The ability of the ancient Egyptians to preserve the human body through embalming has not only fascinated people since antiquity, but also has always raised the question of how this outstanding chemical and ritual process was practically achieved. Here we integrate archaeological, philological and organic residue analyses, shedding new light on the practice and economy of embalming in ancient Egypt. We analysed the organic contents of 31 ceramic vessels recovered from a 26th Dynasty embalming workshop at Saqqara1,2. These vessels were labelled according to their content and/or use, enabling us to correlate organic substances with their Egyptian names and specific embalming practices. We identified specific mixtures of fragrant or antiseptic oils, tars and resins that were used to embalm the head and treat the wrappings using gas chromatography–mass spectrometry analyses. Our study of the Saqqara workshop extends interpretations from a micro-level analysis highlighting the socio-economic status of a tomb owner3–7 to macro-level interpretations of the society. The identification of non-local organic substances enables the reconstruction of trade networks that provided ancient Egyptian embalmers with the substances required for mummification. This extensive demand for foreign products promoted trade both within the Mediterranean8–10 (for example, Pistacia and conifer by-products) and with tropical forest regions (for example, dammar and elemi). Additionally, we show that at Saqqara, antiu and sefet—well known from ancient texts and usually translated as ‘myrrh’ or ‘incense’11–13 and ‘a sacred oil’13,14—refer to a coniferous oils-or-tars-based mixture and an unguent with plant additives, respectively.

Brain tissue is ubiquitous in the archaeological record. Multiple, independent studies report the finding of black, resinous or shiny brain tissue, and Petrone et al. [2020 “Heat-induced Brain Vitrification from the Vesuvius Eruption in C.E. 79.” N Engl J Med. 382: 383–384; doi:10.1056/NEJMc1909867] raise the intriguing prospect of a role for vitrification in the preservation of ancient biomolecules. However, Petrone et al. (2020) have not made their raw data available, and no detailed laboratory or analytical methodology is offered. Issues of contamination and misinterpretation hampered a decade of research in biomolecular archaeology, such that addressing these sources of bias and facilitating validation of specious findings has become both routine and of paramount importance in the discipline. We argue that the evidence they present does not support their conclusion of heat-induced vitrification of human brain tissue, and that future studies should share palaeoproteomic data in an open access repository to facilitate comparative analysis of the recovery of ancient proteins and patterns of their degradation.

Physical interactions between proteins are essential for most biological processes governing life. However, the molecular determinants of such interactions have been challenging to understand, even as genomic, proteomic, and structural data grows. This knowledge gap has been a major obstacle for the comprehensive understanding of cellular protein-protein interaction (PPI) networks and for thede novodesign of protein binders that are crucial for synthetic biology and translational applications. We exploit a geometric deep learning framework operating on protein surfaces that generates fingerprints to describe geometric and chemical features critical to drive PPIs. We hypothesized these fingerprints capture the key aspects of molecular recognition that represent a new paradigm in the computational design of novel protein interactions. As a proof-of-principle, we computationally designed severalde novoprotein binders to engage four protein targets: SARS-CoV-2 spike, PD-1, PD-L1, and CTLA-4. Several designs were experimentally optimized while others were purely generatedin silico, reaching nanomolar affinity with structural and mutational characterization showing highly accurate predictions. Overall, our surface-centric approach captures the physical and chemical determinants of molecular recognition, enabling a novel approach for thede novodesign of protein interactions and, more broadly, of artificial proteins with function.

