1. HAPLOINSUFFICIENCY OF TIE2 IN MUTATED BLOOD CELLS SUPPRESS ANGIOGENESIS IN THE BONE MARROW AND INHIBIT PROGRESSION OF MDS

Abstract

Introduction: Tie2 is a receptor tyrosine kinase and regulates angiogenesis and vascular quiescence. Given that Tie2 modulates microvascular density in cancer, we hypothesized that deletion of Tie2 in blood cells can inhibit progression of myelodysplastic syndrome (MDS). We attempted to understand the role of Tie2 in development of MDS by using an Ezh2/Tet2 double knock out (DKO) mouse model. Methods: We transplanted bone marrow (BM) cells isolated from Cre-ERT2 mice, Tie2flox/wt; Cre-ERT2 mice, Ezh2flox/flox; Tet2flox/flox; Cre-ERT2 mice, Ezh2flox/flox; Tet2flox/flox; Tie2flox/wt; Cre-ERT2 mice and Ezh2flox/flox; Tet2flox/flox; Tie2flox/flox; Cre-ERT2 mice into lethally-irradiated Ly5.1+ recipient mice. Ezh2, Tet2 and Tie2 genes were deleted by administration of tamoxifen one month post the transplantation. Results: We found that Ezh2−/−Tet2−/- DKO, Ezh2−/−Tet2−/- Tie2+/− (DKOTie2+/−) and Ezh2−/−Tet2−/- Tie2−/− TKO mice all developed MDS and MDS/MPN, showing anemia and dysplastic cells in the peripheral blood (PB) and the BM; however, DKOTie2+/− mice showed significantly longer survival than did DKO mice and TKO mice. While DKO mice showed deformed CD31+ endothelial cells and increased vascular density in the BM, DKOTie2+/− mice mitigated the altered vascular formation in the BM. RNA-sequencing revealed that DKOTie2+/− stem cells repressed expression of genes involved in interferon, cell cycles and angiogenesis, compared to DKO stem cells, suggesting that the haploinsufficiency of Tie2 impaired the property of MDS cells to drive angiogenesis in the BM, resulting in the delayed development of MDS. Conclusions: We are now working on the molecular mechanism of how the Tie2 gene in blood cells modulates the angiogenesis to drive the progression of MDS.

Abstract

Personal Feedback Interventions (PFIs) have been widely used to reduce the amount of time and money individuals spend on gambling. A central component of these interventions is personalized information about an individual’s gambling behavior, often in comparison to others’ gambling. The purpose of the present review and meta-analysis was to evaluate these interventions in terms of content, mode of delivery, target sample, and efficacy. Sixteen interventions from 11 studies were reviewed. We found a small, statistically significant effect in favor of PFIs versus control (d = 0.20, 95% CI 0.12, 0.27). Six moderators of intervention efficacy were explored. These interventions appeared to be most efficacious when used in populations of greater gambling severity, when individuals were provided with gambling-related educational information, and when used in conjunction with motivational interviewing. Factors associated with reduced efficacy include in-person delivery of feedback without motivational-interviewing and informing participants of their score on a psychological measure of gambling severity. Efficacy did not vary as a function of college or community samples. PFIs are a low cost, easily disseminated intervention that can be used as a harm-reduction strategy. However, more substantial effects may be attained if used as part of a larger course of therapy.

