Micro-milling has been employed in various fields due to its efficiency, flexibility, material, and structural versatility in the machining of small and high-quality parts. However, when micro-milling highly plastic aluminum alloys, it is difficult to produce a smooth surface due to the easy sticking of tools. To solve this problem, a multi-energy field coupling method of adding nano-lubricant to the micro-milling area by minimum quantity lubrication (MQL) and modifying the aluminum alloy by cold plasma (CP) was introduced. In this paper, the experiments of CP and h-BN nano-lubricant minimum quantity lubrication (NMQL) multi-field coupling assisted (CPNMQL) micro-milling of aluminum alloy 6061-T65 were carried out to analyze 3D surface roughness (Sa), tool texture, milling forces (Fx and Fy), and surface morphology. The results revealed that the CP could increase the surface microhardness and improve material removal rate; Cottonseed oil with h-BN nanoparticles exhibited superior lubricating properties; The milling forces Fx and Fy were significantly reduced; Sa (0.029 µm) under CPNMQL condition decreased by 60 %, 72.1 %, 27.5 %, and 53.2 % comparing to dry (0.104 µm), NMQL (0.04 µm), and CP (0.062 µm) conditions. In addition, the smallest and most uniform tool mark texture height and depth of 0.038 µm was obtained under CPNMQL condition, a 76.3 % reduction over dry condition.

Abstract Various pathogens from oomycete, fungi, and bacteria kingdoms can infect potato and significantly reduce potato yield. The early diagnosis of potato pathogens is important for tracing disease epidemics and the subsequent disease management. Loop-mediated isothermal amplification (LAMP) is a critical technique for pathogen detection, but available LAMP assays do not effectively meet the requirement of field diagnoses due to complexities including co-infection of different pathogens. Hence, this study aims to develop integrated-LAMP assays (iLAMPs) for simultaneous detection of eight common potato pathogens and apply iLAMPs to pathogen detection in field samples from the four main potato-growing regions of China in 2023. Therefore, eight sets of primers showing gene- and genus-specificity were designed and used for iLAMPs to determine their specificity, sensitivity, and visualization. Subsequently, iLAMPs-mediated pathogen detection revealed that 72.82% of 206 diseased leaves and 84.94% of 239 diseased tubers carry more than one pathogen. The detection rate for each pathogen significantly varies from 1.94 to 65.53% in diseased leaves, and ranges from 26.78 to 52.72% in diseased tubers, respectively. In addition, the detection rate of Phytophthora infestans and Alternaria solani positively correlates in both leaves and tubers, especially for those samples from the southwestern and southern regions. Taken together, iLAMPs developed in this study enables simultaneous detection of eight common potato pathogens from field samples and may have broad applications in early management of potato diseases.

Optimizing blasting parameters is of paramount importance in minimizing overbreak during tunneling. Hence, this paper proposed an intelligent approach, which integrates separate parameter optimization based on varying rock mass qualities, with the objective of reducing overbreak. This novel intelligent method constructs a comprehensive model with three distinct functions, which can provide precise overbreak prediction, analyze the mechanisms by which input parameters influence overbreak, and integrate feature importance into the blasting parameters optimization process. First, the hyperparameters of seven tree-based algorithms were optimized using the Sparrow Search Algorithm (SSA), and the best predictive model was selected by comparing various performance metrics. Then, the importance and influencing mechanisms of input features on overbreak were revealed through the utilization of the Shapley Additive Explanation. Subsequently, interactions among crucial parameters were investigated, and their design values were recommended. Finally, a novel parameter optimization method was employed to reduce overbreak, which combines the conclusions drawn from the importance analysis of input features with SSA through three key steps: modification of the initial position, expanding the search scope, and neighborhood perturbation. Compared to the unimproved methods, the proposed approach can significantly reduce post-blast overbreak areas in different sections of the tunnel by 12.8 % and 16.4 %, respectively.

Abstract Background Lung adenocarcinoma (LUAD) is one of the most common malignant diseases worldwide. This study aimed to construct an immunogenic cell death (ICD)-related long non-coding RNA (lncRNA) signature to effectively predict the prognosis of LUAD. Methods The RNA-sequencing and clinical data of LUAD were downloaded from The Cancer Genome Atlas (TCGA). Least absolute shrinkage and selection operator (LASSO) and stepwise multivariate Cox proportional hazard regression analysis were utilized to construct lncRNA signature. Then, the reliability of the signature was evaluated in the training, validation and whole cohorts. The differences in the immune landscape and drug sensitivity between the low- and high-risk groups were analyzed. Finally, the expression level of the selected ICD-related lncRNAs in LUAD cell lines via reverse transcription quantitative PCR (RT-qPCR). CCK-8 and transwell assays were performed to study biological function of AC245014.3. Results A signature consisting of 5 ICD-related lncRNAs was constructed. Kaplan Meier (K-M) survival analysis showed shorter overall survival (OS) in high-risk group. The receiver operating characteristic (ROC) curves and Multivariate Cox regression analysis showed the signature was good predictive and independent prognostic factor in LUAD. Moreover, the high-risk group had a lower level of antitumor immunity and was less sensitive to some chemotherapeutics and targeted drugs. Finally, the expression level of selected ICD-related lncRNAs was validated in LUAD cell lines by RT-qPCR. Knockdown of AC245014.3 significantly suppressed LUAD proliferation, migration and invasion. Conclusions In this study, an ICD-related lncRNA signature was constructed, which could accurately predict the prognosis of LUAD patients and guide clinical treatment.

