- Abstract
Mimosa tenuiflora, popularly known as “Jurema-Preta”, is a perennial tree or shrub native to the tropical regions of the Americas, particularly among Afro-Brazilian and Indigenous Brazilian communities. Known for producing N,N-Dimethyltryptamine, a psychedelic compound with profound psychological effects, Jurema-Preta has been studied for its therapeutic potential in mental health. This study offers a comprehensive analysis of the plastid (ptDNA) and mitochondrion (mtDNA) genomes of M. tenuiflora. The 165,639 bp ptDNA sequence features the classical quadripartite structure with 130 protein-coding genes. Comparative genomics among Mimosa species shows high sequence identity in protein-coding genes, with variation in the rpoC1, clpP, ndhA, and ycf1 genes. The ptDNA junctions display distinct features, such as the deletion of the rpl22 gene, and specific simple sequence repeats highlight genetic variation and unique motifs as valuable genetic markers for population studies. Phylogenetic analysis places M. tenuiflora in the Caesalpinioideae, closely related to M. pigra and M. pudica. The 617,839 bp mtDNA sequence exhibits a complex structure with multiple genomic arrangements due to large repeats, encoding 107 protein-coding genes, including the ptDNA petG and psaA genes, and non-retroviral RNA mitoviruses sequences. Comparative analysis across Fabaceae species reveals limited conservation, emphasizing the dynamic nature of plant mitochondrial genomes. The genomic characterization of M. tenuiflora enhances understanding of its evolutionary dynamics, providing insights for population studies and potential applications in ethnopharmacology and conservation. In recent years, Salmonella enterica subsp. enterica serovar Mbandaka (S. Mbandaka) has beenincreasingly isolated fromlaying hens and shell eggs around theworld. Moreover, this serovar has beenidentified as the causative agent of several salmonellosis outbreaks in humans. Surprisingly, little is knownabout the characteristics of this emerging serovar, and therefore, we investigated antimicrobial resistance,virulence, and prophage genes of six selected Brazilian strains of SalmonellaMbandaka usingWholeGenomeSequencing (WGS).Multi-locus sequence typing revealed that the tested strains belong to Sequence Type413 (ST413), which has been linked to recent multi-country salmonellosis outbreaks in Europe. A totalof nine resistance genes were detected, and themost frequent oneswere aac(6′)-Iaa, sul1, qacE, blaOXA-129,tet(B), and aadA1. A point mutation in ParC at the 57th position (threonine → serine) associated withquinolone resistance was present in all investigated genomes. A 112,960 bp IncHI2A plasmid wasmappedin 4/6 strains. This plasmid harboured tetracycline (tetACDR) andmercury (mer) resistance genes, genescontributing to conjugative transfer, and genes involved in plasmidmaintenance. Most strains (four/six)carried Salmonella genomic island 1 (SGI1). All S.Mbandaka genomes carried seven pathogenicity islands(SPIs) involved in intracellular survival and virulence: SPIs 1-5, 9, and C63PI. The virulence genes csgC, fimY,tcfA, sscA, (two/six), and ssaS (one/six) were absent in some of the genomes; conversely, fimA, prgH, andmgtC were present in all of them. Five Salmonella bacteriophage sequences (with homology to Escherichiaphage phiV10, Enterobacteria phage Fels-2, Enterobacteria phageHK542, Enterobacteria phage ST64T, Salmonellaphage SW9) were identified, with protein counts between 31 and 54, genome lengths of 24.7 bp and 47.7 bp,and averageGCcontent of 51.25%. - Abstract
The genus Utricularia includes around 250 species of carnivorous plants, commonly known as bladderworts. The generic name Utricularia was coined by Carolus Linnaeus in reference to the carnivorous organs (Utriculus in Latin) present in all species of the genus. Since the formal proposition by Linnaeus, many species of Utricularia were described, but only scarce information about the biology for most species is known. All Utricularia species are herbs with vegetative organs that do not follow traditional models of morphological classification. Since the formal description of Utricularia in the 18th century, the trap function has intrigued naturalists. Historically, the traps were regarded as floating organs, a common hypothesis that was maintained by different botanists. However, Charles Darwin was most likely the first naturalist to refute this idea, since even with the removal of all traps, the plants continued to float. More recently, due mainly to methodological advances, detailed studies on the trap function and mechanisms could be investigated. This review shows a historical perspective on Utricularia studies which focuses on the traps and body organization.

