N-methyl-D-aspartate receptor (NMDAR) encephalitis is the most common subtype of autoimmune encephalitis characterized by a complex neuropsychiatric syndrome usually including memory impairment. Patients develop an intrathecal immune response against NMDARs with antibodies that presumably bind to the amino-terminal domain of the GluN1 subunit. The therapeutic response to immunotherapy is often delayed. Therefore, new therapeutic approaches for fast neutralization of NMDAR antibodies are needed. Here, we developed fusion constructs consisting of the Fc part of immunoglobulin G and the amino-terminal domains of either GluN1 or combinations of GluN1 with GluN2A or GluN2B. Surprisingly, both GluN1 and GluN2 subunits were required to generate high-affinity epitopes. The construct with both subunits efficiently prevented NMDAR binding of patient-derived monoclonal antibodies and of patient CSF containing high-titre NMDAR antibodies. Furthermore, it inhibited the internalization of NMDARs in rodent dissociated neurons and human induced pluripotent stem cell-derived neurons. Finally, the construct stabilized NMDAR currents recorded in rodent neurons and rescued memory defects in passive-transfer mouse models using intrahippocampal injections. Our results demonstrate that both GluN1 and GluN2B subunits contribute to the main immunogenic region of the NMDAR and provide a promising strategy for fast and specific treatment of NMDAR encephalitis, which could complement immunotherapy.

Sepsis-associated encephalopathy (SAE) is a severe and frequent complication of sepsis causing delirium, coma, and long-term cognitive dysfunction. We identified microglia and C1q complement activation in hippocampal autopsy tissue of patients with sepsis and increased C1q-mediated synaptic pruning in a murine polymicrobial sepsis model. Unbiased transcriptomics of hippocampal tissue and isolated microglia derived from septic mice revealed an involvement of the innate immune system, complement activation, and up-regulation of lysosomal pathways during SAE in parallel to neuronal and synaptic damage. Microglial engulfment of C1q-tagged synapses could be prevented by stereotactic intrahippocampal injection of a specific C1q-blocking antibody. Pharmacologically targeting microglia by PLX5622, a CSF1-R inhibitor, reduced C1q levels and the number of C1q-tagged synapses, protected from neuronal damage and synapse loss, and improved neurocognitive outcome. Thus, we identified complement-dependent synaptic pruning by microglia as a crucial pathomechanism for the development of neuronal defects during SAE.

To describe earth’s former and predict its expected future climate in a general way, we need to understand at least two basic characteristics of the distribution of earth’s temperature, its mean state and its temporal and spatial variance. While there is some confidence in projections of the mean state, earth system models (ESMs) models are suspected to underestimate natural climate variability on longer time scales, as they are developed and evaluated mainly on observational data that only cover a short time period. At low frequencies they show up to two orders of magnitude less temperature variability than data from proxy archives.However, while climate proxies, such as oxygen isotopes from foraminifera retrieved from marine sediments, provide long records, they are sparse in time and space and contain additional non-climate components such as measurement error. A fundamental criticism of proxy data is that correlation between even nearby records is low and therefore they cannot represent the shared climate signal between the two locations.So how to decide whether climate models simulate enough variability at long timescales? We here suggest a new evaluation method for existing climate models. The idea is to combine proxy data, proxy understanding and climate models in such a way that we can derive a plausibility check on the ability of the climate models to reproduce natural climate variability. Considering the high noise levels that we find, what if the proxy records don’t replicate well but still replicate too well enough to have come from a relatively low-variability climate like the climate models?This is the question we answer in this combined data analysis and simulation study. We do this by developing a hypothesis test where transient ESM simulation TraCE21k forms the null hypothesis on the nature of climate variability. Our proxy data base are multiple and high-resolution Holocene oxygen isotopes and Mg/Ca records from marine sediment cores from the Eastern tropical Indian Ocean - a region with a strong variability mismatch between proxy data and climate model simulations.The result of the hypothesis test is that, based on the correlation we find in our records, we have to reject the null hypothesis. That means the possibility that the climate signal that was imprinted in our records was generated by a climate system with TraCE21k-like variability characteristics is very unlikely. We therefore reject the hypothesis that TraCE21k reproduces the climate variability characteristics.We will expand the analysis on different simulations of last generation of ESMs to see if they perform better.

Paleoclimate research in Indonesia has become increasingly important with growing awareness of the critical role of this region for global climate. By reconstructing the past climate trend and variability, paleoclimate research contributes to a more reliable simulation of future Indonesian climate, which has proven difficult owing to the lack of temporally long instrumental records and model deficiencies in reproducing the complex regional setting. After two decades of paleoclimate research, there is strong evidence that melting of the ice sheets and slowdown of the ocean circulation in the North Atlantic, e.g., during Heinrich Events, result in a cooler Northern Hemisphere, and warmer and drier conditions over Sumatra, Java, and the Lesser Sunda Islands. On the other hand, this region experiences a cooler windy season and a wetter rainy season during periods of a generally warmer Northern Hemisphere summer, e.g., during precession minima, with environmental changes being more severe in monsoonal south Indonesia compared with the ever-wet equatorial regions. Abstrak Penelitian iklim purba di Indonesia telah menjadi semakin penting dengan meningkatnya kesadaran akan peran kritis wilayah ini dalam mempengaruhi iklim global. Dengan merekonstruksi kecenderungan iklim masa lalu dan variasinya, penelitian iklim purba memberikan kontribusi dalam simulasi iklim Indonesia masa depan yang lebih dapat diandalkan, hal tersebut sebelumnya telah terbukti sulit karena kurangnya rekaman pengukuran parameter iklim secara teratur berjangka panjang dan kekurang-mampuan model dalam menggambarkan wilayah regional yang kompleks. Setelah dua dekade penelitian iklim purba, terdapat bukti kuat bahwa pencairan tudung es dan pelambatan sirkulasi lautan di Atlantik Utara, seperti selama Peristiwa Heinrich, menghasilkan iklim yang lebih sejuk di belahan bumi utara dan kondisi lebih hangat dan kering di Sumatra, Jawa dan Kepulauan Sunda Kecil. Sebaliknya daerah-daerah ini mengalami musim angin sejuk dan musim hujan basah selama periode yang umumnya hangat di belahan bumi utara pada musim panas, misalnya selama presesi minimal, yang disertai dengan perubahan lingkungan yang drastis selama musim hujan di selatan Indonesia dibandingkan dengan daerah khatulistiwa yang selalu basah.

