Abstract

Managing wildlife populations in the face of global change requires regular data on the abundance and distribution of wild animals, but acquiring these over appropriate spatial scales in a sustainable way has proven challenging. Here we present the data from Snapshot USA 2020, a second annual national mammal survey of the USA. This project involved 152 scientists setting camera traps in a standardized protocol at 1485 locations across 103 arrays in 43 states for a total of 52,710 trap-nights of survey effort. Most (58) of these arrays were also sampled during the same months (September and October) in 2019, providing a direct comparison of animal populations in 2 years that includes data from both during and before the COVID-19 pandemic. All data were managed by the eMammal system, with all species identifications checked by at least two reviewers. In total, we recorded 117,415 detections of 78 species of wild mammals, 9236 detections of at least 43 species of birds, 15,851 detections of six domestic animals and 23,825 detections of humans or their vehicles. Spatial differences across arrays explained more variation in the relative abundance than temporal variation across years for all 38 species modeled, although there are examples of significant site-level differences among years for many species. Temporal results show how species allocate their time and can be used to study species interactions, including between humans and wildlife. These data provide a snapshot of the mammal community of the USA for 2020 and will be useful for exploring the drivers of spatial and temporal changes in relative abundance and distribution, and the impacts of species interactions on daily activity patterns. 4. Survival and Recurrence Rates Following SBRT or Surgery in Medically Operable Stage I NSCLC.

Abstract

Surgery is the standard of care for early-stage non-small cell lung cancer (NSCLC), with SBRT reserved for patients who are not surgical candidates or decline surgery. The role of SBRT for medically operable early-stage NSCLC is not defined. We hypothesized overall survival (OS), lung cancer-specific survival (LCSS), progression free survival (PFS), and recurrence rates following SBRT or surgery in medically operable patients with Stage I NSCLC from the Veterans' Health Care System (VAHS) would be equivalent. Medically operable patients diagnosed with Stage I NSCLC between 2000-2020 from the VAHS, determined by an FEV1 or DLCO > 60% of predicted and Charlson comorbidity index (CCI) of 0 or 1, treated with SBRT or surgery were identified. SBRT patients were propensity score matched for age, diagnosis year, sex, race, smoking status, tumor stage, FEV1, and CCI in a 1:1:1 ratio (SBRT:lobectomy:sub-lobar resection). OS, LCSS, and PFS were compared using log-rank and Kaplan-Meier analyses. PFS was calculated as the time from treatment start to recurrence or death, whichever occurred first. Local recurrence was defined as any new lung tumor, regional as spread to intrathoracic lymph nodes, and distant as spread beyond these sites. Hazard ratios were calculated using Cox regression. Recurrence was compared via a chi-squared test. 103 patients were included in each cohort. 97% of patients were male, 86% were white, and 89% were diagnosed from 2011-2020. 82% of patients had a CCI of 0 and the mean percent predicted FEV1 was 88%. 52% of SBRT patients received 50-60 Gy in 5 fractions and 69% of sub-lobectomy patients were treated with a wedge resection. Median follow-up was 4.32 years. For all patients, 5-year OS was 51% (95% CI 46-57%). After matching, OS, LCSS, and PFS were significantly worse with SBRT compared to either surgical cohort (Table 1). Regional recurrence was significantly higher following SBRT (15.5% vs 6.8% or 4.9%; p < 0.05), but there was no significant difference in local (28.2% vs 21.4% or 21.4%; p > 0.05) or distant recurrence (10.7% vs 9.7% or 13.6%; p > 0.05) when compared to lobectomy or sub-lobar resection, respectively. In medically operable patients, OS, LCSS, and PFS following either lobectomy or sub-lobar resection were superior to that for SBRT for Stage I NSCLC, likely due in part to higher regional recurrence following SBRT. This suggests that pulmonary function test results and comorbidity calculations alone are insufficient to define a cohort of medically operable patients suited for SBRT. 6. The Effect of the Open Space School on Children's Security and Independence Development, Academic Performance, Teacher Assessments and School Motivation. Final Report.

