This study aimed to analyze the effect of pollution caused by stone quarries on the morpho-anatomy, biochemistry, and physiology of a medicinal wild bramble Rubus plicatus Weihe & Nees. Samples were collected from two natural protected areas: Iron Gates Natural Park and Jiu Gorge National Park, both located in the southwestern part of Romania, and two unpolluted areas from these parks as background sites. We carried out the following analyses on the collected leaves of this taxon: morphology, micromorphology, anatomy, assimilating pigments, heavy metals (Pb, Ni, Cr, Fe), dry mass, bioactive compounds (total phenols and flavonoids), and antioxidant capacity. The results showed more stomata, higher amounts of assimilating pigments, higher amounts of heavy metals (especially lead), less dry mass, less phenols, and more flavonoids in Rubus plicatus leaves from polluted areas compared to areas without sources of pollution. The increased number of stomata and the amounts of assimilator pigments revealed the mechanisms developed by this species in order to survive in polluted conditions.

This study aimed at the relationship between antioxidant capacity, antimicrobial activity, and in vitro evaluation of the wound healing effect of the extract obtained from Echinaceae purpureae folium (EPF). This study’s objective was to assess the bioactive components (total phenol and flavonoid content) and antioxidant activity of EPF extracts using the DPPH test method. The antioxidant capacity and the quantities of the compounds with antioxidant capacity were evaluated by spectrophotometric methods. Antimicrobial activity has been investigated against various pathogenic microorganisms. The minimum inhibitory concentration was determined by the microdilution method. Additionally, our work used a scratch test to examine the in vitro wound healing effects of EPF extract on NHDF cells. Statistical analysis was used to quantify the rate of migration and proliferation of fibroblast cells within the wound. Microscope pictures of fibroblast cells exposed to various EPF extract dosages were processed to estimate the width of the wound, area of the wound, and cell density inside the wound. The study proved that there was a relationship between the antioxidant, antimicrobial, and wound healing ability of EPF extracts.

5. The Influence of Salicylic Acid Treatment on Photosynthetic Pigments, Bioactive Compounds and Antioxidant Capacity in Drought-Stressed Petunia grandiflora

Drought is widely recognized as the most severe environmental danger to horticulture in almost all regions across the globe. The objective of this study was to determine if applying a foliar treatment of salicylic acid (SA) at 400 ppm could mitigate the negative effects of drought on Petunia grandiflora. Four experimental variants were implemented for this purpose, with regular watering or moderate drought conditions. These variants were observed for duration of 14 days. The level of photosynthetic pigments was markedly greater in the samples exposed to drought but treated with foliar SA, in comparison to the samples exposed to drought without any treatment (128.28±8.78 and 38.01±1.15 µg/g total chlorophyll (a+b), respectively). The total polyphenol content and antioxidant capacity of both the leaves and flowers of Petunia were assessed. The polyphenols content was significantly reduced by drought. However, following the application of SA, anthocyanin levels increased in comparison to the sample that had been subjected to drought.The results demonstrate that applying SA to the leaves mitigates the adverse impacts of drought, providing a solution to the issue of water scarcity in ornamental plants, particularly Petunia grandiflora.

Plants are an inexhaustible source of compounds with different medicinal properties, suitable as alternative options for the prevention and treatment of various pathologies. They are safe, effective and economical. In this paper, a combined extract made of Ocimum basilicum and Trifolium pratense extracts (EOT) was used for the first time to demonstrate its healing effect on dermal pathologies. To evaluate the wound healing effect of EOT, a novel gel formulation was prepared and subsequently tested in vitro (using the scratch test assay) and in vivo (on an animal model). The in vitro tests demonstrated the complete recovery of the dermal fibroblast monolayer when treated with EOT in a concentration of 50 µg/mL. In vivo results using a hydrogel formulation based on EOT demonstrated improved wound contraction time and complete healing after 13 days of treatment. Moreover, a clinical case of Psoriasis vulgaris was presented, in which one week of treatment led to the significant improvement of the patient’s health. In conclusion, the topical use of the novel gel formulation containing EOT is a successful therapeutic alternative in the treatment of dermal diseases.

7. Improved Antioxidant Status after Diet Supplementation with Novel Natural-Based Supplement in Combat Athletes

Background: Intense physical activity is known to generate high levels of oxidative stress, and supplementation with bioactive products with powerful antioxidant effects is often recommended. In this context, the present study aims to evaluate the potential benefits of supplementing the diet of professional combat fighters with a new bioactive formulation based on Aronia melanocarpa (black chokeberry) and bee pollen, a natural combination with a balanced ratio of macro- and micronutrients, rich in proteins and polyphenols. Methods: A total of 31 professional combat fighters were selected to participate in this study. Due to allergic medical history, two were excluded, and the remaining 29 were divided into two groups: 14 in the control and 15 in the supplementation group. The supplemented group consumed daily 40 g of novel supplement based on a combination of dried black chokeberry and bee pollen (1:2 ratio) for 4 weeks. Results: A statistically significant increase in plasma antioxidant capacity was observed in the supplemented group compared to control related to the time and group factors at the end of this study. During this study, a significant increase in neutrophils was recorded in both groups. Supplementation with black chokeberry and bee pollen generated no significant modifications in inflammatory markers, body composition, glycemia, maximum aerobic capacity, blood glycemia, hemoglobin or red cell line. Conclusion: This clinical study pointed out a positive result in terms of plasma antioxidant capacity upon administration of the novel nutraceutical formulation.

