Cudrania tricuspidata (Moraceae) is a deciduous tree widely distributed in East Asia, including China, Korea, and Japan. It produces delicious fruit, and its cortex and root bark have been used as a traditional medicine to treat neuritis and inflammation. As C. tricuspidata has become known as a functional food, its cultivation area and production gradually have increased in Korea. However, information of viral disease in C. tricuspidata is very limited. In September 2012, open-field-grown C. tricuspidata trees showing virus-like symptoms of mosaic, yellowing, and distortion on the leaves were found in Naju, Korea. The fruit production in the diseased trees decreased to 20 to 40% of that in healthy trees. To identify causal agent(s), total RNA was isolated from the symptomatic leaves and used to generate a transcriptome library using the TruSeq Stranded Total RNA with Ribo-Zero Plant kit (Illumina, San Diego, CA) according to the manufacturer's instruction. The transcriptome library was analyzed by next-generation sequencing (NGS) using an Illumina HiSeq2000 sequencer. NGS reads were quality filtered and de novo assembled by the Trinity pipeline, and the assembled contigs were analyzed against the viral reference genome database in Genbank by BLASTn and BLASTx searches (3). The entire NGS procedure was perofrmed by Macrogen Inc. (Seoul, South Korea). Among the analyzed contigs, one large contig (10,043 bp) was of viral origin. Nucleotide blast searches showed that the contig has a maximum identity of 89% (with 100% coverage) to the isolate MS1 (Genbank Accession No. EU761198) of Bean common mosaic virus (BCMV), which was isolated from Macroptilium atropurpureum in Australia. The presence of BCMV was confirmed by a commercially available double-antibody sandwich (DAS)-ELISA kit (Agdia, Elkhart, IN). To confirm the BCMV sequence obtained by NGS, two large fragments covering the entire BCMV genome were amplified by reverse transcription-polymerase chain reaction (RT-PCR) using two sets of specific primers (5′-AAAATAAAACAACTCATAAAGACAAC-3′ and 5′-AGACTGTGTCCCAGAGCATTTC-3′ to amplify the 5′ half of the BCMV genome; 5′-GCATCCTGAGATTCACAGAATTC-3′ and 5′-GGAACAACAAACATTGCCGTAG-3′ to amplify the 3′ half of the BCMV genome) and sequenced. To obtain the complete genome sequence, the 5′ and 3′ terminal sequences were analyzed by the 5′ and 3′ rapid amplification of cDNA ends (RACE) method as described previously (1). The assembled full-length sequence of BCMV isolated from C. tricuspidata was 10,051 nucleotides in length without a poly(A) tail. It was deposited in Genbank under the accession number KM076650. BCMV, a member of the genus Potyvirus, is one of the most common viruses naturally infecting legumes, including Phaseolus vulgaris (2). In general, BCMV is known to have a restricted host range outside legume species (2). Therefore, the identification of BCMV from C. tricuspidata in this report is very exceptional. Because BCMV is easily transmitted by various aphids like other potyviruses, a large-scale survey may be required for exact investigation of the BCMV incidence in C. tricuspidata to prevent rapid spread of the virus. A disease forecast model for bacterial grain rot (BGR) of rice, which is caused by Burkholderia glumae, was developed in this study. The model, which was named 'BGRcast', determined daily conduciveness of weather conditions to epidemic development of BGR and forecasted risk of BGR development. All data that were used to develop and validate the BGRcast model were collected from field observations on disease incidence at Naju, Korea during 1998-2004 and 2010. In this study, we have proposed the environmental conduciveness as a measure of conduciveness of weather conditions for population growth of B. glumae and panicle infection in the field. The BGRcast calculated daily environmental conduciveness, Ci , based on daily minimum temperature and daily average relative humidity. With regard to the developmental stages of rice plants, the epidemic development of BGR was divided into three phases, i.e., lag, inoculum build-up and infection phases. Daily average of Ci was calculated for the inoculum build-up phase (Cinf ) and the infection phase (Cinc ). The Cinc and Cinf were considered environmental conduciveness for the periods of inoculum build-up in association with rice plants and panicle infection during the heading stage, respectively. The BGRcast model was able to forecast actual occurrence of BGR at the probability of 71.4% and its false alarm ratio was 47.6%. With the thresholds of Cinc = 0.3 and Cinf = 0.5, the model was able to provide advisories that could be used to make decisions on whether to spray bactericide at the pre- and post-heading stage.

