Drought stress causes multiple feedback responses in plants. These responses span from stomata closure and enzymatic downregulation of photosynthetic activity to structural adjustments of xylem biomass and leaf area. Some of these processes are not easily reversible and may persist long after the stress has ended. Despite a multitude of hydraulic model approaches, simulation models still widely lack an integrative mechanistic description of how this sequence of physiological to structural tree responses may be realized that is also simple enough to be generally applicable. Here, we suggest an integrative, sequential approach to simulate drought stress responses. First, decreasing plant water potential triggers stomatal closure alongside a downregulation of photosynthetic performance, thereby effectively slowing down further desiccation. A second protective mechanism is introduced by increasing the soil–root resistance, represented by a disconnection of fine roots after a threshold soil water potential has been reached. Further decreases in plant water potential due to residual transpiration and loss of internal stem water storage consistently lead to a loss of hydraulic functioning, which is reflected in sapwood loss and foliage senescence. This new model functionality has been used to investigate the responses of tree hydraulics, carbon uptake, and transpiration to soil and atmospheric drought in an extremely dry Aleppo pine (Pinus halepensis Mill.) plantation. Using the hypothesis of a sequential triggering of stress-mitigating responses, the model was able to reflect carbon uptake and transpiration patterns under varying soil water supply and atmospheric demand conditions – especially during summer – and respond realistically regarding medium-term responses, such as leaf and sapwood senescence. We could show that the observed avoidance strategy was only achieved when the model accounted for very early photosynthesis downregulation, and the relatively high measured plant water potentials were well reproduced with a root–soil disconnection strategy that started before major xylem conductance losses occurred. Residual canopy conductance was found to be pivotal in explaining dehydration and transpiration patterns during summer, but it also disclosed the fact that explaining the water balance in the driest periods requires water supply from stem water and deep soil layers. In agreement with the high drought resistance observed at the site, our model indicated little loss of hydraulic functioning in Aleppo pine, despite the intensive seasonal summer drought.

We describe and benchmark a new quantum charge-coupled device (QCCD) trapped-ion quantum computer based on a linear trap with periodic boundary conditions, which resembles a race track. The new system successfully incorporates several technologies crucial to future scalability—including electrode broadcasting, multilayer rf routing, and magneto-optical trap (MOT) loading—while maintaining, and in some cases exceeding, the gate fidelities of previous QCCD systems. The system is initially operated with 32 qubits, but future upgrades will allow for more. We benchmark the performance of primitive operations, including an average state preparation and measurement error of 1.6(1)×10^{-3}, an average single-qubit gate infidelity of 2.5(3)×10^{-5}, and an average two-qubit gate infidelity of 1.84(5)×10^{-3}. The system-level performance of the quantum processor is assessed with mirror benchmarking, linear cross-entropy benchmarking, a quantum volume measurement of QV=2^{16}, and the creation of 32-qubit entanglement in a GHZ state. We also tested application benchmarks, including Hamiltonian simulation, QAOA, error correction on a repetition code, and dynamics simulations using qubit reuse. We also discuss future upgrades to the new system aimed at adding more qubits and capabilities.

Abstract Exercise could prevent physical and psychological deteriorations, especially during pandemic times of lock-down scenarios and social isolation. But to meet both, the common exercise protocols require optimization based on holistic investigations and with respect to underlying processes. This study aimed to explore individual chronic and acute effects of continuous and interval running exercise on physical and cognitive performance, mood, and affect and underlying neurophysiological factors during a terrestrial simulated space mission. Six volunteers (three females) were isolated for 120 days. Accompanying exercise training consisted of a continuous and interval running protocol in a cross-over design. Incremental stage tests on a treadmill were done frequently to test physical performance. Actigraphy was used to monitor physical activity level. Cognitive performance, mood (MoodMeter®), affect (PANAS), brain-derived neurotrophic factor (BDNF), insulin-like growth factor 1 (IGF-1), vascular-endothelial growth factor (VEGF), and saliva cortisol were investigated prior to, four times during, and after isolation, pre- and post-exercise on two separate days, respectively. As a chronic effect, physical performance increased (and IGF-1 tended) in the course of isolation and training until the end of isolation. Subjective mood and affect state, as well as cognitive performance, basal BDNF and VEGF levels, were well-preserved across the intervention. No acute effects of exercise were detected, besides slower reaction time after exercise in two out of nine cognitive tests, testing sensorimotor speed and memory of complex figures. Consistently higher basal IGF-1 concentrations and faster reaction time in the psychomotor vigilance test were found for the continuous compared to the interval running protocol. The results suggest that 120 days of isolation and confinement can be undergone without cognitive and mental deteriorations. Regular, individual aerobic running training supporting physical fitness is hypothesized to play an important role in this regard. Continuous running exercise seems to trigger higher IGF-1 levels and vigilance compared to interval running. Systematic and prolonged investigations and larger sample size are required to follow up on exercise-protocol specific differences in order to optimize the exercise intervention for long-term psycho-physiological health and well-being.

