IntroductionFollowing early cell specification and tenocyte differentiation at the sites of future tendons, very little is known about how tendon maturation into robust load-bearing tissue is regulated. Between embryonic day (E)16 and E18 in the chick, there is a rapid change in mechanical properties which is dependent on normal embryo movement. However, the tissue, cellular and molecular changes that contribute to this transition are not well defined.MethodsHere we profiled aspects of late tendon development (collagen fibre alignment, cell organisation and Yap pathway activity), describing changes that coincide with tissue maturation. We compared effects of rigid (constant static loading) and flaccid (no loading) immobilisation to gain insight into developmental steps influenced by mechanical cues.ResultsWe show that YAP signalling is active and responsive to movement in late tendon. Collagen fibre alignment increased over time and under static loading. Cells organise into end-to-end stacked columns with increased distance between adjacent columns, where collagen fibres are deposited; this organisation was lost following both types of immobilisation.DiscussionWe conclude that specific aspects of tendon maturation require controlled levels of dynamic muscle-generated stimulation. Such a developmental approach to understanding how tendons are constructed will inform future work to engineer improved tensile load-bearing tissues.

Biotoxins produced by harmful algal blooms (HABs) are a substantial global threat to ocean and human health. Domoic acid (DA) is one such biotoxin whose negative impacts are forecasted to increase with climate change and coastal development. This manuscript serves as a review of DA toxicosis after environmental exposure in humans and wildlife, including an introduction to HAB toxins, the history of DA toxicosis, DA production, toxicokinetic properties of DA, susceptibility, clinical signs, DA detection methods and other diagnostic tests, time course of toxicosis, treatment, prognostics, and recommendations for future research. Additionally, we highlight the utility of California sea lions (CSLs; Zalophus californianus) as a model and sentinel of environmental DA exposure.

A framework is presented to quantify the objective-level resilience of reinforced concrete liners of circular tunnels when exposed to enclosed vehicle fire hazards. By assessing the loss of functionality due to fire-induced damage, the framework enables a decision-basis evaluation of the efficiency of various fire mitigation methods for specific tunnel conditions. In this study, the fire-induced damage of concrete tunnel liners due to strength loss and spalling is stochastically simulated and classified based on typical post-fire repair procedures and damage evaluation. The resilience assessment is conducted using Monte Carlo Simulation in combination with a fast-running tool for calculating the thermal impact from vehicle fires on the inside surface of the tunnel liner (developed by the authors in previous work). The proposed approach accounts for uncertainties associated with both the vehicle fire (particularly the combustion energy) and the tunnel conditions (i.e., geometry, dimensions, and the presence of longitudinal ventilation and/or fixed fire-fighting systems (FFFS)). A parametric case study is used to quantitatively demonstrate the effectiveness of FFFS for reducing post-fire losses of tunnel functionality. Other parameters such as tunnel dimensions, traffic restrictions for vehicles with heavy fire hazard risk, and installation or upgrade of the tunnel ventilation system show somewhat less effectiveness for reducing fire-induced damage.

Therapeutic benefit to immune checkpoint blockade (ICB) is currently limited to the subset of cancers thought to possess a sufficient tumor mutational burden (TMB) to allow for the spontaneous recognition of neoantigens (NeoAg) by autologous T cells. We explored whether the response to ICB of an aggressive low-TMB squamous cell tumor could be improved through combination immunotherapy using functionally defined NeoAg as targets for endogenous CD4+ and CD8+ T cells. We found that, whereas vaccination with CD4+ or CD8+ NeoAg alone did not offer prophylactic or therapeutic immunity, vaccines containing NeoAg recognized by both subsets overcame ICB resistance and led to the eradication of large established tumors that contained a subset of PD-L1+ tumor-initiating cancer stem cells (tCSC), provided the relevant epitopes were physically linked. Therapeutic CD4+/CD8+ T cell NeoAg vaccination produced a modified tumor microenvironment (TME) with increased numbers of NeoAg-specific CD8+ T cells existing in progenitor and intermediate exhausted states enabled by combination ICB-mediated intermolecular epitope spreading. We believe that the concepts explored herein should be exploited for the development of more potent personalized cancer vaccines that can expand the range of tumors treatable with ICB.

