A prototype version of the Q & U bolometric interferometer for cosmology (QUBIC) underwent a campaign of testing in the laboratory at Astroparticle Physics and Cosmology laboratory in Paris (APC). The detection chain is currently made of 256 NbSi transition edge sensors (TES) cooled to 320 mK. The readout system is a 128:1 time domain multiplexing scheme based on 128 SQUIDs cooled at 1 K that are controlled and amplified by an SiGe application specific integrated circuit at 40 K. We report the performance of this readout chain and the characterization of the TES. The readout system has been functionally tested and characterized in the lab and in QUBIC. The low noise amplifier demonstrated a white noise level of 0.3 nV.Hz^-0.5. Characterizations of the QUBIC detectors and readout electronics includes the measurement of I-V curves, time constant and the noise equivalent power. The QUBIC TES bolometer array has approximately 80% detectors within operational parameters. It demonstrated a thermal decoupling compatible with a phonon noise of about 5.10^-17 W.Hz^-0.5 at 410 mK critical temperature. While still limited by microphonics from the pulse tubes and noise aliasing from readout system, the instrument noise equivalent power is about 2.10^-16 W.Hz^-0.5, enough for the demonstration of bolometric interferometry., Comment: Accepted for publication in JCAP

The Q $\&$ U Bolometric Interferometer for Cosmology (QUBIC) is a novel kind of polarimeter optimized for the measurement of the B-mode polarization of the Cosmic Microwave Background (CMB), which is one of the major challenges of observational cosmology. The signal is expected to be of the order of a few tens of nK, prone to instrumental systematic effects and polluted by various astrophysical foregrounds which can only be controlled through multichroic observations. QUBIC is designed to address these observational issues with a novel approach that combines the advantages of interferometry in terms of control of instrumental systematic effects with those of bolometric detectors in terms of wide-band, background-limited sensitivity. The QUBIC synthesized beam has a frequency-dependent shape that results in the ability to produce maps of the CMB polarization in multiple sub-bands within the two physical bands of the instrument (150 and 220 GHz). These features make QUBIC complementary to other instruments and makes it particularly well suited to characterize and remove Galactic foreground contamination. In this article, first of a series of eight, we give an overview of the QUBIC instrument design, the main results of the calibration campaign, and present the scientific program of QUBIC including not only the measurement of primordial B-modes, but also the measurement of Galactic foregrounds. We give forecasts for typical observations and measurements: with three years of integration on the sky and assuming perfect foreground removal as well as stable atmospheric conditions from our site in Argentina, our simulations show that we can achieve a statistical sensitivity to the effective tensor-to-scalar ratio (including primordial and foreground B-modes) $\sigma(r)=0.015$., Comment: 34 pages, 16 figures, accepted for publication by JCAP. Bolometric interferometry is a novel technique that has the ability to perform spectral imaging. A bolometric interferometer observes the sky in a wide frequency band and can reconstruct sky maps in several sub-bands within the physical band in post-processing of the data. This provides a powerful spectral method to discriminate between the cosmic microwave background (CMB) and astrophysical foregrounds. In this paper, the methodology is illustrated with examples based on the Q \& U Bolometric Interferometer for Cosmology (QUBIC) which is a ground-based instrument designed to measure the B-mode polarization of the sky at millimeter wavelengths. We consider the specific cases of point source reconstruction and Galactic dust mapping and we characterize the point spread function as a function of frequency. We study the noise properties of spectral imaging, especially the correlations between sub-bands, using end-to-end simulations together with a fast noise simulator. We conclude showing that spectral imaging performance are nearly optimal up to five sub-bands in the case of QUBIC., Comment: 27 pages, 18 figures. Accepted by JCAP on July 6, 2021. We present the design, manufacturing and performance of the horn-switch system developed for the technological demonstrator of QUBIC (the $Q$\&$U$ Bolometric Interferometer for Cosmology). This system is constituted of 64 back-to-back dual-band (150\,GHz and 220\,GHz) corrugated feed-horns interspersed with mechanical switches used to select desired baselines during the instrument self-calibration. We manufactured the horns in aluminum platelets milled by photo-chemical etching and mechanically tightened with screws. The switches are based on steel blades that open and close the wave-guide between the back-to-back horns and are operated by miniaturized electromagnets. We also show the current development status of the feedhorn-switch system for the QUBIC full instrument, based on an array of 400 horn-switch assemblies., Comment: 30 pages, 28 figures. Current experiments aimed at measuring the polarization of the Cosmic Microwave Background (CMB) use cryogenic detector arrays and cold optical systems to boost the mapping speed of the sky survey. For these reasons, large volume cryogenic systems, with large optical windows, working continuously for years, are needed. Here we report on the cryogenic system of the QUBIC (Q and U Bolometric Interferometer for Cosmology) experiment: we describe its design, fabrication, experimental optimization and validation in the Technological Demonstrator configuration. The QUBIC cryogenic system is based on a large volume cryostat, using two pulse-tube refrigerators to cool at ~3K a large (~1 m^3) volume, heavy (~165kg) instrument, including the cryogenic polarization modulator, the corrugated feedhorns array, and the lower temperature stages; a 4He evaporator cooling at ~1K the interferometer beam combiner; a 3He evaporator cooling at ~0.3K the focal-plane detector arrays. The cryogenic system has been tested and validated for more than 6 months of continuous operation. The detector arrays have reached a stable operating temperature of 0.33K, while the polarization modulator has been operated from a ~10K base temperature. The system has been tilted to cover the boresight elevation range 20 deg -90 deg without significant temperature variations. Inflation Gravity Waves B-Modes polarization detection is the ultimate goal of modern large angular scale cosmic microwave background (CMB) experiments around the world. A big effort is undergoing with the deployment of many ground-based, balloon-borne and satellite experiments using different methods to separate this faint polarized component from the incoming radiation. One of the largely used technique is the Stokes Polarimetry that uses a rotating half-wave plate (HWP) and a linear polarizer to separate and modulate the polarization components with low residual cross-polarization. This paper describes the QUBIC Stokes Polarimeter highlighting its design features and its performances. A common systematic with these devices is the generation of large spurious signals synchronous with the rotation and proportional to the emissivity of the optical elements. A key feature of the QUBIC Stokes Polarimeter is to operate at cryogenic temperature in order to minimize this unwanted component. Moving efficiently this large optical element at low temperature constitutes a big engineering challenge in order to reduce friction power dissipation. Big attention has been given during the designing phase to minimize the differential thermal contractions between parts. The rotation is driven by a stepper motor placed outside the cryostat to avoid thermal load dissipation at cryogenic temperature. The tests and the results presented in this work show that the QUBIC polarimeter can easily achieve a precision below 0.1{\deg} in positioning simply using the stepper motor precision and the optical absolute encoder. The rotation induces only few mK of extra power load on the second cryogenic stage (~ 8 K)., Comment: Part of a series of 8 papers on QUBIC to be submitted to a special issue of JCAP

