Carbon dots (CDs), because of their unique properties, are being rapidly developed as important luminescent materials for imaging, sensing, and use in photonic devices. However, most of the reported fundamental properties of the CDs are results of investigations conducted in the solution state, which may be completely different from those conducted in the solid state. In this work, we study the luminescence properties, photostability, and the dynamics of CDs in different matrix environments, from ensemble to the single-particle level. We observed that the properties associated with the emission centers and photostability of CDs were extremely sensitive to the local chemical environment. A better understanding of the dependence of the spectroscopic properties of CDs on the complex local chemical environment is an important step toward finding new ways of controlling the optical properties of CDs and optimizing their use in various applications.

We report what we believe is the first use of 19F NMR spectroscopy to directly measure in-vitro release (IVR) from polymeric nanoparticles (PNPs). Using 19F NMR we selectively measured IVR of AZD2811 from PNPs. Due to rapid nuclear relaxation in solid-like environments only AZD2811 in solution is detected, and physical separation from the PNPs isn't required. The NMR approach and ultra-centrifugation/UHPLC were shown to be equivalent. The selectivity of 19F NMR means it is readily applied to complex IVR media such as recombinant human serum albumin (rHSA).

Aim: The current study dealt with the degradation behavior of Bilastine and degradation kinetics of a drug in solution state. Background: Very limited information on the effect of pH on maximum stability has been published. In order to understand the degradation kinetics of bilastine, aqueous stability studies were carried out, because such studies on bilastine have not been reported in the literature, further no methods have reported about shelf-life determination of bilastine. The study design involves selection of stability indicating RP-HPLC method for estimation of drug then evaluation of degradation kinetics, shelf-life determination and validation of proposed method. Results: The Shimadzu HPLC series 1100 was used for stress degradation analysis of bilastine in tablet dosage form. The analysis was performed using Agilent ZORBAX SB-C8 (4.6×150×5µm) column and Phosphate Buffer: Acetonitrile (pH-5.0) in the ratio of 60:40 as mobile phase; wavelength selected for analysis was 254nm with the flow rate of 1mL/min at which drug showed sharp peak. The analysis was performed on the isocratic pump mode with the injection volume of 20µl. The mobile phase is used as diluent. The proposed method was found to be linear over the range 10 to 50 µg/mL. The analysis was performed by placing standard and samples with 7 different pH buffer, oxidative and neutral hydrolytic solutions in oven at 40ºC, 60⁰C and room temperature for an interval of 30, 60, 90, 120, 150, 180 mints for standard and samples. The results indicated that the pH, temperature, ionic strength and oxidation greatly influence the stability of Bilastine and the degradation behavior of Bilastine followed pseudo-first-order kinetics. Bilastine was most stable in neutral, alkaline, lower temperature conditions and lower ionic strength. Conclusion: The proposed method was found to be specific, selective and robust and successfully applied for its assay, degradation (stress testing) of drug and degradation kinetics in solution state. A series of zinc complexes of 2,7-bis(2-pyridyl)-1,8-naphthyridine (BPNP) has been synthesised and characterised by single crystal X-ray diffraction and solution state NMR spectroscopic studies. Reactions of BPNP with zinc acetate and zinc chloride were found to give mononuclear complexes of the form [Zn(BPNP)X2] (X = OAc, Cl), whereas reactions with zinc triflate led to a mixture of products. Several of these were identified crystallographically as [Zn(BPNP-H)(H2O)4](OTf)3 and [Zn(BPNP-H)(NCMe)(OTf)2]OTf, in which protonation of one pyridyl group occurred, and the dimeric species [Zn2(BPNP)4(μ-H2O)2](OTf)4. A trimetallic complex [Zn3(μ2-BPNP)(μ2-OAc)3(OAc)2(μ3-OH)] was also isolated from reactions involving zinc acetate, and demonstrates the ability of BPNP to coordinate two zinc atoms in the adjacent binding pockets.

