High altitude (HA) poses various dangers to living organisms, the most serious of which are oxidative stress and its accompanying metabolic problems, as well as hypoxia and its associated metabolic abnormalities. Acrylamide is a poisonous chemical produced under the oxidative stress as a result of intracellular reactive oxygen species formation and toxicity. Heating carbohydrates-rich meals give acrylamide that is widely used in the industry. The precise mechanism of acrylamide toxicity is unknown. The current study aimed to examine the impacts of Taify pomegranate juice (TPJ) from Taif area (Punica granatum L.) on acrylamide-induced testicular stress and dysfunction. Twenty four male of adult Wistar rats, were divided into four groups: Group 1 was a negative-control received only saline; Group 2 was a positive-control that received orally acrylamide (20 mg/kg/bw) for consecutive 4 weeks; and Group 3 received TPJ; 2 mL/kg/bw, orally for 4 weeks. Rats in Group 4 received pomegranate juice and acrylamide as described in Groups 2 and 3, with the TPJ delivered 1 week before the acrylamide. Acrylamide elevated serum levels of inflammatory cytokines (interleukin-1 (IL-1), interleukin-6 (IL-6), and tumor necrosis factor alpha (TNF-α)). Moreover, acrylamide decreased concentrations of follicle stimulating hormone (FSH), leutinizing hormone (LH), and testosterone and altered semen characteristics (decreased sperm viability and concentration, and increased sperm abnormalities). Acrylamide increased malondialdehyde levels (MDA), while it decreased antioxidant activities (superoxide dismutase; SOD and reduced glutathione; GSH). The inflammation associated cytokines were restored by TPJ administration to acrylamide rats. GSH and SOD were significantly recovered to near control levels in TPJ plus acrylamide group compared to acrylamide-administered rats. TPJ preadministration to rats restored semen profiles, alteration in testis pathology, and normalized the changes on the male reproductive hormones affected by acrylamide. Furthermore, the TPJ reversed the upregulation in caspases-3 and the decrease in B-cell lymphoma-2 (Bcl-2) gene expressions affected by acrylamide, with significant upregulation of hemoxygenase-1 (HO-1) and nuclear factor erythroid-2 (Nrf2) mRNA expression. Current study examined the protective impact of Rhodiola rosea (R. rosea) extract on hepatic toxicity markers caused by MSG. MSG induced significant increases in hepatic stress biomarkers. There were significant declines in GSH, catalase and SOD levels, which were accompanied by substantial elevations in IL-1β, IL-6, and TNF-α. There was hepatic cellular damage, and upregulation of caspase-3 and a decrease in Bcl-2 expression. The hepatotoxic impacts of MSG were normalized by Rhodiola supplementation. R. rosea downregulated caspase-3 and upregulated HO-1 and Nrf2 mRNA expression. R. rosea regulated the immunoreactivity of COX-2, TGF-β1, and NFkB genes associated with hepatic toxicity. R. rosea extract normalized histopathological changes induced by MSG and restored hepatic architecture. R. rosea extract exhibited anti-apoptotic, antioxidant, and anti-inflammatory properties. In conclusion, R. rosea extract is a promising medication against hepatic toxicity induced by MSG, which works by regulating the different signaling pathways of inflammation-, oxidative-stress-, and apoptosis-associated markers. GA3 is widely used as a growth stimulant in agricultural regions. The long‐term use of GA3 can cause organs damage. Chrysin is a flavonoid found in nature that is commonly used to treat organ toxicity. In this study, we examined the effect of chrysin on the testes function of GA3‐affected rats. A total of 24 male Wistar rats were divided into 4 groups. Saline was given to the control group. The chrysin group was given orally 50 mg/kg/BW of chrysin in saline. The GA3 group received a daily oral gavage of GA3 (55 mg/kg/BW). The protective group (chrysin + GA3) was given chrysin and GA3 as those described in chrysin and GA3 groups. There were an increase in MDA levels in the serum and testicular tissue of GA3‐treated group. Catalase, GSH, and SOD levels were all lowered in the GA3‐treated rats. Chrysin dramatically reduced the harmful effects of GA3 by restoring reproductive hormone levels, altered sperm parameters, and antioxidant capabilities. Furthermore, GA3 reduced the quantitative expression of steroidogenesis genes StAR and 3‐HSD, as well as Bcl2 genes, while it increased the apoptotic marker BAX; all were alleviated by the pre‐administration of chrysin. The pre‐administration of chrysin protected the GA3 group from spermatogenic vacuolation, interstitial edema, necrosis, and depletion. Chrysin inhibited oxidative stress and modulated antioxidant activity, as well as apoptosis‐/anti‐apoptosis‐related mediators in the testes. Chrysin has the potential to repair GA3‐induced testicular dysfunctions. This suggests that chrysin is preferable as a medication to mitigate GA3‐induced oxidative damage in the testes. Thyme (Thymus vulgaris) is an herbal plant with pleiotropic medicinal properties. In this study, we examined the possible protective effect of an ethanolic extract of thyme leaves against the renal oxidative stress induced by sodium nitrite (NaNO2). Male Swiss mice received either saline or thyme extract for 15 days (0.5 g/kg body weight, orally). NaNO2 (60 mg/kg) was injected intraperitoneally at Day 14. The protective group received the thyme extract for 15 days and NaNO2 on Day 14. Blood and kidney samples were taken from all groups to measure serum urea, blood urea nitrogen (BUN), creatinine, serum, tissue antioxidant activity, and the inflammatory cytokines IL‐1β and IL‐6. Quantitative real‐time PCR (qRT‐PCR) was used to examine the expression of kidney injury marker‐1 (Kim‐1), TNF‐α, nuclear factor erythroid‐2 related factor 2 (Nrf2), and hemoxygenase‐1 (HO‐1), all of which are associated with kidney redox and oxidative stress. Pretreatment with thyme extract reduced the effects of NaNO2 on urea, BUN, and creatinine, and reversed its effect on tissue and serum antioxidants. NaNO2‐induced nephritis as demonstrated by the upregulation in mRNA expression of Kim‐1 and TNF‐α, which was, however, recovered and protected by pretreatment with thyme extract. Expression of Nrf2 and HO‐1 was upregulated by treatment with thyme extract and downregulated by NaNO2 intoxication. NaNO2‐induced congestion in glomeruli and dilatation of the renal tubules, conditions that were restored in the group pretreated with thyme extract. NaNO2 upregulated Bax immunoreactivity and caused apoptosis in renal structures. Thus, thyme extract is effective in managing the renal toxicity associated with oxidative stress and renal redox. Practical applications: The results from this study have shown that use of thyme extract may promote better health due to its high antioxidant activity. For instance, it could be ingested to alleviate the symptoms of renal inflammation and oxidative stress associated with nitrite toxicity. In agriculture, gibberellic acid (GA3) is commonly used with extreme dangers for public health. The current research evaluates the improving effects of n‐acetyl cysteine (NAC, 150 mg/kg bw) co‐administered with GA3 (55 mg/kg bw) mediated testicular injury. Twenty‐four male albino rats were split into 4 groups: Negative control (CNT), NAC group, positive GA3 group and protective group, co‐administered NAC plus GA3. On day 21, rats were anesthetised then euthanised by decapitation. Blood samples were collected; testicular samples were taken for semen analysis, serum chemistry, RNA extraction, histological and antioxidants markers examination. Our results revealed a significant decline p <.05 of catalase level and total antioxidant capacity. There was a substantial rise of MDA concentration in GA3‐treated rats along with a considerable decrease of the antioxidant markers (SOD, GSH) and serum male reproductive hormones. In GA3‐treated rats, an overexpression of the inflammatory cytokines (TNF‐α, IL‐1β) and anti‐inflammatory cytokine IL‐10 with boost mRNA expression of nuclear factor‐kappa (NFkB) were confirmed. There was downregulation of steroidogenesis genes and decrease in sperm quality and concentration with an increase in sperm abnormalities, all were reported in GA3‐treated rats. NAC treatment significantly increased the antioxidant state, testicular function beside structural germ cell and seminiferous tubules histology accompanied by upsurge of steroidogenic mRNA expressions (P450scc and 3β‐HSD) and downregulated the pro‐inflammatory cytokines mRNA expression (TNF‐α, IL‐1β). These results confirm the antioxidant capability of NAC and afford robust evidence about the ameliorative effect of the NAC to attenuate the testicular injury induced by GA3 through modulation of the antioxidant defence system, steroidogenic and pro‐inflammatory cytokines mRNA expression. The extensive usage of gibberellic acid (GA3) in agriculture and plant growth is generally associated with enormous human and public health hazards. The present research assesses the impact of n-acetyl cysteine (NAC) on the hepatorenal injury persuaded by GA3 for this purpose, After two weeks of adaptation twenty-four rats allocated into four groups (6 rats/group) as follows: control group, supplied with saline only; n-acetyl cysteine (NAC) group, provided with 150 mg/kg/bw by stomach tube (orally) dissolved in saline; Positive GA3 group, received GA3 (55 mg/kg/bw) orally; Protective group received NAC (150 mg/kg/bw) and GA3 (55 mg/kg/bw) as in NAC and GA3 groups. Rats received their treatments for consecutive 3 weeks. On day 22, rats were anesthetized, then euthanized. Blood and tissue samples were obtained for biochemical, antioxidants markers analysis, gene expression, and histopathological examination. Our results revealed significant changes in serum AST, ALT, urea, uric acid, total protein, and albumin levels with a substantial rise of MDA and NO concentration in GA3 treated rats along