In the era of circular economies, municipal wastewater treatment plants (WWTPs) are viewed as resource recovery facilities. At the very minimum, the targeted resources are water, biogas, and phosphorus. However, municipal wastewater streams (sludge and effluent) need to be adequately treated to eliminate the potential for the transmission of microbial pathogens including protozoa, bacteria, and viruses. This paper presents the results from a study demonstrating the use of electron beam technology for sludge hygenization and enhanced methane (biogas) production using municipal wastewater samples. Cogeneration of heat for fertilizer drying and granulation and electricity for powering the electron beam system are also demonstrated.

Per- and polyfluoroalkyl substances (PFASs) are lipophilic pollutants of increasing health concern due to their environmental persistence, bioaccumulation, and toxicity. Remediation techniques have been set up for PFAS removal, such as electron beam, which exhibits excellent performance due to its cost-effective ability of producing large amounts of both reducing and oxidizing species. However, the degradation efficiency and mechanisms of the shorter-chain PFASs by eBeam irradiation are poorly known. Herein, perfluoroheptanoic acid was exposed to different eBeam doses of 0–75 kGy. We tested the effect of pH (6.0 and 13.0), nitrate ions, and fulvic acids in water. Results show that highly alkaline conditions favored the removal of perfluoroheptanoic acid at 100.0 μg/L with complete elimination at eBeam doses of 50 kGy and 75 kGy. Nitrate, fulvic acid, and CaCO3 alkalinity did not inhibit perfluoroheptanoic acid degradation. Defluorination and decarboxylation are proposed to be the major degradation pathways based on the identification of two degradation products. Overall, our findings show that eBeam irradiation is a promising remediation technique to break down the shorter-chain PFASs.

Ionizing radiation including electron beam (eBeam) technology has been used as a quarantine measure around the world for transboundary shipment of agricultural products. Since the treatment goal of this technology is not to instantly kill insects but to cause them to be sterile, an apparent challenge is how to ensure that the agricultural commodities have been exposed to ionizing radiation if live insects are found. Because irradiation affects DNA synthesis and cell division, we investigated the feasibility of using flow cytometry detection of eBeam irradiation-induced ploidy change as a diagnostic tool. All eBeam-treated holometabolous insects tested including cowpea bruchid (Callosobruchus maculatus), corn earworm (Helicoverpa zea) and fruit fly (Drosophila melanogaster) displayed significantly higher proportions of polyploid cells than their respective unirradiated controls. However, such increased DNA ploidy was mostly absent in irradiated hemimetabolous sand field cricket (Gryllus firmus), green peach aphid (Myzus persicae) and potato/tomato psyllid (Bactericera cockerelli). Further studies with representative holometabolous insects revealed that the eBeam-triggered DNA endoreplicative response was strong in larvae, but became weaker as insects continued to develop into pupae and was even diminished in adults. Therefore, the flow cytometry-based method has the potential to be developed into a diagnostic tool to determine whether juvenile insects found on agricultural shipments, particularly those that undergo complete metamorphosis, have received irradiation as a quarantine treatment. This simple and rapid detection method could potentially resolve the dilemma that occurs in border control and domestic movement of plant material and products from areas under quarantine.

We study a generalized location‐routing problem in which the key decisions are made by supply chain partners with conflicting objectives. The context of our problem is the irradiation by electron beam (eBeam) of fresh produce imported from Mexico to reduce the threat of insects and pests to the U.S. agriculture. Because too few irradiation facilities exist to serve the current demand, we focus on two parties to be coordinated: the eBeam Services Provider, who will choose the facilities’ locations and capacities, and the Distributor, who will determine routes for transporting fruits from Mexico to commercial hubs in the United States via these facilities. We demonstrate the value of cooperation and how that cooperation can be achieved and enforced for a supply chain that must coordinate the independent companies by the strategic decisions of facility location and capacity construction, as opposed to the more common coordination by capacity allocation or pricing. The parties’ interactions are modeled as a sequential game with perfect information. Specifically, we formulate the sequential and cooperative decision‐making problems as mixed‐integer programs, analyze the computational complexity of the problems, and conduct extensive computational experiments. Additionally, we detail three schemes by which the parties can engage in profitable and enforceable cooperation. These methods progressively increase each party’s commitment as trust is built and profits rise. Total cooperation increases overall profit by an average of 8.6%. Further, a stochastic program that uses sample average approximation is applied to demonstrate the results’ robustness to nature’s supply variations while maximizing the supply chain’s overall profit.