Since 2020, Douyin, an app known for its interactive entertainment and vibrant youth cultures, has risen to dominance in the retail sector. Douyin stands out by making paid traffic a significant revenue stream alongside commissions. This strategy, which restricts organic growth, compels sellers to make additional investments in traffic. Drawing from Douyin walkthroughs and the company's business development presentations, this article analyzes how audience attention and platform traffic are manufactured and integrated with retail in the context of China's recent national policy that positions data as a factor of production equal to labor, land, technology, and capital. In contrast to Instagram, traffic conversion into sales takes precedence over product visibility on Douyin. In this process, Douyin actively uses user data to manufacture high-traffic keywords with buying intent. This involves measurements employing surveillance technologies that span image and speech recognition, keywords, performance metrics, and pricing algorithms. The article argues that Douyin e-commerce cannot be fully explained by the current visibility research paradigm centered on metrics such as likes, shares, and comments, which are considered indications of interests and preferences. It is suggested that Douyin uses historical data to invoke momentary interests and produce desired user actions for conversion. Traffic investment alone cannot result in the conversion of momentary interests into sales; it needs to be combined with pricing that incorporates discounts, coupons, and reductions. The integration of traffic investment with pricing strategy has emerged as a dominant e-commerce practice that fosters retail growth.

Bacterial infections significantly threaten human health, leading to severe diseases and complications across multiple systems and organs. Antibiotics remain the primary treatment strategy for these infections. However, the growing resistance of bacteria to conventional antibiotics underscores the urgent need for safe and effective alternative treatments. In response, several approaches have been developed, including carbon dots (CDs), antimicrobial peptides, and antimicrobial polymers, all of which have proven effective in combating bacterial resistance. Among these, CDs stand out due to their unique advantages, including low preparation cost, stable physicochemical properties, high biocompatibility, tunable surface chemistry, strong photoluminescence, and efficient generation of reactive oxygen species. These features make CDs highly promising in antibacterial applications. This review explores the development of antibacterial CDs, focusing on their mechanisms of action—physical destroy, biochemical damage, and synergistic effects—while highlighting their potential for clinical use as antibacterial agents.

Abstract The present study aimed to explore the underlying mechanism of bile reflux-inducing chronic atrophic gastritis (CAG) with colonic mucosal lesion. The rat model of CAG with colonic mucosal lesion was induced by free-drinking 20 mmol/L sodium deoxycholate to simulate bile reflux and 2% cold sodium salicylate for 12 weeks. In comparison to the control group, the model rats had increased abundances of Bacteroidetes and Firmicutes but had decreased abundances of Proteobacteria and Fusobacterium. Several gut bacteria with bile acids transformation ability were enriched in the model group, such as Blautia, Phascolarctobacter, and Enterococcus. The cytotoxic deoxycholic acid and lithocholic acid were significantly increased in the model group. Transcriptome analysis of colonic tissues presented that the down-regulated genes enriched in T cell receptor signaling pathway, antigen processing and presentation, Th17 cell differentiation, Th1 and Th2 cell differentiation, and intestinal immune network for IgA production in the model group. These results suggest that bile reflux-inducing CAG with colonic mucosal lesion accompanied by gut dysbacteriosis, mucosal immunocompromise, and increased gene expressions related to repair of intestinal mucosal injury.

Bacterial infections and antibiotic resistance are global health problems, and current treatments for bacterial infections still rely on the use of antibiotics. Phototherapy based on the use of a photosensitizer has high efficiency, a broad spectrum, strong selectivity, does not easily induce drug resistance, and is expected to become an effective strategy for the treatment of bacterial infections, particularly drug-resistant infections. This article reviews antimicrobial strategies of phototherapy based on photosensitizers, including photodynamic therapy (PDT), photothermal therapy (PTT), and their combination. These methods have significant application potential in combating multi-drug-resistant bacterial and biofilm infections, providing an alternative to traditional antibiotics and chemical antibacterial agents.

Micro light-emitting diode (Micro-LED) is a highly promising technology in the field of new displays, with the mass transfer process involved in its manufacturing process widely regarded as a major barrier to their further development. This study adopts laser transfer technology as the primary solution, using ablation-type transfer release materials that improve chip utilization rates over blister-type release materials. In addition, measurement of the laser transfer parameters, inspection, and laser repair technology are combined to achieve a transfer yield of about 100% to the carrier substrate and a cumulative transfer displacement of less than 1 µm in the Micro-LED inverted chip array. Furthermore, cleaning agents were used to remove adhesive residue from the receiving substrate after transfer, improving the bonding yield between the chip and the thin film transistor driver circuit board. This study provides a feasible solution for Micro-LED mass transfer, which could boost its further development toward commercial application.

BackgroundPatients with heart failure (HF) with preserved ejection fraction (HFpEF) are more prone to atrial fibrillation (AF) compared to those with heart failure with reduced ejection fraction (HFrEF). Nevertheless, a risk prediction model for new-onset atrial fibrillation (NOAF) in HFpEF patients remains a notable gap, especially with respect to imaging indicators.MethodsWe retrospectively analyzed 402 HFpEF subjects reviewed at the Affiliated Hospital of Qingdao University from 2017 to 2023. Cox regression analysis was performed to screen predictors of NOAF. A nomogram was constructed based on these factors and internally validated through the bootstrap resampling method. A performance comparison between the nomogram and the mC2HEST score was performed.ResultsOut of the 402 participants, 62 (15%) developed atrial fibrillation. The risk factors for NOAF were finally screened out to include age, chronic obstructive pulmonary disease (COPD), hyperthyroidism, renal dysfunction, left atrial anterior–posterior diameter (LAD), and pulmonary artery systolic pressure (PASP), all of which were identified to create the nomogram. We calculated the bootstrap-corrected C-index (0.819, 95% CI: 0.762–0.870) and drew receiver operator characteristic (ROC) curves [3-year areas under curves (AUC) = 0.827, 5-year AUC = 0.825], calibration curves, and clinical decision curves to evaluate the discrimination, calibration, and clinical adaptability of the six-factor nomogram. Abstract Background Increasing evidence indicates that four and a half LIM domains 2 (FHL2) plays a crucial role in the progression of various cancers. However, the biological functions and molecular mechanism of FHL2 in lung adenocarcinoma (LUAD) remain unclear. Methods We evaluated the prognostic value of FHL2 in LUAD using public datasets and further confirmed its prognostic value with our clinical data. The biological functions of FHL2 in LUAD were evaluated by in vitro and in vivo experiments. Pathway analysis and rescue experiments were subsequently performed to explore the molecular mechanism by which FHL2 promoted the progression of LUAD. Results FHL2 was upregulated in LUAD tissues compared to adjacent normal lung tissues, and FHL2 overexpression was correlated with unfavorable outcomes in patients with LUAD. FHL2 knockdown significantly suppressed the proliferation, migration and invasion of LUAD cells, while FHL2 overexpression had the opposite effect. Mechanistically, FHL2 upregulated the PI3K/AKT/mTOR pathway and subsequently inhibited autophagy in LUAD cells. The effects FHL2 on the proliferation, migration and invasion of LUAD cells are dependent on the inhibition of autophagy, as of induction autophagy attenuated the aggressive phenotype induced by FHL2 overexpression. Conclusions FHL2 promotes the progression of LUAD by activating the PI3K/AKT/mTOR pathway and subsequently inhibiting autophagy, which can be exploited as a potential therapeutic target for LUAD.