The COVID-19 pandemic is caused by the betacoronavirus SARS-CoV-2. In November 2021, the Omicron variant was discovered and classified as a variant of concern (VOC). Omicron shows substantially more mutations in the spike protein than any previous variant, mostly in the receptor binding domain (RBD). We analyzed the binding of the Omicron RBD to the human ACE2 receptor (hACE2) and the ability of human sera from COVID-19 patients or vaccinees in comparison to Wuhan, Beta or Delta RBDs variants.All RBDs were produced in insect cells. RBD binding to hACE2 was analyzed by ELISA and microscale thermophoresis (MST). Similarly, sera from 27 COVID-19 patients, 58 fully vaccinated individuals and 16 booster recipients were titrated by ELISA on the fixed RBDs from the original Wuhan strain, Beta, Delta and Omicron VOC.Surprisingly, the Omicron RBD showed a weaker binding to ACE2 compared to Beta and Delta, arguing that improved ACE2 binding is not a likely driver of Omicron evolution. Serum antibody titers were significantly lower against Omicron RBD compared to the original Wuhan strain. However, a difference of 2.5 times was observed in RBD binding while in other studies the neutralization of Omicron SARS-CoV-2 was reduced by a magnitude of 10x and more. These results indicate an immune escape focused on neutralizing antibodies.The reduced binding of sera to Omicron RBD adds evidence that current vaccination protocols may be less efficient against the Omicron variant.

Food allergies are a major health issue worldwide. Modern breeding techniques such as genome editing via CRISPR/Cas9 have the potential to mitigate this by targeting allergens in plants. This study addressed the major allergen Bra j I, a seed storage protein of the 2S albumin class, in the allotetraploid brown mustard (Brassica juncea). Cotyledon explants of an Indian gene bank accession (CR2664) and the German variety Terratop were transformed using Agrobacterium tumefaciens harboring binary vectors with multiple single guide RNAs to induce either large deletions or frameshift mutations in both Bra j I homoeologs. A total of 49 T0 lines were obtained with up to 3.8% transformation efficiency. Four lines had large deletions of 566 up to 790 bp in the Bra j IB allele. Among 18 Terratop T0 lines, nine carried indels in the targeted regions. From 16 analyzed CR2664 T0 lines, 14 held indels and three had all four Bra j I alleles mutated. The majority of the CRISPR/Cas9-induced mutations were heritable to T1 progenies. In some edited lines, seed formation and viability were reduced and seeds showed a precocious development of the embryo leading to a rupture of the testa already in the siliques. Immunoblotting using newly developed Bra j I-specific antibodies revealed the amount of Bra j I protein to be reduced or absent in seed extracts of selected lines. Removing an allergenic determinant from mustard is an important first step towards the development of safer food crops.

Upper water column dynamics in the eastern Arabian Sea were reconstructed in order to investigate changes in the activity of the South Asian / Indian monsoon during the early Pleistocene (c. 1.5–2.7 Ma). We used planktic foraminiferal assemblage records combined with isotopic (δ18O and δ13C) data, Mg/Ca-based sea surface temperatures and seawater δ18O records to estimate changes in surface water conditions at International Ocean Discovery Program (IODP) Site U1457. Our records indicate two distinct regimes of monsoon-induced changes in upper water structure during the periodsc. 1.55–1.65 Ma andc. 1.85–2.7 Ma. We infer that a more stratified upper water column and oligotrophic mixed layer conditions prevailed during the period 1.85–2.7 Ma, which may be due to overall weaker South Asian / Indian winter (NE) and summer (SW) monsoon circulations. The period 1.55–1.65 Ma was characterized by enhanced eutrophication of the mixed layer, which was probably triggered by intensified winter (NE) monsoonal winds. The long-term trend in hydrographic changes during 1.55–1.65 Ma appears to be superimposed by short-term variations, probably reflecting glacial/interglacial changes. We suggest that an intensification of the South Asian / Indian winter monsoon circulation occurred between ∼1.65 Ma and 1.85 Ma, which is most likely due to the development of strong meridional and zonal atmospheric circulations (i.e. Walker Circulation and Hadley Circulation) because of strong equatorial East–West Pacific temperature gradients.

Burkholderia encompasses a group of ubiquitous Gram-negative bacteria that includes numerous saprophytes as well as species that cause infections in animals, immunocompromised patients, and plants. Some species of Burkholderia produce colored, redox-active secondary metabolites called phenazines. Phenazines contribute to competitiveness, biofilm formation, and virulence in the opportunistic pathogen Pseudomonas aeruginosa, but knowledge of their diversity, biosynthesis, and biological functions in Burkholderia is lacking. In this study, we screened publicly accessible genome sequence databases and identified phenazine biosynthesis genes in multiple strains of the Burkholderia cepacia complex, some isolates of the B. pseudomallei clade, and the plant pathogen B. glumae. We then focused on B. lata ATCC 17760 to reveal the organization and function of genes involved in the production of dimethyl 4,9-dihydroxy-1,6-phenazinedicarboxylate. Using a combination of isogenic mutants and plasmids carrying different segments of the phz locus, we characterized three novel genes involved in the modification of the phenazine tricycle. Our functional studies revealed a connection between the presence and amount of phenazines and the dynamics of biofilm growth in flow cell and static experimental systems but at the same time failed to link the production of phenazines with the capacity of Burkholderia to kill fruit flies and rot onions. IMPORTANCE Although the production of phenazines in Burkholderia was first reported almost 70 years ago, the role these metabolites play in the biology of these economically important microorganisms remains poorly understood. Our results revealed that the phenazine biosynthetic pathway in Burkholderia has a complex evolutionary history, which likely involved horizontal gene transfers among several distantly related groups of organisms. The contribution of phenazines to the formation of biofilms suggests that Burkholderia, like fluorescent pseudomonads, may benefit from the unique redox-cycling properties of these versatile secondary metabolites.

