Monilinia fructicola has been widely reported as the causal agent of brown rot disease on many Rosaceae family fruits worldwide. It has been reported on stone fruits, e.g., peach, plum, cherry, apricot and mume; as well as pome fruits, e.g., apple, pear and hawthorn. Loquat is a member of the Eriobotrya genus in subfamily Maloideae along with apple, pear and hawthorn. So far, loquat has not been reported as the host of any Monilia species. In June 2019, brown rot symptoms were observed on loquat fruits in an orchard in Wuhan, Hubei Province, China. Thirty single spore isolates were obtained and identified as M. fructicola based on morphological characteristics and molecular analysis. This is the first report of loquat brown rot disease caused by Monilia species in the world. Furthermore, upon artificial inoculation, all three Monilia species from peach in China, i.e., M. fructicola, M. mumecola and M. yunnanensis, could cause typical brown rot disease on loquat fruits. At the same time, M. fructicola isolates from loquat showed virulence similar to those isolates from peach when the pathogenicity test was conducted on peach fruits. These results suggested that loquat could be infected by other Monilia species and that isolates from loquat also have potential to damage other Rosaceae family fruits in practice.

To compare the clinical effect of wheat grain moxibustion combined with rehabilitation training and simple rehabilitation training on finger spasm after stroke.A total of 80 patients with finger spasm after stroke were randomly divided into an observation group and a control group, 40 cases in each group. The control group was given routine rehabilitation training, once a day, 30 min each time. The observation group was given wheat grain moxibustion at Shixuan (EX-UE 11) on the basis of the control group, 8~10 moxibustion cones at each point, once a day. Both groups were treated for 6 days as one course of treatment for 4 courses. The motor function of the affected hand (Fugl-Meyer assessment [FMA] score) and muscle tension (modified Ashworth scale [MAS] grading), surface EMG indexes (wrist dorsiflexor muscle and flexor carpal metacarpal muscle mean square [RMS] value), hand muscle strength (neurological deficit score [NDS]) and daily living ability (modified Barthel index [MBI] score) were compared between the two groups before and after treatment, and clinical efficacy was evaluated.After treatment, FMA and MBI scores in the 2 groups were increased compared with before treatment (Wheat grain moxibustion at Shixuan (EX-UE 11) combined with rehabilitation training can improve the hand motor function and daily living ability of patients with finger spasm after stroke, improve the degree of spasm and the function of wrist dorsiflexor muscle and flexor carpal metacarpal muscle, the clinical effect is better than simple rehabilitation training.

Airport collaborative decision-making has put forward higher requirements for aircraft turnaround operations. In order to improve the efficiency of aircraft turnaround operations, this paper improves the Petri net model based on the characteristics of MATLAB language. It gives the specific steps to analyze the influence of aircraft turnaround operations period on the apron flight viable service route. The experimental analysis shows that the change in passenger cabin cleaning, catering, and air bridge disconnecting within the routine service time does not affect the number of viable service routes and critical operations of aircraft turnaround service. Suppose the time of four operations, the bridge connecting, water clearing, passenger deboarding, and water filling, are respectively set in the interval [1.43, 1.98], [8, 8.7], [10.5, 11] and [8.1, 9.9], the number of viable service routes will only achieve two values. In that case, the bigger one is 4.7543 * 1076 with the 72% generating probability. The other is 3.5715 * 1076, which is 3.11 * 104 times as many as the minimum value obtained by the regular value interval of four operations, improving the quality of the minimum number of viable service routes. The results indicate that setting the time of some aircraft turnaround operations within a reasonable range will improve the number of viable service routes.

The set equation M T = T + D MT = T+\mathcal {D} uniquely defines a nonempty compact set T ⊂ R 3 T\subset \mathbb {R}^3 . If T T has positive Lebesgue measure it is called a 3 3 -dimensional self-affine tile. In the present paper we study topological properties of 3 3 -dimensional self-affine tiles with collinear digit set, i.e., with a digit set of the form D = { 0 , v , 2 v , … , ( | det M | − 1 ) v } \mathcal {D}=\{0,v,2v,\ldots , (|\det M|-1)v\} for some v ∈ Z 3 ∖ { 0 } v\in \mathbb {Z}^3\setminus \{0\} . We prove that the boundary of such a tile T T is homeomorphic to a 2 2 -sphere whenever its set of neighbors in a lattice tiling which is induced by T T in a natural way contains 14 14 elements. The combinatorics of this lattice tiling is then the same as the one of the bitruncated cubic honeycomb, a body-centered cubic lattice tiling by truncated octahedra. We give a characterization of 3 3 -dimensional self-affine tiles with collinear digit set having 14 14 neighbors in terms of the coefficients of the characteristic polynomial of M M . In our proofs we use results of R. H. Bing on the topological characterization of spheres.

