2. Theory of superlocalized magnetic nanoparticle hyperthermia: rotating versus oscillating fields

Abstract

The main idea of magnetic hyperthermia is to increase locally the temperature of the human body by means of injected superparamagnetic nanoparticles. They absorb energy from a time-dependent external magnetic field and transfer it into their environment. In the so-called superlocalization, the combination of an applied oscillating and a static magnetic field gradient provides even more focused heating since for large enough static field the dissipation is considerably reduced. Similar effect was found in the deterministic study of the rotating field combined with a static field gradient. Here we study theoretically the influence of thermal effects on superlocalization and on heating efficiency. We demonstrate that when time-dependent steady state motions of the magnetisation vector are present in the zero temperature limit, then deterministic and stochastic results are very similar to each other. We also show that when steady state motions are absent, the superlocalization is severely reduced by thermal effects. Our most important finding is that in the low frequency range ($\omega \to 0$) suitable for hyperthermia, the oscillating applied field is shown to result in two times larger intrinsic loss power and specific absorption rate then the rotating one with identical superlocalization ability which has importance in technical realisation., Comment: 20 pages, 18 figures, published in J. Abstract

Single-domain ferromagnetic nanoparticle systems can be used to transfer energy from a time-dependent magnetic field into their environment. This local heat generation, i.e., magnetic hyperthermia, receives applications in cancer therapy which requires the enhancement of the energy loss. A possible way to improve the efficiency is to chose a proper type of applied field, e.g., a rotating instead of an oscillating one. The latter case is very well studied and there is an increasing interest in the literature to investigate the former although it is still unclear under which circumstances the rotating applied field can be more favourable than the oscillating one. The goal of this work is to incorporate the presence of a static field and to perform a systematic study of the non-linear dynamics of the magnetisation in the framework of the deterministic Landau-Lifshitz-Gilbert equation in order to calculate energy losses. Two cases are considered: the static field is either assumed to be perpendicular to the plane of rotation or situated in the plane of rotation. In the latter case a significant increase in the energy loss/cycle is observed if the magnitudes of the static and the rotating fields have a certain ratio (e.g. it should be one for isotropic nanoparticles). Abstract

The deterministic Landau-Lifshitz-Gilbert equation has been used to investigate the nonlinear dynamics of magnetization and the specific loss power in magnetic nanoparticles with uniaxial anisotropy driven by a rotating magnetic field. We propose a new type of applied field, which is "simultaneously rotating and alternating", i.e. the direction of the rotating external field changes periodically. We show that a more efficient heat generation by magnetic nanoparticles is possible with this new type of applied field and we suggest its possible experimental realization in cancer therapy which requires the enhancement of loss energies., Comment: 7 pages, 4 figures, to be published in Phys. Abstract

Motivated by a recent experimental observation of a complex magnetic structure [Takada et al. 2013 J. Magn. Magn. Mater. 329 95] we present a theoretical study of the magnetic structure of an Fe monolayer deposited on Rh(001). We use a classical spin Hamiltonian with parameters obtained from ab initio calculations and go beyond the usual anisotropic Heisenberg model by including isotropic biquadratic interactions. Zero-temperature Landau--Lifshitz--Gilbert spin dynamics simulations lead to a complex collinear spin configuration that, however, contradicts experimental finding. We thus conclude that higher order multi-spin interactions are likely needed to account for the magnetic ordering of the system., Comment: 10 pages, 3 figures

Abstract

Gastric mucosal microvessels were isolated after arterial perfusion of the rat stomach with magnetized iron oxide suspension. After homogenization of scrapped gastric mucosa, microvessels were initially separated with a high power magnet and further separated and purified by using a nylon sieve. Aliquots of purified microvessels were assessed for viability, histologic appearance, ultrastructure and generation of prostacyclin. Microvessels were plated on Matrigel and cultured in DMEM with high glucose and 10% FBS for 1, 3 or 5 days. After 1, 3 and 5 days of culturing, endothelial viability was assessed with Fast green exclusion, and the basal and stimulated (with calcium ionophore) generation of prostacyclin was determined by assaying aliquots of the incubating medium for 6-keto PGF(1alpha). At 1 and 3 hrs after isolation, microvessels demonstrated intact morphologic structures as reflected by transmission EM and 92+/-4% of viable endothelial cells. The microvessels plated on Matrigel maintained good viability for at least 5 days and generated prostacyclin at the baseline and following ionophore stimulation. These data demonstrate that isolated microvessels cultured under optimal conditions are fully viable and functional.