Abstract

BackgroundThe number of patients (pts) with rheumatoid arthritis (RA) is highest in persons in their late 60s, and the age at onset has increased in recent years. In many countries, including Japan, ‘elderly’ is defined as 65 years or older, but recently, the Japan Geriatrics Society and Japan Gerontological Society have advocated a newer classification of elderly: pre-old as 65 to 74 years, old as 75-89 years and super old as ≥90 years1). Disease activities, renal functions, treatments and other parameters in thus classified elderly RA pts have not been investigated yet.ObjectivesElderly pts with RA were classified, and then disease activities, treatments, renal functions and other parameters compared between the groups to clarify the real-world picture of pre-old and old RA pts.MethodsWe enrolled 359 RA patients (85 males, mean age 65.0 ± 13.6 years) treated at our hospital and affiliated hospitals. They were divided into three groups: under 65 years as a control group, 65-74 years as a pre-old (group I), and 75 years or older as an old (group II). Urine examination, serum Cr, eGFR, BUN, other biochemical tests, serological tests, disease activity, treatment, smoking, and comorbidities were evaluated. Difficult-to-treat RA pts (D2T RA) were evaluated according to EULAR criteria2).ResultsGroup C contained 148 pts (28 males, mean age 51.7 ± 9.6 years), Group I 123 pts (28 males, mean age 70.1 ± 2.9 years) and Group II 88 pts (29 males, average age 80.2 ± 4.5 years). Among the three groups, the frequency of males was 33.0% in group II, which was significantly higher than in the other groups. There was no significant difference in disease activity among the three groups. There was a significant difference in eGFR among the three groups (77.3 ± 17.3 ml/min. in group C, 69.4 ± 15.8 ml/min. in group I, 62.7 ± 18.7 ml/min. in group II). There was a significant difference in frequencies of treatment with NSAIDs (40/133 (30.0%) in group C, 23/102 (22.5%) in group I, 9/67 (13.4%) in group II), methotrexate (MTX) (87/148 (58.8%) in Group C, 62/123 (50.4%) in Group I, 28/88 (31.4%) in Group II). There was no significant difference in frequencies of treatment with prednisolone (55 / 148 (30.2%) in group C, 55/123 (43.1%) in Group I, 45/88 (51.1%) in Group II), with biologics (44/148 (29.7%) in Group C, 29/123 (23.6%) in Group I, 26/88 (29.5%) in Group II) and with JAK inhibitor (15/148 (10.1%) in Group C, 7/123 (5.7%) in Group I, 5/88 (5.7%) in Group II). There was no significant difference in D2TRA pts among the three groups (21/148 (14.2%) in Group C, 12/123 (9.8%) in Group I, 13/88 (14.8%) in Group II). There were significant differences in the frequency of stage IV and comorbidities among the three groups.ConclusionIn the pre-old and old RA pts, renal function was lower than in the control group. MTX was administered even in them, but less frequently. We were able to clarify the current situation that JAK inhibitors and biologics are actively used even in pre-old and old RA pts with impaired renal function, and the frequency of D2T RA pts could be suppressed to almost that in the control group. Abstract

Sarcoidosis affects multiple organs and rarely has unusual manifestations. A 78-year-old woman was referred to our hospital for coughing symptoms. A chest computed tomography (CT) scan revealed bilateral diffuse miliary patterns and right pleural effusion. Bronchoscopy showed multiple nodules in the carina and the bronchus intermedius. A CT scan of her abdomen revealed hypovascular lesions involving the pancreatic head and body. A transbronchial lung biopsy, bronchial mucosal biopsy, and endoscopic ultrasound-guided fine-needle aspiration of the pancreatic mass demonstrated non-caseating granulomas. We diagnosed the patient with sarcoidosis. She received no treatment for sarcoidosis and has been followed up for one year, during which no pulmonary disease progression had been observed and the pancreatic masses partially regressed.

Abstract

Personal Feedback Interventions (PFIs) have been widely used to reduce the amount of time and money individuals spend on gambling. A central component of these interventions is personalized information about an individual's gambling behavior, often in comparison to others' gambling. The purpose of the present review and meta-analysis was to evaluate these interventions in terms of content, mode of delivery, target sample, and efficacy. Sixteen interventions from 11 studies were reviewed. We found a small, statistically significant effect in favor of PFIs versus control (d = 0.20, 95% CI 0.12, 0.27). Six moderators of intervention efficacy were explored. These interventions appeared to be most efficacious when used in populations of greater gambling severity, when individuals were provided with gambling-related educational information, and when used in conjunction with motivational interviewing. Factors associated with reduced efficacy include in-person delivery of feedback without motivational-interviewing and informing participants of their score on a psychological measure of gambling severity. Efficacy did not vary as a function of college or community samples. PFIs are a low cost, easily disseminated intervention that can be used as a harm-reduction strategy. However, more substantial effects may be attained if used as part of a larger course of therapy.