Beginning with the outbreak of COVID-19 at the dawn of 2020, the continuing spread of the pandemic has challenged the healthcare market and the supply chain of Personal Protective Equipment (PPE) around the world. Moreover, the emergence of the variants of COVID-19 occurring in waves threatens the sufficient supply of PPE. Among the various types of PPE, N95 Respirators, surgical masks, and medical gowns are the most consumed and thus have a high potential for a serious shortage during such emergencies. Considering the unanticipated demand for PPE during a pandemic, re-processing of used PPE is one approach to continue to protect the health of first responders and healthcare personnel. This paper evaluates the viability and efficacy of using FDA-approved electron beam (eBeam) sterilization technology (ISO 11137) to re-process used PPE. PPEs including 3M N95 Respirators, Proxima Sirus gowns, and face shields were eBeam irradiated in different media (air, argon) over a dose range of 0-200 kGy. Several tests were then performed to examine surface properties, mechanical properties, functionality performance, discoloration phenomenon, and liquid barrier performance. The results show a reduction of filtration efficiency to about 63.6% in the N95 Respirator; however, charge regeneration may improve the re-processed efficiency. Additionally, mechanical degradation was observed in Proxima Sirus gown with increasing dose up to 100 kGy. However, no mechanical degradation was observed in the face shields after 10 times donning and doffing. Apart from the face shield, N95 Respirators and Proxima Sirus gown both show significant mechanical degradation with ebeam dose over sterilization doses (25 kGy), indicating that eBeam technology is not appropriate for the re-processing these PPEs.

The potato/tomato psyllid Bactericera cockerelli causes serious damage to several solanaceous crops by direct feeding and vectoring Candidatus Liberibacter solanacearum, a bacterial pathogen. Electron beam (eBeam) irradiation is an environmentally friendly, chemical-free alternative method that is increasing in use for disinfestation of insect pests. We hypothesize that this irradiation technology will have detrimental effects on potato psyllid and thus impede its disease vectoring. To this end, we explored the effects of eBeam treatment ranging from 50 to 500 Gy on survival, development and reproduction of this pest. Impact on psyllids was apparently dose-dependent. When irradiated at 350 Gy, eggs could not hatch, 1st instar nymphs failed to emerge, and although a small portion of irradiated 5th instar nymphs survived, the emerged adults were mostly deformed. Abnormality in eclosed adults suggests harmful effects of eBeam on metamorphosis. Reproduction was seriously impaired when female psyllids were exposed to eBeam at the 5th instar nymphal or young adult stage, presumably due to inability to form oocytes. In addition, reciprocal crosses between irradiated and untreated psyllids indicated that female psyllids were more radiosensitive than males to eBeam. Taken together, these findings indicate that eBeam negatively impacted potato psyllid development and reproduction, which would inevitably compromise its disease transmission capacity. A dose of 350 Gy can be considered as a reference dose for effective control of potato psyllids.

Harmful algal and cyanobacterial blooms pose threats to human and ecological health due to their release of hazardous toxins. Microcystin-LR (MC-LR), a potent hepatotoxin, is the most prevalent cyanotoxin found in freshwater blooms. Although produced by many species of cyanobacteria, Microcystis aeruginosa is most commonly associated with MC-LR production. These blooms are increasing in occurrence in lakes, ponds, and other surface waters and, therefore, require efficient treatment methods to be removed from water supplies. Ionizing radiation technologies offer promising approaches for the removal of organic pollutants in water, including cyanotoxins and cyanobacteria. Gamma irradiation for the degradation of cyano-bacteria and toxins is effective for overall MC-LR degradation as well as reducing cell concentrations. However, gamma irradiation technology involves use of radioactive isotopes and, therefore, may not feasible commercially from a security perspective. Electron beam (eBeam) irradiation technology, which relies on regular electricity to generate highly energetic electrons, is able to achieve the same results without the confounding challenges of radioactive isotopes and related security issues. In this critical review, the current state of the science concerning the remediation of MC-LR and M. aeruginosa with ionizing radiation technologies is presented and future necessary research is discussed.