Abstract Infection is the most common cause of mortality early in life, yet the broad potential of immunization is not fully realized in this vulnerable population. Most vaccines are administered during infancy and childhood, but in some cases the full benefit of vaccination is not realized in-part. New adjuvants are cardinal to further optimize current immunization approaches for early life. However, only a few classes of adjuvants are presently incorporated in vaccines approved for human use. Recent advances in the discovery and delivery of Toll-like receptor (TLR) agonist adjuvants have provided a new toolbox for vaccinologists. Prominent among these candidate adjuvants are synthetic small molecule TLR7/8 agonists. The development of an effective infant Bordetella pertussis vaccine is urgently required because of the resurgence of pertussis in many countries, contemporaneous to the switch from whole cell to acellular vaccines. In this context, TLR7/8 adjuvant based vaccine formulation strategies may be a promising tool to enhance and accelerate early life immunity by acellular B. pertussis vaccines. In the present study, we optimized (a) the formulation delivery system, (b) structure, and (c) immunologic activity of novel small molecule imidazoquinoline TLR7/8 adjuvants towards human infant leukocytes, including dendritic cells. Upon immunization of neonatal mice, this TLR7/8 adjuvant overcame neonatal hyporesponsiveness to acellular pertussis vaccination by driving a T helper (Th)1/Th17 biased T cell- and IgG2c-skewed humoral response to a licensed acellular vaccine (DTaP). This potent immunization strategy may represent a new paradigm for effective immunization against pertussis and other pathogens in early life.

CD4+ T cells play a critical role in antitumor immunity via recognition of peptide antigens presented on MHC class II (MHC-II). Although some solid cancers can be induced to express MHC-II, the extent to which this enables direct recognition by tumor-specific CD4+ T cells is unclear. We isolated and characterized T cell antigen receptors (TCRs) from naturally primed CD4+ T cells specific for 2 oncoproteins, HPV-16 E6 and the activating KRASG12V mutation, from patients with head and neck squamous cell carcinoma and pancreatic ductal adenocarcinoma, respectively, and determined their ability to recognize autologous or human leukocyte antigen–matched antigen-expressing tumor cells. We found in both cases that the TCRs were capable of recognizing peptide-loaded target cells expressing the relevant MHC-II or B cell antigen-presenting cells (APCs) when the antigens were endogenously expressed and directed to the endosomal pathway but failed to recognize tumor cells expressing the source protein even after induction of surface MHC-II expression by IFN-γ or transduction with CIITA. These results suggest that priming and functional recognition of both a nuclear (E6) and a membrane-associated (KRAS) oncoprotein are predominantly confined to crosspresenting APCs rather than via direct recognition of tumor cells induced to express MHC-II.

Binary expression systems are a powerful tool for tissue- and cell-specific research. Many of the currently available Drosophila eye-specific drivers have not been systematically characterized for their expression level and cell-type specificity in the adult eye or during development. Here, we used a luciferase reporter to measure expression levels of different drivers in the adult Drosophila eye, and characterized the cell type-specificity of each driver using a fluorescent reporter in live 10-day-old adult males. We also further characterized the expression pattern of these drivers in various developmental stages. We compared several Gal4 drivers from the Bloomington Drosophila Stock Center (BDSC) including GMR-Gal4, longGMR-Gal4 and Rh1-Gal4 with newly developed Gal4 and QF2 drivers that are specific to different cell types in the adult eye. In addition, we generated drug-inducible Rh1-GSGal4 lines and compared their induced expression with an available GMR-GSGal4 line. Although both lines had significant induction of gene expression measured by luciferase activity, Rh1-GSGal4 was expressed at levels below the detection of the fluorescent reporter by confocal microscopy, while GMR-GSGal4 showed substantial reporter expression in the absence of drug by microscopy. Overall, our study systematically characterizes and compares a large toolkit of eye- and photoreceptor-specific drivers, while also uncovering some of the limitations of currently available expression systems in the adult eye.

