Purpose: The embryonic microenvironment contains many properties that have not yet been fully explored. Our aim in this study is to report an optimized and efficient method that enables investigating the effects of the secretome of pluripotent embryonic stem cells on cancer stem cells. Methods: The study is performed with a chimeric model consisted of mouse blastocysts, human prostate cancer stem cells and non-cancer stem cells. Ovulation induced mice were used for blastocyst collection. DU145 prostate cancer cell line was separated into cancer stem cells and non-cancer stem cells according to cancer stem cells biomarker expressions by fluorescent activated cell sorting method. Human prostate cancer stem cells and non-cancer stem cells were microinjected into 4-day blastocyst culture in vitro by intracytoplasmic sperm injection method. Results: Chimeric models provide us great convenience in basic oncological studies. In this study, using a chimeric model, we were able to study the secretome of mouse embryonic stem cells and their effect on cancer stem cells. The method is efficient and yield promising result; and could be used to study the effects on other cells as well. Conclusion: The embryonic stem cell microenvironment is suggested to have a great regenerative capacity which is, nowadays, the center of attraction for cancer research studies. Ethical issues restrict the human embryo studies, however, mimicking the in vivo human microenvironment with 3D cell cultures or bioprinting are now possible. Finally, optimization of new methods including 3D cell cultures with human cell lines will be a great opportunity for better understanding the reprogramming notion.

Prostat kanserinin tanı ve tedavisine yönelik birçok alanda ilerleme sağlanabilmesine rağmen hastalık bazı vakalar için ölümcül olma niteliğini sürdürmektedir. Hastaların ölümden kurtulması için atılan her adım hedefe yaklaşılmasına yardım etse de halen sonuca ulaşmak için araştırılması gereken pek çok konu bulunmaktadır. Kök hücrenin keşfi ile bu hücrelerin insan sağlığı için önemi anlaşılmış ve tedavide nasıl kullanılacağının belirlenmesine yönelik çalışmalar büyük hız kazanmıştır. İlerleyen yıllarda Kanser Kök Hücresi kavramı ortaya çıkmış ve bu hücrelerin, kök hücre özelliklerini taşıyan ancak tümör dokusu içinde metastazı yapan, tedavi sonrası nükse yol açabilen veya tedaviye direnç̧ geliştiren hücreler oldukları belirlenmiştir. Köklülük özelliğine sahip bu hücreler dışında kalan hücre gurubu kanser kök hücresi olmayan hücre gurubudur ve konvansiyonel kanser tedavisine cevap veren kanser hücrelerdir. Kanserin metastaz yapması ve çevre dokuya invazyonunda adezyon moleküllerinin önemi büyüktür. Yapılan çalışmalar özellikle çoklu ilaç direnci ve epitelial mezenşimal geçişte adezyon moleküllerinin büyük önem kazandığını göstermiştir. Bu çalışmanın amacı prostat kanseri kök hücreleri üzerine zoledronik asit uygulaması sonrası, metastaz geliştirme sürecinde önemli rolü olan adezyon molekülleri üzerine etkisinin incelenmesidir. Bu amaçla DU145 insan prostat kanseri hücre hattından akım sitometri cihazı ile CD133/CD44 yüzey belirteçleri kullanılarak izole edilen kanser kök hücreleri üzerine zoledronik asit tedavisi uygulanmıştır. Kanser kök hücresinde oluşan değişiklikler adezyon molekülleri yönü ile araştırılmıştır. Elde edilen sonuçlar zoledronik asit tedavisi sonrası kanser kök hücresi sayısında önemli bir düşüş olduğunu ve bu uygulamanın CD44, ITGB1, CD29, LAMB1, LAMB3, LAMC1, SPP1, TGFB1, TGFB1, TIMP2, ADAMTS1, ITGB5’de önemli değişimlere yol açtığını göstermiştir. Bu çalışmada in-vitro ortamda zoledronik asit uygulamasının kanser kök hücresi adezyon molekülleri üzerine baskılayıcı etki oluşturduğu ve ilerleyen çalışmalarda bu ilacın klinik kullanımda prostat kanseri tedavisinde uygulanabilme olasılığının olduğunu göstermiştir. Although the diagnosis and treatment of prostate cancer has made a progress, the