Prescottella equi (formerly Rhodococcus equi) is a facultative intracellular bacterial pathogen that causes severe pneumonia in foals 1–6 months of age, whereas adult horses are highly resistant to infection. We have shown that vaccinating pregnant mares against the conserved surface polysaccharide capsule, β-1 → 6-linked poly-N-acetyl glucosamine (PNAG), elicits opsonic killing antibody that transfers via colostrum to foals and protects them against experimental infection with virulent. R. equi. We hypothesized that equine IgG1 might be more important than IgG4/7 for mediating protection against R. equi infection in foals. To test this hypothesis, we compared complement component 1 (C1) deposition and polymorphonuclear cell-mediated opsonophagocytic killing (OPK) mediated by IgG1 or IgG4/7 enriched from either PNAG hyperimmune plasma (HIP) or standard plasma. Subclasses IgG1 and IgG4/7 from PNAG HIP and standard plasma were precipitated onto a diethylaminoethyl ion exchange column, then further enriched using a protein G Sepharose column. We determined C1 deposition by enzyme-linked immunosorbent assay (ELISA) and estimated OPK by quantitative microbiologic culture. Anti-PNAG IgG1 deposited significantly (P < 0.05) more C1 onto PNAG than did IgG4/7 from PNAG HIP or subclasses IgG1 and IgG4/7 from standard plasma. In addition, IgG1 from PNAG HIP mediated significantly (P < 0.05) greater OPK than IgG4/7 from PNAG HIP or IgG1 and IgG4/7 from standard plasma. Our findings indicate that anti-PNAG IgG1 is a correlate of protection against R. equi in foals, which has important implications for understanding the immunopathogenesis of R. equi pneumonia, and as a tool for assessing vaccine efficacy and effectiveness when challenge is not feasible.

This study investigates the microbiological and immunological basis underlying the efficacy of electron beam-inactivated immune modulators. The underlying hypothesis is that exposure to eBeam-based ionization reactions inactivate microorganisms without modifying their antigenic properties and thereby creating immune modulators. The immunological correlates of protection induced by such eBeam based Salmonella Typhimurium (EBST) immune modulators in dendritic cell (DC) (in vitro) and mice (in vivo) models were assessed. The EBST stimulated innate pro inflammatory response (TNFα) and maturation (MHC-II, CD40, CD80 and CD86) of DC. Immuno-stimulatory potential of EBST was on par with both a commercial Salmonella vaccine, and live Salmonella cells. The EBST cells did not multiply under permissive in vitro and in vivo conditions. However, EBST cells remained metabolically active. EBST immunized mice developed Salmonella-specific CD4+ T-cells that produced the Th1 cytokine IFNγ at a level similar to that induced by the live attenuated vaccine (AroA- ST) formulation. The EBST retained stable immunogenic properties for several months at room temperature, 4°C, and -20°C as well as after lyophilization. Therefore, such eBeam-based immune modulators have potential as vaccine candidates since they offer the safety of a “killed” vaccine, while retaining the immunogenicity of an “attenuated” vaccine. The ability to store eBeam based immune modulators at room temperature without loss of potency is also noteworthy.

Clostridium perfringens (Cp) is a Gram-positive anaerobe that is one of the causative agents of necrotic enteritis (NE) in chickens, which leads to high mortality. Owing to the ban of administering antibiotics in feed to chickens, there has been an increase in the number of NE outbreaks all over the world, and the estimated loss is approximately 6 billion U.S. dollars. The best alternative method to control NE without antibiotics could be vaccination. In this study, we exposed three different strains of Cp to electron beam (eBeam) irradiation to inactivate them and then used them as a killed vaccine to control the colonization of Cp in broiler chickens. The vaccine was delivered to 18-day old embryos in ovo and the chickens were challenged with the respective vaccine strain at two different time points (early and late) to test the protective efficacy of the vaccine. The results indicate that an effective eBeam dose of 10 kGy inactivated all three strains of Cp, did not affect the cell membrane or epitopes, induced significant levels of IgY in the vaccinated birds, and further reduced the colonization of Cp strains significantly (p &lt, 0.0001) in late challenge (JGS4064: 4 out of 10, JGS1473: 0 out of 10, JGS4104: 3 out of 10). Further studies are necessary to enhance the efficacy of the vaccine and to understand the mechanism of vaccine protection.

