BackgroundClimate change has recently boosted the severity and frequency of pine bark beetle attacks. The bacterial community associated with these beetles acts as “hidden players,” enhancing their ability to infest and thrive on defense-rich pine trees. There is limited understanding of the environmental acquisition of these hidden players and their life stage-specific association with different pine-feeding bark beetles. There is inadequate knowledge on novel bacterial introduction to pine trees after the beetle infestation. Hence, we conducted the first comparative bacterial metabarcoding study revealing the bacterial communities in the pine trees before and after beetle feeding and in different life stages of two dominant pine-feeding bark beetles, namely Ips sexdentatus and Ips acuminatus. We also evaluated the bacterial association between wild and lab-bred beetles to measure the deviation due to inhabiting a controlled environment.ResultsSignificant differences in bacterial amplicon sequence variance (ASVs) abundance existed among different life stages within and between the pine beetles. However, Pseudomonas, Serratia, Pseudoxanthomonas, Taibaiella, and Acinetobacter served as core bacteria. Interestingly, I. sexdentatus larvae correspond to significantly higher bacterial diversity and community richness and evenness compared to other developmental stages, while I. acuminatus adults displayed higher bacterial richness with no significant variation in the diversity and evenness between the life stages. Both wild and lab-bred I. sexdentatus beetles showed a prevalence of the bacterial family Pseudomonadaceae. In addition, wild I. sexdentatus showed dominance of Yersiniaceae, whereas Erwiniaceae was abundant in lab-bred beetles. Alternatively, Acidobacteriaceae, Corynebacteriaceae, and Microbacteriaceae were highly abundant bacterial families in lab-bred, whereas Chitinophagaceae and Microbacteriaceae were highly abundant in wild I. accuminatus. We validated the relative abundances of selected bacterial taxa estimated by metagenomic sequencing with quantitative PCR.ConclusionOur study sheds new insights into bacterial associations in pine beetles under the influence of various drivers such as environment, host, and life stages. We documented that lab-breeding considerably influences beetle bacterial community assembly. Furthermore, beetle feeding alters bacteriome at the microhabitat level. Nevertheless, our study revisited pine-feeding bark beetle symbiosis under the influence of different drivers and revealed intriguing insight into bacterial community assembly, facilitating future functional studies.

This paper focuses on the examination of the long-run relationship between money supply and selected national and global stock indices. Detailed knowledge of this relationship can be used by analysts, investors and monetary policy makers. Analysis of the relationship was performed using a 2-stage Engle-Granger cointegration. First, the stationarity of the time series was tested, then both the long-term OLS model and the short-term EC model were estimated. Time series were always tested on the longest period for which data were available. The longterm dependence of stock indices on the respective M2 was confirmed for the BOVESPA, FTSE100, S&P/BMV IPC, S&P BSE500, TSX and The 5000 Wilshire Small Cap Price Return indices. In contrast, the dependence between world money supply indicator GlobalM2, the stock index FTSEALL World, and the S&P500 index was not demonstrated. Additionally, no dependence was identified between the respective M2 and the DAX, PX, Nikkei225, KOSPI, SMI, SPCITIC300, Eurostoxx50, Willshire5000PR and ATX indices. Backward dependence of M2 on the stock index was found only for the Chinese SPCITIC300 index.