The Q and U Bolometric Interferometer for Cosmology (QUBIC) is a ground-based experiment that aims to detect B-mode polarisation anisotropies in the CMB at angular scales around the l=100 recombination peak. Systematic errors make ground-based observations of B modes at millimetre wavelengths very challenging and QUBIC mitigates these problems in a somewhat complementary way to other existing or planned experiments using the novel technique of bolometric interferometry. This technique takes advantage of the sensitivity of an imager and the systematic error control of an interferometer. A cold reflective optical combiner superimposes there-emitted beams from 400 aperture feedhorns on two focal planes. A shielding system composedof a fixed groundshield, and a forebaffle that moves with the instrument, limits the impact of local contaminants. A prototype version of the Q & U Bolometric Interferometer for Cosmology (QUBIC) underwent a campaign of testing in the laboratory at Astroparticle Physics and Cosmology in Paris. We report the results of this Technological Demonstrator which successfully shows the feasibility of the principle of Bolometric Interferometry. Characterization of QUBIC includes the measurement of the synthesized beam, the measurement of interference fringes, and the measurement of polarization performance. A modulated and frequency tunable millimetre-wave source in the telescope far-field is used to simulate a point source. The QUBIC pointing is scanned across the point source to produce beam maps. Polarization modulation is measured using a rotating Half Wave Plate. The measured beam matches well to the theoretical simulations and gives QUBIC the ability to do spectro imaging. The polarization performance is excellent with less than 0.5\% cross-polarization rejection. The Q & U Bolometric Interferometer for Cosmology, QUBIC, is an innovative experiment designed to measure the polarization of the Cosmic Microwave Background and in particular the signature left therein by the inflationary expansion of the Universe. The expected signal is extremely faint, thus extreme sensitivity and systematic control are necessary in order to attempt this measurement. QUBIC addresses these requirements using an innovative approach combining the sensitivity of Transition Edge Sensor cryogenic bolometers, with the deep control of systematics characteristic of interferometers. This makes QUBIC unique with respect to others classical imagers experiments devoted to the CMB polarization. In this contribution we report a description of the QUBIC instrument including recent achievements and the demonstration of the bolometric interferometry performed in lab. QUBIC will be deployed at the observation site in Alto Chorrillos, in Argentina at the end of 2019., Comment: Accepted for publication in the Journal of Low Temperature Physics