with a considerable decrease of the GSH and overexpression of the inflammatory hepatic and renal cytokines (IL-10, TNF-α, NOS) and fibrotic gene expression TGF-β1, and α-SMA, with boost expression of nuclear factor-kappa (NFkB). NAC co-administered with GA3 significantly normalized the kidney and liver function and the antioxidant state, besides normal histological structure of both liver and kidney tissue and downregulated expression of the pro-inflammatory cytokines as well as, fibrogenic gene expression. Practical applications: The current study confirmed that GA3 induced hepto-renal dysfunction that was ameliorated by NAC administration. Moreover, our findings confirmed the antioxidant capability of n-acetyl cysteine and afford robust evidence about the ameliorative effect of the n-acetyl cysteine to attenuate the hepatorenal injury induced by gibberellic acid through modulation of the antioxidant defense system fibrogenic, and pro-inflammatory cytokines expression. The herb thyme (Thymus vulgaris) has multiple therapeutic uses. In this study, we explored how T. vulgaris leaf extract protects liver cells against sodium nitrite-(NaNO2) induced oxidative stress. Mice were divided into four groups; each group received one of the following treatments orally: saline; T. vulgaris extract alone; NaNO2 alone; or T. vulgaris extract + NaNO2. Alanine aminotransferase (ALT), aspartate aminotransferase (AST), reduced glutathione (GSH), superoxide dismutase (SOD), malondialdehyde (MDA), IL-1β, IL-6, TNF-α, and total proteins were measured in serum using standard methods. TNF-α, hemooxygenase-1 (HO-1), thioredoxin, SOD, and GSH synthase, all of which are linked to oxidative stress, were measured using quantitative real-time PCR (qRT-PCR). In mice treated with T. vulgaris extract, the effect of NaNO2 on ALT and AST levels and total proteins was reduced, and its effect on antioxidant levels was reversed. Normally, NaNO2 causes hepatocyte congestion and severe hepatic central vein congestion. Tissues in the mice treated with T. vulgaris were restored to normal conditions. Our results demonstrate that NaNO2-induced hepatic injury is significantly reduced by pretreatment with T. vulgaris extract, which protects against hepatic oxidative stress and its associated genes at the biochemical, molecular, and cellular levels. Hyperuricemia is an abnormal metabolic condition characterized by an increase in uric acid levels in the blood. It is the cause of gout, manifested by inflammatory arthritis, pain and disability. This study examined the possible ameliorative impacts of parsley (PAR) and celery (CEL) as hypouricemic agents at biochemical, molecular and cellular levels. PAR and CEL alone or in combination were orally administered to hyperuricemic (HU) mice and control mice for 10 consecutive days. Serum levels of uric acid and blood urea nitrogen (BUN), xanthine oxidase activity, antioxidants, inflammatory (IL-1β and TNF-α) and anti-inflammatory cytokines (IL-10) were measured. mRNA expression of urate transporters and uric acid excretion genes in renal tissues were examined using qRT-PCR (quantitative real time PCR). Normal histology and immunoreactivity of transforming growth factor-beta 1 (TGF-β1) in kidneys was examined. Administration of PAR and CEL significantly reduced serum BUN and uric acids in HU mice, ameliorated changes in malondialdehyde, catalase, and reduced glutathione, glutathione peroxidase (GPX), IL-1β, TNF-α and IL-10 in hyperuricemic mice. Both effectively normalized the alterations in mURAT-1, mGLUT-9, mOAT-1 and mOAT-3 expression, as well as changes in TGF-β1 immunoreactivity. Interestingly, combined administration of PAR and CEL mitigated all examined measurements synergistically, and improved renal dysfunction in the hyperuricemic mice. Objective. The current study was aimed to examine the possible ameliorative impacts of MO leaf extract (MOLE) against MTX-induced alterations on oxidative stress of mouse spleen and explore the possible molecular mechanism that controls such impacts. Methods. Adult male mice were allocated into 4 groups: control, Moringa oleifera leaf extract (MOLE), MTX, and MOLE plus MTX. Mice received MOLE orally for a week before MTX injection and continued for 12 days. Serum and spleen were sampled for biochemical and quantitative gene expressions. Results. As compared with the MTX-injected group, MOLE effectively reduced the changes in total proteins, spleen MDA, SOD and catalase activities, and changes in serum antioxidants levels. Moreover, there is downregulation of antioxidant genes (SOD and catalase) and antiapoptotic genes (XIAP and Bcl-xl) along with upregulation in Bax and caspase-3 mRNA (apoptotic genes) in the MTX-injected group. MTX induced changes in IL-1β, IL-6, TNF-α, and IL-10 expression. MOLE restored and ameliorated the changes induced in biochemical, antioxidants, apoptosis, and apoptosis associated genes that were induced by MTX intoxication. Conclusion. The aim of the current study was to examine the effects of chronic consumption of soft drinks (SDs) on hepatic oxidative stress and cytochrome P450 enzymes (CYPs) expression in the livers of Wistar rats. For 3 consecutive months, the rats had free access to three different soft drinks, Coca-Cola, Pepsi-Cola and 7-UP. The rats were subsequently compared with control group rats that had consumed water. Blood and hepatic tissue samples were assayed for the changes in antioxidants, liver function biomarkers and hepatic gene expression for different isoforms of hepatic CYP. The results indicated that SD consumption (SDC) decreased serum antioxidant levels and increased malondialdehyde secretion, and increased liver biomarkers (glutamate pyruvate transaminase and glutamate oxaloacetate). SD induced alterations in mRNA expression of hepatic antioxidants and cytochrome isoforms. The expression of peroxidase, catalase, CYP1A2, CYP3A2 and CYP2C11 in the liver were upregulated following SDC. By contrast, CYP2B1 was downregulated after 3 months of SDC in liver tissue samples. Thus, the present findings indicate that SDs induced oxidative stress in the liver of Wistar rats and for the first time, to the best of our knowledge, indicate that SDC disrupts hepatic CYP enzymes that may affect drug metabolism. The present study was performed to examine the effects of chronic soft drink consumption (SDC) on oxidative stress, biochemical alterations, gene biomarkers and histopathology of bone, liver and kidney. Free drinking water of adult male Wistar rats was substituted with three different soft drinks: Coca-Cola, Pepsi and 7-Up, for three consecutive months. The serum and organs were collected for examining the biochemical parameters associated with bone, liver and kidney functions. Semi-quantitative reverse transcription polymerase chain reaction was used to observe the changes in the expression of genes in the liver and kidney, which are associated with oxidative stress resistance. Histopathological investigations were performed to determine the changes in bone, liver and kidney tissues using hematoxylin and eosin stains. SDC affected liver, kidney and bone function biomarkers. Soft drinks increased oxidative stress, which is represented by an increase in malondialdehyde and a decrease in antioxidant levels. SDC affected serum mineral levels, particularly calcium and phosphorus. Soft drinks downregulated the expression levels of glutathione-S-transferase and super oxide dismutase in the liver compared with that of control rats. Rats administered Coca-Cola exhibited a hepatic decrease in the mRNA expression of a2-macroglobulin compared with rats administered Pepsi and 7-Up. On the other hand, SDC increased the mRNA expression of a1-acid glycoprotein. The present renal studies revealed that Coca-Cola increased the mRNA expression levels of desmin, angiotensinogen and angiotensinogen receptor compared with the other groups, together with mild congestion in renal histopathology. Deleterious histopathological changes were reported predominantly in the bone and liver of the Coca-Cola and Pepsi groups. In conclusion, a very strict caution must be considered with SDC due to the increase in oxidative stress biomarkers and disruption in the expression of certain genes associated with the bio-vital function of both the liver and kidney. The present study was conducted to test the effect of Origanum Majoranum Extract (OME) of leaves on alterations induced in a model of type 2 diabetic rats. Adult male Wistar rats were fed high fat diet for 3 weeks and injected a single dose of streptozotocin (35 mg/kg) intraperitoneally to induce type 2 diabetic rats. Diabetic rats were given aqueous extract of OME in a dose of 20 mg/kg orally for 3 weeks. Changes in lipid profiles, glucose, insulin, expression of some genes related to glucose metabolism and histopathological changes in liver and kidney were examined. Administration of OME improved and normalized dyslipidemia recorded in type 2 diabetic rats together with reduction in glucose and insulin levels. OME induced up-regulation in gene expression of glucose [adiponectin and glucose transporter-2 (GLUT-2)] and lipid metabolism [lipoprotein lipase (LPL)]. Moreover, OME normalized histopathological changes occurred in liver and kidney of diabetic rats. OME decreased lipids accumulation in liver and kidney and increased regeneration of hepatic parenchyma and restored normal renal architecture with disappearance of fat droplets. Type 2 diabetes mellitus (T2DM) is a serious health issue worldwide. The disease is characterized by insulin resistance (IR), which leads to dyslipidemia and alterations in the expression levels of a number of genes. Metformin is the standard treatment for T2DM; however, the exact mechanism underlying metformin regulation is not fully understood. The aim of the present study was to investigate the effects of metformin on serum lipid profiles and the expression levels of various genes that are associated with IR, as well as the histopathological changes in the liver