Protein engineering and synthetic biology stand to benefit immensely from recent advances in silico tools for structural and functional analyses of proteins. In the context of designing novel proteins, current in silico tools inform the user on individual parameters of a query protein, with output scores/metrics unique to each parameter. In reality, proteins feature multiple "parts"/functions and modification of a protein aimed at altering a given part, typically has collateral impact on other protein parts. A system for prediction of the combined effect of design parameters on the overall performance of the final protein does not exist. Function2Form Bridge (F2F-Bridge) attempts to address this by combining the scores of different design parameters pertaining to the protein being analyzed into a single easily interpreted output describing overall performance. The strategy comprises of (a) a mathematical strategy combining data from a myriad of in silico tools into an OP-score (a singular score informing on a user-defined overall performance) and (b) the F2F Plot, a graphical means of informing the wetlab biologist holistically on designed construct suitability in the context of multiple parameters, highlighting scope for improvement. F2F predictive output was compared with wetlab data from a range of synthetic proteins designed, built, and tested for this study. Statistical/machine learning approaches for predicting overall performance, for use alongside the F2F plot, were also examined. Comparisons between wetlab performance and F2F predictions demonstrated close and reliable correlations. This user-friendly strategy represents a pivotal enabler in increasing the accessibility of synthetic protein building and de novo protein design.

Dental calculus is now widely used to recover information on items ingested in the past. It is particularly valuable in the earlier Paleolithic, where recovered data may represent the only evidence for plant use. Several recovery methods are used and each one produces different results. Biomolecular markers and genetic material recovered from dental calculus is providing new data on identifiable dietary and medicinal items and human microbial communities. The recovery of microfossils, in particular, starch granules, has triggered a new awareness of the role of plants in the diet throughout the Paleolithic. However, the minute amount of material recovered has little relationship with food eaten during a person's life, while salivary amylase breaks down cooked starch. Therefore, broader dietary interpretations and detection of cooked food are problematic. The study of ancient dental calculus holds great potential to recover information about past lives, within realistic parameters.

Besides grid-to-vehicle (G2V) and vehicle-to-grid (V2G) functions, the battery of an electric vehicle (EV) also has the specific feature of mobility. This means that EVs not only have the potential to utilize the storage of cheap electricity for use in high energy price periods, but can also transfer energy from one place to another place. Based on these special features of an EV battery, a new EV energy scheduling method has been developed and is described in this article. The approach is aimed at optimizing the utilization EV energy for EVs that are regularly used in multiple places. The objective is to minimize electricity costs from multiple meter points. This work applies real data in order to analyze the effectiveness of the method. The results show that by applying the control strategy presented in this paper at locations where the EVs are parked, the electricity cost can be reduced without shifting the demand and lowering customer’s satisfaction. The effects of PV size and number of EVs on our model are also analyzed in this paper. This model has the potential to be used by energy system designers as a new perspective to determine optimal sizes of generators or storage devices in energy systems.

Dental calculus (mineralized dental plaque) was first recognised as a potentially useful archaeological deposit in the 1970s, though interest in human dental calculus as a resource material has increased sharply in the past few years. The majority of recent research has focused on the retrieval of plant microfossils embedded in its matrix and interpretation of these finds as largely the result of deliberate consumption of plant-derived food. However, while most of the material described in published works does represent food, dental calculus is in fact a "depositional environment" as material can enter the mouth from a range of sources. In this respect, it therefore represents an archaeological deposit that can also contain extensive non-dietary debris. This can comprise a wide variety of cultural and environmental material which reaches the mouth and can become embedded in dental calculus through alternative pathways. Here, we explore the human behaviors and activities besides eating that can generate a flux of particles into the human mouth, the broad range of additional cultural and environmental information that can be obtained through the analysis and contextualisation of this material, and the implications of the additional pathways by which material can become embedded in dental calculus.