QUBIC (Q \& U Bolometric Interferometer for Cosmology) is an international ground-based experiment dedicated in the measurement of the polarized fluctuations of the Cosmic Microwave Background (CMB). It is based on bolometric interferometry, an original detection technique which combine the immunity to systematic effects of an interferometer with the sensitivity of low temperature incoherent detectors. QUBIC will be deployed in Argentina, at the Alto Chorrillos mountain site near San Antonio de los Cobres, in the Salta province. The QUBIC detection chain consists in 2048 NbSi Transition Edge Sensors (TESs) cooled to 350mK.The voltage-biased TESs are read out with Time Domain Multiplexing based on Superconducting QUantum Interference Devices (SQUIDs) at 1 K and a novel SiGe Application-Specific Integrated Circuit (ASIC) at 60 K allowing to reach an unprecedented multiplexing (MUX) factor equal to 128. The QUBIC experiment is currently being characterized in the lab with a reduced number of detectors before upgrading to the full instrument. - Abstract
Background: Literature suggests colonic resection and primary anastomosis (RPA) instead of Hartmann’s procedure (HP) for the treatment of left-sided colonic emergencies. We aim to evaluate the surgical options globally used to treat patients with acute left-sided colonic emergencies and the factors that leading to the choice of treatment, comparing HP and RPA. Methods: This is a prospective, international, multicenter, observational study registered on ClinicalTrials.gov. A total 1215 patients with left-sided colonic emergencies who required surgery were included from 204 centers during the period of March 1, 2020, to May 31, 2020. with a 1-year follow-up. Results: 564 patients (43.1%) were females. The mean age was 65.9 ± 15.6 years. HP was performed in 697 (57.3%) patients and RPA in 384 (31.6%) cases. Complicated acute diverticulitis was the most common cause of left-sided colonic emergencies (40.2%), followed by colorectal malignancy (36.6%). Severe complications (Clavien-Dindo ≥ 3b) were higher in the HP group (P < 0.001). 30-day mortality was higher in HP patients (13.7%), especially in case of bowel perforation and diffused peritonitis. 1-year follow-up showed no differences on ostomy reversal rate between HP and RPA. (P = 0.127). A backward likelihood logistic regression model showed that RPA was preferred in younger patients, having low ASA score (≤ 3), in case of large bowel obstruction, absence of colonic ischemia, longer time from admission to surgery, operating early at the day working hours, by a surgeon who performed more than 50 colorectal resections. Conclusions: After 100 years since the first Hartmann’s procedure, HP remains the most common treatment for left-sided colorectal emergencies. Treatment’s choice depends on patient characteristics, the time of surgery and the experience of the surgeon. RPA should be considered as the gold standard for surgery, with HP being an exception.

- Abstract
SETBP1 mutations are found in various clonal myeloid disorders. However, it is unclear whether they can initiate leukemia, because SETBP1 mutations typically appear as later events during oncogenesis. To answer this question, we generated a mouse model expressing mutated SETBP1 in hematopoietic tissue: this model showed profound alterations in the differentiation program of hematopoietic progenitors and developed a myeloid neoplasm with megakaryocytic dysplasia, splenomegaly, and bone marrow fibrosis, prompting us to investigate SETBP1 mutations in a cohort of 36 triple-negative primary myelofibrosis (TN-PMF) cases. We identified 2 distinct subgroups, one carrying SETBP1 mutations and the other completely devoid of somatic variants. Clinically, a striking difference in disease aggressiveness was noted, with patients with SETBP1 mutation showing a much worse clinical course. In contrast to myelodysplastic/myeloproliferative neoplasms, in which SETBP1 mutations are mostly found as a late clonal event, single-cell clonal hierarchy reconstruction in 3 patients with TN-PMF from our cohort revealed SETBP1 to be a very early event, suggesting that the phenotype of the different SETBP1+ disorders may be shaped by the opposite hierarchy of the same clonal SETBP1 variants.

