BACKGROUND: Pseudodiastrophic dysplasia (PDD) is a severe skeletal dysplasia associated with prenatal manifestation and early lethality. Clinically, PDD is classified as a 'dysplasia with multiple joint dislocations'; however, the molecular aetiology of the disorder is currently unknown. METHODS: Whole exome sequencing (WES) was performed on three patients from two unrelated families, clinically diagnosed with PDD, in order to identify the underlying genetic cause. The functional effects of the identified variants were characterised using primary cells and human cell-based overexpression assays. RESULTS: WES resulted in the identification of biallelic variants in the established skeletal dysplasia genes, B3GAT3 (family 1) and CANT1 (family 2). Mutations in these genes have previously been reported to cause 'multiple joint dislocations, short stature, and craniofacial dysmorphism with or without congenital heart defects' ('JDSCD'; B3GAT3) and Desbuquois dysplasia 1 (CANT1), disorders in the same nosological group as PDD. Follow-up of the B3GAT3 variants demonstrated significantly reduced B3GAT3/GlcAT-I expression. Downstream in vitro functional analysis revealed abolished biosynthesis of glycosaminoglycan side chains on proteoglycans. Functional evaluation of the CANT1 variant showed impaired nucleotidase activity, which results in inhibition of glycosaminoglycan synthesis through accumulation of uridine diphosphate. CONCLUSION: For the families described in this study, the PDD phenotype was caused by mutations in the known skeletal dysplasia genes B3GAT3 and CANT1, demonstrating the advantage of genomic analyses in delineating the molecular diagnosis of skeletal dysplasias. This finding expands the phenotypic spectrum of B3GAT3-related and CANT1-related skeletal dysplasias to include PDD and highlights the significant phenotypic overlap of conditions within the proteoglycan biosynthesis pathway.

OBJECTIVES: Juvenile idiopathic arthritis (JIA) is an autoimmune disease and a common cause of chronic disability in children. Diagnosis of JIA is based purely on clinical symptoms, which can be variable, leading to diagnosis and treatment delays. Despite JIA having substantial heritability, the construction of genomic risk scores (GRSs) to aid or expedite diagnosis has not been assessed. Here, we generate GRSs for JIA and its subtypes and evaluate their performance. METHODS: We examined three case/control cohorts (UK, US-based and Australia) with genome-wide single nucleotide polymorphism (SNP) genotypes. We trained GRSs for JIA and its subtypes using lasso-penalised linear models in cross-validation on the UK cohort, and externally tested it in the other cohorts. RESULTS: The JIA GRS alone achieved cross-validated area under the receiver operating characteristic curve (AUC)=0.670 in the UK cohort and externally-validated AUCs of 0.657 and 0.671 in the US-based and Australian cohorts, respectively. In logistic regression of case/control status, the corresponding odds ratios (ORs) per standard deviation (SD) of GRS were 1.831 (1.685 to 1.991) and 2.008 (1.731 to 2.345), and were unattenuated by adjustment for sex or the top 10 genetic principal components. Extending our analysis to JIA subtypes revealed that the enthesitis-related JIA had both the longest time-to-referral and the subtype GRS with the strongest predictive capacity overall across data sets: AUCs 0.82 in UK; 0.84 in Australian; and 0.70 in US-based. The particularly common oligoarthritis JIA also had a GRS that outperformed those for JIA overall, with AUCs of 0.72, 0.74 and 0.77, respectively. CONCLUSIONS: A GRS for JIA has potential to augment clinical JIA diagnosis protocols, prioritising higher-risk individuals for follow-up and treatment. Consistent with JIA heterogeneity, subtype-specific GRSs showed particularly high performance for enthesitis-related and oligoarthritis JIA.

This article addresses a critical issue, which has been overlooked, in relation to the design of phase-correcting structures (PCSs) for electromagnetic bandgap (EBG) resonator antennas (ERAs). All the previously proposed PCSs for ERAs are made using either several expensive radio frequency (RF) dielectric laminates or thick and heavy dielectric materials, contributing to very high fabrication cost, posing an industrial impediment to the application of ERAs. This article presents a new industrial-friendly generation of PCS, in which dielectrics, known as the main cause of high manufacturing cost, are removed from the PCS configuration, introducing an all-metallic PCS (AMPCS). Unlike existing PCSs, a hybrid topology of fully metallic spatial phase shifters are developed for the AMPCS, resulting in an extremely lower prototyping cost as that of other state-of-the-art substrate-based PCSs. The APMCS was fabricated using laser technology and tested with an ERA to verify its predicted performance. The results show that the phase uniformity of the ERA aperture has been remarkably improved, resulting in 8.4 dB improvement in the peak gain of the antenna and improved sidelobe levels (SLLs). The antenna system including APMCS has a peak gain of 19.42 dB with a 1 dB gain bandwidth of around 6%.

© 2017 We consider the problem of refill scheduling for a team of vehicles or robots that must contend for access to a single physical location for refilling. The objective is to minimise time spent in travelling to/from the refill station, and also time lost to queuing (waiting for access). In this paper, we present principled results for this problem in the context of agricultural operations. We first establish that the problem is NP-hard and prove that the maximum number of vehicles that can usefully work together is bounded. We then focus on the design of practical algorithms and present two solutions. The first is an exact algorithm based on dynamic programming that is suitable for small problem instances. The second is an approximate anytime algorithm based on the branch and bound approach that is suitable for large problem instances with many robots. We present simulated results of our algorithms for three classes of agricultural work that cover a range of operations: spot spraying, broadcast spraying and slurry application. We show that the algorithm is reasonably robust to inaccurate prediction of resource utilisation rate, which is difficult to estimate in cases such as spot application of herbicide for weed control, and validate its performance in simulation using realistic scenarios with up to 30 robots.
