Normal epithelial cells exert their competitive advantage over RasV12-transformed cells and eliminate them into the apical lumen via cell competition. However, the internal or external factors that compromise cell competition and provoke carcinogenesis remains unknown. In this study, we examined the effect of sequential accumulation of gene mutations, mimicking multi-sequential carcinogenesis on RasV12-induced cell competition in intestinal epithelial tissues. Consequently, we found that directionality of RasV12-cell extrusion in Wnt-activated epithelia is reversed, and transformed cells are delaminated into the basal lamina via non-cell autonomous MMP21 upregulation. Subsequently, diffusively infiltrating, transformed cells develop into highly invasive carcinomas. Elevated production of MMP21 is elicited partly through NF-κB signaling, blockage of which restores apical elimination of RasV12 cells. We further found that the NF-κB-MMP21 axis is significantly bolstered in early colorectal carcinoma in humans. Collectively, this study shows that cells with high mutational burdens exploit cell competition for their benefit by behaving as unfit cells, endowing them with an invasion advantage.

The mechanism of H. pylori-induced atrophy and metaplasia has not been fully understood. Here, we demonstrate the novel role of Apoptosis signal-regulating kinase 1 (ASK1) and downstream MAPKs as a regulator of host immune responses and epithelial maintenance against H. pylori infection. ASK1 gene deficiency resulted in enhanced inflammation with numerous inflammatory cells including Gr-1+CD11b+ myeloid-derived suppressor cells (MDSCs) recruited into the infected stomach. Increase of IL-1β release from apoptotic macrophages and enhancement of TH1-polarized immune responses caused STAT1 and NF-κB activation in epithelial cells in ASK1 knockout mice. Dysregulated immune and epithelial activation in ASK1 knockout mice led to dramatic expansion of gastric progenitor cells and massive metaplasia development. Bone marrow transplantation experiments revealed that ASK1 in inflammatory cells is critical for inducing immune disorder and metaplastic changes in epithelium, while ASK1 in epithelial cells regulates cell proliferation in stem/progenitor zone without changes in inflammation and differentiation. These results suggest that H. pylori-induced immune cells may regulate epithelial homeostasis and cell fate as an inflammatory niche via ASK1 signaling.

河道湾曲部に設置された水制工が水の流れに対してどのような制御効果をもち, また, 水制周辺ではどのような水理現象が生じるのか, 水理模型実験と数値シミュレーションにより検討した. 数値シミュレーションについては, 流れおよび河床変動の解析を幅広く汎用的に行うことができるよう, 既往の平面2次元浅水流モデルを基本とする簡易で実用的なシミュレーションモデルを提案している. また, 水制工の護岸・護床機能を予測してその適切な設置を検討するために, シミュレーションモデルをいかに適用すればよいか, 考察した. 水制工の形式としては, 不透過型水制の例として出し型水制を, 透過型水制の例としてスクリーン枠を対象とした.

本研究は, 土石流中の巨礫の衝突を受ける鋼製透過型砂防構造物の耐衝撃設計法を確立するため, まず, 局部変形とひずみ速度効果の影響を考慮した3次元鋼管骨組の弾塑性衝撃応答解析法を開発した. 次に, 本解析法を用いて部材に発生する塑性回転角を算定し, これを部材の動的塑性回転容量と比較することにより終局限界の判定を行う耐衝撃設計法を提案した. さらに, B型スリットダムに巨礫が衝突した場合の具体的な設計例を示すとともに, 現行設計法との比較検討を行い, 本設計法が現行設計法に比べより合理的かつ経済的な設計法であることを確認した.

本研究は, 土石流中の巨礫の衝突を受ける鋼製透過型砂防ダムの耐衝撃設計に対する基礎的資料を提供するため, 高速載荷荷重を受ける鋼管はり部材の動的終局限界について検討を行ったものである. まず, 鋼製透過型砂防ダムの終局限界を規定する物理量として塑性回転角に着目し, 鋼管片持はりの静的および高速載荷実験を行い, 静的および動的塑性回転容量を実験的に求めた. 次に, 鋼管はり部材の塑性回転容量を解析的に求めるため, 固定端付近に発生する局部座屈が鋼管短柱に発生する局部座屈に等しいと考え, 鋼管の静的および高速短柱圧縮試験を行った. さらに得られた実験結果を基に, 鋼管はり部材の動的塑性回転容量を算定する手法および実験式を提案したものである.

本研究は, 土石流中の巨礫の衝突を受ける鋼製透過型砂防構造物の耐衝撃設計への基礎的資料を提供することを目的とし, まず鋼管固定はりを対象として静的および高速載荷実験を行い, 荷重～局部変形算定式を導いた. 次に衝突点の局部変形後の断面形状をひょうたん形ほか3種類に変化させて, その妥当性を全塑性モーメント (塑性断面係数) の観点から検討した. さらに, ひずみ速度効果を考慮した準静的解析法を開発し, 球状載荷体の衝突を受ける鋼管固定はりの最大応答変位を算定するとともに, 衝突実験結果との比較検討を試みたものである.

The thermal decomposition and hydrogen etching of a 6H-SiC(0001) Si-face were directly monitored using an in situ gravimetric monitoring system. The monitoring of the weight change of the 6H-SiC Si-face using this system clarified the dependences of the thermal decomposition and hydrogen etching rates on the substrate temperature. Although the thermal decomposition of the 6H-SiC Si-face above 1400 °C generated a graphite layer since only the Si atom directly desorbs from the surface, the etching of the 6H-SiC Si-face by hydrogen did not form this layer, and both Si and C atoms react with hydrogen. Moreover, the surface reaction of the 6H-SiC Si face with H2 and the resultant surface morphology were found to change at approximately 1250 °C.