- Abstract
Polycythemia Vera (PV) is typically caused by V617F or exon 12 JAK2 mutations. Little is known about Polycythemia cases where no JAK2 variants can be detected, and no other causes identified. This condition is defined as idiopathic erythrocytosis (IE). We evaluated clinical-laboratory parameters of a cohort of 56 IE patients and we determined their molecular profile at diagnosis with paired blood/buccal-DNA exome-sequencing coupled with a high-depth targeted OncoPanel to identify a possible underling germline or somatic cause. We demonstrated that most of our cohort (40/56: 71.4%) showed no evidence of clonal hematopoiesis, suggesting that IE is, in large part, a germline disorder. We identified 20 low mutation burden somatic variants (Variant allelic fraction, VAF, < 10%) in only 14 (25%) patients, principally involving DNMT3A and TET2. Only 2 patients presented high mutation burden somatic variants, involving DNMT3A, TET2, ASXL1 and WT1. We identified recurrent germline variants in 42 (75%) patients occurring mainly in JAK/STAT, Hypoxia and Iron metabolism pathways, among them: JAK3-V722I and HIF1A-P582S; a high fraction of patients (48.2%) resulted also mutated in homeostatic iron regulatory gene HFE-H63D or C282Y. By generating cellular models, we showed that JAK3-V722I causes activation of the JAK-STAT5 axis and upregulation of EPAS1/HIF2A, while HIF1A-P582S causes suppression of hepcidin mRNA synthesis, suggesting a major role for these variants in the onset of IE.

QUBIC, the QU Bolometric Interferometer for Cosmology, is a novel forthcoming instrument to measure the B-mode polarization anisotropy of the Cosmic Microwave Background. The detection of the B-mode signal will be extremely challenging; QUBIC has been designed to address this with a novel approach, namely bolometric interferometry. The receiver cryostat is exceptionally large and cools complex optical and detector stages to 40 K, 4 K, 1 K and 350 mK using two pulse tube coolers, a novel 4He sorption cooler and a double-stage 3He/4He sorption cooler. We discuss the thermal and mechanical design of the cryostat, modelling and thermal analysis, and laboratory cryogenic testing.

Atmospheric emission is a dominant source of disturbance in ground-based astronomy at mm wavelengths. The Antarctic plateau is recognized to be an ideal site for mm and sub-mm observations, and the French/Italian base of Dome C is among the best sites on Earth for these observations. In this paper we present measurements, performed using the BRAIN-pathfinder experiment, at Dome C of the atmospheric emission in intensity and polarization at 150GHz, one of the best observational frequencies for CMB observations when considering cosmic signal intensity, atmospheric transmission, detectors sensitivity, and foreground removal. Careful characterization of the air-mass synchronous emission has been performed, acquiring more that 380 elevation scans (i.e. "skydip") during the third BRAIN-pathfinder summer campaign in December 2009/January 2010. The extremely high transparency of the Antarctic atmosphere over Dome Concordia is proven by the very low measured optical depth:
=0.050 \pm 0.003 \pm 0.011 where the first error is statistical and the second is systematic error. Mid term stability, over the summer campaign, of the atmosphere emission has also been studied. Adapting the radiative transfer atmosphere emission model "am" to the particular conditions found at Dome C, we also infer the level of the PWV content of the atmosphere, notoriously the main source of disturbance in millimetric astronomy ( =0.77 +/- 0.06 + 0.15 - 0.12 mm). Upper limits on the air-mass correlated polarized signal are also placed for the first time. The degree of circular polarization of atmospheric emission is found to be lower than 0.2% (95%CL), while the degree of linear polarization is found to be lower than 0.1% (95%CL). One of the major challenges of modern cosmology is the detection of B-mode polarization anisotropies in the CMB. These originate from tensor fluctuations of the metric produced during the inflationary phase. Their detection would therefore constitute a major step towards understanding the primordial Universe. The expected level of these anisotropies is however so small that it requires a new generation of instruments with high sensitivity and extremely good control of systematic effects. We propose the QUBIC instrument based on the novel concept of bolometric interferometry, bringing together the sensitivity advantages of bolometric detectors with the systematics effects advantages of interferometry. Methods: The instrument will directly observe the sky through an array of entry horns whose signals will be combined together using an optical combiner. The whole set-up is located inside a cryostat. Polarization modulation will be achieved using a rotating half-wave plate and interference fringes will be imaged on two focal planes (separated by a polarizing grid) tiled with bolometers. We show that QUBIC can be considered as a synthetic imager, exactly similar to a usual imager but with a synthesized beam formed by the array of entry horns. Scanning the sky provides an additional modulation of the signal and improve the sky coverage shape. The usual techniques of map-making and power spectrum estimation can then be applied. We show that the sensitivity of such an instrument is comparable with that of an imager with the same number of horns. We anticipate a low level of beam-related systematics thanks to the fact that the synthesized beam is determined by the location of the primary horns. The SAGACE experiment consists of a mm/sub-mm telescope with a 3-m diameter primary mirror, coupled to a cryogenic multi-beam differential spectrometer. SAGACE explores the sky in the 100-760 GHz frequency range, using four diffraction-limited bolometer arrays. The instrument is designed to perform spectroscopic surveys of the Sunyaev-Zeldovich effects of thousands of galaxy clusters, of the spectral energy distribution of active galactic nuclei, and of the [CII] line of a thousand galaxies in the redshift desert. In 2008 a full phase-A study for a national small mission was completed and delivered to the Italian Space Agency (ASI). We have shown that taking advantage of the differential operation of the Fourier Transform Spectrometer, this ambitious instrument can operate from a Molniya orbit, and can be built and operated within the tight budget of a small mission., Comment: Proceedings of the 12th Marcel Grossman Meeting