IntroductionIn recent years, Norway spruce (Picea abies L.) forests in Central Europe have faced escalating threats from bark beetles, primarily Ips typographus (L.), and other species, such as I. duplicatus (Sahlberg). Outbreaks are partially attributed to weakened tree defense resulting from drought periods induced by climate change. This study examines Norway spruce’s physiological and metabolic reactions to acute drought stress during the growing season and evaluates its susceptibility to I. typographus and I. duplicatus.MethodologyIn order to induce drought stress, mature Norway spruces had their roots covered with a roof in April 2021, depriving them of water. Control trees were left with free access to natural rainwater. Over 5 months of the growing season, soil water potential, bark temperature, tree trunk circumference, and sap flow were monitored. Roofed trees and controls were sampled in July, August, and September and analyzed for non-structural carbohydrates and the two classes of defensive compounds, phenolics and terpenes. Furthermore, two different bioassays in tubes and boxes were performed using adult I. typographus and I. duplicatus beetles to assess host choice and acceptance.ResultsRoofed trees exhibited signs of stress as early as July, resulting in decreased tree trunk and a consequent increase in non-structural carbohydrate content. Defensive metabolites remained largely unaffected except for an increase in diterpenes in September. In bioassays, I. typographus preferred boring into the bark of roofed trees in August in tubes and in September in boxes. This increased tree acceptance correlated with increased levels of soluble carbohydrates in the phloem. I. typographus and I. duplicatus beetles showed higher mobility in boxes in August and September on roofed trees but not in July, even though bark surface temperatures were elevated in roof-covered trees during all three bioassay periods.ConclusionThe study revealed rapid physiological responses of trees to acute drought stress, although not many changes were observed in defense traits. Despite the absence of natural bark beetle attacks, drought trees were more accepted by I. typographus than naturally watered trees. This response may indicate the beetles’ preference for trees with phloem of higher nutritional quality induced by the acute drought stress conditions.

Home is not only a concrete place and a complex system of relations, but also an end-in-view that connects perceived shortcomings in the current versions of home with the desired goals and the means to achieve them. Our case study centres on a dining improvement project which strives to create home in residential institutions for people identified as disabled by serving a 'home-like meal'. We describe three versions of home that are enacted in residential institutions – home as a commune, home as a private space and home as an intimate sphere – and document how they influence the serving of meals. We combine pragmatic theory of valuation with ethnographic research of home-making practices to assess the feasibility of these ends-in-view in relation to the housing options available to the disabled-identified. We show that when the realization of the chosen goals proves unfeasible under present circumstances, the discrepancies between the desired and actual versions of home can be effaced through various re-contextualization strategies. Of the three versions of home encountered during our intervention/research, the home enacted as an intimate sphere is the one most firmly grounded in clients' real wishes and needs, and therefore the one most favourable to positive change. Abstract Bone is a remarkable, living tissue that functionally adapts to external loading. Therefore, bone shape and internal structure carry information relevant to many disciplines, including medicine, forensic science, and anthropology. However, morphometric comparisons of homologous regions across different individuals or groups are still challenging. In this study, two methods were combined to quantify such differences: (1) Holistic morphometric analysis (HMA) was used to quantify morphometric values in each bone, (2) which could then be mapped to a volumetric mesh of a canonical bone created by a statistical free-form deformation model (SDM). Required parameters for this canonical holistic morphometric analysis (cHMA) method were identified and the robustness of the method was evaluated. The robustness studies showed that the SDM converged after one to two iterations, had only a marginal bias towards the chosen starting image, and could handle large shape differences seen in bones of different species. Case studies were performed on metacarpal bones and proximal femora of different primate species to confirm prior study results. The differences between species could be visualised and statistically analysed in both case studies. cHMA provides a framework for performing quantitative comparisons of different morphometric quantities across individuals or groups. These comparisons facilitate investigation of the relationship between spatial morphometric variations and function or pathology, or both.

Abstract Eurasian spruce bark beetle (Ips typographus [L.]) causes substantial damage to spruce forests worldwide. Undoubtedly, more aggressive measures are necessary to restrict the enduring loss. Finishing genome sequencing is a landmark achievement for deploying molecular techniques (i.e., RNA interference) to manage this pest. Gene expression studies assist in understanding insect physiology and deployment of molecular approaches for pest management. RT-qPCR is a valuable technique for such studies. However, accuracy and reliability depend on suitable reference genes. With the genome sequence available and the growing requirement of molecular tools for aggressive forest pest management, it is crucial to find suitable reference genes in Ips typographus under different experimental conditions. Hence, we evaluated the stability of twelve candidate reference genes under diverse experimental conditions such as biotic (developmental, sex and tissues) and abiotic factors (i.e., temperature and juvenile hormone treatment) to identify the reference genes. Our results revealed that ribosomal protein 3a (RPS3-a) was the best reference gene across all the experimental conditions, with minor exceptions. However, the stability of the reference gene can differ based on experiments. Nevertheless, present study provides a comprehensive list of reference genes under different experimental conditions for Ips typographus and contributes to “future genomic and functional genomic research”.