disease remains fatal for some cases. Every step taken to save patients from death helps to approach the goal but there are still many issues that need to be investigated to reach a result. With the discovery of stem cells, the importance of these cells for human health has been understood and studies on how to use them in treatment have gained great momentum. In the following years, the concept of Cancer Stem Cell has emerged. It has been shown that these cells have stem cell characteristics but metastasize within the tumor tissue. They can cause recurrence after treatment or develop resistance to treatment. The other group of cells with rootedness is the non-cancer stem cell group and are cancer cells that respond to conventional cancer treatment. Adhesion molecules are of great importance in the cancer metastasis and invasion into the surrounding tissue. Studies have shown that adhesion molecules has great importance spesifically in multi-drug resistance and epithelial-mesenchymal transition. The aim of this study is to examine the effect of zoledronic acid application on prostate cancer stem cells on adhesion molecules, which have an important role in the metastasis development process. For this purpose, zoledronic acid treatment was applied to cancer stem cells isolated from DU145 human prostate cancer cell line using a flow cytometry device and CD133/CD44 surface markers. Changes in cancer stem cells have been investigated in terms of adhesion molecules. The results showed that there was a significant decrease in the number of cancer stem cells after zoledronic acid treatment and this application led to significant changes in CD44, ITGB1, CD29, LAMB1, LAMB3, LAMC1, SPP1, TGFB1, TGFB1, TIMP2, ADAMTS1, ITGB5. In this study, it has been shown that zoledronic acid administration in-vitro has a suppressive effect on cancer stem cell adhesion molecules and that this drug may be used in the treatment of prostate cancer in clinical use in further studies.

Amaç: Faz 1 çalışmalar aday ilacın ilk defa insan ile karşılaştığı çalışmalar olması nedeni ileönemlidir. Ülkemizde yazım tarihi itibari ile 12 adet onaylı Faz 1 merkezi bulunmaktadır. Biyoteknolojikilaçlar tüm dünyada başarı göstermelerin yanı sıra çok ciddi bir tedavi maliyetine nedenolabilmektedirler. Patent koruma sürelerinin sonlanması ile biyobenzer ilaçlar aynı pazarda yer bularak hem tedavi maliyetlerinde azalmaya hem de daha fazla kişinin ilacı ulaşabilmesine sonuç vermektedir.Gereç ve Yöntem: Bu yazıda 01.01.2017-01.06.2022 yıllarında dünyada biyobenzer ilaçlar ile yapılmış olan Faz 1, Faz 1, Faz 1-2 klinik çalışmaların sistematik değerlendirilmesi yapılmıştır. Son 65 aya ait olan bu veriler başvuru sayıları, çalışma süreleri, gönüllü sayıları ve sağlıklı gönüllü sayıları açısından ilişkilendirilerek değerlendirilmiştir.Bulgular: 01.01.2017-01.06.2022 tarihleri arasında toplamda kayıtlı 87 biyobenzer çalışmasında 7746 gönüllü ile çalışılmıştır. Bu gönüllülerin 4906’sı (%76,08) sağlıklı gönüllülerden oluşmaktadır. Tamamlanmış olan çalışmaların ortalama süresi 293,2 gün ve bu çalışmalara ait ortalama gönüllü sayısı 155,6’dır.Sonuç: Sonuç olarak, biyobenzer ürünler ile yapılan karşılaştırmalı Faz 1 çalışmalar yüksek sayıda sağlıklı gönüllü katılımı gerektirebilen, uzun süreli, her basamağının detaylı olarak planlanması gereken çalışmalardır. Patent sürelerinin sonlanması önümüzdeki dönemde biyobenzer çalışmaları için yeni merkezlerin aranması ihtiyacını doğuracaktır., Aim: Phase 1 studies are important because they are the first studies where the candidate drug is encountered with humans. As of the date of writing, there are 12 approved Phase 1 centers in our country. Bio-technological drugs are not only successful all over the world, but they can cause a very serious treatment cost. With the expiration of patent protection periods, biosimilar drugs find a place in the same market, resulting in both a reduction in treatment costs and more people accessing the drug.Materials and Methods: In this article, a systematic evaluation of Phase 1, Phase 1, and Phase 1-2 clinical studies conducted with biosimilar drugs in the world between 01.01.2017-01.06.2022 was studied. These data belonging to the last 65 months were evaluated in terms of the number of applications, study duration, number of volunteers and the number of healthy volunteers.Results: Between 01.01.2017 and 01.06.2022, 7746 volunteers were studied in 87 registered biosimilar studies. 