and pancreas. A T2DM rat model was established by feeding the rats a high-fat diet for 4 weeks, combined with a dose of streptozotocin (35 mg/kg body weight). Following the successful induction of T2DM, metformin was administered orally (400 mg/kg/day) for 4 weeks. The results indicated that metformin improved the symptoms of IR by normalizing the serum lipid profiles in the diabetic rats. Furthermore, metformin upregulated the expression of insulin receptors and genes associated with lipid metabolism, including acyl-CoA oxidase, carnitine palmitoyl transferase-1 and peroxisome proliferator activated receptor-α. In addition, treatment with metformin downregulated the expression levels of fetuin-A and retinol binding protein-4 (RBP-4), while normalizing the expression of perilipin that had been reduced in the T2DM rats. Metformin administration induced regenerative changes in the hepatocyte cytoplasm and parenchyma. In the pancreas, treatment with metformin was shown to induce positive signaling for insulin and the regeneration of pancreatic β cells. In summary, metformin treatment ameliorated a number of the harmful effects associated with T2DM via the modulation of the expression levels of fetuin-A, RBP-4, perilipin and various genes associated with lipid metabolism, resulting in regenerative changes in the liver and pancreatic cells. Haptoglobin (Hp) is a protein secreted mainly from the liver to bind hemoglobin and thus prevents iron loss and oxidative stress. Bovine Hp in blood increases during diseases such as mastitis and fatty liver syndrome. This study was conducted to examine the expression and regulation of Hp in differentiated mature bovine adipocytes. Hp expression was confirmed in various tissues and in mature adipocytes but not in the stromal vascular cell fraction using qualitative RT -PCR and quantitative northern blot analysis, respectively. Intravenous injection of Holstein cows with recombinant bovine tumor necrosis factor-α (rbTNF-α) and interferon gamma (rbIFN-γ) significantly increased the serum concentrations of Hp. When mature differentiated bovine adipocytes were incubated with short chain fatty acids, cytokines and hormones, Hp expression was altered. Acetate, butyrate and propionate at a dose of 1 mM significantly up-regulated Hp mRNA abundance. Lipopolysaccharide (LPS) and TNF-α also significantly up-regulated Hp mRNA abundance, while troglitazone and norepinephrine down-regulated it. Our study confirmed expression of Hp in differentiated mature adipocytes and identified LPS, TNF-α, short chain fatty acids, as regulators of Hp mRNA expression in these cells. Zinc oxide nanoparticles (ZnONPs), characterized by their nanoscale dimensions and unique properties, have emerged as promising materials in various applications. From electronics to biomedicine, their versatility and biocompatibility makes them valuable assets to address multifaceted challenges. This study aimed to comparatively analyze chemically synthesized ZnONPs (CS-ZnONPs) and Illicium verum-mediated ZnONPs (IV-ZnONPs) in terms of characteristics and biological activities. Characterization of both types of synthesized ZnONPs were done by using various techniques namely XRD, UV-vis spectroscopy, FTIR, TEM, and EDX. TEM and XRD results revealed the variability in size and shape of IV-ZnONPs (∼13.75 nm; triangular) and CS-ZnONPs (∼38.9 nm; flower-shaped). FTIR spectra showed the presence of different phytochemicals present in I. verum extract involved in the capping of IV-ZnONPs. Both types of ZnONPs were found to be biocompatible in nature. Furthermore, variations in their biological properties were observed using different in vitro assays, such as FRAP (242.3 ± 18.9 and 101.5 ± 9.5 μM TEAC), CUPRAC (249.8 ± 8.9 and 149.5 ± 8.2 μM TEAC), ABTS (I36.2 ± 0.5 and 28.2 ± 0.8 μM TEAC), and ORAC cell-free assays (19.9 ± 0.6 and 11.9 ± 0.9 μM TEAC) for anti-oxidant investigations of IV-ZnONPs and CS-ZnONPs, respectively. Percentage Inhibition of Vespelysine-like AGEs and pentosidine-like AGEs for anti-aging analysis were reported. In addition to these, the anti-cancerous potential of both types of ZnONPs was also studied based on MTT assay, ROS/NOS production, mitochondria membrane potential, activity of caspase 3/7 measurements, and expression of caspase-3 gene in mammalian HepG2 cell lines. Our findings revealed that both types of ZnONPs were found to be more active through electron-transfer anti-oxidant mechanism. Likewise, the findings of anti-aging analyses revealed significant difference of inhibitory potential between CS-ZnONPs and IV-ZnONPs. As far as anti-cancerous potential is concerned, both IV-ZnONPs and CS-ZnONPs were found to have promising potential against HepG2 cells. However, CS-ZnONPs were observed to have a slight edge over IV-ZnONPs. Nonetheless, the overall results indicated that ZnONPs synthesized through the green route exhibit outstanding biological potential, positioning them as promising candidates for future biomedical applications, particularly in the realm of cancer therapeutics. Group 1 was a control group that got orally supplied corn oil (vehicle). Group 2 was intoxicated by malathion dissolved in corn oil orally at 135 mg/kg/day. Group 3 orally received both malathion + Panax Ginseng® (300 mg/kg/day). Group 4 was orally given Panax Ginseng® at a 300 mg/kg/day dose. Treatments were administered daily and continued for up to 30 consecutive days. Malathion's toxic effect on both hepatic and renal tissues was revealed by a considerable loss in body weight and biochemically by a marked increase in liver enzymes, LDH, ACP, cholesterol, and functional renal markers with a marked decrease in serum TP, albumin, and TG levels with decreased AchE and Paraoxonase activity. Additionally, malondialdehydes, nitric oxide (nitrite), 8-hydroxy-2-deoxyguanosine, and TNFα with a significant drop in the antioxidant activities were reported in the malathion group. Malathion upregulated the inflammatory cytokines and apoptotic genes, while Nrf2, Bcl2, and HO-1 were downregulated. Ginseng® and malathion co-treatment reduced malathion's harmful effects by restoring metabolic indicators, enhancing antioxidant pursuit, lowering the inflammatory reaction, and alleviating pathological alterations. Moringa Oleifera (MO) is a herbal plant native to South Asia known for its anti-oxidative and anti-inflammatory properties. This study explored the protective effects of MO leaf extract (MOLE) against oxidative stress and hepatic and renal injuries caused by methotrexate (MTX) therapy. Mice received a single intraperitoneal injection of 20 mg/kg body weight MTX to induce hepatic and kidney injuries. They then received 300 mg/kg body weight of MOLE orally for seven days, followed by MTX on day 7 then five more days of MOLE (12 days total). Blood, liver and kidney samples were collected from all groups and the following biochemical parameters were tested: serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), catalase, superoxide dismutase (SOD), malondialdehyde (MDA) and total proteins. Quantitative real time PCR (qRT-PCR) was used to examine Nrf2, HO-1, BAX, TIMP, XIAP, and NFkB, which are associated with apoptosis, anti-apoptosis and oxidative stress. Capase-9 and Bcl2 genes underwent immunohistochemical analysis. Pretreatment with MOLE reduced the effect of MTX on ALT, AST and total proteins, and reversed its effect on serum and tissue antioxidants. Nrf2/HO-1, apoptotic and anti-apoptotic gene expression was regulated, and Bax and TIMP were reduced; XIAP expression was increased in both the liver and kidney samples, and immunoreactivity of caspase-9 and Bcl2 was restored in the MOLE-administered experimental group. Overall, the study concluded that MOLE can inhibit the effects of hepato-renal injuries caused by MTX by regulating oxidative stress, apoptosis and anti-apoptotic genes at biochemical, molecular and cellular levels. Hydroxyurea (HDU) is a widely used medication for various malignancies, thalassemia, and sickle cell anemia with reported side effects. The current study investigated HDU- induced hepatic injury and the protective potential of the royal jelly (RJ) against this hepatotoxic effect in the light of hepatic oxidative/ antioxidative status, pro-inflammatory cytokine, apoptosis signaling pathway, and histopathology. Sixty albino rats were used (n = 10/group) for 60 days: control, RJ (100 mg/kg body weight, orally), HDU (225 mg/kg body weight, orally), 2HDU (450 mg/kg body weight, orally), and HDU + RJ groups. HDU-treated rats showed significant elevation of liver function tests as aspartate aminotransferase, alanine aminotransferase, and alkaline phosphatase, as well as malondialdehyde and nitric oxide (oxidative biomarkers) and significant decreased hepatic antioxidant molecules (reduced glutathione, superoxide dismutase, and glutathione peroxidase), compared to a control group, that more pronounced in the high dose of HDU. In addition, HDU induced significant upregulation of TNF-α and the Caspase-3 apoptotic pathway. Moreover, the liver of HDU treated groups showed various hepatic lesions from mild to severe necrotic changes related to the HDU dose. However, administration of RJ with HDU improved liver function tests, liver histology, and hepatic oxidative/antioxidative status concerning HDU groups. Furthermore, oral RJ administration with HDU significantly lessens the immune-expression area % of TNF-α and Caspase-3. Thus, the royal jelly has antioxidant, anti-inflammatory, and anti-apoptotic properties against HDU- induced hepatic injury and could be, therefore, used as adjuvant therapy in patients with long-term HDU medication. Simple Summary: In modern aquaculture, fish have been subjected to intensive stressful conditions, which threaten their growth rate and increase their susceptibility to bacterial diseases. Innovative and sustainable production strategies in aquaculture have encouraged the utilization of plant-derived bioactive molecules (phytogenics) in fish diets, especially when they were