On the occasion of the reorganization of disciplinary fields (DF) which is under implementation by the MIUR (Italian Ministry for Research and Education), the scientific production of the Italian forest research in two characterizing disciplines (AGR05 Forest management and Silviculture, and AGR06 Wood technology and Forest Operation) has been qualitatively investigated over the period 2005-2017. Using a text mining software, 2.112 scientific articles indexed in the SCOPUS database were analyzed and the evolution over time of the Italian forest research in the above disciplines was examined. The results indicate that ecosystem functions and services have pushed forest research to overcome the aspects linked to wood production (which still remains an important competence of the two DFs), to tackle topics aiming to understand the functioning and functionality of forest ecosystems, provide tools for monitoring forest dynamics and highlight the contribution of forests and urban green areas in contrasting climate change in terms of adaptation and mitigation. As regards the AGR06 sector, studies on forest operations have been mainly focused on issues related to the forest-energy supply chain, while for wood technology the enhancement of structural uses, the modified wood, and the conservation of Wooden Cultural Heritage have emerged. Therefore, the reorganization of disciplinary fields must certainly take into account the traditional Italian forestry thinking and the aspects related to planning, management and close-to-nature forestry, but also the evolution of forestry research in the past 15 years, which led to an intense scientific activity and production, must be considered. In defining the new declarations, it will therefore be appropriate firstly to consider the peculiarity in the area of Agricultural Science, and then the specificity of skills and research that each one of the two sectors carries forward both jointly and separately, thus ensuring to both sectors an adequate representation in autonomous profiles and domains.

- Abstract
Within the context of precision medicine, the scientific community is giving particular attention to early diagnosis and intervention, guided by non-invasive methodologies. Liquid biopsy (LBx) is a recent laboratory approach consisting of a non-invasive blood draw, which allows the detection of information about potential prognostic factors, or markers to be used for diagnostic purposes; it might also allow the clinician to establish a treatment regimen and predict a patient’s response. Since the discovery of circulating tumor cells (CTCs) in the nineteenth century, the possibility of integrating LBx into clinical practice has been explored, primarily because of its safeness and easy execution: indeed, compared to solid biopsy, sampling-related risks are less of a concern, and the quickness and repeatability of the process could help confirm a prompt diagnosis or to further corroborate the existence of a metastatic spreading of the disease. LBx’s usefulness has been consolidated in a narrow range of oncological settings, first of all, non-small cell lung carcinoma (NSCLC), and it is now gradually being assessed also in lymphoproliferative diseases, such as acute lymphocytic leukemia (ALL), B-cell lymphomas, and multiple myeloma. The present review aims to summarize LBx’s overall characteristics (such as its advantages and flaws, collection and analysis methodologies, indications, and targets of the test), and to highlight the applications of this technique within the specific field of B-cell malignancies. The perspectives on how such a simple and convenient technique could improve hemato-oncological clinical practice are broadly encouraging, yet far from a complete integration in routine clinical settings.

- Abstract
The Q & U Bolometric Interferometer for Cosmology (QUBIC) is a cosmology experiment that aims to measure the B-mode polarization of the cosmic microwave background (CMB). Measurements of the primordial B-mode pattern of the CMB polarization are in fact among the most exciting goals in cosmology as it would allow testing of the inflationary paradigm. Many experiments are attempting to measure the B-modes, from the ground and the stratosphere, using imaging Stokes polarimeters. The QUBIC collaboration developed an innovative concept to measure CMB polarization using bolometric interferometry. This approach mixes the high sensitivity of bolometric detectors with the accurate control of systematics due to the interferometric layout of the instrument. We present the calibration results for the Technological Demonstrator, before its commissioning in the Argentinian observing site and preparation for first light.

