BACKGROUND AND AIMS Proteins that recognize epigenetic modifications function as mediators to interpret epigenetic codes. Hypoxia response and metabolic rewiring are two major events during cancer progression. However, whether and how the epigenetic regulator integrates hypoxia response and metabolism together remain open for study. APPROACH AND RESULTS We data mined the clinical association of 33 histone lysine acetylation reader proteins with liver cancer and found that ALL1-fused gene from chromosome 9 (AF9) is up-regulated in cancer and correlates with tumor stage and poor prognosis. Conditional deletion of Af9 in mouse liver resulted in decreased tumor formation induced by c-MET proto-oncogene/β-catenin. Loss of AF9 heavily impaired cell proliferation and completely blocked solid tumor formation. We further discovered that AF9 formed a positive feedback circuit with hypoxia-inducible factor 1 alpha (HIF1α) and also stabilized MYC proto-oncogene (cMyc). Mechanically, AF9 interacted with HIF1α and targeted HIF1A promoter whereas AF9 recognized cMyc acetylation at K148, protected cMyc phosphorylation at S62, and then stabilized cMyc, which, in turn, up-regulates phosphofructokinase, platelet expression. Otherwise, knockout of Af9 in mouse hepatocytes increased the infiltration of CD8+ T cells, which is linked to the down-regulation of lactate dehydrogenase A. CONCLUSIONS AF9 is up-regulated to promote gene expression of hypoxia tolerance and glycolysis by simultaneously forming a complex with HIF1α and recognizing acetylated cMyc. Our results establish the oncogenic role of AF9 in human liver cancer, which could be a potential target for designing drugs against liver cancer.

Blockchain technology has gained growing popularity in recent years. While the technology works well for most applications, the vulnerability of the blockchain-based system is not well understood. The core part of a blockchain-based system is the consensus algorithm. It can be regarded as a protocol used by all network users to make decisions on the growth of the chain. Malicious users can exploit the vulnerabilities of the consensus algorithm to launch block withholding attacks to earn more profit by increasing their winning possibilities, which are significant threats to the security of blockchain-based applications. In this paper, we describe some scenarios of block withholding attacks and suggest effective mitigation methods accordingly. We propose two hybrid consensus algorithms against the block withholding attacks and evaluate the effectiveness of these algorithms, both analytically and through simulation. Simulation results from the SimBlock simulator verified the effectiveness of the proposed algorithms.

Background: Thrombospondin (TSP) is an astrocyte-secreted protein, well-known for its function as a modulator of synaptogenesis and neurogenesis. The mechanism underlying the effects of TSP1 on synaptic activity and formation involved in MHE pathogenesis remains unclear. Methods: The present study explored the effect of TSP1 on neurodegeneration, inflammatory response and the activation of Wnt7a/CNTF signaling in the primary rat neurons and an MHE rat model. Results: When we treated neurons with TSP1, p38MAPK phosphorylation and TNFα expression was increased significantly. Also, the exposure of TSP1 increased the expression and release of Wnt7a and CNTF, upregulated spinophilin, enhanced the interaction of Wnt7a/CNTF with spinophilin, and triggered synaptic activity through p38/TNFα signaling in PC12 cells and primary neurons. The hippocampal injection of TSP1 siRNA in mice decreased the interaction of Wnt7a/CNTF with spinophilin, while injection of Wnt7a and CNTF improved learning and memory dysfunctions. MHE brains showed decreased TSP1 expression. The overexpression of TSP1 in the hippocampus of MHE rats ameliorated the disrupted synaptogenesis, learning, and memory. Conclusions: Taken together, these results indicated that TSP1 is a potential synaptic factor related to the inflammatory response and the activation of Wnt7a/CNTF signaling in MHE pathogenesis.

Background Patients with minimal hepatic encephalopathy (MHE) show mild cognitive impairments. Thrombopoietin (TPO) has been shown to be neuroprotective. This study aimed to explore the therapeutic effect of Thrombopoietin receptor agonist eltrombopag (ELT) on MHE and the involvement of NRG1 signaling using primary rat neurons and a MHE rat model. Methods We explored the effects of ELT stimulation on NRG1/ErbB4 signaling and synapse formation in the primary rat neurons. Furthermore, we explored the cerebral TPO expression level and the effect of TPO replacement therapy in an MHE rat model. Results The results showed that ELT stimulation activated NRG1/ErbB4 signaling and enhanced synaptic protein expression in the primary rat neurons via sirtuin 1. An anti-NRG1 antibody, ErbB4 inhibitor, or knockdown of NRG1 or ErbB4 could significantly abolish ELT-induced upregulation of synaptic protein expression in the primary rat neurons. MHE rats had significantly decreased cerebral ELT expression compared with normal rats. ELT activated NRG1/ErbB4 signaling in MHE rat brains. Administration or overexpression of ELT or TPO promoted synapse formation and alleviated cognitive impairments in MHE rats. Conclusions These data suggest that ELT promotes synapse formation in vitro and in vivo via activating NRG1/ErbB4 signaling, serving as a promising therapeutic agent for MHE treatment.

