12. Mechanisms of ROS-induced mitochondria-dependent apoptosis in Phaeodactylum tricornutum under AgNPs exposure.

Abstract

Introduction: Nanomaterials such as silver nanoparticles (AgNPs) have gained widespread application across various fields. However, the large-scale production and application of AgNPs have raised concerns about their distribution in the environment and potential pollution issues. Methods: This study investigates the toxic effects of AgNPs on the diatom Phaeodactylum tricornutum by employing electron microscopy for cellular observation, quantifying apoptotic cell numbers, and measuring antioxidant indicators. The research examines how varying concentrations of AgNPs induce stress in P. tricornutum and the specific mechanisms of the toxic effect. Results: The findings reveal that AgNPs induce apoptosis in P. tricornutum cells by triggering a mitochondria-mediated pathway, marked by a decrease in mitochondrial membrane potential and the activation of caspase enzymes. Additionally, AgNP exposure results in an overproduction of reactive oxygen species (ROS) within the algal cells, leading to lipid peroxidation of the cell membrane and a consequent increase in malondialdehyde (MDA) levels. This oxidative stress response induces the upregulation of antioxidant enzyme activities in an attempt to mitigate the excessive ROS. Discussion: ROS is identified as the primary factor responsible for inducing mitochondria-mediated apoptosis. Abstract

Seven new nitrogen-containing heptacyclic oligophenalenone dimers, talauromides A–G (1-7), together with known compounds bacillisporins A and B (8and 9), talaromycesones C and B (10and 11), duclauxin (12) and lamellicolic anhydride (13), were isolated from the soil derived-fungus Talaromyces stipitatusBMC-16. Their chemical structures were determined based on spectroscopic analysis data. The absolute configurations were elucidated by chemical approaches and the comparison of CD spectra with related compounds. Compounds 3, 8and 11exhibited inhibitory activity on the Wnt/β-catenin signaling pathway in zebrafish embryos at a concentration of 20 μM.

Abstract

We present an efficient method to solve the quantum discord of two-qubit X states exactly. A geometric picture is used to clarify whether and when the general POVM measurement is superior to von Neumann measurement. We show that either the von Neumann measurement or the three-element POVM measurement is optimal, and more interestingly, in the latter case the components of the postmeasurement ensemble are invariant for a class of states., Comment: 6 pages, 4 figures

29. Geometric picture of quantum discord for two-qubit quantum states

Abstract

Among various definitions of quantum correlations, quantum discord has attracted considerable attention. To find analytical expression of quantum discord is an intractable task. Exact results are known only for very special states, namely, two-qubit X-shaped states. We present in this paper a geometric viewpoint, from which two-qubit quantum discord can be described clearly. The known results about X state discord are restated in the directly perceivable geometric language. As a consequence, the dynamics of classical correlations and quantum discord for an X state in the presence of decoherence is endowed with geometric interpretation. More importantly, we extend the geometric method to the case of more general states, for which numerical as well as analytica results about quantum discord have not been found yet. Based on the support of numerical computations, some conjectures are proposed to help us establish geometric picture. We find that the geometric picture for these states has intimate relationship with that for X states. Thereby in some cases analytical expressions of classical correlations and quantum discord can be obtained., Comment: 9 figures

Abstract

The use of trastuzumab emtansine (T‐DM1) has revealed significant efficacy in HER2‐positive metastatic breast cancer (MBC). However, optimal therapeutic strategies following T‐DM1 failure remain a subject of debate in clinical practice. In this multicenter, retrospective, real‐world study, we sought to examine the effectiveness and safety of tyrosine kinase inhibitors (TKIs) as a therapeutic strategy in HER2‐positive MBC who developed T‐DM1 resistance. Between September 2018 and December 2022, 66 patients were enrolled. The median progression‐free survival of TKIs‐based therapy was 10.1 months (95% CI, 4.7–15.6). Objective response rate and clinical benefit rate were 18.2 and 66.7%, respectively. TKIs‐based therapy demonstrated better effectiveness in patients who had previously derived benefit from T‐DM1 and featured acquired resistance to trastuzumab. The most common adverse events were diarrhea (36, 54.5%), hand‐foot syndrome (31, 47.0%), and leucopenia (30, 45.5%). In conclusion, TKIs‐based therapy showed promising effectiveness and safety in HER2‐positive MBC patients after T‐DM1 failure. [ABSTRACT FROM AUTHOR]