Bovine respiratory disease complex (BRDC) is a serious disease affecting feedlot cattle in China and likely other places worldwide. Bovine viral diarrhea virus (BVDV) and bovine parainfluenza virus type 3 (BPIV3) are principally responsible for causing BRDC, and are a major strain to the industrial economy. Eradication of these viruses/disease requires swift viral identification and treatment. Hence, this study established a fast and easy procedure of BVDV and BPIV3 identification that employs reverse transcription recombinase polymerase amplification (RT-RPA) and lateral flow dipstick (LFD), and uses primers and lateral flow (LF) probe targeting the 5’-UTR gene of BVDV and phosphoprotein P gene of BPIV3, respectively. Our assay was able to successfully amplify BVDV and BPIV3 RNA within 25 minutes at 35 °C using RT-RPA, with products visible on the LFD within 5 min at room temperature (RT). The lowest detection limits were 50 RNA molecules for BVDV and 34 RNA molecules for BPIV3 per reaction. We also demonstrated that the established dual RT-RPA LFD assay was precise and targeted, harboring excellent potential to become an onsite molecular diagnostic tool in the detection of BVDV and BPIV3. This method can detect BVDV (Pestivirus A, B) and BPIV3, and exhibit no cross-reaction with other viruses like the classical swine fever virus (CSFV) and infectious bovine rhinotracheitis virus (IBRV). The assay performance was further assessed with clinical samples, and demonstrated good performance in comparison to real-time RT-PCR (RT-qPCR). Moreover, the RT-RPA LFD assay was comparitively rapid and required minimal training.

To investigate the clinical features, treatment and prognosis of patients with hematological diseases complicated with mucor infection.The risk factors, clinical features, treatment regimen and prognosis of 18 hematological disease patients with mucor infection diagnosed by histopathology in our center from April 2014 to June 2020 were retrospectively analyzed.Thirteen males and five females, with an average age of 30 (13－54) years old, were diagnosed as mucor infection by histopathological examination at the site of infection, including 16 cases of mucor infection alone and 2 cases of mucor + aspergillus mixed infection. There were 12 cases with malignant hematological disease and 6 cases with severe aplastic anemia, all of whom with long-term agranulocytosis, and their clinical manifestations and imaging findings were not specific. The common sites of infection were sinuses and lungs, and some patients showed multiple systemic manifestations. The remission status of hematological diseases and recovery of immune function showed an impact on the prognosis. All the patients were treated with amphotericin B liposome combined with posaconazole, and 15 patients were treated with surgery combined with antifungal drugs, 9 of whom were effective and 6 were ineffective, while intravenous administration in 3 cases was ineffective.It is difficult to diagnose hematological disease complicated with mucor infection. After early diagnosis, prognosis can be improved by amelioration of primary state and combination of drugs and surgery.血液病合并毛霉菌感染患者的临床特征及预后分析.探讨血液病患者合并毛霉菌感染的临床特征、治疗及预后情况.回顾性分析2014年4月至2020年6月在本中心经组织病理结果诊断为毛霉菌感染的18例血液病患者的高危因素、临床特征、治疗方案和预后.18例患者中男13例，女5例，平均年龄30（13－54）岁，均经过感染部位组织病理学检查确诊为毛霉菌感染，其中单纯毛霉菌感染16例，毛霉菌+曲霉菌混合感染2例；12例患者为恶性血液病，6例患者为重型再生障碍性贫血，均为长期粒细胞缺乏患者；临床表现及影像学检查结果无特异性；感染累及的常见部位为鼻窦和肺部，部分患者存在多个系统受累表现；血液病的缓解状态和患者的免疫功能恢复情况对预后有影响，所有患者均给予两性霉素B脂质体联合泊沙康唑治疗，手术联合抗真菌药物治疗15例，有效9例，无效6例；3例静脉用抗真菌药物，均无效.血液病合并毛霉菌感染诊断困难，早期明确诊断后，积极改善患者原发病状态，药物联合手术治疗可改善此类患者的预后.

In order to improve the quality of software development, software testing is indispensable. This paper puts forward a reasonable test scheme for the mutual authentication function of high-level biosafety laboratory management software. The author first describes the test environment in detail and then analyze the functional requirements of the system. Finally, the test examples and corresponding results are given.