Reconstructing detailed aspects of the lives of Lower Palaeolithic hominins, who lived during the Middle Pleistocene, is challenging due to the restricted nature of the surviving evidence, predominantly animal bones and stone tools. Qesem Cave, Israel (420–200 ka) is a site that has produced evidence for a wealth of innovative features including controlled use of fire, represented by a repeatedly used hearth. Numerous charred bone and stone tools as well as wood ash have been found throughout the ten metres of archaeological deposits. Here, we describe the presence of a range of potentially inhaled, and ingested, materials extracted from samples of dental calculus from the Qesem Cave hominins. These finds offer an insight into the environment in and around the cave, while micro-charcoal highlights the need for smoke management in enclosed environments. Plant fibres and a phytolith may be evidence of oral hygiene activities or of using the teeth to work raw materials. Starch granules and chemical compounds provide a direct link to ingested plant food items. This extends the evidence for consumption of plant foods containing essential nutrients including polyunsaturated fatty acids and carbohydrates, into the Lower Palaeolithic. Together, these results represent a significant breakthrough towards a better understanding of Middle Pleistocene dietary breadth and highlight some of the challenges facing the adoption of the habitual use of fire for cooking by the Qesem Cave hominins, as well as offering an insight into their ecological knowledge and technological adaptability.

Analysis of dental calculus is increasingly important in archaeology, although the focus has hitherto been on dietary reconstruction. Non-edible material has, however, recently been extracted from the dental calculus of a Neanderthal population from the 49 000-year-old site of El Sidrón, Spain, in the form of fibre and chemical compounds that indicate conifer wood. Associated dental wear confirms that the teeth were being used for non-dietary activities. These results highlight the importance of dental calculus as a source of wider biographical information, and demonstrate the need to include associated data within research, in particular tooth wear, to maximise this valuable resource., Research and fieldwork at the El Sidrón site is funded by the Principado de Asturias and the Spanish Government (Project CGL2012-36682)

Interdisciplinary scientific investigations utilising chemical analysis, shotgun metagenomics, textile analysis and radiocarbon dating have been applied to the study of an intact prehistoric Egyptian mummy, allowing insights into when this individual lived and died, and the funerary treatments employed in the preparation of the body. Here we present the first evidence for an extant prehistoric mummy that has undergone treatment with notably similar formative complex ‘balms’ that would later constitute the classic embalming recipes employed at the height of pharaonic mummification some 2500 years later. Making the informed assumption that the provenance of the Turin body was Gebelein, Qena or Luxor (Thebes), the findings offer the first indication that this type of funerary recipe was likely to have been employed over a wider geographical area at a time when the concept of a pan-Egyptian identity was supposedly still developing.

The session “‘AV-Ready’ Cities or ‘City-Ready’ AVs” at the Automated Vehicles Symposium 2016 in San Francisco addressed key aspects of road automation from an urban policy perspective. The aim of the session was to raise the awareness of urban policy needs within the automation community, as well as enabling urban policy makers to understand better the opportunities and issues related to automated, connected vehicles when drafting their cities’ policies in the area of automated road transport.

In a recent study, Hardy et al. (2012) identified compounds from two non-nutritional plants, yarrow and camomile, in a sample of Neanderthal dental calculus from the northern Spanish site of El Sidrón. Both these plants are bitter tasting and have little nutritional value but are well known for their medicinal qualities. Bitter taste can signal poison. We know that the bitter taste perception gene TAS2R38 was present among the Neanderthals of El Sidrón (Lalueza-Fox et al. 2009), and their selection of yarrow and camomile was hence probably deliberate. With few nutritional benefits, reasons must be sought for why the Neanderthals collected and ingested these plants. They could have consumed them as flavouring, but this presupposes a degree of complexity in cuisine for which there is little evidence. The widespread evidence for animal self-medication, or zoopharmacognosy, however, offers an attractive behavioural context.We propose, indeed, that these plants were selected and ingested deliberately for the purpose of self-medication.Here, we investigate the implications of this new finding for Neanderthal knowledge of plants and we offer a context for plant knowledge and self-medication among early human and hominin populations.

