7. Assessing the utility of metabolite profiles of human serum, urine and saliva for determining dietary intake

Abstract

The work undertaken in this thesis aimed to use a combined NMR and MS-based metabolomics approach to discover and validate nutritional biomarkers in a cohort of Northern Irish older adults. Serum, urine and saliva samples from 96 participants of the NIDAS dietary validation sub-cohort of the NICOLA study were collected, together with detailed dietary information (food diaries) and biometric data. These samples were aliquoted and separately processed on both a Bruker 600MHz Ascent (1H NMR spectroscopy) and a Waters TQ-S (a triple quadrupole Mass Spectrometer) paired to an Acquity UPLC, the latter of which also employing 4 Biocrates p180 kits for targeted analysis. After adjusting for known confounders, twelve biomarkers of interest for single foods remained across the three biofluids, six of these in serum, four in urine and two in saliva. Of all these, a specific subset of circulating phosphatidylcholines associated with the intake of milk and dairy products, and a group of salivary amino acids and derivatives which strongly correlated with meat were the findings of highest interest. This same metabolomics data was then used to model two dietary scores measuring three key aspects of complex diets: dietary composition, grouped by k-means clustering; a modified FDSK-11 DDS score, which tracks dietary diversity; and the E-NRF score, which tracks the intake of seven key nutrients to encourage in an older population and three to limit. VIP (variable importance in projection) plots of these models showed consistently different features across groups, with higher levels of amino acids and choline-based compounds in lower scores and "unhealthier" clusters much higher in meat and sweet foods. However, these multivariate models also validated poorly, although the saliva-based OPLS-DA (Orthogonal Projections to Latent Structures Discriminant Analysis) models showed better visual separation and appeared to be less overfit. Together, these results show the benefits of a synergistic approach using multiple instruments towards biomarker discovery, the usefulness of metabolomics-based biomarkers as additional tools for the betterment of nutritional epidemiology studies, and the value of saliva as an under-utilized biofluid in metabolomics. Future work should focus on the validation of these biomarkers and models in larger and more heterogenous cohorts, with a specific focus on the creation of metabolomics-based multivariate approaches to modelling complex dietary patterns with better cross-validation.

Abstract

There is growing evidence that studying aquatic acoustic communities can provide ecologically relevant information. Understanding these communities may offer unique insights into species behaviour and ecology, while consolidating passive acoustic monitoring as a tool for mapping the presence of target species or estimating changes in aquatic biodiversity. Fish can be significant soundscape contributors, but most soniferous fish species are yet to be identified. Here, we crossed information of three key fish acoustic communities in the Lusitanian Province of the Temperate Northern Atlantic (the Madeira archipelago, the Azores archipelago and Arrábida in mainland Portugal) to unveil potential sources of unidentified fish sounds. We found that the three communities shared various sound types and we were able to narrow down the list of possible fish sound sources. Several sound types were suggested to be produced by species of the Pomacentridae, Scorpaenidae and Serranidae families. We also observed that the sound type /kwa/, associated with Scorpaena spp., exhibited more variations in the geographic area where more species of this genus are known to be present. This study showcases that, as databases of unidentified fish sounds continue to grow, future comparisons of multiple acoustic communities may provide insights into unknown fish sound sources and sound types. 48. Multimodal vs. Unimodal Physiological Control in Videogames for Enhanced Realism and Depth

Abstract

(arXiv abridged abstract) In the last two decades, videogames have evolved in a nearly explosive way from the pixelated graphics to today's near-realistic 3D environments. The interaction devices traditionally used in videogames have not evolved with the same intensity, but recent HCI studies have explored biofeedback interaction - the explicit manipulation of a person's physiological data as input to a system - as an alternative to them. Traditional biofeedback prototypes apply 1 sensor to each game mechanic (unimodality). In this dissertation, we introduce the combination of 2 physiological sensors simultaneously per game mechanic (multimodality) and present a First-Person Shooter game comprised of 8 game mechanics with three interaction flavours (no biofeedback/vanilla, unimodal and multimodal). An empirical study with 32 regular players was employed to explore and study differences between the three interaction types and where they can be best employed. Players compared the three games in terms of Fun, Ease of Use, Originality, Playability and Favourite Condition. For the sake of completeness, other evaluation methods were used as well: IMI Questionnaire, keywords association and open-ended commentaries. The vanilla version was considered easier to use, but both biofeedback versions were considered the most fun. Both versions were praised differently: the unimodal version for its simplicity of use, and the multimodal for its realism, activation safety of game mechanics and depth added to the game. Our conclusion is that multimodal biofeedback can have a relevant impact in terms of added depth, depending on the way it is used inside the game.