8. Effects of Allium ursinum L. Leaves and Egg Amount on Quality Attributes, Polyphenol Content, and Antioxidant Capacity of Pasta

Pastas enriched with vegetables are premium nutritious products and their consumption can bring significant health benefits. Recent studies have reported a high content of bioavailable phytochemical compounds in Allium ursinum L.; as such, it can be used in the development of functional food products. This study involved the evaluation of 10 varieties of pasta. Five of these varieties were made from white flour and varying amounts of egg (Ctrl_1–Ctrl_5). The other five varieties (AU_1–AU_5) contained fresh shredded wild garlic leaves in equal proportions (20 g) in addition to the corresponding ingredients (white flour and egg). All pasta variants were investigated for their cooking properties and the presence of bioactive compounds and antioxidant capacity in the raw and cooked pasta. Sensory analysis was conducted on all types of cooked pasta, and results indicated that the pasta with the least quantity of eggs was the most favoured variant. Additionally, the pasta that was enhanced with wild garlic leaves received the highest rating for “overall quality”. According to our results, the optimal cooking time (OCT) increased when the quantity of eggs was raised, but when wild garlic leaves were present, OCT decreased in comparison to the control samples. A significant decrease (p < 0.05) in the cooking parameters, swelling index, and water absorption was observed in both egg pasta (Ctrl) and egg pasta enriched with A. ursinum L. (AU). However, compared to the corresponding egg pasta, the presence of wild garlic in pasta generally produced higher values in SI and WA. Pastas enriched with wild garlic are rich in polyphenols (TPh), with a content ranging from 82.44 to 94.62 µg GAE/g dw, and also exhibit antioxidant properties. The amount of total polyphenol significantly decreases (p < 0.05) during cooking varies depending on the composition of the pasta. The pasta with the highest egg content (AU_5) had the lowest loss of polyphenols, equivalent to 55.35%. Our investigations indicate that producing pasta with eggs and fresh wild garlic leaves results in a valuable food product with a good sensory score, enriched in bioactive components, with essential nutrients that maintains cooking qualities.

9. Soybean Response to Seed Inoculation with Bradyrhizobium japonicum and/or Nitrogen Fertilization

Seed inoculation with symbiotic bacteria is a commonly employed practice in soybean cultivation. As a result, nodulation proceeds properly and plants self-supply atmospheric nitrogen, requiring either minimal or no additional nitrogen fertilization. The aim of the study was to investigate the response of soybeans to the application of the recommended or double dose of commercial inoculants (HiStick® Soy or TURBOSOY®) and/or mineral nitrogen fertilization compared to the untreated control. It was demonstrated that a double dose of the tested preparations had the most favorable effect on nodulation. However, the impact of weather conditions modified their effectiveness during the study years, which was especially visible in 2022. Sowing seeds without inoculation (control) resulted in the formation of sparse root nodules and consequently the lowest leaf area index (LAI) and soil plant analysis development (SPAD) measurements. In addition, the values of SPAD and LAI indices varied across the years of the study, indicating that weather conditions modified nitrogen uptake by plants. Overall, seed inoculation and/or nitrogen fertilization positively influenced the chemical composition of seeds compared to the control. The only decrease observed was in the oil content, while the double dose of HiStick® Soy preparation reduced the polyphenol content. The double dose of the tested inoculants had the most favorable impact on yield components and seed yield. However, applying inoculation at the recommended dose or in combination with nitrogen fertilization yielded similar or slightly worse results, depending on the year. Therefore, soybean seed inoculation should be recommended, although the effectiveness of the procedure will depend on various factors, including the type of inoculant, dosage, nitrogen fertilization, and weather conditions.

10. The Formulation of Dermato-Cosmetic Products Using Sanguisorba minor Scop. Extract with Powerful Antioxidant Capacities