Abstract The utilisation of offshore wind turbines has rapidly increased in the last decade, which has resulted in a steady increase in wind turbine sizes. The global average offshore wind turbine size has increased from 1.5 MW to 6 MW in the last two decades. The research community has started to investigate huge 10 to 15 MW offshore wind turbines in recent years, resulting in the study of very innovative floating wind turbines using various substructure technologies. With this backdrop, this paper will investigate and thoroughly compare the power performance of extreme load effects of a large offshore 10 MW turbine installed on the monopile, spar, and semisubmersible substructures. This is performed by using the average conditional exceedance rate (ACER) and Gumbel methods to predict the extreme responses under the operating conditions of 8, 12, and 16 m/s mean wind speed, representing the below‐rated, rated, and above‐rated regions, respectively. The results show that the power performance and extreme loads experienced depends significantly on the operating regions. The mean power generation between the three different types of offshore wind turbines (OWTs) are closely in the whole operating range, which standard deviations differ significantly. Large standard deviations of power generation appear in the spar turbine under the below‐rated condition. Further, it was observed that the spar wind turbine generally experiences larger extreme loads due to larger platform pitch motion. In addition, the ACER method shows a better prediction for the 1, 2 and 5‐year extreme responses than the Gumbel method, which is due to the relatively poor data fitting of the Gumbel method at the upper tail. The study is believed to consolidate and close the knowledge gap in understanding wind turbine responses across the most common offshore substructure technologies and provide a basis for design and deployment of OWTs.

Plastic mulch plays a crucial role in agricultural production in arid and semi-arid regions, positively impacting crop yields, salt suppression, and seedling protection. However, as the usage of plastic mulch extends over time, residue accumulation becomes a significant issue in these regions. To clarify the effects of plastic mulch residue on soil fungi, this study focused on three typical cotton-growing areas in Xinjiang. Using high-throughput sequencing technology, the study analyzed the changes in the fungal community structure and diversity in rhizosphere and non-rhizosphere soils across 27 cotton fields under three different levels of plastic mulch residue: 0–75 kg/ha, 75–150 kg/ha, and 150–225 kg/ha. The results indicated that Ascomycota and Basidiomycota were the dominant fungal phyla across all treatments. Increasing levels of plastic mulch residue reduced the fungal richness in the soil, with a greater effects observed on rhizosphere fungi compared to bulk soil fungi. The α-diversity of cotton rhizosphere fungi showed an increasing trend, followed by a decrease with increasing plastic mulch residue, in Aksu and Bazhou, peaking at 75–150 kg/ha. Conversely, in Changji, the α-diversity decreased with increasing plastic mulch residue. The α-diversity of non-rhizosphere fungi associated with cotton decreased with increasing plastic mulch residue. Plastic mulch residue significantly altered the soil fungal α-diversity and had a greater effects on rhizosphere fungi. Different levels of plastic mulch residue had varying effects on the β-diversity of rhizosphere and non-rhizosphere fungi, significantly influencing rhizosphere fungi in Aksu and Bazhou and non-rhizosphere fungi in Changji and Bazhou. Overall, different levels of plastic mulch residue exerted varying degrees of influence on the community composition and diversity of soil fungi associated with cotton, potentially reducing the fungal richness and altering the community structure with increasing residue levels.

The purpose of this study was to explore the protective effect of SeMet on renal injury induced by AFB1 in rabbits and its molecular mechanism. Forty rabbits of 35 days old were randomly divided into control group, AFB1 group (0.3 mg AFB1/kg b.w), 0.2 mg/kg Se + AFB1 group (0.3 mg AFB1/kg b.w + 0.2 mg SeMet/kg feed) and 0.4 mg/kg Se + AFB1 group (0.3 mg AFB1/kg b.w + 0.4 mg SeMet/kg feed). The SeMet treatment group was fed different doses of SeMet diets every day for 21 days. On the 17–21 day, the AFB1 treatment group, the 0.2 mg/kg Se + AFB1 group and the 0.4 mg/kg Se + AFB1 group were administered 0.3 mg AFB1 /kg b.w by gavage (dissolved in 0.5 ml olive oil) respectively. The results showed that AFB1 poisoning resulted in the changes of renal structure, the increase of renal coefficient and serum biochemical indexes, the ascent of ROS and MDA levels, the descent of antioxidant enzyme activity, and the significant down-regulation of Nrf2, HO-1 and NQO1. Besides, AFB1 poisoning increased the number of renal apoptotic cells, rised the levels of PTEN, Bax, Caspase-3 and Caspase-9, and decreased the levels of PI3K, AKT, p-AKT and Bcl-2. In summary, SeMet was added to alleviate the oxidative stress injury and apoptosis of kidney induced by AFB1, and the effect of 0.2 mg/kg Se + AFB1 is better than 0.4 mg/kg Se + AFB1.

