外部環境からの刺激（ストレス）を受けた多細胞生物の細胞は、周囲の細胞や細胞外液からのシグナルを伝搬・受容する仕組みを備えている。例えば、細胞にあらかじめ低線量放射線を照射した後、高線量の放射線を照射すると、事前照射を行わなかった場合と比較して放射線影響が緩和されることが知られており、一般に放射線適応応答と呼ばれている。この時細胞膜上のEGFなどのレセプターが外部からのシグナルを受容することで、細胞内へ情報が伝搬される。放射線適応応答の情報伝達の過程では、IP3が関与する複雑なCa2+放出反応ネットワークが形成されており、細胞内カルシウムストアである小胞体からCa2+放出を促すポジティブフィードバックを介した時間的な振動構造が脳・神経細胞で知られている。このような細胞内・細胞間の信号伝達には、情報伝達物質の周期的な放出と受容が利用されており、これは一種の波動を介した細胞間の情報伝達と定義される。シグナル伝達物質の時空間的な濃度変化（疎密波）は”細胞の声”とも言える事象であり、この細胞内・外の疎密波によるコミュニケーションの仕組みを明らかにすることが、本研究の目的である。脳・神経細胞の小胞体でCa2+波が確認されているCa2+イオンに着目し、細胞集団に対するCa2+波の蛍光ライブセルイメージング方法を検討した。今回は、ヒト正常繊維芽細胞（BJ-1 h-TERT）に対してCa2+に特異的な化学的蛍光プローブ（Fluo 4-AM）を導入し、蛍光顕微鏡を用いて細胞全体の輝度の時系列変化を約4分半撮影した（図1）。タイムラプス撮影の結果から、細胞内の相対的なCa2+濃度変化を算出した。観察結果より、隣接する細胞群の中には、Ca2+濃度変化のピーク強度や半値幅に細胞間の距離との相関があることが推測でき、これらの値が情報伝達の閾値として機能している可能性があると考えられる。情報伝達の起点となる細胞からの距離に比例/反比例して疎密波の波形が変化すると推測される。今後は細胞に放射線を照射した場合の細胞内Ca2+濃度変化を算出し、非照射群との比較・検討を行う。また、マイクロビームで細胞の部分照射を行い、細胞内外のCa疎密波による情報伝達過程の理解を目指す。, 令和 2 年度日本原子力学会北関東支部リモート若手研究者・技術者発表会

To react to stimuli from a circumstance, such as ionizing irradiation, living systems use various wave-like signals as communication tools. In this study, we focus on fluctuation of the Ca2+ concentration as a second messenger within cells exposed to an ionizing radiation. Normally, Ca2+ ions are stored in endoplasmic reticulum (ER) or mitochondria. In response to the environmental change, ER promptly discharges the ions resulting in a temporal periodic change of Ca2+ concentration in the cell. This oscillation is called Ca2+ wave and its propagation within the cells has been suggested to control various physiological functions. For example, glial cells in a neural system mutually communicate together by the electronical or the chemical waves. Our object is to clarify whether the Ca2+ waves arise when cells are exposed to an ionizing radiation to regulate their stress responses. We tried to establish an experimental method to visualize the Ca2+ concentration in the various mammalian cells. The cells were treated with a Ca2+ specific chemical fluorescent probe, Fluo 4. Time lapse observations were performed for the cells using a fluorescent microscope. In addition to background fluorescence, a progressive increase or a temporal burst-like Ca2+ concentration-change was seen in some of the cells exposed to X-rays. Waves of concentration change of specific signal molecules play an important role in signal exchanges between cells. To response to stimulus from outside of the cellular system they might communicate each other using waves. In this study, we focus on Ca2+ functioning as second messenger within cells. Normally, Ca2+ are stored in endoplasmic reticulum (ER). In response to the environmental stimulus, the Ca2+ channels on the ER membrane could be gated and induce a repetition of temporary ups and downs of the Ca2+ concentration in a cell. This “wave”, so-called Ca2+ concentration oscillation, has been predicted to regulate various physiological functions in cells. We investigate whether this temporal oscillation arises when cells are exposed to ionizing radiation. In this study, first, we tried to establish a measurement technique to visualize the Ca2+ concentration in mammalian cells. Normal human fibroblast cells were treated with a Ca2+ specific chemical fluorescent probe, Fluo 4. Then live-cell observations were performed with a fluorescent microscope to obtain time-lapse movies. Fluorescent intensities of the cells at each time point were converted to relative values to that at the beginning time of observation. Slight changes of the fluorescence in the most cells were recognized. Furthermore, some cells transiently showed intensive ups and downs of their fluorescence, indicating the occurrence of the Ca2+ waves. Using this method, for the next step, we challenge to detect local Ca2+ concentration oscillation in cell nucleus or organelle after exposure of ionizing radiation., 第3回QST国際シンポジウム「Quantum Life Science」

細胞間における情報伝達の過程において、シグナル伝達物質のバリエーションに依存するだけではなく、その物質の疎密波も情報の送受信に利用されている可能性を検討する。本研究では、細胞内でセカンドメッセンジャーとして機能するCa2+に着目し研究を行う。通常細胞内のCa2+は、Ca2+貯蔵部位の小胞体で保持されている。細胞外から刺激を受けると、細胞内Ca2+濃度は小胞体膜上のチャネルの開閉により上昇・下降を繰り返す。この現象はCa振動と呼ばれており、様々な生理現象に関与している。本研究では細胞間の情報伝達の前段階として、放射線刺激による細胞内Ca2+の時間的な振動構造が現れるかを検討した。まず、非照射細胞を用いてCa2+濃度の測定法を検討した。 今回は、ヒト正常繊維芽細胞に対してCa2+特異的な蛍光プローブを導入し、蛍光顕微鏡を用いてタイムラプス撮影を行った。撮影の結果から、観察開始時の蛍光強度を１とした時の相対蛍光強度を算出したところ、多くの細胞で時間経過に伴う若干の蛍光上昇が観察され、一部の細胞では一過性の急激なCa2+濃度の上昇と下降が見られた。 今後は放射線刺激による細胞内Ca2+濃度変化を観察し、細胞核やオルガネラなど、細胞局所でのCa2+濃度変化を合わせて検討する。, 第一回量子生命科学会若手の会