There has been a significant increase in the use of botanical resources for the formulation of topical products designed for medicinal and cosmetic applications. Sanguisorba minor Scop., a botanical species, exhibits a variety of properties and has significant potential for applications in the field of cosmetics. The aim of this study was to formulate topical preparations, incorporating an extract derived from the plant S. minor Scop. comprising a combination of roots, leaves, and flowers. In the initial phase, a total of seven combinations were prepared using extracts derived from the roots, leaves, and flowers of S. minor Scop. (v/v/v). These combinations were subsequently subjected to evaluation for their antioxidant capacity using four distinct methods: 2,2-diphenyl-1-picrylhydrazyl (DPPH), ferric reducing antioxidant power (FRAP), cupric reducing antioxidant capacity (CUPRAC), and Trolox equivalent antioxidant capacity (TEAC). An extract of plant organs in a ratio of 1:2:1 (v/v/v), which had a strong antioxidant capacity and demonstrated synergistic effects according to the DPPH, TEAC, and CUPRAC assays (with values of 1.58 ± 0.1, 1.18 ± 0.09, and 1.07 ± 0.07, respectively), was selected for inclusion in three dermato-cosmetic products (hydrogel, emulgel, and cream). All the prepared preparations were evaluated in terms of topical formulation attributes and organoleptic characteristics. The testing of dermato-cosmetic products included assessments of their topical formulation properties and organoleptic characteristics. The hydrogel, emulgel, and cream exhibited varying degrees of stretchability. In addition, a study was carried out to assess the in vitro release of polyphenols from the cosmetic formulations using a Franz diffusion cell system. The results showed that the emulgel containing the extract of S. minor Scop. had the highest and most significant release of polyphenols, with a release rate of 84.39 ± 1.01%. This was followed by the hydrogel and cream, which had release percentages of 80.52 ± 0.89 and 75.88 ± 0.88, respectively, over an 8 h period. Thus, for the first time in the literature, a topical cosmetic product with high antioxidant potential containing S. minor Scop. extract was developed and optimized.

The purpose of this study is to analyze prospective approaches that emphasize the beneficial biofunctional and metabolic effects of different anatomic parts of the Prunus spinosa L. on maintaining human health and improving some pathophysiological processes. Our research is based on recent data from literature on the biology and ecology of P. spinosa L., focusing on its bioactive compounds. Based on such bifunctional parameters, we aim to conceptualize path-breaking approaches that have as a goal the incorporation of P. spinosa L. fruits into certain food products to make use of their potential metabolic benefits in cardiovascular pathologies and other disorders that are prevalent at present and respond to nutritional intake of antioxidants. It is well known that dietary interventions allow the search for mechanisms leading to the development of effective nutritional therapies. This review has mainly focused on the identification of bioactive and functional nutrients of P. spinosa L. that can be incorporated into diverse food formulations. This is because nutrition plays a pivotal role in the development, validation, and recommendation of the nutritional composition of food, with demonstrated impacts on metabolic processes in specific diet-related pathologies.

Plant Growth Promoting Rhizobacteria (PGPR) has gained immense importance in the last decade due to its in-depth study and the role of the rhizosphere as an ecological unit in the biosphere. A putative PGPR is considered PGPR only when it may have a positive impact on the plant after inoculation. From the various pieces of literature, it has been found that these bacteria improve the growth of plants and their products through their plant growth-promoting activities. A microbial consortium has a positive effect on plant growth-promoting (PGP) activities evident by the literature. In the natural ecosystem, rhizobacteria interact synergistically and antagonistically with each other in the form of a consortium, but in a natural consortium, there are various oscillating environmental conditions that affect the potential mechanism of the consortium. For the sustainable development of our ecological environment, it is our utmost necessity to maintain the stability of the rhizobacterial consortium in fluctuating environmental conditions. In the last decade, various studies have been conducted to design synthetic rhizobacterial consortium that helps to integrate cross-feeding over microbial strains and reveal their social interactions. In this review, the authors have emphasized covering all the studies on designing synthetic rhizobacterial consortiums, their strategies, mechanism, and their application in the field of environmental ecology and biotechnology.

With a high number of athletes using sport supplements targeting different results, the need for complex, natural and effective formulations represents an actual reality, while nutrition dosing regimens aiming to sustain the health and performance of athletes are always challenging. In this context, the main goal of this study was to elaborate a novel and complex nutraceutical supplement based on multiple bioactive compounds extracted from Aronia melanocarpa and bee pollen, aiming to support physiological adaptations and to minimize the stress generated by intense physical activity in the case of professional or amateur athletes. Our proposed formulations are based on different combinations of Aronia and bee pollen (A1:P1, A1:P2 and A2:P1), offering personalized supplements designed to fulfill the individual requirements of different categories of athletes. The approximate composition, fatty acid profile, identification and quantification of individual polyphenols, along with the antioxidant capacity of raw biological materials and different formulations, was performed using spectrophotometric methods, GS-MS and HPLC-DAD-MS-ESI+. In terms of antioxidant capacity, our formulations based on different ratios of bee pollen and Aronia were able to act as complex and powerful antioxidant products, highlighted by the synergic or additional effect of the combinations. Overall, the most powerful synergism was obtained for the A1:P2 formulation.

Since ancient times, many plants have been cultivated for their nutritional and medicinal properties. The genus Sanguisorba has been used for medicinal purposes for more than 2000 years. These species are distributed in temperate, arctic, or alpine areas in the Northern Hemisphere. Elongated, imparipinnate leaves and densely clustered flower heads are characteristics of the genus Sanguisorba. While Sanguisorba officinalis L. is mainly known for its significant medicinal applications, Sanguisorba minor Scop. is beginning to attract greater interest for its chemical composition and biological effects. Our research collected extensive information on Sanguisorba minor, including its history, taxonomy, habitat, and distribution, as well as its bioactive components and biological activities. In addition to electron microscopy of plant parts (root, stems, and leaves), which is described for the first time in the literature in the case of S. minor, the study also provides information on potential pests or beneficial insects that may be present. Our goal was to provide important information that will serve as a solid foundation for upcoming research on Sanguisorba minor Scop.

