The condensation of 4'-amino-1'H-spiro[cyclohexane-1,2'-naphthalene]-3'-carbonitrile with 3-chloro-3-oxopropanoate and subsequent cyclization of ethyl 3-{(3'-cyano-1'H-spiro[cyclohexane-1,2'-naphthalen]-4'-yl)amino}-3-oxopropanoate developed the synthesis method of ethyl 2-(4-oxo-4,6-dihydro-3H-spiro[benzo[h]quinazoline-5,1'-cyclohexane]-2-yl)acetate. It is revealed that, as a result of hydrolysis of ester, the formed acetic acid was exposed to spontaneous self-decarboxylation with formation 2-methyl-3Hspiro[ benzo[h]quinazoline-5,1'-cyclohexane]-4(6H)-one. By hydrazinolysis of ethyl 3-{(3'-cyano-1'H-spiro[cyclohexane-1,2'-naphthalen]-4'-yl)amino}-3-oxopropanoate 2-(4-oxo-4,6-dihydro-3H-spiro[benzo[h]quinazoline-5,1'-cyclohexane]-2-yl)acetohydrazide was obtained, which is a stable compound. Condensation of 2-methyl-3H-spiro[benzo[h]quinazoline-5,1'-cyclohexane]-4(6H)-one with aromatic aldehydes synthesized (E)-2-styryl-3H-spiro[benzo[h]quinazoline-5,1'-cyclohexane]-4(6H)-ones. Antitumor and antibacterial properties of synthesized compounds are studied. [ABSTRACT FROM AUTHOR]

The method of the synthesis of 2-substituted 5,5-dimethyl-5,6-dihydrobenzo[h]quinazolin-4(3H)-ones starting from 1-amino-3,3- dialkyl-3,4-dihydronaphthalene-2-carbonitriles and 3-chloro-3-oxopropanoate has been developed. It was found that 2-(5,5-dialkyl-4- oxo-3,4,5,6-tetrahydrobenzo[h]quinazolin-2-yl)acetic acids are exposed spontaneous self-decarboxylation giving 5,5-dialkyl-2- methyl-5,6-dihydrobenzo[h]quinazolin-4(3H)-ones.2-(5,5-Dimethyl-4-oxo-3,4,5,6-tetrahydrobenzo[h]quinazolin-2-yl)acetohydrazide was prepared by hydrazinolysis of ethyl 2-(5,5-dimethyl-4-oxo-3,4,5,6-tetrahydrobenzo[h]quinazolin-2-yl)acetate. (E)-2-Styryl-5,5- dialkyl-5,6-dihydrobenzo[h]quinazolin-4(3H)-ones have been synthesized by condensation of 2-methyl-5,6- dihydrobenzo[h]quinazolin-4(3H)-ones with aromatic aldehydes. Starting from 5,5-dimethyl-2-thioxo-2,3,5,6-tetrahydrobenzo[h]quinazolin-4(1H)-one the 2-hydrazino-5,5-dimethyl-5,6-dihydrobenzo[ h] quinazolin-4(3H)-one was synthesized, which is transferred to a 6,6-dimethyl-5,6-dihydrobenzo[h][1,2,4]triazolo[3,4- b]quinazolin-7(11H)-one and 9-mercapto-6,6-dimethyl-5,6-dihydrobenzo[h][1,2,4]triazolo[3,4-b]quinazolin-7(11H)-one. It was revealed that the alkylation of the latter, depending on the amount of halide and alkali monoalkylated or dialkylated products can be obtained. Monosubstituted triazoles are converted to 9,11-dialkylated products containing various substituents. Some of the synthesized compounds showed antibacterial activity against Gram-positive and Gram-negative bacteria. [ABSTRACT FROM AUTHOR]