- Abstract
A key task of genomic surveillance of infectious viral diseases lies in the early detection of dangerous variants. Unexpected help to this end is provided by the analysis of deep sequencing data of viral samples, which are typically discarded after creating consensus sequences. Such analysis allows one to detect intra-host low-frequency mutations, which are a footprint of mutational processes underlying the origination of new variants. Their timely identification may improve public-health decision-making with respect to traditional approaches exploiting consensus sequences. We present the analysis of 220,788 high-quality deep sequencing SARS-CoV-2 samples, showing that many spike and nucleocapsid mutations of interest associated to the most circulating variants, including Beta, Delta, and Omicron, might have been intercepted several months in advance. Furthermore, we show that a refined genomic surveillance system leveraging deep sequencing data might allow one to pinpoint emerging mutation patterns, providing an automated data-driven support to virologists and epidemiologists.

- Abstract
QUBIC (Q and U bolometric interferometer for cosmology) is an international ground-based experiment dedicated to the measurement of the polarized fluctuations of the cosmic microwave background (CMB). It is based on bolometric interferometry, an original detection technique which combines the immunity to systematic effects of an interferometer with the sensitivity of low temperature incoherent detectors. QUBIC will be deployed in Argentina, at the Alto Chorrillos mountain site near San Antonio de los Cobres, in the Salta province. The QUBIC detection chain consists of 2048 NbSi transition edge sensors (TESs) cooled to 320 mK. The voltage-biased TESs are read out with time domain multiplexing based on superconducting quantum interference devices (SQUIDs) at 1 K and a novel SiGe application-specific integrated circuit (ASIC) at 60 K allowing an unprecedented multiplexing (MUX) factor equal to 128 to be reached. The current QUBIC version is based on a reduced number of detectors (1/4) in order to validate the detection technique. The QUBIC experiment is currently being validated in the lab in Salta (Argentina) before going to the site for observations. This paper presents the main results of the characterization phase with a focus on the detectors and readout system.

- Abstract
We report on an extensive test campaign of a prototype version of the QUBIC (Q & U Bolometric Interferometer for Cosmology) instrument, carried out at Astroparticle Physics and Cosmology (APC) in Paris. Exploiting the novel concept called bolometric interferometry, QUBIC is designed to measure the CMB polarization at 150 and 220 GHz from a high altitude site at Alto Chorillo, Argentina. The prototype model called QUBIC Technological Demonstrator (QUBIC-TD) operates in a single frequency band (150 GHz) and with a reduced number of baselines, but it contains all the elements of the QUBIC instrument in its final configuration. The test campaign included measurements of the synthesized beam and of the polarization performance, as well as a verification of the interference fringe pattern. A modulated, frequency-tunable millimetre-wave source was placed in the telescope far-field and was used to simulate a point source. The QUBIC-TD field of view was scanned across the source to produce beam maps. Our measurements confirm the frequency-dependent behaviour of the beam profile, which gives QUBIC the possibility to do spectral imaging. The measured polarization performance indicates a cross-polarization leakage less than 0.6%. We also successfully tested the polarization modulation system, which is provided by a rotating half wave plate. We demonstrate the full mapmaking pipeline using data from this measurement campaign, effectively giving an end-to-end checkout of the entire QUBIC system, including all hardware subsystems, their interfaces, and the software to operate the whole system and run the analysis. Our results confirm the viability of bolometric interferometry for measurements of the CMB polarization.

- Abstract
Bolometric interferometry is a novel technique that has the ability to perform spectral imaging. A bolometric interferometer observes the sky in a wide frequency band and can reconstruct sky maps in several sub-bands within the physical band in post-processing of the data. This provides a powerful spectral method to discriminate between the cosmic microwave background (CMB) and astrophysical foregrounds. In this paper, the methodology is illustrated with examples based on the Q & U Bolometric Interferometer for Cosmology (QUBIC) which is a ground-based instrument designed to measure the B-mode polarization of the sky at millimeter wavelengths. We consider the specific cases of point source reconstruction and Galactic dust mapping and we characterize the point spread function as a function of frequency. We study the noise properties of spectral imaging, especially the correlations between sub-bands, using end-to-end simulations together with a fast noise simulator. We conclude showing that spectral imaging performance are nearly optimal up to five sub-bands in the case of QUBIC.