Background Electron beam (eBeam) irradiation and hermetic storage are safe and effective technologies to protect stored products. Although hypoxic environment improves performance of some irradiated insects, whether hypoxia affects irradiation of storage insects and impacts pest control efficacy remains to be investigated. Results Using cowpea bruchid (Callosobruchus maculatus) larvae, we showed that, relative to eBeam irradiation under normoxia, the adult emergence rate increased substantially if they were exposed to hypoxia prior to and during eBeam treatment. Conversely, exposure to hypoxia only after eBeam irradiation did not have this protective effect. eBeam irradiation caused an increase in reactive oxygen species (ROS) in normoxic larvae but not in hypoxic larvae. The activity of citrate synthase, a pace-making enzyme in the citric acid cycle, was suppressed under hypoxia but resumed normal function within hours of reoxygenation, suggesting that reduced mitochondrial activity, and thus less ROS production under hypoxia increased insect tolerance to irradiation. Furthermore, reoxygenation accelerated eBeam-induced glutathione-S-transferase activation and potentiated eBeam-enhanced catalase activities. Faster and stronger detoxification capacity in eBeam-irradiated, hypoxic larvae may have protected them from oxidative damage. Conclusion Hypoxic environment enhanced radiotolerance of bruchid larvae, presumably due to limited ROS production and elevated antioxidant enzymatic activities after reoxygenation. © 2018 Society of Chemical Industry.

To meet the increasing global demand for fresh produce, robust processing methods that ensures both the safety and quality of fresh produce are needed. Since fresh produce cannot withstand thermal processing conditions, most of common safety interventions used in other foods are ineffective. Electron beam (eBeam) is a non-thermal technology that can be used to extend the shelf life and ensure the microbiological safety of fresh produce. There have been studies documenting the application of eBeam to ensure both safety and quality in fresh produce, however, there are still unexplored areas that still need further research. This is a critical review on the current literature on the application of eBeam technology for fresh produce.

As manned space missions extend in length, the safety, nutrition, acceptability, and shelf life of space foods are of paramount importance to NASA. Since food and mealtimes play a key role in reducing stress and boredom of prolonged missions, the quality of food in terms of appearance, flavor, texture, and aroma can have significant psychological ramifications on astronaut performance. The FDA, which oversees space foods, currently requires a minimum dose of 44 kGy for irradiated space foods. The underlying hypothesis was that commercial sterility of space foods could be achieved at a significantly lower dose, and this lowered dose would positively affect the shelf life of the product. Electron beam processed beef fajitas were used as an example NASA space food to benchmark the minimum eBeam dose required for sterility. A 15 kGy dose was able to achieve an approximately 10 log reduction in Shiga-toxin-producing Escherichia coli bacteria, and a 5 log reduction in Clostridium sporogenes spores. Furthermore, accelerated shelf life testing (ASLT) to determine sensory and quality characteristics under various conditions was conducted. Using Multidimensional gas-chromatography–olfactometry–mass spectrometry (MDGC-O-MS), numerous volatiles were shown to be dependent on the dose applied to the product. Furthermore, concentrations of off –flavor aroma compounds such as dimethyl sulfide were decreased at the reduced 15 kGy dose. The results suggest that the combination of conventional cooking combined with eBeam processing (15 kGy) can achieve the safety and shelf-life objectives needed for long duration space-foods.

Remediation technologies are urgently needed to degrade poly – and per fluoroalkyl substances (PFAS) in groundwater and soils. Since high energy electron beam (eBeam) technology involves rapid oxidation and reduction reactions it has the potential to breakdown PFAS. In this study, the technology was applied to PFAS-contaminated groundwater and soil samples to degrade PFOS and PFOA and other PFAS. An eBeam dose of 2000 kGy (kGy) reduced PFOS and PFOA concentrations in groundwater by 87.9% and 53.7% respectively. In soils with 10% moisture content, PFOS and PFOA concentrations were reduced by 99.9% and 86.5%, respectively. Out of 17 different PFAS that were detected in the soil sample, the 2000 kGy dose was able to degrade 10 PFAS to below detectable levels: PFNA (initially 19.4 ng/g), PFDA (3.9 ng/g), PFPeS (12.1 ng/g), PFHxS (365 ng/g), PFHpS (18.0 ng/g), PFNS (4.3 ng/g), 4:2 FTS (16.6 ng/g), 6:2 FTS (146.1 ng/g), 8:2 FTS (165.6 ng/g) and PFOSA (171.8 ng/g). Of the remaining 7 PFAS, the removal efficiency ranged between 49% and 99.9%. It appears that eBeam degradation of PFAS in soil is more effective at reduced soil moisture content.

