This study explored disciplinary writing in grades 4-6 and the potential of writing to learn and learning to write across the curriculum to prepare the pupils for their future writing. Using Ivanic's discourses of writing as an analytical framework, observation protocols from 104 observers in 374 lessons in 76 Swedish schools were analyzed exploring school writing in the different curriculum subjects. Analysis of the data reveals that in most lessons the teachers required their pupils to write with a single focus on reinforcing learning, enacting three of Ivanic's seven discourses of writing: "thinking and learning" discourse, "skills" discourse, and "social practices" discourse. Much less frequently overall but commonly in language lessons, teachers required their pupils to write with a dual focus, developing writing proficiency while reinforcing learning. In these cases, all of Ivanic's discourses were enacted. The results suggest potential for a dual focus on writing to learn and learning to write to further develop the pupils' writing across the curriculum.

Still, in the school year 2018/19, approximately 7% of all Swedish pupils did not achieve the learning goals related to reading competence stated for the subject Swedish in the national curriculum for compulsory school (grades 0-9) (Skolverkets statistikdatabas). In order to identify and support pupils with reading difficulties as early as possible, compulsory reading development assessments are conducted in grades 1, 3, 6 and 9 according to which all pupils displaying a delayed development are to receive additional support. The provision of this support is the responsibility of every municipality, but in what form and to what extent and duration it should be given is not prescribed. The present study aims to give insight into how Swedish policy reforms to improve reading ability among pupils in compulsory school are operationalised in one Swedish municipality during the school year 2018/2019. Further, the study seeks to exemplify how the policy guidelines are implemented at three schools and how policy and practice are related.

In mass disaster events, forensic DNA laboratories may be called upon to quickly pivot their operations toward identifying bodies and reuniting remains with family members. Ideally, laboratories have considered this possibility in advance and have a plan in place. Compared with traditional short tandem repeat (STR) typing, single nucleotide polymorphisms (SNPs) may be better suited to these disaster victim identification (DVI) scenarios due to their small genomic target size, resulting in an improved success rate in degraded DNA samples. As the landscape of technology has shifted toward DNA sequencing, many forensic laboratories now have benchtop instruments available for massively parallel sequencing (MPS), facilitating this operational pivot from routine forensic STR casework to DVI SNP typing. Herein, we present the commercially available SNP sequencing assays amenable to DVI, we use data simulations to explore the potential for kinship prediction from SNP panels of varying sizes, and we give an example DVI scenario as context for presenting the matrix of considerations: kinship predictive potential, cost, and throughput of current SNP assay options. Views about what writing is and how it should be taught have varied over the years as well as across contexts. Studies of curricula, teaching materials, and teaching practices have shown a strong focus on skills, genres, and processes, but few have asked teachers about their perspectives on writing. In this article we explore what views, or discourses, of writing are currently active among teachers in Swedish compulsory education, covering ages from 7 to 15. Sixty teachers answered a questionnaire with open and closed questions. Using Ivanic's framework for discourses of writing, the answers were analyzed holistically in order to define what main discourse, or discourses, each teacher represented. Results show that most teachers represent one main discourse, but that a combination of discourses occur, in particular among teachers from the earliest school years (1-3). The most common discourse was the process discourse, followed by genre, creativity, skills, and thinking. None of the teachers represented the social practice or the sociopolitical discourse. The results concur with findings from studies of curricula, teaching materials, and teaching practices both in Sweden and globally and are discussed in relation to what literacy skills may be necessary in the 21st century in order to participate in social and political life.

This study investigates what discourses dominate teachers’ beliefs about writing education and how these discourses are negotiated among teachers in social media. The empirical material is based on a stratified random sample of interactions, so-called threads, between teachers in three large open Facebook groups for teachers of Swedish (2,500–10,000 members). Taking Ivanič’s 7 discourses of writing and learning to write as a framework, the study analyses discourses about writing visible in the interactions as well as blogs linked to, and school books and apps recommended. The two last steps are data-driven, emanating from previous steps. The result shows that 40% of the interactions concern writing. These interactions are dominated by a skills discourse. Further, a genre discourse challenges a former prominent discourse, the process discourse. Also, discourses in a social context are rare. The results indicate a narrow view of writing education in policy and practice, which, due to Ivanič (2004), can be interpreted as a consequence of a wider societal context where the educational system is questioned and explicit standards for writing are foregrounded. What conditions that would facilitate a wider range of discourses of writing in a school under pressure merits further investigations.