Abstract

This study explores the effects of physical differences in the architectural design of schools by comparing behavioral and performance variables for two groups of pupils: (1) those who had previously attended an open space school, and (2) those who had always attended classes in a traditionally built school. Three major variables are examined: academic performance, motivation for school, and security development. Differences in school marks, teacher assessment, I.Q. scores, sex and grade level are also explored. Four classes of grade 7 pupils and three classes of grade 8 pupils (a total of 210 subjects) are included in the study. The tests employed were the Institute of Child Study Security Test-Elementary Form, the Canadian Tests of Basic Skills, the Frymier Junior Index of Motivation, the Dominion Group Test of Learning Capacity-Intermediate Form, and a pupil rating scale. Results from these tests are discussed in detail. (ED)

24. Fourier transform ion cyclotron resonance MS reveals the presence of a water molecule in an enzyme transition-state analogue complex

Abstract

The structures of several powerful inhibitors of hydrolytic enzymes resemble that of the altered substrate in the transition state, except that a hydrogen atom replaces one substituent (typically the leaving group). To test the hypothesis that a water molecule might be present in the gap resulting from this replacement, we examined a transition-state analogue complex formed by Escherichia coli cytidine deaminase by Fourier transform ion cyclotron resonance MS in electrospray mode. Upon nebularization from aqueous solution under conditions (pH 5.6) where the enzyme is active, cytidine deaminase remains dimeric in the vapor phase. In the presence of inhibitor, the enzyme's exact mass can be used to infer the presence at each active site of zinc, 5-fluoro-3,4-dihydrouridine, and a single water molecule.

39. Physiological and morphological changes in bovine mammary glands following intramammary infusion of recombinant interferon-gamma

Abstract

Eleven lactating dairy cows were used to evaluate the response of bovine mammary glands to increasing doses of recombinant bovine interferon (rBoIFN)-gamma. Right front and rear quarters were intramammarily infused with eight different doses (10(2) U to 2 x 10(8) U/quarter) of rBoIFN-gamma; each dose was tested in at least two quarters. Left udder halves served as within animal controls in which quarters were injected with a saline placebo or were not infused at all. Milk secretion samples for compositional analysis were collected from each quarter prior to infusion and at 6, 24, 36 and 48 h following infusion. Animals were slaughtered immediately following the 48 h sampling period and mammary tissue was obtained for morphometric analyses. Milk composition was similar between control quarters and those quarters infused with up to 10(5) U of rBo-IFN-gamma during the entire sampling period. Quarters infused with 10(6) U and 10(7) U of rBoIFN-gamma had higher milk somatic cell counts (SCC) following treatment compared with preinfusion values. Changes in the composition of mammary secretion were most dramatic in quarters infused with greater than or equal to 10(8) U of rBoIFN-gamma as indicated by the significant increase in SCC and milk pH with a concomitant decrease in lactose concentration when compared with pre-infusion values or with control quarters. Morphometric analysis of tissue demonstrated an increase in stroma, a decrease in luminal area, and a marked increase in the number of infiltrating leukocytes in those quarters infused with the higher doses of rBoIFN-gamma.(ABSTRACT TRUNCATED AT 250 WORDS)

Abstract

Bovine herpes virus type 1 (BHV-1) gIII is a major virion glycoprotein with homology to the immunoglobulin superfamily. We have investigated the possibility that gIII is related to host molecules and have identified a gIII-specific monoclonal antibody (mAb) that cross-reacts with normal bovine cells. The cross-reactive entity was expressed mainly on monocyte/macrophages (M phi), polymorphonuclear leucocytes (PMN) and platelets, and was identified as a 43,000-63,000 molecular weight (MW) cell-surface glycoprotein. For M phi, the glycoprotein appears to be a general lineage marker, rather than a maturation or activation marker, and may be a functional receptor, as evidenced by its endocytosis via coated pits and its involvement in proliferation of mononuclear cells in vitro. This novel leucocyte marker was also detected on subsets of human, ovine and canine M phi. Competitive binding assays with sera from cattle immunized with BHV-1 or gIII revealed apparent low responsiveness to the cross-reactive epitope. The results suggest that BHV-1 gIII is antigenically related to a novel host leucocyte receptor and that evasion and/or interference with leucocyte function may be a consequence of this molecular mimicry relationship.
