The difference between the two competitions is in physical performance. The aim of the present study was to compare and investigate the relationship between physical characteristics and fitness between the two canoe sprint competitors. Methods: Subjects were 11 canoe paddlers (C) and 12 kayak paddlers (K). They underwent anthropometric characteristics, body composition and fitness tests, and 120 s all-out tests using a canoe and kayak ergometer. The unpaired t-test was used to test for significant differences between disciplines, while Pearson's correlation coefficient was used to examine the association between each measurement item and the performance test. Results: The age, height, body mass, body mass index, and total body fat were, C: 20.6±0.9 yr, 172.8±5.2 cm, 70.8±7.8 kg, 23.7±1.9, 14.4%±3.5%; and K: 19.7±1.2 yr, 172.8±5.3 cm, 69.5±7.8 kg, 23.2±2.1, 12.1%±3.6%, respectively. No significant differences were seen in any of the items for physical characteristics or fitness between C and K. A correlation analysis of performance tests and each measurement item revealed a positive correlation with low-speed isokinetic knee extension and flexion strength for C only (extension: r=0.761; flexion: r=0.784; P

We report the case of a 37-year-old woman who had a relapse of acute myeloid leukemia (AML) during treatment for chronic graft versus host disease (cGVHD) after allogeneic bone marrow transplantation. She was originally suspected of having autoimmune pancreatitis. Relapse of AML often occurs at extramedullary sites. Whereas the pancreas is rare as an organ of AML relapse, physicians should be aware that enlargement of the pancreas could be a sign of relapsed AML when excluding autoimmune pancreatitis, particularly during active cGVHD after allogeneic stem cell transplantation. In surgical removal of a malignant brain tumor, which is close to a pyramidal tract, diffusion tensor images obtained by preoperative magnetic resonance imaging (MRI) can display the spatial relationship between the tumor and the pyramidal tract. This image, however, is not appropriate to navigate the surgery because of brain shift due to surgical manipulation. We developed a navigation system employing intraoperative diffusion weighted imaging (DWI) using open MRI, which is the first trial with a 0.3 T scanner to aim at complete removal of cerebral tumors. A newly developed surgical coil has both functions of head fixation in surgery and a receiver coil in scan. The DWI of volunteers could display nerve fibers such as pyramidal tracts and corpus callosum under the optimized scan parameters. By refining navigation software, a physical position directed by the pointing device could be displayed correctly on three cross-sectional T1 (or T2) weighted images and three-dimensional (3D) DWI. Initial clinical evaluation of this system revealed that the spatial relationship between the tumor, the pyramidal tract, and the treatment position could be displayed correctly. Furthermore, the surgeons could recognize these positions intraoperatively and visually. The navigation system employing intraoperative DWI for a tumor and nerve fibers helped the surgeons remove the tumor and obviated postoperative disturbance such as motor paralysis. Background We aimed to investigate the association between protein intake and quadriceps quantity and quality on the paretic and non-paretic sides during rehabilitation in people with subacute stroke. Method Eighty-six people with stroke were recruited from a rehabilitation ward. We measured quadriceps muscle quantity and quality on the paretic and non-paretic sides using ultrasonography at admission and after 4weeks. Protein intake was assessed 2 or 3weeks after admission, and participants were classified into two groups: adequate and inadequate protein intake groups. Analysis of covariance was used to determine the effects of protein intake on quadriceps muscle quantity and quality. Results The mean age (standard deviation), median interval between stroke onset and admission (interquartile range) and male proportion of the study participants were 67.6 (13.5) years, 22.5 (16.8-31.3) days, and 54.7%, respectively. The adequate protein intake group showed significantly greater improvements in paretic-side quadriceps thickness than the inadequate group (group-by-time interaction, F =8.771, P =0.004). In contrast, no significant interactions were observed in quadriceps thickness on the non-paretic side (F =2.383, P =0.127) and quadriceps echo intensity on both sides (paretic-side: F =0.020, P =0.887, non-paretic side: F =0.181, P =0.672). Conclusions Adequate protein intake may be useful for improving quadriceps quantity on the paretic side in people with subacute stroke undergoing rehabilitation. However, quadriceps quantity on the non-paretic side and quadriceps quality on both sides were not significantly associated with adequate protein intake.

