Abstract

Imaging technologies such as positron emission tomography (PET) and magnetic resonance imaging (MRI) present unparalleled opportunities to investigate the neural basis of autism spectrum disorder (ASD). However, challenges such as deficits in social interaction, anxiety around new experiences, impaired language abilities, and hypersensitivity to sensory stimuli make participating in neuroimaging studies challenging for individuals with ASD. In this commentary, we describe the existent training protocols for preparing individuals with ASD for PET/MRI scans and our own experience developing a training protocol to facilitate the inclusion of low-functioning adults with ASD in PET-MRI studies. We hope to raise awareness of the need for more information exchange between research groups about lessons learned in this context in order to include the entire disease spectrum in neuroimaging studies.

Abstract

We present calculations showing that upcoming Cosmic Microwave Background (CMB) experiments will have the power to improve on current constraints on neutrino masses and provide new limits on neutrino degeneracy parameters. The latter could surpass those derived from Big Bang Nucleosynthesis (BBN) and the observationally-inferred primordial helium abundance. These conclusions derive from our Monte Carlo Markov Chain (MCMC) simulations which incorporate a full BBN nuclear reaction network. This provides a self-consistent treatment of the helium abundance, the baryon number, the three individual neutrino degeneracy parameters and other cosmological parameters. Our analysis focuses on the effects of gravitational lensing on CMB constraints on neutrino rest mass and degeneracy parameter. We find for the PLANCK experiment that total (summed) neutrino mass $M_{\nu} > 0.29$ eV could be ruled out at $2\sigma$ or better. Likewise neutrino degeneracy parameters $\xi_{\nu_{e}} > 0.11$ and $| \xi_{\nu_{\mu/\tau}} | > 0.49$ could be detected or ruled out at $2\sigma$ confidence, or better. For POLARBEAR we find that the corresponding detectable values are $M_\nu > 0.75 {\rm eV}$, $\xi_{\nu_{e}} > 0.62$, and $| \xi_{\nu_{\mu/\tau}}| > 1.1$, while for EPIC we obtain $M_\nu > 0.20 {\rm eV}$, $\xi_{\nu_{e}} > 0.045$, and $|\xi_{\nu_{\mu/\tau}}| > 0.29$. Our forcast for EPIC demonstrates that CMB observations have the potential to set constraints on neutrino degeneracy parameters which are better than BBN-derived limits and an order of magnitude better than current WMAP-derived limits., Comment: 27 pages, 11 figures, matches published version in JCAP

8. Methodological issues when investigating the prevalence and incidence of antiretroviral-related toxicities amongst HIV positive individuals

Abstract

The introduction of highly active antiretroviral therapy (HAART) for the treatment of HIV infection has led to dramatic reductions in morbidity. However, these highly potent drugs have been associated with a number of side effects. It is important to accurately quantify the prevalence and incidence of these toxicities, and to be able to compare the impact of specific antiretrovirals on toxicity rates in an unbiased manner. As we have become aware of potential HAART-related toxicities, there has been an increase in the frequency of monitoring of associated laboratory markers. Thus, diagnoses are made more quickly, and randomly abnormal values are more likely to be observed. There have also been changes over time in the specific antiretrovirals prescribed in HAART regimens. Consequently, newer antiretrovirals may seem to be associated with greater toxicity, even if this is not the case. Data simulations performed suggest that, when comparing the impact of specific HAART regimens on the occurrence of toxicity, considering the first measurement in a specified window leads to the most unbiased results. The choice of most appropriate cut-off of a surrogate laboratory marker of toxicity can also lead to differences in prevlance estimates. My investigations suggest that the most appropriate cut-off for each toxicity and each laboratory marker must be considered individually, but that definitions in which a confirmatory measurement is required are often appropriate. Results of a simulation study investigating a method proposed to account for unmeasured confounding, sample selection models, found this method gives unbiased treatment estimates in the situations we investigated. However, this method can lead to a lack of precision, and requires identification of a variable that is associated with treatment allocation. Thus, the use of these models in real-life settings may be limited. Mis-specification of non-linear associations between variables in regression models can lead to biased estimates. I have found that the use of multi-fractional polynomials to systematically consider the most appropriate relationship between variables is a method that is easy to apply and leads to plausible results.

Abstract

The IPCC's scientific assessment of the timing of net-zero emissions and 2030 emission reduction targets consistent with limiting warming to 1.5°C or 2°C rests on large scenario databases. Updates to this assessment, such as between the IPCC's Special Report on Global Warming of 1.5°C (SR1.5) of warming and the Sixth Assessment Report (AR6), are the result of intertwined, sometimes opaque, factors. Here we isolate one factor: the Earth System Model emulators used to estimate the global warming implications of scenarios. We show that warming projections using AR6-calibrated emulators are consistent, to within around 0.1°C, with projections made by the emulators used in SR1.5. The consistency is due to two almost compensating changes: the increase in assessed historical warming between SR1.5 (based on AR5) and AR6, and a reduction in projected warming due to improved agreement between the emulators' response to emissions and the assessment to which it is calibrated.