used in a unique blend offering enumerable growth and health benefits. Therefore, our research was conducted to uncover the potential nutritional and immunological impacts of thymol (Thy) and/or thymoquinone (ThQ) with a disease shielding effect against Aeromonas sobria. Herein, the integration of dietary Thy and ThQ provided an efficient way to improve fish growth-related parameters and muscle antioxidant capacity. Moreover, we presented evidence for the unique immune-stimulating role of a Thy and ThQ combination with a successful forefront defense against A. sobria experimental infection in Nile tilapia. Our key findings motived the application of a dietary blend comprising Thy and ThQ as a functional feed that fights the challenges facing the aquaculture industry with maximized fish productivity. Plant-derived bioactive compounds with promising nutritional and therapeutic attributes (phytogenics) are among the top priorities in the aquaculture sector. Therefore, the impact of thymol (Thy) and/or thymoquinone (ThQ) on the growth, immune response antioxidant capacity, and Aeromonas sobria (A. sobria) resistance of Nile tilapia was investigated. Four fish groups were fed a control diet and three basal diets supplemented with 200 mg/kg diet of Thy or ThQ and a blend of both Thy and ThQ at a level of 200 mg/kg diet each. At the end of the feeding trial (12 weeks), the tilapias were challenged intraperitoneally with virulent A. sobria (2.5 × 108 CFU/mL) harboring aerolysin (aero) and hemolysin (hly) genes. The results revealed that tilapias fed diets fortified with a combination of Thy and ThQ displayed significantly enhanced growth rate and feed conversion ratio. Notably, the expression of the genes encoding digestive enzymes (pepsinogen, chymotrypsinogen, α-amylase and lipase) and muscle and intestinal antioxidant enzymes (glutathione peroxidase, catalase and superoxide dismutase) was significantly upregulated in Thy/ThQ-fed fish. An excessive inflammatory response was subsided more prominently in the group administrated Thy/ThQ as supported by the downregulation of il-β, il-6 and il-8 genes and in contrast, the upregulation of the anti-inflammatory il-10 gene. Remarkably, dietary inclusion of Thy/ThQ augmented the expression of autophagy-related genes, whilst it downregulated that of mtor gene improving the autophagy process. Furthermore, Thy/ThQ protective effect against A. sobria was evidenced via downregulating the expression of its aero and hly virulence genes with higher fish survival rates. Overall, the current study encouraged the inclusion of Thy/ThQ in fish diets to boost their growth rates, promote digestive and antioxidant genes expression, improve their immune responses and provide defense against A. sorbia infections with great economic benefits. Wheat germ oil (WGO) is a well-known product with anti-inflammatory and antioxidant properties. The current study aimed to investigate the impacts of WGO against ethanol-induced liver and kidney dysfunction at the serum, anti-inflammatory, antioxidants and anti-apoptotic signaling pathways. Rats received saline orally as a negative control or WGO in a dose of 1.5 mL/kg (1400 mg/kg body weight orally) for 15 days. The affected group received ethanol 50% v/v 10 mL/kg (5 g/kg) body weight orally once a day for consecutive 15 days to induce hepatorenal injuries in ethanolic non-treated group. The protective group received WGO daily 1 h before ethanol administration. Serum (1.5 mL) from blood was extracted and examined for the changes in biochemical assessments in serum alkaline phosphatase (ALP), alanine aminotransferase (ALT), bilirubin, serum γ-glutamyl transpeptidase (GGT), total protein, serum albumin, butyrylcholinesterase (BChE), total cholesterol (TC), total triglyceride (TG), urea, creatinine, uric acid, potassium (K+), Beta-2 microglobulin (β2M), malondialdehyde (MDA), catalase (CAT), reduced glutathione (GSH), superoxide dismutase (SOD) and aspartate aminotransferase (AST). Kidney and liver homogenate was used to measure MDA, GSH and catalase activities. Quantitative real time PCR (qRT-PCR) was used to express Nrf2 and HO-1 in liver, and NF-kB and kidney injury molecule (KIM-1) in kidneys, which are correlated with oxidative stress and inflammation. Capase-3 and Bcl2 genes were examined using immunohistochemical analysis in the kidney and liver. Ethanol administration induced significant alteration in examined liver and kidney markers (AST, ALT, GGT, ALP, total proteins, urea, creatinine and uric acid). Moreover, alcohol administration decreased antioxidant activities at serum and hepatorenal tissues (GSH, catalase and SOD), while MDA was increased as a tissue degradation marker. Inflammatory cytokines, together with genes of oxidative stress markers (Nrf2 and HO-1), were all affected. At cellular levels, apoptotic marker caspase-3 was upregulated, while antiapoptotic marker B-cell lymphoma 2 (Bcl2), was down regulated using immunohistochemical analysis. Of interest, pretreatment with WGO improved the side effects induced by ethanol on hepatic, renal biomarkers and reversed its impact on serum and tissue antioxidant parameters. Nrf2/HO-1 were upregulated, while NFk-B and KIM-1 were downregulated using real time PCR. Immune reactivities of caspase-3 and Bcl2 genes were restored in the protective group. In conclusion, WGO ameliorated ethanol-induced hepatic and renal dysfunction at the biochemical, molecular and cellular levels by regulating some mechanisms that controls oxidative stress, apoptosis, inflammation and anti-apoptotic pathways. Abnormal uric acid level result in the development of hyperuricemia and hallmark of various diseases, including renal injury, gout, cardiovascular disorders, and non‐alcoholic fatty liver. This study was designed to explore the anti‐inflammatory potential of stevia residue extract (STR) against hyperuricemia‐associated renal injury in mice. The results revealed that STR at dosages of 150 and 300 mg/kg bw and allopurinol markedly modulated serum uric acid, blood urea nitrogen, and creatinine in hyperuricemic mice. Serum and renal cytokine levels (IL‐18, IL‐6, IL‐1Β, and TNF‐α) were also restored by STR treatments. Furthermore, mRNA and immunohistochemistry (IHC) analysis revealed that STR ameliorates UA (uric acid)‐associated renal inflammation, fibrosis, and EMT (epithelial‐mesenchymal transition) via MMPS (matrix metalloproteinases), inhibiting NF‐κB/NLRP3 activation by the AMPK/SIRT1 pathway and modulating the JAK2‐STAT3 and Nrf2 signaling pathways. In summary, the present study provided experimental evidence that STR is an ideal candidate for the treatment of hyperuricemia‐mediated renal inflammation. Practical applications: The higher uric acid results in hyperuricemia and gout. The available options for the treatment of hyperuricemia and gout are the use of allopurinol, and colchicine drugs, etc. These drugs possess several undesirable side effect. The polyphenolic compounds are abundantly present in plants, for example, stevia residue extract (STR) exert a positive effect on human health. This work aimed to identify novel angiotensin‐converting‐enzyme (ACE) inhibitory peptides from Macadamia integrifolia antimicrobial protein 2 (MiAMP2). The MiAMP2 protein was hydrolyzed through in silico digestion, and the generated peptides were screened for ACE inhibitory activity. The in silico enzyme digestion results revealed that 18 unreported peptides were obtained using AHTPDB and BIOPEP‐UWM, and none were thought to be toxic based on absorption, distribution, metabolism, and excretion (ADMET) prediction. PGPR, RPLY, MNPQR, and AAPR were predicted to exhibit good biological activity. The molecular docking results revealed that the four peptides tightly bound to the active pocket of ACE via hydrogen bonds and hydrophobic interactions, among which RPLY and MNPQR bound to ACE more strongly. The in vitro assay results confirmed that RPLY and MNPQR peptides inhibited ACE via competitive manner. These results provide theoretical guidance for the development of novel foodborne antihypertensive peptides from Macadamia nut proteins. The present study aimed to investigate the molecular mechanism underlying the hepatoprotective effects of pomegranate (POM) against oxidative stress in a rat model of carbon tetrachloride (CCl4)-induced liver damage. Injection of rats with CCl4 resulted in hepatic inflammation and lipid accumulation via the upregulation of interleukin (IL)-6 and sterol regulatory element-binding protein 1c (SREBP-1c) mRNA expression. CCl4 induced downregulation of the anti-inflammatory factors alpha 2-macroglobulin (α-2M) and IL-10 in comparison with the POM treated group. In addition, CCl4 induced downregulation of superoxide dismutase (SOD), glutathione S-transferase (GST) and catalase (CAT) expression. Conversely, prior administration of POM counteracted the deleterious alterations induced by CCl4. POM downregulated CCl4-induced IL-6 upregulation, normalized the increase in SREBP-1c expression, and prevented CCl4-induced α-2M downregulation. POM counteracted CCL4-induced alterations via immunosuppressive, anti-inflammatory and regenerative effects by upregulating transforming growth factor-β1, HSP70 and IL-10 mRNA expression. In addition, POM increased reactive oxygen species scavenging activity by augmenting the antioxidant defense mechanism against CCl4 hepatotoxicity, as demonstrated by detecting SOD, CAT and GST expression. The limit of tolerance to salinity in freshwater fish has become an important environmental factor influencing growth performance. Therefore, this study aimed to explore the impact of different levels of salinity on behavioural responses, growth performance, haematological, oxidative and biochemical stress parameters, in addition to immune parameters of African catfish. Three‐hundred and Sixty juvenile African catfish were randomly distributed into six groups and exposed to different salinity levels (0, 4, 8, 12, 16 and 20 ppt) with three replicate tanks per treatment group; each