4906 (76.08%) of these volunteers were healthy volunteers. The average duration of the completed studies is 293.2 days and the average number of volunteers for these studies is 155.6. Conclusion: As a result, comparative Phase 1 studies with biosimilar products are long-term studies that require the participation of a high number of healthy volunteers, and each step must be planned in detail. The expiration of patent periods will necessitate the search for new centers for biosimilar studies in the coming period.

Amaç: Deneysel olarak sıçanlarda Streptozotosin ile oluşturulmuş diyabet modelinde bir mitokondri spesifik antioksidan olan Mitotempo’nun endotelial ve erektil disfonksiyona etkisinin incelenmesi amaçlanmıştır.Gereç ve Yöntem: Deney için 280-320 g ağırlığında Wistar-Albino soyu sıçanlar kullanılmıştır. Diyabet grubuna 50 mg/kg intraperitoneal tek doz streptozotosin uygulanmıştır. 1 hafta sonunda 300 mg/dl kan glikozu seviyesinin üstünde olanlar diyabet kabul edilmiştir. 4 hafta boyunca kan glikozu takip edilmiştir. Bu 4 hafta boyunca tedavi grubuna 0.7 mg/kg/gün oral mitotempo uygulanmıştır. 4. Hafta sonunda dekapite edilen hayvanların aort ve korpus kavernozum dokuları alınarak kasılma ve gevşeme yanıtları izole organ banyosunda değerlendirilmiştir.Bulgular: Çalışma sonuçlarımıza göre diyabet modelinde mitotemponun 0.7 mg/kg/gün dozunda 4 hafta süre ile uygulanması hem torasik aorta hem de korpus kavernosum endotel yanıtlarını korumuştur. KCl kasılmasına göre rölatif olarak değerlendirilen fenilefrin kasılma yanıtları gruplar arasında farklılık göstermemiştir.Sonuç: Endotel hücreleri, dolaşımdaki maddelere ilk maruz kalan organlardan biri olarak tanımlanabilir. Hipergliseminin neden olduğu endotel disfonksiyonda mROS’un etkisi bilinmektedir. Diyabete bağlı oluşan endotel disfonksiyonun engellenebilmesi hastalığa bağlı mortalite ve morbiditenin önüne geçebilir. Çalışmamızda mitotemponun diyabetik endotelial disfonksiyon üzerine olası etkileri değerlendirilmiş ve sınırlı koruyucu etkileri gösterilmiştir., Objective: In this study we aimed to evaluate the effect of mitotempo, a mitochondria-specific antioxidant, on endothelial and erectile dysfunction in a Streptozotocin-induced rat diabetes model.Material and Methods: Wistar Albino rats weighing 280-320 g were used in the study. Diabetes was induced by intraperitoneal 50 mg/kg single dose streptozotocin injection. Blood glucose levels above 300 mg/dl at the end of 1 week were considered diabetes. Blood glucose was monitored for 4 weeks. The treatment group received mitotempo orally at 0.7 mg/kg/ day for 4 weeks. At the end of the 4th week, the aorta and corpus cavernosum contraction and relaxation responses were evaluated in an isolated organ bath after decapitation.Results: According to our study results, mitotempo 0.7 mg/kg/day for 4 weeks in a diabetes model preserved endothelial relaxation responses in both the thoracic aorta and corpus cavernosum. Phenylephrine contractions calculated according to KCl contraction did not differ between the groups.Conclusion: Endothelial cells can be identified as one of the first organs to be exposed to circulating substances. The effects of mROS on endothelial dysfunction caused by hyperglycemia is known. In our study, we found that a 0.7 mg/kg/day mitotempo treatment for 4 weeks showed protective effects on STZ-induced diabetic endothelial dysfunction.