Introduction: Autonomous delivery robots are a promising alternative for last-mile delivery. To realise successful implementation of delivery robots in public spaces, it is important to study the interaction between robots and the traffic environment. The traffic environment includes the physical infrastructure and the objects using it like cars and people.Methods: This research proposes an assessment method to determine the readiness of a traffic environment for autonomous delivery robots. A conceptual model is proposed that includes the factors that determine this so-called “roboreadiness”. The two key components of the model are the performance of the robot in the traffic environment and its social acceptance. A real-life experimental test case, expert interviews, and a survey are used to refine and validate the framework.Results: The real-life test case showed for the basic variant a sufficient level both on performance and social acceptance. All other variants such as pillars, road narrowing, and bends did not lead to sufficient performance or social acceptance levels.Discussion: The main outcome of this research is an assessment framework which allows to quantitatively assess traffic performance and social acceptance of sidewalk automated delivery robots. Suggestions for future work include further detailing and elaboration of the approach, scaling up experiments, and researching the possible influence of social acceptance on traffic performance.

Abstract Matrix metalloproteinases (MMPs) modify bioactive factors via selective processing or degradation resulting in tumour-promoting or tumour-suppressive effects, such as those by MMP8 in various cancers. We mapped the substrates of MMP8 to elucidate its previously shown tumour-protective role in oral tongue squamous cell carcinoma (OTSCC). MMP8 overexpressing (+) HSC-3 cells, previously demonstrated to have reduced migration and invasion, showed enhanced cell-cell adhesion. By analysing the secretomes of MMP8 + and control cells with terminal amine isotopic labelling of substrates (TAILS) coupled with liquid chromatography and tandem mass spectrometry (LC-MS/MS), we identified 36 potential substrates of MMP8, including FXYD domain-containing ion transport regulator 5 (FXYD5). An anti-adhesive glycoprotein FXYD5 has been previously shown to predict poor survival in OTSCC. Cleavage of FXYD5 by MMP8 was confirmed using recombinant proteins. Furthermore, we detected a loss of FXYD5 levels on cell membrane of MMP8 + cells, which was rescued by inhibition of the proteolytic activity of MMP8. Silencing (si) FXYD5 increased the cell-cell adhesion of control but not that of MMP8 + cells. siFXYD5 diminished the viability and motility of HSC-3 cells independent of MMP8 and similar effects were seen in another tongue cancer cell line, SCC-25. FXYD5 is a novel substrate of MMP8 and reducing FXYD5 levels either with siRNA or cleavage by MMP8 increases cell adhesion leading to reduced motility. FXYD5 being a known prognostic factor in OTSCC, our findings strengthen its potential as a therapeutic target.

Reliable quantitative vegetation reconstructions for Europe during the Holocene are crucial to improving our understanding of landscape dynamics, making it possible to assess the past effects of environmental variables and land-use change on ecosystems and biodiversity, and mitigating their effects in the future. We present here the most spatially extensive and temporally continuous pollen-based reconstructions of plant cover in Europe (at a spatial resolution of 1° × 1°) over the Holocene (last 11.7 ka BP) using the ‘Regional Estimates of VEgetation Abundance from Large Sites’ (REVEALS) model. This study has three main aims. First, to present the most accurate and reliable generation of REVEALS reconstructions across Europe so far. This has been achieved by including a larger number of pollen records compared to former analyses, in particular from the Mediterranean area. Second, to discuss methodological issues in the quantification of past land cover by using alternative datasets of relative pollen productivities (RPPs), one of the key input parameters of REVEALS, to test model sensitivity. Finally, to validate our reconstructions with the global forest change dataset. The results suggest that the RPPs.st1 (31 taxa) dataset is best suited to producing regional vegetation cover estimates for Europe. These reconstructions offer a long-term perspective providing unique possibilities to explore spatial-temporal changes in past land cover and biodiversity.