During July to November 2014, severe rust infection was consistently found on Peucedanum japonicum growing farm in Yeosu, Korea. The rust was observed mainly on lower leaf surfaces. Symptoms of typical plants included yellow-orange rust pustules were observed on the petiole and leaf surface with small yellowish to chlorotic lesions on the upper surface. No symptom was observed on flowers. Uredinia were occurred amphigenous on leaf surface, and occasionally caulicolous, scattered or loosely aggregate, rounded to oblong, 0.4 to 4 mm in diameter, covered by epidermis, then naked, surrounded by ruptured epidermis, pulverulent, and brown. Urediniospores were ovate-ellipsoid, ellipsoid or subglobose, light brown, 20 to 45 ×15 to 35 mm, walls 2 to 4 mm thick. The resulting sequences were deposited in GenBank with accession No. KT778808, KT778809, and KT778810, respectively. Since this was the first accession of 28S sequence Puccinia jogashimensis, there was no exact match in GenBank nucleotide database. On the basis of the morphological characteristics and phylogenetic analyses of 28S rDNA, the fungus was identified as P. jogashimensis. To our knowledge, this is the first confirmed report on the occurrence of P. jogashimensis on P. japonicum in Korea.

Fruit drop due to Eurytoma maslovskii infestations of Japanese apricot (Prunus mume) is a serious economic issue in most parts of Jeonnam Province, with the exception of the coastal areas such as Wando, Shinan, Yeosu, and Muan. The average incidence of fruit drop was 67% in 2013 and 33.3% in 2014. E. maslovskii larvae overwinter inside the pits of Japanese apricots. Larval survival rates decrease to less than 30% after a rainy summer season. The eggs are long oval of 0.68 mm, 0.29 mm and ivory white. Mature larva is of 6.56 mm, 3.18 mm. Free pupa are black, adult females and males of 6.97 mm, 4.90 mm lengths, respectively, while the ovipositor is 0.64 mm in length. Adult emergence occurs from early April to early May, when Japanese apricot fruits are from 4 mm to 17 mm in diameter. Adults persist for 13.5 d after emergence, and the sex ratio of females to males was found th be 45.9:54.1. Although up to four eggs may be oviposited per fruit, only one larva will ultimately survive, ans the larvae are cannibalistic. The period during which E. maslovskii is able to oviposit on Japanese apricots only lasts from mid- to late April (fruit diameter: 12~16 mm). When the diameter of the fruit is >16 mm, the pit of the Japanese apricot hardens and larvae have difficultly penetrating the stone. Therefore, the most effective method of controlling this pest is to spray orchards with a control agent 2~3 times, at 5 d intervals, beginning in mid- April.

The hotpepper (Capsicum annuum), bell pepper (Capsicum annuum va .r angulosum) and tomato (Solanumlycopersicon) in greenhouse and hot pepper in open field were infected by TSWV. Especially, hot pepper wasseverely damaged by TSWV infection. The survey data indicated that 1.1 −30% in the nursery field at Naju,Suncheon, and Jangheung were infected by TSWV. Plants were infected by TSWV from early June toAugust. However, TSWV-infected seedlings from nursery fields showed the disease symptoms from May aftertransplanting. In pepper greenhouses, Frankliniella occidentalis was more dominant insect vector thanFrankliniella intonsa. But in open field, the population of insect vector was opposed to greenhouse. Inaddition, the removal of weeds was able to delay the incidence of TSWV via side-window of greenhouse inWinter. An outbreak of Ricania sp. occurred in the Kurye, Jeonnam area in 2011. This outbreak damaged many kinds of fruit trees such as Cornus, Persimmon and Chestnut. This experiment was conducted to survey the occurrence ecology of Ricania sp. such as host plants, oviposition characters, morphological characters and life cycle, as well as to select environmental friendly control agents. Ricaina sp. host plants included 51 species such as 32 xylophytes, and 19 herbaceous plants. Ricaina sp. preferred Cornus officinalis, Diospyros kaki, Castanea crenata, Eucommia ulmoides, Styrax japonicus for oviposition. Adults laid eggs on new inner twigs with 28.8 eggs per egg-mass. Egg size was 1.24 mm(length), 0.55 mm(width) in an oval shape. Nymphs molted four times. Every nymph stage had an x shape of yellow or white beeswax around the anus. Overwintered eggs of Ricania sp. hatched from the mid May to early June. Nymphal periods were from mid May to mid August and adults appeared from mid July but spawning began in mid August. Ricania sp. damaged new twigs by oviposition and retarded growth by sucking nutrients and producing a sooty mold. Sophora and natural plant extracts were effective environmentally friendly agricultural materials used to control the nymph and adult Ricania sp. Mortality was > 80%.