Background: The clinical importance of skeletal muscle characteristics for improving gait ability of stroke survivors is increasing. We aimed to examine the association between muscle quantity and quality at discharge and changes in gait independence at the time of 1 year after discharge in patients with stroke., Methods: This prospective observational study included 100 patients with stroke who were admitted to a convalescent rehabilitation ward. We defined muscle quantity and quality operationally as muscle thickness and echo intensity observed in ultrasonography images, respectively, and measured quadriceps muscle on the paretic and non-paretic sides at the time of discharge. The outcome measured in our study was changes in gait independence 1 year after discharge, as assessed by the Functional Independence Measure gait assessment tool score., Results: Among the study participants, 23 (23.0 %) were assessed to have reduced gait independence, while 77 (77.0 %) were evaluated to have improved or maintained gait independence. Our multivariate logistic regression analysis revealed that only muscle quantity on the paretic side was significantly associated with an improvement or maintenance of gait independence (odds ratios 3.32; 95 % confidence interval 1.01-10.95; p = 0.049)., Conclusions: Our findings revealed that an improvement in gait independence 1 year after discharge was influenced by quadriceps muscle quantity on the paretic side at the time of discharge in patients with subacute stroke. The correlation between early attrition and school sports clubs has been underexplored. This study aims to clarify the correlates of early attrition from school sports clubs at a private male senior high school in Fukuoka Prefecture, Japan. Of the 928 first-year students, 331 belonging to the school's sports clubs were included in this study. A baseline survey was conducted in May 2017 and a follow-up survey was conducted in October 2019. This study defined early attrition as a student who quit a sports club earlier than April in their third year. Examined correlates for early attrition consisted of biological, intrapersonal, interpersonal, and institutional factors. We used univariate logistic regression analysis, in which early attrition was the response variable and the examined correlates were the explanatory variables, and calculated the odds ratios (OR) and 95% confidence intervals (CI). Overall, 232 students (85.0%) continued to participate in sports clubs after April of their third year, and 41 students (15.0%) experienced early attrition. Statistically significant correlates for early attrition were weight (OR = 0.94, 95% CI = 0.90-0.98), body mass index (OR = 0.84, 95% CI = 0.74-0.97), experience of injury or disability (OR = 0.40, 95% CI = 0.19-0.87), athletic achievement (OR = 0.29, 95% CI = 0.13-0.62), and duration of experience of the sport (OR = 0.99, 95% CI = 0.98-1.00). Our findings suggest that the school officers or family members of students should recognize the possibility of early attrition and provide appropriate support if a student experiences early attrition. Background: Stroke is a disease that easily impairs skeletal muscle characteristics, resulting in low quantity, low quality, and poor strength. However, the recovery process of skeletal muscles in patients with stroke is not well established., Objectives: We aimed to determine the longitudinal changes in skeletal muscle characteristics in patients with stroke during intensive rehabilitation and examine their relationship with functional recovery., Methods: Eighty-five patients with stroke were prospectively recruited. We defined muscle quantity and quality as ultrasonographic muscle thickness and echo intensity, respectively, and muscle strength as knee extension strength. These skeletal muscle characteristics were compared at four time points: on admission, after 2 weeks, after 4 weeks, and at discharge. We also explored the relationship between changes in skeletal muscle characteristics and functional recovery., Results: Non-paretic knee extension torque (F = 5.838, P = .001), paretic quadriceps thickness (F = 2.943, P = .039), and paretic tibialis anterior thickness (F = 4.654, P = .004) improved over time, and changes in the paretic side quadriceps thickness were significantly associated with recovery for balance ability (odds ratio, 1.05; 95% confidence interval, 1.00-1.09; P = .048)., Conclusions: The present study revealed that knee extension muscle strength on the non-paretic side and quadriceps and tibialis anterior thickness on the paretic side significantly improved over time and that changes in the paretic side quadriceps thickness were associated with the recovery of balance ability.