15. Bioactive Compounds and Antioxidant Capacity of Several Blackberry (Rubus spp.) Fruits Cultivars Grown in Romania

Blackberry fruit (Rubus spp.) has a powerful antioxidant capacity due to the high levels of anthocyanins and other phenols it contains. In this work, we investigated the physico-chemical characteristics, the bioactive compounds (total phenols, flavonoids, and monomeric anthocyanins), and the antioxidant capacity of seven blackberry cultivars belonging to the Rubus fruticosus L. and Rubus laciniatus L. genera growing in the NW region of Romania. In addition, the wild blackberry cultivar from the same area was also evaluated. Anthocyanins from the blackberry fruit were extracted using SPE (Solid Phase Extraction), and the anthocyanin profile was identified and quantified using HPLC-PDA analysis. In terms of polyphenol content and antioxidant capacity, two of the cultivars examined stood out. The majority of anthocyanin found in blackberries was cyanidin-3-glucoside, with the highest amount recorded in the ‘Thorn Free’ cultivar at 329.26 ± 9.36 mg/g dw. Comparatively, ‘Loch Ness’ and ‘Thorn Free’ fruits exhibited total phenol contents of 1830.98 ± 13.55 and 1687.14 ± 62.41 mg GAE/100 g dw, respectively. Antioxidant capacities varied significantly among the eight blackberry cultivars, with cultivars ‘Loch Ness’ and ‘Thorn Free’ achieving high values in comparison to the others. The findings of the multivariate analysis also supported the experimental results. Knowing the phytochemical composition and antioxidant potential of different blueberry cultivars, one can use them as fresh, functional foods or for commercial purposes to produce products derived from fruits with a high concentration of bioactive components.

Natural compounds are used in modern dermal treatments to avoid side effects commonly associated with conventional treatments. The aim of our study was to develop a liposomal formulation including black mulberry extract and to highlight its potential on the healing of normal human dermal fibroblasts (NHDF) in vitro using the scratch test. Mulberry-loaded liposomes (MnL) were prepared using a thin-film hydration method based on cholesterol (C) and phosphatidylcholine (PC) in a 1:3 (w/w) ratio. The liposomal formulation was characterized by analyzing its size, electric surface potential, morphology, entrapment efficiency, and in vitro healing effects. Also, the black mulberry fruits (Morus nigra L.) were characterized from point of view of polyphenolic compounds and antioxidant capacity by Ferric-Reducing Antioxidant Power (FRAP) assay. HPLC-DAD-MS (ESI+) (high performance liquid chromatography-photodiode array detection-mass spectrometry (electrospray ionization)) analysis indicated the presence of phenolic compounds namely from hydroxybenzoic and hydroxycinnamic acids and flavonols. Among flavonols, quercetin-glucoside represented 50.56%, and chlorogenic acid was the predominant compound among hydroxycinnamic acids (37.06%). In vitro fibroblast wound closure was more effective with mulberry-loaded liposomes (MnL) than extracts of mulberries. According to our study, mulberry-loaded liposomes have been shown to be effective in wound healing and can be used as a natural treatment.

Azelaic acid (AA), as a natural product, was proven to be effective in targeting multiple causes of acne and related dermatological conditions, as it is well tolerated using different classical formulations (gel, cream, etc.). However, its limited aqueous solubility and inadequate penetration across the stratum corneum might be related to different possible side effects such as itching and burning. The aim of our work was to elaborate a novel liposomal formulation based on azelaic acid, with enhanced biocompatibility, bio-availability, antimicrobial, antigenotoxic, and anti-inflammatory properties. The liposomal formulations were prepared by the lipid film hydration method with different concentrations of azelaic acid (15%, 20%, 25%) and characterized in terms of morphological features, physico-chemical properties, antimicrobial, cytotoxic, and in vitro wound healing effect. Successful encapsulation with 80.42% efficiency, with a size of up to 500 nm and good stability, was achieved, as demonstrated by FTIR spectroscopy (Fourier Transform Infrared Spectroscopy), DLS (dynamic light scattering), and zeta-potential measurements. In terms of antibacterial activity, all the liposomal formulations exhibited a better effect compared to free AA solution against Staphylococcus aureus and Enterococcus faecalis. Cytotoxicity assays and an in vitro “scratch” test performed with normal human dermal fibroblasts revealed an accelerating healing effect, while a comet assay evidenced the protective effect of AA liposomal formulations against hydrogen-peroxide-induced DNA damage in fibroblasts. The optimum formulation in terms of both the antimicrobial and wound healing effect was AALipo20% (liposomes with 20% azelaic acid included).