In order to develop a environmentally friendly control protocol for managing tomato leaf mold disease in the field, we employed bacteria- and fungi-based commercially available microbial preparations. The field experiment was conducted from April to July in 2010. Average incidence rates tomato leaf mold caused by Fulvia fulva were 13.1% at the two plastic houses located in Jangsung, Jeonnam area. Initially 11 microbial preparations were tested for antifungal activity against F. fulva in vitro. Among them, 7 selected preparations showed to be inhibited the mycelial growth of the fungal pathogen over 50%. Four microbes suppressed disease incidence as much 50% under greenhouse condition. Eventually in the field two microbial products including Bacillus subtilis GB-0365 and B. subtilis KB-401 respectively were showed control value up to 71.8% for four times sprays from 20 days to 70 days after transplanting. Furthermore, the control value of three times spray program demonstrated 79.3%. Efficacy of the three and four spray programs was more effective than that of non-spray control treatment. Our results indicated that adjustment of application method of commercially available microbial preparation could be used to control a target plant disease as an effective and efficient crop protection system for organic farming.

Bacterial culture filtrates of an aggressive rhizobacterium, Pseudomonas chlororaphis O6, displayed strong nematicidal activity. The nematicidal activity of P. chlororaphis O6 was markedly reduced in the gacS mutant of P. chlororaphis O6 grown in the presence of glycine, but no reduction of nematicidal activity in the gacS mutant was noted in the absence of glycine. The results of bioassay with P. chlororaphis O6 mutants showed that phenazine and pyrrolnitrin production was not a major factor, but the effects of glycine in the culture medium suggest that formation of hydrogen cyanide might be important. Assessments in greenhouse studies with tomatoes growing in nematode-infested soils confirmed that the application of P. chlororaphis O6 resulted in the control of the root-knot nematode. Our results demonstrated that P. chlororaphis O6 could be employed as a biocontrol agent for the control of the root-knot nematode, and the global regulator, GacS, functions as a positive regulator of the expression of nematicidal compounds and enzymes in P. chlororaphis O6.

A virus causing stunt, yellowing, severe mosaic, malformation symptoms on leaves and uneven development and malformation on fruits of zucchini was prevalent around Goseong, Gyeongsangnam-do, Korea. A survey conducted (2004) in the Goseong area revealed about 20% virus infection rate. The disease causative identified as Cucumber mosaic virus (CMV-Z1) was further characterized. The isolate induces mosaic symptoms on Cucumis sativus, while severe mosaic, stunt and malformation on C. pepo. Thin section analyses have shown that virus inclusions are formed in the cuticle layers as well as epidermal, parenchyma and collenchymas cells in virus-infected Nicotiana tabacum. CMV-Z1 isolate induced specific cytoplasmic inclusion bodies such as irregular clumps (IC), crystal (Cr) and irregular chloroplasts (ICh). IC was made up of virus particles interspersed with a darkly stained amorphous material and found both in the cytoplasm and vacuoles, whereas ICh and Cr were rarely found in the vacuoles. The genome of CMV-Z1 RNA-1 consists of 3359 nucleotide (nt) encoding 1a protein of 993 amino acids (aa). The CMVZ1 RNA-2 was 3050 nt in length containing 2a (857 aa) and 2b (110 aa), while RNA-3 encoding 3a movement protein (279 aa) and coat protein (218 aa) was 2215 nt in length. Phylogenetic analyses of nucleotide sequences of CMV-Z1 isolate appeared it is more closely related to subgroup IA than to subgroup IB or II.