Transcription factor EVI1 is essential for normal hematopoiesis in embryos but is aberrantly elevated in bone marrow cells of myelodysplastic syndrome (MDS) patients. EVI1 and its downstream GATA-2 appear to be a possible therapeutic target of MDS. Here we found that treatment of EVI1-expressing K562 cells with arsenite (As(III)) reduced the mRNA and protein levels of EVI1 and GATA-2. A gel shift assay using the nuclear extract of K562 cells showed that As(III) suppressed the DNA-binding activity of EVI1. The DNA-binding activity of the recombinant EVI1 protein was also suppressed by As(III) but was recovered by excess amounts of dithiothreitol, suggesting the involvement of cysteine residues of EVI1. Since the 7th Zn finger domain of EVI1, having a motif of CCHC, is known to be involved in DNA-binding, the synthetic peptide of 7th Zn finger domain was reacted with As(III) and subjected to MALDI-TOF-MS analysis. The results showed that As(III) binds to this peptide via three cysteine residues. As(III)-induced reduction of the DNA-binding activity of the recombinant EVI1 was abolished by the mutations of each of three cysteine residues to alanine in the 7th Zn finger domain. Otitis media, the most common disease of childhood, is characterized by extensive changes in the morphology of the middle ear cavity. This includes hyperplasia of the mucosa that lines the tympanic cavity, from a simple monolayer of squamous epithelium into a greatly thickened, respiratory-type mucosa. The processes that control this response, which is critical to otitis media pathogenesis and recovery, are incompletely understood. Given the central role of protein phosphorylation in most intracellular processes, including cell proliferation and differentiation, we screened a library of kinase inhibitors targeting members of all the major families in the kinome for their ability to influence the growth of middle ear mucosal explants in vitro. Of the 160 inhibitors, 30 were found to inhibit mucosal growth, while two inhibitors enhanced tissue proliferation. The results suggest that the regulation of infection-mediated tissue growth in the ME mucosa involves multiple cellular processes that span the kinome. While some of the pathways and processes identified have been previously implicated in mucosa hyperplasia others are novel. The results were used to generate a global model of growth regulation by kinase pathways. Like other sensory systems, olfactory function deteriorates with age. Epidemiological studies have revealed that the incidence of olfactory dysfunction increases at the age of 60 and older and males are more affected than females. Moreover, smoking, heavy alcohol use, sinonasal diseases, and Down's syndrome are associated with an increased incidence of olfactory dysfunction. Although the pathophysiology of olfactory dysfunction in humans remains largely unknown, studies in laboratory animals have demonstrated that both the peripheral and central olfactory nervous systems are affected by aging. Aged olfactory neuroepithelium in the nasal cavity shows the loss of mature olfactory neurons, replacement of olfactory neuroepithelium by respiratory epithelium, and a decrease in basal cell proliferation both in the normal state and after injury. In the central olfactory pathway, a decrease in the turnover of interneurons in the olfactory bulb (OB) and reduced activity in the olfactory cortex under olfactory stimulation is observed. Recently, the association between olfactory impairment and neurodegenerative diseases, such as Alzheimer's disease (AD) and Parkinson's disease (PD), has gained attention. Evidence-based pharmacotherapy to suppress or improve age-related olfactory dysfunction has not yet been established, but preliminary results suggest that olfactory training using odorants may be useful to improve some aspects of age-related olfactory impairment., (Copyright © 2020 Kondo, Kikuta, Ueha, Suzukawa and Yamasoba.)

The effects of caloric restriction (CR) on cell dynamics and gene expression in the mouse olfactory neuroepithelium are evaluated. Eight-week-old male C57BL/6 mice were fed either control pellets (104 kcal/week) or CR pellets (67 kcal/week). The cytoarchitecture of the olfactory neuroepithelium in the uninjured condition and its regeneration after injury by an olfactotoxic chemical, methimazole, were compared between mice fed with the control and CR diets. In the uninjured condition, there were significantly fewer olfactory marker protein (OMP)-positive olfactory receptor neurons and Ki67-positive proliferating basal cells at 3 months in the CR group than in the control group. The number of Ki67-positive basal cells increased after methimazole-induced mucosal injury in both the control and the CR groups, but the increase was less robust in the CR group. The recovery of the neuroepithelium at 2 months after methimazole administration was less complete in the CR group than in the control group. These histological changes were region-specific. The decrease in the OMP-positive neurons was prominent in the anterior region of the olfactory mucosa. Gene expression analysis using a DNA microarray and quantitative real-time polymerase chain reaction demonstrated that the expression levels of two inflammatory cytokines, interleukin-6 and chemokine ligand 1, were elevated in the olfactory mucosa of the CR group compared with the control group. These findings suggest that CR may be disadvantageous to the maintenance of the olfactory neuroepithelium, especially when it is injured.

Objectives: This open-label, multicenter, phase 1b/2 study assessed necitumumab plus gemcitabine and cisplatin (GC + N) in patients with previously untreated squamous non-small cell lung cancer in Japan., Materials and Methods: The phase 1b part determined the gemcitabine dose for the phase 2 part, in which patients were randomized 1:1 to GC + N or gemcitabine and cisplatin (GC) (gemcitabine 1250 mg/m 2 on days 1 and 8; cisplatin 75 mg/m 2 on day 1 of maximum four 3-week cycles; nectimumab 800 mg on days 1 and 8 of a 3-week cycle continued until progressive disease or unacceptable toxicity). The primary endpoint of the phase 2 part was overall survival., Results: In the phase 2 part, 181 patients received GC + N (N = 90) or GC (N = 91). Overall survival was significantly improved with GC + N versus GC (median, 14.9 months vs 10.8 months; hazard ratio [HR] = 0.66, 95% CI: 0.47 - 0.93, p = 0.0161). Improvements were also observed in progression-free survival (median, 4.2 months vs 4.0 months; HR = 0.56; p = 0.0004) and objective response rate (51% vs 21%; p < 0.0001). Survival was also significantly prolonged with GC + N versus GC for patients with epidermal growth factor receptor-positive tumors. Grade ≥3 treatment-emergent adverse events at ≥5% higher incidence with GC + N than GC were neutrophil count decreased (42% vs 35%), febrile neutropenia (12% vs 3%), decreased appetite (11% vs 4%), and dermatitis acneiform (6% vs 0%)., Conclusion: GC + N is well tolerated and has significant and clinically meaningful treatment benefit in the first-line treatment of patients with squamous non-small cell lung cancer in Japan.
