Denne masteroppgaven er et kvalitativt forskningsprosjekt om kreativitet i møte med alternative verktøy som maleredskap i grunnskolen. Hensikten med studien er å undersøke hva som skjer med den kreative prosessen i det man utforsker andre verktøy enn pensel som maleredskap, gjennom egne og læreres erfaringer. Hovedproblemstillinger er: Hvordan kan utforskning av ulike maleredskap påvirke den kreative prosessen? Den undersøkes gjennom to forskningsspørsmål: Hvilke erfaringer har et utvalg kunstlærere med elevenes kreative prosess ved bruk av alternative verktøy som maleredskap? og Hvilke didaktiske potensial synliggjøres i forskningsprosjektet mitt? Med hermeneutisk fenomenologi som vitenskapsteoretisk tilnærming har jeg valgt praksisledet forskning (også kalt practice-led research) som metodologi for å skaffe ny kunnskap gjennom skaping. Datamaterialet er generert gjennom eget utforskende arbeid med alternative maleredskap og intervju av et utvalg kunstlærerer med relevant erfaring. Gjennom forskerlogg fra eget utforskende arbeid har jeg skrevet narrativer for hvert maleredskap, og analysert narrativene gjennom en narrativ analyse. De semistrukturerte intervjuene har blitt analysert gjennom en tematisk analyse, hvor jeg har kodet og lett etter mønster. Analysen drøftes gjennom tre valgte analysetema, som også presenterer fagteoretiske perspektiver; kreativitet gjennom utforskning, engasjement og utforskertrang, og flow. Hovedkonklusjonene i forskningsprosjektet er at gjennom oppdagelser, nye ideer og erfaringer, kan utforskning av maleredskap bidra til å utvikle kreativiteten, enten man er elev, lærer eller kunstner. Utforskningen kan by på utfordringer og negative følelser som stagnering eller frustrasjon, men åpne for refleksjoner og videreutvikling. The purpose of this thesis is to examine how exploration of other tools than a painting brush can affect creativity, through my own and teachers experience. My main research question is: how can exploration of alternative painting tools affect the creative process? This question is examined through two sub-questions: What experience do art teachers have with pupils’ creative process when exploring with alternative painting tools? Also: what didactic potential does my research project highlight? With hermeneutic phenomenology as scientific theory, I’ve chosen practice-led research as methodology to acquire knowledge through creating. The research material is generated through my own exploration with alternative painting tools and interviews with teachers. I’ve written narratives for each tool and analyzed them through a narrative analysis. The interviews have been analyzed through a thematic analysis, where I’ve been coding and looking for patterns. The analysis is discussed through three chosen themes, that also includes theoretical perspectives: creativity through exploration, engagement and exploratory desire and flow. The main conclusion of this study is that through discoveries, ideas and experiences, exploration (of alternative painting tools) can contribute to developing creativity for pupils, teachers, and artists. Exploration can offer challenges and negative feelings such as stagnation or frustration but opens for reflection and further development. This article explores how physical surroundings may be integrated as a supportive measure in social work efforts. Drawing on ecological psychology and the concept of liminality, the article presents a case study of Kofoed’s School (KS), a social institution in Copenhagen, Denmark. In recent years, KS has undergone a major renovation, opening up previously sheltered workshops to the public. By creating liminal spaces of possibility, where students can take up “both/and” positions allowing for a multitude of ways to participate, students are experiencing increased support and inclusion, which contributes to a growing feeling of citizenship and well-being. Drawing on participant observations and interviews with students, staff members, as well as customers at the school’s shops, we explore how the architectural layout may facilitate students’ flexible and fluid movements between more or less sheltered positions and further discuss how this flexibility may become supportive for their personal development and well-being. We propose to think of such spaces of possibility as enabling spaces, where inclusive architecture contributes to the creation of new possibilities for participation for people in marginalized life positions. This, we suggest, holds a great potential for social work efforts for people experiencing complex social vulnerability.

Ocean warming along with nutrient enrichment are major stressors causing global seagrass decline. While the effects of global warming on metabolic parameters in seagrasses are well described, the effect of increasing temperature on the epiphytic overgrowth of seagrass leaves and the consequences for the seagrass plant are poorly understood. Here, we investigated the effects of elevating temperature on the photosynthetic efficiency of the seagrass species Zostera marina L. and its associated epiphytes, to explore how ocean warming might affect epiphytism in seagrasses. Gas exchange and final pH measurements on bare seagrass leaves, leaves with epiphytes, and epiphytes separated from seagrass leaves were used to quantify photosynthesis and respiration rates, and the inorganic carbon extraction capacity of leaves and epiphytes as a function of photon scalar irradiance and temperature (12, 17, 22, and 27°C). Seagrass without epiphytic biofilm had a high ability to exploit the incoming irradiance regardless of the light intensity and temperature, shown as continuously high light use efficiency and maximum net photosynthesis rates. The presence of epiphytic biofilm on the seagrass leaves impaired plant photosynthesis by increasing light requirements and reducing the photosynthetic efficiency (especially at 27°C). Epiphytes showed the lowest respiration rates in darkness and had the highest oxygen surplus over diel cycles up to 22°C, whereas bare leaves had the highest diel oxygen surplus at 27°C. Both bare leaves and epiphytes lost the ability to utilize bicarbonate at 27°C, and epiphytes also did not show use of bicarbonate at 12°C. Our results indicate a competitive advantage for epiphytes in cold CO2-rich environments, whereas seagrass with bare leaves could be less affected under elevated seawater temperatures.

