Chronic stress and elevated levels of glucocorticoids (GCs), the main stress hormones, accelerate Alzheimer’s disease (AD) onset and progression. A major driver of AD progression is the spreading of pathogenic Tau protein between brain regions, precipitated by neuronal Tau secretion. While stress and high GC levels are known to induce intraneuronal Tau pathology ( i.e. hyperphosphorylation, oligomerization) in animal models, their role in trans-neuronal Tau spreading is unexplored. Here, we find that GCs promote secretion of full-length, vesicle-free, phosphorylated Tau from murine hippocampal neurons and ex vivo brain slices. This process occurs via type 1 unconventional protein secretion (UPS) and requires neuronal activity and the kinase GSK3β. GCs also dramatically enhance trans-neuronal Tau spreading in vivo , and this effect is blocked by an inhibitor of Tau oligomerization and type 1 UPS. These findings uncover a potential mechanism by which stress/GCs stimulate Tau propagation in AD.

Το Καθιερωμένο Πρότυπο (Standard Model - SM), παρά την αδιαμφισβήτητη επιτυχία του στην περιγραφή των δομικών συστατικών της ύλης και των μεταξύ τους αλληλεπιδράσεων, αφήνει αναπάντητα σημαντικά ερωτήματα, με αποτέλεσμα να έχουν προταθεί πολλές νέες θεωρίες προκειμένου να απαντήσουν ορισμένα από αυτά. Μία από αυτές είναι η Υπερσυμμετρία (Supersymmetry – SUSY), η οποία αποτελεί προέκταση του Καθιερωμένου Προτύπου, προβλέποντας για κάθε SM σωμάτιο τον υπερσυμμετρικό του εταίρο με σπιν μειωμένο κατά ħ/2. Σε κάποια SUSY μοντέλα, στα οποία προβλέπεται η διατήρηση μίας νέας συμμετρίας, της R - parity, το ελαφρύτερο υπερσυμμετρικό σωματίδιο (Lightest Supersymmetric Particle – LSP), το οποίο ονομάζεται neutralino - 1 και είναι υποψήφιο για τη σύσταση της σκοτεινής ύλης, πρέπει να είναι σταθερό. Έτσι, μια επιβεβαίωση της ύπαρξης των SUSY σωματιδίων και ειδικότερα του LSP, θα ήταν υψίστης σημασίας. Από πειραματικής άποψης, περισσότερο ενδιαφέρον παρουσιάζουν οι αλυσίδες διάσπασης, στις οποίες η διαφορά μάζας μεταξύ των μητρικών (π.χ. neutralino - 2, chargino - 1) και των θυγατρικών (LSPs) SUSY σωματιδίων είναι πολύ μικρή, σενάρια τα οποία αναφέρονται ως compressed. Τη δυσκολία σε αυτά, όμως, συνιστά το γεγονός ότι η ορμή των τελικών προϊόντων μειώνεται δραματικά, ομοίως και η ελλείπουσα εγκάρσια ορμή του γεγονότος, με συνέπεια η διάκριση αυτών των διασπάσεων από το σύνολο των γνωστών αλληλεπιδράσεων (SM background) να καθίσταται μεγάλη πρόκληση. Ωστόσο, όπως θα δούμε, η δυσκολία αυτή μπορεί να μετριαστεί σε περιπτώσεις όπου το σύστημα των μητρικών SUSY σωματιδίων ανακρούεται από μία ή περισσότερες δέσμες γλουονίων (Initial State Radiation jets – ISR jets). Επιπλέον δυσκολία αποτελεί το γεγονός ότι, αν παραχθούν σε έναν επιταχυντή αδρονίων, τα LSPs διαφεύγουν χωρίς να ανιχνευτούν, όπως και τα SM νετρίνα. Ως αποτέλεσμα, προκύπτουν κινηματικές και πιθανώς και συνδυαστικές ασάφειες που δεν μας επιτρέπουν να λάβουμε πληροφορίες για όλες τις ιδιότητες των ενδιάμεσων σωματιδίων της αλυσίδας διάσπασης. Η λύση στο παραπάνω πρόβλημα δίνεται μέσω μιας μεθόδου που ονομάζεται Recursive Jigsaw Reconstruction (RJR). Αυτή η μέθοδος χρησιμοποιεί ένα συνδυασμό αλγορίθμων-κανόνων, γνωστών ως Jigsaw Rules (JR’s), οι οποίοι εφαρμόζονται αναδρομικά και καταφέρνουν να επιλύσουν με μεγάλη επιτυχία τις ασάφειες που εμφανίζονται σε κάθε διεργασία. Στην παρούσα πτυχιακή εργασία, κάνοντας χρήση του λογισμικού πακέτου RestFrames, θα εφαρμόσουμε τη μέθοδο RJR στην περίπτωση της εξής αντίδρασης: neutralino-2 chargino-1 -> Z(ll) neutralino-1 W(lν) neutralino-1. Στόχος μας είναι η ορθή παραγωγή και ανακατασκευή της, καθώς και ο υπολογισμός - μελέτη της κινηματικής ποσότητας R_ISR, η οποία είναι "ευαίσθητη" στην παρουσία βαρέων LSPs και κατάλληλη να χρησιμοποιηθεί για την αναζήτηση σημάτων της συγκεκριμένης αντίδρασης, αλλά και σημάτων από compressed SUSY τοπολογίες γενικότερα. The Standard Model (SM), despite its undeniable success in describing the structural components of matter and the interactions between them, leaves important questions unanswered, resulting in many new theories being proposed to answer some of them. One of these is Supersymmetry (SUSY), which is an extension of