Abstract Background This study was conducted to investigate the long-term outcomes of laparoscopic total gastrectomy (LTG) versus open total gastrectomy (OTG) in patients with advanced gastric cancer (AGC) after neoadjuvant chemotherapy (NACT). Methods Patients with AGC who received NACT before surgery were enrolled in either the LTG or OTG group. Propensity score matching (PSM) (1:2) was performed between the two groups based on the propensity score using a 0.15 calliper width. Three-year overall survival (OS) and disease-free survival (DFS) were compared between these two groups before and after PSM. OS and DFS rates were calculated by the Kaplan‒Meier method, and any differences in survival were evaluated with a log-rank test. Univariate and multivariate Cox proportional hazards analyses were used to estimate the simultaneous effects of prognostic factors on survival and the hazard ratio (HR) between LTG and OTG patients. Results A total of 144 patients completed the follow-up, with 24 patients in the LTG group and 120 patients in the OTG group. After a mean follow-up of 64.40 months, there were no significant differences in the 3-year OS or DFS rates between the two groups before (P = 0.453 and P = 0.362, respectively) or after PSM (P = 0.972 and P = 0.884, respectively). Multivariate Cox proportional hazards analysis indicated that ypN stage was an independent risk factor for worse OS (P = 0.013). Conclusions This study showed that LTG with D2 lymphadenectomy performed by an experienced surgical team resulted in comparable 3-year OS and DFS compared with OTG in patients with AGC after NACT. BackgroundPostoperative complications may lower the quality of life of patients, consequently leading to a reduction in their overall survival (OS). In our previous investigations, we found that patients with gastric cancer (GC) with postoperative complications who underwent direct surgery had a significantly lower OS than patients without complications. We observed no significant difference in OS among patients who underwent neoadjuvant chemotherapy (NAC), regardless of complications. We propose that for patients who underwent reoperation following NAC, downstaging (reduction of clinical stage) and postoperative complications exerted contrasting effects on the OS. Further, we hypothesize that post-NAC downstaging and the absence of postoperative complications lead to a longer OS.MethodsWe conducted a retrospective analysis to collect the clinical data of patients with GC who underwent surgery after receiving NAC at the First Hospital of Lanzhou University from January 2016 to December 2022. Based on the presence of a post-NAC downstaging period and postoperative complications, we categorized the patients into group A (downstaging without complications), group B (downstaging with complications), group C (non-downstaging with complications), and group D (non-downstaging without complications). First, we assessed the OS disparity between the groups. Subsequently, we performed a comparative analysis of the body composition and hematological indexes of patients from the four groups.ResultsWe included 295 patients in the study and categorized them into four subgroups: group A comprised 83 patients (28.1%), group B comprised 32 patients (10.8%), group C comprised 83 patients (28.1%), and group D comprised 97 patients (32.9%). Group A patients had the longest OS of 40.1 ± 20.53, whereas group C patients had the shortest OS of 32.15 ± 25.09. The OS of patients in the other two groups was between these values. Pairwise comparisons revealed significant differences between the OS of group A patients and that of groups C (32.15 ± 25.09) and D (33.06 ± 20.89) patients (p 0.05). With respect to hematological indexes, the prognostic nutritional index (PNI) was highest in group A and lowest in group C. The PNI in group A (417.89 ± 37.58) was significantly higher than that in group C (397.62 ± 47.56) (p 0.05).ConclusionPatients with advanced GC who experienced post-NAC downstaging and no postoperative complication had the longest OS. Patients with better body composition demonstrated more significant downstaging, fewer postoperative complications, and a longer OS.

Abstract Translational control exerts immediate effect on the composition, abundance, and integrity of the proteome. Ribosome-associated quality control (RQC) handles ribosomes stalled at the elongation and termination steps of translation, with ZNF598 in mammals and Hel2 in yeast serving as key sensors of translation stalling and coordinators of downstream resolution of collided ribosomes, termination of stalled translation, and removal of faulty translation products. The physiological regulation of RQC in general and ZNF598 in particular in multicellular settings is underexplored. Here we show that ZNF598 undergoes regulatory K63-linked ubiquitination in a CNOT4-dependent manner and is upregulated upon mitochondrial stresses in mammalian cells and Drosophila. ZNF598 promotes resolution of stalled ribosomes and protects against mitochondrial stress in a ubiquitination-dependent fashion. In Drosophila models of neurodegenerative diseases and patient cells, ZNF598 overexpression aborts stalled translation of mitochondrial outer membrane-associated mRNAs, removes faulty translation products causal of disease, and improves mitochondrial and tissue health. These results shed lights on the regulation of ZNF598 and its functional role in mitochondrial and tissue homeostasis.

Background Nonylphenol (NP) is widely recognized as a crucial environmental endocrine-disrupting chemical and persistent toxic substance. The remediation of NP-contaminated sites primarily relies on biological degradation. Compound microbial products, as opposed to pure strains, possess a greater variety of metabolic pathways and can thrive in a wider range of environmental conditions. This characteristic is believed to facilitate the synergistic degradation of pollutants. Limited research has been conducted to thoroughly examine the potential compatibility of compound microbial agents with indigenous microflora, their ability to function effectively in practical environments, their capacity to enhance the dissipation of NP, and their potential to improve soil physicochemical and biological characteristics. Methods In order to efficiently eliminate NP in contaminated soil in an eco-friendly manner, a simulation study was conducted to investigate the impact of bioaugmentation using the functional compound microbial agent NP-M2 at varying concentrations (50 and 200 mg/L) on the dynamics of the soil microbial community. The treatments were set as follows: sterilized soil with 50 mg/kg NP (CK50) or 200 mg/kg NP (CK200); non-sterilized soil with 50 mg/kg NP (TU50) or 200 mg/kg NP (TU200); non-sterilized soil with the compound microbial agent NP-M2 at 50 mg/kg NP (J50) or 200 mg/kg NP (J200). Full-length 16S rRNA analysis was performed using the PacBio Sequel II platform. Results Both the indigenous microbes (TU50 and TU200 treatments) and the application of NP-M2 (J50 and J200 treatments) exhibited rapid NP removal, with removal rates ranging from 93% to 99%. The application of NP-M2 further accelerated the degradation rate of NP for a subtle lag period. Although the different treatments had minimal impacts on the soil bacterial α-diversity, they significantly altered the β-diversity and composition of the bacterial community. The dominant phyla were Proteobacteria (35.54%–44.14%), Acidobacteria (13.55%–17.07%), Planctomycetes (10.78%–11.42%), Bacteroidetes (5.60%–10.74%), and Actinobacteria (6.44%–8.68%). The core species were Luteitalea_pratensis, Pyrinomonas_methylaliphatogenes, Fimbriiglobus_ruber, Longimicrobium_terrae, and Massilia_sp003590855. The bacterial community structure and taxon distribution in polluted soils were significantly influenced by the activities of soil catalase, sucrase, and polyphenol oxidase, which were identified as the major environmental factors. Notably, the concentration of NP and, to a lesser extent, the compound microbial agent NP-M2 were found to cause major shifts in the bacterial community. This study highlights the importance of conducting bioremediation experiments in conjunction with microbiome assessment to better understand the impact of bioaugmentation/biostimulation on the potential functions of complex microbial communities present in contaminated soils, which is essential for bioremediation success.