Rehmannia glutinosa (family Scrophulariaceae) is a widely used traditional medicinal plant. In Korea, the area under the cultivation of this plant, as well as its production, has gradually increased because of its medicinal properties. R. glutinosa is conventionally propagated using its tuberous roots rather than its seeds, which have poor viability and low propagation rate. However, vegetative propagation is vulnerable to the accumulation of viruses over subsequent generations. Viral diseases cause significant loss of yield and deterioration of quality in R. glutinosa (Ling and Liu, 2009). In 2010, three R. glutinosa plants showing virus-like symptoms, namely chlorotic and necrotic local spots, vein necrosis and mosaic on the leaves, and stunting, were collected from a greenhouse in the Hwasun area of Korea. The causal virus was identified as Cucumbermosaic virus (CMV) using characteristics from bio-logical, serological and molecular analyses and namedas CMV-Rb. CMV-Rb caused mosaic on Nicotianabenthamiana, N. tabacum, Capsicum annuum, and Lyco-persicon esculentum. However, typical local lesions didnot develop on inoculated Pisum sativum, Cucurbitamoschata, Datura stramonium and Tetragonia expansaplants. Full-length genome sequences of CMV-RbRNAs 1, 2 and 3 were obtained using 12 primer pairs byRT-PCR analysis. The genome of CMV-Rb RNA seg-ments 1, 2, and 3 consists of 3363nt, 3049nt, and 2214ntin length, respectively. In order to ascertain their taxo-nomic identity, nucleotide and the deduced amino acidsequence analyses RNAs 1, 2 and 3 of CMV-Rb isolateswere conducted with previously reported sequences ofCMV strains and/or isolates. CMV-Rb RNAs showedabout 90 to 99% sequence identity to those of subgroupI strains suggesting that CMV-Rb is more closely relat-ed to CMV isolates belong to subgroup I. In infected host plants,it also produces two subgenomic RNAs (4 and 4A; Palukaitiset al., 1992). Based upon biological, serological, and, mole-cular biological characteristics of CMV isolates, they wereclassified into two subgroups, i.e., subgroup I and II (Owenand Palukaitis, 1988). The nucleotide sequence identitybetween subgroup I and II isolates range from 69 to 77%,while showing above 90% nucleotide sequence identitywithin each subgroup isolates. More detailed nucleotidesequence analyses, especially with the coat protein (CP)and 5' nontranslated region (NTR) of RNA 3 further sug-gested subdivision of subgroup I into subgroups IA and IB(Roossinck et al., 1999). This subgrouping was more clear-ly supported from the analysis of nucleotide sequences of1a and 3a open reading frames (ORFs) than those of the 2aand CP ORFs (Roossinck, 2002). We investigated the effect of the infection times and infection degrees at transplanting time of Zucchini yellow mosaic virus (ZYMV) on the growth and yield of cucumber plants at the semi-forcing and the retarding culture in 2007. When cucumber was inoculated with ZYMV at transplanting time, 15, 30, 45, 60, and 75 days after transplanting, vine length, internode length, number of leaf of the plants and marketable yield largely decreased as the cucumber infected earlier. The symptom variations among Korean Pepper mottle virus (PepMoV) isolates infecting pepper, tomato and potato were described and the cause of variations in relation to molecular variability were investigated. In addition, the entire genome of the 13 PepMoV isolates, collected from five provinces (Kyonggi, Chungnam, Gyeongnam, Jeonbuk and Jeonnam) in Korea, were determined and compared including the previously reported Korean-Vb isolate and 2 other PepMoV isolates isolated from America (CA and FL). Our results showed that the nucleotide sequence of all Korean isolates tested were nearly identical (98-99%) and only 94% similar to American isolates. In general, the complete nucleotide sequences and deduced polyprotein sequences indicated low genetic variation among isolates showing 0.1-3% nucleotide changes per site. However, based on ratio between nucleotide diversity values in nonsynonymous and synonymous position (dN/dS ratio) surprisingly, P1 and 6K2 genes showed relatively high nucleotide substitution ratio (0.8 and 1.0 nucleotide, respectively). When the 6K2 amino acid were aligned, there were 15 amino acid substitutions found in PepMoV-infected potato and only 1 amino acid change from two isolates of PepMoV-infected bell pepper. Interestingly, three isolates including isolate numbers 731, 205135 and 205136 that possessed different aa changes at 6K2 region also showed distinct symptom differentiation in indicator hosts and cosegregated in the phylogenetic analysis. These results further proved previous studies that P1 and 6K2 genes with other proteins might have some involvement on host specificity and pathogenicity.

The incidence and occurrence of virus infecting paprika (Capsicum annuum var. grossum) in Jeonnam province, the main areas of cultivation in Korea is undetermined. In this study, a total of 1,020 samples with virus-like symptoms were collected in Jeonnam province during summer season for 3 consecutive years (2002-2005) and were tested using enzyme linked immunosorbent assay (ELISA). Results showed that Pepper mottle virus (PepMoV), Broad bean wilt virus (BBWV), and Cucumber mosaic virus (CMV) were found to be the most prevalent viruses with a 3-year average percent incidence of 41.3, 19.8, and 4.4 respectively. Mixed infection with more than two viruses was also found with 3.5%, 17.0%, and 8.3%, respectively. Symptoms of these virus diseases were not evident at the seedling stage but slowly appeared at the transplanting stage and increased to the middle stage (4-5 months after transplanting) during the 3-year cultivation periods. Symptom appearance of infected plants however varied largely with transplanting time. Those plants transplanted from November to January were found to be infected with viruses in June, whereas symptoms appeared with in a month those plants transplanted from June to August. There were differences in the virus incidence from primary factor such as district, type of green house and variety, but these were not statistically significant (data not shown). Recommended control measures of paprika against these viruses is also discussed in this paper.