- Abstract
A prototype version of the Q & U bolometric interferometer for cosmology (QUBIC) underwent a campaign of testing in the laboratory at Astroparticle Physics and Cosmology laboratory in Paris (APC). The detection chain is currently made of 256 NbSi transition edge sensors (TES) cooled to 320 mK. The readout system is a 128:1 time domain multiplexing scheme based on 128 SQUIDs cooled at 1 K that are controlled and amplified by a SiGe application specific integrated circuit at 40 K. We report the performance of this readout chain and the characterization of the TES. The readout system has been functionally tested and characterized in the lab and in QUBIC. The low noise amplifier demonstrated a white noise level of 0.3 nV/√Hz. Characterizations of the QUBIC detectors and readout electronics includes the measurement of I-V curves, time constant and the noise equivalent power. The QUBIC TES bolometer array has approximately 80% detectors within operational parameters. It demonstrated a thermal decoupling compatible with a phonon noise of about 5 × 10-17 W/√Hz at 410 mK critical temperature. While still limited by microphonics from the pulse tubes and noise aliasing from readout system, the instrument noise equivalent power is about 2 × 10-16 W/√Hz, enough for the demonstration of bolometric interferometry.

- Abstract
Current experiments aimed at measuring the polarization of the Cosmic Microwave Background (CMB) use cryogenic detector arrays with cold optical systems to boost their mapping speed. For this reason, large volume cryogenic systems with large optical windows, working continuously for years, are needed. The cryogenic system of the QUBIC (Q & U Bolometric Interferometer for Cosmology) experiment solves a combination of simultaneous requirements: very large optical throughput (1/440 cm2sr), large volume (1/41 m3) and large mass (1/4165 kg) of the cryogenic instrument. Here we describe its design, fabrication, experimental optimization and validation in the Technological Demonstrator configuration. The QUBIC cryogenic system is based on a large volume cryostat that uses two pulse-tube refrigerators to cool the instrument to 1/43 K. The instrument includes the cryogenic polarization modulator, the corrugated feedhorn array, and the lower temperature stages: a 4He evaporator cooling the interferometer beam combiner to 1/41 K and a 3He evaporator cooling the focal-plane detector arrays to 1/40.3 K. The cryogenic system has been tested and validated for more than 6 months of continuous operation. The detector arrays have reached a stable operating temperature of 0.33 K, while the polarization modulator has operated at a 1/410 K base temperature. The system has been tilted to cover the boresight elevation range 20°-90° without significant temperature variations. The instrument is now ready for deployment to the high Argentinean Andes.

- Abstract
The Q and U Bolometric Interferometer for Cosmology (QUBIC) is a ground-based experiment that aims to detect B-mode polarization anisotropies [1] in the CMB at angular scales around the â.,"≃100 recombination peak. Systematic errors make ground-based observations of B modes at millimetre wavelengths very challenging and QUBIC mitigates these problems in a somewhat complementary way to other existing or planned experiments using the novel technique of bolometric interferometry. This technique takes advantage of the sensitivity of an imager and the systematic error control of an interferometer. A cold reflective optical combiner superimposes the re-emitted beams from 400 aperture feedhorns on two focal planes. A shielding system composed of a fixed groundshield, and a forebaffle that moves with the instrument, limits the impact of local contaminants. The modelling, design, manufacturing and preliminary measurements of the optical components are described in this paper.

- Abstract
Within the chromatin, distal elements interact with promoters to regulate specific transcriptional programs. Histone acetylation, interfering with the net charges of the nucleosomes, is a key player in this regulation. Here, we report that the oncoprotein SET is a critical determinant for the levels of histone acetylation within enhancers. We disclose that a condition in which SET is accumulated, the severe Schinzel-Giedion Syndrome (SGS), is characterized by a failure in the usage of the distal regulatory regions typically employed during fate commitment. This is accompanied by the usage of alternative enhancers leading to a massive rewiring of the distal control of the gene transcription. This represents a (mal)adaptive mechanism that, on one side, allows to achieve a certain degree of differentiation, while on the other affects the fine and corrected maturation of the cells. Thus, we propose the differential in cis-regulation as a contributing factor to the pathological basis of SGS and possibly other the SET-related disorders in humans.

- Abstract
We present a large-scale analysis of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) substitutions, considering 1,585,456 high-quality raw sequencing samples, aimed at investigating the existence and quantifying the effect of mutational processes causing mutations in SARS-CoV-2 genomes when interacting with the human host. As a result, we confirmed the presence of three well-differentiated mutational processes likely ruled by reactive oxygen species (ROS), apolipoprotein B editing complex (APOBEC), and adenosine deaminase acting on RNA (ADAR). We then evaluated the activity of these mutational processes in different continental groups, showing that some samples from Africa present a significantly higher number of substitutions, most likely due to higher APOBEC activity. We finally analyzed the activity of mutational processes across different SARS-CoV-2 variants, and we found a significantly lower number of mutations attributable to APOBEC activity in samples assigned to the Omicron variant.