tank contained 20 individuals for 42 days. Behavioural responses of fish were monitored during the experimental period. Growth performance, haematological, oxidative and biochemical stress parameters, in addition to immune parameters and osmotic pressure, were also determined at the end of the treatment period. Results revealed that the proportion of fish performing abnormal stress behavioural responses increased directly to the salinity level. In contrast, the survival rate and growth performance were decreased with high feed conversion at the higher salinities. Inconsistent, the haematological, oxidative, biochemical, immune and osmotic pressure parameters were altered gradually with the increase in the salinity level. The study was conducted to assess the long-term effects of predominant land uses on physicochemical properties, nutrient status and their interactions in soils of south-western Punjab representing the semi-arid soils of India. From each site, soil samples of three predominant land use viz. croplands, horticultural lands and uncultivated lands were collected from 0–15, 15–30, 30–60 and 60–90 cm depths. Soils of both croplands and horticultural lands were classified as sandy loam whereas uncultivated lands showed loamy sand texture with relatively higher pH, electrical conductivity (EC) and bulk density (Bd). Greater soil organic carbon (SOC), available nitrogen (N), phosphorus (P) and micronutrients (Zn, Cu, Fe and Mn) in horticulture might be due to the higher addition of OC and mineral nutrients through the decomposition of leaf litterfall and root deposits over their removal from soils while long-term use of potassic fertilizer raised the available K contents in croplands. Profile study up to 90 cm depicted the largest sequestration of 74.89 Mg C ha−1 under orchards which was 40 and 70% higher than croplands and uncultivated lands respectively. Significant variability in water-stable aggregates (WSA) (R2 = 0.5843, p < 0.05) and mean weighted diameter (MWD) (R2 = 0.6497, p < 0.01) with SOC indicated better soil stability in horticulture due to the presence of higher SOC. Positive relations of soil available micronutrients with SOC and finer soil particles were supported by the results of correlation, Principal component analysis and dendrogram indicating horticulture as a potent source of available micronutrients. An overall superiority of horticultural land use over the other two land uses in terms of nutrient status and soil stability suggests its inclusion as a positive strategy that could be taken into account in policymaking for maintaining productivity along with the sustainability of the concerned land degradation prone area. This study aimed to investigate the oxidative neurotoxicity induced by silver nanoparticles (AgNPs) and assess the neuroprotective effects of quercetin against this toxicity. Forty adult male rats were divided into four equal groups: control, AgNPs (50 mg/kg intraperitoneally), quercetin (50 mg/kg orally), and quercetin + AgNPs. After 30 days, blood and brain tissue samples were collected for further studies. AgNP exposure increased lipid peroxidation and decreased glutathione peroxidase, catalase, and superoxide dismutase activities in brain tissue. AgNPs decreased serum acetylcholine esterase activity and γ-aminobutyric acid concentrations. AgNPs upregulated tumor necrosis factor-α, interleukin-1β, and Bax transcript levels. AgNPs reduced the transcripts of claudin-5, brain-derived neurotrophic factor, paraoxonase, nuclear factor-erythroid factor 2 (Nrf2), and Bcl-2. Histopathologically, AgNPs caused various degenerative changes and neuronal necrosis associated with glial cell reactions. AgNPs increased the immunohistochemical staining of glial fibrillary acidic protein (GFAP) in the cerebrum and cerebellum. Oral treatment with quercetin efficiently counteracted the opposing effects of AgNPs on brain tissue via modulation of tight junction proteins, Nrf2, and paraoxonase, and its positive mechanism in modulating pro-inflammatory cytokines and the downregulation of GFAP expression, and the apoptotic pathway. The poultry farms need a safe and effective alternative for antibiotics that can counteract the negative impacts of necrotic enteritis (NE), which causes severe mortalities and economic losses. The current study was aimed to examine the influence of antibiotic (Flagymox) and the microbial‐based administration on carcass traits in Clostridium(C.)perfringens‐infected Cobb and Arbor Acres broilers. A total number of 360 Cobb and Arbor Acres broiler chicks (180 numbers per breed) were allocated to four groups; negative control group (without any treatments); positive control group (administration of C. perfringens at the rate of 1 × 109cfu/bird via crop gavage twice daily from day 16 to 18 post‐hatch); C. perfringens challenge plus antibiotic (Flagymox®) group, and Clostridiumperfringens challenge plus microbial‐based treatment (Big‐lactoα®) group. The results indicated that the Flagymox and Big‐lactoα treated Cobb breed group achieved a significant increase in their body weight (BW) than the positive control group at the third week post‐infection. Also, the Arbor Acres breed gained significantly higher weight compared to the Cobb breed at the third week. Total weight gain (TWG) from 0 to the fifth week in the Cobb and Arbor Acres breeds were higher in the groups treated with Flagymox and Big‐lactoα compared to the birds challenged with C. perfringens without any treatment, thus, increasing the total return (TR) in the treated groups. Economic efficiency showed no significant differences (p <.05) between the treatment groups of both the breeds. Although the treatment cost of Flagymox is higher than the microbial‐based treatment (0.86 versus 0.35 LE), there were no mortalities reported in the microbial‐based groups in both the breeds resulting in significantly low losses compared to the Flagymox treated groups. The groups treated with the microbial‐based products in both breeds were superior in dressing percentage (75.16 and 77.06% for Cobb and Arbor Acres, respectively) compared to that of the other groups. In conclusion, microbial‐based therapy improved the growth rate, carcass traits, survival rate, and economic efficiency in necrotic enteritis induced in Cobb and Arbor Acres broilers. Simple Summary: The use of date palm pollen ethanolic extract (DPPE) is a conventional approach in improving the side-effects induced by Doxorubicin (DOX).DPPE mitigated DOX-induced body and heart weight changes and ameliorated DOX-induced elevated cardiac injury markers. In addition, serum cardiac troponin I concentrations (cTnI), troponin T (cTnT), and N-terminal NBP and cytosolic (Ca+2) were amplified by alleviating the inflammatory and oxidative injury markers and decreasing histopathological lesions severity. DPPE decreased DOX-induced heart injuries by mitigating inflammation, fibrosis, and apoptosis through its antioxidant effect. To reduce DOX-induced oxidative stress injuries and other detrimental effects, a combined treatment of DPPE is advocated. Doxorubicin (DOX) has a potent antineoplastic efficacy and is considered a cornerstone of chemotherapy. However, it causes several dose-dependent cardiotoxic results, which has substantially restricted its clinical application. This study was intended to explore the potential ameliorative effect of date palm pollen ethanolic extract (DPPE) against DOX-induced cardiotoxicity and the mechanisms underlying it. Forty male Wistar albino rats were equally allocated into Control (CTR), DPPE (500 mg/kg bw for 4 weeks), DOX (2.5 mg/kg bw, intraperitoneally six times over 2 weeks), and DPPE + DOX-treated groups. Pre-coadministration of DPPE with DOX partially ameliorated DOX-induced cardiotoxicity as DPPE improved DOX-induced body and heart weight changes and mitigated the elevated cardiac injury markers activities of serum aminotransferases, lactate dehydrogenase, creatine kinase, and creatine kinase-cardiac type isoenzyme. Additionally, the concentration of serum cardiac troponin I (cTnI), troponin T (cTnT), N-terminal pro-brain natriuretic peptide (NT-pro BNP), and cytosolic calcium (Ca+2) were amplified. DPPE also alleviated nitrosative status (nitric oxide) in DOX-treated animals, lipid peroxidation and antioxidant molecules as glutathione content, and glutathione peroxidase, catalase, and superoxide dismutase activities and inflammatory markers levels; NF-κB p65, TNF-α, IL-1β, and IL-6. As well, it ameliorated the severity of histopathological lesions, histomorphometric alteration and improved the immune-staining of the pro-fibrotic (TGF-β1), pro-apoptotic (caspase-3 and Bax), and anti-apoptotic (Bcl-2) proteins in cardiac tissues. Collectively, pre-coadministration of DPPE partially mitigated DOX-induced cardiac injuries via its antioxidant, anti-inflammatory, anti-fibrotic, and anti-apoptotic potential. Objective: The aim of this study was to examine the effect of some natural compounds against multidrug-resistant bacteria. Methods: Forty-three bacterial strains were collected. Disc diffusion and minimum inhibitory concentration (MIC) tests were carried out for natural compounds including quercetin, Acacia nilotica, Syzygium aromaticum, and Holothuria atra. Scanning electron microscope analysis and bacterial DNA apoptosis assays were performed. Results: Staphylococcus aureus strains were resistant to imipenim, ampicillin, and penicillin. Most Escherichia coli strains were resistant to amoxicillin, clavulanat, and ampicillin. Finally, tigecycline was effective with Klebsiella pneumoniae and was resistant to all antibiotics. Only S aromaticum had an antibacterial effect on K pneumoniae. Most S aureus strains were sensitive to S aromaticum, A nilotica, and quercetin. All examined natural extracts had no effect on E coli . Holothuria atra had no effect on any of the strains tested. Minimum inhibitory concentration and minimum bactericidal concentration values for examined plants against S aureus were 6.25 to 12, 1.6 to 