Rice plays a crucial role in global food security, serving as a staple for more than half of the world's population. Rice blast, caused by the fungus Magnaporthe oryzae, is one of the most devastating fungal diseases, resulting in substantial economic losses in terms of rice yield and grain quality. The resistance provided by individual blast resistance genes tends to become ineffective within a few years of extensive agricultural use due to the rapid evolution of the fungus. Therefore, continuous efforts in resistance breeding in rice are required to enrich the pool of new resistance genes and alleles. A large-scale screening of new blast resistance alleles was conducted across 2000 rice accessions from major rice-producing areas in China. Field trials were employed in two rice uniform blast nurseries located in Enshi and Yichang. Approximately 153 rice accessions showed at least moderate resistance to the rice blast. Sequence-based allele mining was used to identify the allelic variants of major rice blast resistance genes at the Pi5 locus of chromosome 9. Six novel alleles were identified from 64 accessions. Field tests at five locations, including Enshi, Yichang, Lvtian, Taojiang, and Yaan, and greenhouse inoculation using 33 blast strains from Hubei, Hunan, Guangzhou, and Sichuan, were employed to verify the resistance of these six alleles. Three near-isogenic lines (types 2, 4, and 6) were constructed and inoculated with 31 blast isolates from Heilongjiang to evaluate their blast resistance. Furthermore, bulk segregant analysis was used to confirm that Pi5-type2 and Pi5-type6 are dominant alleles in the analyzed populations. These novel alleles expand the allelic series, enriching the genetic resources for rice blast resistance breeding programs, especially in the north of China. Furthermore, they provide valuable insights into the molecular interactions between rice and rice blast.

Yunjing 29 bred by Institute of Food Crops of Yunnan Academy of Agricultural Sciences is a new conventional japonica rice variety with fragrant and soft rice, it was examined and approved by Yunnan Provincial Variety Examination and Approval Committee in March of 2011. The variety is early-maturing, high-yield and good-quality, and has resistances to lodging, blast and bacterial leaf blight, its rice is sweet soft, goluptious and lucidus as well as not coarse when it is cold, so it is a good commodity. To further promote the popularization and cultivation of the variety, maintain its characteristics of high quality and high yield, and prevent commingling and degeneration, the purification and rejuvenation as well as breeding technology of high-quality seeds were proposed after continuous exploration and study. This paper presents investigations of segregation distortion of six rice F2 populations generated from reciprocal F1 hybrids grown at three locations varied at altitudes from 400 to 2200 m. The F1s were derived from reciprocal crosses between cv. XMG, which is a japonica landrace traditionally grown at 2650 m altitude, and cv. N34, which is a japonica restorer possessing a fertility restoring (Rf ) gene and cytoplasm of male sterility (CMS) donated by an indica cultivar. Among nine morphological traits of the F2 populations, only one was in normal distribution, eight were distorted in all or at least one population. Out of 16 polymorphic PCR markers, 10 markers distributed on 7 chromosomes were significantly distorted. Among these markers, RMAN7 and RM257 were distorted in both of the reciprocal populations, which suggested that nuclear genes had strong effects on segregation distortion. The other makers were distorted only in the populations with cytoplasm donated by XMG or N34. The results indicated that segregation of DNA markers was affected by cytoplasm background. Segregation distribution was also affected by altitude, since segregation distortions of most of the markers were detected not in all the three populations generated from F1 grown at the three altitudes, but only in one population from F1 grown at one altitude. Marker M45461, which is located within Rf-1 locus, was severely distorted towards N34 in all the populations with cytoplasm donated by N34, but not in the populations with cytoplasm provided by XMG. The results indicated that interaction between CMS and Rf gene had strong effects on distortion. Results of this study indicated that japonica cytoplasm did not cause distortion favouring a special parent, but indica cytoplasm made distortion favouring a maternal parent. The results suggested that indica cytoplasm was not well compatible with japonica nuclear background, while japonica cytoplasm did not have such trouble with indica nuclei.
