Abstract The development of efficient and robust non‐Pt and low‐Pt catalysts with equivalent or even superior performance to commercial Pt‐based catalysts for hydrogen evolution reaction (HER) is highly desired, but challenging, in the field of water electrolysis. Herein, we report a facile and cost‐effective in situ electrochemical approach for the synthesis of atomically dispersed metal sites including platinum (Pt), ruthenium (Ru), and palladium (Pd) on the polyaniline (PANI) support. The PANI exhibits not only high electrochemical conductivity but also efficient H+ capture from hydronium ions, leading to the formation of protonated amine groups that can be easily electrochemically reduced to H2 on atomically dispersed metal active sites. As an example, the atomically dispersed Pt sites anchored on carbon cloth‐supported PANI (PANI‐Pt/CC) demonstrate excellent activity and durability toward the HER. The mass activity of PANI‐Pt‐10/CC reaches 25 A mgPt−1, exhibiting a significant enhancement of 50‐fold compared to that of the commercial Pt/C (0.5 A mgPt−1). Therefore, this study presents a universally applicable approach for the design of atomically dispersed metal sites/conducting polymer heterostructures for highly efficient catalysts toward HER and beyond.

Abstract Background Protein cysteine oxidation is substantially involved in various biological and pathogenic processes, but its implications in pancreatic cancer development remains poorly understood. Methods and results In this study, we performed a global characterization of protein oxidation targets in PDAC cells through iodoTMT-based quantitative proteomics, which identified over 4300 oxidized cysteine sites in more than 2100 proteins in HPDE6c7 and PANC-1 cells. Among them, 1715 cysteine residues were shown to be differentially oxidized between HPDE6c7 and PANC-1 cells. Also, charged amino acids including aspartate, glutamate and lysine were significantly overrepresented in flanking sequences of oxidized cysteines. Differentially oxidized proteins in PANC-1 cells were enriched in multiple cancer-related biological processes and signaling pathways. Specifically, the HIF-1 signaling proteins exhibited significant oxidation alterations in PANC-1 cells, and the reduced PHD2 oxidation in human PDAC tissues was correlated with lower survival time in pancreatic cancer patients. Conclusion These investigations provided new insights into protein oxidation-regulated signaling and biological processes during PDAC pathogenesis, which might be further explored for pancreatic cancer diagnosis and treatment.

Abstract Objective Infectious pancreatic necrosis (IPN) is a serious complication of acute pancreatitis, and early recognition and timely intervention are the keys to improving clinical outcomes. The purpose of this study was to investigate the predictive capacity of the neutrophil CD64 index (nCD64 index) on IPN in patients with acute pancreatitis Methods This study comprises two independent cohorts: the training cohort consisted of 202 patients from Hunan Provincial People's Hospital, and the validation cohort consisted of 100 patients from Changsha Central Hospital. Peripheral blood samples were collected on the day of admission and on the 3rd, 5th, 7th, and 10th days of hospitalization, and the nCD64 index was detected by flow cytometry. Additionally, relevant clinical characteristics and laboratory biomarkers were collected and analyzed. [Objective] To observe the effect that extracting solution of radix euphorbiae lantu has on the proliferation and apoptosis of K562 cell lines. [Methods] The growth inhibition ratio of the extracting solution of Euphorbia fischeriana Steud. on K562 cell line was determined by MTT. Apoptosis and its morphology were observed by Hoechst fluorescence staining. Changes of cell membrane potential and cell cycle were detected by FCM after treated with extracting solution of Ephorbia fischeriana Steud. [Results] The extracting solution of Ephorbia fischeriana Steud. had significant inhibiting effect on the growth of K562 cell line. Observed by fluorescence microscope, we found that the extracting solution of Euphorbia fischeriana Steud. could change the morphology of K562 cell. The mitochondrial transmembrane potential (Δψm) lowered and the number of cells in G0/G1 phase increased significantly which were detected by FCM. [Conclusions] The extracting solution of Euphorbia fischeriana Steud. could promote the apoptosis of K562 cell line and inhibit its growth by changing the cell cycle. [ABSTRACT FROM AUTHOR]