To address the lack of reliable measurement methods for identifying wear mechanisms and predicting the state of mechanical seal tribo-parts, this study proposes a method for characterizing tribological behavior based on measuring face vibration acceleration. It aims to uncover the source mechanism of mechanical seal face vibration acceleration influenced by tribology and dynamic behavior. This research delves into the dynamic behavior characteristics and vibration acceleration of the mechanical seal stationary ring. We explored the variation pattern of face vibration acceleration root mean square (RMS) with rotation speed, sealing medium pressure, and face surface roughness. The results indicate that under constant medium pressure, an increase in rotation speed leads to a decrease in acceleration RMS and an increase in face temperature. Similarly, under constant rotation speed, an increase in medium pressure results in nonlinear changes in acceleration RMS, forming an “M” shape, along with an increase in face temperature. Furthermore, under conditions of constant medium pressure and rotation speed, an increase in the surface roughness of the rotating ring face corresponds to an increase in acceleration RMS and face temperature. Upon starting the mechanical seal, both acceleration RMS and temperature initially increase before decreasing, a trend consistent with the Stribeck curve.

Spring-energized seals demonstrate good sealing performance over a wide range of pressures and temperatures and can compensate for installation eccentricity, high-temperature aging, etc. However, as a contact seal, its polytetrafluoroethylene (PTFE) jacket material is easily worn during the rotation of the end face, which leads to a decline in sealing performance and, ultimately, seal failure. Based on the Archard wear model, a performance prediction model of the spring-energized seal was established by combining tests and numerical analyses. In order to improve the tribological performance of spring-energized seals made of PTFE, varied fillers were added to modify the PTFE, and the tribological and mechanical properties of PTFE composites with varied fillers were measured in experiments. Using a performance prediction model for spring-energized seals, the variation in the friction performance of seals made of these filled PTFEs during the operating cycle was analyzed. The results showed that the performance prediction model can accurately simulate this variation. After a certain amount of wear, the deviation between the simulated data and the experimental data was within ±5%. Compared with spring-energized seals made of pure PTFE, the friction torque of spring-energized seals made of GF/PTFE was reduced by 28.97% at most, and the friction torque reduction rate was lowered by 22.25%.

Abstract Background Peritoneal dialysis (PD) remains limited due to dialysis failure caused by peritoneal fibrosis. Tamoxifen (TAM), an inhibitor of estrogen receptor 1 (ESR1), has been reported to treat fibrosis, but the underlying mechanism remains unknown. In this study, we sought to explore whether tamoxifen played an anti-fibrotic role by affecting transcription factor ESR1. Methods ESR1 expression was detected in the human peritoneum. Mice were daily intraperitoneally injected with 4.25% glucose PD dialysate containing 40 mM methylglyoxal for 2 weeks to establish PD-induced peritoneal fibrosis. Tamoxifen was administrated by daily gavage, at the dose of 10 mg/kg. Chromatin immunoprecipitation (ChIP) and dual‐luciferase reporter assay were performed to validate ESR1 bound H19 promoter. Gain-of-function and loss-of-function experiments were performed to investigate the biological roles of H19 on the mesothelial-mesenchymal transition (MMT) of human peritoneal mesothelial cells (HPMCs). Intraperitoneal injection of nanomaterial-wrapped 2′-O-Me-modified small interfering RNA was applied to suppress H19 in the mouse peritoneum. RNA immunoprecipitation and RNA pull-down assays demonstrated binding between H19 and p300. Exfoliated peritoneal cells were obtained from peritoneal dialysis effluent to analyze the correlations between ESR1 (or H19) and peritoneal solute transfer rate (PSTR). Results ESR1 was increased significantly in the peritoneum after long-term exposure to PD dialysate. Tamoxifen treatment ameliorated high glucose-induced MMT of HPMCs, improved ultrafiltration rate, and decreased PSTR of mouse peritoneum. Tamoxifen reduced the H19 level by decreasing the ESR1 transcription of H19. Depletion of H19 reversed the pro-fibrotic effect of high glucose while ectopic expression of H19 exacerbated fibrotic pathological changes. Intraperitoneal injection of nanomaterial-wrapped 2′-O-Me-modified siRNAs targeting H19 mitigated PD-related fibrosis in mice. RNA immunoprecipitation (RIP) and RNA pull-down results delineated that H19 activated VEGFA expression by binding p300 to the VEGFA promoter and inducing histone acetylation of the VEGFA promoter. ESR1 and H19 were promising targets to predict peritoneal function. Conclusions High glucose-induced MMT of peritoneal mesothelial cells in peritoneal dialysis via activating ESR1. In peritoneal mesothelial cells, ESR1 transcribed the H19 and H19 binds to transcription cofactor p300 to activate the VEGFA. Targeting ESR1/H19/VEGFA pathway provided new hope for patients undergoing peritoneal dialysis. Graphic Abstract

Abstract Background Porcine epidemic diarrhea (PED) is a contagious intestinal disease caused by porcine epidemic diarrhea virus (PEDV) characterized by vomiting, diarrhea, anorexia, and dehydration, which has caused huge economic losses around the world. However, it is very hard to find completely valid approaches to control the transmission of PEDV. At present, vaccine immunity remains the most effective method. To better control the spread of PED and evaluate the validity of different immunization strategies, 240 PED outbreak cases from 577 swine breeding farms were collected and analyzed. The objective of the present study was to analyze the epidemic regularity of PEDV and evaluate two kinds of different immunization strategies for controlling PED. Results The results showed that the main reasons which led to the outbreak of PED were the movement of pig herds between different pig farms (41.7%) and delaying piglets from the normal production flow (15.8%). The prevalence of PEDV in the hot season (May to October) was obviously higher than that in the cold season (January to April, November to December). Results of different vaccine immunity cases showed that immunization with the highly virulent live vaccine (NH-TA2020 strain) and the commercial inactivated vaccine could significantly decrease the frequency of swine breeding farms (5.9%), the duration of PED epidemic (1.70 weeks), and the week batches of dead piglets (0.48 weeks weaned piglets), compared with immunization with commercial attenuated vaccines and inactivated vaccine of PED. Meanwhile, immunization with the highly virulent live vaccine and the commercial inactivated vaccine could bring us more cash flows of Y̶275,274 per year than immunization with commercial live attenuated vaccine and inactivated vaccine in one 3000 sow pig farm within one year. Conclusion Therefore, immunization with highly virulent live vaccine and inactivated vaccine of PED is more effective and economical in the prevention and control of PED in the large-scale swine farming system.

Abstract Background The prognosis of gastric cancer (GC) patients with positive peritoneal cytology (CY1) without other distant metastasis is poor, and there are no standard treatment strategies. Our study aimed to compare the survival outcomes of CY1 GC patients receiving chemotherapy or surgery as initial treatment. Methods From February 2017 to January 2020, clinical and pathological data of patients diagnosed with CY1 GC without other distant metastasis in the Peking University Cancer Hospital was reviewed. Patients were divided into two groups: chemotherapy-initial group and surgery-initial group. In chemotherapy-initial group, patients received preoperative chemotherapy initially. According to the treatment response, the patients were divided into three subgroups: conversion gastrectomy group, palliative gastrectomy group, and further systematic chemotherapy group. In surgery-initial group, patients underwent gastrectomy followed by postoperative chemotherapy. Results A total of 96 CY1 GC patients were included with 48 patients in each group. In chemotherapy-initial group, preoperative chemotherapy yielded an objective response rate of 20.8% and disease control rate of 87.5%. Conversion to CY0 after preoperative chemotherapy was obtained in 24 (50%) patients. The median overall survival was 36.1 months in chemotherapy-initial group and 29.7 months in surgery-initial group (p = 0.367). The median progression-free survival was 18.1 months in chemotherapy-initial group and 16.1 months in surgery-initial group (p = 0.861). The 3-year overall survival rates were 50.0% and 47.9%, respectively. In chemotherapy-initial group, twenty-four patients who converted to CY0 by preoperative chemotherapy and received surgery obtained a significantly better prognosis. The median overall survival was still not reached in these patients. Conclusion There was no significant difference in survival outcomes between chemotherapy-initial group and surgery-initial group. CY1 GC patients who converted to CY0 by preoperative chemotherapy and received radical surgery could obtain a favorable long-term prognosis. Further investigation should focus on preoperative chemotherapy to eliminate peritoneal cancer cell. Trial registration This study is retrospectively registered.