The Mre11-Rad50-Nbs1/Xrs2 protein complex plays a pivotal role in the detection and repair of DNA double strand breaks. Through traditional and emerging structural biology techniques, various functional structural states of this complex have been visualized; however, relatively little is known about the transitions between these states. Indeed, it is these structural transitions that are important for Mre11-Rad50-mediated DNA unwinding at a break and the activation of downstream repair signaling events. Here, we present a brief overview of the current understanding of the structure of the core Mre11-Rad50 complex. We then highlight our recent studies emphasizing the contributions of solution state NMR spectroscopy and other biophysical techniques in providing insight into the structures and dynamics associated with Mre11-Rad50 functions.

Nuclear magnetic resonance (NMR) spectroscopy is a well-established method for analyzing protein structure, interaction, and dynamics at atomic resolution and in various sample states including solution state, solid state, and membranous environment. Thanks to rapid NMR methodology development, the past decade has witnessed a growing number of protein NMR studies in complex systems ranging from membrane mimetics to living cells, which pushes the research frontier further toward physiological environments and offers unique insights in elucidating protein functional mechanisms. In particular, in-cell NMR has become a method of choice for bridging the huge gap between structural biology and cell biology. Herein, we review the recent developments and applications of NMR methods for protein analysis in close-to-physiological environments, with special emphasis on in-cell protein structural determination and the analysis of protein dynamics, both difficult to be accessed by traditional methods.

In practical applications, fixing a fluorescent sensor on a solid composite film matrix can settle the boundaries of keeping the sensor in the solution state. Herein, we first presented a preparation of anime-response membrane by utilizing the edible gelatin as a film-forming skeleton to immobilize the dual phenazine derivative (PD-6), acting as amine-responsive functional units to fabricate a novel solid sensor. In order to ensure the sensor (PD-6 film) is available in the actual application, the mechanical properties, anime-responsive performance, and further application for monitoring shrimp freshness were fully characterized. Those observations indicated that the PD-6 film not only enhanced the sensitivity to anime, but also strengthened the application potential as a solid fluorescent sensor with flexible recyclability and favorable antibacterial properties. It is hoped that this valuable strategy could open a new avenue to overcome the application limitations of the sensor in the solution state.

A series of three aza-Bodipy donor molecules namely Aza-Bthp, Aza-Sty, and Aza-Fhdt have been synthesized. The compounds exhibit panchromatic absorption spanning approximately 280–1000 nm in the solution state and possess suitable energy levels for their usefulness as donors in organic solar cells. Under the optimized conditions, using PC70BM as an acceptor, power conversion efficiencies (PCE%) of 2.44, 1.2, and 2.52 were afforded for Aza-Bthp, Aza-Fhdt, and Aza-Sty respectively.

Supramolecular low molecular weight gels have potential in many areas from medicine to optoelectronics. Of the different applications, visual sensing of chemical analytes using gels is a fairly new concept in materials chemistry research. Among various analytes, cyanide is considered the most threatening to the environment and human life because of its acute toxicity. Generally, the detection of cyanide involves the use of sophisticated and expensive instrumentation, which complementary entails highly trained personnel to operate it. In contrast, a gel-based visual detection technique provides an advantage as it is simple, cost-effective, and instrument-free whilst the sensing event can be executed either by observing a phase transformation or by a naked eye detectable color change of the gel. Further, as the gel formation requires a high concentration of gelator (≥10−3 M), for the same chemical compound, the gel phase interactions often exhibit better selectivity for the ionic guests than in the solution state. Despite these facts, limited attempts have been made to synthesize gel-based cyanide sensors. In this review, we discuss an up-to-date summary of various reports on cyanide responsive gels emphasizing the approaches, design principles, and reaction mechanisms. We also highlight the advantages, limitations, and challenges as well as the necessity of further exploration of gels in this domain.

