Abstract Background Spinal cord motor neurons (MNs) from human iPS cells (iPSCs) have wide applications in disease modeling and therapeutic development for amyotrophic lateral sclerosis (ALS) and other MN-associated neurodegenerative diseases. We need highly efficient MN differentiation strategies for generating iPSC-derived disease models that closely recapitulate the genetic and phenotypic complexity of ALS. An important application of these models is to understand molecular mechanisms of action of FDA-approved ALS drugs that only show modest clinical efficacy. Novel mechanistic insights will help us design optimal therapeutic strategies together with predictive biomarkers to achieve better efficacy. Methods We induce efficient MN differentiation from iPSCs in 4 days using synthetic mRNAs coding two transcription factors (Ngn2 and Olig2) with phosphosite modification. These MNs after extensive characterization were applied in electrophysiological and neurotoxicity assays as well as transcriptomic analysis, to study the neuroprotective effect and molecular mechanisms of edaravone, an FDA-approved drug for ALS, for improving its clinical efficacy. Results We generate highly pure and functional mRNA-induced MNs (miMNs) from control and ALS iPSCs, as well as embryonic stem cells. Edaravone alleviates H2O2-induced neurotoxicity and electrophysiological dysfunction in miMNs, demonstrating its neuroprotective effect that was also found in the glutamate-induced miMN neurotoxicity model. Guided by the transcriptomic analysis, we show a previously unrecognized effect of edaravone to induce the GDNF receptor RET and the GDNF/RET neurotrophic signaling in vitro and in vivo, suggesting a clinically translatable strategy to activate this key neuroprotective signaling. Notably, edaravone can replace required neurotrophic factors (BDNF and GDNF) to support long-term miMN survival and maturation, further supporting the neurotrophic function of edaravone-activated signaling. Furthermore, we show that edaravone and GDNF combined treatment more effectively protects miMNs from H2O2-induced neurotoxicity than single treatment, suggesting a potential combination strategy for ALS treatment. Conclusions This study provides methodology to facilitate iPSC differentiation and disease modeling. Our discoveries will facilitate the development of optimal edaravone-based therapies for ALS and potentially other neurodegenerative diseases. Graphical abstract

Nerve regeneration in diabetes is essential for reversal of neuropathy as well as the recovery of nerves from injury due to acute nerve compression. Here, we investigated the effects of treadmill training (TMT) on nerve regeneration and myelination of injured nerve, restoration of spinal cord motor neuron, glucose metabolism and functional recovery of innervated muscles after sciatic nerve injury in the streptozotocin-induced diabetic rats. Using immunofluorescenestaining, anterograde and retrogradetracing techniques, we identified enhanced axonal regrowth in the sciatic nerve and number of Dil-labeled spinal cord motor neurons after crush injury in diabetes rats with low intensity treadmill exercise. Primary DRG cell culture cells were Immunofluorescence stained with tubulin, neuron-specific antibody and Hoechst staining for nuclear visualization. We also carried out western blot analysis to investigate axonal growth-associated protein, GAP-43 expression in injured sciatic nerve and glucose transporter (GLUT) 4 expression in innervated muscles. The neural functional assessment was determined by the Sciatic Functional Index (SFl) for hind-foot prints. The numbers of total myelinated axon and degenerated myelin in the sciatic nerves were measured by the examination of semithin sections made from epon-embedded tissue blocks under light microscope and transmission electron microscope (TEM) with toluidine blue staining and analyzed with image analysis system to make a morphological analysis of the effect of TMT on injured nerves. GAP-43 levels were highly induced in DIE (diabetes + sciatic nerve injury + low intensity treadmill exercise) group compared with sedentary group. Total number of Dil-labeled motor neurons in time spinal cord was increased in DIE group compared with DI (diabetes + sciatic never injury) group. Total number of myelinated axons was increased in DIE group. GLU'F-4 levels also were highly induced in DIE group compared with DI group. Neurite outgrowth of DRG sensory neurons was increased after TMT. SFI values in DIE group were increased more rapidly than DI group. We found that low intensity treadmill exercise promoted axonal regeneration and myelination of injured nerve, glucose metabolism and functional recovery of innervated muscles after sciatic nerve injury in the streptozotocin-induced diabetic rats. Thus, the present results suggest that low intensity treadmill exercise may be used for adjuvant therapy to the regeneration of the peripheral nerve injury with diabetes. [ABSTRACT FROM AUTHOR]

Peripheral nerve injury (PNI) impacts millions annually, often leaving debilitated patients with minimal repair options to improve functional recovery. Our group has previously developed tissue engineered nerve grafts (TENGs) featuring long, aligned axonal tracts from dorsal root ganglia (DRG) neurons that are fabricated in custom bioreactors using the process of axon "stretch-growth." We have shown that TENGs effectively serve as "living scaffolds" to promote regeneration across segmental nerve defects by exploiting the newfound mechanism of axon-facilitated axon regeneration, or "AFAR," by simultaneously providing haptic and neurotrophic support. To extend this work, the current study investigated the efficacy of living versus nonliving regenerative scaffolds in preserving host sensory and motor neuronal health following nerve repair. Rats were assigned across five groups: naïve, or repair using autograft, nerve guidance tube (NGT) with collagen, NGT + non-aligned DRG populations in collagen, or TENGs. We found that TENG repairs yielded equivalent regenerative capacity as autograft repairs based on preserved health of host spinal cord motor neurons and acute axonal regeneration, whereas NGT repairs or DRG neurons within an NGT exhibited reduced motor neuron preservation and diminished regenerative capacity. These acute regenerative benefits ultimately resulted in enhanced levels of functional recovery in animals receiving TENGs, at levels matching those attained by autografts. Our findings indicate that TENGs may preserve host spinal cord motor neuron health and regenerative capacity without sacrificing an otherwise uninjured nerve (as in the case of the autograft) and therefore represent a promising alternative strategy for neurosurgical repair following PNI., (© 2020 John Wiley & Sons, Ltd.)
