4. Game Theoretic Modelling of a Ransom and Extortion Attack on Ethereum Validators

Abstract

Consensus algorithms facilitate agreement on and resolution of blockchain functions, such as smart contracts and transactions. Ethereum uses a Proof-of-Stake (PoS) consensus mechanism, which depends on financial incentives to ensure that validators perform certain duties and do not act maliciously. Should a validator attempt to defraud the system, legitimate validators will identify this and then staked cryptocurrency is `burned' through a process of slashing. In this paper, we show that an attacker who has compromised a set of validators could threaten to perform malicious actions that would result in slashing and thus, hold those validators to ransom. We use game theory to study how an attacker can coerce payment from a victim, for example by deploying a smart contract to provide a root of trust shared between attacker and victim during the extortion process. Our game theoretic model finds that it is in the interests of the validators to fully pay the ransom due to a lack of systemic protections for validators. Financial risk is solely placed on the victim during such an attack, with no mitigations available to them aside from capitulation (payment of ransom) in many scenarios. Such attacks could be disruptive to Ethereum and, likely, to many other PoS networks, if public trust in the validator system is eroded. We also discuss and evaluate potential mitigation measures arising from our analysis of the game theoretic model.

Abstract

Modern ransomware often generate and manage cryptographic keys on the victim's machine, giving defenders an opportunity to capture exposed keys and recover encrypted data without paying the ransom. However, recent work has raised the possibility of future enclave-enhanced malware that could avoid such mitigations using emerging support for hardware-enforced secure enclaves in commodity CPUs. Nonetheless, the practicality of such enclave-enhanced malware and its potential impact on all phases of the ransomware lifecyle remain unclear. Given the demonstrated capacity of ransomware authors to innovate in order to better extort their victims (e.g. through the adoption of untraceable virtual currencies and anonymity networks), it is important to better understand the risks involved and identify potential mitigations. As a basis for comprehensive security and performance analysis of enclave-enhanced ransomware, we present RansomClave, a family of ransomware that securely manage their cryptographic keys using an enclave. We use RansomClave to explore the implications of enclave-enhanced ransomware for the key generation, encryption and key release phases of the ransomware lifecycle, and to identify potential limitations and mitigations. We propose two plausible victim models and analyse, from an attacker's perspective, how RansomClave can protect cryptographic keys from each type of victim. We find that some existing mitigations are likely to be effective during the key generation and encryption phases, but that RansomClave enables new trustless key release schemes that could potentially improve attacker's profitability and, by extension, make enclaves an attractive target for future attackers.

Abstract

The evolution of the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) virus leads to new variants that warrant timely epidemiological characterization. Here we use the dense genomic surveillance data generated by the COVID-19 Genomics UK Consortium to reconstruct the dynamics of 71 different lineages in each of 315 English local authorities between September 2020 and June 2021. This analysis reveals a series of subepidemics that peaked in early autumn 2020, followed by a jump in transmissibility of the B.1.1.7/Alpha lineage. The Alpha variant grew when other lineages declined during the second national lockdown and regionally tiered restrictions between November and December 2020. A third more stringent national lockdown suppressed the Alpha variant and eliminated nearly all other lineages in early 2021. Yet a series of variants (most of which contained the spike E484K mutation) defied these trends and persisted at moderately increasing proportions. However, by accounting for sustained introductions, we found that the transmissibility of these variants is unlikely to have exceeded the transmissibility of the Alpha variant. Finally, B.1.617.2/Delta was repeatedly introduced in England and grew rapidly in early summer 2021, constituting approximately 98% of sampled SARS-CoV-2 genomes on 26 June 2021.

Abstract

We report severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) spike ΔH69/V70 in multiple independent lineages, often occurring after acquisition of receptor binding motif replacements such as N439K and Y453F, known to increase binding affinity to the ACE2 receptor and confer antibody escape. In vitro, we show that, although ΔH69/V70 itself is not an antibody evasion mechanism, it increases infectivity associated with enhanced incorporation of cleaved spike into virions. ΔH69/V70 is able to partially rescue infectivity of spike proteins that have acquired N439K and Y453F escape mutations by increased spike incorporation. In addition, replacement of the H69 and V70 residues in the Alpha variant B.1.1.7 spike (where ΔH69/V70 occurs naturally) impairs spike incorporation and entry efficiency of the B.1.1.7 spike pseudotyped virus. Alpha variant B.1.1.7 spike mediates faster kinetics of cell-cell fusion than wild-type Wuhan-1 D614G, dependent on ΔH69/V70. Therefore, as ΔH69/V70 compensates for immune escape mutations that impair infectivity, continued surveillance for deletions with functional effects is warranted.

Abstract

Plerixafor is a CXCR4 antagonist approved in 2008 by the FDA for hematopoietic stem cell collection. Subsequently, plerixafor has shown promise as a potential pathogen‐agnostic immunomodulator in a variety of preclinical animal models. Additionally, investigator‐led studies demonstrated plerixafor prevents viral and bacterial infections in patients with WHIM syndrome, a rare immunodeficiency with aberrant CXCR4 signaling. Here, we investigated whether plerixafor could be repurposed to treat sepsis or severe wound infections, either alone or as an adjunct therapy. In a Pseudomonas aeruginosa lipopolysaccharide (LPS)‐induced zebrafish sepsis model, plerixafor reduced sepsis mortality and morbidity assessed by tail edema. There was a U‐shaped response curve with the greatest effect seen at 0.1 μM concentration. We used Acinetobacter baumannii infection in a neutropenic murine thigh infection model. Plerixafor did not show reduced bacterial growth at 24 h in the mouse thigh model, nor did it amplify the effects of a rifampin antibiotic therapy, in varying regimens. While plerixafor did not mitigate or treat bacterial wound infections in mice, it did reduce sepsis mortality in zebra fish. The observed mortality reduction in our LPS model of zebrafish was consistent with prior research demonstrating a mortality benefit in a murine model of sepsis. However, based on our results, plerixafor is unlikely to be successful as an adjunct therapy for wound infections. Further research is needed to better define the scope of plerixafor as a pathogen‐agnostic therapy. Future directions may include the use of longer acting CXCR4 antagonists, biased CXCR4 signaling, and optimization of animal models. [ABSTRACT FROM AUTHOR]

Abstract

There is a need to clarify relationships within the actinobacterial genus Micromonospora, the type genus of the family Micromonosporaceae, given its biotechnological and ecological importance. Here, draft genomes of 40 Micromonospora type strains and two non-type strains are made available through the Genomic Encyclopedia of Bacteria and Archaea project and used to generate a phylogenomic tree which showed they could be assigned to well supported phyletic lines that were not evident in corresponding trees based on single and concatenated sequences of conserved genes. DNA G+C ratios derived from genome sequences showed that corresponding data from species descriptions were imprecise. Emended descriptions include precise base composition data and approximate genome sizes of the type strains. antiSMASH analyses of the draft genomes show that micromonosporae have a previously unrealised potential to synthesize novel specialized metabolites. Close to one thousand biosynthetic gene clusters were detected, including NRPS, PKS, terpenes and siderophores clusters that were discontinuously distributed thereby opening up the prospect of prioritising gifted strains for natural product discovery. The distribution of key stress related genes provide an insight into how micromonosporae adapt to key environmental variables. Genes associated with plant interactions highlight the potential use of micromonosporae in agriculture and biotechnology.