3.2, and 9.12 to 18.24 mg/mL, respectively. Syzygium aromaticum was active against K pneumoniae with an MIC of 12.5 mg/mL. Scanning electron microscope analysis performed after 24 and 48 hours of incubation showed bacterial strains with distorted shapes and severe cell wall damage. Syzygium aromaticum, quercetin, and A nilotica showed clear fragmentations of S aureus DNA. Juniperus procera , a coniferous tree in the cypress family, is one of the famous medicinal plants traditionally used in the southern area of the Arabian peninsula. This study examined the anti-hyperglycemic action of Juniperus procera extract (JPE) on diabetic rats. Sixty male rats were divided into 6 equal groups: control, control treated with JPE (200 mg/kg), diabetic, diabetic treated with insulin (1 U/kg), diabetic treated with JPE (200 mg/kg), and diabetic treated with both insulin and JPE. Blood and tissue samples were collected for serum chemistry, gene expression, and immunohistochemistry analyses, the results of which revealed hyperglycemia and inflammation following diabetes induction. Administration of JPE alone or with insulin reduced the hyperglycemia reported in diabetic rats by 25 %. The immunohistochemical examination of pancreatic tissues demonstrated a moderate restoration of insulin and NF-κB expression in pancreatic and hepatic tissues. Significant recovery was observed for glutathione-S-transferase (GST), superoxide dismutase (SOD), and glutathione peroxidase (GPx) mRNA expression in the livers of rats treated with JPE. Administration of JPE led to similar amelioration of the mRNA expression of pyruvate kinase (PK) and phosphoenol pyruvate carboxy kinase (PEPCK) in the livers of diabetic rats. In addition, diabetic rats treated with insulin, JPE, or a combination of these agents demonstrated an improvement in the mRNA expression of IRS-1 and IRS-2 in hepatic and pancreatic tissues, reaching levels approaching normal. Levamisole (LEVA) and garlic are prevalent immunomodulators in humans and animals. Therefore, the present study aimed to examine the immunomodulatory effects of LEVA and garlic oil (GO) alone or in combination on the immune response of Wistar rats. A total of 24 male Wistar rats were allocated into four equal groups: Control group, which was given ad libitum access to food and water; and groups 2-4, which were orally administered LEVA [2.5 mg/kg body weight (BW) every 2 days], GO, (5 ml/kg BW daily), or LEVA plus GO, respectively for 4 consecutive weeks. Serum immunoglobulin (Ig)G and IgM levels were measured using a radial immunodiffusion assay. Serum cytokine levels, including interferon (IFN)-γ, interleukin (IL)-5 and tumor necrosis factor (TNF)-α, were measured using enzyme-linked immunosorbent assay kits. Total blood counts were measured automatically using a cell counter. Serum lysozyme enzymatic activity was determined by measuring the diameters of the zones of clearance relative to lysozyme. Immunohistochemical detection of CD4 and CD8 was carried out using the streptavidin-biotinperoxidase method. Furthermore, the mRNA expression levels of IL-4, IL-5 and IL-12 were measured in the leukocytes and thymus gland by semi-quantitative polymerase chain reaction. The results revealed that LEVA increased serum levels of IFN-γ, IL-5 and TNF-α cytokines, whereas co-administration of LEVA and GO decreased the stimulatory action of LEVA alone. LEVA and GO alone increased the serum levels of IgG, IgM and total blood cell counts, and co-administration of GO and LEVA inhibited the effects of LEVA. At the cellular level, in the spleen, LEVA increased immunoreactivity of CD4 and CD8, whereas co-administration of GO with LEVA decreased this strong expression. At the molecular level, in leukocytes, LEVA upregulated the mRNA expression levels of IL-2, IL-4 and IL-5, whereas GO alone downregulated mRNA expression. Co-administration of GO with LEVA inhibited the LEVA-induced upregulation of IL-2, IL-4 and IL-5 mRNA expression. In the thymus, both LEVA and GO upregulated the mRNA expression levels of IL-4 and IL-5, whereas LEVA alone did not affect IL-12 mRNA expression. Co-administration of GO with LEVA inhibited LEVA-induced upregulation of IL-4 and GO-induced upregulation of IL-12 expression, and had an additive upregulatory effect on IL-5 expression. The planning and design of an airport is subject to many difficulties. One of these difficulties stems from uncertainty about the future demands of air traffic evolution. The growth of and change of potential air traffic impact significantly on airport facilities development. Current airport development paradigms seem inadequate to deal with the volatility of air traffic and site conditions. To overcome this dilemma, variables such as the characteristics of airport movement area parts and aircraft, as well as site conditions, have been extensively examined. Then analytical models based on the fleet mix of aircraft type selection and site conditions have been formulated. The main parameters considered in the analytical models are critical design, aircraft, site information and the reference code of the airport. Depending on these parameters and analytical models, a framework for computerised simulation software has been developed. The developed simulation software combines several types of model with different capabilities and features. The objective of these models is to reach an optimum analysis for the planning and design of airport facilities with the greatest degree of flexibility and expansibility. The newly created models are used to determine runway turn pad dimensions, runway exit locations, taxiway fillet and widening dimensions, apron size, visual marking, PAPI sitting distance, firefighting facilities and airspace obstacle limitations for all sizes and types of airport. The improved models can also be used to determine aeroplane reference length and its adjustments as well as the physical characteristics of movement areas. A sample empirical exercise was carried out to verify the benefits obtained from the proposed models. A comparison between the outcomes of this approach and previous methodologies proves the reliability and effectiveness of this technique. Practically, the analysis and calculation result derived from the program models was used to supplement three supporting case studies. In the first case study, the sitting distances of visual aid devices fixed on the runway approaches at five international airports are analysed and assessed. In the second case study, the cumulative usage and landing segments of runway turn-off points was drawn from interactions among the various approach speeds of the different studied aircrafts. The last case study provides the geometrical design charts of taxiway intersection widening portions for different aeroplane groups and manoeuvring systems. The present study aimed to investigate the prevalence, antibiotic susceptibility profiles, and some toxin genes of Panton-Valentine leukocidin (PVL)-positive Staphylococcus aureus (S. aureus) in unpasteurized raw cow's milk collected from retail outlets located at Mansoura, Dakahliya governorate, Egypt. In that context, a total of 700 raw cow's milk samples were investigated for the presence of S. aureus, which was identified in 41.1% (288/700) of the samples. Among the S. aureus isolates, 113 PVL-positive S. aureus were identified and subjected for further analysis. The PVL-positive S. aureus were investigated for the existence of toxin-related genes, including hemolysin (hla), toxic shock syndrome toxin-1 (tst), and enterotoxins (sea, seb, sec, see, seg, sei, and selj). Genotypic resistance of PVL-positive strains was performed for the detection of blaZ and mecA genes. Among the PVL-positive S. aureus, sea, seb, and sec were detected in 44.2, 6.2%, and 0.9%, respectively, while the hla and tst genes were identified in 54.9% and 0.9%, respectively. The blaZ and mecA genes were successfully identified in 84.9 (96/113) and 32.7% (37/113) of the total evaluated S. aureus isolates, respectively. PVL-positive S. aureus displayed a high level of resistance to penicillin, ampicillin, and trimethoprim-sulfamethoxazole. Multidrug resistance (resistant to ≥3 antimicrobial classes) was displayed by all methicillin-resistant S. aureus (MRSA) and 38.2% of methicillin-sensitive S. aureus (MSSA) isolates. The obtained findings are raising the alarm of virulent PVL-positive MRSA clones in retail milk in Egypt, suggesting the requirement for limiting the use of β-lactam drugs in food-producing animals and the importance of implementing strong hygiene procedures in dairy farms and processing plants. The Acrylamide is a toxic compound generated under oxidative stress arising from intracellular ROS production and induced toxicity. It is frequently used in industry and generated through the heating of tobacco and foods high in carbohydrates. The exact mechanism of its toxicity is still unclear. In this study, an extract of the peels of pomegranate (Punica granatum L.), a nutritious and visually appealing fruit with a diverse bioactive profile, was examined for its potential anti-apoptotic, antioxidant, and anti-inflammatory effects. A total of 40 adult male Wistar rats were allocated into four groups of 10 rats each: Group 1 was a negative-control group (CNT) and received normal saline; Group 2 was a positive-control acrylamide group and received acrylamide orally at a dose of 20 mg/kg/bw; in Group 3, the rats were supplemented with pomegranate-peel extract (P.P; 150 mg/kg/bw) orally on a daily basis for 3 weeks, administered simultaneously with the acrylamide treatment described for Group 2; Group 4 was a protective group, and the animals received the pomegranate-peel extract and acrylamide as stated for Groups 2 and 3, with the pomegranate-peel extract (P.P. extract) administered 1 week earlier than the acrylamide. The results indicate that acrylamide exposure increased the serum levels of AST, ALT, creatinine, interleukin-1 beta, and interleukin-6 in an extraordinary manner. In