I denne oppgaven skal jeg ta for meg følgende problemstilling; Hvordan kan lederskap påvirke sykefraværet i Norge? Oppgaven tar for seg en studie av IA-avtalens første delmål som handler om å minimere sykefraværet med 10% i forhold til 2018 og lederskap. Den tar for seg hvordan lederens erfaringer og leders stiler kan påvirke mediearbeidene, og ulike faktorer som omhandlet dette. Studie tar for seg ulike perspektiver til hvordan disse kan være en faktor for individets psykiske og fysiske helse. Studiet tar for seg organisasjoner, sett fra medarbeidernes ståsted, og ikke hvilke meninger ledelsene har rundt dette. Min konklusjon etter å ha sett på ulike studier og observasjoner at lederskap kan ha en stor påvirkningsfaktor for sykefraværet i en bedrift. Dette basert de ulike studiene jeg har tatt for meg.

Denne artikel tager udgangspunkt i recovery-tilgangens implicitte fordring om selvansvar ift. psykisk lidelse og problematiserer måden, hvorpå dette individualiseres som en del af arbejdet med recovery. Med afsæt i kulturhistorisk virksomhedsteori og økologisk psykologi søger artiklen at udforske og diskutere, hvordan det individuelle og kollektive ansvar er intimt forbundet, og hvad dette betyder i en socialpsykiatrisk praksissammenhæng. Det empiriske materiale udgøres af deltagerobservationer fra det japanske socialpsykiatriske tilbud, Bethel House, hvor man arbejder ud fra et motto om ”by myself, with others”. Her er arbejdet til dels centreret omkring ’tohjisha kenkyu’, en udforskende tilgang til psykiske vanskeligheder, og til dels et stærkt praksisfællesskab med fokus på fælles produktion af bl.a. jordbær og kombu. Artiklen påpeger nødvendigheden af en tydeligere prioritering af meningsfulde sociale fællesskaber i det socialpsykiatriske recovery-arbejde og et blik for betydningen af noget (meningsfuldt) at forpligte sig på – ellers risikerer fordringen om selv- ansvar at blive et tiltag, der fastlåser mennesker yderligere i deres tilværelser.

Contribution to DUT Time Capsule

Objective: This study introduced a lens of liminal theory, drawn from anthropological classical ritual theory, to explore how a preparatory teaching format using video cases influenced medical students’ patient approaches in their subsequent psychiatric clerkship.The video cases portrayed simulated patient-doctor encounters in diagnostic interview situations and were hypothesized to function as a liminal trickster. Methods: The study applied a qualitative explorative design using individual rich picture interviews. We asked the students to draw their experiences, which we investigated using a semi-structured interview guide designed to capture and unfold the students’ perspectives. We explored how students navigated insights from the preparatory teaching in their clerkship using liminal theory concepts in a mixed inductive and deductive thematic analysis. Results: The results from 8 rich picture interviews demonstrated that students’ ability to navigate insight gained from the video cases in their clerkship varied according to their roles in the clinical diagnostic interview situations. Students having active roles in the diagnostic interview situation adopted a patient-centred focus demonstrating empathic engagement and self-reflexivity related to their learning experiences with the video cases. Students with passive roles described a focus on how to adopt an appropriate appearance and copied the behaviour of the simulated doctors in the video cases. Conclusion: The liminal ritual theory perspective to explore the influence of preparatory teaching was useful for demonstrating how video cases could affect students’ patient-centred learning. Without guidance and active roles in clerkship, medical students’ learning experiences may lead to a prolonged liminal phase and may not capitalise on the potentially positive effects of the preparatory teaching. Liminal theory may further inform our understanding of students’ learning considering patient cases in educational technology arrangements as tricksters.