For the eco-friendly control of fruit rot of fig, disease incidences according to the cultural practices at 51 fig orchards in south coast area of Korea were surveyed in 2002, and the effect of soil mulch and potassium phosphonate on fruit rot of fig was evaluated from 2002 to 2004. Diseased fruits of fig orchards of open field, rain shading, and vinyl house were 33.3%, 7.5%, and 0%, respectively. Diseased fruits were rated at 34.6%, 35.7%, and 27% as the tree spacing of , respectively. Among the soil mulching materials, barley straw showed disease incidence of 1.8% and 3.5% of diseased fruits compared to 42.1% and 48% of no mulching in 2002 and 2003, respectively. Diseased fruits in dark paper film mulching were recorded at 4.8% and 12% in 2002 and 2003, respectively, which were lower than those in dark polyethylene film mulching at 11.2% and 26.2%. When potassium phosphonate(1,000 ppm) was sprayed 3 times with 10 days intervals from the beginning of disease occurrence, disease incidences were rated at 3.0% and 2.0% in 2003 and 2004, respectively, which were much lower than those of control at 45.9 % and 39.3%.

A peculiar virus-like disease of tomato showing yellow mosaic and necrotic spots on leaves and necrosis on veins, petioles and stems was observed at the Tomato Experimental Station (TES), Buyeo, Chungcheongnamdo, Korea. The disease incidence at TES fields ranged from 21 to 35% infecting different tomato cultivars. For this reason, to identify the virus infecting tomato and to characterize the virus based on biology, serology, cytology and at molecular level. Here, leaf samples were randomly collected from different infected tomato cultivars at TES fields and greenhouses and tested by ELISA using Pepper mottle virus (PePMoV) and Tomato mosaic virus (ToMV) antisera. Infected saps were mechanically inoculated in different host plants to test for pathogenicity, symptomatology and host ranges. Infected tissues and ultrathin sections were examined by electron microscopy. Finally, putative coat protein and 3`-untranslated region (CP/3`-UTR) fragment was amplified and cloned for sequence determination and analyzed its genetic relationship to existing PepMoV and PVY sequences at the Genbank. Results showed 69% of the samples were positive with PepMoV, 13% with ToMV and 19 % were doubly infected with PepMoV and ToMV. Symptoms greatly varied from different host plants inoculated with tomato leaf sap infected with PepMoV alone and discussed in detailed in this paper. Electron microscopy from infected tissues showed filamentous particles of 720-750nm in length, a typical morphology and size of PepMoV. In addition, cylindrical inclusion bodies, pinwheels, scrolls and laminates with masses of fibrillar inclusions were also found in ultrathin sections. A new virus-like disease of tomato showing chlorotic spots, malformation and necrosis on leaves, and chlorotic blotching, rings, and necrosis on fruits was observed around Sacheon, Gyeongsangnam-do, Korea in 2004. Host range analysis could not differentiate 4 field isolates collected from tomatoes showing different symptoms but identified them as Tomato bushy stunt virus (TBSV). TBSV-tsf2 isolate induced symptoms in indicator plants similar to those caused by the TBSV-C, -S and -Nf. As the isolate could not systemically infect Chenopodium quinoa, the isolate might belong to the previously described TBSV-S isolate. TBSV-tsf2 isolate caused similar cytological alterations that were similar to that generally caused by previously reported TBSV isolates. TBSV-tsf2 isolate, however, could be considered to belong to new strain of TBSV because masses of small electron-dense patches that were not observed from the previously described TBSV. The complete nucleotide sequences of the genomic RNA of 4739 nt excluding non-translated sequences at both termini have been determined and compared to sequences of other TBSV strains. The complete nucleotide sequence identity among TBSV isolates was 98.9% to 99.7%, and to the other tombusviruses ranged from 80.8% to 94.9%. Comparison of the amino acid sequences all five ORFs with those of other TBSV strains shows a similar genomic organization, and high percentage of amino acid sequence homology with TBSV-Nf than TBSV-S isolate. Since the TBSV symptoms were only observed in Sacheon fields where imported seeds from Japan were planted, the TBSV incidence probably caused by the planting contaminated tomato seeds and thus require more through quarantine procedure to prevent settlement of TBSV in Korea. Altogether, these results support that the Korean isolate of TBSV infecting tomato might be new strain.