Sulfonylhydrazones are a novel hydrazone-based organic molecular photoswitch. The four derivatives with unsubstituted and o-, m- and p-nitro substituted sulfonylhydrazone (SH-1, SH-2, SH-3, and SH-4, respectively) derived from p-toluenesulfonyl hydrazide and corresponding benzaldehydes were examined with respect to their solution state photoswitching and photomechanical response in the crystalline state. All the compounds displayed UV-induced E → Z isomerization in the solution state, whereas the back conversion was slow. Single crystals of SH-3 displayed rapid and large photomechanical bending, whereas SH-1 and SH-4 crystals underwent slow bending to a comparatively lesser extent. On the contrary, SH-2 crystals did not show any photomechanical effects. The photomechanical deflection of the crystal tip of SH-1 increased linearly; for SH-3, the deflection increased in a sub-linear manner for few seconds, after which it started to revert towards the light source. For SH-4 crystals, the photo-induced tip deflection initially increased in a sub-linear fashion, after which no motion was observed even after continuous exposure to UV light irradiation. The photomechanical behavior of the crystals was also inspected with the aid of velocity-time and acceleration-time plots that unravelled the instantaneous photomechanical motion at various time intervals.

A series of group 13 dichloride species, supported by monoanionic NNN-pincer ligands, LMCl2 (L = 2,5-(iPr2PNAr)2N(C4H2); Ar = 4-iPrC6H4 (LPipp), 2,4,6-Me3C6H2 (LMes), 2,6-iPr2C6H3 (LDipp); M = Ga (1), In (2)), were prepared via the reaction of NaL with MCl3 and characterized by multinuclear NMR spectroscopy, elemental analysis and X-ray crystallography. Related organoaluminum complexes, LAlMe2 (3), were synthesized by addition of AlMe3 to L. Treatment with the Lewis acid B(C6F5)3 afforded well-defined cationic aluminum methyl species, [LAlMe]+[MeB(C6F5)3]− (4), which were found to be thermally stable in both solid and solution state for days.

A method was developed for rapid qualitative determination of lignocellulose in the tobacco cell wall by utilizing 2D heteronuclear single quantum coherence NMR spectra (2D HSQC NMR). Traditional methods for analyzing the structure of lignocellulose involve many steps of separation and extraction, which is labor-intensive. In this work, the whole cell wall was milled and dissolved in deuterium solvent. The solvent dimethylsulfoxide (DMSO-d6) containing hexamethylphosphoramide (HMPA-d18) enhanced swelling of the sample and gave high-resolution spectra. The tobacco samples are ball milled at different ball milling times, and the state of the particles is observed through an electron microscope, and then the probability of the particles being less than 5 µm is counted. Through the comparison of the abundance and integration of the peak signals in the spectra under different transmittances, it was determined that when the milling time was 6 h, the quality of the NMR spectra was the best. The optimum conditions of characterizing tobacco structure were DMSO-d6/HMPA-d18 solution and 6 h milling time. Under these conditions, complete representation of the structure of lignocellulose and simplified process could be achieved.

Interaction of [Pb(SiPr)2]n with 1.5 equiv. of AgNO3 in DMF under aerobic conditions at room temperature gave a cationic lead-centered thiolate cluster [Pb13(SiPr)6O8](NO3)4 (1). While treatment of [Pb(SiPr)2]n in refluxing DMF in air afforded a two-dimensional layered structure of [Pb14(SiPr)16O6]n (2), comprising of [Pb14(SiPr)18O6]2− units bridged the iPrS− moieties. Molecular structures of the two oxidation products of μ4-oxygen-bridged high-nuclearity lead(II) clusters have been determined by single-crystal X-ray analysis. The UV/Vis properties of the two clusters in both solid state and solution state were also investigated in this paper.

Controlling the solution-state aggregation of conjugated polymers for producing specific microstructures remains challenging. Herein, a practical approach is developed to finely tune the solid-state microstructures through temperature-controlled solution-state aggregation and polymer crystallization. High temperature generates significant conformation fluctuation of conjugated backbones in solution, which facilitates the polymer crystallization from solvated aggregates to orderly packed structures. The polymer films deposited at high temperatures exhibit less structural disorders and higher electron mobilities (up to two orders of magnitude) in field-effect transistors, compared to those deposited at low temperatures. This work provides an effective strategy to tune the solution-state aggregation to reveal the relationship between solution-state aggregation and solid-state microstructures of conjugated polymers.