The novel betacoranavirus SARS-CoV-2 causes a form of severe pneumonia disease, termed COVID-19 (coronavirus disease 2019). Recombinant human antibodies are proven potent neutralizers of viruses and can block the interaction of viral surface proteins with their host receptors. To develop neutralizing anti-SARS-CoV-2 antibodies, antibody gene libraries from convalescent COVID-19 patients were constructed and recombinant antibody fragments (scFv) against the receptor binding domain (RBD) of the S1 subunit of the viral spike (S) protein were selected by phage display. The selected antibodies were produced in the scFv-Fc format and 30 showed more than 80% inhibition of spike (S1-S2) binding to cells expressing ACE2, assessed by flow cytometry screening assay. The majority of these inhibiting antibodies are derived from the VH3-66 V-gene. The antibody STE90-C11 showed a sub nM IC50 in a plaque-based live SARS-CoV-2 neutralization assay. The in vivo efficacy of the antibody was demonstrated in the Syrian hamster and in the hACE2 mice model using a silenced human IgG1 Fc part. The crystal structure of STE90-C11 Fab in complex with SARS-CoV-2-RBD was solved at 2.0 Å resolution showing that the antibody binds at the same region as ACE2 to RBD. The binding and inhibtion of STE90-C11 is not blocked by many known RBD mutations including N439K, L452R, E484K or L452R+E484Q (emerging B.1.617). STE90-C11 derived human IgG1 with FcγR silenced Fc (COR-101) is currently undergoing Phase Ib/II clinical trials for the treatment of moderate to severe COVID-19.In BriefHuman antibodies were selected from convalescent COVID-19 patients using antibody phage display. The antibody STE90-C11 is neutralizing authentic SARS-CoV-2 virus in vitro and in vivo and the crystal structure of STE90-C11 in complex with SARS-CoV-2-RBD revealed that this antibody is binding in the RBD-ACE2 interface. S1 binding of STE90-C11 and inhibition of ACE2 binding is not blocked by many known RBD mutations.

A 1108.6 m long core was recovered at Site U1457 located on the Indus Fan in the Laxmi Basin of the eastern Arabian Sea during IODP Expedition 355. Shipboard examinations defined five lithologic units (I to V) of the lower Paleocene to Holocene sedimentary sequence. In this study, δ13C values of sedimentary organic matter (SOM) confirm the differentiation of the lithologic units and further divide units III and IV into two subunits (1 and 2). Based on the underlying assumption that the SOM is decided primarily by a mixture of marine and terrestrial origins, δ13CSOM values at Site U1457 provide information on the terrestrial catchment conditions since late Miocene time. Low δ13CSOM values from late Miocene to late Pleistocene times are similar (c. −22.0 ‰) for the most part, reflecting a consistent contribution of terrestrial organic matter from the catchment areas characterized by dominant C3 land plants. Significantly lower δ13CSOM values (c. −24.0 ‰) in Unit III-2 (∼8 to ∼7 Ma) might be due to a greater input of C3 terrestrial organic matter. The increase in δ13CSOM values at ∼7 Ma and the appearance of high δ13CSOM values (c. −18.0 ‰) within Unit III-1 (∼7 to ∼2 Ma) indicate that C4 biomass overwhelmed the terrestrial catchment environment as a result of enhanced terrestrial aridity in the Himalayan foreland. The three-end-member simple mixing model, estimating the relative contributions of SOM from terrestrial C3 and C4 plants and marine phytoplankton, supports our interpretation of the distribution of C3 and C4 land plants in the terrestrial catchment environment.

The Dansgaard-Oeschger oscillations and Heinrich events described in Greenland ice core records are also expressed in the climate of the tropical realm as for instance documented in Arabian Sea sediments. However, little is known about these fluctuations beyond the reach of the Greenland ice cores. Here, we present high-resolution organic- and inorganic geochemical, sedimentological as well as micropaleontological data from two cores retrieved off the coast of Pakistan, extending the monsoon record to the past 200,000 years in millennial scale resolution.The stable oxygen isotope (δ18O) record of the planktic foraminifera G. ruber shows a strong correspondence to Greenland ice core δ18O, whereas the deepwater δ18O signal of benthic foraminifera (U. peregrina and G. affinis) reflects patterns similar to those observed in Antarctic ice core records. Strong shifts in benthic δ18O during stadials are interpreted to show frequent injections of oxygen-rich intermediate water masses of Southern Ocean origin into the Arabian Sea. Alkenone-derived SSTs vary between 23 and 28°C. Highest temperatures were encountered during interglacial MIS 5. Millennial scale SST changes of 2°C magnitude are modulated by long-term SST fluctuations. Interstadials (of glacial phases) and the cold phases of interglacials are characterized by sediments enriched in organic carbon (TOC) whereas sediments with low TOC contents appear during stadials. Abrupt shifts (50-60 year duration) at climate transitions, such as interstadial inceptions, correlate with changes in productivity-related and anoxia-indicating proxies. Interstadial inorganic data consistently show that enhanced fluxes of terrestrial-derived sediments are paralleled by productivity maxima, and are characterized by an increased fluvial contribution from the Indus River. The hydrogen isotopic composition of terrigenous plant waxes indicates that stadials are dry phases whereas humid conditions seem to have prevailed during interstadials. In contrast, stadials are characterized by an increased contribution of aeolian dust probably from the Arabian Peninsula. Heinrich events are especially dry and dusty, indicating a dramatically weakened Indian summer monsoon and increased continental aridity.These results strengthen the evidence that North Atlantic temperature changes and shifts in the hydrological cycle of the Indian monsoon system are closely coupled, and had a massive impact on regional environmental conditions such as river discharge and ocean margin anoxia. These shifts were modulated by changes in the supply of water masses from the Southern Hemisphere.

To identify environmental causes for past changes in vegetation in subtropical East Asia, we present carbon isotope compositions of plant-wax n-alkanes and provide estimates of the C4-plant contribution across the past four glacial terminations and interglacials, based on cores recovered from the northern South China Sea. Our results show a comparable C4-plant contribution between the Last Glacial Maximum (LGM) and the Holocene. An increase of the C4-plant contribution by 15–20% is found for Terminations IV, II and I relative to subsequent interglacial peaks, coeval with an expansion of Cyperaceae and Poaceae. In contrast, Termination V reveals a lower C4-plant contribution than Marine Isotope Stage (MIS) 11c. The data exhibit a long-term trend, with a stepwise increase of the C4-plant contribution across interglacials MIS 11c, 9e, 7e and 1. We suggest that no substantial changes in humidity levels over glacial-interglacial cycles occurred facilitating a similar C3/C4-plant ratio for the LGM and the Holocene. Instead, deglacial sea-level rises caused an extensive development of floodplains and wetlands on the exposed continental shelf, providing habitats for the spread of C4 sedges and grasses. The progressive subsidence of Chinese coastal areas and the broadening of the continental shelf over the late Quaternary explains the nearly absence of C4 plant occurrence during Termination V and a gradual increase of the C4-plant contribution across interglacial peaks. Taken together, changes in coastal environments should be considered when interpreting marine-based vegetation reconstructions from subtropical Asia.