Single cell RNA-sequencing (scRNA-seq) technology, a powerful tool for analyzing the entire transcriptome at single cell level, is receiving increasing research attention. The presence of dropouts is an important characteristic of scRNA-seq data that may affect the performance of downstream analyses, such as dimensionality reduction and clustering. Cells sequenced to lower depths tend to have more dropouts than those sequenced to greater depths. In this study, we aimed to develop a dimensionality reduction method to address both dropouts and the non-negativity constraints in scRNA-seq data. The developed method simultaneously performs dimensionality reduction and dropout imputation under the non-negative matrix factorization (NMF) framework. The dropouts were modeled as a non-negative sparse matrix. Summation of the observed data matrix and dropout matrix was approximated by NMF. To ensure the sparsity pattern was maintained, a weighted ℓ1 penalty that took into account the dependency of dropouts on the sequencing depth in each cell was imposed. An efficient algorithm was developed to solve the proposed optimization problem. Experiments using both synthetic data and real data showed that dimensionality reduction via the proposed method afforded more robust clustering results compared with those obtained from the existing methods, and that dropout imputation improved the differential expression analysis.

The present climate in the Changbai Mountain area, NE China is warm with abundant rainfall. It is mainly affected by the wet air mass from the Sea of Japan. Local vegetation belongs to the warm temperate mixed conifer and broad-leaved forest, with a mountain cold temperate coniferous forest belt occurring at an altitude of over 1100 m. The Neogene vegetation history of this region is not yet well understood because of the lack of palaeobotanical data. Here we report a systematic palynological result of a plant-bearing diatomite sequence at Badaogou of Changbai Mountains in Jilin Province, which shows distinct vegetation and environment changes during the late Neogene. Seven palynological assemblages are recognized in the diatomite deposits from a freshly closed mine (Zones M-I and M-II), and an outcrop (Zones I–V) respectively in ascending order. Pollen data indicate a dominant woody angiosperm plants such as Ulmus, Quercus, Pterocarya and Fagus etc. at first (Zone M-I). Subsequently, gymnosperm plants mainly represented by Pinus, Tsuga, Picea and Abies show an increase (Zone M-II), and become predominant at a lowest sample in the outcrop (Zone I), distinguished by rich Picea, along with large amount of Tsuga, Pinus and Abies. Pinus increases significantly, while Picea remains higher value, and Abies and Tsuga become limited in the following assemblage (Zone II). Woody angiosperm pollen Tilia, and terrestrial herbs and shrubs including Asteraceae and Chenopodiaceae show their first sign of increase in Zone III, and become more distinguishable in Zone IV. Picea rises again in Zone V. Pollen assemblages display a successive change of vegetation between the mine (Zones M-I and M-II) and the outcrop (Zones I–V), reflecting their continuity in time. Based on the Ar–Ar dating, the outcropping deposits (Zones I–V) are the Latest Pliocene in age, approaching to the Plio-Pleistocene boundary. Consequently, pollen data in this study indicate a major turnover from the warm temperate mixed conifer and broad-leaved forest (Zones M-I and M-II) to the cold temperate coniferous forest (Zone I) during the Late Pliocene in the Changbai Mountain area. It also displays some subsequent fluctuations including the warm temperate mixed conifer and broad-leaved forest partly recovered in Zones III and IV, and decreased again in Zone V. It shows for the first time how local vegetation responses to the global climate deterioration in the Late Pliocene of NE China. The overall changing process is more or less comparable with some simultaneous vegetation changes in North China and Central Japan. The terrestrial herbs and shrubs in the assemblages are not significant comparing with other areas in North China, reflecting the influence of wet air mass from the Sea of Japan. Meanwhile, Tsuga is gradually disappearing in the upper assemblages as a result of continuous lower temperature.

Pestivirus are responsible for cosmopolitan diseases affecting cattle, pigs and other ruminants, presenting a wide range of clinical manifestations, with relevant impact on zootechnic production. Understanding genomic characteristic and virus taxonomy is fundamental in order to sustain control and prophylactic programs. Given the recent various studies reporting a relatively high number of new strains, in particular from Asian countries, in the present study, six hundred-fifty-one genomic sequences have been considered applying the palindromic nucleotide substitutions method for genotyping. Based on the secondary structure analysis of the 5’ untranslated region of RNA, sequence characteristics among Asian genomic clusters within the different Pestivirus species suggested geographic segregation and occurrence of micro-evolutive steps in the genus evolutionary history. This aspect was particularly evident in atypical sequences originated from China or Turkey, indicating risk of diffusion by animals and products trade or contamination of biological products as bovine calf serum, with potential diagnostic and control difficulties.