A series of polynuclear gold(I) sulfido complexes of bis(diphenylphosphino)amine ligands has been synthesized and characterized. A rather small variation in the conformation of the bis(diphenylphosphino)amine ligands has led to distinct differences in the identity of the polynuclear gold(I) sulfido complexes formed. Unprecedented concentration-dependent and solvation-dependent reversible cluster-to-cluster transformation between a dodecanuclear gold(I) sulfido complex (LMe-Au12) and a hexanuclear gold(I) sulfido complex (LMe-Au6) has been observed. The transformation process has been monitored not only by 1H and 31P{1H} NMR spectroscopy but also by UV-vis absorption spectroscopy and high-resolution electrospray ionization mass spectrometry (HR-ESI-MS) in the solution state. This work has provided a simple approach to achieve structure modulation of gold(I) sulfido complexes and an understanding of supramolecular transformations via external stimuli.

Mechanistic understanding of the asymmetric autocatalysis of pyrimidyl alkanol is a highly attractive and challenging topic due to its unique feature of amplification of enantiomeric excess. Circular dichroism spectroscopic analysis of this reaction allows monitoring of the structual changes of possible catalyst precursors in the solution state and shows characteristic temperature and solvent dependence. TD-DFT calculations suggest that these spectral changes are induced by a dimer–tetramer equilibrium of zinc alkoxides.

The degree of oxidation of graphene oxide (GO) has been shown to be important for its toxicity and drug-loading efficiency. However, the effect of its variations on GO-protein interaction remains unclear. Here, we evaluate the effect of the different oxidation degrees of GO on its interaction with human ubiquitin (8.6 kDa) using solution state nuclear magnetic resonance (NMR) spectroscopy in combination with other biophysical techniques. Our findings show that the interaction between the protein and the different GO samples is weak and electrostatic in nature. It involves fast dynamic exchange of the protein molecules from the surface of the GO. As the oxidation degree of the GO increases, the extent of the interaction with the protein changes. The interaction of the protein with GO can thus be modulated by tuning the degree of oxidation. This study opens up new avenues to design appropriate graphenic materials for use in various biomedical fields such as drug delivery, biomedical devices and imaging.

High yield synthesis and spectroscopic isolation of two hitherto unknown highly stable single conformers of meso-aryl substituted dithia/diselena trans-doubly N-confused porphyrins with fully π-conjugated [18] annulene structures are reported. In-depth solution state spectroscopic measurements and DFT level theoretical calculations strongly show the distinct aromaticity with strong NIR absorption of these new macrocycles.

Intramolecular rotations are known to interfere with the excited-state processes resulting in luminescence quenching. Efforts aimed at restricting intramolecular rotations have led to the development of highly luminescent systems. Here we report the luminescence properties of a BODIPY molecule (I6) containing six iodo-substituents. The luminescence of I6 was highly quenched in the solution state whereas the conversion of this molecule into its nanoaggregates enhanced the emission intensity. Detailed photophysical studies and molecular simulations were employed to study the underlying mechanism which showed that the electron-withdrawing nature of iodine atoms and the peculiar structure of I6 contributed towards the anomalous luminescence behaviour. Further, the luminescence and photosensitization properties of I6 were exploited for cell imaging and photodynamic therapy. Thus, our simple but intelligent molecular design yielded a multi-faceted molecule having excellent photophysical properties for potential photobiological applications.

The interplay of organic dications bearing various n-alkylimidazolium termini substituted at different positions on a bis(methylene)phenyl ring and p-sulfonatocalix[4]arene, along with tetraphenylphosphonium cations and hydrated lanthanides (Ce3+, Er3+ and Gd3+), afforded various self-assembled solid-state structures. The host–guest binding behaviour of the dications and p-sulfonatocalix[4]arene was carefully investigated by crystallographic studies including their interactions in the solution state, but in the absence of phosphonium and lanthanide cations via1H NMR experiments.

