It is demonstrated that electrochemistry (EC) coupled to liquid chromatography (LC) and electrospray ionization tandem mass spectrometry (LC/EC/ESI-MS/MS) can be used to rapidly obtain information about the antioxidant activity (i.e., oxidation potential) and capacity (i.e., amount) of polyphenolic compounds, including catechin, kaempferol, resveratrol, quercetin, and quercetin glucosides. The described on-line LC/EC/ESI-MS/MS method facilitates the detection and characterization of individual antioxidants based on a combination of the obtained m/z values for the antioxidants and their oxidation products, the potential dependences for the ion intensities, and correlations between the retention times in the LC, EC, and MS chromatograms. As these results provide patterns that can be used in rapid screening for antioxidants in complex samples, the method should be a valuable complement to chemical assays commonly used to determine the total antioxidant capacity of samples. It is shown that the antioxidant capacity for a mixture of polyphenolic compounds depends on the redox potential employed in the evaluation, and this should consequently be taken into account when comparing results from different total antioxidant capacity assays. It is also demonstrated that the inherent antioxidant capacities of phenolic compounds increase with an increasing number of hydroxyl groups and that the potential needed to oxidize the remaining hydroxyl groups increases successively upon oxidation of the compound. Unlike chemical assays, which generally do not provide any information about the identities of the compounds on the molecular level, the present screening method can be used to identify individual antioxidants, rank compounds with respect to their ease of oxidation, and to study the antioxidant capacity at any redox potential of interest. 10.1021/ac901397c

The oxidation of 4-chloroaniline (4-CA) has been studied by electrochemistry (EC) coupled on-line with electrospray ionization mass spectrometry (ESI-MS) using two electrochemical flow cells of different design. The experimental results, which generally verify previously suggested oxidation pathways for 4-CA, also indicate the presence of an up to now unrecognized comproporfionation reaction. The oxidation of 4-CA (m/z 128.2) was found to give rise to the formation of both an oxidized dimer, 4-[(4-chlorophenyl)imino]-2,5-cyclohexadien-1-imine (m/z 217.2), and a reduced dimer, 4-amino-4'-chlorodiphenylamine (m/z 219.2), in addition to a dimer intermediate (m/z 253.2). The unexpected formation of the reduced dimer is shown to stem from a comproportionation reaction involving 4-CA and the oxidized dimer. The presence of the latter reaction was clearly seen by comparing results obtained with two thin-layer flow cells, both with conversion efficiencies of 50% under mass transport controlled conditions but of different design with respect to the influence of the counter electrode reaction on the reaction at the working electrode. The experimental results demonstrate that the formation of the reduced dimer is favored by a decrease in the local pH in the flow cell, and the influence of the pH on the oxidation of 4-CA was also investigated in the pH range between 2.0 and 6.0 using off-line voltammetry. It is concluded that EC/ESI-MS is a powerful tool for the study of the present type of reactions and that studies of the reaction pathways of these systems are best carried out under noncoulometric experimental conditions as the latter facilitates the detection of reaction intermediates. Comproportionation reactions, similar to the reaction present in the 4-CA system, can also be expected to be present during the formation of conducting polymers such as polyaniline and polypyrrole.