This study aims to compare the phenotype, management, and outcome of familial DCM (FDCM) and non‐familial (sporadic) DCM (SDCM) across Europe.Methods and resultsPatients with DCM that were enrolled in the prospective ESC EORP Cardiomyopathy & Myocarditis Registry were included. Baseline characteristics, genetic testing, genetic yield, and outcome were analysed comparing FDCM and SDCM; 1260 adult patients were studied (238 FDCM, 707 SDCM, and 315 not disclosed). Patients with FDCM were younger (P < 0.01), had less severe disease phenotype at presentation (P < 0.02), more favourable baseline cardiovascular risk profiles (P ≤ 0.007), and less medication use (P ≤ 0.042). Outcome at 1 year was similar and predicted by NYHA class (HR 0.45; 95% CI [0.25–0.81]) and LVEF per % decrease (HR 1.05; 95% CI [1.02–1.08]. Throughout Europe, patients with FDCM received more genetic testing (47% vs. 8%, P < 0.01) and had higher genetic yield (55% vs. 22%, P < 0.01).ConclusionsWe observed that FDCM and SDCM have significant differences at baseline but similar short‐term prognosis. Whether modification of associated cardiovascular risk factors provide opportunities for treatment remains to be investigated. Our results also show a prevalent role of genetics in FDCM and a non‐marginal yield in SDCM although genetic testing is largely neglected in SDCM. Limited genetic testing and heterogeneity in panels provides a scaffold for improvement of guideline adherence.

Abstract

Background & Aims: Celiac disease (CeD), an immune-mediated disease with enteropathy triggered by gluten, affects ~1% of the general European population. Currently, there are no biomarkers to predict CeD development. MicroRNAs (miRNAs) are short RNAs involved in post-transcriptional gene regulation, and certain disease- and stage-specific miRNA profiles have been found previously. We aimed to investigate whether circulating miRNAs can predict the development of CeD. Methods: Using next-generation miRNA-sequencing, we determined miRNAs in >200 serum samples from 53 participants of the PreventCD study, of whom 33 developed CeD during follow-up. Following study inclusion at 3 months of age, samples were drawn at predefined ages, diagnosis (first anti-transglutaminase antibody (TGA) positivity or diagnostic biopsy) and after the start of a gluten-free diet (GFD). This allowed identification of circulating miRNAs that are deregulated before TGA positivity. For validation of the biomarkers for CeD and GFD response, two additional cohorts were included in subsequent meta-analyses. Additionally, miRNAs were measured in duodenal biopsies in a case-control cohort. Results: 53 circulating miRNAs were increased (27) or decreased (26) in CeD versus controls. We assessed specific trends in these individual miRNAs in the PreventCD cohort by grouping the pre-diagnostic samples of the CeD patients (all had negative TGA) by how close to seroconversion (first sample positive TGA) the samples were taken. 8/53 miRNAs differed significantly between controls and samples taken <1 year before TGA positivity: miR-21-3p, miR-374a-5p, 144-3p, miR-500a-3p, miR-486-3p let-7d-3p, let-7e-5p and miR-3605-3p. 6/26 downregulated miRNAs reconstituted upon GFD, including miR-150-5p/-3p, whereas no upregulated miRNAs were downregulated upon GFD. 15/53 biomarker candidates also differed between CeD biopsies and controls, with a concordant direction, indicating that these circulating miRNAs migh

