Abstract

A kinetic study of vanadate inhibition was performed in combination with spectroscopic studies to characterize uteferrin's diiron center and its binding sites. Results reveal that vanadate inhibited uteroferrin in a noncompetitive and time-dependent manner in anaerobic and anaerobic conditions. However, this effect was observed only in the presence of the diiron enzyme and did not appear with the FeZn enzyme. Two distinct vanadium-uteroferrin complexes were formed during the reaction. Uteroferrin's interaction with phosphate and molybdate were found to differ significantly from the interaction seen for bovine purple acid phosphatase.

4. Chemically induced modifications of cofactor specificity of glucose-6-phophate dehydrogenase

Abstract

A chemical scheme for changing the cofactor specificity of NADP dependent enzymes is reported. Vanadium exhibits properties that may modify the cofactor specificity of 2'-NADP dependent dehydrogenases. The ability of 2'-NADV to imitate 2'-NADP in biological systems is tested with bakers' yeast glucose-6-phosphate dehydrogenase. Bakers' yeast is added top a solution of NAD and varying concentrations of vanadate. Results show that reaction rates increase by as much as 20 times in the presence of vanadate, while reaction rates are negligible without bakers' yeast.