The occurrence and relative incidence of viruses including Cucumber mosaic virus (CMV), Zucchini yellow mosaic virus (ZYMV), Papaya rings pot virus (PRSV), and Watermelon mosaic virus (WMV), Cucumber green mottle mosaic virus (CGMMV), Kyuri green mottle mosaic virus (KGMMV), and Melon necrotic spot virus (MNSV) were surveyed from main melon (Cucumis melo L.) production areas in Jeonnam province during 2000-2002. Virus disease incidences of melon cultivating fields were 0% and 11% in spring and fall 2000; 40%, 2.1%, and 8.8% in spring, summer, and fall 2001; and 6.3 % in spring 2002 in main cultivated areas in Jeonnam province, respectively. Field disease incidences of melon virus infections were 0% and 18.8% in spring and fall 2000; 50%, 38.5%, and 82.6% in spring, summer, and fall 2001; and 47.4% in spring 2002, respectively. Total of 101 melon samples showing typical disease symptoms were collected from 2000 to 2002 and tested for virus infection by RT-PCR. Potyvirus-specific DNA fragments for WMV, ZYMV, and PRSV were amplified from 46, 5, and 4 samples, respectively. MNSV specific DNA fragment was amplified from 18 samples. CMV-specific DNA fragment was detected from only 3 samples.

Sweet potato feathery mottle virus(SPFMV) is one of the most prevalent viruses infecting sweet potatoes and occurs widely in sweet potato cultivating areas in Korea. To assess their genetic variation, a total of 28 samples infected with SPFMV were subjected to restriction fragment length polymorphism(RFLP) analysis using DNAs amplified by RT-PCR with specific primer sets corresponding to the coat protein(CP) region of the virus. The similarity matrix by UPGMA procedure indicated that 28 samples infected with SPFMV were classified into three groups based on the number and size of DNA fragments by digestion of CP-encoding regions with 7 enzymes including SalI, AluI, EcoRI, HindIII, FokI, Sau3AI, and DraI bands. Four primer combinations out of 5 designed sets were able to differentiate SPFMV and sweet potato virus G infection, suggesting that these specific primers could be used to differentiate inter-groups of SPFMV. Sequence analysis of the CP genes of 17 SPFMV samples were 97-99% and 91-93% identical at the intra-group and inter-groups of SPFMV, respectively. The N-terminal region of the CP is highly variable and examination of the multiple alignments of amino acid sequences revealed two residues(residues 31 and 32) that were consistently different between SPFMV-O and SPFMV-RC.

Disease incidences of cucumber virus diseases in Jeonnam province were 52.5%, 16.1%, 35.2%, and 50.9% in 1999, 2000, 2001, and 2002, respectively. Rod- and flexuous rod-shaped virus particles were observed with the frequencies of 63.2% and 10.5%, respectively from the samples collected in 1999 under EM observation. Rod-shaped virus particles are considered as tobamovirus while flexuous rod shaped particles are considered as potyviruses. To further confirm their nature, total of 312 diseased virus samples were collected from 2000 to 2002, and tested by RT-PCR. Disease incidences of tobamoviruses including Cucumber green mottle mosaic virus and Kyuri green mottle mosaic virus were 48.7% and 3.8%, respectively while those of potyviruses including Zucchini yellow mosaic virus, Papaya ringspot virus, and Watermelon mosaic virus were 15.7%, 9.3%, and 5.1%, respectively. Interestingly, Cucumber mosaic virus was hardly detected. About 5.8% of tested samples were infected with more than one virus. Tobamovirus infection was consistently observed from September to December regardless of planting time, whereas infection of potyviruses was observed in many cucumber cultivating areas where it was planted in September and October.

A virus isolate collected from infected paprika (Capsicum annuum var. grossum) was characterized as Potato virus Y (PVY) based on biological, serological, cytopathological, and molecular properties. In host range studies, the paprika isolate produced the mosaic symptom on some tobacco, tomato and pepper (Capsicum annuum). A new paprika isolate also infected potato cultivars which is different biological characteristic compared to the other popular potyvirus infecting paprika, Pepper mottle virus (PepMoV). Previously reported PVY strains, and did not infect pepper and typical PepMoV isolates did not infect potato. Distinctive inclusion patterns of the scroll, pinwheel, long laminated inclusions, and helper components in the cytoplasm of infected cells were also different to those observed by the typical PVY isolate infections. However, the paprika isolate reacted to the monoclonal antibody of strain with high absorbance readings. RT-PCR amplification, cloning, and sequencing of the 3` untranslated region and a part of coat protein gene also added additional evidence of the paprika isolate as the -related isolate. Multiple alignments as well as cluster dendrograms of PVY-paprika isolate revealed close phylogenetic relationship to the subgroup. Altogether, these results suggest that a new PVY isolate infecting paprika contained distinct characteristics compared to the other previously described PVY strains with closer relationship to the strain.