Background/aim: In this study, the e ects of resveratrol as a natural polyphenol compound, gemcitabine as an antimetabolite that has nucleoside structure analogous to deoxycytidine, and para-aminophenol-derived paracetamol were investigated with single and combined applications in monolayers of the MDAH-2774 human ovarian cancer cell line. Materials and methods: Drugs were evaluated in cell culture with respect to cell proliferation, cell cytotoxicity (trypan blue dye exclusion test), synthesis phase of cell cycle, and cell structure in 24, 48, 72, and 96 h. Result: Resveratrol and gemcitabine diminished both cell proliferation and cell cycle synthesis phase indication in monolayer cell cultures (P < 0.05). All combination groups showed similar e ects that were mainly more e ective in respect to single usage of resveratrol and gemcitabine in monolayer cell cultures. Conclusion: The effects of gemcitabine, resveratrol, and paracetamol were investigated in monolayers of the MDAH-2774 human ovarian cancer cell line and a decrease in cell number in cell cycle synthesis phase, prevention of cell proliferation, and destruction of cell structure were observed.

Farmakogenetik (PGx), genetik varyasyonlar ve bunların bireyler arasında oluşturduğu ilaç yanıtı farklılıkları ile ilgilenir. İlaç metabolizmasından sorumlu enzimlerin keşfi ve bu enzimleri kodlayan DNA dizilimlerinin araştırılması bireysel tedavi stratejilerinin oluşturulmasını sağlar. İlaç metabolizmasından sorumlu sitokrom 2B6, sitokrom 2C9, sitokrom 2C19, sitokrom 2D6, sitokrom 3A4, N-asetil transferaz, dihidropirimidin dehidrogenaz, tiopurin metiltransferaz, 5’-difosfat (UDP)-glukuronoziltransferaz ve katekol-O-metil transferaz enzim polimorfizleri ilacın farmokokinetik özelliğini etkileyerek bireyler arası ilaç yanıtı farklılıklarına neden olabilirler. PGx çalışmaların temelini oluşturan ilk örnekler N- asetil transferaz ve sitokrom 2D6 polimorfizmleridir. İlaç seçiminde ciddi farmakokinetik farklılıklara neden olan enzim polimorfizmlerinin göz önünde bulundurulması tedavi başarısını artırmak ve advers/toksik reaksiyon riskini önlemek açısından önemlidir. Yaklaşık 50 yıl önce temelleri atılan farmakogenetik ile ilgili çalışmalar gen teknolojisinin gelişmesi ile günümüzde daha önemli bir hale gelmiştir. Teknolojik gelişmeler sayesinde hızlanan farmakogenetik çalışmalar ile bireye özgü ilaç seçimi farmakogenetiğin ikinci 50 yılı içerisinde daha fazla gelişme kaydederek önemli bir parametre olacaktır., Pharmacogenetics deals with genetic variations and individual response differences of drugs . The discovery of enzymes responsible for drug metabolism and the research to DNA sequences encoding these enzymes enable the creation of individual treatment strategies. Cytochrome 2B6, cytochrome 2C9, cytochrome C19, cytochrome 2D6, cytochrome 3A4, N-acetyltransferase, dihydropyrimidine dehydrogenase, thiopurine methyltransferase, UDP-glucuronyl transferase and catechol-O-methyltransferase enzymes are mainlyresponsible from drug metabolism and polymorphisms in enzyme activities affect the pharmacokinetic properties of the drug which leads to differences in drug response between individuals. First examples that form of the basis of pharmacogenetic studies are N-acetyltransferase and cytochrome 2D6 polymorphisms. Consideration of enzyme polymorphisms that may result with pharmacokinetic differences during drug choice is important to increase the success of treatment and to prevent adverse/toxic reaction risk. The studies about pharmacogenetics, which was studied about 50 years ago, has become more important nowadays with the development of gene technology. Individual drug choice will be an important parameter by further development of pharmacogenetics in the second 50 years through pharmacogenetic studies that is accelerated due to technological developments.