The most widespread Sanguisorba species are Sanguisorba officinalis L. and Sanguisorba minor Scop. which are also found in the Romanian flora and classified as medicinal plants because of hemostatic, antibacterial, antitumor, antioxidant and antiviral activities. This study aimed to characterize and compare Sanguisorba species in order to highlight which species is more valuable according to phenolic profile and antimicrobial activity. Based on high-performance liquid chromatography equipped with photodiode array detection and mass spectrometry (electrospray ionization) (HPLC–DAD-MS (ESI+)) analysis, it was evident that the ethanol extract obtained from the leaves of S. minor Scop. contains the highest content of phenolic compounds at 160.96 mg/g p.s., followed by the flower and root extract (131.56 mg/g dw and 121.36 mg/g dw, respectively). While in S. officinalis, the highest amount of phenols was recorded in the root extract (127.06 mg/g), followed by the flower and leaves extract (102.31 mg/g and 81.09 mg/g dw, respectively). Our results show that among the two species, S. minor Scop. is richer in phenolic compounds compared with the S. officinalis L. sample. In addition, the antimicrobial potential of each plant organ of Sanguisorba species was investigated. The ethanol extract of S. minor Scop. leaves exhibited better antibacterial activity against all of the bacteria tested, especially on Staphylococcus aureus, with an inhibition zone of 15.33 ± 0.83 mm. Due to the chemical composition and antimicrobial effect, the Sanguisorba species can be used as food supplements with beneficial effects on human health.

Viscum album L. subsp. album is a hemiparasitic plant that is recognized as a medicinal plant due to its beneficial effects, including anti-tumor activity, antioxidant, anti-inflammatory, anti-hepatotoxic, hypoglycemic, and antimicrobial properties as well as for lowering blood pressure. On the other hand, mistletoe is a biotic stressor for both deciduous trees and conifers. Our main aim was to evidence the influence of mistletoe on the content of chlorophylls, proline, total phenols, flavonoids, and antioxidant capacity of leaves from tree host trees (Malus domestica, Prunus domestica, and Populus alba) that grow on the northwest of Romania. In addition, HPLC-DAD-MS-ESI+ was used to analyze the phenolic acid and flavonoid profiles of V. album L. subsp. album leaves according to their parasitized hosts. A significant decrease in chlorophyll a level of approximately 32% was detected in poplars infested with mistletoe, followed by infested apples and plums with pigment reductions of 29.25% and 9.65%, respectively. The content of total phenols and flavonoids in the parasitized trees was higher compared to the non-parasitized ones. In the case of poplar, which presented the highest incidence of mistletoe infestation (70.37%), the content of total phenols in the leaves was two times higher compared to non-infested leaves. Based on HPLC chromatographic analysis, leaves of mistletoe growing on apple (VAM) had the highest content of phenolic acids (7.833 mg/g dw), followed by mistletoe leaves on poplar (VAO) and plum (VAP) (7.033 mg/g dw and, respectively, 5.559 mg/g dw). Among the flavonols, the predominant component was Rhamnazin glucosides in the amount of 1.025 ± 0.08 mg/g dw in VAO, followed by VAP and VAM (0.514 ± 0.04 and 0.478 ± 0.04 mg/g dw, respectively). Although our results show that mistletoe negatively influences the host trees, it is still a valuable plant that must be exploited to bring benefits to human health.

The somatic embryogenesis is an advanced method for clonal propagation and a useful tool for ex situ conservation of genetic resources. In this paper an experimental device was designed, composed of: one oak species (Quercus petraea), one provenance, four types of explants (developmental stages of the zygotic embryo) and four variants of culture medium. It was released the two-way statistical analyses of the two analyzed factors affecting the embryo induction efficiency: the stage of explant and culture medium (4 stages x 4 culture media). The most important factor affecting the efficiency of somatic embryogenesis was found to be the developmental stage of the zygotic embryos used as juvenile explants. The high embryogenic ability of young zygotic embryos and low embryogenic ability of maturing zygotic ones showed the strong positive relationship between somatic embryo induction and accumulation of reserve nutrients in cotyledons. The best results were obtained with the youngest embryos (stage 1), with a linear decrement towards the stage 4. The embryo induction efficiency ranged between 44% and 10% in Q. petraea. Two nutritive culture media and two growth hormone combinations were tested. The effect of culture medium proved to be totally insignificant, besides the differences observed among the variants cultivated on different media. The situation was the same as well for the nutrients as for growth regulators.

21. Antigenotoxic and Antimutagenic Potentials of Proline in Allium cepa Exposed to the Toxicity of Cadmium

This study was conducted to evaluate whether the application of proline as a potential osmoprotectant at different doses could improve the genotoxic and mutagenic effects caused by plant exposure to cadmium salts. For this purpose, the Comet assay was used, which allows the rapid detection of DNA damage shortly after its occurrence, before the DNA is repaired, as well as the discrimination of the DNA damage limited to specific cells in a heterogeneous population. After treatment of Allium cepa roots with 75µM CdSO4·H2O (Cd sample), a DNA percentage of 35.24% was recorded in the tail. In the samples treated first with proline and then with cadmium (pre-treatment group), the percentage DNA in the tail was reduced by 24.8% compared with the Cd sample. Instead, in the post-treatment group (samples treated first with cadmium and then with proline), the percentage DNA in the tail was reduced by 69.04% compared with the Cd sample. All cadmium treatments induced chromosomal aberrations (CAs). Compared with the CAs values obtained after Cd treatment, the reduction was 75.6% in the pre-treatment group and 55.39% in the post-treatment group. The results of this study highlighted that exogenous application of proline alleviated the genotoxic effect of cadmium.