Harmful cyanobacterial blooms (cyanoHABs) pose threats to human and animal health due to the production of harmful cyanotoxins. Microcystis aeruginosa is a common cyanobacterium associated with these blooms and is responsible for producing the potent cyclic hepatotoxin microcystin-LR (MC-LR). Concerns over the public health implications of these toxins in water supplies have increased due to rising occurrence of these blooms. High energy electron beam (eBeam) irradiation technology presents a promising strategy for the mitigation of both cyanobacterial cells and cyanotoxins within the water treatment process. However, it is imperative that both cellular and chemical responses to eBeam irradiation are understood to ensure efficient treatment. We sought to investigate the effect of eBeam irradiation on M. aeruginosa cells and MC-LR degradation. Results indicate that doses as low as 2 kGy are lethal to M. aeruginosa cells and induce cell lysis. Even lower doses are required for degradation of the parent MC-LR toxin. However, it was observed that there is a delay in cell lysis after irradiation where M. aeruginosa cells may still be metabolically active and able to synthesize microcystin. These results suggest that eBeam may be suitable for cyanoHAB mitigation in water treatment if employed following cell lysis.

There is a growing need for increased efficiency in the sterilization of single use medical devices and other products that contain polymer components. Gamma radiation is widely used for devices suited for radiation sterilization; however, safety, throughput and cobalt-60 source availability are challenging the prospect of relying on gamma radiation to meet the anticipated needs of the industry. Use of electron beam (e-beam) and X-rays as alternatives to gamma for radiation sterilization have been hampered in part by a concern that these modalities may adversely affect polymer integrity and performance relative to the gamma method, for which the industry has had much more experience. To address this concern, the effects of sterilization-relevant doses of e-beam, X-ray and gamma radiation were directly compared using common medical device polymers found in two prototypical commercial devices currently sterilized using cobalt-60 gamma irradiation. The Becton, Dickinson and Company (BD) Vacutainer™ Plus tube contains low-density polyethylene and chlorobutyl rubber components, while the BD Vacutainer™ Push Button Blood Collection Set contains polypropylene homopolymer and polyolefin elastomer components. Injection-molded samples prepared from the polymers used in these products were exposed to target doses of 15, 35, 50 and 80 kGy using gamma, e-beam and X-ray radiation. Changes in coloration, tensile properties and hardness were measured for each condition, and the effects of e-beam and X-ray irradiation compared with the effects of gamma irradiation on these properties. Both e-beam and X-ray appear as viable alternatives to gamma irradiation for sterilization of the polymers tested.

Raw milk is known to contain relatively high numbers of microorganisms, some of which include microbial pathogens. Electron beam (eBeam) processing is a nonthermal pasteurization food processing technology. The underlying hypothesis was that eBeam processing will not negatively influence the composition, nutrient content, and aroma profile of raw milk. Raw milk samples were exposed to eBeam doses of 1 and 2 kGy, since our studies had shown that 2 kGy is suitable for raw milk pasteurization. The untreated and eBeam-treated raw milk samples were analyzed to detect changes in lactose, vitamin B2 , vitamin B12 , and calcium concentrations. The possible breakdown of casein and whey proteins and lipid oxidation were investigated along with the formation of volatile aroma compounds. Even though vitamin B2 showed a 31.6% decrease in concentration, the B2 content in eBeam-pasteurized raw milk met all USDA nutritional guidelines. Even though there were no indications of lipid oxidation after the 2.0-kGy eBeam treatment, there was lipid oxidation (58%) after 7 d of refrigerated storage. However, based on the GC-olfactory analysis, the lipid oxidation did not necessarily result in the development of a wide variety of off-odors.