To investigate the cause, diagnosis and therapeutic method of the neurological complication with the main manifestation of paraplegia after the hematopoietic stem cell transplantation (HSCT).The clinical features, the process of diagnosis and treatment and the prognosis follow-up of 9 cases, who received HSCT in our department during January 2014 and January 2017 and had the neurological complication with the main symptom of paraplegia after the transplantation, were summarized.The incidence rate of paraplegia was 2.96% (9/304). The median onset time was 245 days (50 days-772 days) after transplantation. The cause of paraplegia determined by examination was extramedullary recurrence of leukemia in 3 cases, cyclosporin neurotoxicity in 1 case, GBS in 1 case, CIDP in 2 cases and autoimmune myeleterosis in 2 cases. One patient abandoned the treatment. The rest 8 patients received empirical or targeted treatment. The median follow-up period was 11 months. There were 5 dead cases and 4 survival cases.Paraplegia is a serious post-HSCT complication. The cause of paraplegia should be determined as early as possible to perform targeted treatment. Empirical preemptive treatment should be given if necessary, so as to improve the survival rate and the quality of life of HSCT patients.造血干细胞移植术后以截瘫为主要表现的神经系统合并症.探讨造血干细胞移植术后出现以截瘫为主要表现的神经系统合并症的病因、诊断和治疗方法.总结自2014年1月至2017年1月在我院血液科接受造血干细胞移植治疗后出现以截瘫为主要表现的神经系统合并症9例的临床特点，诊疗经过以及预后随访.截瘫的发病率为2.96%（9/304），中位发病时间为移植后245（50-772）d，完善检查后确定截瘫病因，3例为白血病髓外复发累及脊髓，1例为环孢素神经毒性，1例为格林-巴利综合征（GBS），2例为慢性炎症脱髓鞘性多神经病（CIDP），2例为自身免疫性脊髓病变。除1例患者放弃治疗外，其余8例患者均接受经验性或针对性治疗，中位随访时间为11个月，死亡5例，存活4例.截瘫是HSCT术后严重的并发症，应尽早明确病因后针对性治疗，必要时可予以经验性抢先治疗，以改善HSCT患者的生存率及生活质量.

Objective To explore the effects of human umbilical cord mesenchymal stem cells (hUCMSCs) on the organ protection of sepsis mice and the regulation of inflammatory factors. Methods hUCMSCs were isolated and established from normal newborn infants. To induce sepsis in mice, lipopolysaccharide (LPS) was injected into the abdominal cavity at 10mg/kg. Animals were randomly divided into 3 groups: control group (n=5), LPS group (n=12), LPS+hUCMSCs group (n=12). Sepsis was induced by intraperitoneal injection of LPS in the LPS group and the hUCMSCs group. After 1 hour of injection, hUSMSCs were injected into the tail vein of mice in LPS+hUCMSCs group (2.5×105/mouse). Mice were euthanized at 3, 6, 12 and 24 hours after LPS injection. Then, the liver, lung, kidneys, and blood specimen were harvested. HE staining was used to detect the injury and recovery of organs in the mice. The expression of inflammatory cytokines IL-10 and TNF-α in the liver and peripheral blood were detected by immunohistochemistry and ELISA, respectively. Results hUCMSCs were successfully established. hUCMSCs significantly alleviated the lung, kidney, and liver tissue damage in LPS+hUCMSCs group compared with LPS group. We study the topological properties of a class of planar crystallographic replication tiles. Let M ∈ Z 2 × 2 be an expanding matrix with characteristic polynomial x 2 + A x + B ( A , B ∈ Z , B ≥ 2 ) and v ∈ Z 2 such that ( v , M v ) are linearly independent. Then the equation M T + B − 1 2 v = T ∪ ( T + v ) ∪ ( T + 2 v ) ∪ ⋯ ∪ ( T + ( B − 2 ) v ) ∪ ( − T ) defines a unique nonempty compact set T satisfying T o ¯ = T . Moreover, T tiles the plane by the crystallographic group p 2 generated by the π -rotation and the translations by integer vectors. It was proved by Leung and Lau in the context of self-affine lattice tiles with collinear digit set that T ∪ ( − T ) is homeomorphic to a closed disk if and only if 2 | A | B + 3 . However, this characterization does not hold anymore for T itself. In this paper, we completely characterize the tiles T of this class that are homeomorphic to a closed disk.

In this paper, we study a nonlinear ψ−Hilfer fractional integro-differential equation on finite interval [ a , b ] . Sufficient conditions for the existence of solution and stability for the initial value problem are obtained. Firstly, the existence of solution for the equation is proved by using Banach contraction mapping principle. Then the Ulam-Hyers-Rassias stability, Ulam-Hyers stability, Semi-Ulam-Hyers-Rassias stability of the system are discussed.