Climate model emulators are widely used to generate temperature projections for climate scenarios, including in the recent Intergovernmental Panel on Climate Change Sixth Assessment Report. Here we evaluate the performance of a two-layer energy balance model in emulating historical and future temperature projections from Coupled Model Intercomparison Project Phase 6 models. We find that emulation errors can be large (>0.5°C for SSP2-4.5) and differ markedly between climate models, forcing scenarios and time periods. Errors arise in emulating the near-surface temperature response to both greenhouse gas and aerosol forcing; in some periods the errors due to these forcings oppose one another, giving the spurious impression of better emulator performance. Climate feedbacks are assumed constant in the emulator, introducing time-varying or state dependent feedbacks may reduce prediction errors. Close emulations can be produced for a given period but, crucially, this does not guarantee reliable emulations of other scenarios and periods. Therefore, rigorous out-of-sample evaluation is necessary to characterize emulator performance.

Abstract

BACKGROUND: Immediate initiation of antiretroviral therapy (ART) regardless of CD4 cell count reduces risk for AIDS and non-AIDS-related events in asymptomatic, HIV-positive persons and is the standard of care. However, most HIV-positive persons initiate ART when their CD4 count decreases below 500 × 10 9 cells/L. Consequences of delayed ART on risk for non-AIDS-defining and AIDS-defining cancer, one of the most common reasons for death in HIV, are unclear. OBJECTIVE: To estimate the long-term risk difference for cancer with the immediate ART strategy.DESIGN: Multinational prospective cohort study.SETTING: The D:A:D (Data collection on Adverse events of anti-HIV Drugs) study, which included HIV-positive persons from Europe, Australia, and the United States.PARTICIPANTS: 8318 HIV-positive persons with at least 1 measurement each of CD4 cell count and viral load while ART-naive (study period, 2006 to 2016).MEASUREMENTS: The parametric g-formula was used, with adjustment for baseline and time-dependent confounders (CD4 cell count and viral load), to assess the 10-year risk for non-AIDS-defining and AIDS-defining cancer of immediate versus deferred (at CD4 counts RESULTS: During 64 021 person-years of follow-up, 231 cases of non-AIDS-defining cancer and 272 of AIDS-defining cancer occurred among HIV-positive persons with a median age of 36 years (interquartile range, 29 to 43 years). With immediate ART, the 10-year risk for non-AIDS-defining cancer was 2.97% (95% CI, 2.37% to 3.50%) and that for AIDS-defining cancer was 2.50% (CI, 2.37% to 3.38%). Compared with immediate ART initiation, the 10-year absolute risk differences when deferring ART to CD4 counts less than 500 × 10 9 cells/L and less than 350 × 10 9 cells/L were 0.12 percentage point (CI, -0.01 to 0.26 percentage point) and 0.29 percentage point (CI, -0.03 to 0.73 percentage point), respectively, for non-AIDS-defining cancer and 0.32 percentage point (CI, 0.21 to 0.44 percentage point) and 1.00 percentage point (CI, 0.67 to 1.44 percentage points), respectively, for AIDS-defining cancer. LIMITATION: Potential residual confounding due to observational study design.CONCLUSION: In this young cohort, effects of immediate ART on 10-year risk for cancer were small, and further supportive data are needed for non-AIDS-defining cancer.PRIMARY FUNDING SOURCE: Highly Active Antiretroviral Therapy Oversight Committee.

Abstract

Rotational spectra of thioacetic acid (CH3COSH) have been observed by pulsed-nozzle Fourier transform microwave spectroscopy. Spectroscopic constants are reported for both the syn and anti conformers of the parent species, as well as the 34S and 13C carbonyl isotopologues. Transitions arising from the lowest A and E internal rotor states of the methyl group have been observed and analyzed. Experimental values of the three-fold internal rotation barrier, V3, for the syn and anti conformers of the parent isotopologue are 76.300(12) and 358.056(51) cm−1, respectively, indicating a large effect of the S−H orientation on the CH3 internal rotation potential. M06-2X/6-311+G(d,p) calculations are in good agreement with these results. The block localized energy decomposition method has been applied to understand the origins of this strong dependence of V3 on conformation. The results indicate that π conjugation from the SH to the carbonyl group and steric repulsion between the SH and the methyl group in the anti form are main contributors to the difference. Abstract

Most genes associated with neurodevelopmental disorders (NDDs) were identified with an excess of de novo mutations (DNMs) but the significance in case–control mutation burden analysis is unestablished. Here, we sequence 63 genes in 16,294 NDD cases and an additional 62 genes in 6,211 NDD cases. By combining these with published data, we assess a total of 125 genes in over 16,000 NDD cases and compare the mutation burden to nonpsychiatric controls from ExAC. We identify 48 genes (25 newly reported) showing significant burden of ultra-rare (MAF, For many neurodevelopmental disorder (NDD) risk genes, the significance for mutational burden is unestablished. Here, the authors sequence 125 candidate NDD genes in over 16,000 NDD cases; case-control mutational burden analysis identifies 48 genes with a significant burden of severe ultra-rare mutations.
