluded that watershed ecosystem ET increased as the vegetation cover shifted from low stock forests to shrub and grasslands that had higher ET rates. A conceptual model was developed for the study watershed to describe the vegetation cover-streamflow relationships during a 50-year time frame. This paper highlighted the importance of eco-physiologically based studies in understanding transitory, nonstationary effects of deforestation or forestation on watershed water balances. Vanadium dioxide (VO2) is a well-known thermochromic material with large IR modulating ability, promising for energy-saving smart windows. The main drawbacks of VO2 are its high phase transition temperature (τc = 68 °C), low luminous transmission (Tlum), and weak solar modulating ability (ΔTsol). In this paper, the terbium cation (Tb3+) doping was first reported to reduce τc and increase Tlum of VO2 thin films. Compared with pristine VO2, 2 at. % doping level gives both enhanced Tlum and ΔTsol from 45.8% to 54.0% and 7.7% to 8.3%, respectively. The Tlum increases with continuous Tb3+ doping and reaches 79.4% at 6 at. % doping level, representing ~73.4% relative increment compared with pure VO2. Simulating the dynamics of soil water content and modeling soil water evaporation are critical for many environmental and agricultural strategies. The present study aims to develop an analytical solution to simulate soil water redistribution during the evaporation process. This analytical solution was derived utilizing an exponential function to describe the relation of hydraulic conductivity and water content on pressure head. The solution was obtained based on the initial condition of saturation and an exponential function to model the change of surface water content. Also, the evaporation experiments were conducted under a climate control apparatus to validate the theoretical development. Comparisons between the proposed analytical solution and experimental result are presented from the aspects of soil water redistribution, evaporative rate and cumulative evaporation. Their good agreement indicates that this analytical solution provides a reliable way to investigate the interaction of evaporation and soil water profile. [ABSTRACT FROM AUTHOR]

Low-grade concrete-cored sand-gravel (CCSG) pile composite foundation is a new kind of composite foundation for thick and soft clay ground treatment. An analytical solution was derived for calculating the consolidation process of this composite foundation by considering coefficients of horizontal permeability in smear zone, the radial flow within the sand-gravel shell, and the impervious property of concrete-cored pile. The results show that Terzaghi's one-dimensional consolidation solution and the consolidation analytical solution of ordinary composite foundation were special cases of this solution. Curves of the average consolidation degree of the composite foundation under various nondimensional parameters were observed using the program based on the theoretical formula. Meanwhile, a series of in situ measurements including the settlement of pile and soil, the pore water pressure, and the total stress under embankment load were obtained on the CCSG pile composite foundation on a section of Zhenjiang-Liyang highway. The analyzed results show that the new style composite foundation patent technology has many advantages such as small differential postconstruction settlement (differential is not good, small is), reliable quality, high bearing capacity, and stability. And the consolidation of composite foundation is largely affected by the nondimensional parameters. The analytical solution is finally verified with the actual measurement data. [ABSTRACT FROM AUTHOR]