The Global Strategy for Plant Conservation (GSPC) was established by the Conference of Parties in 2002 to decrease the loss of plant diversity, reduce poverty and contribute to sustainable development. To achieve this overarching goal, the GSPC has established a series of targets, one of which is to ensure that plant diversity is well understood, so that it can be effectively conserved and used in a sustainable manner. Brazil hosts more than 46,000 species of plants, algae and fungi, representing one of the most biodiverse countries on Earth, and playing a key role in the GSPC. To meet the GSPC goals of Target 1 and facilitate access to plant diversity, Brazil committed to preparing the List of Species of the Brazilian Flora (2008-2015) and the Brazilian Flora 2020 (2016-present). Managing all the information associated with such great biodiversity has proven to be an extremely challenging task. Here, we synthesize the history of these projects, focusing on the multidisciplinary and collaborative approach adopted to develop and manage the inclusion of all the knowledge generated though digital information systems. We further describe the methods used, challenges faced, and strategies adopted, as well as summarize advances to date and prospects for completing the Brazilian flora in 2020.

- Abstract
The shortage of reliable primary taxonomic data limits the description of biological taxa and the understanding of biodiversity patterns and processes, complicating biogeographical, ecological, and evolutionary studies. This deficit creates a significant taxonomic impediment to biodiversity research and conservation planning. The taxonomic impediment and the biodiversity crisis are widely recognized, highlighting the urgent need for reliable taxonomic data. Over the past decade, numerous countries worldwide have devoted considerable effort to Target 1 of the Global Strategy for Plant Conservation (GSPC), which called for the preparation of a working list of all known plant species by 2010 and an online world Flora by 2020. Brazil is a megadiverse country, home to more of the world's known plant species than any other country. Despite that, Flora Brasiliensis, concluded in 1906, was the last comprehensive treatment of the Brazilian flora. The lack of accurate estimates of the number of species of algae, fungi, and plants occurring in Brazil contributes to the prevailing taxonomic impediment and delays progress towards the GSPC targets. Over the past 12 years, a legion of taxonomists motivated to meet Target 1 of the GSPC, worked together to gather and integrate knowledge on the algal, plant, and fungal diversity of Brazil. Overall, a team of about 980 taxonomists joined efforts in a highly collaborative project that used cybertaxonomy to prepare an updated Flora of Brazil, showing the power of scientific collaboration to reach ambitious goals. This paper presents an overview of the Brazilian Flora 2020 and provides taxonomic and spatial updates on the algae, fungi, and plants found in one of the world's most biodiverse countries. We further identify collection gaps and summarize future goals that extend beyond 2020. Our results show that Brazil is home to 46,975 native species of algae, fungi, and plants, of which 19,669 are endemic to the country. The data compiled to date suggests that the Atlantic Rainforest might be the most diverse Brazilian domain for all plant groups except gymnosperms, which are most diverse in the Amazon. However, scientific knowledge of Brazilian diversity is still unequally distributed, with the Atlantic Rainforest and the Cerrado being the most intensively sampled and studied biomes in the country. In times of “scientific reductionism”, with botanical and mycological sciences suffering pervasive depreciation in recent decades, the first online Flora of Brazil 2020 significantly enhanced the quality and quantity of taxonomic data available for algae, fungi, and plants from Brazil. This project also made all the information freely available online, providing a firm foundation for future research and for the management, conservation, and sustainable use of the Brazilian funga and flora.