Vegetation dynamics during previous warm interglacial periods shed light on the human impacts on natural ecosystems during the Holocene. However, reliable terrestrial records that span such periods are rare and provide little information on regional scale. Here we present a high-resolution marine pollen record from the northern South China Sea, which reveals that during five peak interglacial periods, Marine Isotope Stages 13a, 11c, 9c, 5e and 1 (the Holocene), the vegetation successions in southern China were similar. At the beginning of each interglacial period, tropical rainforest conifers, which include Dacrydium, Dacrycarpus and Podocarpus, and associated broadleaved taxa, such as Altingia, expanded quickly at the expense of the subtropical/temperate montane conifer Pinus. Near the end of the warm periods, Pinus recovered and the tropical taxa retreated. However, the Holocene displays subtle but significant differences in which the species turnover was interrupted and the rainforest conifers did not fully expanded. The Mg/Ca-based sea surface temperature record from the same site reveals that temperature was the major control of the rise and fall of the peak interglacial vegetation. However, exceptionally high charcoal fluxes during the Holocene suggest that human activities through land-use modifications completely, and possibly permanently, altered the natural vegetation trend five to six thousand years ago. Human land use changed the evolution of vegetation in southern China 6,000 years ago, according to analyses of a high-resolution marine pollen record.

Precipitation isotope reconstructions derived from speleothems and plant waxes are important archives for understanding hydroclimate dynamics. Their climatic significance in East Asia, however, remains controversial. Here we present terrestrial plant-wax stable hydrogen isotope (δDwax) records over periods covering the last four interglacials and glacial terminations from sediment cores recovered from the northern South China Sea (SCS) as an archive of regionally-integrated precipitation isotope changes in Southeast China. Combined with previous precipitation isotope reconstructions from China, we find that the SCS δDwax and Southwest-Central China stalagmite δ 18 O records show relatively enriched and depleted isotopic values, respectively, during interglacial peaks; but relatively similar isotopic variations during most sub-interglacials and glacial periods over the past 430 thousand years. During interglacial peaks, strong summer insolation should have intensified the convection intensity, the isotopic fractionation along moisture trajectories and the seasonality, which are all in favor of causing isotopically-depleted rainfall over the East Asian monsoon regime. These effects in combination with a relatively high proportion of Indian Ocean- versus Pacific-sourced moisture influx should have resulted in strongly depleted precipitation isotopes (stalagmite δ 18 O) over most parts of China. However, Southeast China should have been affected by a relatively low ratio of Indian Ocean- versus Pacific-sourced moisture influx, which dominated over effects yielding depleted precipitation isotopes and led to enriched precipitation isotopes (δDwax). It is thus concluded that glacial boundary conditions and insolation forcing are the two most important factors for causing regional differences in precipitation isotope compositions over subtropical East Asia on orbital timescales.

El Nino/Southern Oscillation (ENSO) events and latitudinal shifts of the Intertropical Convergence Zone (ITCZ) cause large scale hydrologic changes in the western tropical Pacific (WTP). However, there is still a debate on the role of ENSO and ITCZ shifts on the hydroclimate in the WTP and their linkage to Northern Hemisphere climate change on glacial-interglacial timescales. For that reason, we present ice volume-corrected seawater δ18O (δ18Osw-ice, a proxy for salinity) and sea surface temperature (SST) records from IMAGES Core MD06-3047B recovered from the northern WTP to investigate the hydroclimate in the WTP during the last 700 kyr. Planktic foraminifera Globigerinoides ruber based Mg/Ca-SST and δ18Osw-ice records reveal distinct glacial-interglacial variations. The δ18Osw-ice record shows that our study site is characterized by fresher conditions and thus higher precipitation during glacials than during interglacials. Our results are in good agreement with numerical model simulations, which also suggest a glacial freshening in the northern WTP. A glacial freshening of the WTP is most likely related to a substantial weakening of the Atlantic Meridional Overturning Circulation (AMOC) and/or exposure of the Maritime Continent due to sea level changes which resulted in an enhanced Pacific Walker circulation and/or southward shift of the ITCZ. In addition, SST records from the WTP and the eastern tropical Pacific (ETP) suggest an enhanced tropical Pacific zonal SST gradient and thus La Nina-like conditions, which may have contributed to fresher conditions in the WTP during glacial periods. Besides, our rainfall record is in antiphase to the Chinese speleothem δ18O records, supporting the idea of an opposing trend in precipitation changes between the WTP and East Asia on orbital timescales.

Sea surface temperature (SST) and seawater δ 18 O sw records were generated from sediment cores located in the southern Bay of Bengal (SBOB) and the northeastern Arabian Sea (NEAS) to understand glacial to Holocene changes in the hydrography of these regions. This was accomplished through the use of paired δ 18 O and Mg/Ca measurements in planktic foraminifera ( Globigerinoides ruber ) from two sediment cores; SK157-14 in the SBOB (lat. 5°11′N; long. 90°05′E; water-depth 3306 m) and SK148-21 in the NEAS (lat. 21°29′N, long. 67°01′E; water-depth 1900 m). The results suggest significant changes in SST and δ 18 O sw since last glacial period. The glacial SSTs in the SBOB and the NEAS were lower by ~ 2–2.5 °C relative to the Holocene. The deglacial period in both cores is characterized by significant variations in SSTs and seawater δ 18 O sw . The Dansgaard-Oeschger (DO) cycles and Heinrich events are strongly expressed in SST and δ 18 O sw records of the SBOB core. These records support a strong control of atmospheric-oceanic changes in the northern high latitude on thermal state of the SBOB. The SST time series in both these regions indicates a sudden and abrupt increase in temperature at the end of last glacial maximum. The SST time series in the NEAS core SK 148-21 reveals an intensification of the northeast monsoon during the last glacial maximum.

