Objective: This study aimed to investigated the effect of ice-temperature vacuum drying on the powder quality of selenium-rich Rugao black pakchoi. Methods: After pretreatment, the fresh black pakchoi was dried to the moisture content of (5.0±0.5) %, and the physical and chemical indexes were investigated, comparing to the control groups of hot-air drying, vacuum drying and freeze-vacuum drying. Results: The vitamin C content of vacuum drying at ice temperature was 554.60 mg/100 g, and the retention rate reached 83.96%, which was 43.11%, 33.57% and 1.61% higher than that of hot-air drying, vacuum drying and freeze-vacuum drying, respectively. Glutathione peroxidase had little activity loss and the best purity maintenance. The retention rates of total phenols and total flavonoids were 59.69% and 60.58%, respectively, which were better than the other three groups. In terms of microstructure, the powder of ice-temperature vacuum drying group was well-distributed, with a smooth and flat surface and less debris. Compared with the other three groups, the ice-temperature vacuum drying group could maintain the original dark green color, and the rehydration and solubility were improved. Conclusion: The ice-temperature vacuum drying process can minimally affect the Rugao black pakchoi powder, and the nutritional quality of this powder is the highest.

Objective: The effect of frozen temperature on the storage characteristics of ready-to-eat snakehead sausages were investigated. Methods: In this study, The freezing temperature dehydration technique was used to control the moisture content of snakehead sausage at about 50%, and then the temperature was lowered to-5.6 ℃. Subsequently, the sausage was stored at -4 ℃, and those stored at 4 ℃ were set as the control. The changes in the quality and microstructure of the snakehead sausages were analyzed, by measuring the physicochemical indexes such as total bacterial colony, color, water holding capacity, textural properties, biogenic amines and scanning electron microscopy during storage. Results: The logarithmic values of TVC were （5.37±0.06）; TVB-N were （17±0.03） mg/100 g; the total amine mass fraction were （324.93±1.89） mg/kg at 42 d. The results of colony count and TVB-N analysis showed that, compared to those stored at 4 ℃, frozen storage could extend the shelf life of snakehead sausage by 1-fold. The low-field NMR results showed that the moisture distribution of snakehead sausage during frozen storage was better than that of the control. Scanning electron microscopy showed that the gel network of the frozen-storage group had a higher degree of binding to fat particles. The frozen storage of snakehead sausage was superior to the refrigerated storage. Conclusion: The preservation effect of snakehead sausage stored at freezing temperature is better than that of refrigerated storage.

Emerging studies have focused on ways to treat cancers by modulating T cell activation. However, whether B cell receptor signaling in the tumor microenvironment (TME) can be harnessed for immunotherapy is unclear. Here, we report that an Asia-specific variant of human IgG1 containing a Gly396 to Arg396 substitution (hIgG1-G396R) conferred improved survival of patients with colorectal cancer (CRC). Mice with knockin of the murine functional homolog mIgG2c-G400R recapitulated the alleviated tumorigenesis and progression in murine colon carcinoma models. Immune profiling of the TME revealed broad mobilizations of IgG1+ plasma cells, CD8+ T cells, CD103+ DCs, and active tertiary lymphoid structure formation, suggesting an effective antitumor microenvironment in hIgG1-G396R CRC patients. Mechanistically, this variant potentiated tumor-associated antigen–specific (TAA-specific) plasma cell differentiation and thus antibody production. These elevated TAA-specific IgG2c antibodies in turn efficiently boosted the antibody-dependent tumor cell phagocytosis and TAA presentation to effector CD8+ T cells. Notably, adoptive transfer of TAA-specific class-switched memory B cells harboring this variant exhibited therapeutic efficacy in murine tumor models, indicating their clinical potential. All these results prompted a prospective investigation of hIgG1-G396R in patients with CRC as a biomarker for clinical prognosis and demonstrated that manipulating the functionality of IgG1+ memory B cells in tumors could improve immunotherapy outcomes.

Objective: This study was to investigate the role of hsa-miR-155-3p and hsa-miR-155-5p as biomarkers and regulators of biological behavior in Systemic Sclerosis. Methods: A total of 10 SSc patients and 10 healthy controls were selected for the study. The expression levels of hsa-miR-155-3p and hsa-miR-155-5p in peripheral blood mononuclear cells of SSc patients and healthy controls were measured using RT-qPCR. The diagnostic value of these miRNAs was explored using Receiver Operating Characteristic curve analysis. Pearson or Spearman correlation analysis was performed to assess the correlation between miRNAs and clinical indicators in SSc patients. Potential target genes of hsa-miR-155-3p and hsa-miR-155-5p were predicted using miRDB, Targetscan, and miRDIP databases. GO functional annotation, KEGG pathway enrichment analysis, protein-protein interaction network construction, and selection of central genes were conducted. Results: The expression levels of hsa-miR-155-3p and hsamiR- 155-5p were significantly higher in PBMCs of SSc patients compared to healthy controls (P<0.001). The ROC curve analysis showed that hsa-miR-155-3p and hsa-miR-155-5p had a high diagnostic value for SSc (AUC=1, P<0.001). Correlation analysis revealed that hsamiR- 155-3p, hsa-miR-155-5p, and clinical indicators such as high-resolution CT, neutrophil percentage, lymphocyte percentage, and albumin to globulin ratio were correlated (P<0.05). The signaling pathways enriched with target genes of hsa-miR-155-3p and hsa-miR-155- 5p were closely associated with the occurrence and development of SSc fibrosis, immunity, and inflammation. Conclusions: hsa-miR-155-3p and hsa-miR-155-5p may be involved in regulating the occurrence and development of SSc fibrosis, immunity, and inflammation. They have the potential to serve as biomarkers for clinical diagnosis and treatment of SSc. [ABSTRACT FROM AUTHOR]

Curved ducts are widely used in aircraft engines to improve some capability of aero-engines. Complex internal flow characteristics would be induced by the curvature in such components. In this study, the influence of parameters, including the arc angle α , the curvature radius R i , and the height H, on the local accelerating and transonic flow in the curved ducts with equal area were studied numerically and theoretically under different nozzle pressure ratios (NPRs). The range of the Re number based on the height of the duct and the velocity at the inlet was 0.8 × 10 6 ∼ 1.6 × 10 6 . The shear stress transport κ-ω turbulent model was proved by the test data to suitably simulate the flow field in curved ducts because it could accurately predict the flow separations under adverse pressure gradients. The uncertainty of the pressure scan value to obtain the test data was ±0.05%. Numerical results showed that the effect of α on the flow characteristics of the curved ducts is little. The maximum Ma number in the curved section reduces with the increase of R i , and that grows with the increase of H. The range of the maximum Ma number was 1.20∼1.80. The critical NPRs, which decided the special flow features, were found in the curved ducts. The critical NPR rises with the increase of R i ; however, the effect of H on the critical NPR is irregular due to the flow separations located near the lower wall induced by the large adverse pressure gradient. The theoretical results based on the small perturbation theory of transonic flow in the polar coordinate system proved that the distribution of sonic line was just dependent on the inner diameter R 1, the outer diameter R 2, and the arc angle θ max of the curved section.