Corrosion inhibitors play a vital role in impeding the corrosion process of steel bars within concrete structures exposed to corrosive environments. Nevertheless, conventional corrosion inhibitors pose environmental risks. In contrast, contemporary studies have explored corrosion inhibitors that are eco-friendly. However, these inhibitors are burdened by high costs and complex production processes, impeding the widespread application in concrete structures. Consequently, this study presents an innovative solution by incorporating uniconazole, an agricultural fungicide, as a corrosion inhibitor for steel bars in concrete structures. The steel bars were exposed to corrosion within a simulated concrete pore solution containing 0.6 mol/L NaCl, both with and without the presence of uniconazole. The morphology and hydrophilicity of the steel bar surface were investigated via optical microscope and contact angle experiments. Electrochemical tests (open circuit potential, potentiodynamic polarization, electrochemical impedance spectroscopy, and Mott-Schottky analysis) and X-ray photoelectron spectroscopy were employed to investigate the corrosion inhibition performance and mechanism of uniconazole. The results demonstrate that uniconazole elevates the hydrophobicity and contributes to the corrosion inhibition of steel bars. Electrochemical test results indicate that as the concentration of uniconazole increases from 1 × 10−4 mol/L to 1 × 10−3 mol/L, the inhibition efficiency likewise demonstrates a corresponding increase, escalating from around 50 %–90 %. Uniconazole molecules function as mixed-type inhibitors, exhibiting characteristics of both anode-type and cathode-type inhibitors. The adsorption of uniconazole enhances the stability and thickness of the passive-adsorbed layer on the steel surface, effectively impeding the charge transfer process and obstructing the interaction of corrosive substances with the base metal. In summary, the application of uniconazole exhibits the highlights of efficient, cost-effective, environmentally friendly, and the potential for scalable production. This positions uniconazole as a promising candidate for use as a corrosion inhibitor in the domain of concrete structures.

Diatomaceous earth (DE), a kind of natural and environmentally friendly concrete admixture with good pozzolanic and water absorption properties, is a potential high-quality internal curing material. DE internal curing agent was prepared by calcining excavated diatomaceous earth and applied in cement-based materials with a low water–binder ratio of 0.35 to study its effects on the autogenous shrinkage and early crack resistance of the materials. The results showed that DE was a kind of microfine powder with a unique and orderly microporous structure, and its main component was SiO2, which had good water-holding and -releasing properties. Pre-absorbed DE could effectively improve the internal relative humidity of cement-based materials with a low water–binder ratio. Under the standard of not reducing the 28 d compressive strength of mortar, compared to the benchmark group without DE, when 1% of cement is replaced by DE of equal mass, DE could effectively reduce the 7 d autogenous shrinkage of mortar by about 36.7% and delay the initial cracking time of mortar under the restraint condition of the ring by 10.7%. Therefore, it is an internal curing material with excellent performance.

The hydraulic concrete in the alpine region is subjected to alternating actions of freeze–thaw (F) and abrasion (W) during operation, resulting in significant deterioration of concrete durability. In this paper, the water/binder ratio (W/B) was employed as the test variable, the working condition F group and W group were set as the control group, and the working condition F-W group was used as the test group. Fast-freezing and underwater methods are used for the alternating test. By measuring the mass loss, relative dynamic elastic modulus (RDEM), surface morphological characteristics, fractal dimension of concrete in each alternating cycle, and the evolution law of concrete performance under the alternating action of F and W was explored. The results show that compared with the control group, the alternating action will accelerate the mass loss of concrete, reduce the RDEM, and cause the deterioration of surface wear. The maximum increase in mass loss and RDEM of concrete is 1.92% and 20.11%, respectively. During this process, the fractal dimension of the concrete increases as the number of alternating cycles increases, but it still does not exceed the limit of 2.4. In addition, a relationship function between the fractal dimension and the mass loss rate, volume loss, was established. It was found that the experimental group had a good linear correlation, and the correlation was close to 95%, which was about 20% higher than that of the control group.

A model based on a three-dimensional fractal function is developed and used in conjunction with experiments to analyze the evolutionary pattern of sealing performance during the start-up process of dynamic pressure seals, and the influence of end-face microscopic features on the evolution law is discussed. It is found that the opening state of the seal is divided into three stages: the non-opened stage, transition stage, and full-opened stage. The isotropic dimensions of the cavities have a coupling effect on the leakage, and they diminish as the speed increases. In order to enhance the sealing performance during start-up, it is suggested that the seal faces have a fractal dimension of 2.4 to 2.6, and a characteristic factor of less than 1 × 10−9 m.

Key Clinical Message Systemic lupus erythematosus is difficult to diagnose in patients who are antinuclear antibody (ANA) negative and lack typical clinical manifestations. For such patient who presented ANA‐negative severe lupus‐like manifestations, the diagnosis and treatment are a huge challenge. Histological findings may provide clues to diagnosis. Abstract Systemic lupus erythematosus (SLE) is a multisystem autoimmune disease characterized by formation of autoantibodies to nuclear and cytoplasmic antigens. It was reported that a small subset of patients had typical clinical features of SLE with consistently negative antinuclear antibody (ANA), but such disease is usually mild and rarely involves multisystem. At present, there are no reports about severe lupus with ANA continued negative. Our report describes a 34‐year‐old Chinese woman who presented renal failure, multiple serous cavity effusion, and epilepsy, without malar rash, photosensitivity, lymphopenia, and arthritis. Further renal biopsy pathology revealed lupus‐like nephritis. Autoantibodies, including ANA, antibodies against Smith and against double stranded DNA, were negative. Such a ANA negative and lack of typical clinical symptoms of SLE patient, but with severe lupus‐like manifestations, whether it was lupus or not is worth discussing.