Activated platelets contribute to thrombosis and inflammation by the release of extracellular vesicles (EVs) exposing P-selectin, phosphatidylserine (PS) and fibrinogen. P2Y12 receptor antagonists are routinely administered to inhibit platelet activation in patients after acute myocardial infarction (AMI), being a combined antithrombotic and anti-inflammatory therapy. The more potent P2Y12 antagonist ticagrelor improves cardiovascular outcome in patients after AMI compared to the less potent clopidogrel, suggesting that greater inhibition of platelet aggregation is associated with better prognosis. The effect of ticagrelor and clopidogrel on the release of EVs from platelets and other P2Y12-exposing cells is unknown. This study compares the effects of ticagrelor and clopidogrel on (1) the concentrations of EVs from activated platelets (primary end point), (2) the concentrations of EVs exposing fibrinogen, exposing PS, from leukocytes and from endothelial cells (secondary end points) and (3) the procoagulant activity of plasma EVs (tertiary end points) in 60 consecutive AMI patients. After the percutaneous coronary intervention, patients will be randomized to antiplatelet therapy with ticagrelor (study group) or clopidogrel (control group). Blood will be collected from patients at randomization, 48 hours after randomization and 6 months following the index hospitalization. In addition, 30 age- and gender-matched healthy volunteers will be enrolled in the study to investigate the physiological concentrations and procoagulant activity of EVs using recently standardized protocols and EV-dedicated flow cytometry. Concentrations of EVs will be determined by flow cytometry. Procoagulant activity of EVs will be determined by fibrin generation test. The compliance and response to antiplatelet therapy will be assessed by impedance aggregometry. We expect that plasma from patients treated with ticagrelor (1) contains lower concentrations of EVs from activated platelets, exposing fibrinogen, exposing PS, from leukocytes and from endothelial cells and (2) has lower procoagulant activity, when compared to patients treated with clopidogrel. Antiplatelet therapy effect on EVs may identify a new mechanism of action of ticagrelor, as well as create a basis for future studies to investigate whether lower EV concentrations are associated with improved clinical outcomes in patients treated with P2Y12 antagonists.

There is a great interest and demand for green-type energy storage in Sweden for both short- and long-term (hours, days, weeks and seasons) periods. While there are a number of approaches proposed (e.g., compressed air, geothermal and thermal), only a few have commercially been demonstrated through upscaling projects. Among these, the thermal energy storage (TES) that stores energy (excess heat or cold) in fluids is particularly interesting. The excess energy can be stored underground in excavated caverns and used for large district heating and cooling purposes as well as for balancing and regulating electrical energy in power grids. For an upscaling underground TES project within the Tornquist suture zone of Scania in the southwest of Sweden, three high-resolution seismic profiles, each approximately 1 km long, were acquired. Geologically, the site sits within the southern margin of the Romeleåsen fault zone in the Sorgenfrei–Tornquist Zone (STZ), where dolerite dyke swarms of Carboniferous–Permian age are observed striking in the SE–NW direction for hundreds of kilometers both on land and in offshore seismic and magnetic data (from Scania to Midland Valley in the UK). These dykes, 10–50 m thick, in the nearby quarries (within both Precambrian gneiss and quartzite) express themselves mostly in a subvertical manner. They can therefore act as a good water/fluid barrier, which can be an important geological factor for any TES site. For the data acquisition, combined cabled and wireless recorders were used to provide continuity on both sides of a major road running in the middle of the study area. Bedrock depressions are clearly depicted in the tomograms, suggesting the possibility of zones of weaknesses, highly fractured and/or weathered, in the bedrock and confirmed in several places by follow-up boreholes. Several steeply dipping (60–65°) reflections were imaged down to 400 m depth and interpreted to originate from dolerite dykes. This interpretation is based on their orientations, strong amplitudes, regular occurrences and correlation with downhole logging data. In addition, groundwater flow measurements within the unconsolidated sediments and in bedrock suggest steeply dipping structures are the dominant factor in directing water mainly along a SE–NW trend, which is consistent with the strike of the dyke swarm within the STZ. To provide further insight on the origin of the reflections, even the historical crustal-scale offshore BABEL (Baltic and Bothnian Echoes from the Lithosphere) lines (A-AA-AB) were revisited. Clear multiphase faults and signs of intrusions or melt source in the lower crust are observed, as well as a Moho step across the Tornquist zone. Overall, we favor that the reflections are of dolerite origin and their dip component (i.e., not subvertical) may imply a Precambrian basement (and dykes) tilting, block rotation, towards the NE as a result of the Romeleåsen reverse faulting. In terms of thermal storage, these dykes then may be encountered during the excavation of the site and can complicate underground water flow should they be used as a fluid barrier in case of leakage.