Advanced nutritional interventions are one of the key components of elite sports performance in general. Combat sports require a high percentage of muscle mass with minimum body weight to generate the maximum power possible. An adequate level of nutrition knowledge, particularly with respect to identifying energy needs while avoiding confusion over dietary supplements and false perceptions of steroid requirement, which may compromise the health condition, is of crucial importance. In this context, the aim of our work is to highlight nutritional requirements/nutritional assessment, the importance of daily dietary intake in combat players, which increasingly includes a broad range of sports nutrition supplements, and the roles of vitamins, minerals and proteins, combined with antioxidants and strength training, in muscular performance. The main nutrients required in the daily diet of combat players, the mechanisms of action, the main outcomes and possible side effects are summarized. Special attention is paid to natural supplements and their importance and advantages over synthetic ones, along with future trends of development.

23. Effects of Different Edible Coatings on the Shelf Life of Fresh Black Mulberry Fruits (Morus nigra L.)

The aim of this study was to evaluate the effects of edible coatings based on gelatine, oils (sunflower and coconut) and beeswax on the physicochemical properties, bioactive compounds (total phenols and monomeric anthocyanin content), antioxidant capacity (DPPH and FRAP assays were used) and sensorial attributes of mulberry fruits during a period of storage (0, 2, 5 and 8 days) at 4 °C. The results showed that, in all samples treated with edible films, the degree of firmness was higher and the deterioration weaker compared to the control at day 8 of storage. Edible coatings significantly reduced the rate of deterioration, in terms of total phenolics and anthocyanins, in mulberry fruits over time, and the gelatine-coated mulberry samples (G_Mn) exhibited the best results. In terms of the antioxidant capacity of the coated mulberry, after 5 and 8 days of storage, only the G_Mn samples maintained significantly high DPPH radical scavenging and FRAP values compared to control. Coating improved the sensorial attributes of the mulberry during storage, and gelatine-coated fruits recorded the highest score, followed by layer-by-layer samples (O+W_G_Mn). All edible coatings used to cover black mulberry in this study extended the shelf life of the fruits, while maintaining high levels of bioactive compounds and, consequently, high antioxidant capacity, along with improved sensory qualities, during cold storage.

24. The Effect of Sustainable Feeding Systems, Combining Total Mixed Rations and Pasture, on Milk Fatty Acid Composition and Antioxidant Capacity in Jersey Dairy Cows

This study was carried out to assess the effect of using pasture combined with total mixed ration (TMR) on milk production and composition, fatty acid (FA) profiles, fat-soluble antioxidant content, and total milk antioxidant capacity (TAC). In addition, the effect of milk pasteurization and storage at 2 °C for 4 days, lipophilic antioxidants and TAC were assessed. Two homogeneous groups of Jersey cows (n = 8) were constituted, which were randomly assigned to one of the two treatments: TMR (without access to pasture), or partial mixed diet (pTMR: grazing 8 h/day + TMR indoors). To establish FA profiles and lipophilic antioxidants’ changes in milk during the grazing period, in case of the pTMR group the experimental period was spilt in three grazing periods: P1 (May), P2 (June), and P3 (June/July). Milk yielded from cows having limited access on pasture (pTMR diet) showed an improved FA profile, with higher concentrations of FAs considered benefic for human health (vaccenic acid (VA), conjugated linoleic acid (CLA), omega-3 FA (n-3 FA)) (p < 0.01) and a lower concentration of FAs with hypercholesterolemiant potential (C12:0, C14:0, and C16:0) (p < 0.05), compared to that of the TMR diet. This change in FA profile was correlated with improved sanogenous lipid indices of milk fat (n-6/n-3 FA atherogenic index and thrombogenic index). Milk yielded during the P1 grazing period had higher concentrations of VA, CLA, and n-3 FA (p < 0.05) and lower concentrations of C14:0 and C16:0 (p < 0.01); it exhibited the best values for the main sanogenous fat lipid indices of fat. Moreover, pTMR milk showed a higher α-tocopherol, retinol, and β-carotene content (p < 0.05), positively correlated with TAC values in milk (P1 ˃ P2 ˃ P3). By comparison, cows fed using the TMR diet yielded a higher quantity of milk (p < 0.05), but a lower fat and protein content (p < 0.01), and also a higher saturated FAs and n-6 FA content (p < 0.05) together with a lower concentration of lipophilic antioxidants in milk. Thermal treatment showed no effect on α-tocopherol and retinol content in milk, but their concentrations decreased during the storage, at the same time a TAC decrease. The results of this study prove the positive effect of using pasture combined with TMR on FA profiles, milk antioxidant content, and antioxidant capacity, with beneficial effects on nutrition and health in humans.