Giriş: Öğrencilerin klasik eğitimle aldıkları temel tıp bilimleri bilgilerini klinik yıllara ve klinik pratiğe taşıyamadıkları ve probleme dayalı öğrenimin (PDÖ) bu eksiği kapatabileceği ifade edilmektedir. Teorik farmakoloji eğitimini tamamlamış olan üçüncü sınıf öğrencilerinin çocukluk çağı zehirlenmeleri hakkındaki bilgi düzeylerinin hazırlanmış PDÖ senaryosu ile ölçülmesi amaçlandı. Gereç ve Yöntem: Bu çalışmada 2009-2010 akademik yılının 2. yarıyılı ortasında 2. PDÖ senaryosu olarak uygulanan bir çocukluk çağı zehirlenme olgusu öncesinde ve sonrasında araştırmacılar tarafından hazırlanan bir anket formu rastgele örnekleme yöntemiyle seçilen PDÖ gruplarına isimsiz ve gönüllülük temelinde uygulandı. Demografik bilgiler, zehirlenme vakası karşısında tutumlarını ve bilgi düzeylerini ölçmeye yönelik açıkkapalı uçlu sorular ile öncelik sıralaması yapma şeklindeki ifadelerden oluşan anket formunun değerlendirilmesinde sayılar, yüzdeler ki-kare ve Student-t testi kullanıldı. Bulgular: Çalışma grubunda, ön testte 89 öğrenci, son testte 96 öğrenci yer aldı. Çalışmamızda amaçlananhiperkolesterolemik ortamda oral resveratrolün korpus kavernozum (KK) dokusunda,torakal aorta (TA), mezenterik arter (MA) ve renal arter (RA)'de endotel disfonksiyonuönleyici etkisini karşılaştırmalı olarak araştırmaktır.YÖNTEMGruplar 6 hafta süre ile normal yem, %2 a/a'lik kolesterol diyeti ve %2 a/akolesterol diyeti+4mg/kg/gün resveratrol ile beslenmiştir. Bu süre sonunda plazmakolesterol düzeyleri değerlendirilmiştir. RA, TA, MA ve KK dokuları alınarak izoleorgan banyosunda kümülatif dozlarda asetilkolin (ACh) ve sodyum nitroprussit (SNP)ile sırası ile endotel bağımlı ve endotel bağımsız yanıtları değerlendirilmiştir.BULGULARPlazma total kolesterol düzeyleri hiperkolesterolemik grup ve resveratrol grubuarasında anlamlı farklılık göstermezken; her 2 grup da kontrol grubu ile anlamlıfarklılık göstermiştir. Resveratrol+kolesterol grubunda ACh'e bağlı gevşeme yanıtlarıhiperkolesterolemik grup ve kontrol grubu ile anlamlı farklılık göstermiştir. Gruplararasında endotel bağımsız gevşeme yanıtlarında herhangi bir farklılıkgözlenmemiştir. %2 a/a 6 hafta süre ile hiperkolesterolemik diyet sonucu oluşanendotel bağımlı gevşeme yanıtları en fazla sırası ile MA>KK>TA>RA'de bozulmagöstermektedir. Tavşanların 6 hafta süre ile %2 kolesterol ile beraber 4mg/kg/gün5resveratrol ile beslenmeleri ACh yanıtlarında düzelmeye neden olmuştur. Budüzelme değerlendirildiğinde ise sırası ile en fazla MA>RA>KK>TA' da olmaktadır.SONUÇ4mg/kg/gün resveratrol ile beraber %2 kolesterol diyeti RA, TA, MA ve KKdokularını hiperkolesteroleminin endotel disfonksiyon oluşturucu etkilerindenkorumuştur. Hiperkolesterolemik ve resveratrol gruplarının plazma kolesteroldüzeyleri arasında herhangi bir anlamlı farklılık oluşmaması resveratrolün bukoruyucu etkisini plazma kolesterol düzeyinde bağımsız bir mekanizma ile yaptığınıgöstermektedir. In our study; we aimed to study and compare the protective effects ofresveratrol in hypercholesterolemia on thoracic aorta (TA), mesenteric artery (MA),renal artery (RA) and corpus cavernosum (CC).MATERIAL AND METHODSAnimals