Spores of Bacillus thuringiensis contain a toxin active against lepidopterous larvae. This toxin can be solubilized by extraction with reagents which dissolve the protein crystal of B thuringiensis. It is inactivated by crystal-specific antiserum. Spores of Bacillus cereus contain a similar toxin although the specific activity is much lower than the spores of B. thuringiensis. The B. cereus toxin contains a single major polypeptidecomponent. Toxic activity can be solubliized from spores of both species by incubation with gut juices from Pieris brassicae.

Crystals were purified from four serotypes of the insect pathogen Bacillus thuringiensis. Crystals from these serotypes were similar in amino acid and N-terminal analyses, but differed in their toxicity to two species of Lepidoptera and in their immunological properties. Toxic polypeptides were obtained following trypsin digestion of solutions of the crystals. In two strains (serotypes 3 and 9) this fraction contained only one polypeptide. Similar results were obtained when dissolved crystals were digested with other proteolytic enzymes or with gut contents from Pieris brassicae. The trypsin-resistant polypeptide was further purified by gel and ion-exchange chromatograhy and had a molecular weight of about 70,000, estimated by gel chromatogrpahy and gel electrophoresis. No evidence was obtained for a toxin of lower molecular weight. This purified toxin accounted for most, if not all, of the toxic activity originally present in the crystal solution and was active by injection and ingestion. The purified toxic fraction from serotype 1 appeared to contain two polypeptides, one of which corresponded to that found with serotypes 3 and 9. There were no major differences in the composition of crystals from different serotypes of B. thuringiensis and it is concluded that the trypsin-resistant polypeptide represents the active insecticidal toxin of the crystal.

Two membrane fractions, F1 and F2, have been purified from the outer layers of spores of Bacillus thuringiensis. Both fractions contain 6-7% cysteine and appear to be similar in composition. Amino acids account for about 75% of the dry weight, carbohydrate for about 2% and lipids for about 25%. The fractions are both toxic to Pieris brassicae and the toxicity is inactivated by antiserum to the toxic crystal of Bacillus thuringiensis. The fractions can be distinguished by examination under the electron microscope; both fractions show similar hexagonal patterns but with different spacings. The same fractions from an acrystaliferous mutant (cr) were prepared. These were identical in density and in appearance under the electron microscope; the amino acid analysis of fraction F2 from both strains was identical. However, the spores and fractions F1 and F2 from this strain lacked toxicity. Fraction F2 from the cr strain was used to prepare antiserum specific to fraction F2. Using this anti-serum and anticrystal serum, crystal and F2 antigens were shown to appear simultaneously in sporulating cultures. Crystal and F2 antigens appeared some time before the maximum rate of uptake of [35s]cysteine. It is concluded that fraction F2 is derived from the exosporium and that fraction F1 probably originates from the spore coat. The exosporium in Bacillus thuringiensis appears to be synthesised during stages II and III of sporulation although uptake of [35S]cysteine occurs much later.

Spore suspensions containing about 0.3% crystals and crystal suspensions containing about 0.1% spores were obtained from cultures of Bacillus thuringiensis by extraction with a two-phase system. Both preparations were tested for the presence of contaminating material from vegetative cells and were judged to be clean. Solutions of spore protein were obtained by extracting broken spores with phosphate buffer followed by extraction with either alkali- or urea-mercaptoethanol. The alkali spore or urea spore extracts had the same isoelectric point as crystal protein solubilized with these reagents. An antiserum prepared against alkali crystal solution precipitated alkali or urea spore extracts and crystal solutions but not phosphate spore extracts or extracts of whole cells. Lines of identity between spore and crystal precipitates were observed by using the Ouchterlony double-diffusion technique. Absorption of the antiserum with an excess of urea spore extract caused a disappearance of the precipitin bands originating from the spore protein and the homologous bands from the crystal protein. The results suggest that the crystal and endospore contain one or more common proteins.