Recent advances in developing and screening candidate pharmacotherapies for psychiatric disorders have depended on rodent models. Eating disorders are a set of psychiatric disorders that have traditionally relied on behavioral therapies for effective long-term treatment. However, the clinical use of Lisdexamfatamine for binge eating disorder (BED) has furthered the notion of using pharmacotherapies for treating binge eating pathologies. While there are several binge eating rodent models, there is not a consensus on how to define pharmacological effectiveness within these models. Our purpose is to provide an overview of the potential pharmacotherapies or compounds tested in established rodent models of binge eating behavior. These findings will help provide guidance for determining pharmacological effectiveness for potential novel or repurposed pharmacotherapies.

During embryonic development, tendons transform into a hypocellular tissue with robust tensile load-bearing capabilities. Previous work suggests that this mechanical transformation is due to increases in collagen fibril length and is dependent on mechanical stimulation via muscle activity. However, the relationship between changes in the microscale tissue structure and changes in macroscale tendon mechanics is still unclear. Additionally, the specific effect of mechanical stimulation on the multiscale structure-function relationships of developing tendons is also unknown. Therefore, the objective of this study was to measure the changes in tendon mechanics and structure at multiple length scales during embryonic development with and without skeletal muscle paralysis. Tensile testing of tendons from chick embryos was performed to determine the macroscale tensile modulus as well as the magnitude of the fibril strains and interfibrillar sliding with applied tissue strain. Embryos were also treated with either decamethonium bromide or pancuronium bromide to produce rigid or flaccid paralysis. Histology was performed to assess changes in tendon size, spacing between tendon subunits, and collagen fiber diameter. We found that the increase in the macroscale modulus observed with development is accompanied by an increase in the fibril:tissue strain ratio, which is consistent with an increase in collagen fibril length. Additionally, we found that flaccid paralysis reduced the macroscale tendon modulus and the fibril:tissue strain ratio, whereas less pronounced effects that were not statistically significant were observed with rigid paralysis. Finally, skeletal paralysis also reduced the size of collagen fibril bundles (i.e., fibers). Together, these data suggest that more of the applied tissue strain is transmitted to the collagen fibrils at later embryonic ages, which leads to an increase in the tendon macroscale tensile mechanics. Furthermore, our data suggest that mechanical stimulation during development is necessary to induce structural and mechanical changes at multiple physical length scales. This information provides valuable insight into the multiscale structure-function relationships of developing tendons and the importance of mechanical stimulation in producing a robust tensile load-bearing soft tissue.

Exposure of common bottlenose dolphins (Tursiops truncatus) to brevetoxins (PbTx) produced by blooms of the toxic phytoplankton Karenia brevis frequently results in severe health impacts, including illness and large-scale mortality events. Although PbTx accumulation in dead-stranded dolphins is well documented, there are limited data for corresponding brevetoxin exposure in live dolphins. In addition, the severity of impacts on living survivors of such toxic blooms is difficult to assess due to a lack of data on the relationship between K. brevis bloom severity and corresponding PbTx concentrations in exposed animals. Here we present results of PbTx analysis of urine, serum, milk, gastric fluid, and feces samples collected from live, free-ranging dolphins (n = 253) from Sarasota Bay, Florida during 2000–2018, and investigate the relationship between PbTx concentrations detected and corresponding K. brevis cell abundances that are temporally (within 30 days) and spatially (within 16 km) associated with each individual. We found that 28% of dolphins were associated with elevated K. brevis abundances (10,000–60,000,000 cells/L), with 41% (n = 104) of dolphins testing positive for PbTx in at least one sample type. Harmful algal blooms (HABs) such as those produced by Karenia brevis have acute negative impacts on common bottlenose dolphins (Tursiops truncatus) in Florida coastal waters, frequently causing illness and death. However, much less is known about chronic, sub-acute effects on these important sentinel species. This study investigates whether bottlenose dolphin behavior in Sarasota Bay, Florida is influenced by the presence of severe red tide events, focusing on respiratory and other behaviors likely affected by abundant toxin aerosols produced during these blooms. Through focal animal behavioral follows, we observed free-ranging dolphin respiratory behavior, activity budgets, and movement patterns relative to K. brevis abundance in the study area. We compared behavior from dolphins observed during a 2005 K. brevis bloom to those observed during inter-bloom conditions where K. brevis was present at background concentrations. We found that the rate of “chuffing”, an explosive type of exhalation, was significantly greater in dolphins observed during the bloom. No apparent effect on respiratory rate, heading change rate or activity budgets was observed. We propose that this chuffing behavior is analogous to symptoms of respiratory irritation observed in humans exposed to such red tide events, and suggest that this may be a type of disturbance response. With an observed increase in both the frequency and severity of HABs, such disturbance responses may have large-scale chronic impacts to the health and fitness of bottlenose dolphins in regions where such HABs are common.