Purpose/objectives: Instructional videos may demonstrate the execution of complex clinical procedures and the cooperation between members of the dental team better than traditional slide-based teaching materials. The aim of the present study was to compare the effect of a procedural video on student ratings to a traditional still-image-based presentation in a course on rubber dam application. Methods: In a randomized, double-blind, parallel arm design, participants (46 dental students) completed a seven-item, five-step Likert-scale questionnaire at baseline (t1), after a video-based or slide-based demonstration of rubber dam application (t2) and after hands-on training (t3). The students’ judgement on the benefits of rubber dam (items 1–3), their motivation to use rubber dam (item 4), their self-efficacy (items 5–6) and their expected use of the teaching material (item 7) were assessed. Changes in the students’ individual answers were analyzed for each item and comparison between intervention groups made. Moreover, the impact of the teaching format on in-class discussions was analyzed qualitatively using a thematic approach. Results: Both interventions arose comparable significant improvement in the students’ Likert-scale ratings from t1 to t2, and again from t2 to t3. No significant differences between intervention groups were found in the students’ ratings or in the qualitative analysis. Conclusions: Procedural videos have proven to be a valuable learning aid in a variety of teaching formats, but in the context of a live lecture, they may not constitute an improvement over traditional text- and still-image-based presentations.

We report high resolution measurements of the stable isotope ratios of ancient ice (δ18O, δD) from the North Greenland Eemian deep ice core (NEEM, 77.45° N, 51.06° E). The record covers the period 8–130 ky b2k (y before 2000) with a temporal resolution of ≈0.5 and 7 y at the top and the bottom of the core respectively and contains important climate events such as the 8.2 ky event, the last glacial termination and a series of glacial stadials and interstadials. At its bottom part the record contains ice from the Eemian interglacial. Isotope ratios are calibrated on the SMOW/SLAP scale and reported on the GICC05 (Greenland Ice Core Chronology 2005) and AICC2012 (Antarctic Ice Core Chronology 2012) time scales interpolated accordingly. Easily accessible biomarkers for Alzheimer&rsquo, s dementia (AD) are lacking and established clinical markers are limited in applicability. Blood is a common biofluid for biomarker discoveries, and extracellular vesicles (EVs) may provide a matrix for exploring AD related biomarkers. Thus, we investigated proteins related to neurological and inflammatory processes in plasma and EVs. By proximity extension assay (PEA), 182 proteins were measured in plasma and EVs from patients with AD (n = 10), Mild Cognitive Impairment (MCI, n = 10), and healthy controls (n = 10). Plasma-derived EVs were enriched by 20,000&times, g, 1 h, 4 &deg, C, and confirmed using nanoparticle tracking analysis (NTA), western blotting, and transmission electron microscopy with immunolabelling (IEM). Presence of CD9+ EVs was confirmed by western blotting and IEM. No group differences in particle concentration or size were detected by NTA. However, significant protein profiles were observed among subjects, particularly for EVs. Several proteins and their ratios could distinguish cognitively affected from healthy individuals. For plasma TGF-&alpha, │CCL20 (AUC = 0.96, 95% CI = 0.88&ndash, 1.00, p = 0.001) and EVs CLEC1B│CCL11 (AUC = 0.95, 95% CI = 0.86&ndash, 1.00, p = 0.001) showed diagnostic capabilities. Using PEA, we identified protein profiles capable of distinguishing healthy controls from AD patients. EVs provided additional biological information related to AD not observed in plasma alone.

De seneste års politiske og institutionelle fokus på kvalitetssikring i undervisningen på de videregående uddannelser har afdækket et behov for at anerkende undervisning på lige fod med forskning. Peer-review anvendes til anerkendelse af forskning, men kan en lignende metode også anvendes til anerkendelse af undervisning? Og hvad sker der i en organisationskultur, når peer-review af undervisningen introduceres som pædagogisk kompetenceudvikling? I denne artikel præsenteres resultaterne af et forskningsprojekt, der udviklede, afprøvede og undersøgte en pædagogisk kompetenceudviklingsmodel inspireret af peer-review – en Faciliteret Multi-Source Feedback-model (FMSF-model). I et kvalitativt interviewstudie undersøgte vi deltagernes (n=15) oplevelse af FMSF-modellen og dens anvendelighed i et organisatorisk læringsperspektiv. Resultaterne peger på, at FMSF-modellen kan facilitere handlingsorienteret refleksion til gavn for undervisernes pædagogiske kompetenceudvikling og understøtte en organisatorisk læringsstruktur qua dens målrettede og strukturerede feedbackformat. Modellens anvendelighed påvirkes dog af lokalt forankrede kulturelle normer og praksisser, f.eks. sparsom peer-feedback og -observation samt manglende transparens blandt deltagerne.

