Abstract

Background The cause of kidney failure is unknown in approximately 10% of patients with stage 5 chronic kidney disease (CKD). For those who first present to nephrology care with kidney failure, standard investigations of serology, imaging, urinalysis and kidney biopsy are limited differentiators of etiology. We aimed to determine the diagnostic utility of whole-genome sequencing (WGS) with analysis of a broad kidney gene panel in patients with kidney failure of unknown cause. Methods We prospectively recruited 100 participants who reached CKD stage 5 at 50 years of age and had an unknown cause of kidney failure after standard investigation. Clinically-accredited WGS was performed in this national cohort after genetic counselling. The primary analysis was targeted to 388 kidney-related genes with second-tier genome-wide and mitochondrial analysis. Results The cohort was 61% male and the average age of participants at stage 5 CKD was 32 years (9 months to 50 years). A genetic diagnosis was made in 25% of participants. Disease-causing variants were identified across autosomal dominant tubulointerstitial kidney disease (6), glomerular disorders (4), ciliopathies (3), tubular disorders (2), Alport syndrome (4) and mitochondrial disease (1). Most diagnoses (80%) were in autosomal dominant, X-linked or mitochondrial conditions (UMOD; COL4A5; INF2; CLCN5; TRPC6; COL4A4; EYA1; HNF1B; WT1; NBEA; m.3243A>G). Patients with a family history of CKD were more likely to have a positive result (OR 3.29, 95% CI 1.10-11.29). Thirteen percent of participants without a CKD family history had a positive result. In those who first presented in stage 5 CKD, WGS with broad analysis of a curated kidney-disease gene panel was diagnostically more informative than kidney biopsy, with biopsy being inconclusive in 24 of 25 participants. Conclusions In this prospectively ascertained Australian cohort, we identified a genetic diagnosis in 25% of patients with kidney failure of unknown cause.

Abstract

The tissue‐specific heart decellularized extracellular matrix (hdECM) demonstrates a variety of therapeutic advantages, including fibrosis reduction and angiogenesis. Consequently, recent research for myocardial infarction (MI) therapy has utilized hdECM with various delivery techniques, such as injection or patch implantation. In this study, a novel approach for hdECM delivery using a wet adhesive paintable hydrogel is proposed. The hdECM‐containing paintable hydrogel (pdHA_t) is simply applied, with no theoretical limit to the size or shape, making it highly beneficial for scale‐up. Additionally, pdHA_t exhibits robust adhesion to the epicardium, with a minimal swelling ratio and sufficient adhesion strength for MI treatment when applied to the rat MI model. Moreover, the adhesiveness of pdHA_t can be easily washed off to prevent undesired adhesion with nearby organs, such as the rib cages and lungs, which can result in stenosis. During the 28 days of in vivo analysis, the pdHA_t not only facilitates functional regeneration by reducing ventricular wall thinning but also promotes neo‐vascularization in the MI region. In conclusion, the pdHA_t presents a promising strategy for MI treatment and cardiac tissue regeneration, offering the potential for improved patient outcomes and enhanced cardiac function post‐MI. 36. Fabrication and characterization of corrosion resistant amorphous-nanocrystalline alloy Ni49Co3Mo22B26by galvanostatic deposition

Abstract

Amorphous materials have unique physical properties but have limited practical use due to their low ductility and thermal instability. A quaternary Ni49Co3Mo22B26amorphous-nanocrystalline plating was prepared by electrodeposition to obtain good hardness, corrosion resistance, and thermal stability. Multiple electrolyte baths with boric acid concentration ranging from 0.3 M to 0.8 M were investigated to study the co-deposition pattern of molybdenum (Mo) and boron (B) with iron-group elements. Experimentations suggest that an array of binary, ternary, and quaternary alloys can be fabricated by adjusting boric acid concentration in an electrolyte bath. Energy Dispersive X-ray Spectroscopy (EDS) analysis shows that co-deposition of Mo requires sluggish mass transport of iron group element nickel (Ni) and cobalt (Co) and high boric acid concentration retards Mo deposition entirely. X-ray diffraction (XRD) patterns were examined to identify the formation of amorphous-nanocrystalline film deposition. An average hardness for quaternary Ni-Co-Mo-B deposit samples was measured between the range of 380 HV- 450 HV, and the hardness increases with increased B content. Electrochemical corrosion tests reveal that the addition of Mo in quaternary Ni-Co-Mo-B increases corrosion performance in saline environments but reduces corrosion performance in acidic environments over Ni-Co-B amorphous alloy. Differential scanning calorimetry (DSC) scan shows that prepared amorphous-nanocrystalline material has thermal stability up to 346 °C without undergoing phase change.

AI-based face recognition for missing person searches is a promising strategy that has the potential to significantly speed up and improve accuracy. The system makes use of artificial intelligence algorithms to match surveillance camera realtime video footage with facial images of people who have gone missing. Face recognition technology in video surveillance systems is used in this project to come up with a strategy for locating people who have gone missing. The system involves gathering information about the missing person, creating a database of facial images, and matching those images to real-time video footage with the help of artificial intelligence algorithms. The goal of the field of computer science known as artificial intelligence (AI) is to create intelligent machines that can carry out tasks that typically call for human intelligence. This includes tasks like translating languages, visual perception, speech recognition, and decision-making. Machine learning algorithms are used by AI systems to learn from data and allow them to improve their performance over time. A powerful method for training artificial neural networks with many layers, deep learning, a subset of machine learning, has emerged, allowing. The system can be used to quickly identify and locate missing people in public places like airports and train stations. The speed and accuracy of missing person searches could be significantly enhanced by the proposed system, increasing the likelihood of successful reunions. Finding missing people in view of face acknowledgment utilizing Convolutional Brain Organization (CNN) calculation is a well-known approach that has shown promising outcomes. CNN is a deep learning algorithm that works well for face recognition because it is widely used for image recognition and classification.