addition, it increased the lipid peroxidation marker malondialdehyde (MDA) and simultaneously weakened antioxidant biomarker activities (SOD, GSH, and catalase) and reduced the levels of interleukin-10. The pomegranate-peel extract was shown to reduce the inflammatory blood markers of interleukin-1 beta and IL-6. Glutathione peroxidase, superoxide dismutase, catalase, and interleukin-10 were all significantly elevated in comparison to the acrylamide-treatment group as a result of the significant reduction in MDA levels induced by the P.P extract. In addition, the pomegranate-peel extract normalized the cyclooxygenase-2 (COX2), transforming growth factor-beta 1 (TGF-β1), and caspase-3 levels, with a significant upregulation of the mRNA expression of heme oxygenase-1 (HO-1), nuclear factor erythroid 2 (Nrf2), and Bcl-2. Therefore, these data reveal that pomegranate peel has anti-inflammatory, antiapoptotic, free-radical-scavenging, and powerful antioxidant activity that protects against acrylamide toxicity. The impact of zinc oxide nanoparticles (ZnO-NPs) on the pathogenesis of coccidiosis in broiler chickens was tested. A total of 160 1-day-old broiler chicks (Ross 308) were randomly allocated into 4 groups (n = 40). Group 1: unchallenged, unmedicated; Group 2: challenged, unmedicated; Group 3: challenged, supplemented with diclazuril (1 ppm); Group 4: challenged, supplemented with ZnO-NPs (20 ppm). Mixed Eimeria species (E. maxima, E. acervulina, E. mivati, and E. tenella) of a commercial coccidial vaccine (FORTEGRA®) were used to perform the coccidial challenge by 15× of its vaccinal dose on the 14th day of age. Diclazuril and ZnO-NPs supplementation in Group 3 and 4, respectively, reduced the mortality rate due to coccidial challenge to 5.8% compared to 11.9% in Group 2. The growth performance was improved by ZnO-NPs in coccidiosis-infected group (p ≤ 0.05) compared to Group 2 and was comparable to that of Group 3 (p ≥ 0.05). The average oocyst count was lower in Groups 3 and 4 (7.8 × 103 and 14.3 × 103, respectively) than in Group 2 (67 × 103 oocysts). Group 3 had a decreased gross lesion score in duodenum and caecum (p ≤ 0.05) as well as jujenum and ileum (p ≥ 0.05) compared to Group 2; while the average lesion scores of all intestinal parts in Group 4 were significantly decreased (p ≤ 0.05). However, diclazuril was superior to ZnO-NPs in reducing caecal lesion score (p ≤ 0.05). Plasma carotenoids levels were increased by diclazuril (p ≥ 0.05) and ZnO-NPs (p ≤ 0.05) supplementation compared to Group 2. Oxidative stress appeared on the fourth week post-challenge (pc) in Group 2 (p ≤ 0.05) compared to Group 1, while the dietary supplementation with either diclazuril or ZnO-NPs numerically decreased Malondialdhyde (p ≥ 0.05) and statistically increased antioxidant activity (p ≤ 0.05). Both medications significantly improved the PCV%, Hb% and RBCs count on the 6th-day and 4th-week pc (p ≤ 0.05) compared to Group 2, though this improvement was higher significantly in Group 4 than Group 3 on the 6th day pc (p ≤ 0.05). Neither coccidial challenge nor medications had an impact on the total WBCs count as well as organ index, except Bursa of fabricious index that significantly improved by ZnO-NPs on the 4th-week pc compared to Group 2. Coccidial challenge reduced total protein and globulin levels and increased the serum alanine aminotransferase, serum cholesterol, and low-density lipoprotein levels (p ≤ 0.05) compared to Group 1, while those of both medicated groups (Group 3 and 4) were comparable to Group 1 (p ≥ 0.05). Simple Summary: Sustainable aquaculture requires natural alternative substances with high potential in enhancing the performance and wellbeing of aquatic animals. In this regard, the present study tested the possibility of using mannan oligosaccharides (MOS) in the diets of grey mullet as functional additives. For 8 weeks, fish were fed with enriched diets containing 0, 0.5, 1, and 2% MOS. The results showed marked improvements in the growth performance, digestive enzyme activity, blood chemistry, and antioxidative capacity. In conclusion, dietary MOS at 0.5–1% is required to enhance the productivity of grey mullet. Mannan oligosaccharide (MOS) is prebiotic with high functionality in aquaculture. The current study investigated the potential roles of MOS on the growth performance, digestive enzyme activity, carcass composition, and blood chemistry of Thinlip grey mullet (Liza ramada). Four tested diets with 34.49% crude protein and 6.29% of total lipids were prepared and fortified with 0, 0.5, 1, and 2% MOS. Fish of initial weight = 5.14 ± 0.11 g/fish were distributed in 12 hapas (0.5 × 0.5 × 1 m) at 15 fish per hapa (triplicates) and fed the test diets to the satiation level two times a day (08:00 and 15:00) for eight weeks. At the end of the trial, all fish were weighed individually for growth performance calculation. Blood was collected to check blood chemistry traits, and intestines were dissected for digestive enzyme analysis. Fish treated with MOS had marked enhancement in the final body weight, feed conversion ratio, protein gain, and protein retention regardless of inclusion dose (p < 0.05). The weight gain, specific growth rate, and protein efficiency ratio were meaningfully enhanced by including MOS at 0.5 and 1%, followed by fish fed with 2% MOS, while the lowest values were in the control group (p < 0.05). Insignificant influences of MOS were seen on the chemical composition of carcass components (moisture, crude protein, total lipids, and ash) (p > 0.05). Fish treated with MOS at 0.5 and 1% had marked enhancement in the amylase, lipase, and protease activities regardless of inclusion dose (p < 0.05). The blood total protein and albumin levels were meaningfully enhanced by including MOS at 0.5 and 1%, followed by fish fed with 2% MOS, while the lowest values were in the control group (p < 0.05). The blood globulin was significantly enhanced in fish fed 1% MOS than fish treated with 0, 0.5, and 2% of MOS (p < 0.05). The blood lysozyme activity was meaningfully enhanced by including MOS at 1%, followed by fish treated with 0.5 and 2%, while the lowest values were in the control group (p < 0.05). Phagocytic activity and phagocytic index were markedly improved in fish treated with 1 and 2% MOS, followed by those fed 0.5% compared with fish fed MOS-free diet (p < 0.05). Superoxide dismutase and glutathione peroxidase were markedly improved in fish treated with 1, and 2% MOS, followed by those fed 0.5% compared with fish fed MOS-free diet (p < 0.05). Dietary MOS (0.5, 1, and 2%) meaningfully enhanced catalase activity while decreased the malondialdehyde concentration (p < 0.05). The bioavailability and pharmacokinetics in turkeys of cefquinome (CFQ), a broad-spectrum 4th-generation cephalosporin antibiotic, were explored after a single injection of 2 mg/kg body weight by intravenous (IV) and intramuscular (IM) routes. In a crossover design and 3-weeks washout interval, seven turkeys were assigned for this objective. Blood samples were collected prior to and at various time intervals following each administration. The concentration of CFQ in plasma was measured using HPLC with a UV detector set at 266 nm. For pharmacokinetic analysis, non-compartmental methods have been applied. Following IV administration, the elimination half-life (t1/2ʎz), distribution volume at steady state (Vdss), and total body clearance (Cltot) of CFQ were 1.55 h, 0.54 L/kg, and 0.32 L/h/kg, respectively. Following the IM administration, CFQ was speedily absorbed with an absorption half-life (t1/2ab) of 0.25 h, a maximum plasma concentration (Cmax) of 2.71 μg/mL, attained (Tmax) at 0.56 h. The bioavailability (F) and in vitro plasma protein binding of CFQ were 95.56% and 11.5%, respectively. Results indicated that CFQ was speedily absorbed with a considerable bioavailability after IM administration. In conclusion, CFQ has a favorable disposition in turkeys that can guide to estimate optimum dosage regimes and eventually lead to its usage to eradicate turkey's susceptible bacterial infections. The present study was conducted to evaluate the analgesic potential of the new triamilide macrolide antibiotic, tulathromycin, at 20 and 40 mg/kg of body weight (BW), subcutaneously against acute pain in mice. Acute pain was induced either chemically (using acetic acid-induced writhing and formalin-induced pain tests) or thermally (using hot-plate, and tail-flick tests). In the acetic acid-induced writhing test, tulathromycin induced a dose-dependent and significant decrease in the number of writhes compared with the control group. In the late phase of the formalin test, a significant decline in hind paw licking time compared with the control group was observed. In the hot-plate and tail-flick tests, tulathromycin caused a dose-dependent and significant prolongation of latency of nociceptive response to heat stimuli, compared with the control group. These findings may indicate that tulathromycin possesses significant peripheral and central analgesic potentials that may be valuable in symptomatic relief of pain, in addition to its well-established antibacterial effect. Simple Summary: Coccidiosis, one of the most contagious diseases among domestic rabbits, negatively affects production and results in massive economic losses. This study evaluates the therapeutic efficacy of treatment with aqueous neem leaf extract and ethanolic pomegranate peel extract (PPE) individually and in combination on the intestinal coccidiosis caused by Eimeria spp. in New Zealand white and V-line (VL) rabbits. Rabbits from two breeds were divided into ten equal groups (five groups each for NZ and VL). All rabbits were inoculated with Eimeria spp. oocysts except for the rabbits in the first group (G1) (negative control). The remaining groups were: G2, positive control, G3, treated with neem leaf extract, G4, treated with pomegranate peel extract (PPE), and G5, treated with a combination of neem leaf extract and PPE. Our results showed that the use of neem