Biological applications of nanomaterials as delivery carriers have been embedded in traditional biomedical research for decades. Despite lagging behind, recent significant breakthroughs in the use of nanocarriers as tools for plant biotechnology have created great interest. In this Perspective, we review the outstanding recent works in nanocarrier-mediated plant transformation and its agricultural applications. We analyze the chemical and physical properties of nanocarriers determining their uptake efficiency and transport throughout the plant body.

The successful delivery of nanoparticles (NPs) to cancer cells is dependent on various factors, including particle size, shape, surface properties such as hydrophobicity/hydrophilicity, charges, and functional moieties. Tailoring these properties has been explored extensively to enhance the efficacy of NPs for drug delivery. Single-chain polymer nanoparticles (SCNPs), notable for their small size (sub-20 nm) and tunable properties, are emerging as a promising platform for drug delivery. However, the impact of surface charge on the biological performance of SCNPs in cancer cells remains underexplored. In this study, we prepared a library of SCNPs with varying charge types (neutral, anionic, cationic, and zwitterionic), charge densities, charge positions, and crosslinking densities to evaluate their effects on cellular uptake in MCF-7 breast cancer cells. Key findings include that cationic SCNPs are more likely to translocate into cells than neutral, anionic, or zwitterionic counterparts. Furthermore, cellular uptake was enhanced with increased charge density (from 10 to 15 mol%) before reaching a critical point (20 mol%) where excessive positive charge led to NP adhesion to the cell membrane, resulting in cell death. We also found that the position of the charge on the polymer chain also impacted the delivery of NPs to cancer cells, with tadpole-shaped SCNPs achieving the highest uptake. Furthermore, crosslinking density significantly influenced cellular uptake, with SCNPs at 50% crosslinking conversion showing the highest cytosolic localization, while other densities resulted in retention primarily at the cell membrane. This study offers valuable insights into how charge type, density, position, and crosslinking density affect the biological performance of SCNPs, guiding the rational design of more effective and safer drug delivery systems. Covalent drugs can offer significant advantages over non-covalent drugs in terms of pharmacodynamics (i.e., target-binding properties). However, the development of covalent drugs is sometimes hampered by pharmacokinetic limitations (e.g., low bioavailability, rapid metabolism and toxicity due to off-target binding). Polymeric nanoparticles offer a potential solution to these limitations. Delivering covalent drugs via polymeric nanoparticles provides myriad benefits in terms of drug solubility, permeability, lifetime, selectivity, controlled release and the opportunity for synergistic administration alongside other drugs. Antimicrobial resistance is a global healthcare challenge that urgently needs the development of new therapeutic agents. Antimicrobial peptides and mimics thereof are promising candidates but mostly suffer from inherent toxicity issues due to the non‐selective binding of cationic groups with mammalian cells. To overcome this toxicity issue, this work herein reports the synthesis of a smart antimicrobial dendron with masked cationic groups (Gal‐Dendron) that could be uncaged in the presence of β‐galactosidase enzyme to form the activated Enz‐Dendron and confer antimicrobial activity. Enz‐Dendron show bacteriostatic activity toward Gram‐negative (P. aeruginosa and E. coli) and Gram‐positive (S. aureus) bacteria with minimum inhibitory concentration values of 96 µm and exerted its antimicrobial mechanism via a membrane disruption pathway, as indicated by inner and outer membrane permeabilization assays. Crucially, toxicity studies confirmed that the masked prodrug Gal‐Dendron exhibited low hemolysis and is at least 2.4 times less toxic than the uncaged cationic Enz‐Dendron, thus demonstrating the advantage of masking the cationic groups with responsive immolative linkers to overcome toxicity and selectivity issues. Overall, this study highlights the potential of designing new membrane‐disruptive antimicrobial agents that are more biocompatible via the amine uncaging strategy. [ABSTRACT FROM AUTHOR]