fed a %2 a/a cholesterol diet and %2 a/a high cholesteroldiet+resveratrol (4 mg/kg/day) for 6 week duration. Total cholesterol levels in theplasma were measured after the cholesterol challenge. Vascular and endothelialfunctions in RA, TA, MA and CC were assessed by isolated tissue bath withcumulative doses of acetylcholine and sodium nitroprusside.RESULTSEven there have been no significant changes on plasma total cholesterol levelsbetween cholesterol and cholesterol+resveratrol treated groups; both groups showedsignificant changes when compared with control group. Vasorelaxation responses toACh in resveratrol treated group showed significant changes when compared withhypercholesterolemia and control group. No statistically significant differences wereseen between endothelium independent vasorelaxation responses between threegroups. %2 a/a cholesterol diet for 6 weeks has diminished endothelium dependentvasorelaxation responses mostly on MA>CC>TA>RA respectively. Resveratrol hasproved endothelial responses when given with %2 a/a cholesterol for 6 weeks. Theprotective effect of resveratrol was seen most on MA>RA>KK>TA respectively.7CONCLUSIONThe results also provide support that 4mg/kg/day resveratrol can suppressendothelial dysfunction in hypercholesterolemic rabbits. Resveratrol is showing thisprotective effect without changing plasma total cholesterol levels. Teorik farmakoloji eğitimini tamamlamış olan üçüncü sınıf öğrencilerinin çocukluk çağı zehirlenmeleri hakkındaki bilgi düzeylerinin hazırlanmış PDÖ senaryosu ile ölçülmesi amaçlandı. Gereç ve Yöntem: Bu çalışmada 2009-2010 akademik yılının 2. yarıyılı ortasında 2. PDÖ senaryosu olarak uygulanan bir çocukluk çağı zehirlenme olgusu öncesinde ve sonrasında araştırmacılar tarafından hazırlanan bir anket formu rastgele örnekleme yöntemiyle seçilen PDÖ gruplarına isimsiz ve gönüllülük temelinde uygulandı. Demografik bilgiler, zehirlenme vakası karşısında tutumlarını ve bilgi düzeylerini ölçmeye yönelik açıkkapalı uçlu sorular ile öncelik sıralaması yapma şeklindeki ifadelerden oluşan anket formunun değerlendirilmesinde sayılar, yüzdeler ki-kare ve Student-t testi kullanıldı. Bulgular: Çalışma grubunda, ön testte 89 öğrenci, son testte 96 öğrenci yer aldı. Refik Saydam Hıfzıssıhha Ulusal Zehir Danışma Merkezi UZEM verilerine göre çok sık zehirlenme başvurusu yapılan maddelerden oluşturulan listeye verilen yanıtlarda örneğin; losyon, banyo köpüğü, kozmetikler, suluboya ve kalaminli losyonlar gibi öğrencilerin ön testte toksik dedikleri maddeler son teste anlamlı olarak doğru cevaplanmıştı p, Introduction: It is stated that students cannot carry their knowledge on basic medical sciences that they gained with classical education to clinical classes and clinical practice and problem based learning PBL can compensate this drawback. It was aimed to evaluate the effect of PBL scenario written on this topic on level of knowledge about childhood poisoning of third class students who completed the most of theoretical pharmacology education. Materials and Method: A questionnaire form prepared by researchers was applied to randomly chosen