Objective: To study the effects of Atorvastatin calcium combined with Aspirin on serum levels of homocysteine (Hcy), neuron-specific enolase (NSE), uric acid (UA), high sensitity C-reactive protein (hs-CRP) and inflammatory factors of patients with cerebral infarction. Methods: 100 cases of patients with cerebral infarction from March 2014 to May 2016 were treated in the Department of Neurology of our hospital and affiliated to Huazhong University of Science and Technology of traditional Chinese medicine and Western Medicine. The subjects were divided into the control group (n=50) and the treatment group (n=50) randomly. The control group was treated with Aspirin, the treatment group were treated with Atorvastatin calcium combined with Aspirin. The two groups were treated for 28 d. The serum levels of Hcy, NSE, UA, hs- CRP, interleukin-6 (IL-6), interleukin-8 (IL-8) and tumor necrosis factor-α (TNF-α) of the two groups before and after treatment were compared. Results: There were no significantly differences of the serum levels of the Hcy, NSE, UA and hs-CRP of the two groups before treatment (P>0.05). After treatment, the serum levels of the Hcy, NSE, UA and hs-CRP of the two groups were significantly lower than before treatment, and that of the treatment group were significantly lower than the control group (P0.05). After treatment, the serum levels of the IL-6, IL-8 and TNF-α of the two groups were significantly lower than before treatment, and that of the treatment group were significantly lower than the control group (P

Effects of polyvinylchloride (PVC) sheet thickness (t), feed speed (υ), spindle speed (ω), Z-axis feed rate (p) and tool head diameter (Φ) as well as their interactions during the single point incremental forming (SPIF) on forming performance of the PVC sheet material were studied through an orthogonal experimental test. In this experiment, the angle-variable cone was used and the maximum forming limiting angle was taken as the experimental index. Results showed that and ω×Φ influence forming performance of PVC sheet material significantly. υ is the main influencing factor of SPIF performance of PVC sheet material. Small υ is good for sheet material forming. p and Φ are proportional to forming performance of sheet material. Over ω will cause material wear-out. Effect of t could be neglected.

We demonstrate cascaded multiple Fabry–Perot interferometers in multimode fiber with a waveguide, inscribed by femtosecond laser micromachining. The device can sustain high temperature up to 1000 °C, and the temperature sensitivity obtained is 10.1 pm/°C and 17.1 pm/°C within the temperature region 100–400 °C and 400–1000 °C respectively. Compared with single-mode fiber, multimode fiber has a larger core diameter and a higher numerical aperture, allowing more optical power to be coupled from the light source. Thus the optical fiber sensors based on multimode fiber do not have high requirements on the light source used, the system cost can be reduced.

Background Many mathematical and statistical models and algorithms have been proposed to do biomarker identification in recent years. However, the biomarkers inferred from different datasets suffer a lack of reproducibilities due to the heterogeneity of the data generated from different platforms or laboratories. This motivates us to develop robust biomarker identification methods by integrating multiple datasets. Methods In this paper, we developed an integrative method for classification based on logistic regression. Different constant terms are set in the logistic regression model to measure the heterogeneity of the samples. By minimizing the differences of the constant terms within the same dataset, both the homogeneity within the same dataset and the heterogeneity in multiple datasets can be kept. The model is formulated as an optimization problem with a network penalty measuring the differences of the constant terms. The L1 penalty, elastic penalty and network related penalties are added to the objective function for the biomarker discovery purpose. Algorithms based on proximal Newton method are proposed to solve the optimization problem. Results We first applied the proposed method to the simulated datasets. Both the AUC of the prediction and the biomarker identification accuracy are improved. We then applied the method to two breast cancer gene expression datasets. By integrating both datasets, the prediction AUC is improved over directly merging the datasets and MetaLasso. And it’s comparable to the best AUC when doing biomarker identification in an individual dataset. The identified biomarkers using network related penalty for variables were further analyzed. Meaningful subnetworks enriched by breast cancer were identified. Conclusion A network-based integrative logistic regression model is proposed in the paper. It improves both the prediction and biomarker identification accuracy. Skeleton is a new notion designed for constructing space-filling curves of self-similar sets. In a previous paper by Dai and the authors [Space-filling curves of self-similar sets (II): Edge-to-trail substitution rule, Nonlinearity 32(5) (2019) 1772–1809] it was shown that for all the connected self-similar sets with a skeleton satisfying the open set condition, space-filling curves can be constructed. In this paper, we give a criterion of existence of skeletons by using the so-called neighbor graph of a self-similar set. In particular, we show that a connected self-similar set satisfying the finite-type condition always possesses skeletons: an algorithm is obtained here.