Abstract Background Laparoscopic gastrectomy (LG) for gastric cancer has rapidly developed and become more popular in recent decades. Additional high-quality randomized controlled trial (RCT) studies comparing LG versus open gastrectomy (OG) for gastric cancer (GC) have been published in recent years. An updated systematic review is warranted. The aim of our meta-analysis was to comprehensively evaluate the short- and long-term outcomes of LG versus OG for GC. Materials and methods The PubMed, Embase, Web of Science, and Cochrane Center Register of Controlled Trials databases were comprehensively searched to identify RCTs comparing LG versus OG for GC published between January 1994 and December 7, 2021. This study was conducted in accordance with the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) and Cochrane Collaboration and the Quality of Reporting of Meta-analyses (QUORUM) guidelines. All RCTs comparing the short- and long-term outcomes of LG with those of OG were included. A random effects model was adopted with significant heterogeneity (I 2 > 50%), while a fixed effects model was employed in all other cases (I 2 ≤ 50%). Results A total of 26 RCTs with 8301 patients were included in this meta-analysis. The results indicated that the intraoperative complication rate was comparable between the LG group and the OG group (OR=1.14, 95% CI [0.76, 1.70], I 2=0%, p=0.53). Abstract Background The safety and short-term outcomes of gastrectomy after preoperative chemotherapy plus immunotherapy (PCIT) versus preoperative chemotherapy (PCT) in patients with advanced gastric cancer (AGC) remain unclear. This study was conducted to compare the safety and short-term efficacy of PCIT with those of PCT in patients with AGC. Methods We retrospectively reviewed the data of patients with AGC who received PCIT or PCT at Peking University Cancer Hospital and Institute Gastrointestinal Cancer Center I between January 2019 and June 2021. The clinical characteristics were recorded, and short-term oncological outcomes were compared. IntroductionPorcine circovirus type 2 (PCV2) is the primary etiological agent of porcine circovirus diseases (PCVD), which are widespread in most pig herds, causing huge economic losses in the global pig industry. Therefore, it is critical to assess the infection characteristics of PCV2 in different swine herds to develop effective strategies against PCVD.MethodsIn this study, routine diagnostic and monitoring protocols were used to collect 12,714 samples from intensive farms in China, and PCV2 was tested for by qPCR to determine positivity rates and viral loads in samples from different herds and materials.ResultsPCV2 was found to be prevalent throughout China, and fattening farms had higher positivity rates than breeding farms. The PCV2 positivity rates in breeding farms in Southern China were higher than those in Northern China. Growing–finishing pigs demonstrated the highest positivity rate in the tested samples, while pre-weaning piglets and adult sows had the lowest. Meanwhile, samples with viral loads exceeding 106 copies/mL in growing–finishing pigs had 27.2% positivity, compared to 1.9% and 3.3% in sows and piglets, respectively. The results of the viral loads in the serum samples followed a similar trend.DiscussionThe findings reveal that PCV2 circulates in different herds from intensive farms, with positivity increasing from pre-weaning to growing–finishing herds. It is urgent to develop effective strategies to reduce PCV2 positivity in growing–finishing herds and prevent viral circulation among pigs.

Abstract Aim To determine the tolerance and acceptance of hepatic venous pressure gradient (HVPG) measurements in patients with liver cirrhosis. Methods This prospective international multicenter study included 271 patients with cirrhosis who were scheduled to undergo HVPG measurement between October 2019 and June 2020. Data related to the tolerance and acceptance of HVPG measurements were collected using descriptive questionnaires. Objective: To summarize and think about the surgical process of Da Vinci Xi™ robot-assisted liver resection. Methods: To summarize the clinical data, preoperative preparations, surgical procedures, and postoperative recovery of a patient who underwent Da Vinci Xi™ robot-assisted left hepatectomy in the third ward of the Department of Hepatobiliary and Pancreatic Surgery of the Second Affiliated Hospital of Kunming Medical University in 2021. Discussion to guide future clinical work. Results: The left hemihepatectomy was performed safely and effectively on patients with left intrahepatic bile duct stones with the assistance of Da Vinci Xi™ robot. Conclusion: Da Vinci Xi™ robot's three-dimensional field of vision, stable and flexible manipulator operation and other advantages make it suitable for liver resection surgery.

Background:. HVPG measurement is the gold standard for assessing portal hypertension. Many patients decline HVPG measurements due to associated pain. According to previous studies, propofol sedation during HVPG measurements potentially alters HVPG readings. However, opioid analgesics’ effects on HVPG await full elucidation. This study aimed to evaluate fentanyl analgesia’s effects on HVPG measurement accuracy in patients with cirrhosis. Methods:. This prospective, multicenter study included patients with cirrhosis undergoing HVPG measurements, which were performed preanalgesia and under analgesia with fentanyl injection (1.0 or 1.5 μg/kg). Results:. Of the 48 enrolled patients with cirrhosis, 23 were administered 1.0 μg/kg fentanyl analgesia during HVPG measurement. The HVPG was 13.4±4.9 mm Hg in preanalgesia and 13.5±5.2 mm Hg under analgesia. HVPG measurement accuracy was not altered after fentanyl analgesia (p = 0.801). The following measures also did not change: heart rate (p = 0.132), mean arterial pressure (p = 0.348), and blood oxygen saturation (p = 0.748); however, respiratory rate (p = 0.001) changes occurred. The Verbal Numerical Rating Score for comfort under analgesia was higher than that in preanalgesia (p = 0.001). Twenty-five patients were administered 1.5 μg/kg fentanyl analgesia during HVPG measurement. The HVPG was 19.5±5.7 mm Hg in preanalgesia and 19.6±5.6 mm Hg under analgesia. HVPG measurement accuracy did not alter after fentanyl analgesia (p = 0.469). Similarly, the following measures did not change: mean arterial pressure (p = 0.871) and oxygen saturation (p = 0.327); nevertheless, respiratory rate (p = 0.015) and heart rate (p = 0.019) changes occurred. The Verbal Numerical Rating Score for comfort under analgesia was higher than that in preanalgesia (p < 0.001). Conclusion:. Fentanyl analgesia did not alter HVPG measurement accuracy, and fentanyl improved comfort in patients with cirrhosis during HVPG measurements.

The physicochemical property, volatile flavor compounds, and microbial community structure of Jinhua fatty ham (FH) and lean ham (LH) were investigated and compared by high-throughput sequencing and HS-GC-IMS. Results showed that FH had higher pH and slightly lighter and yellower color than LH. Meanwhile, 33 volatile flavor compounds were identified from FH and LH, among which LH showed higher abundance of total alcohols and acids, but FH had generally richer aldehydes, ketones, esters, heterocyclic, and sulfur-containing compounds. Moreover, FH and LH did not have significant difference in α-diversity of bacterial community, but LH presented a much lower α-diversity of fungal community than FH. Besides, the dominant microorganisms (relative abundance >2%) in FH were Ruminococcaceae UCG-005, Staphylococcus, Ruminococcaceae UCG-014, Meyerozyma, and Aspergillus at the genus level, while in LH were Staphylococcus, Psychrobacter, Halomonas, Propionicicella, Ruminococcaceae UCG-005, Meyerozyma, Yamadazyma, and Aspergillus. Furthermore, the analysis of Pearson’s correlation and metabolic network confirmed that the discriminative flavor compounds of FH were mainly β-oxidation and degradation products of fatty acids, while those of LH were mostly derived from the Strecker reaction or microbial metabolism of amino acids. The present study could help understand the potential pathway of characteristic microorganisms affecting flavor formation of fat-deficient dry-cured hams and provide theoretical supports for developing healthier fermented meat products.

Abstract Amyloid precursor protein (APP) metabolism is central to Alzheimer’s disease (AD) pathogenesis, but the key etiological driver remains elusive. Recent failures of clinical trials targeting amyloid-β (Aβ) peptides, the proteolytic fragments of amyloid precursor protein (APP) that are the main component of amyloid plaques, suggest that the proteostasis-disrupting, key pathogenic species remain to be identified. Previous studies suggest that APP C-terminal fragment (APP.C99) can cause disease in an Aβ-independent manner. The mechanism of APP.C99 pathogenesis is incompletely understood. We used Drosophila models expressing APP.C99 with the native ER-targeting signal of human APP, expressing full-length human APP only, or co-expressing full-length human APP and β-secretase (BACE), to investigate mechanisms of APP.C99 pathogenesis. Key findings are validated in mammalian cell culture models, mouse 5xFAD model, and postmortem AD patient brain materials. We find that ribosomes stall at the ER membrane during co-translational translocation of APP.C99, activating ribosome-associated quality control (RQC) to resolve ribosome collision and stalled translation. Stalled APP.C99 species with C-terminal extensions (CAT-tails) resulting from inadequate RQC are prone to aggregation, causing endolysosomal and autophagy defects and seeding the aggregation of amyloid β peptides, the main component of amyloid plaques. Genetically removing stalled and CAT-tailed APP.C99 rescued proteostasis failure, endolysosomal/autophagy dysfunction, neuromuscular degeneration, and cognitive deficits in AD models. Our finding of RQC factor deposition at the core of amyloid plaques from AD brains further supports the central role of defective RQC of ribosome collision and stalled translation in AD pathogenesis. These findings demonstrate that amyloid plaque formation is the consequence and manifestation of a deeper level proteostasis failure caused by inadequate RQC of translational stalling and the resultant aberrantly modified APP.C99 species, previously unrecognized etiological drivers of AD and newly discovered therapeutic targets.