This study was conducted to investigate the sap transmission pattern of CGMMV and to select the disinfectant of sap transmission on watermelon cultivation. Being investigated CGMMV occurrence change after transplanting two diseased plants among 47 plants on the watermelon house, disease spread rapidly from early growth stage of 4 % to late growth stage of 83 %. When the grafting knife and topping scissor was dipped onto the several disinfectants before grafting or topping, disease suppressed 87-95 % on whole milk, skim milk, tri-sodium phosphate, and sodium hypo-chloride solution. Disinfection effects of 88.2 % and 91.2 % on whole milk and skim milk were showed when the hand dipped on the disinfectants.

To analyze the relationship between CGMMV occurrence and seed contamination by cucumber cultivars, cucumber cultivars of CGMMV infected field and diseased degree were investigated in cucumber main cultivation regions of Jeonnam province from 1999 to 2002. While the diseased degree was different by years and cultivars, CGMMV occurred for 4 years in `Janghyeongnakhap`, for 3 years in `Jangjukcheongjang`, and for 2 years in `Gyeoulsali` and `Gyeoulnagi`. CGMMV was detected in seeds of `Janghyeongnakhap` and `Jangjukcheongjang` by ELISA test, and seed of `Jangjukcheongjang` showed positive reaction in bioassay. As a result of resistant test of 34 cultivars to CGMMV, all cultivars showed mosaic symptoms in pot test but only `Hanboksamcheok` showed mild mosaic symptom in field test.

Recent occurrence of virus diseases on watermelon plants cultivated in Jeonnam province was investigated from 1998 to 2002. While virus diseases were severely occurred on watermelon cultivated in green house in 1998, those of open field were severer than in green house since 2000. When 128 samples collected from different fields were examined by electron microscopy, 87.8％ of the samples contained rod-shaped or filamentous virus particles. RT-PCR analysis of the samples revealed that Cucumber green mottle mosaic virus (CGMMV) was only detected from collected samples at May. Watermelon mosaic virus (WMV) was most frequently found and CGMMV and Zucchini yellow mosaic virus (ZYMV) were slightly at June and July. However Cucumber mosaic virus (CMV) and Papaya ringspot virus (PRSV) have not been detected.

We investigated the effect of infection time of CGMMV on the growth and quality of watermelon and cucumber plants. The effect (damages by CGMMV) was estimated on the watermelon where CGMMV had been inoculated at different growth stages, vegetative (transplanting stage, vegetative growth stage) and reproductive growth stage (fruiting stage and fruit hypertrophy stage). In the case of cucumber, CGMMV was inoculated at transplanting stage and Erst flowering stage, respectively. When watermelon was infected with CGMMV at vegetative growth stage, vine length, internode length, leaf area, and fruit weight of the plants largely decreased compared with control plants, while the infected plant growth was not very different from control plants when it was infected at reproductive growth stage. Brix of the fruit of watermelon also decreased when the plants was infected with the virus earlier than fruiting stage. The occurrence of `Pisubag`, internal discoloration and decomposition of watermelon fruits, tended to be increased as earlier infection time with CGMMV In the case of cucumber infection time with CGMMV did not influence earlier growth of the plants, but did later growth showing that plant height, vine length, internode length, number of leaf, leaf wide, and leaf length of the plants decreased as infection time became to be earlier.

It was conducted to investigate the effect of rice downy mildew (RDM) infection to plant growth and yield components in water seeding stage, and to screen of varietal resistance to downy mildew. Being infected by rice downy mildew, chlorotic spot appeared in the leaf and leaf length was shortened. As the infected rice was growing, internode was not elongated properly and was deformed, and then panicle was not arised or mal-formed. Plant height of infected rice was shortened at all growth stage, and while the number of tillers of infected rice was more decreased than that of healthy plant before maximum tillering stage, and that of infected rice was more increased after heading stage. While the number of internode of infected tiller was much increased than that of healthy tiller internode length of infected tiller was shorter. As the rice infected by RDM severely, the number of panicles per square meter and ripening of rice was more decreased and yield of rice was extremely much decreased. As result of the varietal resistance screening with rice seedling, Geyh-wabyeo and Donjinbyeo were resistant varieties to downy mildew.