This paper presents a hybrid electromechanical transformer that passively transfers electrical power between galvanically isolated ports by coupling electrodynamic and piezoelectric transducers. The use of these two complementary electromechanical transduction methods along with a high-Q mechanical resonance affords very large transformations of voltage, current, or impedance at particular electrical frequencies. A chip-size prototype is designed, simulated, fabricated, and experimentally characterized. The 7.6 mm × 7.6 mm × 1.65 mm device achieves an open-circuit voltage gain of 31.4 and 48.7 when operating as a step-up transformer at 729.5 Hz and 1015 Hz resonance frequencies, respectively. When operating as a step-down transformer, the resonance frequencies and the corresponding voltage gains are 728 Hz, 1002 Hz, and 0.0097, 0.0128, respectively. In one operational mode, the system shows a minimum power dissipation of only 0.9 µW corresponding to a power conversion efficiency of 11.8%.

Abstract Background Aging is associated with functional decline of neurons and increased incidence of both neurodegenerative and ocular disease. Photoreceptor neurons in Drosophila melanogaster provide a powerful model for studying the molecular changes involved in functional senescence of neurons since decreased visual behavior precedes retinal degeneration. Here, we sought to identify gene expression changes and the genomic features of differentially regulated genes in photoreceptors that contribute to visual senescence. Results To identify gene expression changes that could lead to visual senescence, we characterized the aging transcriptome of Drosophila sensory neurons highly enriched for photoreceptors. We profiled the nuclear transcriptome of genetically-labeled photoreceptors over a 40 day time course and identified increased expression of genes involved in stress and DNA damage response, and decreased expression of genes required for neuronal function. We further show that combinations of promoter motifs robustly identify age-regulated genes, suggesting that transcription factors are important in driving expression changes in aging photoreceptors. However, long, highly expressed and heavily spliced genes are also more likely to be downregulated with age, indicating that other mechanisms could contribute to expression changes at these genes. Lastly, we identify that circular RNAs (circRNAs) strongly increase during aging in photoreceptors. Conclusions Overall, we identified changes in gene expression in aging Drosophila photoreceptors that could account for visual senescence. Further, we show that genomic features predict these age-related changes, suggesting potential mechanisms that could be targeted to slow the rate of age-associated visual decline.

We report the design, fabrication, and characterization of a prototype that meets the form, fit, and function of a household 1.5 V AA battery, but which can be wirelessly recharged without removal from the host device. The prototype system comprises a low-frequency electrodynamic wireless power transmission (EWPT) receiver, a lithium polymer energy storage cell, and a power management circuit (PMC), all contained within a 3D-printed package. The EWPT receiver and overall system are experimentally characterized using a 238 Hz sinusoidal magnetic charging field and either a 1000 µF electrolytic capacitor or a lithium polymer (LiPo) cell as the storage cell. The system demonstrates a minimal operating field as low as 50 µTrms (similar in magnitude to Earth’s magnetic field). At this minimum charging field, the prototype transfers a maximum dc current of 50 µA to the capacitor, corresponding to a power delivery of 118 µW. The power effectiveness of the power management system is approximately 49%; with power effectiveness defined as the ratio between actual output power and the maximum possible power the EWPT receiver can transfer to a pure resistive load at a given field strength.