Abstract

BACKGROUND AND AIMS: Screening for celiac disease (CD) is recommended in children with affected first-degree relatives (FDR). However, the frequency of screening and at what age remain unknown. The aims of this study were to detect variables influencing the risk of CD development and develop and validate clinical prediction models to provide individualized screening advice. METHODS: We analyzed prospective data from the 10 years of follow-up of the PreventCD-birth cohort involving 944 genetically predisposed children with CD-FDR. Variables significantly influencing the CD risk were combined to determine a risk score. Landmark analyses were performed at different ages. Prediction models were created using multivariable Cox proportional hazards regression analyses, backward elimination, and Harrell's c-index for discrimination. Validation was done using data from the independent NeoCel cohort. RESULTS: In March 2019, the median follow-up was 8.3 years (22 days-12.0 years); 135/944 children developed CD (mean age, 4.3 years [range, 1.1-11.4]). CD developed significantly more often in girls (P = .005) and in Human Leukocyte Antigen (HLA)-DQ2 homozygous individuals (8-year cumulative incidence rate of 35.4% vs maximum of the other HLA-risk groups 18.2% [P < .001]). The effect of homozygosity DR3-DQ2/DR7-DQ2 on CD development was only present in girls (interaction P = .04). The prediction models showed good fit in the validation cohort (Cox regression 0.81 [0.54]). To calculate a personalized risk of CD development and provide screening advice, we designed the Prediction application https://hputter.shinyapps.io/preventcd/. CONCLUSION: Children with CD-FDR develop CD early in life, and their risk depends on gender, age and HLA-DQ, which are all factors that are important for sound screening advice. These children should be screened early in life, including HLA-DQ2/8-typing, and if genetically predisposed to CD, they should get further personalized screening advice using our Prediction application. Abstract

Exercise Doppler echocardiography (EDE) is a well-validated tool in ischemic and valvular heart diseases. However, its use in the assessment of the right heart and pulmonary circulation unit (RH-PCU) is limited. The aim of this study is to assess the semi-recumbent bicycle EDE feasibility for the evaluation of RH-PCU in a large multi-center population, from healthy individuals and elite athletes to patients with overt or at risk of developing pulmonary hypertension (PH). From January 2019 to July 2019, 954 subjects [mean age 54.2 ± 16.4years, range 16–96, 430 women] underwent standardized semi-recumbent bicycle EDE with an incremental workload of 25 watts every 2min, were prospectively enrolled among 7 centers participating to the RIGHT Heart International NETwork (RIGHT-NET). EDE parameters of right heart structure, function and pressures were obtained according to current recommendations. Right ventricular (RV) function at peak exercise was feasible in 903/940 (96%) by tricuspid annular plane systolic excursion (TAPSE), 667/751 (89%) by tissue Doppler-derived tricuspid lateral annular systolic velocity (S′) and 445/672 (66.2%) by right ventricular fractional area change (RVFAC). RV—right atrial pressure gradient [RV–RA gradient = 4 × tricuspid regurgitation velocity2 (TRV)] was feasible in 894/954 patients (93.7%) at rest and in 816/954 (85.5%) at peak exercise. The feasibility rate in estimating pulmonary artery pressure improved to more than 95%, if both TRV and/or right ventricular outflow tract acceleration time (RVOT AcT) were considered. In high specialized echocardiography laboratories semi-recumbent bicycle EDE is a feasible tool for the assessment of the RH-PCU pressure and function.

Abstract

Conventional fuels for vehicles, which are non-renewable energy sources, are still currently in use. To reduce noise and CO2 emissions, electric vehicles dictate a remarkable research and developments in order to reach a viable solution. Electrified vehicles have a dedicated energy store, usually batteries, to improve emissions or replace the conventional energy source with electric power. For almost a decade now, the use of Li-ion batteries as a source of energy for electric vehicles has exploded, but there are also many questions about their safety. This study is dedicated to compare the results obtained by experiment with the result obtained by simulation about mechanical behavior of an 18650 Li-ion cell under pinch indentation. Abstract

Lithium-ion batteries (LIBs) are an important component of the automotive industry in terms of electrification. Regarding their safety, the researchers perform different scenarios in which they follow closely and carefully their behavior under the action of induced requests. The most common mechanical abuse tests are three-point bending, simple bending, deformation with hemispherical punch head and nail penetration. In this article, the behavior of the 21700 lithiumion cell is debated and the results are compared. First, experimental tests are performed and the results are validated with FEM model, to check the veracity of the experiment.