Compared to conventional vertical wells, horizontal wells with Ultra-Long horizontal sections greatly increase the oil drainage area and can significantly increase oil and gas production from individual wells. In order to analyze the limit of horizontal well extension, an extension limit model was developed, which takes into account formation, rated pump pressure, rated pressure and string buckling, and is used to predict the limit of horizontal well extension under different constraints. The results show that increasing the inside and outside diameter of drill pipe can effectively reduce pressure loss and facilitate the extension of the horizontal section while keeping the hole size unchanged. As the cuttings density, cuttings diameter, mud density, mud performance, pump rates, and ROP increase, the limit extension length of the horizontal section gradually decreases. As the WOB, RPM, friction factor increase, the torque gradually increases and the limit extension length of the horizontal section gradually decreases. Selecting the right combination of drilling tools can reduce the torque, which is beneficial for mechanical extension. This study is instructive for the limit of horizontal well extension with ultra-long horizontal sections in the field.

Soil salinization has a significant impact on agricultural production and ecology. There is an urgent demand to establish an effective method that monitors the spatial and temporal distribution of soil salinity. In this study, a multi-indicator soil salinity monitoring model was proposed for monitoring soil salinity in Bachu County, Kashgar Region, Xinjiang, from 2002 to 2022. The model was established by combining multiple predictors (spectral, salinity, and composite indices and topographic factors) and the accuracy of the four models (Random Forest [RF], Partial Least Squares [PLS], Classification Regression Tree [CART], and Support Vector Machine [SVM]) was compared. The results reveal the high accuracy of the optimized prediction model, and the order of the accuracy is observed as RF > PLS > CART > SVM. The most accurate model, RF, exhibited an R2 of 0.723, a root mean square error (RMSE) of 2.604 g·kg−1, and a mean absolute error (MAE) of 1.95 g·kg−1 at a 0–20 cm depth. At a 20–40 cm depth, RF had an R2 value of 0.64, an RMSE of 3.62 g·kg−1, and an MAE of 2.728 g·kg−1. Spatial changes in soil salinity were observed throughout the study period, particularly increased salinization from 2002 to 2012 in the agricultural and mountainous areas within the central and western regions of the country. However, salinization declined from 2012 to 2022, with a decreasing trend in salinity observed in the top 0–20 cm of soil, followed by an increasing trend in salinity at a 20–40 cm depth. The proposed method can effectively extract large-scale soil salinity and provide a practical basis for simplifying the remote sensing monitoring and management of soil salinity. This study also provides constructive suggestions for the protection of agricultural areas and farmlands.

Engineering projects are confronted with many problems resulting from overbreak in tunnel blasting, necessitating the optimization of design parameters to minimize overbreak. In this study, an AI-based model for overbreak prediction and optimization is proposed, aiming to mitigate the hazards associated with overbreak. Firstly, the Extreme Gradient Boosting (XGBoost) model is integrated with three distinct metaheuristic algorithms, namely Particle Swarm Optimization (PSO), Whale Optimization Algorithm (WOA), and Sparrow Search Algorithm (SSA), respectively. Consequently, the hyperparameters are optimized, and the performance of predictions is enhanced. Meanwhile, to overcome the limitations of a small dataset and enhance the generalization ability of the three developed models, a 5-fold cross-validation is employed. Then, the performance of the different models with five distinct swarm sizes is evaluated via four metrics, including coefficient of determination (R2), mean square error (MSE), mean absolute error (MAE), and variance accounted for (VAF). Subsequently, by comparing the aforementioned developed models, the optimal prediction model with the highest accuracy can be obtained, which is then used for parameter optimization research. Finally, individual studies are conducted to address the issue of overbreak caused by the adoption of identical blasting parameters due to geological variations, aiming to minimize overbreak in different sections of the tunnel. By comparing the optimization abilities of PSO, WOA, and SSA, the objective of finding the minimum value of overbreak within a short timeframe is achieved. The results indicate that the model developed in this study accurately predicts overbreak, and effectively optimizes blast parameters for different sections of the tunnel.

Genome-wide analyses of maize populations have clarified the genetic basis of crop domestication and improvement. However, limited information is available on how breeding improvement reshaped the genome in the process of the formation of heterotic groups. In this study, we identified a new heterotic group (X group) based on an examination of 512 Chinese maize inbred lines. The X group was clearly distinct from the other non-H&L groups, implying that X × HIL is a new heterotic pattern. We selected the core inbred lines for an analysis of yield-related traits. Almost all yield-related traits were better in the X lines than those in the parental lines, indicating that the primary genetic improvement in the X group during breeding was yield-related traits. We generated whole-genome sequences of these lines with an average coverage of 17.35× to explore genome changes further. We analyzed the identity-by-descent (IBD) segments transferred from the two parents to the X lines and identified 29 and 28 IBD conserved regions (ICRs) from the parents PH4CV and PH6WC, respectively, accounting for 28.8% and 12.8% of the genome. We also identified 103, 89, and 131 selective sweeps (SSWs) using methods that involved the π, Tajima’s D, and CLR values, respectively. Notably, 96.13% of the ICRs co-localized with SSWs, indicating that SSW signals concentrated in ICRs. We identified 171 annotated genes associated with yield-related traits in maize both in ICRs and SSWs. To identify the genetic factors associated with yield improvement, we conducted QTL mapping for 240 lines from a DH population (PH4CV × PH6WC, which are the parents of X1132X) for ten key yield-related traits and identified a total of 55 QTLs. Furthermore, we detected three QTL clusters both in ICRs and SSWs. Based on the genetic evidence, we finally identified three key genes contributing to yield improvement in breeding the X group. These findings reveal key loci and genes targeted during pedigree breeding and provide new insights for future genomic breeding.