Metabolomic fingerprinting is a growing strategy for characterizing complex biological samples without detailed prior knowledge about the metabolic system. A two-way analysis system with liquid separation and mass spectrometric detection provides detail-rich data suitable for such fingerprints. As a model study, human urine samples, obtained after intake of coffee, tea, or water, were analyzed with capillary electrophoresis electrospray ionization time-of-flight mass spectrometry (CE-ESI-TOF-MS). In-house-developed software (in Matlab) was utilized to manage and explore the large amount of data acquired (230 CE-MS runs, each with 50-100 million nonzero data points). After baseline and noise reduction, followed by suitable binning in time and m/z, the data sets comprised 9 and 14 million data points in negative and positive ESI mode, respectively. Finally, a signal threshold was applied, further reducing the number to about 100 000 data points per data set. A set of interactive exploratory tools, utilizing principal component analysis (PCA) and analysis of variance (ANOVA) results based on a general linear model, facilitated visual interpretation with score plots (for group assessment) and differential fingerprints (for 'hot spot' detection). In the model study highly significant differences due to beverage intake were obtained among the 10 first principal components (p < [10.sup.-6] for two of the components in both ESI modes). Especially, the contrasts between 'coffee' and 'tea or water' indicated several 'hot spots' with highly elevated intensities (e.g., for uncharged masses 93, 94, 109, 119, 123, 132, 148, 169, 178, 187, 190, and 193) suitable for further analysis, for example, with tandem MS.

The structures of organic phosphorous (P) compounds in aquatic sediments are to a large extent unknown although these compounds are considered to play an important role in regulating lake trophic status. To enhance identification of these compounds, a liquid chromatography (LC) method for their separation was developed. The stationary phase was porous graphitic carbon (PGC), and the mobile phases used in the gradient elution were compatible with both inductive coupled plasma atomic emission spectroscopy (ICP-AES) and electrospray ionization tandem mass spectrometry (ESI-MS/MS). With LC-ICP-AES, eight different P containing peaks could be observed in the P chromatogram indicating that at least eight different P compounds were separated. With the setup of an information dependent acquisition (IDA) with ESI-MS/MS, the mass over charge (m/z) of compounds containing a phosphate group ([H.sub.2][PO.sub.3.sup.-], m/z 97) could be measured and further fragmentation experiments gave additional information on the structure of almost 40 separated P compounds, several were verified to be nucleotides. ICP-AES was very suitable in the development of the LC method and allowed screening and quantification of P compounds. The presented LC-ESI-MS/MS technique was able to identify several sediment organic P compounds.

The tracer-pulse method provides the real adsorption data points directly from simple, straightforward calculations and is therefore a superior method for multicomponent adsorption isotherm determination in HPLC. Only one important problem has restricted its use so far: the tracer peaks are invisible using any conventional detection principle. We present a solution to this problem with an approach with a firm base in analytical chemistry, utilizing stable isotopes and mass spectrometric detection. The new approach was used for the determination of binary adsorption isotherms, and a systematic investigation was made of its main sources of error. With this modification, the tracer method can be a prime choice for future characterizations of multicomponent separation systems and of competitive drug binding studies.

Capillary online restricted-access media-liquid chromatography-electrospray ionization-tandem mass spectrometry (RAM-LC-ESI-MS/MS) for direct analysis of drugs and metabolites spiked in biological fluids was developed. Using a column switching setup it was possible to perform effective sample preparation and analysis of raw biological fluids (plasma and urine) without matrix effects in the electrospray mass spectrometric detection step. The peak focusing efficiency of the extraction column was more effective in backflush compared to foreflush mode. The system was able to concentrate diminished samples of polar drugs and their metabolites reaching quantifiable results as low as I ng/mL utilizing a sample volume of only 333 nL of biofluids. New column hardware was developed to circumvent clogging problems experienced with plasma injections. The glass fiber filter frit, which is commonly used, was replaced with a short piece of 20 [micro]m i.d. fused silica capillary. The extraction columns were able to handle up to 60 injections and showed a high loading capacity, making the saturation of the MS detector the limiting factor on the linear dynamic range. The simultaneous separation and detection of 10 drugs and metabolites was obtained in 8 min of analysis, including the online sample preparation and enrichment step.

Electrospray ionization mass spectrometry (ESI-MS) and subsequent MS/MS analyses were employed to directly detect palladium-containing cationic reaction intermediates in a ligand controlled palladium (II)-catalyzed oxidative Heck arylation. The study supports previous mechanistic propositions and also provides new information regarding the composition of aryl-containing palladium complexes in such reactions.

