Abstract

Peanuts (Arachis hypogaea L.) contain nonnutritive proactive and bioactive phytochemicals that have been associated with a decreased risk of cardiovascular disease and certain cancers. This study was designed to determine the effect of dietary peanuts at 10, 15, and 20% on colon tumors at the initiation (I), promotion (P), and initiation + promotion (I + P) stages (42 rats/treatment) in Fisher 344 male rats. After a 1-wk acclimatization period, rats were divided into 8 groups and fed AIN93G (Control-C) and 3 experimental diets. At ages 7 and 8 wk, all rats were treated with subcutaneous injections of azoxymethane (AOM) in saline (16 mg/kg body wt). The rats were fed the assigned diets until they were killed by C[O.sub.2] at age 45 wk. Cecal pH, cecal weight, feed intake, weight gain, tumors in the colon and liver, and glutathione S-transferase activity were determined. There were no significant (P < 0.05) differences in cecal weight, although cecal pH differed. Tumor incidences (%) in the colon of rats in the control, I, P, and I + P groups at 10, 15, and 20% were 100, 75, 42, and 36; 100, 46, 41, and 41; and 100, 54, 36, and 23, respectively. Colon tumors per tumor-bearing rat were 3.22, 1.7, 2.0, and 1.0; 3.22, 3.1, 2.5, and 2.1; and 3.22, 2.2, 1.1, and 0.6, respectively. Glutathione S-transferase values were 12.5, 29.3, 30.4, and 31.4; 12.5, 29.4, 30.3, and 31.8; and 12.5, 29.5, 30.5, and 31.2, respectively. The results indicate that dietary peanuts suppress colon tumors, particularly at the promotion stages in AOM-induced colon tumors. Peanuts may therefore play a role in chemoprevention if incorporated in the daily diet.

37. Insulin resistance during androgen deprivation therapy in men with prostate cancer.

Abstract

Background Methods Results Conclusions Androgen deprivation therapy (ADT) in prostate cancer (PCa) has been associated with development of insulin resistance. However, the predominant site of insulin resistance remains unclear.The ADT & Metabolism Study was a single‐center, 24‐week, prospective observational study that enrolled ADT‐naive men without diabetes who were starting ADT for at least 24 weeks (ADT group, n = 42). The control group comprised men without diabetes with prior history of PCa who were in remission after prostatectomy (non‐ADT group, n = 23). Prevalent diabetes mellitus was excluded in both groups using all three laboratory criteria defined in the American Diabetes Association guidelines. All participants were eugonadal at enrollment. The primary outcome was to elucidate the predominant site of insulin resistance (liver or skeletal muscle). Secondary outcomes included assessments of body composition, and hepatic and intramyocellular fat. Outcomes were assessed at baseline, 12, and 24 weeks.At 24 weeks, there was no change in hepatic (1.2; 95% confidence interval [CI], −2.10 to 4.43; p = .47) or skeletal muscle (−3.2; 95% CI, −7.07 to 0.66; p = .10) insulin resistance in the ADT group. No increase in hepatic or intramyocellular fat deposition or worsening of glucose was seen. These changes were mirrored by those observed in the non‐ADT group. Men undergoing ADT gained 3.7 kg of fat mass.In men with PCa and no diabetes, 24 weeks of ADT did not change insulin resistance despite adverse body composition changes. These findings should be reassuring for treating physicians and for patients who are being considered for short‐term ADT. [ABSTRACT FROM AUTHOR]
