Abstract

Cold-hardening of cabbage plants, involves changes in Fraction-1-protein. Such changes are correlated with factors affecting hardening e.g. temperature, light, mineral nutrition and age. Abstract

Phosphoenolpyruvate carboxylase (PEP carboxylase EC 4.1.1.31) was extracted from various halophytic, semi-halophytic and glycophytic plant species. When the enzyme of those extracts was substrate protected, and in the presence of 1.6 mM PEP in the reaction mixture, the activity of PEP carboxylase was increased by 100 mM NaCI, and the activity range in the presence of NaCI was expanded. No correlation could be established between the response of the enzyme to ions and various plant characteristics, such as taxonomic status, salt tolerance or carbon fixation pathways. Salt activation of PEP carboxylase was substrate (PEP) dependent, but the minimal substrate concentration varied in different species. Effects of the stabilizing solutes PEP, betaine, proline and glycerol on the kinetic properties of PEP carboxylase from Zea mays (L.) cv. Hazera were analyzed. In the absence of NaC1 the slope of the Hill plot (nH) tended to rise in the presence of these solutes. Stabilization of the enzyme with betaine or glycerol caused a decrease in K' while K' and Vmax increased in the presence of PEP. NaCl (100mM) caused an increase in both K' and Vmax in the protected as well as in the unprotected enzyme, except for PEP protection, where K' decreased somewhat. In the presence of the protectants, glycerol and PEP, the effect of NaCl on Vmax was 2-4 times higher than its effect on the non-protected enzyme. [ABSTRACT FROM AUTHOR]

Abstract

The effects of NaCl on the kinetic properties of desalted phosphoenolpyruvate carboxylase (PEP carboxylase, EC 4.1.1.31) from two halophytes, Suaeda monoica Forssk. ex. J.F. Gmel and Chloris gayana Kunth. were investigated. The tolerance of PEP carboxylase to NaCl in the reaction medium depends on the enzyme pre‐conditioning as well as on the concentration of its substrate PEP in the assay medium. Addition of PEP to the extraction and the storage medium, stabilizes the enzyme. Such a pre‐treated enzyme is inhibited by NaCl in the presence of low concentrations of PEP in the assay medium but is activated by NaCl in the presence of PEP at concentrations above 1.0 mM. Abstract

Young leaves of salt-depleted Aeluropus litoralis Parl. plants show CO2 fixation by the C3-carbon fixation pathway. No detectable activity of phosphoenol pyruvate (PEP) carboxylase was found. When A. litoralis plants were exposed to a NaCl solution, the leaves showed a high activity of PEP carboxylase as well as a significant CO2 fixation by the C4-pathway. - Also in Zea mays L. and Chloris gayana Kunth., the presence of NaCl in the medium influences the balance between phosphoenol pyruvate carboxylase and ribulose-1 ,5-diphosphate carboxylase. Abstract

The effects of NaCl and other salts, in vivo and in vitro, on the activity of phosphoenolpyruvate carboxylase from the coastal C3 halophyte Cakile maritima Scop. were investigated. Plants grown with 100 mM NaCl in their growth medium yielded some 30% higher rates of phosphoenolpyruvate carboxylase activity than did salt-depleted plants. Activity of the enzyme was stimulated when NaCl was added to the reaction mixture in concentrations of up to 200 mM. The magnitude of this in vitro stimulation was similar for plants grown in the presence or absence of NaCl. 32. Carbon Metabolism in Seagrasses: II. PATTERNS OF PHOTOS YNTHETIC CO2 INCORPORATION

Abstract

Patterns of initial photosynthetic CO2 incorporation were determined for some seagrasses and were related to activities of primary carbon fixing enzymes, carbonic anhydrase activities, and δ13C values. According to the incorporation patterns, Cymodocea nodosa was a C4 species while Thalassia hemprichli and Thalassodendron ciliatum were C3 plants. Halophila stipulacea showed an unusual incorporation pattern which could be viewed as intermediate between typical C3 and C4 pathways. The activity ratios of ribulose-l, 5-bisphosphate carboxylase (RUBPcase) to phosphoenolpyruvate carboxylase (PEPcase) were about 3 for Thalassodendron ciliatum and 1 for Cymodocea nodosa and Halophila stipulacea. The latter value, which is intermediate to ratios found in terrestrial C3 and C4 plants, may correlate with the incorporation patterns found for Halophila stipulacea. Since the C4 seagrass lacked the Kranz anatomy, it may, in addition, point to a flexible incorporation potential for these plants. The high δ13C values found in these and other seagrasses did not correlate with their photosynthetic pathways as in terrestrial plants. This discrepancy is probably due to a ‘closed system’ type of photosynthesis in which CO2 is efficiently utilized. The C3 species which utilize CO2 enzymatically must convert exogenous HCO-3 to CO2 internally. Even though carbonic anhydrase activities were very low, conversion rates seemed to be sufficient for high rates of photosynthesis. Since enzymatic fixation rates approached photosynthetic rates even at CO2 saturation, the limitation for these seagrasses to express their high photosynthetic potential is most probably the HCO−3 uptake system.