Background Minimal hepatic encephalopathy (MHE) is characterized by impaired cognitive function and memory loss, which are often the result of synaptic pathology. Insulin resistance has been reported to be closely correlated with the pathogenesis of MHE. Methods The effect of thrombopoietin receptor agonist eltrombopag (ELT) on neurodegeneration and insulin resistance was examined in the primary rat neurons and an MHE rat model. Results We found that the level of thrombopoietin receptor c-MPL (MPL) expression was decreased in MHE brains, and ELT administration improved insulin resistance, alleviated the destruction of synaptic formation and enhanced learning and memory in the MHE rats, indicating the relationship between dowregulated ELT and insulin resistance. Then in vitro, ELT treatment ameliorated the impairment of glucose uptake, indicating the reduction of insulin resistance. High dose of glucose inhibited insulin-stimulated downregulation of Hypoxia-inducible factor-1α (HIF1α) expression, the phosphorylation of GSK3β (pGSK3 β), upregulation of sirtuin-1 (Sirt1), destruction of synaptic formation and activity, which were all reversed by ELT treatment in insulin resistant neurons. Conclusions These results indicate that ELT is a promising potential therapeutic agent for insulin resistance and defect in learning and memory.

Our previous study revealed that minimal hepatic encephalopathy (MHE) is strongly associated with neuroinflammation. Nevertheless, the underlying mechanism of the induction of inflammatory response in MHE astrocytes remains unclear. In this study, we further investigated the effect and mechanism of S100B, predominant isoform expressed and released from mature astrocytes, on MHE-like neuropathology in the MHE rat model. We discovered that S100B expressions and autocrine were significantly increased in MHE rats and astrocytes isolated from MHE rats. Furthermore, we found that S100B stimulates VEGF expression via the interaction between TLR2 and RAGE in an autocrine manner. S100B-facilitated VEGF autocrine expression further led to a VEGFR2 and COX-2 interaction, which in turn induced the activation of NFƙB, eventually resulting in inflammation and oxidative stress caused by MHE astrocytes. Compared to WT astrocytes, impairment of MHE astrocytes supported neuronal growth in co-culture. To sum up, comprehensive-understanding of the impact of S100B-overexpressed MHE astrocyte on MHE pathology may provide insights into the etiology of MHE. Background— Cardiac interstitial fibrosis is a major cause of the deteriorated performance of the heart in patients with chronic myocardial infarction. MicroRNAs (miRs) have recently been proven to be a novel class of regulators of cardiovascular diseases, including those associated with cardiac fibrosis. This study aimed to explore the role of miR-101 in cardiac fibrosis and the underlying mechanisms. Methods and Results— Four weeks after coronary artery ligation of rats, the expression of miR-101a and miR-101b (miR-101a/b) in the peri-infarct area was decreased. Treatment of cultured rat neonatal cardiac fibroblasts with angiotensin II also suppressed the expression of miR-101a/b. Forced expression of miR-101a/b suppressed the proliferation and collagen production in rat neonatal cardiac fibroblasts, as revealed by cell counting, MTT assay, and quantitative reverse transcription–polymerase chain reaction. The effect was abrogated by cotransfection with AMO-101a/b, the antisense inhibitors of miR-101a/b. c-Fos was found to be a target of miR-101a because overexpression of miR-101a decreased the protein and mRNA levels of c-Fos and its downstream protein transforming growth factor-β1. Silencing c-Fos by siRNA mimicked the antifibrotic action of miR-101a, whereas forced expression of c-Fos protein canceled the effect of miR-101a in cultured cardiac fibroblasts. Strikingly, echocardiography and hemodynamic measurements indicated remarkable improvement of the cardiac performance 4 weeks after adenovirus-mediated overexpression of miR-101a in rats with chronic myocardial infarction. Furthermore, the interstitial fibrosis was alleviated and the expression of c-Fos and transforming growth factor-β1 was inhibited. Conclusion— Overexpression of miR-101a can mitigate interstitial fibrosis and the deterioration of cardiac performance in postinfarct rats, indicating the therapeutic potential of miR-101a for cardiac disease associated with fibrosis.

The purpose of this study was to investigate the effect of long-term administration of scutellarin (Scu) on cardiac interstitial fibrosis of myocardial infarct rats and the underlying mechanisms.