The objective of this study was to characterize two varieties of apple (Florina and Liberty) grown in the North-West of Romania, with a focus on their volatile compounds, bioactive compounds and their antioxidant capacity. The content of bioactive compounds and antioxidant capacity was also tracked during pasteurization at different times (15, 20 and 25 minutes). Among bioactive compounds, the total phenol content was determined, as well as the total flavonoid and vitamin C content. The antioxidant capacity of fruits and pasteurized juice was evaluated by three different methods (DPPH, FRAP and ABTS). Of the 2 apple cultivars, Florina showed a remarkably higher content of total phenolic compounds (657.97 mg GAE/kg fw), flavonoids (122.07 mg QE/kg fw) and vitamin C (94.62 mg/kg), compared to the Liberty cultivar. Following pasteurization, the vitamin C contents decreased significantly relative to pasteurization time. However, in the case of total phenols content, only insignificant decreases were registered, compared to unpasteurized juice. The content of total flavonoids increased significantly after 15 minutes of pasteurization in apple cultivars juices. The apple varieties investigated are rich in bioactive compounds, and pasteurization treatment does not lead to drastic decreases in these compounds and in the antioxidant capacity of apple juices.

In recent years, mulberry has acquired a special importance due to its phytochemical composition and its beneficial effects on human health, including antioxidant, anticancer, antidiabetic and immunomodulatory effects. Botanical parts of Morus sp. (fruits, leaves, twigs, roots) are considered a rich source of secondary metabolites. The aim of our study was to highlight the phytochemical profile of each of the botanical parts of Morus tree, their health benefits and applications in food industry with an updated review of literature. Black and white mulberries are characterized in terms of predominant phenolic compounds in correlation with their medical applications. In addition to anthocyanins (mainly cyanidin-3-O-glucoside), black mulberry fruits also contain flavonols and phenolic acids. The leaves are a rich source of flavonols, including quercetin and kaempferol in the glycosylated forms and chlorogenic acid as predominant phenolic acids. Mulberry bark roots and twigs are a source of prenylated flavonoids, predominantly morusin. In this context, the exploitation of mulberry in food industry is reviewed in this paper, in terms of developing novel, functional food with multiple health-promoting effects.

The healing of skin wounds remains an important concern in medicine, especially in chronic wounds caused by various diseases such as diabetes. Using herbs or herbal products to heal skin wounds is a therapeutic challenge for traditional medicine. In this context, the main aim of our work was to highlight the in vitro healing potential of Stellaria media (L.) Vill. (SM) extract using the scratch assay on normal human dermal fibroblasts (NHDF). The ability to stimulate cell migration and proliferation under the influence of different concentrations of SM extract (range between 12.5 and 200 µg/mL) was determined compared to the control (untreated in vitro-simulated wound) and positive control (allantoin 50 µg/mL). Our results showed that the concentration of 100 µg/mL SM extract applied on the simulated wound recorded the strongest and fastest (24 h) migration (with wound closure) and proliferation of NHDF compared with the control. In addition, the SM extract was characterized in terms of bioactive compounds (total phenols and flavonoids content), antioxidant capacity (FRAP (The Ferric-Reducing Antioxidant Power) assay and electrochemical method), and antimicrobial activity. The results show that the SM extract contains a considerable amount of polyphenols (17.19 ± 1.32 mg GAE/g dw and 7.28 ± 1.18 mg QE/g dw for total phenol and flavonoid content, respectively) with antioxidant capacity. Antimicrobial activity against Gram-positive bacteria (S. aureus) is higher than E. coli at a dose of 15 µg/mL. This study showed that Stellaria media is a source of polyphenols compounds with antioxidant capacity, and for the first time, its wound healing potential was emphasized.

The aim of this work was to study the effect of pulsed electric field (PEF) treatment on the content of bioactive compounds (total phenols, total flavonoids, monomeric anthocyanin content), antioxidant capacity, color intensity (CI), and sensory quality of wines derived from three varieties of grapes (Muscat Ottonel, Pinot Noir, and Merlot). PEF treatment resulted in an increase in all bioactive compounds, and consequently, an increase in their antioxidant capacity, which was evaluated by different methods. Applying PEF treatment to red grapes resulted in obtaining wines with higher CI compared to the control sample. For a better interpretation of the results and a proper discrimination of the samples in terms of the variety of grapes and PEF treatment, multivariate analyses, such as the principal component analysis, multivariate analysis of variance, and hierarchical cluster analysis, were applied.

Nowadays, the tendency in pharmaceutical and food industries is to replace synthetic antioxidants with the natural ones. For this reason, there is a growing interest in analyzing natural, healthy and non-toxic additives as potential antioxidants. Some plants, which contain high levels of phenolic compounds, present an increasing interest for medicine due to their ability to scavenge free radicals, along with other pharmacological activities, such as antibacterial activity, wound healing and anti-inflammatory effect, to mention only a few. The aim of this review is to explore the therapeutic potential of Ocimum basilicum and Trifolium pratense in relation with their phytochemical profile and to highlight the pharmacological activity of aqueous or ethanol extracts. Special attention was devoted to the dermal pathology and wound healing effects, in the context of multiple skin conditions such as acne, eczema boils, psoriasis and rashes. Additionally, both extracts (Trifolium sp. and Ocimum sp.) are characterized by high content of antioxidant compounds, which are responsible for the radiance and resistance of the skin and slowing down of the aging process by maintaining estrogen levels. Moreover, the potential combined effect of the mixed extract is pointed out in terms of future applications for wound healing, based on some preliminary results obtained from a “scratch tests” assay performed with respect to human dermal fibroblasts.