On Douyin, China's domestic version of TikTok, straight users are being recommended queer content, and videos created by straight men are being recommended to queer viewers. As Douyin recommends videos based on users’ online activities (e.g., liking, sharing, or spending time watching a video) and networks (e.g., connections made on the platform), sexuality comes to be algorithmically interpreted and defined. This process differs from an understanding of gender and sexuality as more or less fixed classifications that are the result of what people register when entering a platform concerning their gender and sexual identifications. This article analyzes viewers’ and creators’ experiences and reflections on the algorithmic grouping of sexual orientation and erotic curiosity through the relational lens of configurations. Using two years of online observational data and in-depth interviews with 18 Douyin users, both straight and same-sex orientated, we found that a language-centered, semiotic approach alone cannot assist in capturing the reconfiguration of sexual identifications presently occurring in China, and most likely elsewhere. Sexual identifications are the result of a relational process in which desires (regardless of sexual orientation) and intimacy, content creation and consumption, platform vernaculars and affordances, and data and algorithms converge and clash. This process allows for an erotic curiosity that has not yet been named or normalized in language systems and therefore reconfigures how sexual identity or orientation come to be understood in relation to the ever-increasing presence of computational power.

To obtain timely, accurate, and reliable information on wheat yield dynamics. The UAV DJI Wizard 4-multispectral version was utilized to acquire multispectral images of winter wheat during the tasseling, grouting, and ripening periods, and to manually acquire ground yield data. Sixteen vegetation indices were screened by correlation analysis, and eight textural features were extracted from five single bands in three fertility periods. Subsequently, models for estimating winter wheat yield were developed utilizing multiple linear regression (MLR), partial least squares (PLS), BP neural network (BPNN), and random forest regression (RF), respectively. (1) The results indicated a consistent correlation between the two variable types and yield across various fertility periods. This correlation consistently followed a sequence: heading period > filling period > mature stage. (2) The model’s accuracy improves significantly when incorporating both texture features and vegetation indices for estimation, surpassing the accuracy achieved through the estimation of a single variable type. (3) Among the various models considered, the partial least squares (PLS) model integrating texture features and vegetation indices exhibited the highest accuracy in estimating winter wheat yield. It achieved a coefficient of determination (R2) of 0.852, a root mean square error (RMSE) of 74.469 kg·hm−2, and a normalized root mean square error (NRMSE) of 7.41%. This study validates the significance of utilizing image texture features along with vegetation indices to enhance the accuracy of models estimating winter wheat yield. It demonstrates that UAV multispectral images can effectively establish a yield estimation model. Combining vegetation indices and texture features results in a more accurate and predictive model compared to using a single index.

ObjectiveBile reflux plays a key role in the development of gastric intestinal metaplasia (GIM), an independent risk factor of gastric cancer. Here, we aimed to explore the biological mechanism of GIM induced by bile reflux in a rat model.MethodsRats were treated with 2% sodium salicylate and allowed to freely drink 20 mmol/L sodium deoxycholate for 12 weeks, and GIM was confirmed by histopathological analysis. Gastric microbiota was profiled according to the 16S rDNA V3–V4 region, gastric transcriptome was sequenced, and serum bile acids (BAs) were analyzed by targeted metabolomics. Spearman's correlation analysis was used in constructing the network among gastric microbiota, serum BAs, and gene profiles. Real-time polymerase chain reaction (RT-PCR) measured the expression levels of nine genes in the gastric transcriptome.ResultsIn the stomach, deoxycholic acid (DCA) decreased the microbial diversity but promoted the abundances of several bacterial genera, such as Limosilactobacillus, Burkholderia–Caballeronia–Paraburkholderia, and Rikenellaceae RC9 gut group. Gastric transcriptome showed that the genes enriched in gastric acid secretion were significantly downregulated, whereas the genes enriched in fat digestion and absorption were obviously upregulated in GIM rats. The GIM rats had four promoted serum BAs, namely cholic acid (CA), DCA, taurocholic acid, and taurodeoxycholic acid. Further correlation analysis showed that the Rikenellaceae RC9 gut group was significantly positively correlated with DCA and RGD1311575 (capping protein-inhibiting regulator of actin dynamics), and RGD1311575 was positively correlated with Fabp1 (fatty acid-binding protein, liver), a key gene involved in fat digestion and absorption. Finally, the upregulated expression of Dgat1 (diacylglycerol acyltransferase 1) and Fabp1 related to fat digestion and absorption was identified by RT-PCR and IHC.ConclusionDCA-induced GIM enhanced gastric fat digestion and absorption function and impaired gastric acid secretion function. The DCA–Rikenellaceae RC9 gut group–RGD1311575/Fabp1 axis might play a key role in the mechanism of bile reflux-related GIM.

Since 2016, China’s online LGBTQ platforms, such as Blued and Aloha, have been seeking to increase user acquisition and retention by introducing new functionalities to their interfaces. Although these attempts were promising at first, most of their endeavors proved unsustainable, largely due to the condition in which their businesses operate: a niche audience segment in a winner-take-all mobile market and a highly regulatory market in which LGBTQ content is often made a site for the execution of power. This article investigates the now-defunct role-playing social game Werewolf embedded in Aloha, one of the popular dating apps for queer men in China. By bringing together scholarship on game studies and queer media studies, it is argued that social games embedded in dating apps foster a new form of sexual sociality in which desires become increasingly gamified and intimacy networked, which are essential for queer social media to retain users. Although Werewolf was ultimately closed due to Aloha’s budgetary controls, queer players have emerged as a promising market for their unique engagement in social games. LGBTQ platforms, however, are not the main beneficiary of the queer gaming market because of the winner-take-all mobile ecosystem. The article highlights the inherent limits of “outward” expansion of China’s LGBTQ platforms into the mainstream market and suggests that prioritizing the unaddressed needs of their niche audience through an inward approach may be a more viable strategy for business growth in a mobile ecosystem dominated by a handful of major players.