Mg/Ca and stable oxygen isotope compositions (δ18O) of planktic foraminifera tests are commonly used as proxies to reconstruct past ocean conditions including variations in the vertical water column structure. Accurate proxy calibrations require thorough regional studies, since parameters such as calcification depth and temperature of planktic foraminifera depend on local environmental conditions. Here we present radiocarbon‐dated, modern surface sediment samples and water column data (temperature, salinity, and seawater δ18O) from the Western Pacific Warm Pool. Seawater δ18O (δ18OSW) and salinity are used to calculate individual regressions for western Pacific surface and thermocline waters (δ18OSW = 0.37 × S‐12.4 and δ18OSW = 0.33 × S‐11.0). We combine shell δ18O and Mg/Ca with water column data to estimate calcification depths of several planktic foraminifera and establish regional Mg/Ca‐temperature calibrations. Globigerinoides ruber, Globigerinoides elongatus, and Globigerinoides sacculifer reflect mixed layer conditions. Pulleniatina obliquiloculata and Neogloboquadrina dutertrei and Globorotalia tumida preserve upper and lower thermocline conditions, respectively. Our multispecies Mg/Ca‐temperature calibration (Mg/Ca = 0.26exp0.097*T) matches published regressions. Assuming the same temperature sensitivity in all species, we propose species‐specific calibrations that can be used to reconstruct upper water column temperatures. The Mg/Ca temperature dependencies of G. ruber, G. elongatus, and G. tumida are similar to published equations. However, our data imply that calcification temperatures of G. sacculifer, P. obliquiloculata, and N. dutertrei are exceptionally warm in the western tropical Pacific and thus underestimated by previously published calibrations. Regional Mg/Ca‐temperature relations are best described by Mg/Ca = 0.24exp0.097*T for G. sacculifer and by Mg/Ca = 0.21exp0.097*T for P. obliquiloculata and N. dutertrei.

The novel betacoranavirus SARS-CoV-2 causes a form of severe pneumonia disease, termed COVID-19 (coronavirus disease 2019). Recombinant human antibodies are proven potent neutralizers of viruses and can block the interaction of viral surface proteins with their host receptors. To develop neutralizing anti-SARS-CoV-2 antibodies, antibody gene libraries from convalescent COVID-19 patients were constructed and recombinant antibody fragments (scFv) against the receptor binding domain (RBD) of the S1 subunit of the viral spike (S) protein were selected by phage display. The selected antibodies were produced in the scFv-Fc format and 30 showed more than 80% inhibition of spike (S1-S2) binding to cells expressing ACE2, assessed by flow cytometry screening assay. The majority of these inhibiting antibodies are derived from the VH3-66 V-gene. The antibody STE90-C11 showed an IC50 of 0.56 nM in a plaque-based live SARS-CoV-2 neutralization assay. The crystal structure of STE90-C11 in complex with SARS-CoV-2-RBD was solved at 2.0 A resolution showing that the antibody binds at the same region as ACE2 to RBD. In contrast to other published anti-SARS-CoV-2 antibodies, the binding of STE90-C11 is not blocked by known RBD mutations, endowing our antibody with higher intrinsic resistance to those possible escape mutants. The complex, named the Nataraja Slide, was drilled by International Ocean Discovery Program (IODP) Expedition 355 in two locations where it is similar to 300 m (Site U1456) and similar to 200 m thick (Site U1457). The top of this mass-transport complex is defined by the presence of both reworked microfossil assemblages and deformation structures, such as folding and faulting. The deposit consists of two main phases of mass wasting, each consisting of smaller pulses, with generally fining-upward cycles, all emplaced just prior to 10.8 Ma based on biostratigraphy. The base of the deposit at each site is composed largely of matrix-supported carbonate breccia that is interpreted as the product of debris-flows. In the first phase, these breccias alternate with well-sorted calcar-enites deposited from a high-energy current, coherent limestone blocks that are derived directly from the Indian continental margin, and a few clastic mudstone beds. In the second phase, at the top of the deposit, muddy turbidites dominate and become increasingly more siliciclastic. At Site U1456, where both phases are seen, a 20-m section of hemipelagic mudstone is present, overlain by a similar to 40-m-thick section of calcarenite and slumped interbedded mud and siltstone. Bulk sediment geochemistry, heavy-mineral analysis, clay mineralogy, isotope geochemistry, and detrital zircon U-Pb ages constrain the provenance of the clastic, muddy material to being reworked, Indus-derived sediment, with input from western Indian rivers (e.g., Narmada and Tapti rivers), and some material from the Deccan Traps. The carbonate blocks found within the breccias are shallow-water limestones from the outer western Indian continental shelf, which was oversteepened from enhanced clastic sediment delivery during the mid-Miocene. The final emplacement of the material was likely related to seismicity as there are modern intraplate earthquakes close to the source of the slide. Although we hypothesize that this area is at low risk for future mass wasting events, it should be noted that other oversteepened continental margins around the world could be at risk for mass failure as large as the Nataraja Slide. Sediment depositional patterns along the upper continental slope of the northern South China Sea (SCS) have been studied using two sediment cores (GeoB16601-6, 20°09.07′N, 116°14.38′E, 1012 m water depth and GeoB16602-4, 18°57.12′N, 113°42.64′E, 951 m water depth) recovered during RV SONNE Cruise SO-221 “INVERS”. Sediment cores were analyzed for bulk sediment element composition by X-Ray Fluorescence (XRF) core scanning and clay mineral assemblage by X-Ray Diffraction (XRD). The bulk sediment element and the clay mineral compositions of the two cores suggest similar depositional patterns between 50 kyr BP and ~ 29 kyr BP, and ~ 14 kyr BP to present, but a clear difference in depositional patterns during the last glacial and deglacial (~ 29–14 kyr BP) when sea level was below − 90 m. Between ~ 29–14 kyr BP, a higher kaolinite percentage in core GeoB16602-4 in comparison to core GeoB16601-6 is interpreted to reflect a higher contribution of clay supplied by the Pearl River to core site GeoB16602-4. In contrast, core GeoB16601-6 received less Pearl River supplied clayey material during the same period. When the sea level was below − 90 m during the last glacial lowstand, the detrital fine-grained materials supplied by the Pearl River were mostly transported by coastal currents to the southwest, resulting in a higher contribution of Pearl River discharged material at site GeoB16602-4 than at site GeoB16601-6. We suggest that sea-level induced modifications of the land-ocean distribution together with changes in the palaeo-physiographic conditions, such as the proximity of the palaeo-rivers to the individual core sites might be responsible for the different sediment depositional patterns in the study area. Thus, the overriding control of sea-level induced changes on the sediment depositional environment might mask climate-related changes in sediment depositional pattern in the northern SCS.

The molar ratio of magnesium to calcium (Mg/Ca) in foraminiferal calcite is a widely used proxy for reconstructing past seawater temperatures. Thorough cleaning of tests is required before analysis to remove contaminant phases such as clay and organic matter. We have adapted a commercial pipette robot to automate an established cleaning procedure, the “Mg-cleaning” protocol of Barker et al. (2003). Efficiency of the automated nine-step method was assessed through monitoring Al/Ca of trial samples (GeoB4420-2 core catcher). Planktonic foraminifera Globigerinoides ruber, Globigerinoides sacculifer, and Neogloboquadrina dutertrei from this sample gave Mg/Ca consistent with the habitat range of the three species, and 40–60% sample recovery after cleaning. Monsoons are the dominant seasonal mode of climate variability in the tropics and are critically important conveyors of atmospheric moisture and energy at a global scale. Predicting monsoons, which have profound impacts on regions that are collectively home to more than 70 per cent of Earth's population, is a challenge that is difficult to overcome by relying on instrumental data from only the past few decades. Palaeoclimatic evidence of monsoon rainfall dynamics across different regions and timescales could help us to understand and predict the sensitivity and response of monsoons to various forcing mechanisms. This evidence suggests that monsoon systems exhibit substantial regional character.