A novel setup for the coupling of a commercially available thin-layer cell to electrospray mass spectrometry (ESI-MS) which allows the electrochemical reactions at the counter electrode to be straightforwardly separated from the flow into the mass spectrometer has been developed. In this way, interferences from reaction products formed at the counter electrode can be minimized. This reduces the risk of changes in the mass spectra as a result of electrochemical reactions in the solution. The described setup also enables the working electrode to be positioned close to the electrospray (ESI) emitter without the need for a grounding point or a long transfer line between the electrochemical cell and the electrospray emitter. By decoupling the electrochemical reactions in the flow cell and those in the electrospray emitter, improved facilities for studies of electrochemical reactions are obtained through a better control of the potential of the working electrode. The setup has been used to study the oxidation of a drug (Olsalazine), which previously has been found to involve chemical follow-up reactions. It is also demonstrated that uncharged thiols can be detected in ESI-MS after spontaneous adsorption on a gold working electrode, followed by oxidative desorption to yield sulfinates or sulfonates. This adsorption and potential-controlled desorption has been used for the preconcentration of micromolar concentrations of 1-hexanethiol as well as for desalting of solutions containing micromolar concentrations of thiols. The results indicate that the present online coupling of an electrochemical cell to ESI-MS provides promising possibilities for sample preconcentration, matrix exchange (including desalting), and ionization of neutral compounds, such as thiols.

Extensive multimer formation of small and uncharged analytes was observed in electrospray mass spectrometry. The concentration-dependent aggregation behavior, including species up to tetramers, resulted in highly nonlinear calibration curves. For some analytes, these effects were operating down into the nanomolar level. Varying the drift potential failed to selectively dissociate the multimer complexes in favor of the monomer, which severely obstructed quantitative work. However, by incorporating certain cationic additives into the electro-sprayed solution, complete suppression of the multimers could be obtained. In addition, a gain in the signal intensifies by a factor up to 7 was achieved. A variety of additives were investigated, and the structural requirements for optimum performance will be outlined.

A simplified method for determination of ion distribution within electrospray ionization droplets has been developed. The method is based on an electrospray ionization mass spectrometry equilibrium partitioning model recently developed by Enke (Enke, C. G. Anal Chem. 1997, 69, 4885-4893). This 'simple' method required only two samples to be analyzed at each solution composition compared to the method previously reported. Furthermore, as the same m/z value is monitored in both experiments, possible effects of the mass-dependent ion transmission in the quadrupole were eliminated. Tetraalkylammonium compounds with variable hydrophobicity were used as model compounds and the effect of methanol--water composition in the electrosprayed solution was studied. It was found, as expected, that the signal optimized at a high content of methanol in the electrosprayed solution. The distribution coefficient for analyte ions between the droplet surface and bulk solution maximize, however, at a lower content of methanol in the electrosprayed solution.

In this paper, we present the work carried out in the SOBITS project executed within the framework of the European Measurement and Testing program. The goal of the project was to compare different methods for characterization of the shielding performance of board-to-backplane and backplane-to-cable connectors used in telecommunication and computer systems. Four measurement methods have been selected. Dedicated test samples have been designed and developed in order to enable the measurement of the shielding characteristics of the connectors under test with all methods. Different shielding configurations were considered, in order to determine the influence of the quality of the shield. Single-ended as well as differential excitations have been considered. Among the test samples, a nonshielded version was designed for each signal/ground configurations. This sample was used as a reference. The definition of the quantities that describe the shielding performance is different for each of the measurement methods. Conversion formulas between these quantities have been formulated. They were verified by measuring a reference device that is easy to measure by all methods. Thus we were able to compare the relative shielding effectiveness of the various shielded board-to-backplane and backplane-to-cable connectors for all four measurement methods. The relative shielding effectiveness of a shielded connector was calculated with respect to the nonshielded version. We also compared the different shielding performance results obtained with each measurement method individually. Index Terms--Connectors, measurement, shielding.