The recently discovered superconductor UTe_{2}, with a superconducting transition temperature T_{c} between 1.5 and 2 K, is attracting much attention due to strong suspicion of spin-triplet and topological superconductivity. Its properties under magnetic field are also remarkable, with field-reinforced (H∥b) and field-induced [H in the (b,c) plane] superconducting phases. Here, we report the first complete thermodynamic determination of the phase diagram for fields applied along the three crystallographic directions. For field along the easy a axis, we uncover a strong negative curvature of the upper critical field very close to T_{c}, revealing a strong suppression of the pairing strength at low magnetic fields. By contrast, measurements performed up to 36 T along the hard magnetization b axis confirm a bulk field-reinforced superconducting phase. Most of all, they also reveal the existence of a phase transition line within the superconducting phase. Drastic differences occur between the low-field and high-field phases pointing to different pairing mechanisms. Detailed analysis suggests a possible transition between a low-field spin-triplet to high-field spin-singlet state, a unique case among superconductors.

Ocean circulation likely played a major role in past episodes of global warming. Here the authors show that converging neodymium isotope values indicate intensification of the Atlantic overturning circulation at 59 million years ago, which may have contributed to a more efficient distribution of heat over the planet.

The electrodynamics of superconducting devices make them suitable for applications as detectors, amplifiers, and qubits. Here the authors show that resonators made from granular aluminum, which naturally realizes a network of Josephson junctions, have practically useful impedances and nonlinearities.

The full H-T phase diagram in the nematic superconductor FeSe is mapped out using heat-capacity and thermal-expansion measurements down to 0.7 K and up to 30 T for both field directions. A clear thermodynamic signal of an underlying vortex-melting transition is found in both datasets and could be followed down to low temperatures. The existence of significant Gaussian thermal superconducting fluctuations is demonstrated by a scaling analysis, which also yields the mean-field upper critical field H_{c2}(T). For both field orientations, H_{c2}(T) shows Pauli-limiting behavior. Whereas the temperature dependence of the vortex-melting line is well described by the model of Houghton et al. [Phys. Rev. B 40, 6763 (1989)PRBMDO0163-182910.1103/PhysRevB.40.6763] down to the lowest temperatures for H ⊥ FeSe layers, the vortex-melting line exhibits an unusual behavior for fields parallel to the planes, where the Pauli limitation is much stronger. Here, the vortex-melting anomaly is only observed down to T^{*}≈2–3 K, and then merges with the H_{c2}(T) line as predicted by Adachi and Ikeda [Phys. Rev. B 68, 184510 (2003)PRBMDO0163-182910.1103/PhysRevB.68.184510]. Below T^{*}, H_{c2}(T) also exhibits a slight upturn possibly related to the occurence of a Fulde-Ferrell-Larkin-Ovchinnikov state.

Left-sided diaphragmatic hernia (CDH) as well as ductus venosus agenesis (ADV) are rare complex congenital malformations. We present a case of coincidence of these malformations and an abnormally high lung-head-ratio (LHR). The left-sided liver-up CDH and the ADV were diagnosed in prenatal ultrasound examination. In CDH cases lung volume is decreased due to the herniation of abdominal organs into the thorax. With 1.4 the LHR of our patient exceeded the normal ratio in liver-up CDH cases considerably. One explanation for this unusually high LHR might be an altered blood flow due to the coinciding ADV. In ADV cases less blood bypasses the lung through the foramen ovale. Consecutively pulmonary circulation is improved which may constitute as an advantage in CDH cases. Diagnosis, prognostic factors, physiology, and therapy strategy are discussed.

Numerical models that combine weather forecasting and atmospheric chemistry are here referred to as chemical weather forecasting models. Eighteen operational chemical weather forecasting models on regional and continental scales in Europe are described and compared in this article. Topics discussed in this article include how weather forecasting and atmospheric chemistry models are integrated into chemical weather forecasting systems, how physical processes are incorporated into the models through parameterization schemes, how the model architecture affects the predicted variables, and how air chemistry and aerosol processes are formulated. In addition, we discuss sensitivity analysis and evaluation of the models, user operational requirements, such as model availability and documentation, and output availability and dissemination. In this manner, this article allows for the evaluation of the relative strengths and weaknesses of the various modelling systems and modelling approaches. Finally, this article highlights the most prominent gaps of knowledge for chemical weather forecasting models and suggests potential priorities for future research directions, for the following selected focus areas: emission inventories, the integration of numerical weather prediction and atmospheric chemical transport models, boundary conditions and nesting of models, data assimilation of the various chemical species, improved understanding and parameterization of physical processes, better evaluation of models against data and the construction of model ensembles.