Asexual spore serves as essential inoculum of rice blast during the disease cycle, and differentiation of young conidium from conidiophore is intimately regulated by cell cycle. Mih1 encodes a dual-specificity phosphatase that involved in the G2/M transition of the mitotic cell cycle by regulating the Cdk1 activity in eukaryotes. Till now, the roles of Mih1 homologue, however, remain unclear in Magnaporthe oryzae. We here functionally characterized the Mih1 homologue MoMih1 in M. oryzae. MoMih1 is localized to both the cytoplasm and nucleus and can physically interact with the CDK protein MoCdc28 in vivo. Loss of MoMih1 led to delayed nucleus division and a high level of Tyr15 phosphorylation of MoCdc28. The MoMih1 mutants showed retarded mycelial growth with a defective polar growth, less fungal biomass, and shorter distance between diaphragms, compared with the KU80. Asexual reproduction altered in MoMih1 mutants, with both abnormal conidial morphogenesis and decreased conidiation. The MoMih1 mutants severely attenuated the virulence to host plants due to the impaired ability of penetration and biotrophic growth. The incapability of scavenging of host-derived reactive oxygen species, which was possibly ascribed to the severely decreased extracellular enzymes activities, were partially associated with deficiency of pathogenicity. Besides, the MoMih1 mutants displayed also improper localization of retromer protein MoVps26 and polarisome component MoSpa2, and defects of cell wall integrity (CWI), melanin pigmentation, chitin synthesis, and hydrophobicity. In conclusion, our results demonstrate that MoMih1 plays pleiotropic roles during fungal development and plant infection of M. oryzae.

The global average size of offshore wind turbines has increased steadily from 1.5 MW to 6 MW from 2000 to 2020. With this backdrop, the research community has recently looked at huge 10–15 MW class floating offshore wind turbines (FOWTs). The larger rotor, nacelle structure and tower have more significant structural flexibility. The larger structural flexibility, controller dynamics, aerodynamics, hydrodynamics, and various environmental conditions result in complex structural responses. The structural load effects of a very large FOWT could be more severe than that of the lower MW classes. Accurate quantification of the extreme dynamic responses of FOWT systems is essential in the design of the Ultimate Limit State (ULS) due to the fully-coupled interaction between the FOWT system and environmental conditions. Motivated by this, extreme responses of the 10 MW semi-submersible type FOWT are investigated using the average conditional exceedance rate (ACER) and Gumbel methods. Three operating conditions representing below-rated (U = 8 m/s), rated (U = 12 m/s) and above-rated (U = 16 m/s) regions were considered. The aim is to guide future research on large FOWTs by indicating the expected ULS loads.

IntroductionStaphylococcus epidermidis infections are an important concern in worldwide, especially when associated with biofilms, and resistance of this agent to many drugs makes the situation even worse. We investigated the inhibitory effect of Disaspidin BB obtained from plant extracts and purifications on clinical S. epidermidis strains and their biofilms, and preliminarily investigated its mechanism of of its anti-biofilm activity.Methods and ResultsThe broth dilution method was used to determine the minimum inhibitory concentrations (MIC) of Disaspidin BB on 11 clinical S. epidermidis strains (MIC value of 0.63 ~ 2.5 μg/ml). SEP-05 was found to be erythromycin-resistant (MIC value>8 μg/ml) and Disaspidin BB sensitive with an MIC value of 0.63 μg/ml. The time-kill curve assay indicated that the antibacterial activity of Disaspidin BB against SEP-05 with concentration dependence. The metabolic activity and total biomass of the drug-treated SEP-05 biofilm in each stage were significantly inhibited by the crystalline violet and XTT assay, and the scavenging effect of Disaspidin BB on SEP-05 biofilm was also confirmed by SEM observation. The results of real-time quantitative PCR showed that subinhibitory concentrations Disaspidin BB can inhibit biofilm formation by affecting the expression level of key genes (aap, atlE, icaA, luxS, recA) in SEP-05 biofilm formation. In addition, the content of polysaccharides, proteins and extracellular DNA in biofilm matrix after the intervention of Disaspidin BB was significantly reduced, and it was tentatively determined that the ability of SEP-05 biofilm formation and its stability were thus disturbed.DiscussionThe results show that Disaspidin BB has promising antibacterial effect on erythromycin-resistant S. epidermidis and significant scavenging effect on its biofilm, which provides a theoretical basis for the further development of BB as a new drug for the treatment of skin infections caused by S. epidermidis.

Genic male sterility (GMS) is one of the most important resources for exploiting heterosis in crop breeding, so that identifying genomic loci regulating GMS is desirable. However, many regulatory genes controlling GMS have not yet been characterized in maize, owing partly to a lack of genetic materials. We generated a recessive male-sterile maize mutant in the Jing 724 genetic background via ethyl methanesulfonate treatment, and found the male sterility to be due to a single gene mutation. Bulk-segregant RNA sequencing of three replicates indicated that one genomic region located at the end of chromosome 4 was associated with the observed mutant phenotype. Among genes with nonsynonymous mutations, Zm00001d053895 (bHLH51) showed abolished expression in the sterile bulks and was annotated as a bHLH transcription factor orthologous to Arabidopsis AMS, suggesting an association with the male sterility of the mutant. Kompetitive Allele-Specific PCR assays further validated the exclusive correlation of male sterility with the single C-to-T mutation in the fifth exon. The new maize mutant and the potential SNP locus provide novel genetic material for investigating the molecular mechanism underlying tapetal development and may facilitate the improvement of hybrid production systems.