Whole-genome sequencing (WGS) data present a readily available resource for mitochondrial genome (mitogenome) haplotypes that can be utilized for genetics research including population studies. However, the reconstruction of the mitogenome is complicated by nuclear mitochondrial DNA (mtDNA) segments (NUMTs) that co-align with the mtDNA sequences and mimic authentic heteroplasmy. Two minimum variant detection thresholds, 5% and 10%, were assessed for the ability to produce authentic mitogenome haplotypes from a previously generated WGS dataset. Variants associated with NUMTs were detected in the mtDNA alignments for 91 of 917 (~8%) Swedish samples when the 5% frequency threshold was applied. The 413 observed NUMT variants were predominantly detected in two regions (nps 12,612–13,105 and 16,390–16,527), which were consistent with previously documented NUMTs. The number of NUMT variants was reduced by ~97% (400) using a 10% frequency threshold. Furthermore, the 5% frequency data were inconsistent with a platinum-quality mitogenome dataset with respect to observed heteroplasmy. These analyses illustrate that a 10% variant detection threshold may be necessary to ensure the generation of reliable mitogenome haplotypes from WGS data resources.

Background: In 2001, we studied the presence and coagulant properties of “microparticles” in the blood of healthy humans. Since then, multiple improvements in detection, isolation and functional characterization of the now called “extracellular vesicles” (EVs) have been made, and shortcomings were identified. Aim: To revisit the presence and function of EVs in blood from healthy humans. Methods: Blood was collected from 20 healthy donors. EV-containing plasma was prepared according to new guidelines, and plasma was diluted to prevent swarm detection. Single EVs were measured by flow cytometry with known sensitivity of fluorescence and light scatter. The haemostatic properties of EVs were measured by thrombin-, fibrin-, and plasmin generation. Plasma concentrations of thrombin-antithrombin complexes and prothrombin fragment 1 + 2 were measured to assess the coagulation status in vivo. Results: Compared to 2001, the total concentrations of detected EVs increased from 190- to 264-fold. In contrast to 2001, however, EVs are non-coagulant which we show can be attributed to improvements in blood collection and plasma preparation. No relation is present between the plasma concentrations of EVs and either TAT or F1 + 2. Finally, we show that EVs support plasmin generation. Discussion: Improvements in blood collection, plasma preparation and detection of EVs reveal that results from earlier studies have to be interpreted with care. Compared to 2001, higher concentrations of EVs are detected in blood of healthy humans which promote fibrinolysis rather than coagulation.

Because procedures of handling and storage of body fluids affect numbers and composition of extracellular vesicles (EVs), standardization is important to ensure reliable and comparable measurements of EVs in a clinical environment. We aimed to develop standard protocols for handling and storage of human body fluids for EV analysis. Conditions such as centrifugation, single freeze–thaw cycle, effect of time delay between blood collection and plasma preparation and storage were investigated. Plasma is the most commonly studied body fluid in EV research. We mainly focused on EVs originating from platelets and erythrocytes and investigated the behaviour of these 2 types of EVs independently as well as in plasma samples of healthy subjects. EVs in urine and saliva were also studied for comparison. All samples were analysed simultaneously before and after freeze–thawing by resistive pulse sensing, nanoparticle tracking analysis, conventional flow cytometry (FCM) and transmission (scanning) electron microscopy. Our main finding is that the effect of centrifugation markedly depends on the cellular origin of EVs. Whereas erythrocyte EVs remain present as single EVs after centrifugation, platelet EVs form aggregates, which affect their measured concentration in plasma. Single erythrocyte and platelet EVs are present mainly in the range of 100–200 nm, far below the lower limit of what can be measured by conventional FCM. Furthermore, the effects of single freeze–thaw cycle, time delay between blood collection and plasma preparation up to 1 hour and storage up to 1 year are insignificant (p>0.05) on the measured concentration and diameter of EVs from erythrocyte and platelet concentrates and EVs in plasma, urine and saliva. In conclusion, in standard protocols for EV studies, centrifugation to isolate EVs from collected body fluids should be avoided. Freezing and storage of collected body fluids, albeit their insignificant effects, should be performed identically for comparative EV studies and to create reliable biorepositories.

