Phytoplasmas are plant pathogenic bacteria, naturally transmitted by insects and confined in the phloem of the host plant, where they take up nutrients and eventually cause plant death. Their control is mainly based on insecticide treatments. The aim of this work was to study the effect of two AM fungi in modifying plant response to chrysanthemum yellows phytoplasma (CY) infection in chrysanthemum plants. Inoculation of Glomus intraradices BB-E and G. mosseae BEG12 reduced the damage caused by this plant pathogen in the aerial part of the plant, increased plant tolerance to the infection and reduced the severity of symptom expression, probably in a systemic way. Inoculation with G. mosseae did not alter CY multiplication and viability in young leaves, whilst the morphology of CY mature leaves was typical of senescent cells. Possible mechanisms involved are discussed.

Rhizoctonia solani root-rot is a major soilborne disease causing growth and yield depression. The ability of Glomus mosseae BEG12 and Pseudomonas fluorescens A6RI to suppress this soilborne disease in tomato was assessed by comparing the shoot and root growth of plants infested with R. solani 1556 when protected or not by these beneficial strains. The epiphytic and parasitic growth of the pathogenic R. solani 1556 was compared in the presence and absence of the biocontrol agents by microscopical observations allowing the quantification of roots with hyphae appressed to epidermal cells (epiphytic growth) and of roots with intraradical infection (parasitic growth). The root architecture of the tomato plants under the different experimental conditions was further characterized by measuring total root length, mean root diameter, number of root tips and by calculating degree of root branching. G. mosseae BEG12 and P. fluorescens A6RI fully overcame the growth depression caused by R. solani 1556. This disease suppression was associated with a significant decrease of the epiphytic and parasitic growth of the pathogen together with an increase of root length and of the number of root tips of inoculated tomato plants. The combined effects of G. mosseae BEG12 and P. fluorescens A6RI on pathogen growth and on root morphogenesis are suggested to be involved in the efficient disease suppression.

Soil microfungal communities of two ad- jacent, but differently aged Alnus viridis coenoses were studied using the soil dilution plate method. A total of 84 taxa were isolated: 59 from the young com- munity, 51 from the old and 26 taxa shared. Morti- erella parvispora and Pythium sp. had the highest den- sity values in the young and the old alder community, respectively. Species compositions were compared be- tween plots of the two communities using a metric multidimensional scaling and a correspondence anal- ysis. Both analyses grouped plots from the young and the old community separately, suggesting a correla- tion between the changes in the fungal species com- position and the age of the alder communities. The correspondence analysis produced two species groups more related to one set of plots or the other, together with another group formed of Mortierella, Micromucor, Geomyces and Trichoderma species, whose distributions were more closely related to the general abiotic conditions than the age of the alder com- munities. It is proposed that the significant differ- ences in the composition of the two fungal commu- nities mirror the existence of a seral fungal succes- sion paralleling the aging of the alder communities.

We have isolated a cDNA clone, Le-MI-13 (Lycopersicon esculentum mycorrhizal induced) by differential screening of a cDNA library prepared from mRNA extracted from tomato roots colonized by the arbuscular mycorrhizal (AM) fungus Glomus mosseae. The Le-MI-13 clone encodes a polypeptide that shows a high degree of amino acid sequence similarity with members of the recently identified multigene family, the cullins. Northern blot analyses demonstrated that the Le-MI-13 transcript accumulated in tomato roots forming arbuscular mycorrhizal symbiosis. Only very little Le-MI-13 transcript was detected in control roots. Tomato roots infected by the pathogenic fungus Phytophthora nicotianae var. parasitica did not accumulate Le-MI-13 transcript, indicating that upregulation of the Le-MI-13 gene is specific to roots forming arbuscular mycorrhizal symbiosis. Indirect evidence suggesting that a Le-MI-13-mediated cell-cycle-like control might operate in AM-colonized cells came from flow cytometry and static micro fluorimetry analysis. There was a strong correlation between nuclear polyploidization and AM colonization.Key words: tomato, arbuscular mycorrhizae, Phytophthora, cullins, polyploidy.

Nuclear changes in roots of tomato (Lycopersicon esculentum), a plant with a small genome, during the establishment of arbuscular mycorrhizal (AM) colonization were studied using light and electron microscopy, as well as flow and static cytometry. Nuclei of mycorrhizal root cortex cells were larger and had more decondensed chromatin than those of controls. Significant ploidy distribution differences were observed between nuclei of AM colonized and control roots, and a strong correlation between nuclear polyploidization and AM colonization was found. Polyploidization and decondensation are usually associated with high metabolic activity. The metabolic activity of mycorrhizal root cells, evaluated in this work as respiratory activity by using a cytochemical assay for succinate dehydrogenase combined with image analysis, increased in comparison to controls. The meaning of polyploidization is discussed in relation to the structural and metabolic modifications induced by mycorrhization.