Strawberries are vulnerable to harboring microbial pathogens because they are generally not washed due to their perishable nature. The focus of this study was to quantify the reduction in infection risks associated with non-O157 Shiga toxin producing Escherichia coli serotypes contaminated strawberries if the strawberries are exposed to low doses ∼1 kGy (kiloGray) of electron beam (eBeam) irradiation. A cocktail of six serotypes of non O157 E. coli namely, O26:H11, O45:H2, O103:H2, O111:NM, O121:H19, and O145 was employed. Strawberry puree rather than whole strawberries were used in this study to ensure dose uniformity that is critical for accurate interpretation of microbial reduction. The results show that when these serotypes are exposed to ≤1 kGy eBeam dose, there is approximately 4-log reduction in their numbers when they are present within a strawberry matrix (puree). Quantitative microbial risk assessments suggest that if a typical strawberry serving (150 g) was heavily contaminated (∼105 CFU/serving size), 2 out of 10 susceptible individuals (20%) would get sick (without eBeam treatment). However, if these contaminated strawberries had been treated with 1 kGy of eBeam dose, the infection risks would have be significantly reduced to approximately 4 out of every 100,000 individuals (0.004%). Similarly, even at low levels of contamination (∼102 CFU/serving), the infection risks would be reduced from 6 out of 10,000 susceptible individuals to approximately 4 out of 100 million susceptible individuals.

Ionizing radiation such as Electron beam (EB) and gamma irradiation inactivate microbial cells preventing their multiplication. These cells, however, are structurally intact and appear to have residual metabolic activity. We were interested in understanding the metabolic pathways that were still functional in EB-inactivated cells. Therefore, the primary objective of this study was to compare the metabolites accumulating in EB-inactivated pathogens E. coli 026:H11 and S. Typhimurium immediately after EB inactivation and 24 h post inactivation. Defined aliquots (109 CFU/mL) of E. coli O26-H11 (TW 1597) and S. Typhimurium (ATCC 13311) suspended in phosphate-buffered saline were exposed to lethal EB doses of 3 kGy and 2 kGy, respectively. Complete inactivation (inability of cells to multiply) was confirmed by traditional plating methods. An untargeted analysis of the primary metabolites accumulating in un-irradiated (control) cells, EB-inactivated cells immediately after irradiation, and EB-inactivated cells that were incubated at room temperature for 24 h post EB inactivation was performed using gas chromatography/mass spectrometry. A total of 349 different metabolites were detected in the EB-inactivated S. Typhimurium and E. coli O26:H11 cells, out of which, only 50% were identifiable. In S. Typhimurium, 98 metabolites were expressed at statistically different concentrations (P < 0.05) between the three treatment groups. In E. coli O26:H11, 63 metabolites were expressed at statistically different concentrations (P < 0.05) between the three treatment groups. In both these pathogens, the β-alanine, alanine, aspartate, and glutamate metabolic pathways were significantly impacted (P < 0.01). Furthermore, the metabolomic changes in EB-inactivated cells were amplified significantly after 24 h storage at room temperature in phosphate-buffered saline. These results suggest that EB-inactivated cells are very metabolically active and, therefore, the term Metabolically Active yet Non-culturable is an apt term describing EB-inactivated bacterial cells.

Electron beam (eBeam) irradiation is one of the main three principal ionizing radiation techniques that are available to myriads of different industries including the food industry. A magnet focuses the generated beam of electrons in order to control the pattern in which the beam leaves the gun. Considering the source of energy, speed, cost, and safety, electron beam accelerators are becoming preferred to cobalt-60-based gamma ray food irradiation technology. The earliest type of irradiation introduced to the food industry was gamma irradiation in 1960 as an alternative method to canning. Although the original development of electron beam linear accelerators took place during the 1930s, it took some time for the technology to be commercialized. When adopting electron beam technology for food irradiation, one has to have a very clear idea of the primary goal of the application. The penetration of eBeam into food packages depends on the areal density of the package.

Various proteins or protein fractions reportedly positively affect gastrointestinal integrity and inflammation in diets providing &gt, 45% energy as fat. This study tested whether benefits were seen in diets providing 30% of energy as fat. Purified diets (PD) with isolated soy protein (ISP), dried whole milk powder (DWMP), milk fat globule membrane (MFGM), or milk protein concentrate (MPC) as protein sources were fed to C57BL/6J mice (n = 15/diet group) for 13 weeks. MFGM-fed mice were heaviest (p &lt, 0.005) but remained within breeder norms. Growth rates and gut motility were similar for all PD-fed mice. FITC-dextran assessed gut permeability was lowest in DWMP and MFGM (p = 0.054), overall, plasma endotoxin and unprovoked circulating cytokines indicated a non-inflammatory state for all PD-fed mice. Despite differences in cecal butyrate and intestinal gene expression, all PDs supported gastrointestinal health. Whole milk provided more positive effects compared to its fractions. However, ISP-fed mice showed a &gt, 370%, (p &lt, 0.006) increase in colonic myeloperoxidase activity indicative of tissue neutrophil infiltration. Surprisingly, FITC-dextran and endotoxin outcomes were many folds better in PD-fed mice than mice (strain, vendor, age and sex matched) fed a “chow-type” nutritionally adequate non-PD. Additional variables within a diet’s matrix appear to affect routine indicators or gastrointestinal health.