The aim of this article is to shed light on how environmental standards in the life of youths influence the development of self. We propose the concept of 'subjectified subjectivity' to grasp these person-environment dialectics in a general form. By elaborating on these conceptual understandings of youth life, the article also seeks to understand young people from their own perspectives on life and from their developing life-perspectives, rather than from general categories. Based on one of the author's data from her study of young people in their transition to (and through the first year of) high school, we carry out an analysis of a 16-year old high school student and how her approach to beer, to beer drinking as a part of Danish high school life-style, and to herself changes over time. We suggest a dialectical-ecological model to analyze the dialectical and synthetic movements over time of the girl and her environments.

Employing student-driven project work in a higher education setting challenges not only the way in which we understand students&rsquo, learning and how we define the expected learning outcomes, it also challenges our ways of assessing students&rsquo, learning. This paper will address this question specifically and illustrate with a case that highlights some of the challenges that may arise in practice when assessing student-driven, problem-based projects. The case involved an assessment situation in which a discrepancy arose between the internal and external examiner in relation to what was valued. The discrepancy had consequences not only for the concrete assessment of students&rsquo, work, but also for the validity of the problem-based university pedagogy in general, and it raised the question of how to assess students&rsquo, work adequately. The research focus of this study was to explore the implications of assessing student-driven projects within a progressive approach to higher education teaching, along with the potential underlying issues. We found a need for clear assessment criteria while insisting on a space for students&rsquo, creativity and reflexivity as essential parts of a learning process. The paper thus makes a case for the notion of reflexivity as an assessment criterion to be integrated into learning objectives.

Objectives The aim of this study was to explore medical stu- dents’ learning experiences from the didactic teaching formats using either text-based patient cases or video-based patient cases with similar content. The authors explored how the two different patient case formats influenced students’ perceptions ofpsychiatric patients and students’ reflections on meeting and communicating with psychiatric patients. Methods The authors conducted group interviews with 30 medical students who volunteered to participate in interviews and applied inductive thematic content analysis to the tran- scribed interviews. Results Students taught with text-based patient cases empha- sized excitement and drama towards the personal clinical nar- ratives presented by the teachers during the course, but never referred to the patient cases. Authority and boundary setting were regarded as important in managing patients. Students taught with video-based patient cases, in contrast, often referred to the patient cases when highlighting new insights, including the importance of patient perspectives when communicating with patients. Conclusion The format of patient cases included in teaching may have a substantial impact on students’ patient-centered- ness. Video-based patient cases are probably more effectivethan text-based patient cases in fostering patient-centered per- spectives in medical students. Teachers sharing stories from their own clinical experiences stimulates both engagement and excitement, but may also provoke unintended stigma and influence an authoritative approach in medical students towards managing patients in clinical psychiatry.

Summary Background Myocardial injury after non-cardiac surgery (MINS) increases the risk of cardiovascular events and deaths, which anticoagulation therapy could prevent. Dabigatran prevents perioperative venous thromboembolism, but whether this drug can prevent a broader range of vascular complications in patients with MINS is unknown. The MANAGE trial assessed the potential of dabigatran to prevent major vascular complications among such patients. Methods In this international, randomised, placebo-controlled trial, we recruited patients from 84 hospitals in 19 countries. Eligible patients were aged at least 45 years, had undergone non-cardiac surgery, and were within 35 days of MINS. Patients were randomly assigned (1:1) to receive dabigatran 110 mg orally twice daily or matched placebo for a maximum of 2 years or until termination of the trial and, using a partial 2-by-2 factorial design, patients not taking a proton-pump inhibitor were also randomly assigned (1:1) to omeprazole 20 mg once daily, for which results will be reported elsewhere, or matched placebo to measure its effect on major upper gastrointestinal complications. Research personnel randomised patients through a central 24 h computerised randomisation system using block randomisation, stratified by centre. Patients, health-care providers, data collectors, and outcome adjudicators were masked to treatment allocation. The primary efficacy outcome was the occurrence of a major vascular complication, a composite of vascular mortality and non-fatal myocardial infarction, non-haemorrhagic stroke, peripheral arterial thrombosis, amputation, and symptomatic venous thromboembolism. The primary safety outcome was a composite of life-threatening, major, and critical organ bleeding. Analyses were done according to the intention-to-treat principle. This trial is registered with ClinicalTrials.gov, number NCT01661101. Findings Between Jan 10, 2013, and July 17, 2017, we randomly assigned 1754 patients to receive dabigatran (n=877) or placebo (n=877); 556 patients were also randomised in the omeprazole partial factorial component. Study drug was permanently discontinued in 401 (46%) of 877 patients allocated to dabigatran and 380 (43%) of 877 patients allocated to placebo. The composite primary efficacy outcome occurred in fewer patients randomised to dabigatran than placebo (97 [11%] of 877 patients assigned to dabigatran vs 133 [15%] of 877 patients assigned to placebo; hazard ratio [HR] 0·72, 95% CI 0·55–0·93; p=0·0115). The primary safety composite outcome occurred in 29 patients (3%) randomised to dabigatran and 31 patients (4%) randomised to placebo (HR 0·92, 95% CI 0·55–1·53; p=0·76). Interpretation Among patients who had MINS, dabigatran 110 mg twice daily lowered the risk of major vascular complications, with no significant increase in major bleeding. Patients with MINS have a poor prognosis; dabigatran 110 mg twice daily has the potential to help many of the 8 million adults globally who have MINS to reduce their risk of a major vascular complication. In the current study, our aim was to demonstrate that EmbryoScope incubation conditions is comparable to standard laboratory incubation circumstances by comparing embryo quality, development and ongoing pregnancy rates between the EmbryoScope (ES) and a standard incubator (SI). We analyzed 478 embryos from 60 couples undergoing oocyte donation were included in the study.All embryos retrieved from a patient were randomly distributed in the ES or SI. We calculated blastocyst development rate, blastocyst viability and ongoing pregnancy rate for embryo transfers from ES, SI and mixed (one embryo from the ES and one from the SI). Statistical analysis was conducted by Chi square tests, considering p0.05 significant.No significant differences were found between the ES and SI from all the parameters evaluated.Thus we concluded that time-lapse monitoring in the EmbryoScope does not impair embryo quality while allowing for morphological, spatial and temporal analysis of embryo development.