The present work aims to shed light on recent literature reports suggesting that ionic species are implicated in the electrical conductivity of 1-octanol and its mixtures with hydrocarbons. Other workers have questioned this interpretation, and herein, based on new experimentation and with reference to various literature studies, we consider that molecular interactions are more likely to be responsible. To investigate this, we have studied mixtures of 1-octanol and either silicone oil (SO) or n-dodecane as nonpolar components, using dielectric (in particular electrical conductivity) and viscometric measurements. With reference to the literature, the self-association of alcohols is known to create microheterogeneity in the neat liquids and in mixtures with nonpolar, low dielectric constant liquids, and it has previously been considered to be responsible for the particular solvent properties of alcohols. The present results suggest that the electrical conductivity of alkane/alcohol systems may have similar origins, with percolating pathways formed from octanol-rich nanodomains comprising polar regions containing hydrogen-bonded hydroxyl groups and nonpolar regions dominated by alkyl chains. The percolation threshold found for dodecane/octanol mixtures, in which interactions between the component molecules are found from viscosity measurements to be repulsive, agrees well with results from experimental and theoretical studies of disordered arrangements of packed spheres, and moreover, it is consistent with other published alkane/alcohol results. On the other hand, the situation is more complex for SO/octanol mixtures, in which interactions between the two components are attractive, based on viscosity data, and in which the phase separation of SO occurs at high octanol concentrations. Overall, we have concluded that electrical conductivity in octanol (and potentially all liquid alcohols) and its mixtures with nonpolar molecules, such as alkanes, is consistent with the presence of conducting networks comprising octanol-rich nanodomains formed by self-association, and not as a result of ionic conduction.

On the basis of dynamic interfacial tension measurements, Ca2+ has been shown specifically to interact with naphthenic acid (NA) at the n-heptane/water interface, consistent with NA adsorption followed by interfacial complexation and formation of a more ordered interfacial film. Optimum concentrations of Ca2+ and NA have been found to yield lower, time-dependent interfacial tensions, not evident for Mg2+ and Sr2+ or for several alkali metal ions studied. The results reflect the specific hydration and coordination chemistry of Ca2+ seen in biology. Owing to the ubiquitous presence of Ca2+ in oilfield waters, this finding has potential relevance to the surface chemistry underlying crude oil recovery. For example, “locking” acidic components at water/oil interfaces may be important for crude oil emulsion stability, or in bonding bulk oil to mineral surfaces through an aqueous phase, potentially relevant for carbonate reservoirs. The relevance of the present results to low salinity waterflooding as an enhanced crude oil recovery technique is also discussed.

Few long-term multicenter investigations have evaluated the relationships between aggregate antimicrobial drug use in hospitals and bacterial resistance. We measured fluoroquinolone use from 1999 through 2003 in a network of US hospitals. The percentages of fluoroquinolone-resistant Pseudomonas aeruginosa and methicillin-resistant Staphylococcus aureus (MRSA) were obtained from yearly antibiograms at each hospital. Univariate linear regression showed significant associations between a hospital's volume of fluoroquinolone use and percent resistance in most individual study years (1999–2001 for P. aeruginosa, 1999–2002 for S. aureus). When the method of generalized estimating equations was used, a population-averaged longitudinal model incorporating total fluoroquinolone use and the previous year's resistance (to account for autocorrelation) did not show a significant effect of fluoroquinolone use on percent resistance for most drug-organism combinations, except for the relationship between levofloxacin use and percent MRSA. The ecologic relationship between fluoroquinolone use and resistance is complex and requires further study.

