Abstract

The present study examined the dietary substitution effect of chicken by‐product meal (CBM) for fish meal (FM) on growth, feed utilization, body composition, haematology and non‐specific immune responses of olive flounder, Paralichthys olivaceus. Six isonitrogenic and isolipidic experimental diets were prepared. The control (Con) diet contained 650 g/kg FM. One hundred, 200, 300, 400 and 500 g/kg FM were replaced with CBM, referred to as the CBM10, CBM20, CBM30, CBM40 and CBM50 diets, respectively. A total of 400 juvenile fish averaging 14.7 g was distributed into 20 tanks (20 fish/tank) and fed triplicate groups of each diet, except for quadruplicate groups for the Con and CBM30 diets twice daily to satiation for 8 weeks. Arginine, phenylalanine and threonine content of the experimental diets increased with dietary increased substitution of FM with CBM. The experimental diets did not affect survival, weight gain, specific growth rate, feed consumption and feed utilization of fish. The chemical composition, whole body amino acid profiles, haematological variables and non‐specific immune responses of fish were not changed by the experimental diets. These findings suggest that FM up to 500 g/kg can be substituted with CBM without retardation in growth, feed utilization, haematology and non‐specific immune responses of olive flounder. Abstract

The gonadosomatic index (GSI), gonadal development and changes in hormones in plasma level of the indoor cultured grunt (Hapalogenys nitens) were investigated by histological study from August 2011 to October 2012. The GSI showed similar trends with gonad developmental stages during the culture periods. Changes in plasma level of estradiol-17β of female H. nitens reached the highest value before the spawning period, and seasonal changes in plasma level of estradiol-17β were similar in trends of oocyte developments and GSI changes. Testosterone levels of male H. nitens reached the highest value before and after the spent stage. Ovarian developmental stages of H. nitens could be classified into early growing stage, late growing stage, mature stage, ripe and spawning stage, recovery and resting stage. The testicular developmental stages could be divided into growing stage, mature stage, ripe and spent stage, and recovery and resting stage.

Abstract

The effects of diluents composition on cold storage for olive flounder (Paralichthys olivaceus) sperm were examined in terms of the swimming speed of sperm, the percentage of mobile sperm, and fertilization rates. The following results indicated that cold storage methods with fresh conditions could be employed in olive flounder milt preservation. The preserved sperm of olive flounder that was diluted I: 10 with artificial seminal plasma II (ASP II) and Stein`s solution (SS) at remained motile for 30 days. The most effective condition for cold storage was ASP II and SS at 0 for the sperm, although there is no significant difference statistically. No difference in the fertilization rate was found between fresh sperm and the preserved ones with ASP I, II and SS at 10 days post-storage.

Abstract

Olive flounder (Paralichthys olivaceus) is one of the most economically important aquaculture fish. However, its production is often affected by various diseases, especially viral hemorrhagic septicemia virus (VHSV) that cause serious economic losses. In this study, we sequenced the whole transcriptome of the P. olivaceus using Illumina RNA-sEq. De novo assembly of control and virus-infected cDNA libraries of head kidney at 13 and 20 °C was accomplished with 2,007,532,438 raw reads, resulting in 244,578 unigenes with an average length of 533 bp and found 65,535 candidate coding unigenes with homology to other species by BLAST analysis. DEG analysis among control and virus-infected head kidney samples of 13 and 20 °C revealed that 1290 up-regulated and 162 down-regulated genes (p ≤ 0.01), linked to metabolism, virulence factors, adhesion and immune-response. We constructed an expressed gene catalog for the P. olivaceus to serve as a resource for marine environmental genomic and immuno-genetic/genomic studies focused on uncovering the molecular mechanisms underlying the responses of P. olivaceus to VHSV under different temperature.