Amyotrophic lateral sclerosis (ALS) and frontotemporal lobar degeneration (FTLD) are neurodegenerative disorders that are characterized by cytoplasmic aggregates and nuclear clearance of TAR DNA-binding protein 43 (TDP-43). Studies in Drosophila, zebrafish and mouse demonstrate that the neuronal dysfunction of TDP-43 is causally related to disease formation. However, TDP-43 aggregates are also observed in glia and muscle cells, which are equally affected in ALS and FTLD; yet, it is unclear whether glia- or muscle-specific dysfunction of TDP-43 contributes to pathogenesis. Here, we show that similar to its human homologue, Drosophila TDP-43, Tar DNA-binding protein homologue (TBPH), is expressed in glia and muscle cells. Muscle-specific knockdown of TBPH causes age-related motor abnormalities, whereas muscle-specific gain of function leads to sarcoplasmic aggregates and nuclear TBPH depletion, which is accompanied by behavioural deficits and premature lethality. TBPH dysfunction in glia cells causes age-related motor deficits and premature lethality. In addition, both loss and gain of Drosophila TDP-43 alter mRNA expression levels of the glutamate transporters Excitatory amino acid transporter 1 (EAAT1) and EAAT2. Taken together, our results demonstrate that both loss and gain of TDP-43 function in muscle and glial cells can lead to cytological and behavioural phenotypes in Drosophila that also characterize ALS and FTLD and identify the glutamate transporters EAAT1/2 as potential direct targets of TDP-43 function. These findings suggest that together with neuronal pathology, glial- and muscle-specific TDP-43 dysfunction may directly contribute to the aetiology and progression of TDP-43-related ALS and FTLD.

A study of the hair and wig of Meyrit. As part of a long-term study of the collections of Turin's Museo Egizio, the authors have particularly focused on the contents of the tomb of the 14th-century BC couple Kha and Meryt, discovered intact in 1906 at the workman's village of Deir el-Medina, modern Luxor (Schiaparelli 1927; Vassilika 2010). As the most impressive woman's wig to have survived from pharaonic times (Schiaparelli 1927, 101), Meryt's long wig closely replicates the numerous portrayals of the long full 'enveloping' style found particularly in 14th-century BC sculpture and tomb scenes (Fletcher 1995, 260-3).

Sima del Elefante, Atapuerca, Spain contains one of the earliest hominin fragments yet known in Europe, dating to 1.2 Ma. Dental calculus from a hominin molar was removed, degraded and analysed to recover entrapped remains. Evidence for plant use at this time is very limited and this study has revealed the earliest direct evidence for foods consumed in the genus Homo. This comprises starchy carbohydrates from two plants, including a species of grass from the Triticeae or Bromideae tribe, meat and plant fibres. All food was eaten raw, and there is no evidence for processing of the starch granules which are intact and undamaged. Additional biographical detail includes fragments of non-edible wood found adjacent to an interproximal groove suggesting oral hygiene activities, while plant fibres may be linked to raw material processing. Environmental evidence comprises spores, insect fragments and conifer pollen grains which are consistent with a forested environment.

Archaeological work in advance of construction at a site on the edge of York, UK, yielded human remains of prehistoric to Romano-British date. Amongst these was a mandible and cranium, the intra-cranial space of which contained shrunken but macroscopically recognizable remains of a brain. Although the distinctive surface morphology of the organ is preserved, little recognizable brain histology survives. Though rare, the survival of brain tissue in otherwise skeletalised human remains from wet burial environments is not unique. A survey of the literature shows that similar brain masses have been previously reported in diverse circumstances. We argue for a greater awareness of these brain masses and for more attention to be paid to their detection and identification in order to improve the reporting rate and to allow a more comprehensive study of this rare archaeological survival.

