Abstract

In this paper, we consider a free boundary problem modeling the growth of spherically symmetric necrotic tumors with angiogenesis and a $\omega$-periodic supply $\phi(t)$ of external nutrients. In the model, the consumption rate of the nutrient and the proliferation rate of tumor cells $S(\sigma)$ are both general nonlinear functions. The well-posedness and asymptotic behavior of solutions are studied. We show that if the average of $S(\phi(t))$ is nonpositive, then all evolutionary tumors will finally vanish; the converse is also ture. If instead the average of $S(\phi(t))$ is positive, then there exists a unique positive periodic solution and all other evolutionary tumors will converge to this periodic state., Comment: 14 pages

Abstract

This paper concerns a free boundary problem modeling tumor growth with angiogenesis and two time delays. The two delays represent the time taken for cells to undergo mitosis and modify the rate of cell loss because of apoptosis, respectively. We study the stability of stationary solutions and find that Hopf bifurcation occurs under some conditions, which extends the results of Xu. Furthermore, numerical simulations are performed to investigate the relationship among the rate of angiogenesis, two time delays and Hopf bifurcation., Comment: 14 pages, 6 figures

Abstract

We consider a free boundary tumor model under the presence of angiogenesis and time delays in the process of proliferation, in which the cell location is incorporated. It is assumed that the tumor attracts blood vessels at a rate proportional to $\alpha$, and a parameter $\mu$ is proportional to the `aggressiveness' of the tumor. In this paper, we first prove that there exists a unique radially symmetric stationary solution $\left(\sigma_{*}, p_{*}, R_{*}\right)$ for all positive $\alpha$, $\mu$. Then a threshold value $\mu_\ast$ is found such that the radially symmetric stationary solution is linearly stable if $\mu<\mu_\ast$ and linearly unstable if $\mu>\mu_\ast$. Our results indicate that the increase of the angiogenesis parameter $\alpha$ would result in the reduction of the threshold value $\mu_\ast$; adding the time delay would not alter the threshold value $\mu_\ast$, but result in a larger stationary tumor, and the larger the tumor aggressiveness parameter $\mu$ is, the greater impact of time delay would have on the size of the stationary tumor.

Abstract

This paper is concerned with a nonlinear free boundary problem modeling the growth of spherically symmetric tumors with angiogenesis, set with a Robin boundary condition. In which, both nonnecrotic tumors and necrotic tumors are taken into consideration. The well-posedness and asymptotic behavior of solutions are studied. It is shown that there exist two thresholds, denoted by $\tilde\sigma$ and $\sigma^*$, on the surrounding nutrient concentration $\bar\sigma$. If $\bar\sigma\leq\tilde\sigma$, then the considered problem admits no stationary solution and all evolutionary tumors will finally vanish, while if $\bar\sigma>\tilde\sigma$, then it admits a unique stationary solution and all evolutionary tumors will converge to this dormant tumor; moreover, the dormant tumor is nonnecrotic if $\tilde\sigma<\bar\sigma\leq\sigma^*$ and necrotic if $\bar\sigma>\sigma^*$. The connection and mutual transition between the nonnecrotic and necrotic phases are also given.

Abstract

We investigate a one-dimensional nonlinear wave system which arises from a variational principle modeling a type of cholesteric liquid crystals. The problem treated here is the Cauchy problem for the same wave speed case with initial data on the parabolic degenerating line. By introducing a partial hodograph transformation, we establish the local existence of smooth solutions in a weighted metric space based on the iteration method. A classical solution of the primary problem is constructed by converting the solution in the partial hodograph variables to that in the original variables.

Abstract

In this paper, we present a rigorous mathematical analysis of a free boundary problem modeling the growth of a vascular solid tumor with a necrotic core. If the vascular system supplies the nutrient concentration $\sigma$ to the tumor at a rate $\beta$, then $\frac{\partial\sigma}{\partial\bf n}+\beta(\sigma-\bar\sigma)=0$ holds on the tumor boundary, where $\bf n$ is the unit outward normal to the boundary and $\bar\sigma$ is the nutrient concentration outside the tumor. The living cells in the nonnecrotic region proliferate at a rate $\mu$. We show that for any given $\rho>0$, there exists a unique $R\in(\rho,\infty)$ such that the corresponding radially symmetric solution solves the steady-state necrotic tumor system with necrotic core boundary $r=\rho$ and outer boundary $r=R$; moreover, there exist a positive integer $n^{**}$ and a sequence of $\mu_n$, symmetry-breaking stationary solutions bifurcate from the radially symmetric stationary solution for each $\mu_n$ (even $n\ge n^{**})$., Comment: 29 pages

Abstract

Tissue repair is an extremely crucial part of clinical treatment. During the course of disease treatment, surgery, chemotherapy, and radiotherapy cause tissue damage. On the other hand, Normal tissue from accidental or therapeutic exposure to high-dose radiation can cause severe tissue damage. There is an urgent need for developing medical countermeasures against radiation injury for tissue repair. Tissue repair involves the regeneration, proliferation, differentiation, and migration of tissue cells; imbalance of local tissue homeostasis, progressive chronic inflammation; decreased cell activity and stem cell function; and wound healing. Although many clinical treatments are currently available for tissue repair, they are expensive. The long recovery time and some unavoidable complications such as cell damage and the inflammatory reaction caused by radiotherapy have led to unsatisfactory results. Extracellular vesicles (EVs) derived from mesenchymal stem cells (MSCs) have similar tissue repair functions as MSCs. In tissue damage, EVs can be used as an alternative to stem cell therapy, thereby avoiding related complications such as immunological rejection. EVs play a major role in regulating tissue damage, anti-inflammation, pro-proliferation, and immune response, thus providing a diversified and efficient solution for the repair of disease- and radiotherapy-induced tissue damage. This article reviews the research progress of mesenchymal stem cell-derived EVs in promoting the repair of tissue including heart, lung, liver, intestine, skin, blood system, central nervous system, and tissue damage caused by radiotherapy, thereby aiming to offer new directions and ideas for the radiotherapy and regenerative applications. Abstract

The safety and efficacy of the lipid nanoparticle (LNP) delivery system are crucial for the successful development of messenger RNA vaccines. We designed and synthesized a series of ketal ester lipids (KELs), featuring a biodegradable ketal moiety in the linker and ester segments in the tail. Through iterative optimization of the head and tail groups of KELs, we tuned the pKa and molecular shapes, and identified (4S)‐KEL12 as a safe and efficient ionizable lipid for mRNA delivery. (4S)‐KEL12 LNP showed significantly higher delivery efficacy and lower toxicity than the DLin‐MC3‐DMA LNP. In comparison to SM‐102 LNP, (4S)‐KEL12 LNP exhibited better spleen tropism, reduced liver tropism, and hepatotoxicity. Additionally, (4S)‐KEL12 demonstrated good biodegradability following intramuscular or intravenous injection. Notably, (4S)‐KEL12 LNP encapsulated with a therapeutic mRNA cancer vaccine elicited robust cellular immune responses leading to substantial tumor regression along with prolonged survival in tumor‐bearing mice. Our results suggest that (4S)‐KEL12 LNP holds great promise for mRNA vaccine delivery. The comprehensive analysis of the structure‐activity relationship, toxicity, biodegradability, distribution, expression, efficacy, and stereochemistry of these LNPs will greatly contribute to the rational design and discovery of novel lipid‐based delivery systems.
