ilustraciones, fotografías, gráficas, tablas La marchitez vascular causada por Fusarium oxysporum f. sp. lycopersici (Fol) es una de las enfermedades más predominante y destructivas en el cultivo del tomate. Su control ha sido ampliamente dependiente del uso de plaguicidas. Una de las alternativas más promisorias para el manejo de la enfermedad es el uso de sustancias bioactivas, como aquellas basadas en fosfitos y silicios. En este trabajo, se propuso establecer un método de inoculación efectivo para el modelo Tomate-Fol, además de evaluar el efecto de fosfitos y silicio en la reducción de la severidad ocasionada por Fol a nivel in vitro y en planta. 119 aislamientos fueron obtenidos de cuatro regiones productoras. Se realizó el análisis comparativo de tres métodos de inoculación y se estandarizó un protocolo para la infección de plántulas. A nivel in vitro, se observó una disminución del área de crecimiento del patógeno con sustancias bioactivas a concentraciones específicas. En las evaluaciones in planta, las plantas tratadas con fosfitos y silicio minimizaron la severidad hasta en un 42%. La información generada constituye un insumo para el entendimiento del modelo Tomate-Fol, además provee insumos para nuevas investigaciones en el uso del método de inoculación para otras solanáceas, además del uso de sustancias bioactivas, como fosfitos y silicios que permitan mitigar el efecto negativo del patógeno Fusarium en suelos y plantas de otros cultivos. (Texto tomado de la fuente). Tomato is one of the most important vegetable crops worldwide. However, its production is severely affected by diseases caused by pathogens. Among these, the vascular wilt caused by the formae speciales of the fungus Fusarium oxysporum f. sp. lycopersici (Fol) is responsible for substantial yield losses up to 80% in severe epidemics. Due to the constant risk of developing fungicide resistance and environmental contamination, alternative methods such as the use of resistance inducers and bioactive substances should be studied. In this project, we wanted to establish an effective inoculation method for the tomato-Fol pathosystem. Them, to evaluate the effect of two bioactive substances, phosphites and silicon, on Fol using and in vitro analysis, and to determine the effect of such bioactive substances in the reduction of severity caused by Fol in tomato plants. 119 fungal isolates were obtained in four different departments of the Central Andean Region in Colombia, from tomato crops with disease wilting symptoms. A comparative analysis of three inoculation methods: soil suspension (SS), root dipping (RD) and SS-RD combination, was performed infecting tomato seedlings (two, three and four weeks old), from the cultivar Santa Cruz Kada (susceptible to all three races of Fol), with the Colombian isolates UDCFOL10 and Fol59. A protocol for the infection of tomato seedlings with Fol59 was standardized. At the in vitro level, a decrease of Fol growth was observed after treatment with bioactive substances at specific concentrations. In in plant evaluations, plants treated with phosphites up to 42%. The knowledge generated in this study will contribute to the understanding of bioactive substances, such as phosphites and silicon, as alternative methods to control vascular wilt that could mitigate the negative effect of Fusarium on tomato crops. Su manejo se basa principalmente en el uso de fumigantes químicos, con un alto impacto sobre el ambiente y la salud humana; por lo que el tratamiento con inductores de resistencia como quitosán y Trichoderma spp., es una alternativa con alto potencial en agricultura porque actúan directamente sobre el patógeno e inducen la defensa en la planta. El objetivo de este trabajo fue identificar un aislamiento de Fol para evaluar la respuesta fisiológica de plantas de tomate infectadas y tratadas con quitosán y Trichoderma spp., como alternativas de manejo de la enfermedad. Se caracterizó un aislamiento patogénico y altamente virulento de Fol raza 2. Se evaluó la actividad controladora in vitro e in planta de quitosán y Trichoderma spp. El quitosán de bajo peso molecular a 2,5 mg.mL-1, redujo la marchitez vascular en un 70% en plantas y disminuyó el daño que ocasiona Fol sobre Fv/Fm, Y(II) y el contenido de clorofila a, en 22%, 35% y 39% respectivamente; y activó la respuesta de defensa de la planta a través del gen PR1a. Este estudio aporta al conocimiento de las alteraciones ocasionadas por Fol y el efecto del tratamiento con quitosán sobre los procesos fisiológicos y moleculares en la planta de tomate, como una alternativa confiable para mitigar el daño en la planta. Este es uno de los primeros esfuerzos en Colombia para identificar y caracterizar molecularmente un aislamiento virulento de Fol. (Texto tomado de la fuente). Fusarium oxysporum f sp. lycopersici (Fol) is the causal agent of tomato wilt disease, a high importance disease causing severe physiological disorders in the plant. Its presence has not been completely described in Colombia and its control is mainly based on chemical management that is hazardous for the environment and human health. Plant resistance induction by treatment with elicitors such as Chitosan and Trichoderma spp., that improve the plant physiology, is a promising management strategy. The aim of this work was to identify an isolate of Fol and to evaluate the physiological response of tomato plants infected with Fol and treated with chitosan and Trichoderma spp., as alternatives for the management of the disease. A highly virulent Colombian isolate of Fol (pathogenic race 2) was obtained and morphologically and molecularly characterized. The control activity in vitro and in planta of chitosan and Trichoderma spp. was also evaluated. Chitosan at low molecular weight (2.5 mg.mL-1), reduced vascular wilt by 70% in plants and decreased damage caused by Fol on Fv/Fm, Y(II) and chlorophyll a content, in 22%, 35% and 39% respectively; and activated the defense response of the plant through the PR1a gene. This study offers knowledge of the alterations caused by Fol and the effect of treatment with elicitors on the physiological and molecular processes in the tomato plant. Chitosan is a reliable and consistent alternative for the mitigation of the damage caused by this disease. In addition, it is one of the first efforts in Colombia to identify and molecularly characterize a virulent isolation of Fol.