Variations in thermoclinal conditions play an important role in the evolution of the Western Pacific Warm Pool (WPWP) and in connecting the tropical and extratropical regions of the Pacific Ocean. However, secular thermoclinal changes in the northwestern part of the WPWP and their forcing mechanisms are not yet well understood, particularly on orbital timescales. In this study, we report thermoclinal variation at Site MD06-3047 B in the western Philippine Sea over the last 700 kyr. Our data show that variations in thermocline temperature and salinity in the study area were correlated with changes in Earth’s obliquity, with warmer and saltier (colder and fresher) thermocline waters corresponding to high (low) Earth-axial obliquity. At high obliquity, increases in thermocline temperature and salinity may have been linked to advection of warm North Pacific tropical water. In the study area, a lower (higher) vertical temperature gradient in the upper water column suggests a deeper (shallower) thermocline during glacials (interglacials). The main factor leading to thermoclinal deepening during glacials was strengthening of the East Asian winter monsoon (EAWM), and more La Nina-like conditions may have contributed to concurrent subsurface warming and thus thermoclinal deepening. In-phase thermoclinal deepening in the northern and peri-equatorial southern WPWP during glacial stages may have been triggered by changes in glacial boundary conditions, such as increased ice sheet thickness, through intensification of the EAWM and strengthening of the tropical Trade Winds.

International audience; Surface and thermocline conditions of the Western Pacific Warm Pool (WPWP) reflect changes in regional and basin scale ocean and atmosphere circulations and in turn may affect climate globally. Previous studies suggest that a range of factors influences the WPWP on different timescales, however the precise forcings and mechanisms are unclear. Combining surface and thermocline records from sediment cores offshore Papua New Guinea we explore the influence of local and remote processes on the WPWP in response to astronomical forcing and changing glacial-interglacial boundary conditions over the past 110 kyr. We find that thermocline temperatures change with variations in Earth's obliquity with higher temperatures coinciding with high obliquity, which is attributed to variations in subduction and advection of the South Pacific Tropical Water. In contrast, rainfall variations associated with meridional migrations of the Intertropical Convergence Zone are primarily driven by changes in insolation due to precession. Records of bulk sedimentary Ti/Ca and foraminiferal Nd/Ca indicate an additional influence of obliquity, which, however, cannot unambiguously be related to changes in precipitation. Finally, our results suggest a thermocline deepening during the Last Glacial Maximum (LGM). A compilation of available proxy records illustrates a dipole-like pattern of LGM thermocline depth anomalies with a shoaling (deepening) in the northern (southern) WPWP. A comparison of the proxy compilation with an ensemble of Paleoclimate Model Intercomparison Project (PMIP) climate model simulations reveals that the spatial pattern of LGM thermocline depth anomalies is mainly attributable to a contraction of the Pacific Walker circulation on its western side.

Climate phenomena like the monsoon system, El Nino Southern Oscillation (ENSO) and the Indian Ocean Dipole (IOD) are interconnected via various feedback mechanisms and control the climate of the Indian Ocean and its surrounding continents on various timescales. The eastern tropical Indian Ocean is a key area for the interplay of these phenomena and for reconstructing their past changes and forcing mechanisms. Here we present records of upper ocean thermal gradient, thermocline temperatures (TT) and relative abundances of planktic foraminifera in core SO 189-39KL taken off western Sumatra (0°47.400′ S, 99°54.510′ E) for the last 8 ka that we use as proxies for changes in upper ocean structure. The records suggest a deeper thermocline between 8 ka and ca 3 ka compared to the late Holocene. We find a shoaling of the thermocline after 3 ka, most likely indicating an increased occurrence of upwelling during the late Holocene compared to the mid-Holocene which might represent changes in the IOD-like mean state of the Indian Ocean with a more negative IOD-like mean state during the mid-Holocene and a more positive IOD-like mean state during the past 3 ka. This interpretation is supported by a transient Holocene climate model simulation in which an IOD-like mode is identified that involves an insolation-forced long-term trend of increasing anomalous surface easterlies over the equatorial eastern Indian Ocean.

Modern variability in upwelling off southern Indonesia is strongly controlled by the Australian-Indonesian monsoon and the El Nino–Southern Oscillation, but multidecadal to centennial-scale variations are less clear. We present high-resolution records of upper water column temperature, thermal gradient, and relative abundances of mixed layer- and thermocline-dwelling planktonic foraminiferal species off southern Indonesia for the past two millennia that we use as proxies for upwelling variability. We find that upwelling was generally strong during the Little Ice Age (LIA) and weak during the Medieval Warm Period (MWP) and the Roman Warm Period (RWP). Upwelling is significantly anticorrelated to East Asian summer monsoonal rainfall and the zonal equatorial Pacific temperature gradient. We suggest that changes in the background state of the tropical Pacific may have substantially contributed to the centennial-scale upwelling trends observed in our records. Our results implicate the prevalence of an El Nino-like mean state during the LIA and a La Nina-like mean state during the MWP and the RWP.