Social influence affects college students’ gambling behaviours. However, few studies have experimentally investigated the influence of social interaction on college students’ gambling behaviour, and those studies that have yielded mixed findings. Women college students (n = 109) who endorsed recreational gambling behaviour were randomly assigned to gamble on electronic gambling machines (EGMs) in three conditions: warm social interaction from a confederate (i.e., initiating and maintaining conversation), cold social interaction from a confederate (i.e., refraining from initiating and maintaining conversation), or gambling alone. On average, participants in the warm social interaction condition placed significantly fewer spins and spent more time placing bets on the EGMs compared to the cold social interaction and no confederate conditions. When examining gambling behaviour over time, participants in the warm social interaction condition increased their bet size and the time between their bets over time compared to the cold interaction and no confederate conditions. These results suggest that interpersonal interactions significantly affect gambling behaviour. However, future research is needed to investigate these social processes in other forms of gambling and other gambling experiences.RésuméL’influence sociale affecte les comportements de jeu des étudiants collégiaux et universitaires. Cependant, peu d’études ont analysé de manière expérimentale l’influence de l’interaction sociale sur le comportement de jeu des étudiants, et les études ont donné des résultats mitigés. Les étudiantes (n = 109) qui ont adopté un comportement de jeu récréatif ont été affectées au hasard à des jeux électroniques en fonction d’une des trois conditions suivantes : avec interaction sociale amicale d’un camarade (c.-à-d., qui amorce et entretient la conversation), avec interaction sociale froide d’un camarade (qui s’abstient d’amorcer et d’entretenir la conversation) ou en solitaire. En moyenne, les participantes en condition d’interaction sociale amicale ont joué beaucoup moins de tours et ont consacré plus de temps à parier sur les appareils de jeu électroniques, comparées à ceux qui étaient en interaction sociale froide ou en solitaire. En examinant le comportement de jeu sur une période donnée, les participantes en condition d’interaction sociale amicale ont augmenté la taille des paris et la durée entre les paris par rapport à celles qui étaient dans une interaction sociale froide et en solitaire. Ces résultats font ressortir que les interactions interpersonnelles affectent de manière importante le comportement du jeu. Cependant, d’autres recherches doivent être effectuées pour analyser ces processus sociaux dans d’autres formes de jeux de hasard et d’autres expériences de jeu.

Abstract

The antral follicle count (AFC) in mammalian ovaries positively correlates with female fertility. We have reported previously that cumulus-oocyte complexes derived from high-AFC ovaries produce more oestradiol-17β (E2) and have higher developmental competence than those from low-AFC ovaries in in vitro growth (Sakaguchi et al. WCRB2017, Okinawa, Japan; 1-15). We also indicated that follicular growth was different between high- and low-AFC cows, and that follicular deviation occurred earlier in high-AFC cows (Nagai et al. 2015 Anim. Reprod. Sci. 163, 172-178). To clarify the causes of the higher E2 production and earlier follicular deviation in high-AFC cows, we investigated circulating steroid hormones and FSH during the oestrous cycle, as well as steroidogenesis in follicles at different phases of the oestrous cycle. Four high-AFC cows (>40 follicles) and 3 low-AFC cows (8mm) was similar between groups (0.8v. 0.8) at any time. Although the number of medium-sized follicles (4-8mm) was not affected in the low-AFC group (4.2-6.7) after follicle ablation, the number increased from 1 to 4 days (10.5 to 17.8) then decreased 6 days after follicle ablation (11.5) in the high-AFC group, which means that degradation of follicles occurs at selection phase in high-AFC cows. Reifarth et al: ; 12 págs.; 9 figs.; Open Access funded by Creative Commons Atribution Licence 3.0 ; Nuclear Physics in Astrophysics VI (NPA6), The nucleosynthesis of elements beyond iron is dominated by neutron captures in the s and r processes. However, 32 stable, proton-rich isotopes cannot be formed during those processes, because they are shielded from the s-process ow and r-process -decay chains. These nuclei are attributed to the p and rp process. For all those processes, current research in nuclear astrophysics addresses the need for more precise reaction data involving radioactive isotopes. Depending on the particular reaction, direct or inverse kinematics, forward or time-reversed direction are investigated to determine or at least to constrain the desired reaction cross sections. The Facility for Antiproton and Ion Research (FAIR) will oer unique, unprecedented opportunities to investigate many of the important reactions. The high yield of radioactive isotopes, even far away from the valley of stability, allows the investigation of isotopes involved in processes as exotic as the r or rp processes., This project was supported by the HGF Young Investigators Project VH-NG-327, EMMI, H4F, HGS-HIRe, JINA, NAVI, DFG and ATHENA.