This study aims to explore the temperature field and mechanical characteristics of hydraulic tunnels during operation in an area with large temperature differences throughout a year. By relying on the measured displacement field change results of a water transmission tunnel in Xinjiang, a finite element calculation model applicable to the hydraulic tunnel-surrounding rock under the action of large temperature differences was established. Then, the FEM model was used to analyze the temperature field distribution and stress field variation of hydraulic tunnel-surrounding rock under the effect of large temperature differences. The results showed that, under the action of large temperature differences, the displacement of the cave top was the largest, followed by the cave bottom, and finally the cave waist. The obtained displacement field data using the calculation model in this study were basically consistent with the on-site measured data, and the displacement field change law was the same, indicating that the established finite element calculation model is reliable. When the ambient temperature was below zero, the freezing front gradually expanded to the depth of the surrounding rock along the tunnel radial direction, while the frozen rock at the intersection of the lining and the surrounding rock melted first when the ambient temperature was above zero. The stress in various parts of the surrounding rock of the hydraulic tunnel increased continuously. To be specific, the growth rate of the tensile stress at the cave bottom, the tensile stress at the cave top, and the compressive stress at the cave waist of the tunnel reached 0.052 MPa/year, 0.053 MPa/year, and 0.096 MPa/year, respectively, in the first year, but the increasing rate subsequently tended to slow down continuously. Therefore, in actual projects, attention should be focused on the effect of large temperature differences on the stress growth of tunnel-surrounding rocks at an early stage, especially the growth of compressive stress at the cave waist. Notably, there are a large number of symmetrical and asymmetric phenomena related to engineering in this study.

Abstract Background The prognostic values of preoperative tumor markers (TMs) remain elusive in patients with locally advanced gastric cancer (LAGC) after neoadjuvant chemotherapy treatment (NACT). This study aimed to assess and establish a novel scoring system incorporating carcinoembryonic antigen (CEA), carbohydrate antigen 19-9 (CA19-9), carbohydrate antigen 72-4 (CA72-4) to enhance prognostic accuracy for progression-free survival (PFS) and pathological response (pCR). Methods Patients' data were retrospectively analyzed from December 2006 to December 2017 in our center. The cutoff value of TMs was determined using the time-dependent receiver operating test characteristics method. These three TMs were allocated 1 point each for the post neoadjuvant chemotherapy combination of tumor markers (post-NACT CTM) scores. The training group comprised 533 patients, responsible for full analysis, and the validation group comprised 137 patients based on the selection protocol. Results Of 533 enrolled patients, 138, 233, 117, and 45 patients scored 0, 1, 2, 3 respectively. ObjectiveLiver cancer is a significant contributor to global burden of cancer. Transcatheter arterial chemoembolization (TACE) is the standard of care for patients with unresectable liver cancer, and CalliSpheres, as novel drug-eluting bead (DEB) microspheres, have been found to be associated with a high tumor response rate. However, the outcomes after DEB-TACE treatment are not always satisfactory with tumor recurrence. Herein, we attempt to compare the clinical efficacy and safety of DEB-TACE with sorafenib and conventional TACE in treating advanced liver cancer.MethodsThe study retrospectively reviewed clinical records of 96 patients with liver cancer, among which there were 48 cases receiving DEB-TACE with sorafenib and 48 cases receiving conventional TACE. The physical properties of Callispheres were evaluated in HepG2 cells and a B6/J mouse model.ResultsDEB-TACE with Callispheres were demonstrated to effectively maintain stability and prolong the half-life of epirubicin. Compared with the patients receiving conventional TACE, those receiving DEB-TACE with sorafenib exhibited better patient outcomes with increased survival rate, reduced tumor volume, and declined levels of tumor markers. Additionally, DEB-TACE with Callispheres could effectively protect liver function, as well as reduce the toxic effects of loaded epirubicin, and its combination with sorafenib would not increase the incidence of adverse reactions.ConclusionDEB-TACE using CalliSpheres combined with sorafenib could prevent the progression of liver cancer and bring a better prognosis.

Chemical fertilizer reduction combined with novel and green agricultural inputs has become an important practice to improve microecological health in agricultural production. Given the close linkages between rhizosphere processes and plant nutrition and productivity, understanding how fertilization impacts this critical zone is highly important for optimizing plant–soil interactions and crop fitness for agricultural sustainability. Here, by using a pot experimental system, we demonstrated that nitrogen fertilizer reduction and microbial agent application promoted plant fitness and altered the microbial community structure in the rhizosphere soil with the following treatments: no fertilization, CK; conventional chemical fertilizer, CF; 30% reduced nitrogen fertilizer, N; 30% reduced nitrogen fertilizer with pure γ-PGA, PGA; 30% reduced nitrogen fertilizer with Bacillus subtilis A-5, A5; 30% reduced nitrogen fertilizer with γ-PGA fermentation broth, FJY. The PGA, A5, and FJY treatments all significantly promoted crop growth, and the FJY treatment showed the strongest positive effect on Chinese cabbage yield (26,385.09 kg/hm2) (P < 0.05). Microbial agents affected the α diversity of the rhizosphere bacterial community; the addition of B. subtilis A-5 (A5 and FJY treatments) significantly affected rhizospheric bacterial community structure. Urease activity and soil pH were the key factors affecting bacterial community structure and composition. The FJY treatment seemed to influence the relative abundances of important bacterial taxa related to metabolite degradation, predation, and nitrogen cycling. This discovery provides insight into the mechanism underlying the effects of microbial agent inputs on rhizosphere microbial community assembly and highlights a promising direction for the manipulation of the rhizosphere microbiome to yield beneficial outcomes.

Summary: Microalgae-based technology is an effective and environmentally friendly method for antibiotics-contaminated wastewater treatment. To assess the tolerance and removal ability of Chlorella sorokiniana to ciprofloxacin (CIP), this study comprehensively revealed the responses of C. sorokiniana to CIP exposure and its degradation processes through physiological and transcriptomic analyses. Although the photosynthetic system was inhibited, the growth of C. sorokiniana was not negatively affected by CIP. Dissolved organic matter was analyzed and indicated that humic-like substances were released to alleviate the stress of CIP. In addition, the maximum removal of CIP was 83.3% under 20 mg L−1 CIP exposure. HPLC-MS/MS and RNA-Seq analyses suggested that CIP could be bioaccumulated and biodegraded by C. sorokiniana through the reactions of hydroxylation, demethylation, ring cleavage, oxidation, dehydrogenation, and decarboxylation with the help of intracellular oxidoreductases, especially cytochrome P450. Collectively, this research shows that C. sorokiniana have a great potential for removing CIP from wastewater.