Inbreeding is expected to increase the variability in size and shape within populations. The distinct effects of inbreeding on size and shape suggest that they are governed by different developmental pathways. One unresolved question is whether the non-allometric shape component is partially unconstrained developmentally and therefore whether shape is evolvable. In the present study, we utilized a mass outbred population of Drosophila melanogaster maintained at standard laboratory conditions. Eight lines with equivalent expected levels of inbreeding (F a 0.67) were obtained by restricting the size of each population to two pairs for nine generations. Nine landmarks were measured on Drosophila wings of the inbreed lines and compared with those of the mass population. Wing landmarks comprise an excellent model system for studying evolution of size and shape. Landmark measurements were analyzed with a Procrustes generalized least squares procedure. To visualize global shape changes among samples, we reconstructed the mean shape and the shape changes related to both the allometric and non-allometric components. An increased variability in the non-allometric shape component was found with inbreeding. This indicated that shape was not entirely developmentally constrained, and therefore that shape appears to be evolvable. © 2011 The Linnean Society of London, Biological Journal of the Linnean Society, 2011, 102, 626-634.

Mechanistic and functional studies using ‘omic' technologies can increase the understanding of the consequences of inbreeding, from the level of DNA to that of population growth. This gives added power to unravelling the causes of inbreeding depression, results that we suggest will be useful in animal and plant breeding and in evolutionary and conservation biology. First results from ‘omic' investigations of inbreeding indicate that inbreeding affects cellular processes that are also observed with aging and exposure to environmental stress. Genetic variation that is expressed only under specific environmental conditions can contribute to additional adverse effects of inbreeding if environmental conditions change. We present a proteomic characterization of a conditional lethal found in an inbred line of Drosophila melanogaster. The lethal effect is apparent as a large increase in early mortality at the restrictive temperature (29 °C) as opposed to normal survival at the permissive temperature (20 °C). The increased mortality in response to the restrictive temperature is probably caused by a single recessive major locus. A quantitative trait locus (QTL) region segregating variation affecting the lethal effect has been identified, allowing for a separation of primary/causal effects and secondary consequences in the proteome expression patterns observed. In this study, the proteomic response to the restrictive temperature in the lethal-line (L-line) was compared with the response in an inbred-control-line (IC-line) and an outbred-control-line (OC-line). Quantitative protein changes were detected using isobaric tags for relative and absolute quantitation (iTRAQ) two-dimensional liquid chromatography-tandem mass spectrometry. In all, 45 proteins were found to be significantly differently regulated in response to the restrictive temperature in the L-line as compared with the IC-line. No proteins were significantly differently regulated between the IC-line and the OC-line, verifying that differential protein regulation was specific to a genetic defect in the L-line. Proteins associated with oxidative phosphorylation and mitochondria were significantly overrepresented within the list of differentially expressed proteins. Quantitative protein changes were detected using isobaric tags for relative and absolute quantitation (iTRAQ) two-dimensional liquid chromatography-tandem mass spectrometry. In all, 45 proteins were found to be significantly differently regulated in response to the restrictive temperature in the L-line as compared with the IC-line. No proteins were significantly differently regulated between the IC-line and the OC-line, verifying that differential protein regulation was specific to a genetic defect in the L-line. Proteins associated with oxidative phosphorylation and mitochondria were significantly overrepresented within the list of differentially expressed proteins. Proteins related to muscle contraction were also found to be differentially expressed in the L-line in response to the restrictive temperature, supporting phenotypic observations of moribund muscle hyper-contraction.