BMC values from dCBCT were higher than DXA measurements (2.34 g vs. 1.5 g, p < 0.001). Cortical thickness strongly correlated to bone mineral density (BMD) from dCBCT (r = 0.83, p < 0.001). No statistically significant correlations were found between trabecular bone microstructure and FE predictions. The results indicate the feasibility to analyse osteoporosis related bone properties of human wrists from corrected dCBCT data. The dCBCT values of BMD and BMC were strongly correlated with DXA. The "gold standard" for the assessment of trabecular bone structure is high‐resolution micro‐CT. In this technical note, we test the influence of initial scan resolution and post hoc downsampling on the quantitative and qualitative analysis of trabecular bone in a Gorilla tibia. We analyzed trabecular morphology in the right distal tibia of one Gorilla gorilla individual to investigate the impact of variation in voxel size on measured trabecular variables. For each version of the micro‐CT volume, trabecular bone was segmented using the medical image analysis method. Holistic morphometric analysis was then used to analyze bone volume (BV/TV), anisotropy (DA), trabecular thickness (Tb.Th), spacing (Tb.Sp), and number (Tb.N). Increasing voxel size during initial scanning was found to have a strong impact on DA and Tb.Th measures, while BV/TV, Tb.Sp, and Tb.N were found to be less sensitive to variations in initial scan resolution. All tested parameters were not substantially influenced by downsampling up to 90 μm resolution. Color maps of BV/TV and DA also retained their distribution up to 90 μm. This study is the first to examine the effect of variation in micro‐CT voxel size on the analysis of trabecular bone structure using whole epiphysis approaches. Our results indicate that microstructural variables may be measured for most trabecular parameters up to a voxel size of 90 μm for both scan and downsampled resolutions. Moreover, if only BV/TV, Tb.Sp or Tb.N is measured, even larger voxel sizes might be used without substantially affecting the results. [ABSTRACT FROM AUTHOR]

The exocyst complex regulates the last steps of exocytosis, which is essential to organisms across kingdoms. In humans, its dysfunction is correlated with several significant diseases, such as diabetes and cancer progression. Investigation of the dynamic regulation of the evolutionarily conserved exocyst-related processes using mutants in genetically tractable organisms such as Arabidopsis thaliana is limited by the lethality or the severity of phenotypes. We discovered that the small molecule Endosidin2 (ES2) binds to the EXO70 (exocyst component of 70 kDa) subunit of the exocyst complex, resulting in inhibition of exocytosis and endosomal recycling in both plant and human cells and enhancement of plant vacuolar trafficking. An EXO70 protein with a C-terminal truncation results in dominant ES2 resistance, uncovering possible distinct regulatory roles for the N terminus of the protein. This study not only provides a valuable tool in studying exocytosis regulation but also offers a potentially new target for drugs aimed at addressing human disease.

Ips sexdentatus (Coleoptera: Curculionidae: Scolytinae) is one of the most destructive and economically important forest pests. A better understanding of molecular mechanisms underlying its adaptation to toxic host compounds may unleash the potential for future management of this pest. Gene expression studies could be considered as one of the key experimental approaches for such purposes. A suitable reference gene selection is fundamental for quantitative gene expression analysis and functional genomics studies in I. sexdentatus. Twelve commonly used reference genes in Coleopterans were screened under different experimental conditions to obtain accurate and reliable normalization of gene expression data. The majority of the 12 reference genes showed a relatively stable expression pattern among developmental stages, tissue-specific, and sex-specific stages; however, some variabilities were observed during varied temperature incubation. Under developmental conditions, the Tubulin beta-1 chain (β-Tubulin) was the most stable reference gene, followed by translation elongation factor (eEF2) and ribosomal protein S3 (RPS3). In sex-specific conditions, RPS3, β-Tubulin, and eEF2 were the most stable reference genes. In contrast, different sets of genes were shown higher stability in terms of expression under tissue-specific conditions, i.e., RPS3 and eEF2 in head tissue, V-ATPase-A and eEF2 in the fat body, V-ATPase-A and eEF2 in the gut. Under varied temperatures, β-Tubulin and V-ATPase-A were most stable, whereas ubiquitin (UbiQ) and V-ATPase-A displayed the highest expression stability after Juvenile Hormone III treatment. The findings were validated further using real-time quantitative reverse transcription PCR (RT-qPCR)-based target gene expression analysis. Nevertheless, the present study delivers a catalog of reference genes under varied experimental conditions for the coleopteran forest pest I. sexdentatus and paves the way for future gene expression and functional genomic studies on this species.