Purpose To analyze the application of a lung ultrasound (LUS)-based diagnostic approach to patients suspected of COVID-19, combining the LUS likelihood of COVID-19 pneumonia with patient’s symptoms and clinical history. Methods This is an international multicenter observational study in 20 US and European hospitals. Patients suspected of COVID-19 were tested with reverse transcription-polymerase chain reaction (RT-PCR) swab test and had an LUS examination. We identified three clinical phenotypes based on pre-existing chronic diseases (mixed phenotype), and on the presence (severe phenotype) or absence (mild phenotype) of signs and/or symptoms of respiratory failure at presentation. We defined the LUS likelihood of COVID-19 pneumonia according to four different patterns: high (HighLUS), intermediate (IntLUS), alternative (AltLUS), and low (LowLUS) probability. The combination of patterns and phenotypes with RT-PCR results was described and analyzed. Results We studied 1462 patients, classified in mild (n = 400), severe (n = 727), and mixed (n = 335) phenotypes. HighLUS and IntLUS showed an overall sensitivity of 90.2% (95% CI 88.23–91.97%) in identifying patients with positive RT-PCR, with higher values in the mixed (94.7%) and severe phenotype (97.1%), and even higher in those patients with objective respiratory failure (99.3%). The HighLUS showed a specificity of 88.8% (CI 85.55–91.65%) that was higher in the mild phenotype (94.4%; CI 90.0–97.0%). Current experiments aimed at measuring the polarization of the Cosmic Microwave Background (CMB) use cryogenic detector arrays and cold optical systems to boost the mapping speed of the sky survey. For these reasons, large volume cryogenic systems, with large optical windows, working continuously for years, are needed. Here we report on the cryogenic system of the QUBIC (Q and U Bolometric Interferometer for Cosmology) experiment: we describe its design, fabrication, experimental optimization and validation in the Technological Demonstrator configuration. The QUBIC cryogenic system is based on a large volume cryostat, using two pulse-tube refrigerators to cool at ~3K a large (~1 m^3) volume, heavy (~165kg) instrument, including the cryogenic polarization modulator, the corrugated feedhorns array, and the lower temperature stages; a 4He evaporator cooling at ~1K the interferometer beam combiner; a 3He evaporator cooling at ~0.3K the focal-plane detector arrays. The cryogenic system has been tested and validated for more than 6 months of continuous operation. The detector arrays have reached a stable operating temperature of 0.33K, while the polarization modulator has been operated from a ~10K base temperature. The system has been tilted to cover the boresight elevation range 20 deg -90 deg without significant temperature variations. The Q and U Bolometric Interferometer for Cosmology (QUBIC) is a ground-based experiment that aims to detect B-mode polarisation anisotropies in the CMB at angular scales around the l=100 recombination peak. Systematic errors make ground-based observations of B modes at millimetre wavelengths very challenging and QUBIC mitigates these problems in a somewhat complementary way to other existing or planned experiments using the novel technique of bolometric interferometry. This technique takes advantage of the sensitivity of an imager and the systematic error control of an interferometer. A cold reflective optical combiner superimposes there-emitted beams from 400 aperture feedhorns on two focal planes. A shielding system composedof a fixed groundshield, and a forebaffle that moves with the instrument, limits the impact of local contaminants. The modelling, design, manufacturing and preliminary measurements of the optical components are described in this paper., Part of a series of 8 papers on QUBIC to be published in a special issue of JCAP. Bolometric interferometry is a novel technique that has the ability to perform spectral imaging. A bolometric interferometer observes the sky in a wide frequency band and can reconstruct sky maps in several sub-bands within the physical band in post-processing of the data. This provides a powerful spectral method to discriminate between the cosmic microwave background (CMB) and astrophysical foregrounds. In this paper, the methodology is illustrated with examples based on the Q \& U Bolometric Interferometer for Cosmology (QUBIC) which is a ground-based instrument designed to measure the B-mode polarization of the sky at millimeter wavelengths. We consider the specific cases of point source reconstruction and Galactic dust mapping and we characterize the point spread function as a function of frequency. We study the noise properties of spectral imaging, especially the correlations between sub-bands, using end-to-end simulations together with a fast noise simulator. We conclude showing that spectral imaging performance are nearly optimal up to five sub-bands in the case of QUBIC., 27 pages, 18 figures. Accepted by JCAP on July 6, 2021.