Purpose: To assess irradiance and total energy dose from different microscopes during the in-vitro embryonic developmental cycle in mouse and pig and to evaluate its effect on embryonic development and quality in pig. Method: Spectral scalar irradiance (380-1050 nm) was measured by a fiber-optic microsensor in the focal plane of a dissection microscope, an inverted microscope and a time-lapse incubation system. Furthermore, the effect of three different red light levels was tested in the time-lapse system on mouse zygotes for 5 days, and on porcine zona-intact and zona-free parthenogenetically activated (PA) embryos for 6 days. Results: The time-lapse system used red light centered at 625 nm and with a lower irradiance level as compared to the white light irradiance levels on the dissection and inverted microscopes, which included more energetic radiation

The complete genome of Chlamydia pneumoniae contains a total of 21 genes encoding polymorphic membrane proteins (Pmp). From this large Pmp family three genes, pmp8, pmp10 and pmp11, were cloned and antibodies against recombinant full-length Pmp proteins were produced. Indirect immunofluorescence microscopy of HEp-2 cells infected with C. pneumoniae CWL029 was performed with the Pmp antibodies in combination with a Chlamydia-specific anti-lipopolysaccharide (LPS) antibody. This double staining technique clearly showed that expression of Pmp10 was differential. Additional double staining with monoclonal antibodies to the surface of C. pneumoniae elementary bodies and the anti-LPS antibody resulted in identification of seven monoclonal antibodies that reacted identically to the Pmp10 antibody indicating that Pmp10 is an immunodominant protein. Finally, the molecular mechanism responsible for differential expression is suggested to be variation in the guanine residues in the polyG tract of pmp10.

Inbreeding depression is a widespread phenomenon of central importance to agriculture, medicine, conservation biology and evolutionary biology. Although the population genetic principles of inbreeding depression are well understood, we know little about its functional genomic causes. To provide insight into the molecular interplay between intrinsic stress responses, inbreeding depression and temperature tolerance, we performed a proteomic characterization of a well-defined conditional inbreeding effect in a single line of Drosophila melanogaster, which suffers from extreme cold sensitivity and lethality. We identified 48 differentially expressed proteins in a conditional lethal line as compared to two control lines. These proteins were enriched for proteins involved in hexose metabolism, in particular pyruvate metabolism, and many were found to be associated with lipid particles. These processes can be linked to known cold tolerance mechanisms, such as the production of cryoprotectants, membrane remodeling and the build-up of energy reserves. We checked mRNA-expression of seven genes with large differential protein expression. Although protein expression poorly correlated with gene expression, we found a single gene (CG18067) that, after cold shock, was upregulated in the conditional lethal line both at the mRNA and protein level. Expression of CG18067 also increased in control flies after cold shock, and has previously been linked to cold exposure and chill coma recovery time. Many differentially expressed proteins in our study appear to be involved in cold tolerance in non-inbred individuals. This suggest the conditional inbreeding effect to be caused by misregulation of physiological cold tolerance mechanisms.

Mating between closely related individuals often causes reduced fitness, which is termed ‘inbreeding depression’. Inbreeding is, therefore, a threat towards the persistence of animal and plant populations. Here we present methods and results from a practical for high-school and first-year university students and discuss learning outcomes of the exercise as an example of inquiry-based science teaching. We use the model organism Drosophila melanogaster to test the ability of inbred and control (non-inbred) females to survive heat stress exposure. Flies were anaesthetised and collected into vials before exposure to 38°C heat stress in a water bath for 1 h. Half an hour later the number of comatose inbred and control flies were scored and chi-square statistic procedures were used to test for different degrees of heat stress tolerance between the two lines of flies. The practical introduces the students to central topics in population genetics and demonstrates potential consequences of inbreeding and environmental stress on fitness. Furthermore, the practical gives insight into how to design, analyse, and interpret the results of a scientific study.

Brugerinddragelse og recovery står øverst på dagsordenen hos de fleste so- cialforvaltninger i de her år. Men hvordan skabes og understøttes brugerinddragelse og recovery side om side med et neoliberalistisk tankegods i form af individualisering og isole- ring af problemstillingerne, managementlogik- ker og økonomisk rationalisering? Vor erfaring med indarbejdning af netop brugerinddragelse og elementer af recovery- tanken (ved genetablering af solide forbindel- ser til samfundet og udvidelse af handleevnen; med henblik på at gøre videre institutiona- lisering unødvendig) har været, at så længe managementlogikkerne og den økonomiske ra- tionalisering ikke udfordres eller anfægtes, så er der åbenhed for både brugerinddragelse og recovery-orienteret arbejde. Vore erfaringer giver anledning til at reflektere over, hvorvidt vi har at gøre med et system af sociale hjæl- peforanstaltninger, der, helt selvmodsigende med sine indbyggede logikker, forhindrer det egentlige formål – nemlig at hjælpe mennesker i en udsat og marginaliseret position til en god og meningsfuld tilværelse.