The “Different Hearing” program (DHP) is an educational activity aimed at stimulating musical creativity of children and adults by group composing in the classroom, alternative to the mainstream model of music education in Czechia. Composing in the classroom in the DHP context does not use traditional musical instruments or notation, instead, the participants use their bodies, sounds originating from common objects as well as environmental sounds as the “elements” for music composition by the participants’ team, with the teacher initiating and then participating and coordinating the creative process, which ends with writing down a graphical score and then performing the composition in front of an audience. The DHP methodology works with a wide definition of musical composition. We hypothesized that the DHP short-term (2 days) intense workshop would induce changes in subjective appreciation of different classes of music and sound (including typical samples of music composed in the DHP course), as well as plastic changes of the brain systems engaged in creative thinking and music perception, in their response to diverse auditory stimuli. In our study, 22 healthy university students participated in the workshop over 2 days and underwent fMRI examinations before and after the workshop, meanwhile 24 students were also scanned twice as a control group. During fMRI, each subject was listening to musical and non-musical sound samples, indicating their esthetic impression with a button press after each sample. As a result, participants’ favorable feelings toward non-musical sound samples were significantly increased only in the active group. fMRI data analyzed using ANOVA with post hoc ROI analysis showed significant group-by-time interaction (opposing trends in the two groups) in the bilateral posterior cingulate cortex/precuneus, which are functional hubs of the default mode network (DMN) and in parts of the executive, motor, and auditory networks. The findings suggest that DHP training modified the behavioral and brain response to diverse sound samples, differentially changing the engagement of functional networks known to be related to creative thinking, namely, increasing DMN activation and decreasing activation of the executive network.

Plant defence mechanisms, including physical barriers like toughened bark and chemical defences like allelochemicals, are essential for protecting them against pests. Trees allocate non-structural carbohydrates (NSCs) to produce secondary metabolites like monoterpenes, which increase during biotic stress to fend off pests like the Eurasian spruce bark beetle, ESBB (Ips typographus). Despite these defences, the ESBB infests Norway spruce, causing significant ecological damage by exploiting weakened trees and using pheromones for aggregation. However, the mechanism of sensing and resistance towards host allelochemicals in ESBB is poorly understood. We hypothesised that the exposure of ESBB to spruce allelochemicals, especially monoterpenes, leads to an upsurge in the important detoxification genes like P450s, GSTs, UGTs, and transporters, and at the same time, genes responsible for development must be compromised. The current study demonstrates that exposure to monoterpenes like R-limonene and sabiene effectively elevated detoxification enzyme activities. The differential gene expression (DGE) analysis revealed 294 differentially expressed (DE) detoxification genes in response to R-limonene and 426 DE detoxification genes in response to sabiene treatments, with 209 common genes between the treatments. Amongst these, genes from the cytochrome P450 family 4 and 6 genes (CP4 and CP6), esterases, glutathione S-transferases family 1 (GSTT1), UDP-glucuronosyltransferase 2B genes (UDB), and glucose synthesis-related dehydrogenases were highly upregulated. We further validated 19 genes using RT-qPCR. Additionally, we observed similar high expression levels of detoxification genes across different monoterpene treatments, including myrcene and α-pinene, suggesting a conserved detoxification mechanism in ESBB, which demands further investigation.