Queen Nefertari, the favourite Royal Consort of Pharaoh Ramses II (Ancient Egypt, New Kingdom, 19th Dynasty c. 1250 BC) is famous for her beautifully decorated tomb in the Valley of the Queens. Her burial was plundered in ancient times yet still many objects were found broken in the debris when the tomb was excavated. Amongst the found objects was a pair of mummified legs. They came to the Egyptian Museum in Turin and are henceforth regarded as the remains of this famous Queen, although they were never scientifically investigated. The following multidisciplinary investigation is the first ever performed on those remains. The results (radiocarbon dating, anthropology, paleopathology, genetics, chemistry and Egyptology) all strongly speak in favour of an identification of the remains as Nefertari’s, although different explanations—albeit less likely—are considered and discussed. The legs probably belong to a lady, a fully adult individual, of about 40 years of age. The materials used for embalming are consistent with Ramesside mummification traditions and indeed all objects within the tomb robustly support the burial as of Queen Nefertari., PLoS ONE, 11 (11), ISSN:1932-6203

The mummies of Kha and his wife Merit were found intact in an undisturbed tomb in western Thebes near the ancient workers’ village of Deir el-Medina. Previous MDCT (this abbreviation needs spelling out) investigations showed that the bodies of Kha and Merit did not undergo classical royal 18th Dynasty artificial mummification, which included removal of the internal organs. It was, therefore, concluded that the retention of the viscera in the body, combined with an absence of canopic jars in the burial chamber, meant the couple underwent a short and shoddy funerary procedure, despite their relative wealth at death. Nevertheless, all internal organs - brain, ocular bulbs/ocular nerves, thoracic and abdominal organs - showed a very good state of preservation, which contradicts the previous interpretation above. In order to better understand the type of mummification used to embalm these bodies, both wrapped mummies were reinvestigated using new generation X-ray imaging and chemical microanalyses Here we provide evidence that both individuals underwent a relatively high quality of mummification, fundamentally contradicting previous understanding. Elucidated “recipes”, whose components had anti-bacterial and anti-insecticidal properties, were used to treat their bodies. The time and effort undoubtedly employed to embalm both Kha and Merit and the use of imported costly resins, notably Pistacia, do not support the previously held view that the two individuals were poorly mummified. Despite a lack of evisceration, the approach clearly allowed their in situ preservation as well as affording a fairly successful mummification.

Traditional theories on ancient Egyptian mummification postulate that in the prehistoric period (i.e. the Neolithic and Chalcolithic periods, 5th and 4th millennia B.C.) bodies were naturally desiccated through the action of the hot, dry desert sand. Although molding of the body with resin-impregnated linen is believed to be an early Pharaonic forerunner to more complex processes, scientific evidence for the early use of resins in artificial mummification has until now been limited to isolated occurrences during the late Old Kingdom (c. 2200 B.C.), their use becoming more apparent during the Middle Kingdom (c. 2000-1600 BC). We examined linen wrappings from bodies in securely provenanced tombs (pit graves) in the earliest recorded ancient Egyptian cemeteries at Mostagedda in the Badari region (Upper Egypt). Our investigations of these prehistoric funerary wrappings using a combination of gas chromatography-mass spectrometry (GC-MS) and thermal desorption/pyrolysis (TD/Py)-GC-MS have identified a pine resin, an aromatic plant extract, a plant gum/sugar, a natural petroleum source, and a plant oil/animal fat in directly AMS-dated funerary wrappings. Predating the earliest scientific evidence by more than a millennium, these embalming agents constitute complex, processed recipes of the same natural products, in similar proportions, as those utilized at the zenith of Pharaonic mummification some 3,000 years later. The antibacterial properties of some of these ingredients and the localized soft-tissue preservation that they would have afforded lead us to conclude that these represent the very beginnings of experimentation that would evolve into the famous mummification practice of the Pharaonic period.

Sports ranking systems are often viewed as inadequate for judging the quality of the teams or players involved. Meanwhile, statistical models have been shown to produce more accurate ratings for those competitors, based on their ability to forecast future results. However, whilst predictive power is a desirable property of any official ranking system, these systems must also be fair, transparent and insensitive to bias. Additional requirements may also be required, such as promoting major tournaments and deciding seedings. By considering rankings for ATP tennis players, we propose that statistical models can be used to improve the existing ranking system, in such a way that the resulting rankings are fair and usable by the governing body. In many cases, there is a trade-off between predictive power and other desirable properties, and so compromise is required to produce a system that can be implemented successfully.