To study the effect and mechanism of mTOR signaling on adipogenesis of bone marrow mesenchymal stem cells(BM-MSCs) from aplastic anemia (AA) patients through regulation of PPARγ.BM-MSCs were isolated from 24 newly diagnosed AA patients and 24 healthy controls. The surface antigen expression of BM-MSCs was identified by flow cytometry. The capacity of adipogenic differentiation of BM-MSCs was determined by lipid droplets based on Oil Red O staining and by the expression of FABP4 based on Western blot. Protein levels of mTOR signaling and PPARγ were tested by immunofluorescence and Western blot.AA BM-MSCs displayed an enhanced capacity of differentiating into adipocytes, compared with control BM-MSCs. It was found that mTOR was activated in AA BM-MSCs. Moreover, the expression levels of p-mTOR and PPAR-γ in AA BM-MSCs showed a parallel differentiation-dependent increase during adipogenic differentiation, which were significantly higher than that of control BM-MSCs at the same time point of adipogenic differentiation. mTOR inhibitor rapamycin did not only inhibit the adipogenic differentiation of BM-MSCs from AA pateints at the early-middle stage, but also partly reversed the adipogenic differention of BM-MSCs from AA pateints at the late stage by PPARγ regulation.mTOR signaling may play a critical role in the adipogenic differentiation of BM-MSCs from AA patients by positively regulating PPARγ expression.

To investigate the value of flow cytometry (FCM) detection in prognostic evaluation of minimal residual disease (MRD) of acute myeloid leukemia (AML) patients after allogeneic hematopoietic stem cell transplantation (allo-HSCT).Eighty-two cases of AML (except M3) after allo-HSCT who accord to enrolled condition (no MRD positive after allo-HSCT confirmed by regular flow cytometry detection and followed-up for 2 years) from April 2012 to September 2016 in our department were selected. Among 82 cases males were 50 and females were 32 with average age of 27.27 (2-57) years old. Clustering methods for multiple graphs explore and exploit multiple graphs simultaneously to obtain a more accurate and robust partition of the data than that using single graph clustering methods. In this paper, we study the clustering of multiple graphs with inter-relation among vertices in different graphs. The main contribution is to propose and develop a block spectral clustering method for multiple graphs with inter-relation. Our idea is to construct a block Laplacian matrix for multiple graphs and make use of its eigenvectors to perform clustering very efficiently. Global optimal solutions are obtained in the proposed method and they are solutions of relaxation of multiple graphs ratio cut and normalized cut problems. In contrast, existing clustering methods cannot guarantee optimal solutions and their solutions are dependent on initial guesses. Experimental results on both synthetic and real-world data sets are given to demonstrate that the clustering accuracy achieved and computational time required by the proposed block clustering method are better than those by the testing clustering methods in the literature.

The rabies virus (RV) vector LBNSE expressing foreign antigens have shown considerable promise as vaccines against viral and bacteria diseases, which is effective and safe. We produced a new RV-based vaccine vehicle expressing 1.824 kb hemagglutinin (H) gene of the canine distemper virus (CDV) by reverse genetics technology. The recombinant virus LBNSE-CDV-H retained growth properties similar to those of vector LBNSE both in BSR and mNA cell culture. The H gene of CDV was expressed and detected by immunostaining. To compare the immunogenicity of LBNSE-CDV-H, dogs were immunized with each of these recombinant viruses by intramuscular (i.m.). The dogs were bled at third weeks after the immunization for the measurement of virus neutralizing antibody (VNA) and then challenged with virulent virus (ZJ 7) at fourth weeks. The parent virus (LBNSE) without expression of any foreign molecules was included for comparison. Dogs inoculated with LBNSE-CDV-H showed no any signs of disease and exhibited seroconversion against both RV and CDV H protein. The LBNSE-CDV-H did not cause disease in dogs and conferred protection from challenge with a lethal wild type CDV strain, demonstrating its potential value for wildlife conservation efforts. Together, these studies suggest that recombinant RV expressing H protein from CDV stimulated high levels of adaptive immune responses (VNA), and protected all dogs challenge infection.

Bovine viral diarrhea virus (BVDV) is often detected in commercial bovine serum. BVDV genetic diversity was investigated in commercial bovine serum of Chinese origin. Twenty-two batches of bovine serum were obtained from 10 suppliers with different geographic origins in China, and 20 batches of bovine serum were positive by reverse-transcription polymerase chain reaction (RT-PCR) and sequencing. Phylogenetic reconstructions of partial 5'UTR sequences indicated that the samples examined in this work clustered within the BVDV type 1 and BVDV type 2 genotypes. Interestingly, 3 sample sequences clustered into CSFV. These results suggest a high genetic diversity in Chinese BVDV field isolates. This study will benefit epidemiological surveys of BVDV detected in China.