Effective sterilization methods for single-use devices are a growing need for the medical industry. Concerns with safety, throughput and source availability, however, prompt prudent contingency planning for gamma irradiation of devices suited for radiation sterilization. Electron beam (e-beam) and X-ray represent two alternatives to gamma radiation if they can be confirmed to be compatible with sterilization of the devices. To address this question, the effects of sterilization-relevant doses of e-beam and X-ray radiation are directly compared to the effects of gamma radiation using two prototypical commercial devices currently sterilized using cobalt-60 gamma radiation. These devices include components that comprise six distinct polymer materials commonly used in the medical device industry. The devices investigated are the Becton, Dickinson and Company (BD) VacutainerTM Plus tube, comprised of low-density polyethylene, chlorobutyl rubber, and polyethylene terephthalate components; and the BD VacutainerTM Push Button Blood Collection Set, containing polypropylene, polyolefin elastomer, and polyvinyl chloride components. Changes in functionality, discoloration and select mechanical properties of components of each device were measured following exposure to targeted doses of 15, 35, 50 and 80 kGy. A statistical analysis was performed to determine if the effects of e-beam or X-ray radiation differ from the effects of gamma radiation for the properties considered. No devices were found to fail the functional performance tests at any of the doses considered. Small, but statistically significant differences were observed in device discoloration from e-beam, X-ray and gamma radiation following processing for certain materials at certain dose levels. Both e-beam and X-ray irradiation appear as viable alternatives to gamma irradiation for sterilization of the medical devices and materials considered.

In recent years, perfluorooctanoic acid (PFOA) and perfluorooctane sulfonate (PFOS) have been recognized as widespread persistent organic pollutants and identified as a great concern to public health. Electron beam (eBeam) technology is a form of ionizing radiation that had been previously successful in mediating the breakdown of PFOA and PFOS. These studies report complete breakdown of PFOA and partial breakdown of PFOS in samples with high pH, nitrate content, alkalinity, and anoxic conditions when exposed to eBeam technology at a dose of 50 kilograys (kGy). In lab-spiked water and sand, the concentrations of PFOA were reduced by 87% and 86% respectively. PFOS was reduced by 16% in lab-spiked water and 28% in lab-spiked sand. In spiked IDW groundwater exposed to 50 kGy, PFOA and PFOS were reduced by 98% and 41% respectively. Groundwater samples were collected from a monitoring well at Randolph AFB in San Antonio, TX, USA. Similarly, PFOA was reduced by 94% and PFOS by 41% in spiked post-UV wastewater effluent collected at the Texas A&M water treatment plant in College Station, TX, USA. Although the aforementioned conditions facilitated the complete breakdown of PFOA, more optimization and higher doses will be needed to achieve the same in PFOS.

Harmful algal and cyanobacterial blooms pose threats to human and ecological health due to their release of hazardous toxins. Microcystin-LR (MC-LR), a potent hepatotoxin, is the most prevalent cyanotoxin found in freshwater blooms. Although produced by many species of cyanobacteria, Microcystis aeruginosa is most commonly associated with MC-LR production. These blooms are increasing in occurrence in lakes, ponds, and other surface waters and, therefore, require efficient treatment methods to be removed from water supplies. Ionizing radiation technologies offer promising approaches for the removal of organic pollutants in water, including cyanotoxins and cyanobacteria. Gamma irradiation for the degradation of cyano-bacteria and toxins is effective for overall MC-LR degradation as well as reducing cell concentrations. However, gamma irradiation technology involves use of radioactive isotopes and, therefore, may not feasible commercially from a security perspective. Electron beam (eBeam) irradiation technology, which relies on regular electricity to generate highly energetic electrons, is able to achieve the same results without the confounding challenges of radioactive isotopes and related security issues. In this critical review, the current state of the science concerning the remediation of MC-LR and M. aeruginosa with ionizing radiation technologies is presented and future necessary research is discussed.