EUROGRID is the vision of the European network of meteorological services (EUMETNET) about a European service for gridded climate monitoring information and products responding to the needs of users from a variety of domains. The EUMETNET project Showcase EUROGRID (S-EUROGRID) presented in this paper has the main objective to provide a better understanding of the EUROGRID idea and the way forward towards its realization. In addition to the identification of user requirements on gridded climate monitoring data and services S-EUROGRID aims to demonstrate example products and services building on existing shared gridded climate monitoring information available from European meteorological and hydrological services and other initiatives. The latter includes a harmonized visualization of data, the generation of example products/services and the dissemination of raw or processed data to the user community. For these purposes S-EUROGRID needed a simple and low-cost technical solution allowing for the seamless integration and dissemination of maps and data contributed by distributed resources, leading to Open Geospatial Consortium (OGC)-protocols as one promising option. In this concept, data providers can set up their own OGC-compliant services for climate data, which then can be viewed/accessed in a harmonized way, e.g. through the S-EUROGRID portal or by a user's specific client software (e.g. a GIS). Dynamic (i.e. real-time) generation of products is beyond the scope of S-EUROGRID, but the architectural choice of OGC-standards offers easy ways for client-side data processing, e.g., using the end user's GIS capacity. In addition, S-EUROGRID hosts a selection of pre-processed example products, illustrating the potential of combining gridded climate monitoring information with data and information sources from other thematic domains.

The COST ES0602 action provides a forum for benchmarking approaches and practices in data exchange and multi-model capabilities for chemical weather forecasting and near real-time information services in Europe. The action includes approximately 30 participants from 19 countries, and its duration is from 2007 to 2011 (http://www.chemicalweather.eu/). Major efforts have been dedicated in other actions and projects to the development of infrastructures for data flow. We have therefore aimed for collaboration with ongoing actions towards developing near real-time exchange of input data for air quality forecasting. We have collected information on the operational air quality forecasting models on a regional and continental scale in a structured form, and inter-compared and evaluated the physical and chemical structure of these models. We have also constructed a European chemical weather forecasting portal that includes links to most of the available chemical weather forecasting systems in Europe. The collaboration also includes the examination of the case studies that have been organized within COST-728, in order to inter-compare and evaluate the models against experimental data. We have also constructed an operational model forecasting ensemble. Data from a representative set of regional background stations have been selected, and the operational forecasts for this set of sites will be inter-compared and evaluated. The Action has investigated, analysed and reviewed existing chemical weather information systems and services, and will provide recommendations on best practices concerning the presentation and dissemination of chemical weather information towards the public and decision makers.

Latest with the publication of the Baker Report (2007) triggered by the Texas City tragedy, it became obvious to the chemical industry that process safety leadership and incorporation of process safety into management decisions cannot be achieved without measuring and reporting process safety performance. To sustainably maintain and continuously improve process safety culture it is at the same time essential that process safety performance indicators (PSPIs) are meaningful and easy to understand for the entire organization from shift operators, plant engineers and plant managers to the top management. Bayer MaterialScience initiated its process safety performance indicator program several years ago. After piloting in selected plants it was globally rolled out and implemented. Factors supporting the successful implementation were face-to-face meetings with Production and Technology (P&T) managers, incl. plant managers and engineers in most of our facilities explaining the concept and the full integration of the selected indicators into the existing management system and software as far as possible. The selected performance indicators are reported to the BMS management and discussed in safety meetings. The Bayer MaterialScience community of plant and process safety experts is regularly reviewing and analysing data to further improve consistency and reporting efficiency but also discussing options for further PSPIs as well as measures to improve process safety performance. This paper provides insight in the system of PSPIs developed at Bayer MaterialScience as well as in major conclusions drawn and actions taken to maintain and improve excellence in process safety.

Por consequência, a padronização de fitomedicamentos é um pré-requisito para a garantia da qualidade, bem como para a constância dos efeitos terapêuticos e segurança do usuário. A validação de processo analítico deve garantir, através de evidências experimentais, que o método atenda às exigências das aplicações analíticas, assegurando a confiabilidade dos resultados. Assim, os equipamentos e materiais de laboratório devem ser devidamente calibrados e o analista qualificado. As substâncias químicas de referência devem ser certificadas por compêndios oficiais, como as Farmacopeias ou por outros códigos autorizados pela legislação vigente. Tão importante quanto o desenvolvimento e validação de uma metodologia analítica é o posterior estudo de estabilidade, a fim de garantir que o produto mantenha sua qualidade durante toda vida útil. Para a obtenção de registro de um medicamento fitoterápico dentro dos padrões requeridos pela legislação faz-se necessário, portanto, a realização de diferentes testes para validação deste medicamento de forma a garantir sua segurança no uso, eficácia na utilização e qualidade do produto.