Antimicrobial function of Ag nanoparticles (NPs) has a strong correlation with the released Ag+ cations that are produced by oxidation of Ag NPs in a solution state under ambient condition. Therefore, in order to develop anti-infective materials for biomedical applications, one needs to include Ag NPs inside biocompatible materials, which can allow slow release of Ag+ cations. Hydrogels of natural polymers could be an ideal choice for the purpose because (a) the physicochemical properties of hydrogels resemble with biological tissue, and (b) the inclusion of Ag NPs inside hydrogels prevents the direct release of Ag NPs, while allowing the release of Ag+ cations out of the hydrogels. In this regard, we present a simple strategy for producing Ag NPs-containing hydrogel based on natural alginate polymers. The chemical modification of alginate, blending with Ag NPs, gelation by photo-crosslinking process have been discussed in connection with antimicrobial reaction on model bacterium.

Finely controlled circularly polarized luminescence (CPL) supramolecular polymerization based on a tetraphenylethene core with four l- or d-alanine branch side chains (l-1 and d-1) in the solution state is presented, resulting from the tuning of mechanical stimulus. Weak, green emissions of l-1 and d-1 in tetrahydrofuran (THF) were converted into strong blue emissions by tuning the mechanical stimulus. The strong blue emissions were caused by an aggregation-induced emission (AIE) effect during the formation of a supramolecular polymer. Lag time in the supramolecular polymerization was drastically reduced by the mechanical stimulus, which was indicative of the acceleration of the supramolecular polymerization. A significant enhancement of circular dichroism (CD) and CPL signals of l-1 and d-1 was observed by tuning the rotational speed of the mechanical stimulus, implying that the chiral supramolecular polymerization was accelerated by the mechanical stimulus.

Careful pulse sequence design and optimization is critical to the success of a given NMR experiment. Over the past several decades the level of sophistication of NMR pulse sequences has increased tremendously, leading to large spectral sensitivity and resolution improvements, to data sets with far fewer artifacts, and to much more rapid acquisition times, opening up a wide range of applications. Here I briefly highlight how pulse sequence ‘engineering’ has evolved, focusing on liquid state NMR, and, in particular, on the HNCA-class of triple-resonance experiment. In many respects, the evolution of triple-resonance NMR mirrors the evolution of solution state NMR experiments in general, with ‘tricks’ that first appeared in triple-resonance pulse sequences or that were motivated by them now incorporated into a broad range of experiments.

The development of a high-flux beamline for X-ray footprinting at NSLS-II is presented., Hydroxyl-radical mediated synchrotron X-ray footprinting (XF) is a powerful solution-state technique in structural biology for the study of macromolecular structure and dynamics of proteins and nucleic acids, with several synchrotron resources available to serve the XF community worldwide. The XFP (Biological X-ray Footprinting) beamline at the NSLS-II was constructed on a three-pole wiggler source at 17-BM to serve as the premier beamline for performing this technique, providing an unparalleled combination of high flux density broadband beam, flexibility in beam morphology, and sample handling capabilities specifically designed for XF experiments. The details of beamline design, beam measurements, and science commissioning results for a standard protein using the two distinct XFP endstations are presented here. XFP took first light in 2016 and is now available for general user operations through peer-reviewed proposals. Currently, beam sizes from 450 µm × 120 µm to 2.7 mm × 2.7 mm (FWHM) are available, with a flux of 1.6 × 1016 photons s−1 (measured at 325 mA ring current) in a broadband (∼5–16 keV) beam. This flux is expected to rise to 2.5 × 1016 photons s−1 at the full NSLS-II design current of 500 mA, providing an incident power density of >500 W mm−2 at full focus.