PBL groups without writing names and on the basis of voluntariness before and after a case of childhood poisoning which was applied as the second PBL scenario in the second midterm of 2009-2010 academic years. Numbers, percentages, chisquare and student’s t-test were used for evaluation of the questionnaire form comprised of demographic data, open-closed ended questions for measuring attitudes and level of knowledge against case of poisoning and statements as making a priority ranking. Results: In the study group, 89 students took part in pre-test and 96 students took part in post-test. Yüksek Lisans Tezi, Bu çalışmada streptozotosin ile indüklenen diyabetli sıçanlara 4 hafta boyunca 0.7 mg/kg oral gavaj yoluyla mitotempo uygulanarak diyabetin oluşturduğu endotelyal disfonksiyon ve erektil disfonksiyon üzerine etkileri incelenmiştir. Kontrol grubu, diyabet grubu ve diyabet + mitotempo uygulanmış olan gruplardan sakrifiye edildikten sonra elde edilen torasik aortlar ve korpus kavernosumlar 37 oC'de sabit tutulan ve %95 O2 + %5 CO2 karışımı ile gazlandırılan 10 ml Krebs-Henseleit (KHS) içeren organ banyolarına alındı. Uygulanan prosedürlere verilen cevaplar izometrik olarak kaydedildi. 10-6 M fenilefrin FE uygulanması ile doku kasılma yanıtlarında diyabetes mellitus (DM) grubu ile kontrol ve Mitotempo grupları arasında hem aort hem de KK yanıtları anlamlı bir fark görülmemiştir. Doku kasılmasının ardından aort ve KK dokularında 10-9 M ve 10 -5 M kümülatif Asetilkolin (Ach) uygulaması ile gevşeme yanıtları incelenmiştir. Ach kontrol grubunda ve diyabet + mitotempo grubunda diyabet grubuna kıyasla anlamlı olarak daha fazla gevşemeye neden olmuştur. Bu sonuçlar streptozotosin ile indüklenen diyabetli sıçanlarda mitotempo uygulamasının torasik aortlar ve korpus kavernosumlarda Ach ile gevşeme yanıtlarını artırarak diyabetin oluşturduğu endotelyal disfonksiyon ve erektil disfonksiyonda etkili olduğunu göstermektedir., The aim of this study was to investigate the effects of endothelial dysfunction and erectile dysfunction by administering mitotempo by oral gavage of 0.7 mg / kg for 4 weeks to streptozotocin-induced diabetes rats. After sacrificing the control group, the diabetic group and the diabetic and mitotempo treated groups, the obtained thoracic aortas and corpus cavernosums were taken containing 10ml Krebs-Henseleit (KHS), kept constant at 37° C and gasified with 95% O2 and %5 CO2 organ baths. Responses to the procedures were recorded isometric. There was no significant difference between DM and control and Mitotempo groups in the response to tissue contraction with 10-6 M phenylephrine FE. After tissue contraction, relaxation responses of aortic and KK tissues by using 10-9 M and 10 -5 M cumulative acetylcholine were examined. Ach caused significantly more relaxation in the control group and in the diabetes + mitotempo group than in the diabetes group. These results indicate that mitotempo administration in streptozotocininduced diabetic rats is effective in endothelial dysfunction and erectile dysfunction by increasing Ach relaxation responses in thoracic aorta and corpus cavernosum.