The final phase of the closure of the Panamanian Gateway and the intensification of Northern Hemisphere Glaciation (NHG) both occurred during the Late Pliocene. Glacial–interglacial (G–IG) variations in sea level might, therefore, have had a significant impact on the remaining connections between the East Pacific and the Caribbean. Here, we present combined foraminiferal Mg/Ca and δ 18O measurements from Ocean Drilling Program (ODP) Site 1241 from the East Pacific and ODP Site 999 from the Caribbean. The studied time interval covers the first three major G–IG Marine Isotope Stages (MIS 95–100, ∼2.5 Ma∼2.5 Ma) after the intensification of NHG. Analyses were performed on the planktonic foraminifera Neogloboquadrina dutertrei and Globigerinoides sacculifer, representing water mass properties in the thermocline and the mixed-layer, respectively. Changes in sea water temperature, relative salinity, and water column stratification strongly suggest that the Panamanian Gateway temporarily closed during glacial MIS 98 and 100, as a result of changes in ice volume equivalent to a drop in sea level of 60–90 m. Reconstructed sea surface temperatures (SST) from G. sacculifer show a glacial decrease of 2.5 °C at Site 1241, but increases of up to 3 °C at Site 999 during glacial MIS 98 and 100 suggesting that the Panamanian Gateway closed during these glacial periods. The Mg/Ca-temperatures of N. dutertrei remain relatively stable in the East Pacific, but do show a 3 °C warming in the Caribbean at the onset of these glacial periods suggesting that the closing of the gateway also changed the water column stratification. We infer that the glacial closure of the gateway allowed the Western Atlantic Warm Pool to extend into the southern Caribbean, increasing SST (G. sacculifer) and deepening the thermocline (N. dutertrei). Additionally, ice volume appears to have become large enough during MIS 100 to survive the relatively short lasting interglacial MIS 99 so that the gateway remained closed. Towards the end of MIS 98, during MIS 97 and into MIS 96 temperatures on both sides are mostly similar suggesting water masses exchanged again. Additionally, Caribbean variations in SST and δ18Owater follow a precession-like cyclicity rather than the obliquity-controlled variations characteristic of the East Pacific and many other tropical areas, suggesting that regional atmospheric processes related to the trade winds and the InterTropical Convergence Zone (ITCZ) had a dominant impact in the Caribbean.

The Australian–Indonesian monsoon has a governing influence on the agricultural practices and livelihood in the highly populated islands of Indonesia. However, little is known about the factors that have influenced past monsoon activity in southern Indonesia. Here, we present a ∼6000 years high-resolution record of Australian-Indonesian summer monsoon (AISM) rainfall variations based on bulk sediment element analysis in a sediment archive retrieved offshore northwest Sumba Island (Indonesia). The record suggests lower riverine detrital supply and hence weaker AISM rainfall between 6000 yr BP and ∼3000 yr BP compared to the Late Holocene. We find a distinct shift in terrigenous sediment supply at around 2800 yr BP indicating a reorganization of the AISM from a drier Mid Holocene to a wetter Late Holocene in southern Indonesia. The abrupt increase in rainfall at around 2800 yr BP coincides with a grand solar minimum. An increase in southern Indonesian rainfall in response to a solar minimum is consistent with climate model simulations that provide a possible explanation of the underlying mechanism responsible for the monsoonal shift. We conclude that variations in solar activity play a significant role in monsoonal rainfall variability at multi-decadal and longer timescales. The combined effect of orbital and solar forcing explains important details in the temporal evolution of AISM rainfall during the last 6000 years. By contrast, we find neither evidence for volcanic forcing of AISM variability nor for a control by long-term variations in the El Nino-Southern Oscillation (ENSO).

The response of the tropical climate in the Indian Ocean realm to abrupt climate change events in the North Atlantic Ocean is contentious. Repositioning of the intertropical convergence zone is thought to have been responsible for changes in tropical hydroclimate during North Atlantic cold spells, but the dearth of high-resolution records outside the monsoon realm in the Indian Ocean precludes a full understanding of this remote relationship and its underlying mechanisms. Here we show that slowdowns of the Atlantic meridional overturning circulation during Heinrich stadials and the Younger Dryas stadial affected the tropical Indian Ocean hydroclimate through changes to the Hadley circulation including a southward shift in the rising branch (the intertropical convergence zone) and an overall weakening over the southern Indian Ocean. Our results are based on new, high-resolution sea surface temperature and seawater oxygen isotope records of well-dated sedimentary archives from the tropical eastern Indian Ocean for the past 45,000 years, combined with climate model simulations of Atlantic circulation slowdown under Marine Isotope Stages 2 and 3 boundary conditions. Similar conditions in the east and west of the basin rule out a zonal dipole structure as the dominant forcing of the tropical Indian Ocean hydroclimate of millennial-scale events. Results from our simulations and proxy data suggest dry conditions in the northern Indian Ocean realm and wet and warm conditions in the southern realm during North Atlantic cold spells.

Despite the significance of denitrification in the Arabian Sea, we have no long-term record of its evolution spanning the past several million years. Here, we present the first record of denitrification evolution since Late Miocene (∼10.2 Ma) in the Eastern Arabian Sea, where the SAM generates moderate surface water productivity, based on the samples retrieved during the International Ocean Discovery Program (IODP) Expedition 355. We find that (i) the SAM was persistently weaker from ∼10.2 to 3.1 Ma; it did not intensify at ∼8 Ma in contrast to a few previous studies, (ii) on tectonic timescale, both the SAM and the East Asian Monsoon (EAM) varied synchronously, (iii) the first evidence of denitrification and productivity/SAM intensification was at ∼3.2-2.8 Ma that coincided with Mid-Pliocene Warm Period (MPWP), and (iv) the modern strength of the OMZ where denitrification is a permanent feature was attained at ∼1.0 Ma. The Indian summer monsoon (ISM) brings most of the annual precipitation to the densely populated region in southern Asia. For the agricultural development and economic prosperity of the region, it is therefore vital to assess the variability of the monsoon system on societal relevant decadal- to centennial time scales. This might help to better understand how potential driving forces might be controlling ISM variability and how it might develop under future climate scenarios. Here we present a study of a sediment core from the northern Oman margin, revealing early- to mid Holocene ISM conditions on a near 20-year resolution. We assess multiple independent proxies indicative of sea surface temperatures (SST) during the upwelling season together with bottom water conditions. We use geochemical parameters, transfer functions of planktic foraminiferal assemblages and Mg/Ca paleothermometry and find evidence corroborating previous studies that upwelling intensity varies significantly in coherence to solar sunspot cycles. The dominant ~80–90-year Gleissberg cycle was apparently also affecting bottom water oxygen conditions. Although the interval from 8.4 to 5.8 ka B.P. is relatively short, the gradually decreasing trend of summer monsoon conditions was interrupted by short events of intensified ISM conditions. Results from both independent SST proxies are linked to phases of weaker OMZ conditions and enhanced carbonate preservation. This indicates that atmospheric forcing was intimately linked to bottom water properties and state of the OMZ on decadal time scales.