This paper demonstrates the ability of a functional food based on probiotics and selenium nanoparticles (SeNPs) to annihilate the toxic effect of cadmium on the kidneys. SeNPs were obtained by eco-friendly method used Lactobacillus casei. The morphological features and size of SeNPS were characterized by Atomic Force Microscopy (AFM) and Dynamic Light Scattering (DLS). Two kind of SeNPs were used, purified and Lacto-SeNPs (LSeNPs), administered by gavage at three concentrations (0.1, 0.2, and 0.4 mg/Kg b.w.) for 30 days in a mouse model of cadmium renal toxicity. The blood marker of renal injury (creatinine) significantly decreased in groups where the mice were treated with both form of SeNPs. The antioxidant capacity of plasma was evaluated by Trolox Equivalent Antioxidant Capacity (TEAC) assay and revealed that SeNPs in co-treatment with Cd, promotes maintaining antioxidant activity at the control level. Histopathological analysis of kidneys demonstrated morphological alteration in the group that received only cadmium and restored after administration of SeNPs or LSeNPs. In addition, immunohistochemical analysis revealed anti-apoptotic effects through reduction of pro-apoptotic bax and increasing of anti-apoptotic Bcl-2 protein expressions. Moreover, co-administration of Cd with SeNPs significantly decreased gene expression of kidneys inflammatory markers (TNF-α, IL-6, NF-ĸB) in a dose dependent manner, with the best results for LSeNPs at highest dose (0.4 mg/kg). Therefore, the L. casei strain is a potential SeNPs-enriched probiotic for application as functional food in the future to annihilate cadmium-induced kidneys toxicity.

31. Extraction of Bioactive Compounds from Two Grape Varieties Using Pulsed Electric Field

In this paper, our objective was to investigate extraction efficiency of polyphenols with antioxidant capacity from two varieties of grapes (white-Muscat Ottonel (MO) and red-Pinot Noir (PN)) using pulsed electric field (PEF). Grapes were harvested in the Crisana Santimreu vineyard in 2015. The grapes were declustered and crushed, and have been divided into three parts. A part was treated in PEF, the second one was homogenized, and then was treated in PEF and the last part remained untreated, representing the control samples. The bioactive compounds, like total polyphenol content and total flavonoid content, were determined by the spectrophotometric method. The antioxidant capacity of the samples was determined by three methods (DPPH, FRAPand TEAC). Our results show that the application of PEF to the crushed grapes increases the total polyphenol content 2.28 and 3.15 times and the total flavonoid content 7.17 and 5.29 times for MO and PN, respectively. Also, the antioxidant capacity was significantly increased when it was compared with the control sample. When the treatment in PEF was applied on homogenized grapes both bioactive compounds and antioxidant capacity were significantly increased compared to the samples treated in PEF for crushed grapes. In the present research work, we demonstrated that the application of PEF to two different grape varieties significantly increased the level of total polyphenols and flavonoids and also the antioxidant capacity. The application of PEF to homogenized grapes was more efficient.

Viscum album L. is a medicinal plants used for many years as a remedy in the traditional medicine and in complementary cancer therapies. We investigated the influence of some host trees: Acer campestre (VAA), Fraxinus excelsior (VAF), Populus nigra (VAP), Malus domestica (VAM), and Robinia pseudoacacia (VAR), on the chemical composition and antioxidant activity of leaves and stems from V. album, as well the influence of the solvent (water and ethanol) used for extraction on biological activity. HPLC with photodiode array detector analysis of bioactive compounds from leaves and stems of mistletoes (V. album) hosted by five differents trees was performed. Antioxidant activities, determined by the 2,2-diphenyl-1-picrylhydrazyl (DPPH), Oxygen Radical Absorbance Capacity (ORAC), (Trolox-equivalent antioxidant capacity (TEAC) methods were compared with the quantification of total phenolics using the Folin -Ciocalteu reagent. V. album hosted by Fraxinus excelsior (VAF), recorded the highest level of total phenolic acids (108.64 μg/g dry matter) while the mistletoe hosted by Malus domestica (VAM) had the lowest level of total polyphenols (39.37 μg/g dry matter). In general, higher antioxidant activity was detected in the alcoholic than in the aqueous extracts. The values obtained for total phenolics in both, aqueous and ethanol extracts, decreased in the order: VAR > VAF > VAP > VAA > VAM. The mistletoe stem extracts contained lower levels of phenolics, as compared to leaves, in both solvents. Our results suggest that mistletoe extracts possess significant antioxidant activities that may be due to their bioactive compounds content, suggesting that this plant may be an important source of natural products with chemopreventive and chemotherapeutic activities.