Background:Phenotypic variations have been observed in populations of west CubanPinguiculaspecies. Such populations occur in patches under different ecological conditions associated with specialised habitats and separated by geographical and ecological barriers, which can lead to morphological differentiation. Aims:To analyse morphological diversity among species and populations of CubanPinguicula; and to test if morphological variability is associated with geographical distribution and distance between populations. Methods:We sampled a total of eight populations ofP. albida,P. cubensisandP. filifoliaand tested for morphometric differences among them by Principal Component Analysis (PCA) and analysis of variance (ANOVA) based on 31 quantitative traits. We also assessed the geographical isolation with respect to morphological distance. Results:The studied species showed high morphological variability at both species and population level. Reproductive characteristics appeared to be more promissory than vegetative ones in distinguishing geographical groups since they clearly defined populations; nevertheless a strong component of individual variation was observed. According to the distance analysis, such morphological variations were correlated with fragmented populations and isolation and allowed the differentiation of two morphotypes ofP. albidaand two ofP. filifolia. Conclusions:We conclude that high phenotypic variation of the analysed species is related to geographical and ecological isolation which have led to the differentiation of morphotypes within species. Utricularia belongs to Lentibulariaceae, a widespread family of carnivorous plants that possess ultra-small and highly dynamic nuclear genomes. It has been shown that the Lentibulariaceae genomes have been shaped by transposable elements expansion and loss, and multiple rounds of whole-genome duplications (WGD), making the family a platform for evolutionary and comparative genomics studies. To explore the evolution of Utricularia, we estimated the chromosome number and genome size, as well as sequenced the terrestrial bladderwort Utricularia reniformis (2n = 40, 1C = 317.1-Mpb). Here, we report a high quality 304 Mb draft genome, with a scaffold NG50 of 466-Kb, a BUSCO completeness of 87.8%, and 42,582 predicted genes. Compared to the smaller and aquatic U. gibba genome (101 Mb) that has a 32% repetitive sequence, the U. reniformis genome is highly repetitive (56%). The structural differences between the two genomes are the result of distinct fractionation and rearrangements after WGD, and massive proliferation of LTR-retrotransposons. Moreover, GO enrichment analyses suggest an ongoing gene birth–death–innovation process occurring among the tandem duplicated genes, shaping the evolution of carnivory-associated functions. We also identified unique patterns of developmentally related genes that support the terrestrial life-form and body plan of U. reniformis. Collectively, our results provided additional insights into the evolution of the plastic and specialized Lentibulariaceae genomes. [ABSTRACT FROM AUTHOR]

Introduction: The fiber of the Gynerium sagittatum Aubl. P. Beauv is raw material for the elaboration of several handcrafts, which are symbols of Colombian cultural identity. In the manufacture process, different genotypes are used according to the fiber quality and the type of craftsmanship, but it is believed that Gynerium is a complex species, and to date, there is no agreement on whether these genotypes belong to the same species or to different species. Objective: The aim of this study was to quickly and accurately identify wild cane plants using the nuclear ribosomal internal transcribed spacer (ITS1+ITS2), three chloroplast regions (matK, rbcL, ycf1), and their combinations. Methods: Different tests were used for discrimination: (1) inter and intraspecific distances, (2) Best Match (BM), Best Close Match (BCM), and tree-based method (3) Neighbor Joining (NJ) and (4) maximum likelihood and bayesian inference in molecular data. Results: The results showed that BM and BCM approaches revealed the low rate of correct species identification for ITS+matK (33.3%) and ITS (28.6%) loci, showing similarity among sequences. These results were further supported by tree-based analyses, where all individual regions and the different gene combinations had a zero discrimination rate. Conclusions: all genotypes belong to the same species of wild cane, therefore existing morphological differences can be related to phenotypic plasticity. [ABSTRACT FROM AUTHOR]

- Abstract
The sacred ayahuasca brew, utilized by indigenous communities in the Amazon and syncretic religious groups in Brazil, primarily consists of a decoction of two plants: (i) the Amazonian liana known as Mariri or Jagube (Banisteriopsis caapi), and (ii) the shrub referred as Chacrona or Rainha (Psychotria viridis). While Chacrona leaves are rich in N,N-Dimethyltryptamine (DMT), a potent psychedelic, the macerated vine of Mariri provides beta-carboline alkaloids acting as monoamine oxidase inhibitors, preventing DMT's degradation. This study sequenced, assembled, and analyzed the complete genome of B. caapi's mitochondrion, yielding a circular structure spanning 503,502 bp. Although the mtDNA encompasses most plant mitochondrial genes, it lacks some ribosomal genes, presents some atypical genes, and contains plastid pseudogenes, suggesting gene transfer between organelles. The presence of a 7-Kb repetitive segment containing copies of the rrnL and trnfM genes suggests mitogenome isomerization, supporting the hypothesis of dynamic mitogenome maintenance in plants. Phylogenetics and phylogenomics across 24 Malpighiales confirms the sample's placement in the "Tucunacá" ethnovariety, aligning with morphological identification. This study spearheads efforts to decode the genome of this esteemed Malpighiaceae.