This research was conducted to develop a simple and effective method for pathogenicity assay of the causal agent of bacterial canker on kiwifruit. The developed method is a modified version of syringe-infiltration method that is used in the assay fer the hypersensitive response assay. Bacterial cell suspensions in 50 mM potassium phosphate buffer(pH 7.5) were infiltrated using a plastic syringe with 25G needle into primary leaves of five-year-old kiwifruit. Typical symptoms of bacterial canker were observed five days after infiltration. Symptoms developed on the leaves were detected in these inocula that treated above 10cfu/ml or above. Using this technique, host range of Pseudomonas syringae pv. actinidiae and three other plant pathogenic pseudomonads were investigated for 25 different plant species. The various symptoms were showed depend-ing on different plant species and inoculated pathogen combinations. This method has the advantage that symptoms can be showed faster compared to other methods and high humid conditions are not required.

to investigate occurrence of kiwifruit bacterial canker disease,172 kiwifmit orchards in Jeonnam and Gyeong-nam provinces were surveyed from March to April in 1999. In the south coast region of Korea, such as Haenam, Wando, Jindo, Jangheung, and Bosuns, red-rusty brown bacterial oozes were observed in 17 kiwifvuit orchards. Disease incidences varied from 2.2% to 100% depending on various regions. Total 22.8 ha of kiwifruit orchards were destroyed by occurrence backerial canker at 1999 in Korea. Orchard in Wando, Jindo, and Goheng were severely damaged in 1999. The seasonal variation of bacterial canker incidence was also investigated from 1996 to 1997 on leaves, vines and twigs of kiwifruit. Red-rusty brown bacterial ooze was exuded from mid February or early March to late April in creaked site of vine and twig. The seasonal incidence of bacterial canker on leaves appeared from late April to late June, and rapidly increased during May, Optimum growth temperature of Pseudomonas sytingae pv. actinidiae was at on King`s B medium and did not grow at 33. We suggest that spreading of bacterial canker was suppressed under the plastic flim and windbreak net house.

Field disease incidences of squash virus diseases in Jeonnam province were estimated to be 76.1% and of delayed planting on August-September (retarding culture) and on February-March (semi-forcing culture) on glass house were 55.0% and 0%, respectively, in 2000. Disease incidences of individual squash plant within a field were 100% and 3.6%, respectively, in wild culture and retarding culture. Total of 61 samples suspected to be infected with viruses were collected in 2000 and tested by RT-PCR using specific oligonulceotide primer sets designed for the detection of Cucumber green mottle mosaic virus (CGMMV), Kyuri green mottle mosaic virus (KGMMV), Zucchini yellow mosaic virus (ZYMV), Papaya ring spot virus (PRSV), Watermelon mosaic virus (WMV), and Cucumber mosaic virus (CMV). Each specific primer set for WMV, ZYMV, and PRSV amplified expected size of DNA fragments from 16, 10, and 2 samples in wild culture, respectively. Double or triple infection were observed in 7 samples tested. In contrast, each specific primer set for WMV, ZYMV, and PRSV confirmed virus infection from 7, 6, and 6 samples, respectively, in samples collected from semi-forcing culture. Double infection of WMV and PRSV was observed in only one sample. However, no DNA fragment was amplified from RT-PCR using CGMMV, KGMMV, and CMV specific oligonucleotide primer sets indicating no CGMMV, KGMMV, or CMV infection in squash fields in Jeonnam province in 2000.

A bacterium C1010, isolated from the rhizospheres of cucumbers in fields in Korea, degraded the microbial quorum-sensing molecules, hexanoyl homoserine lactone (HHSL), and octadecanoyl homoserine lactone (OHSL). Morphological characteristics and 16S rRNA sequence analysis identified C1010 as Acinetobacter sp. strain C1010. This strain was able to degrade the acyl-homoserine lactones (AHLs) produced by the biocontrol bacterium, Pseudomonas chlororaphis O6, and a phytopathogenic bacterium, Burkholderia glumae. Co-cultivation studies showed that the inactivation of AHLs by C1010 inhibited production of phenazines by P. chlororaphis O6. In virulence tests, the C1010 strain attenuated soft rot symptom caused by Erwinia carotovora ssp. carotovora. We suggest Acinetobacter sp. strain C1010 could be a useful bacterium to manipulate biological functions that are regulated by AHLs in various Gram-negative bacteria.Key words: quorum sensing, biological control, attenuation of bacterial diseases.