We use new and published data to discuss the East Asian winter monsoon (EAWM) development and the relationship between the EAWM and the East Asian summer monsoon (EASM) since the Last Glacial Maximum (LGM) with special emphasis on the Holocene. The depth of the mixed layer in the northern South China Sea (SCS) is strongly modulated by the seasonal reversing monsoon winds with deeper vertical mixing during the winter monsoon season and less mixing during the summer monsoon season. Reconstructing the relative depth of the mixed layer thus represents a sensitive diagnostic variable of past changes in the EAWM strength. We derived estimates of the vertical thermal structure from the difference (∆T) in Mg/Ca-based temperatures between surface- ( Globigerinoides ruber ) and thermocline-dwelling ( Pulleniatina obliquiloculata) planktonic foraminifera, and the temperature difference between alkenone and P. obliquiloculata in core MD05-2904 located in the northern SCS. Our ∆T estimates suggest enhanced upper water column mixing due to strengthened EAWM winds during the last glacial, consistent with other marine and terrestrial records as well as model simulations that also suggest a dominance of the cold and dry northwesterly winter monsoon system during the last glacial. Our ∆T estimates reveal that the EAWM was weaker during the Holocene compared to the last glacial. We find that EAWM and EASM changes show an inverse behaviour during the last glacial that is consistent with previous studies but co-varied during the Holocene. It can be inferred from these findings that the variability of the East Asian monsoon during the Holocene is more complex than the simple anti-phase relationship between summer and winter monsoon as assumed previously. The early-mid Holocene strengthening of both the EAWM and the EASM is most likely attributed to the higher summer insolation that resulted in a stronger seasonal contrast.

The present study is the first study on the stable oxygen isotope composition of the photosynthetic calcareous-walled dinoflagellate species Thoracosphaera heimii off NW Africa during the last 45,000 yr. T. heimii based temperature estimates of sediment core GeoB 8507-3 were compared with those obtained from the stable oxygen isotopes of the planktic foraminifera Globigerina bulloides and Globigerinoides ruber (pink), and the Mg/Ca ratio of G. ruber (pink). We show that the isotopic composition of T. heimii and the temperature estimates based on the equation for inorganically precipitated calcite provide comparable results to those obtained from G. ruber (pink) isotopes and Mg/Ca ratios with exception of the Early Holocene and the Younger Dryas. The recently proposed palaeotemperature equation of Zonneveld et al. (2007), however, provides unrealistic temperature reconstructions that are about 16 °C lower than those based on planktic foraminifera. Thus, this equation needs to be revised. The difference between T. heimii and G. bulloides isotopic and temperature reconstructions can be ascribed to differences in the ecology of both species, especially with regard to their depth habitat and/or seasonal production in the research area. All temperature proxies suggest comparable conditions during the last glacial and Holocene. Small differences between the reconstructed temperature values of T. heimii and the other proxies can be explained by differences in seasonal production of the individual species. The relatively low temperatures recorded by T. heimii at about 15,000 to 8,000 yr BP are interpreted to reflect an increase in duration and/or intensity of the upwelling in the vicinity of the core site in comparison to the last glacial, with an abrupt and strong decrease of upwelling intensity and/or duration during the Early Holocene and the Younger Dryas.

Millennial-scale variations of the East Asian winter monsoon (EAWM) remain elusive due to sparse and controversial reconstructions. By compiling a variety of alkenone-based sea surface temperature (SST) estimates, we find that the west–east SST gradient in the southern South China Sea (SCS) well documents the temporal dynamics of the winter “cold tongue” off the southern Vietnam and by inference, variations in the EAWM intensity over the past 26 ka. Our results reveal that the winter “cold tongue” SSTs were significantly colder during Heinrich event 1 and the Younger Dryas event, resulting in an increased west–east SST gradient in the southern SCS due to a strengthened EAWM. Within dating uncertainties, an intensified EAWM during cold stadials was coeval with the shutdown or a reduction in strength of the Atlantic meridional overturning circulation (AMOC), exhibiting a strong linkage between the AMOC and the EAWM system. The west–east SST gradient also indicates an enhanced EAWM during the early Holocene, which may be induced by postglacial ice-sheet dynamics and a strong seasonal contrast in solar insolation. Our findings suggest that the EAWM was probably modulated by a complex interplay between the AMOC, solar insolation and ice-sheet dynamics on sub-orbital time scales.

The Late Pliocene is thought to be characterized by the simultaneous intensification of both the East Asian winter monsoon (EAWM) and East Asian summer monsoon (EASM). However, the evolution of the EASM during the Pliocene remains still controversial and only little is known about the dynamics of the EASM during the Pliocene on orbital time scales. Here we use clay mineral assemblages in sediments from Ocean Drilling Program (ODP) Site 1143 in the southern South China Sea (SCS) to obtain proxy records of past changes in the EASM climate during the Pliocene. Provenance analysis suggests that illite, chlorite and kaolinite originated mainly from the Mekong River drainage area. Smectite was derived mainly from the Indonesian islands. The kaolinite/illite ratio and the chemical index of alteration (CIA) of siliciclastic sediments allowed us to reconstruct the history of chemical weathering and physical erosion of the Mekong River drainage area and thus, the evolution of,the EASM during the Pliocene. Our clay minerals proxy data suggests a stronger EASM during the Early Pliocene than during the Late Pliocene. We propose that the long-term evolution of the EASM has been driven by global cooling rather than the uplift of the Tibetan Plateau. Spectral analysis of kaolinite/ illite ratio displays a set of strong periodicities at 100 ka, 30 ka, 28 ka, 25 ka, and 22 ka. with no clear obliquityrelated signal. Our study suggests that the Pliocene EASM intensity on orbital time scales is not only controlled by the Northern Hemisphere summer insolation, but also strongly influenced by equatorial Pacific ENSO-like ocean atmosphere dynamics. Surface and thermocline conditions of the eastern tropical Indian Ocean were reconstructed through the past glacial–interglacial cycle by using Mg/Ca and alkenone-paleothermometry, stable oxygen isotopes of calcite and seawater, and terrigenous fraction performed on sediment core GeoB 10038-4 off SW Sumatra (∼6°S, 103°E, 1819 m water depth). Results show that annual mean surface and thermocline temperatures varied differently and independently, and suggest that surface temperatures have been responding to southern high-latitude climate, whereas the more variable thermocline temperatures were remotely controlled by changes in the thermocline temperatures of the North Indian Ocean. Except for glacial terminations, salinity proxies indicate that changing intensities of the boreal summer monsoon did not considerably affect annual mean conditions off Sumatra during the past 133,000 years. Our results do not show a glacial–interglacial pattern in the thermocline conditions and reject a linear response of the tropical Indian Ocean thermocline to mid- and high-latitude climate change. Alkenone-based surface temperature estimates varied in line with the terrigenous fraction of the sediment and the East Asian winter monsoon proxy records at the precession band suggestive of monsoon (sea level) to be the dominant control on alkenone temperatures in the eastern tropical Indian Ocean on sub-orbital (glacial–interglacial) timescales.