Udgivelsesdato: 2010-Oct The purpose of this study was to evaluate the relevance of long-term endothelial cell culture as a model system of vascular ageing. Micro- and macrovascular endothelial cells were serially passaged until replicative senescence and their ability to form tube-like structures when cultured on Matrigel was assessed throughout their lifespan. For both cell types low passage cultures adopted a homogeneous cobblestone morphology, while senescent cultures were extremely heterogeneous. Furthermore, both cell types showed a reduction in tube formation ability with in vitro ageing, which is in accordance with the reduction in angiogenic potential observed with ageing in vivo. Examination of senescence associated β-galactosidase activity revealed an increased activity in cells forming tubes as compared to cells cultured on plastic, which could be attributed to an increased lysosomal content of cells undergoing tube formation. As this increased senescence associated β-galactosidase activity was unrelated to the replicative age of the cells, senescence associated β-galactosidase activity may not be a relevant senescence marker for differentiating endothelial cells. The age-related reduction in tube formation ability suggested that long-term culture of endothelial cells may be a valid model system of vascular ageing, which makes it an ideal platform for high throughput screening of compounds influencing angiogenesis.

The majority of experimental studies of the effects of population bottlenecks on fitness are performed under laboratory conditions, which do not account for the environmental complexity that populations face in nature. In this study, we test inbreeding depression in multiple replicates of inbred when compared with non-inbred lines of Drosophila melanogaster under different temperature conditions. Egg-to-adult viability, developmental time and sex ratio of emerging adults are studied under low, intermediate and high temperatures under laboratory as well as semi-natural conditions. The results show inbreeding depression for egg-to-adult viability. The level of inbreeding depression is highly dependent on test temperature and is observed only at low and high temperatures. Inbreeding did not affect the developmental time or the sex ratio of emerging adults. However, temperature affected the sex ratio with more females relative to males emerging at low temperatures, suggesting that selection against males in pre-adult life stages is stronger at low temperatures. The coefficient of variation (CV) of egg-to-adult viability within and among lines is higher for inbred flies and generally increases at stressful temperatures. Aims: To examine sensitivities of various Drosophila melanogaster strains towards human pathogenic and nonpathogenic gram-positive bacteria.Methods and Results: The D. melanogaster Oregon R strain was infected by injecting the thorax with a needle containing Escherichia coli (negative control), Listeria monocytogenes, Staphylococcus aureus (both food-borne pathogens), Listeria innocua, Bacillus subtilis, Carnobacterium maltaromaticum, Lactobacillus plantarum or Pediococcus acidilactici (all nonpathogenic bacteria). Listeria monocytogenes and S. aureus killed the host rapidly compared with the negative control. Infection with L. innocua, B. subtilis or C. maltaromaticum also resulted in a high fly mortality, whereas Lact. plantarum and P. acidilactici resulted in a slightly increased mortality. Four additional D. melanogaster lines, three of which had been selected for heat, cold and desiccation resistance respectively, were subjected to infection by L. monocytogenes, S. aureus and E. coli. Mortality rates were comparable with that of the Oregon R strain.Conclusions: Use of the injection method shows the limitation of D. melanogaster as a model host for gram-positive bacteria as opportunistic infection by nonpathogenic gram-positive bacteria results in partial or high mortality. In addition, lines of fruit flies resistant to various stress exposures did not show an increased resistance to infection by gram-positive pathogens under the conditions tested.Significance and Impact of the Study: This study demonstrates the inadequacy of D. melanogaster infected by the injection method in order to distinguish between virulent and nonvirulent gram-positive bacteria.

Genomewide gene expression patterns were investigated in inbred and noninbred Drosophila melanogaster lines under benign and stressful (high temperature) environmental conditions in a highly replicated experiment using Affymetrix gene chips. We found that both heat-shock protein and metabolism genes are strongly affected by temperature stress and that genes involved in metabolism are differentially expressed in inbred compared with noninbred lines, and that this effect is accentuated after heat stress exposure. Furthermore we show that inbreeding and temperature stress cause increased between-line variance in gene expression patterns. We conclude that inbreeding and environmental stress both independently and synergistically affect gene expression patterns. Interactions between inbreeding and the environment are often observed at the phenotypic level and our results reveal some of the genes that are involved at the individual gene level. Our observation of several metabolism genes being differentially expressed in inbred lines and more so after exposure to temperature stress, together with lower fitness in the investigated inbred lines, supports the hypothesis that superiority of heterozygous individuals partly derives from increased metabolic efficiency.