the Standard Model, predicting for each SM particle its supersymmetric partner with a spin reduced by ħ/2. In some SUSY models, in which the conservation of a new symmetry, R - parity, is considered, the Lightest Supersymmetric Particle (LSP), which is called neutralino - 1 and is a candidate for the composition of dark matter, must be stable. Thus, a confirmation of the existence of SUSY particles, and in particular of LSP, would be of utmost importance. From an experimental point of view, of more interest are the decay chains, in which the mass difference between the parent (e.g. neutralino - 2, chargino - 1) and the daughter (LSPs) SUSY particles is very small, scenarios which are referred to as being compressed. The difficulty in these, however, is the fact that the momentum of the final products decreases dramatically, as does the missing transverse momentum of the event, with the consequence that distinguishing these decays from all known interactions (SM background) becomes a great challenge. However, as we will see, this difficulty can be mitigated in cases where the system of the parent SUSY particles recoils against one or more gluon beams (Initial State Radiation jets - ISR jets). An additional difficulty is the fact that, if produced at a hadron collider, LSPs escape undetected, as do SM neutrinos. As a result, kinematic and possibly combinatoric ambiguities arise that do not allow us to obtain information about all the properties of intermediate particles of the decay chain. The solution to the above problem is given through a method called Recursive Jigsaw Reconstruction (RJR). This method uses a combination of algorithms-rules, known as Jigsaw Rules (JR’s), which are applied recursively and manage to successfully resolve the ambiguities that occur in each process. In this thesis, using the RestFrames software package, we will apply the RJR method in the case of the following reaction: neutralino-2 chargino-1 -> Z(ll) neutralino-1 W(lν) neutralino-1. Our goal is its correct production and reconstruction, as well as the calculation - study of the kinematic quantity R_ISR, which is "sensitive" to the presence of massive LSPs and suitable to be used to search for signals of this specific reaction, but also signals of compressed SUSY topologies in general.

The current article attempts to investigate the causal relationship between human capital and economic growth in postwar Greece. For this purpose, we use annual time series data from 1952 to 2017 retrieved from the Penn World Table (PWT) version 9.1 and we apply the Toda –Yamamoto (1995) approach. Our findings indicate a one-way causality that runs fromhuman capital to economic growth. This is consistent with proposals that the Greek government, despite budget cuts due to the recent fiscal financial crisis, should focus its efforts on the development of human capital in order to achieve sustainable economic growth. Objective: The suggested link between major depression disorder (MDD) and blood–brain barrier (BBB) alterations supports an impact on the neurovascular unit in this disease condition. Here we investigate how pericytes, a major component in the neurovascular unit, respond to stress, stress hormones, proinflammatory cytokine and depression. Method: Hippocampal sections of chronic unpredictable stressed (CMS) rats, MDD patients and respective controls were immuno-stained against NG2, where the number of NG2+ pericytes in the molecular layer was counted. Proliferation of cultured pericytes after treatment with cortisol and IL-1β was analyzed using radioactive-labeled thymidine. Findings: The number of NG2+ pericytes was significantly higher in CMS animals than controls. Higher number of NG2+ pericytes was also detected in MDD patients, but the increase did not reach significance. IL-1β, but not cortisol, induced a significant increase in proliferation of cultured pericytes. Conclusion: Our results indicate that exposure to stressful conditions affects the hippocampal pericyte population. These findings add to our knowledge about the impact of stress on the neurovascular unit, which might be relevant for understanding the alterations in BBB found in MDD patients.