Bovine viral diarrhea virus (BVDV) can contaminate biological products produced in bovine or porcine cells or manufactured using bovine sera. A rapid, specific, sensitive, and practical method of detecting BVDV in bio-products is needed. The purpose of this study was to compare three assays with respect to their ability to accurately detect BVDV in biological samples, namely reverse-transcription loop-mediated isothermal amplification (RT-LAMP), SYBR green I-based real-time RT-PCR, and conventional RT-PCR. All assays detected BVDV nucleotide and differentiated between BVDV-free and -contaminated bovine sera successfully. In addition, the results were specific to BVDV: the amplification of samples containing the closely related classical swine fever virus or other pathogenic bovine viruses yielded negative results. The lowest detection threshold, 10(1) copies, was displayed by the SYBR green I-based real-time RT-PCR and RT-LAMP assay. This assay was also the most effective in the detection of BVDV contamination in a set of commercially available bovine sera. The field conditions suggest that RT-LAMP is specific and sensitive to detecting BVDV in biological samples and may be used for quality control of biomaterials.

Astroviruses are becoming a growing concern in veterinary and public health. Many astrovirus species are associated with enteric diseases have been described in both mammalian and avian hosts. In the present study, 23 fecal samples from diarrheic minks were collected in Liaoning and Shandong Province, and an investigation of astrovirus was performed using biochemical methods and RT-PCR assay with specific primers. A total of four mink astroviral isolates were detected from sick minks with diarrhea problems. Further sequencing and characterization of the partial ORF1b gene and ORF2 gene segments revealed low sequence identities (20.0-85.3 and 31.8-87.2%) with known astroviral strains, indicating the emergence of a novel clade of astroviruses. Some new features of the astroviral genome have also been discovered. The phylogenetic tree revealed that all samples were distantly related to mink astrovirus and were closely related to chicken astroviruses and turkey astroviruses. MK/DL-1, MK/DL-2, MK/SD-1, and MK/SD-2 formed a new clade and were found to be more closely related to astroviruses from birds than to other mink strains, indicating past cross-species transmission and considerable zoonotic potential.

NH4+-N was choiced as an influence factor for analysis the effect on the laccase activity from the white rot fungus Pleurotus ostreatus and indole degradation. The experiments were performed with different NH4+-N concentration, with a focus on the indole degradation by laccase in the presence of 2,2-Azino-bis (3-ethylbenzthiazoline-6-sulphonic acid) (ABTS) or not. Results showed that NH4+-N had a positive effect on laccase activity and also enhanced the degradation of indole. Meanwhile, NH4+-N was oxidized to NO2-.

Porcine circovirus type1 (PCV1), described initially as a contaminant of a porcine kidney cell line, is ubiquitous within the swine population The presence of PCV1 in porcine cell lines can lead to contamination during both human and porcine vaccine production. Therefore, a rapid, specific, sensitive and practical method is needed for the detection of PCV1 in bio-products. The aim of this study was to compare three assays in their ability to accurately quantify PCV1 virus in biological samples, namely loop-mediated isothermal amplification (LAMP), SYBR green I-based real-time polymerase chain reaction (PCR) and conventional PCR. All assays yielded successful quantitation of PCV1 DNA and differentiated between PCV1-free and–contaminated cells. In addition, the results were specific for PCV1, since amplification of samples containing closely-related PCV2 or other pathogenic swine viruses yielded negative results. The lowest detection threshold of 10 2 copies was displayed by the SYBR green I-based real-time PCR assay. In addition, this assay was the most effective in detecting PCV1 contamination in a set of commercially available porcine vaccines. Therefore we conclude that SYBR green I–based real-time PCR is specific and sensitive for detecting PCV1 in biological samples and maybe used for quality control of vaccine and biomaterial production.

Drug-target interaction (DTI) prediction is a challenging step in further drug repositioning, drug discovery and drug design. The advent of high-throughput technologies brings convenience to the development of DTI prediction methods. With the generation of a high number of data sets, many mathematical models and computational algorithms have been developed to identify the potential drug-target pairs. However, most existing methods are proposed based on the single view data. By integrating the drug and target data from different views, we aim to get more stable and accurate prediction results. In this paper, a multiview DTI prediction method based on clustering is proposed. We first introduce a model for single view drug-target data. The model is formulated as an optimization problem, which aims to identify the clusters in both drug similarity network and target protein similarity network, and at the same time make the clusters with more known DTIs be connected together. Then the model is extended to multiview network data by maximizing the consistency of the clusters in each view. An approximation method is proposed to solve the optimization problem. We apply the proposed algorithms to two views of data. Comparisons with some existing algorithms show that the multiview DTI prediction algorithm can produce more accurate predictions. For the considered data set, we finally predict 54 possible DTIs. From the similarity analysis of the drugs/targets, enrichment analysis of DTIs and genes in each cluster, it is shown that the predicted DTIs have a high possibility to be true.