Abstract In fuel cells, the generally low oxygen reduction reactions (ORR) rate at the cathode is a major factor limiting the overall performance of fuel cell, so the development of high‐performance, stable and inexpensive ORR electrocatalysts is a major researching direction. Nano‐electrocatalysts, especially one‐dimensional (1D) electrocatalysts, exhibit good catalytic behavior in fuel cell reactions due to their unique anisotropic structure, large specific surface area, high elasticity and excellent stability. To date, many advanced 1D electrocatalysts have been reported for optimizing the cathode ORR of fuel cells. Therefore, a comprehensive review of the structural design of 1D nano‐electrocatalysts and a systematic analysis of their enhanced performance still need further summarized and concluded. In this review, we firstly introduce the main synthetic methods of 1D electrocatalysts, and then discuss the recent advances and advantages of different structured 1D electrocatalysts involving nanofibers, nanotubes, nanorods, nanochains, nanowires, nanobelts, nanoneedles, nanowhiskers, and coaxial nanocables. In addition, we also introduce 1D electrocatalysts while highlighting advanced strategies and preparation methods for these different structured 1D electrocatalysts to improve ORR performance. Finally, we prospect the future development of 1D ORR electrocatalysts.

Abstract This study compares the dynamic behaviour of a conventional and a compact gearbox for the DTU 10 MW wind turbine supported on a monopile offshore structure. The conventional gearbox configuration is composed of two planetary epicyclic stages and one parallel stage, while the compact gearbox configuration consists of a fixed planetary stage and a differential compound epicyclic stage. The design methodology for these two gearboxes is described, and the final gearbox specifications show a lighter weight and smaller volume for the compact gearbox design compared to the conventional one. Computational gearbox models are established using the multi‐body system dynamic analysis method. A decoupled approach is employed for the gearbox load effect analysis. Comparisons of the dynamic behaviour between these two gearboxes are conducted under pure torque load cases, tangential pin position error conditions and non‐torque load cases. The results demonstrate that the compact gearbox has better dynamic performance under different torque load cases and is more robust to withstand the effects of manufacturing errors and rotor non‐torque loads compared to the conventional gearbox. It is believed that the proposed compact gearbox concept is promising and would be a good alternative for multi‐megawatt floating wind turbines, although challenging with respect to the design and operation complexity.

Satellite signals are easily lost in urban areas, which causes difficulty in vehicles being located with high precision. Visual odometry has been increasingly applied in navigation systems to solve this problem. However, visual odometry relies on dead-reckoning technology, where a slight positioning error can accumulate over time, resulting in a catastrophic positioning error. Thus, this paper proposes a road-network-map-assisted vehicle positioning method based on the theory of pose graph optimization. This method takes the dead-reckoning result of visual odometry as the input and introduces constraints from the point-line form road network map to suppress the accumulated error and improve vehicle positioning accuracy. We design an optimization and prediction model, and the original trajectory of visual odometry is optimized to obtain the corrected trajectory by introducing constraints from map correction points. The vehicle positioning result at the next moment is predicted based on the latest output of the visual odometry and corrected trajectory. The experiments carried out on the KITTI and campus datasets demonstrate the superiority of the proposed method, which can provide stable and accurate vehicle position estimation in real-time, and has higher positioning accuracy than similar map-assisted methods.

Abstract O-GalNAc glycans (or mucin O-glycans) play pivotal roles in diverse biological and pathological processes, including tumor growth and progression. Structurally defined O-GalNAc glycans are essential for functional studies but synthetic challenges and their inherent structural diversity and complexity have limited access to these compounds. Herein, we report an efficient and robust chemoenzymatic modular assembly (CEMA) strategy to construct structurally diverse O-GalNAc glycans. The key to this strategy is the convergent assembly of O-GalNAc cores 1–4 and 6 from three chemical building blocks, followed by enzymatic diversification of the cores by 13 well-tailored enzyme modules. A total of 83 O-GalNAc glycans presenting various natural glycan epitopes are obtained and used to generate a unique synthetic mucin O-glycan microarray. Binding specificities of glycan-binding proteins (GBPs) including plant lectins and selected anti-glycan antibodies towards these O-GalNAc glycans are revealed by this microarray, promoting their applicability in functional O-glycomics. Serum samples from colorectal cancer patients and healthy controls are assayed using the array reveal higher bindings towards less common cores 3, 4, and 6 than abundant cores 1 and 2, providing insights into O-GalNAc glycan structure-activity relationships.

Extreme value prediction of the load-effect responses of complex offshore structures such as the floating wind turbine (FWT) is crucial in ultimate limit state (ULS) design. This paper considers two cases to understand the feasibility of the bivariate correction on the extreme load and motion responses of a 10-MW semi-submersible type FWT. The empirical anchor tension force and surge motion used in this study are obtained from the FAST simulation tool (developed by the National Renewable Energy Laboratory) with the load cases stimulated at under-rated, rated and above rated speeds. Then, the bivariate correction method is applied to model FWT extreme response for a 5-years return period prediction with a 95% confidence interval (CI), based on just 2 min short response record. The proposed methodology permits accurate correction of the bivariate extreme value in case of, for example, corrupted measurement sensor data. Based on the proposed novel method’s performance, it is concluded that the bivariate correction method can offer better robust and precise bivariate predictions of coupled surge motion and anchor tension of the FWT.