Induction of heat shock proteins (Hsp) is a well-known mechanism through which cells cope with stressful conditions. Hsp are induced by a variety of extrinsic stressors. However, recently intrinsic stressors (aging and inbreeding) have been shown to affect expression of Hsp. Increased homozygosity due to inbreeding may disrupt cellular homeostasis by causing increased expression of recessive deleterious mutations and breakdown of epistatic interactions. We investigated the effect of inbreeding and the rate of inbreeding on the expression of Hsp70, larval heat resistance and fecundity. In Drosophila melanogaster we found that inbred lines (F approximately 0.67) had significantly up-regulated expression of Hsp70, and reduced heat resistance and fecundity as compared with outbred control lines. A significant negative correlation was observed between Hsp70 expression and resistance to an extreme heat stress in inbred lines. We interpreted this as an increased requirement for Hsp70 in the lines suffering most from inbreeding depression. Inbreeding depression for fecundity was reduced with a slower rate of inbreeding compared with a fast rate of inbreeding. Thus, the effectiveness of purging seems to be improved with a slower rate of inbreeding.

Inbreeding is expected to decrease the heritability within populations. However, results from empirical studies are inconclusive. In this study, we investigated the effects of three breeding treatments (fast and slow rate of inbreeding - inbred to the same absolute level - and a control) on heritability, phenotypic, genetic and environmental variances of sternopleural bristle number in Drosophila melanogaster. Heritability, and phenotypic, genetic and environmental variances were estimated in 10 replicate lines within each of the three treatments. Standard least squares regression models and Bayesian methods were used to analyse the data. Heritability and additive genetic variance within lines were higher in the control compared with both inbreeding treatments. Heritabilities and additive genetic variances within lines were higher in slow compared with fast inbred lines, indicating that slow inbred lines retain more evolutionary potential despite the same expected absolute level of inbreeding. The between line variance was larger with inbreeding and more than twice as large in the fast than in the slow inbred lines. The different pattern of redistribution of genetic variance within and between lines in the two inbred treatments cannot be explained invoking the standard model based on selective neutrality and additive gene action. Environmental variances were higher with inbreeding, and more so with fast inbreeding, indicating that inbreeding and the rate of inbreeding affect environmental sensitivity. The phenotypic variance decreased with inbreeding, but was not affected by the rate of inbreeding. No inbreeding depression for mean sternopleural bristle number was observed in this study. Considerable variance between lines in additive genetic variance within lines was observed, illustrating between line variation in evolutionary potential.

The deleterious consequences of inbreeding, especially in the form of inbreeding depression, are well known. However, little is known about how inbreeding affects genome-wide gene expression. Here, we show that inbreeding changes transcription levels for a number of genes. Gene expression profiles of Drosophila melanogaster lines inbred to F ≈ 0.67 at different rates changed relative to those of noninbred lines, but the rate of inbreeding did not significantly affect gene expression patterns. Genes being differentially expressed with inbreeding are disproportionately involved in metabolism and stress responses, suggesting that inbreeding acts like an environmental stress factor.

Chlamydia pneumoniae is an obligate intracellular bacterium that causes upper and lower respiratory tract infection in humans. C. pneumoniae harbors the polymorphic membrane protein (Pmp) family with 21 different proteins with a molecular mass around 100 kDa. The Pmps are species-specific, abundant and, together with major outer membrane protein and outer membrane protein 2, the dominant proteins in the C. pneumoniae outer membrane complex. Nevertheless, it is unknown whether Pmps are recognized by the cell-mediated immune response. To address this issue, C57BL/6J mice were infected intranasally with C. pneumoniae and the immune response to primary infection was investigated. We demonstrate, as expected, that the primary response is of the Th1 type by IgG2a- and IgG1-specific sELISA (Medac) on serum. In vivo-primed spleen lymphocytes were found to be reactive to Pmp8, Pmp20 and Pmp21 in an interferon-gamma ELISpot assay. The responses were shown to be mediated by CD4(+) T cells. To our knowledge, this is the first identification of antigens recognized by CD4(+) T cells during murine C. pneumoniae infection.

Background Chlamydiae are obligate intracellular bacteria, which are important human pathogens. Genome sequences of C. trachomatis and C. pneumoniae have revealed the presence of a Chlamydia specific gene family encoding polymorphic outer membrane proteins, Pmps. In C. pneumoniae the family comprises twenty-one members, which are all transcribed. In the present study, the expression, processing and localisation of the sixteen full-length Pmps in C. pneumoniae strain CWL029 have been further investigated by two-dimensional gel electrophoresis and immunofluorescence microscopy.