We introduce the time dynamic random conductance model and consider the heat kernel for the random walk on this environment. In the case where conductances are bounded above, an example environment is presented which exhibits heat kernel decay that is asymptotically slower than in the well studied time homogeneous case - being close to $O\left( n^{-1}\right) $ as opposed to $O\left( n^{-2}\right) $. The example environment given is a modification of an environment introduced in Berger, Biskup, Hoffman and Kozma (2008).

Hardy, Karen et al., Neanderthals disappeared sometime between 30,000 and 24,000 years ago. Until recently, Neanderthals were understood to have been predominantly meat-eaters; however, a growing body of evidence suggests their diet also included plants. We present the results of a study, in which sequential thermal desorption-gas chromatography-mass spectrometry (TD-GC-MS) and pyrolysis-gas chromatographymass spectrometry (Py-GC-MS) were combined with morphological analysis of plant microfossils, to identify material entrapped in dental calculus from five Neanderthal individuals from the north Spanish site of El Sidrón. Our results provide the first molecular evidence for inhalation of wood-fire smoke and bitumen or oil shale and ingestion of a range of cooked plant foods. We also offer the first evidence for the use of medicinal plants by a Neanderthal individual. To explore the influence of level of functioning and cognitive status on outcome after rehabilitation for older people with different types of impairment. Design: An inception cohort study. Subjects: A total of 560 older people, mean age 80.0 years, participating in rehabilitation programmes in the Sydney area. Methods: Level of functioning using the Functional Independence Measure (FIM TM), the Barthel Index, cognitive status using the Mini Mental State Examination (MMSE), and type of impairment were assessed on admission. Level of functioning and length of hospital stay were assessed on discharge and after 6 months. Results: Disability on admission was the strongest independent predictor for functioning at discharge (B = 0.35,R 2= 0.49 p < 0.001) and follow-up (B = 0.22,R 2= 0.27, p < 0.001), and for length of hospital stay (B = -0.63,R 2= 0.12, p < 0.001). Cognitive status at admission (mean MMSE 25 (standard deviation 5.3)) was also a significant predictor of functioning at discharge and at follow-up (B = 0.30,R 2= 0.42, p < 0.01). Functional status prior to injury, joint replacement impairment category, and type of hospital had marginal, but statistically significant, impacts on functioning after discharge. Conclusion: Functional status on admission to a rehabilitation facility has stronger predictive value than type of impairment for rehabilitation outcome for older people. Cognitive impairment may have a small adverse effect on rehabilitation outcome.

Background. We earlier reported that patients with recurrent respiratory papillomatosis responded to six months of treatment with lymphoblastoid interferon alfa-n1. Because another study of patients treated for one year with leukocyte interferon alfa-n3 found that the growth rate of papillomas was slowed in the first six months but returned to base line during months 7 through 12 despite persistent interferon treatment, we now report the long-term results in our original study patients who were followed for a median of four years after the original one-year crossover study. Methods. After the patients in our study had completed the first study year, their physicians could continue or recommence treatment with lymphoblastoid interferon alfa-n1 in a dose of either 2 MU per square meter of body-surface area per day or 4 MU per square meter every other day. The extent of disease was measured by endoscopy when clinically indicated. Results. Data on late-follow-up were obtained for 60 of the 66 patient...

We dene a notion of inversion valid in the general metric space setting. We establish several basic facts concerning inversions; e.g., they are quasimöbius homeomorphisms and quasihyperbolically bilipschitz. In a certain sense, inversion is dual to sphericalization. We demonstrate that both inversion and sphericalization preserve local quasiconvexity and annular quasiconvexity as well as uniformity.