Abstract The diversity and community structure of soil fungi play an important role in crop production and ecosystem balance, especially in paddy-upland vegetable field systems. High-throughput sequencing was used to study changes in the soil fungal community structure and function in paddy-upland vegetable field systems. The results showed that compared with traditional planting, the diversity and community structure of soil fungi were changed by the combination of flooding and drought, the Shannon index increased by 11.07%, and the proportion of the dominant species, Mortierella, decreased by 22.74%. Soil available nitrogen, total phosphorus, available phosphorus, total nitrogen and organic matter played a leading role in the initial stage of the experiment, while the dominant factor changed to total potassium 3 years later and then to soil pH and water content 6 years later. FUNGuild analysis showed that the proportion of three independent trophic modes of soil fungi were increased by the combined flooded-drought model, and there were multiple interaction factors, For example, nutrient supply, pH and planting pattern. This study showed that soil fertility, crop yield and economic benefits were better than the traditional model after three years of planting and breeding. The longer the time, the better the effect.

The gas–liquid miscible backflow pumping seal (G-LMBPHS) is a non-contact mechanical seal that is suitable for high-speed bearing chambers. However, the tribological properties and wear mechanisms of the frictional pair of G-LMBPHS in an oil–air environment have not yet been comprehensively studied. In this study, the tribological properties of six frictional pairs, consisting of three hard materials (18Cr2Ni4WA, Al2O3 coating, and Cr2O3 coating) and two soft materials (metal-impregnated graphite [Metal-IG] and resin-impregnated graphite [Resin-IG]), were analyzed using a disc-on-disc tribometer. An oil–air environment was created using a minimal quantity lubrication (MQL) system and a closed chamber. The results show that the COF of the four frictional pairs consisting of two coatings and two graphites decreases gradually with increasing rotational speed, and the frictional pairs composed of Al2O3 coating and Resin-IG and Cr2O3 coating and Resin-IG have the lowest COF between 0.022 and 0.03. Therefore, the frictional pairs of G-LMBPHS are in a mixed lubrication condition. The lubricant in the oil–air environment is adsorbed and stored in pits on the surface of graphite and coatings, enhancing the hydrodynamic effect of the spiral grooves and reducing the COF by up to 45%. Metal-IG has better wear resistance than Resin-IG, and the frictional pair consisting of Cr2O3 coating and Metal-IG has the lightest wear. This study provides an important basis for the selection of G-LMBPHS frictional pairs in oil–air environments.

In this study, the mechanical behavior of fully grouted rock bolts in hydraulic tunnels subjected to elevated ground temperatures was investigated. A differential equation for axial displacement of the rock bolt was formulated, which considers the force equilibrium of infinitesimal bolt segments and the stress transfer mechanism at the anchor–rock interface. The distribution functions for axial stress within the bolt and the interfacial shear stress were obtained by solving the differential equation, which incorporated the displacement of the surrounding rock mass as a parameter. This study showed that the effectiveness of the bolt–shotcrete support system decreases over time, considering the displacement relaxation rate of the surrounding rock mass. The mechanical model’s variation laws at 20 °C, 50 °C, and 80 °C were summarized by integrating the thermal deformation equation for material parameters, and the numerical simulation results were compared and analyzed. The findings revealed that the bond strength between the rock bolt and the rock mass diminishes as the temperature of the surrounding rock increases, leading to a reduction of interfacial shear stress at both extremities of the bolt. Moreover, the maximum axial force within the bolt escalates as the neutral point migrates farther from the tunnel wall.

Summary: Environmental pollution caused by petroleum hydrocarbons is being paid more and more attention worldwide. Surfactants are able to improve the solubility of petroleum hydrocarbons, but their effects on petroleum hydrocarbon degradation in composting systems are still unclear. In this study, the effects on microbial community succession were investigated by adding petroleum hydrocarbons and rhamnolipids during composting of organic wastes. The results showed that the compost and the addition of rhamnolipids could effectively reduce the petroleum hydrocarbon content with an efficiency of 73.52%, compared to 53.81% for the treatment without addition. Network analyses and Structural Equation Model suggested that there were multiple potential petroleum degraders microbes that might be regulated by nitrogen. The findings in this study can also provide an implication for the treatment of petroleum hydrocarbon pollutants from oil-polluted soil, and the technology can be potentially applied on an industrial scale in practice.

Abstract Impregnated graphite has attracted considerable attention and has been widely used as an ideal friction material in many fields. However, the influence of the friction temperature on its tribological properties has not been clearly studied; furthermore, the evolution mechanism of transferred tribofilm is unknown. In this study, the tribological properties of impregnated graphite were investigated at different friction temperatures, and the evolution of the carbon-based tribofilm was also determined. The results revealed that the tribological properties significantly improved with an increase in friction temperature. The friction coefficient and wear depth of impregnated graphite reduced by 68% and 75%, respectively, at a high temperature of 160 °C compared with those of non-impregnated graphite. The significant properties of the impregnated graphite can be attributed to a transferred carbon-based tribofilm with an ordered structure induced by the friction temperature, which uniformly and stably adsorbs on friction interfaces. This study provides an important basis for designing graphite-based friction materials with improved properties suited for industrial applications.

The reliability and stability of the seal at the fuel-supply end of a rocket-engine turbopump are important factors in determining safety. Conventional single-stage floating rings used for the isolation-sealing of cryogenic media are highly susceptible to operational instability during startup and shutdown, which places demands on the seals’ structure, size, and material properties. In this study, a two-stage floating-ring isolation seal with a non-slotted main sealing surface was designed using a tangential air-intake mode. Based on the full-size three-dimensional finite-volume model, the leakage characteristics of the floating ring during operation were calculated, taking into account the effect of the “inlet effect” on the seal’s performance. Combining the temperature and pressure distributions of the sealing system under cryogenic operating conditions, calculated using a numerical simulation, a reliability analysis of various inlet directions and two kinds of floating-ring schemes was carried out on a self-constructed service-performance test bench. The results indicate that the main wear location of the non-slotted floating ring occurs on the auxiliary sealing surface, with stable working performance. When the inlet direction and spindle-rotation direction are the same, this is more conducive to ensuring the stability of seal performance in practical applications. The results of the current research are instructive for designing floating-ring isolation seals for turbine pumps.

A spring-energized seal, whose PTFE plastic shell has excellent self-lubrication and a low temperature stability, is used widely in liquid fuel valves’ rotating end-face seals. However, in practical application, temperature has a larger effect on not only the physical and tribological properties of materials, but also on the leakage performance of spring-energized rings. Thus, a high and low temperature sealing test of the spring-energized seal that applies to an engine was carried out. In this paper, the leakage characteristics, friction torque and wear characteristics of a spring-energized ring under different temperatures were studied. The friction torque at high temperature was less than that at normal temperature, and a low temperature could effectively reduce the wear amount of PTFE material. In order to study the influence of temperature on PTFE filled with graphite, the friction and wear test of PTFE-2 was carried out. The results showed that the amount of wear of PTFE-2 was only 27.8% of that at the normal temperature but the friction coefficient was three times larger when the temperature was −45 °C.