The A-A/B-B step-growth copolymerization between a monomer immobilized in the crystalline state and a monomer mobile in the solution state is demonstrated. One of the two monomers was immobilized as organic ligands of the metal-organic framework (MOF) and polymerized with the mobile guest monomer, resulting in the formation of linear polymers. The polymerization behavior was completely different from that of the solution polymerizations. In particular, the degrees of polymerization (DP) converged to a specific value depending on the MOF structures. The inevitable termination is caused not by imperfectness of the polymerization reaction, but by the selection of the two polymerization partners among the several adjacent immobilized monomers. This is fully supported by the Monte Carlo simulation on the basis of the polymerization mechanism. Precise immobilization of monomers in the supramolecular assemblies is a promising way for the controlled A-A/B-B step-growth polymerization.

A series of novel phenothiazine based pyridylhydrazones displaying large Stokes shifts, acidochromism and pH sensing in aqueous media was synthesized in good yields. Substituents with different electronic and steric effects were appended on the phenothiazine ring in order to fine tune its donor ability. The photophysical studies revealed the existence of enhanced ICT character in the compound bearing electron withdrawing 4-cyanophenyl group. Effects of steric hindrance and increased conjugation were also examined by spectrophotometric studies. Acid sensing behavior in solution with pKa values in the range of 4.7–5.8 was observed in the hydrazones bearing one pyridyl group. These pKa values are similar to the pH of acidic intracellular organelles. Hydrazone bearing two pyridyl groups displayed an increased pH sensing range with two pKa values at 4.5 and 6.8. Reversible changes in color and spectrophotometric properties were observed in solution state. Also, reversible solid state acidochromism was displayed by all the hydrazones upon consecutive exposure of highly volatile TFA and TEA vapors. The reversibility of hydrazones was further utilized for the construction of molecular logic gates with inhibit (INH) function using TFA/TEA or HCl/NaOH as the chemical inputs.

We report the self-assembled structure formed by Fmoc protected charge single amino acid Fmoc-L-glutamic acid 5-tert-butyl ester (Fmoc-Glu(OtBu)-OH), Fmoc-L-aspartic acid 4-tert-butyl ester (Fmoc-Asp(OtBu)-OH), and Na-Fmoc-Ne-Boc-L-lysine (Fmoc-Lys(Boc)-OH). The self-assembled architecture formed by the charge aliphatic amino acids were assessed under different conditions such as concentration, temperature and pH. Fmoc-Glu(OtBu)-OH assembled to spheres at both lower and higher concentration under room temperature condition. However, it forms a broom stick like morphology at both lower and higher concentration on heating. Fmoc-Asp(OtBu)-OH on the other hand formed rod like at both both low and high concentration and also on hhealing. Fmoc-Lys(Boc)-OH also self-assemble to sphere like morphology in all conditions irrespective of concentration and heating. Since these structure are very intriguing, our future endevorous is to study these structure through different microscopic techniques such as scanning electron microscopy (SEM), Transmission Electron microscopy (TEM). The mechanisms of the structure formation by these amino acids will be characterized by using solution state NMR, FTIR and TGA in future. The self-assembled structures formed by modified amino acids are easy and facile route to design novel nanoarchitectures which may be potentially useful in future for various type of applications in the field of material chemistry, bioscience, biomedical.

We report the self-assembly of carbobenzoxyphenylalanine (Z-Phe-OH), carbobenzoxytryptophan (Z-Trp-OH), carbobenzoxytyrosine (Z-Tyr-OH) and N-(9-Fluorenylmethoxycarbonyl)-O-tert-butyl-L-tyrosine (Fmoc-Tyr(tbu)-OH) to well-defined morphologies such as fibers, spherical and flower-like self-assembled structure. The self-assembled structure formed by modify single amino acids were characterized through various microscopic and spectroscopic techniques. The self-assembled structure formation was studied extensively under varying concentrations and temperature and the mechanism of self-assembled structure formation was extensively studied through solution state 1H NMR. The self-assembly studies on modified amino acid provide an important avenue for the simple fabrication and design of novel materials with immense applications. Hence, Z-Phe-OH, Z-Trp-OH, Z-Tyr-OH, and Fmoc-Tyr(tbu)-OH self-assembly studies may be a pertinent extension in this direction.