Whether or not diabetic choroidopathy (DC) is involved in the initiation and progression of diabetic ocular complications needs to be explored. We included 54 diabetic eyes from 36 diabetic patients, and 54 healthy eyes from 32 control subjects after propensity scores matching. As a multi-target drug to treat ischemic stroke, N-butylphthalide (NBP) is extremely water-insoluble and exhibits limited oral bioavailability, impeding its wide oral application. Effective treatment of ischemic stroke by NBP requires timely and efficient drug exposure, necessitating the development of new oral formulations. Herein, liposomes containing biosurfactant sodium cholate (CA-liposomes) were systemically investigated as an oral NBP delivery platform because of its high biocompatibility and great potential for clinical applications. The optimized liposomes have a uniform hydrodynamic size of 104.30 ± 1.60 nm and excellent encapsulation efficiency (93.91 ± 1.10%). Intriguingly, NBP-loaded CA-liposomes produced rapid drug release and the cumulative release was up to 88.09 ± 4.04% during 12 h while that for NBP group was only 6.79 ± 0.99%. Caco-2 cell monolayer assay demonstrated the superior cell uptake and transport efficiency of NBP-loaded CA-liposomes than free NBP, which was mediated by passive diffusion via transcellular and paracellular routes. After oral administration to rats, NBP-loaded CA-liposomes exhibited rapid and almost complete drug absorption, with a tmax of 0.70 ± 0.14 h and an absolute bioavailability of 92.65% while NBP suspension demonstrated relatively low bioavailability (21.7%). Meanwhile, NBP-loaded CA-liposomes produced 18.30-fold drug concentration in the brain at 5 min compared with NBP suspension, and the brain bioavailability increased by 2.48-fold. As expected, NBP-loaded CA-liposomes demonstrated significant therapeutic efficacy in a middle cerebral artery occlusion rat model. Our study provides new insights for engineering oral formulations of NBP with fast and sufficient drug exposure against ischemic stroke in the clinic.

Although Fraxinellone (Frax) isolated from Dictamnus albus L. possessed excellent anti-hepatic fibrosis activity, oral administration of Frax suffered from the inefficient therapeutic outcome in vivo due to negligible oral absorption. At present, the oral formulation of Frax is rarely exploited. For rational formulation design, we evaluated preabsorption risks of Frax and found that Frax was rather stable while poorly dissolved in the gastrointestinal tract (78.88 μg/mL), which predominantly limited its oral absorption. Further solubility test revealed the outstanding capacity of cyclodextrin derivatives (CDs) to solubilize Frax (6.8–12.8 mg/mL). This led us to study the inclusion complexes of Frax with a series of CDs and holistically explore their drug delivery performance. Characterization techniques involving 1H-NMR, FT-IR, DSC, PXRD, and molecular docking confirmed the most stable binding interactions when Frax complexed with 6-O-α-D-maltosyl-β-cyclodextrin (G2-β-CD-Frax). Notably, G2-β-CD-Frax exhibited the highest solubilizing capacity, fast dissolution rate, and superior Caco-2 cell internalization with no obvious toxicity. Pharmacokinetic studies demonstrated markedly higher oral bioavailability of G2-β-CD-Frax (5.8-fold that of free drug) than other Frax-CDs. Further, long-term administration of G2-β-CD-Frax (5 mg/kg) efficiently inhibited CCl4-induced hepatic fibrosis in the mouse without inducing any toxicity. Our results will inspire the continued advancement of optimal oral Frax formulations for anti-fibrotic therapy.

Abstract Background Stalk fracture caused by strong wind can severely reduce yields in maize. Stalks with higher stiffness and flexibility will exhibit stronger lodging resistance. However, stalk flexibility is rarely studied in maize. Stalk fracture of the internode above the ear before tasseling will result in the lack of tassel and pollen, which is devastating for pollination in seed production. In this study, we focused on stalk lodging before tasseling in two maize inbred lines, JING724 and its improved line JING724A1 and their F2:3 population. Results JING724A1 showed a larger stalk fracture angle than JING724, indicating higher flexibility. In addition, compared to JING724, JING724A1 also had longer and thicker stalks, with a conical, frustum-shaped internode above the ear. Microscopy and X-ray microcomputed tomography of the internal stalk architecture revealed that JING724A1 had more vascular bundles and thicker sclerenchyma tissue. Furthermore, total soluble sugar content of JING724A1, especially the glucose component, was substantially higher than in JING724. Using an F2:3 population derived from a JING724 and JING724A1 cross, we performed bulk segregant analysis for stalk fracture angle and detected one QTL located on Chr3: 14.00–19.28 Mb. Through transcriptome data analysis and ∆ (SNP-index), we identified two candidate genes significantly associated with high stalk fracture angle, which encode a RING/U-box superfamily protein (Zm00001d039769) and a MADS-box transcription factor 54 (Zm00001d039913), respectively. Two KASP markers designed from these two candidate genes also showed significant correlations with stalk fracture angle. Conclusions The internode shape and glucose content are possibly correlated with stalk flexibility in maize. Two genes in the detected QTL are potentially associated with stalk fracture angle. These novel phenotypes and associated loci will provide a theoretical foundation for understanding the genetic mechanisms of lodging, and facilitate the selection of maize varieties with improved flexibility and robust lodging resistance.

Abstract Background Chronic obstructive pulmonary disease (COPD) is the third leading cause of death in the United States with no effective treatment. The current diagnostic method, spirometry, does not accurately reflect the severity of COPD disease status. Therefore, there is a pressing unmet medical need to develop noninvasive methods and reliable biomarkers to detect early stages of COPD. Lipids are the fundamental components of cell membranes, and dysregulation of lipids was proven to be associated with COPD. Lipidomics is a comprehensive approach to all the pathways and networks of cellular lipids in biological systems. It is widely used for disease diagnosis, biomarker identification, and pathology disorders detection relating to lipid metabolism. Methods In the current study, a total of 25 serum samples were collected from 5 normal control subjects and 20 patients with different stages of COPD according to the global initiative for chronic obstructive lung disease (GOLD) (GOLD stages I ~ IV, 5 patients per group). After metabolite extraction, lipidomic analysis was performed using electrospray ionization mass spectrometry (ESI-MS) to detect the serum lipid species. Later, the comparisons of individual lipids were performed between controls and patients with COPD. Orthogonal projections to latent structures discriminant analysis (OPLS-DA) and receiver operating characteristic (ROC) analysis were utilized to test the potential biomarkers. Finally, correlations between the validated lipidomic biomarkers and disease stages, age, FEV1% pack years and BMI were evaluated. Results Our results indicate that a panel of 50 lipid metabolites including phospholipids, sphingolipids, glycerolipids, and cholesterol esters can be used to differentiate the presence of COPD.