BackgroundsPerioperative chemotherapy (PEC) and neoadjuvant chemotherapy (NAC) have become a vital part of locally advanced gastric cancer (LAGC) treatment, but the optimal duration of PEC has not been studied. The aim of this study was to demonstrate the possibility of duration reduction in PEC in the adjuvant chemotherapy (AC) phase for ypN0 patients.MethodsWe included LAGC patients who achieved ypN0 after NAC in our institution from 2005 to 2018. The risk/benefit of AC and other covariates were majorly measured by overall survival (OS) and progression-free survival (PFS). We developed a survival-tree-based model to determine the optimal PEC duration for ypN0 patients in different classes.ResultsA total of 267 R0 resection patients were included. There were 55 patients who did not receive AC. The 5-year OS was 74.34% in the non-AC group and 83.64% in the AC group with a significant difference (p = 0.012). Multivariate Cox regression revealed that both AC (AC vs. non-AC: HR, 0.49; 95%CI, 0.27–0.88; p = 0.018) and ypT stages (ypT3-4 vs. ypT0-2: HR, 2.00; 95%CI, 1.11–3.59; p = 0.021) were significant protective/risk factors on patients OS and PFS. A decision tree model for OS indicated an optimal four to six cycles of PEC, which was recommended for ypT0-2N0 patients, while a minimum of five PEC cycles was recommended for ypT3-4N0 patients.ConclusionAC treatment is still necessary for ypN0. The duration reduction could be applied for the ypT0-2N0 stage patients but may not be suitable for higher ypT stages and beyond. A multicenter-based study is required.

Background and Objectives: This study aims to explore the safety of preoperative chemotherapy and clarify whether preoperative chemotherapy with oxaliplatin + S-1 (SOX) regimen and its adverse events are associated with higher risks of postoperative complications.Methods: We included consecutive patients with gastric cancer who underwent gastrectomy in our department between July 1 2018, and January 31 2020. Patients with preoperative SOX regimen chemotherapy were included in the analysis.Results: In the 343 included patients, 77 cases underwent preoperative chemotherapy. In total, surgical complications were found in 117 patients (34.1%), and there was no significant difference between the patients with and without preoperative chemotherapy before and after propensity score matching (p > 0.05, respectively). Multivariate analysis showed that preoperative comorbidities (p = 0.026) and the preoperative cT4b (p = 0.028) were independent risk factors in postoperative complications. In patients with preoperative chemotherapy, neither the occurrence of adverse events nor their severity was associated with postoperative complications (p > 0.05). However, the patients who received five to six cycles were more prone to postoperative complications than those who received three to four cycles (62.5 vs. 27.9%, OR = 4.306, 95% Cl = 1.282–14.464, p = 0.018).Conclusions: Occurrence of postoperative complications was not influenced by preoperative SOX chemotherapy. However, increased cycles of chemotherapy may lead to higher incidence of postoperative complications.

The Dendrobium officinale flower is a non-medicinal part of the plant, rich in a variety of nutrients and bioactive ingredients. The purpose of this article was to explore the preparation conditions of anthocyanins (ACNs) from the D. officinale flower. Subsequently, its anti-aging effects were evaluated with Caenorhabditis elegans. Results showed that the ACNs had antioxidant activities on scavenging free radicals (DPPH· and ABTS+·), and the clearance rate was positively correlated with the dose. Additionally, ACNs significantly increased the activity of superoxide dismutase (SOD) in C. elegans, which was 2.068-fold higher than that of the control. Treatment with ACNs at 150 μL extended the lifespan of C. elegans by 56.25%, and treatment with ACNs at 50 μL promoted fecundity in C. elegans. Finally, the protective effect of ACNs enhanced stress resistance, thereby increasing the survival numbers of C. elegans, which provided insights for the development and practical application of functional products.

The application of biogas slurry and chemical fertilizer in paddy fields can be a practical method to reduce the environmental risk and utilize the nutrients of biogas slurry. The responses of bacterial and fungal communities to the application of biogas slurry and chemical fertilizer are important reflections of the quality of the ecological environment. In this study, based on a 3-year field experiment with different ratios of biogas slurry and chemical fertilizer (applying the same pure nitrogen amount), the Illumina MiSeq platform was used to investigate the bacterial and fungal community diversity and composition in paddy soil. Our results revealed that compared with the observations under regular chemical fertilization, on the basis of stable paddy yield, the application of biogas slurry combined with chemical fertilizer significantly enhanced the soil nutrient availability and bacterial community diversity and reduced the fungal community diversity. Dissolved organic carbon (DOC), DOC/SOC (soil organic carbon), available nitrogen (AN) and available phosphorus (AP) were positively correlated with the bacterial community diversity, but no soil property was significantly associated with the fungal community. The bacterial community was primarily driven by the application of biogas slurry combined with chemical fertilizer (40.78%), while the fungal community was almost equally affected by the addition of pure biogas slurry, chemical fertilizer and biogas slurry combined with chemical fertilizer (25.65–28.72%). Biogas slurry combined with chemical fertilizer significantly enriched Proteobacteria, Acidobacteria, Planctomycetes, Rokubacteria, and Ascomycota and depleted Chloroflexi, Bacteroidetes, Crenarchaeota, Basidiomycota, and Glomeromycota. The observation of the alteration of some bacteria- and fungus-specific taxa provides insights for the proper application of biogas slurry combined with chemical fertilizer, which has the potential to promote crop growth and inhibit pathogens.

Abstract Background Pathological stage is considered as the best prognosis indicator for gastric cancer. With the increasing use of neoadjuvant chemotherapy (NACT), the latest TNM staging included a new pathological stage of ypTNM for patients with NACT. However, no study has investigated if ypTNM stage has the same prognostic implication as pTNM stage for gastric cancer. Methods We retrospectively selected eligible patients within a prospectively maintained database containing all patients treated with gastric cancer in Peking University Cancer Hospital from 2007 to 2015 using overall survival as the outcome. Patients using ypTNM and pTNM were 1:1 matched by propensity scores (PS) calculated from a model containing variables associated with ypTNM use or survival. Overall survival was compared by unconditional Cox regression. Conventional multivariate analysis was conducted to corroborate PS matching results. Results 1441 patients were included in the analysis with a median follow-up of 37 months (range = 2–106). The matched sample contained 756 patients. After PS matching, patients with specific ypTNM stage were 1.34 (95%CI = 1.05–1.72, P = 0.019) times more likely to die than patients with the same pTNM stage. Similar to the results of PS matching, multivariate Cox regression yielded a hazard ratio (HR) of 1.35 (95%CI = 1.09–1.67, P = 0.006). Subgroup analysis indicated this survival difference between ypTNM and pTNM stage varied by the specific TNM stage of patients. The HR was 3.44 (95%CI = 1.06–11.18, P = 0.040) and 1.28 (95%CI = 1.00–1.62, P = 0.048) for patients in stage I and III, respectively; whereas for stage II patients, no significant difference was observed (HR = 1.37, 95%CI = 0.78–2.38, P = 0.27). Conclusion Gastric cancer patients with specific ypTNM stage had worse prognosis compared to those at the same stage defined by pTNM.

Diabetic nephropathy (DN) is one of the main complications of diabetes and the main cause of diabetic end-stage renal disease, which is often fatal. DN is usually characterized by progressive renal interstitial fibrosis, which is closely related to the excessive accumulation of extracellular matrix and oxidative stress. Non-coding RNAs (ncRNAs) are RNA molecules expressed in eukaryotic cells that are not translated into proteins. They are widely involved in the regulation of biological processes, such as, chromatin remodeling, transcription, post-transcriptional modification, and signal transduction. Recent studies have shown that ncRNAs play an important role in the occurrence and development of DN and participate in the regulation of oxidative stress in DN. This review clarifies the functions and mechanisms of ncRNAs in DN-related oxidative stress, providing valuable insights into the prevention, early diagnosis, and molecular therapeutic targets of DN.