Two novel compounds, 8–[2–(2–thienyl)vinyl]–10,10–dimethyl–10H–pyrido[1,2–a] indolium perchlorate (3a) and 8–[2–(5–phenyl–2–thienyl)vinyl]–10,10–dimethyl–10H–pyrido[1,2–a]indolium perchlorate (3b) were synthesized and characterized by IR, 1H–NMR, elemental analyses, and X–ray diffraction. Crystal structural analysis suggested that either 3a or 3b exhibited good coplanarity and rings and vinyl in the target molecule could make up a large conjugated system. Ultraviolet–visible absorption analysis indicated both 3a and 3b possessed large maximum absorptions, and 3b underwent a significant redshift (43.0 nm) in comparison with 3a.

Based on experiment of sawing Wulian red granite with diamond circular saw, the relations between the cutting force with machining parameters are studied. Cutting speed, feed rate and cutting depth are considered as the process parameters. The cutting force in sawing granite operation are measured and the experimental results are then analyzed using response surface methodology. From the analysis, it is seen that the cutting force Fx , Fy and Fz are reduced with the increase of cutting speed and increased with the increase of feed rate and cutting depth, and the mathematical models of the cutting force are developed. By ANOVA for the cutting force models, It is concluded that the models are significant at 95% confidence level and the significant effects are the first-order of cutting speed, feed speed, cutting depth and the quadratic of cutting depth.

There were 4 Acinetobacter lwoffii obtained from soil samples. The antimicrobial susceptibility of the strains to 16 antimicrobial agents was investigated using K-B method. Three isolates showed the multi-drug resistance. The presence of resistance genes and integrons was determined using PCR. The aadA1, aac(3')-IIc, aph(3')-VII, aac(6')-Ib, sul2, cat2, floR, and tet(K) genes were detected, respectively. Three class 1 integrons were obtained. The arr-3-aacA4 and blaPSE-1 gene cassette, which cause resistance to aminoglycoside and beta-lactamase antibiotics. Our results reported the detection of multi-drug resistant and carried resistant genes Acinetobacter lwoffii from soil. The findings suggested that we should pay close attention to the prevalence of multi-drug resistant bacterial species of environment.

According to microstructures in eutectic composite ceramic, the finite element model of composite with eutectic interphase is established. The mechanical stress field of eutectic composite ceramic containing lamellae is simulated. APDL programming in ANSYS is used to analyze the damage process of eutectic composite ceramic. Results show that the failure of eutectic composite ceramic is determined by the damage of matrix. As load is increasing, the damage will elongate along the interphase and extend to the internal of matrix. At last the damage arouses matrix fracture.

Modeling genetic regulatory interactions is an important issue in systems biology. Probabilistic Boolean networks (PBNs) have been proved to be a useful tool for the task. The steady-state probability distribution of a PBN gives important information about the captured genetic network. The computation of the steady-state probability distribution involves the construction of the transition probability matrix of the PBN. The size of the transition probability matrix is 2n×2n where n is the number of genes. Although given the number of genes and the perturbation probability in a perturbed PBN, the perturbation matrix is the same for different PBNs, the storage requirement for this matrix is huge if the number of genes is large. Thus an important issue is developing computational methods from the perturbation point of view. In this paper, we analyze and estimate the steady-state probability distribution of a PBN with gene perturbations. We first analyze the perturbation matrix. We then give a perturbation matrix analysis for the captured PBN problem and propose a method for computing the steady-state probability distribution. An approximation method with error analysis is then given for further reducing the computational complexity. Numerical experiments are given to demonstrate the efficiency of the proposed methods.

Ni base alloy composite coating containing spheroidal ceramic grains can be fabricated by a vacuum fusion sintering method. Composite coating was mainly composed of Ni base alloy and spheroidal ceramic grains with random orientation. The three-phase model is used to determine the thermal expansion coefficient of the composite coating. First, Eshebly-Mori-Tanaka method was used to determine thermal disturbance strain in two-phase cell aroused thermal inconsistency. Then, average thermal strain in the two-phase cell aroused by thermal inconsistency is gained by the means of volume equilibration. The two-phase cell is transverse isotropy and has two independent thermal expansion coefficients. Finely, based on mean strain of Ni base alloy ceramic composite coating containing spheroidal ceramic grains, the effective thermal expansion coefficient of the composite coating is obtained by considering random orientation of two-phase cells. Ni base alloy composite coating containing spheroidal ceramic grains is isotropy and has one independent thermal expansion coefficient.
