In the quest for bioactive compounds from Ganoderma, artificially cultivated fruiting bodies of Ganoderma cf. mastoporum, strain TBRC-BCC 47851 were chemically investigated. The study led to the isolation of three undescribed lanostane triterpenoids (1–3) together with twelve known compounds. The structures were elucidated on the basis of NMR spectroscopic and mass spectrometry data. The new compounds were inactive in the antimalarial and antitubercular activity assays. Seven undescribed lanostane triterpenoids (1 – 7) were isolated from artificially cultivated fruiting bodies of Ganoderma wiiroense , strain TBRC-BCC 60613. The structures were elucidated on the basis of NMR spectroscopic and mass spectrometry data. The C-23 absolute configuration of 1 was determined to be 23 S using the modified Mosher's method. The isolated compounds were evaluated for their antitubercular and antimalarial activities. Compound 7 showed activity against Mycobacterium tuberculosis H37Ra (MIC 50 μg/ml). Colossolactone J (1), an undescribed lanostane triterpenoid was isolated from a natural fruiting body of Ganoderma colossus using silica gel column chromatography and preparative HPLC. Its structure was elucidated on the basis of the spectroscopic method. The absolute configuration was determined by the combination of the modified Mosher's method and detailed NMR data analysis. A maleic anhydride derivative, botryoanhydride (1), and a chromone derivative, botryochromone (2), together with three known chromones, eugenitin (3), 6-hydroxymethyleugenin (4) and 6-methoxymethyleugenin (5), were isolated from cultures of the endophytic fungus BCC 54265 of the family Botryosphaeriaceae. A new seco-tremulane, 11,12-epoxy-5,6-secotremula-1,6(13)-dien-5,12-olide ( 1), was isolated together with the known compounds, conocenolides A ( 2) and B ( 3), tremulenediol A ( 4), tremulenolide A ( 5), and two lanostane triterpenoids, trametenolic acid B ( 6), and pinicolic acid A ( 7), from cultures of the basidiomycete Flavodon flavus BCC 17421. Interconversion of conocenolides A/B was demonstrated. Compound 1 exhibited weak cytotoxic activities, whereas tremulenediol A showed antiplasmodial activity ( IC50 8.6 μg/ml). Six new oblongolides, W1, W2, X, Y, and Z (1-3, 6, 7) and 2-deoxy-4alpha-hydroxyoblongolide X (4), and the known compounds oblongolide (8), oblongolides T, C, and Q (5, 9, 10), and (-)-5-methylmellein were isolated from the endophytic fungus Phomopsis sp. BCC 9789. Compound 7 showed anti-HSV-1 activity (IC50=14 microM) and cytotoxic activities against KB, BC, NCI-H187, and nonmalignant (Vero) cell lines with respective IC50 values of 37, 26, 32, and 60 microM. Cytotoxic activity against the BC cell line was also observed for compound 6, with an IC50 value of 48 microM.