Introduction: The size of extracellular vesicles (EVs) can be determined with a tunable resistive pulse sensor (TRPS). Because the sensing pore diameter varies from pore to pore, the minimum detectable diameter also varies. The aim of this study is to determine and improve the reproducibility of TRPS measurements. Methods: Experiments were performed with the qNano system (Izon) using beads and a standard urine vesicle sample. With a combination of voltage and stretch that yields a high blockade height, we investigate whether the minimum detected diameter is more reproducible when we configure the instrument targeting (a) fixed stretch and voltage, or (b) fixed blockade height. Results: Daily measurements with a fixed stretch and voltage (n=102) on a standard urine sample show a minimum detected vesicle diameter of 128±19 nm [mean±standard deviation; coefficient of variation (CV) 14.8%]. The vesicle concentration was 2.4·109±3.8·109 vesicles/mL (range 1.4·108–1.8·1010). When we compared setting a fixed stretch and voltage to setting a fixed blockade height on 3 different pores, we found a minimum detected vesicle diameter of 118 nm (CV 15.5%, stretch), and 123 nm (CV 4.5%, blockade height). The detected vesicle concentration was 3.2–8.2·108 vesicles/mL with fixed stretch and 6.4–7.8·108 vesicles/mL with fixed blockade height. Summary/conclusion: Pore-to-pore variability is the cause of the variation in minimum detected size when setting a fixed stretch and voltage. The reproducibility of the minimum detectable diameter is much improved by setting a fixed blockade height.

Background: Isolation of extracellular vesicles from plasma is a challenge due to the presence of proteins and lipoproteins. Isolation of vesicles using differential centrifugation or density-gradient ultracentrifugation results in co-isolation of contaminants such as protein aggregates and incomplete separation of vesicles from lipoproteins, respectively. Aim: To develop a single-step protocol to isolate vesicles from human body fluids. Methods: Platelet-free supernatant, derived from platelet concentrates, was loaded on a sepharose CL-2B column to perform size-exclusion chromatography (SEC; n=3). Fractions were collected and analysed by nanoparticle tracking analysis, resistive pulse sensing, flow cytometry and transmission electron microscopy. The concentrations of high-density lipoprotein cholesterol (HDL) and protein were measured in each fraction. Results: Fractions 9–12 contained the highest concentrations of particles larger than 70 nm and platelet-derived vesicles (46%±6 and 61%±2 of totals present in all collected fractions, respectively), but less than 5% of HDL and less than 1% of protein (4.8%±1 and 0.65%±0.3, respectively). HDL was present mainly in fractions 18–20 (32%±2 of total), and protein in fractions 19–21 (36%±2 of total). Compared to the starting material, recovery of platelet-derived vesicles was 43%±23 in fractions 9–12, with an 8-fold and 70-fold enrichment compared to HDL and protein. Conclusions: SEC efficiently isolates extracellular vesicles with a diameter larger than 70 nm from platelet-free supernatant of platelet concentrates. Application SEC will improve studies on the dimensional, structural and functional properties of extracellular vesicles.

Extracellular vesicles (EVs) facilitate intercellular communication by carrying bioactive molecules such as proteins, messenger RNA, and micro (mi)RNAs. Recently, high-density lipoproteins (HDL) isolated from human plasma were also reported to transport miRNA to other cells. HDL, when isolated from human plasma, ranges in density between 1.063 and 1.21 g/mL, which grossly overlap with the reported density of EVs. Consequently, HDL and EV will be co-isolated when using density gradient ultracentrifugation. Thus, more stringent isolation/separation procedures of EV and HDL are essential to know their relative contribution to the pool of circulating bioactive molecules.

One way to support the development of new safety practices in testing and field failure situations of electric vehicles and their lithium-ion (Li-ion) traction batteries is to conduct studies simulating plausible incident scenarios. This paper focuses on risks and hazards associated with venting of gaseous species formed by thermal decomposition reactions of the electrolyte and electrode materials during thermal runaway of the cell. A test set-up for qualitative and quantitative measurements of both major and minor gas species in the vented emissions from Li-ion batteries is described. The objective of the study is to measure gas emissions in the absence of flames, since gassing can occur without subsequent fire. Test results regarding gas emission rates, total gas emission volumes, and amounts of hydrogen fluoride (HF) and CO2 formed in inert atmosphere when heating lithium iron phosphate (LFP) and lithium nickel-manganese-cobalt (NMC) dioxide/lithium manganese oxide (LMO) spinel cell stacks are presented and discussed. Important test findings include the large difference in total gas emissions from NMC/LMO cells compared to LFP, 780 L kg−1 battery cells, and 42 L kg−1 battery cells, respectively. However, there was no significant difference in the total amount of HF formed for both cell types, suggesting that LFP releases higher concentrations of HF than NMC/LMO cells.