The aims of this study were to determine the associations between anemia of critical illness, erythropoietin stimulating agents (ESA), packed red blood cell transfusions and varying degrees of renal dysfunction with mortality, and ICU- and hospital length of stay (LOS). This was a cross-sectional retrospective study of 5,314 ICU patients from USA hospitals. Hospital, patient demographics, and clinical characteristics were collected. Predictors of mortality and hospital and ICU LOS were evaluated using multivariate logistic regression models. The mean ICU admission hemoglobin in this study was 9.4 g/dL. The prevalence of ESA use was 13% and was associated with declining renal function; 26% of the ICU patients in this study received transfusion. Intravenous (IV) iron and Erythropoiesis Stimulating Agents (ESAs) are recommended for anemia management in chronic kidney disease (CKD). This retrospective cohort study analyzed utilization patterns of IV iron and ESA in patients over 18 years of age admitted to University Health System Hospitals with a primary or secondary diagnosis of CKD between January 1, 2006 to December 31, 2008. A clustered binomial logistic regression using the GEE methodology was used to identify predictors of IV iron utilization. Only 8% (n = 6678) of CKD patients on ESA therapy received IV iron supplementation in university hospitals. Those receiving iron used significantly less amounts of ESAs. Patient demographics (age, race, primary payer), patient clinical conditions (admission status, severity of illness, dialysis status), and physician specialty were identified as predictors of IV iron use in CKD patients. Use of IV iron with ESAs was low despite recommendations from consensus guidelines. The low treatment rate of IV iron represents a gap in treatment practices and signals an opportunity for healthcare improvement in CKD anemic patients.

Objective: To determine (1) how patients currently dispose of used insulin needles, (2) whether patients were educated about disposal of their used insulin needles, and (3) who educated patients about the disposal of their used insulin needles. Methods: A self-administered questionnaire was designed for this study. The survey assessed patient knowledge about disposal of used insulin needles and the patient-reported source and location of education about disposal techniques. The questionnaire was administered to a convenience sample of patients from four locations in Richmond, Virginia. Any patient who used insulin, was at least 18 years old, and was willing to complete the survey was eligible for inclusion. Results: Fifty responses were received with 40% indicating that education had been received on the disposal of used needles. From that 40%, nurses were identified as the source of education 60% of the time and pharmacists 25% of the time. Approximately 50% of the respondents reported disposing of used needles directly in the trash when at home. While away from home, 22% reported placing used needles in the trash, and 38% took them home for disposal. Conclusion: Patients are not consistently educated regarding the proper disposal of used needles. Health care practitioners should play a larger role in educating patients about the potential risks of inappropriate needle disposal and appropriate disposal methods. Future research is still needed to understand fully the magnitude of the problems associated with inappropriate needle disposal by patients. Type: Original Research

Researchers find that students struggle to learn this critical engineering design practice, particularly when tackling real-world engineering design problems. Purpose/Hypothesis: To improve our ability to teach iteration, this study contributes (i) a new teaching approach to improve student teams' expert iterative practices, and (ii) provides support to existing frameworks--chiefly the Design Risk Framework--that predict the key metacognitive processes we should support to help students to engage in expert iterative practices in real-world engineering design. Design/Method: In a 3-year design-based research study, we developed a novel approach to teaching students to take on real-world engineering design projects with real clients, users, and contexts to engage in expert iterative practices. Results: Study 1 confirms that student teams struggle to engage in expert iterative practices, even when supported by problem-based learning (PBL) coaching. Study 2 tests our novel approach, Planning-to-Iterate, which uses (i) templates, (ii) guiding questions to help students to define problem and solution elements, and (iii) risk checklists to help student teams to identify risks. We found that student teams using Planning-to-Iterate engaged in more expert iterative practices while receiving less PBL coaching. Conclusions: This work empirically tests a design argument--a theory for a novel teaching approach--that augments PBL coaching and helps students to identify risks and engage in expert iterative practices in engineering design projects.