Two-dimensional (2D) nanomaterials have attracted much attention for lubrication enhancement of grease. It is difficult to disperse nanosheets in viscous grease and the lubrication performances of grease under harsh conditions urgently need to be improved. In this study, the 2D talc nanosheets are modified by a silane coupling agent with the assistance of high-energy ball milling, which can stably disperse in grease. The thickness and size of the talc nanosheet are about 20 nm and 2 µm. The silane coupling agent is successfully grafted on the surface of talc. Using the modified-talc nanosheet, the coefficient of friction and wear depth can be reduced by 40% and 66% under high temperature (150 °C) and high load (3.5 GPa), respectively. The enhancement of the lubrication and anti-wear performance is attributed to the boundary adsorbed tribofilm of talc achieving a repairing effect of the friction interfaces, the repairing effect of talc on the friction interfaces. This work provides green, economical guidance for developing natural lubricant additives and has great potential in sustainable lubrication.

BackgroundThe relationship between time to surgery (TTS) and survival benefit is not sufficiently demonstrated by previous studies in locally advanced gastric cancer (LAGC). This study aims to assess the impact of TTS after neoadjuvant chemotherapy (NACT) on long-term and short-term outcomes in LAGC patients.MethodsData were collected from patients with LAGC who underwent NACT between January 2007 and January 2018 at our institution. Outcomes assessed were long-term survival, pathologic complete response (pCR) rate, and postoperative complications.ResultsThis cohort of 426 patients was divided into five groups by weeks of TTS. Under cox regression, compared to other groups, the 22–28 days and 29–35 days groups revealed a better OS (≤21 vs. 22–28 days: HR 1.54, 95% CI = 0.81–2.93, P = 0.185; 36–42 vs. 22–28 days: HR 2.20, 95% CI = 1.28−3.79, P = 0.004; 43–84 vs. 22–28 days: HR 1.83, 95% CI = 1.09–3.06, P = 0.022) and PFS (≤21 vs. 22–28 days: HR 1.54, 95% CI = 0.81–2.93, P = 0.256; 36–42 vs. 22–28 days: HR 2.20, 95% CI = 1.28−3.79, P = 0.111; 43–84 vs. 22–28 days: HR 1.83, 95% CI = 1.09–3.06, P = 0.047). Further analysis revealed a better prognosis in patients with TTS within 22–35 days (OS: HR 1.78 95% CI = 1.25−2.54, P = 0.001; PFS: HR 1.49, 95% CI = 1.07−2.08, P = 0.017). Postoperative stay was significantly higher in the ≤21 days group, while other parameters revealed no statistical significance (P > 0.05). Restricted cubic spline depicted the nonlinear relationship between TTS and OS/PFS.ConclusionPatients who received surgery within 3−5 weeks experienced the maximal survival benefit without an increase in postoperative complications or lowering the rate of pCR. Further investigations are warranted.

Summary: Amyotrophic lateral sclerosis (ALS) manifests pathological changes in motor neurons and various other cell types. Compared to motor neurons, the contribution of the other cell types to the ALS phenotypes is understudied. G4C2 repeat expansion in C9ORF72 is the most common genetic cause of ALS along with frontotemporal dementia (C9-ALS/FTD), with increasing evidence supporting repeat-encoded poly(GR) in disease pathogenesis. Here, we show in Drosophila muscle that poly(GR) enters mitochondria and interacts with components of the Mitochondrial Contact Site and Cristae Organizing System (MICOS), altering MICOS dynamics and intra-subunit interactions. This impairs mitochondrial inner membrane structure, ion homeostasis, mitochondrial metabolism, and muscle integrity. Similar mitochondrial defects are observed in patient fibroblasts. Genetic manipulation of MICOS components or pharmacological restoration of ion homeostasis with nigericin effectively rescue the mitochondrial pathology and disease phenotypes in both systems. These results implicate MICOS-regulated ion homeostasis in C9-ALS pathogenesis and suggest potential new therapeutic strategies.

The mechanical analysis of interlayer bonding problem of asphalt pavement is performed by the elastic layered system theory or finite element method (FEM); then, a lot of specialized programs based on the above theories emerged successively, of which BISAR3.0 and EverStressFE are quite representative. In order to further clarify the characteristics of BISAR3.0 and EverStressFE for investigating interlayer bonding problem of asphalt pavement, this paper will carry out a comprehensive comparison from the specific realization viewpoint, such as the principle of interlayer bonding, modeling, calculation processing, and result treatment, and a specific example will be given to compare and analyze their functions. The results indicate that the two programs have certain comparability in analyzing the interlayer bonding problem of asphalt pavement, which will contribute to the foundation for the rational selection of asphalt pavement structure mechanical analysis program.

Steel fiber ultrahigh performance concrete (UHPC) and rubber ultrahigh performance concrete (UHPC) adopt the methods of “rigidity overcomes rigidity” and “softness overcomes rigidity,” respectively, to resist the abrasion and cavitation erosion caused by water flow carrying large solid particles. The above two have been applied in engineering successfully, but there are differences in material properties, mechanical properties, and microstructures. In this study, we will analyze the difference of abrasion resistance performance of the above two kinds of UHPC, in terms of, for example, compressive strength, abrasion resistance strength, abrasion rate, and microstructure in different ages, so as to provide a new material and method. The results indicate that the steel fiber UHPC compressive strength is higher than that of rubber UHPC (containing 1% steel fiber), and the abrasion resistance performance of steel fiber UHPC is lower than that of rubber UHPC (containing 1% steel fiber) when the rubber content is 10% and 12.5%. With the increase of steel fiber content, the compressive strength of steel fiber UHPC is not significantly improved, and the influence of water-binder ratio (W/B) on UHPC is higher than that of steel fiber content. With the increase of rubber content, the rubber UHPC compressive strength decreases, abrasion resistance strength decreases, and abrasion rate increases. The cement paste-aggregate interface transition zone (ITZ) of steel fiber UHPC and rubber UHPC has few internal voids and high compactness; however, the ITZ of steel fiber UHPC is denser than that of rubber UHPC.

The Hippo signaling pathway controls tissue growth by regulating the subcellular localization of Yorkie /Yap. Here the authors show that PRP4K, a kinase involved in mRNA splicing, phosphorylates Yki/Yap in the nucleus, which prevents its nuclear accumulation and inhibits Hippo signaling.

Graphite-based composites are well recognized as ideal functional materials in mechanical seals, bearings of canned pumps, and electrical contact systems because of their outstanding self-lubricating ability, thermostability, and chemical stability. Working in harsh conditions is a huge challenge for the graphite materials, and their tribological properties and wear mechanisms are not well studied. In this study, the tribological performance of metal-impregnated graphite, resin-impregnated graphite, and non-metal-impregnated graphite under high temperature and high load are studied using a ball-on-disc tribometer. The results show that the metal-impregnated graphite (Metal-IG) has a stable friction regime and exhibits better anti-friction and anti-wear properties than that of resin-impregnated graphite (Resin-IG) and non-impregnated graphite (Non-IG) under extreme pressure (200~350 MPa) and high temperature (100–350 °C). The Metal-IG and Resin-IG can reduce the wear depth by 60% and 80%, respectively, when compared with Non-IG substrate. The impregnated materials (metal or resin) can enhance the strength of the graphite matrix and improve the formation of graphite tribofilm on the counterpart surfaces. Friction-induced structural ordering of graphite and slight oxidation of metal in the formed mechanically mixed layer is also beneficial for friction and wear reduction. This study demonstrates the tribological characteristics of impregnated graphite under harsh conditions and provides the experimental basis for the advanced usage of high-reliability and self-lubrication graphite composites.