leaf and/or pomegranate peel extract for both breeds resulted in improved growth performance, a significant reduction in mean oocyst count, no mortalities, an anticoccidial index > 120, and significantly improved economic efficiency measures when compared to the positive control group. Healthy, weaned, coccidial-free male rabbits from two breeds (New Zealand white (NZ) and V-line (VL)) were divided into 10 equal groups (5 groups each for NZ and VL) (3 replicates/group, 6 rabbits/replicate, 18 rabbits/group). All rabbits were inoculated with 5 × 104 Eimeria spp. oocysts (E. intestinalis (67%), E. magna (22%), and E. media (11%)) except for the rabbits in the first group (G1), which were inoculated with a sterile solution and served as a negative control. The remaining four groups were treated as follows: G2, no treatment/positive control, G3, treated with neem leaf extract, G4, treated with pomegranate peel extract (PPE), and G5, treated with a combination of neem leaf extract and PPE. For both breeds, our results showed that the use of neem leaf and/or pomegranate peel extract resulted in improved growth performance, with a significant improvement in relative feed conversion ratio (FCR) compared to the positive control groups, which recorded the worst values, as well as a significant (p ≤ 0.05) reduction in mean oocyst count compared to the positive control groups. We also observed downregulation of mRNA levels of IL-1βα, IL6, and TNF-α in the herbal treatment groups compared with the mRNA levels of these genes in the positive control groups. Herbal treatment with neem leaf and/or pomegranate peel extracts had positive effects on the NZ and VL rabbits experimentally infected with mixed Eimeria species, as evidenced by their healthy appearance, good appetite, no mortalities, an anticoccidial index > 120, and a significantly higher total return and net profit when compared to the positive control groups of both breeds. Simple Summary: Despite the beneficial role of antibiotics in reducing bacterial infection in rabbits, there is an indirect harmful influence on human health. Thus, replacing antibiotics with friendly alternatives is a suitable strategy to protect the performance and welfare of rabbits. This study aimed at comparing the effects of including dietary egg lysozyme and zinc bacitracin antibiotic (ZnB) on the productivity and health conditions of rabbits. The results show a marked enhancement of the growth performance, antibacterial capacity, blood health, and antioxidative status in rabbits treated with egg lysozyme compared with those treated with ZnB. Thus, using egg lysozyme is recommended to replace the usage of ZnB in rabbit production. Dietary egg lysozyme has beneficial roles in the growth performance and health conditions of animals. The study was performed using 90 multicolored rabbits in three groups (each replicate with thirty rabbits). In the control group, rabbits were fed a diet without zinc bacitracin (ZnB) or egg lysozyme, while the second and third groups were treated with ZnB and lysozyme additive at 100 mg/kg, respectively. After eight weeks, the final weight and body weight gain (BWG) of rabbits fed dietary egg lysozyme and ZnB additives were meaningfully increased (p < 0.05). Nevertheless, the feed conversion ratio (FCR) was markedly decreased by dietary egg lysozyme and ZnB (p < 0.05). Interestingly, dietary egg lysozyme resulted in higher final weight and BWG and lower FCR than rabbits treated with ZnB (p < 0.05). Rabbits treated with egg lysozyme and ZnB additives had markedly lower populations of Clostridium spp. and Escherichia coli (p < 0.05) compared with the control. However, the counts of Lactobacillus and total bacteria were meaningfully increased in the the intestines of rabbits treated with egg lysozyme and ZnB (p < 0.05). The blood total protein and globulin of rabbits fed dietary egg lysozyme and ZnB additives were meaningfully increased (p < 0.05). Blood creatinine was significantly lowered by dietary egg lysozyme compared with the control and ZnB-treated rabbits (p < 0.05). The levels of blood urea, ALT, and AST were markedly lowered (p < 0.05) by dietary egg lysozyme and ZnB. The gene expressions of superoxide dismutase 1 (SOD1) and glutathione peroxidase (GPX) in the liver of rabbits fed dietary egg lysozyme and ZnB additives were markedly upregulated (p < 0.05) compared with the control. Dietary egg lysozyme resulted in higher expression of SOD1 and GPX genes than rabbits treated with ZnB (p < 0.05). Two-way ANOVA analysis revealed that the individual effects of ASP in Nile tilapia exposed to salinity levels presented a significant decrease (p < 0.05) in values of haemato-biochemical indices (such as glucose, cortisol, alanine transaminase, aspartate transaminase, and malondialdehyde) compared to those in the CTR group exposed to the same salinity levels. Moreover, significant increases (p < 0.05) of blood protein profile (albumin, globulin, and total protein), non-specific immune responses (lysozyme activity, phagocytic activity, and phagocytic index), and antioxidant enzymes activities (glutathione peroxidase, catalase, and superoxide dismutase) were observed in ASP-supplemented groups. Interestingly, there was significant (p < 0.05) downregulation of the mRNA expression values of heat shock protein 70 and interferon-gamma genes, alongside upregulation of the mRNA expression values of interleukin 1 beta and interleukin 8 genes, in the hepatic tissues of Nile tilapia in ASP-supplemented groups exposed to different salinities compared to those in the CTR group exposed to the same salinity levels. Taken together, these findings supported the potential efficacy of dietary supplementation with ASP in alleviating salinity stress-induced haemato-biochemical alterations, immune suppression, and oxidative stress in the exposed Nile tilapia. Simple Summary: Finding a suitable feed supplement is important for maintaining fish health and sustainability of the aquaculture industry. From these supplements, research studies have shown that activated charcoal (AC) has been extensively used for veterinary and aquaculture objectives as a "Universal Antidote" against several toxicants and aquatic pollutants. Therefore, the mitigating roles of dietary supplementation with different AC levels on physiological responses of Nile tilapia exposed to sub-lethal imidacloprid (IMID) toxicity were evaluated. The findings of this study revealed that dietary supplementation with 14.30 g AC/kg diet positively modulated the toxic impacts of IMID-intoxicated fish. The existing study was designed to assess the influences of dietary activated charcoal (AC) on the growth performance, immune responses, antioxidative status, and its mitigating roles against the physiological responses of Nile tilapia exposed a sub-lethal dose of a neonicotinoid agriculture pesticide, namely, as imidacloprid (IMID). Nile tilapia juveniles were fed on diets supplemented with graded AC levels as 0 (control), 5, 10, 15, and 20 g/kg diet for eight weeks. Growth, hemato-biochemical indices, and antioxidant and immune responses of fish in all groups were evaluated at the end of the feeding experiment. Afterward, fish in all experimental groups were subjected to a sub-lethal dose of IMID (0.0109 μg/L) for two weeks. Then, fish mortalities, stress indicators, and IMID residual levels in liver and flesh were examined. Results of the feeding experiment showed that total feed intake, weight gain, final body weights, and feed efficiency ratio were significantly increased in all AC groups compared with the control group. The survival rate was 100% in all experimental groups. No statistical differences were observed in the hematological picture of all experimental groups except the lymphocyte count, which was significantly increased in all AC groups compared to the control group. Total protein, albumin, globulin, nitric oxide levels, lysozyme, and respiratory burst activities were significantly increased in all AC groups. Serum alanine transaminase, aspartate transaminase, alkaline phosphatase activities, and malondialdehyde (MDA) levels were significantly decreased in all AC groups compared with the AC0 group. After exposure to a sub-lethal dose of IMID, survival rates were significantly elevated, and IMID residual levels in liver and flesh were significantly decreased in all AC groups than in the control group. Moreover, second-order polynomial regression showed that dietary supplementation with 14.30 g AC/kg diet resulted in the lowest blood glucose and serum MDA levels. Simple Summary: Corn silk meal (CSM), the outer thread-like material found under corn husks, is a by-product of corn cultivation, which has been documented as a traditional medicine with multiple benefits for human health. Broilers cannot produce the enzymes needed to hydrolyze non-starch polysaccharide (NSP), which has been found to increase gut viscosity, and reduce the digestion and utilization of nutrients, thereby resulting in poor animal performance. The objective of this study was to examine the effects of diets supplemented with CSM and NSP on growth performance, blood biochemistry, immunological and growth-related gene expression in broiler chickens. A total of 270 broiler chickens were divided into six experimental groups: (1) basal diets (BD) as control; (2) BD supplemented with 0.5 g/kg feed NSP enzyme; (3) and (4) fed a diet containing 40 and 80 kg/ton of CSM; (5) and (6) fed a diet containing 40 and 80 kg/ton CSM and supplemented with 0.5 g/kg NSP enzyme. The results showed that the dietary inclusion of CSM with the NSP enzyme can improve growth performance, intestinal histopathology parameters, modify plasma lipids, and enhance immune response in broilers. Interestingly, the relative mRNAs of the growth hormone receptor (GHR) and insulin growth factor (IGF) were increased (p < 0.05) with the CSM and NSP enzyme: the relative mRNA expressions of cholecystokinin (CCK) and leptin were decreased by feeding CSM diets with the NSP enzyme.