Peptostreptococcus elsdenii, a strict anaerobe from the rumen, was grown on a medium containing yeast extract and [1-(14)C]- or [2-(14)C]-lactate. Radioisotope from lactate was found in all cell fractions, but mainly in the protein. The label in the protein fraction was largely confined to a few amino acids: alanine, serine, aspartic acid, glutamic acid and diaminopimelic acid. The alanine, serine, aspartic acid and glutamic acid were separated, purified and degraded to establish the distribution of (14)C from lactate within the amino acid molecules. The labelling patterns in alanine and serine suggested their formation from lactate without cleavage of the carbon chain. The pattern in aspartic acid suggested formation by condensation of a C(3) unit derived directly from lactate with a C(1) unit, probably carbon dioxide. The distribution in glutamic acid was consistent with two possible pathways of formation: (a) by the reactions of the tricarboxylic acid cycle leading from oxaloacetate to 2-oxoglutarate, followed by transamination; (b) by a pathway involving the reaction sequence 2 acetyl-CoA-->crotonyl-CoA-->glutaconate-->glutamate.

The crystalline inclusion of Bacillus thuringiensis, dissolved in 8 m urea containing 10% 2-mercaptoethanol and dialyzed to pH 8.3 to 8.5, was compared with a fraction obtained by the same extraction procedure from spores broken by dry rupture. The two fractions behaved similarly on chromatography with Sephadex G-100 and diethylaminoethyl cellulose. The preparations behaved identically on acrylamide gel electrophoresis at pH 12 and pH 9.5. Further, peptide maps of the two fractions obtained after digestion with trypsin were almost superimposable. Amino acid analyses of the crystal and spore fraction were closely similar; discrepancies are attributed to contamination of the spore extract with small amounts of other proteins. It is concluded that a significant portion of the spore protein is identical with the crystal protein.

Thin sections of sporulating cells of Bacillus thuringiensis show a marked association between the crystalline inclusion and the exosporium.

Treatment with urea-mercaptoethanol of purified spores of Bacillus thuringiensis, other Bacillus species, and Clostridium roseum solubilizes a protein fraction between 5 and 12% of the dry weight of the spores. This fraction behaves identically to the crystal protein of B. thuringiensis on acrylamide-gel electrophoresis. The protein from all of the Bacillus species shows partial homology with crystal protein, using the Ouchterlony immunodiffusion technique. A further fraction, similar in amount, can be removed from spores of B. thuringiensis by the addition of sodium lauryl sulfate to the urea-mercaptoethanol. Spores of B. thuringiensis extracted in these ways show no difference when compared to untreated spores with respect to viability or resistance to heat and ultraviolet-irradiation. The extracted spores do show differences in their germination requirements and their susceptibility to phase-darkening by lysozyme. It is concluded that an urea-mercaptoethanol-soluble protein or class of protein is a widespread component of bacterial spores, possibly located in the spore coat, and that this protein may be related to the crystal protein of B. thuringiensis.

Cell-free extracts of Peptostreptococcus elsdenii, a strict anaerobe from the rumen, were examined for enzymes catalysing the steps in the biosynthesis from lactate of alanine, serine, aspartate and glutamate. Extracts contain the enzymes necessary for the formation of alanine from lactate via pyruvate. The presence of enzymes catalysing the interconversion of phosphoglycerate and phosphohydroxypyruvate, the transamination of the latter to phosphoserine and the cleavage of phosphoserine to serine and inorganic phosphate was demonstrated, suggesting that serine is formed via these intermediates. ;Malic' enzyme, malate dehydrogenase and glutamate-oxaloacetate transaminase are present in extracts and could account for aspartate formation. The extracts catalyse all of the steps of the tricarboxylic acid pathway leading from oxaloacetate plus acetate to glutamate. Together with substantive data from previous radioactive tracer studies the results provide strong evidence that these four amino acids are synthesized in this strict anaerobe by pathways closely similar to those operating in aerobic and facultatively aerobic organisms.
