Since the mid-1970s, study has focused on reproductive biology of female bowhead whales, while little has been described for males. This study evaluates testicular morphology (mass and length) and histology in relation to body length to determine the onset of male sexual maturity. Mean testis mass and mean testis length were highly correlated. Body length and mean testis mass were significantly correlated and an inflection of increased testicular mass occurred at approximately 12.5-13.0m suggesting the onset of puberty, and also indicated by histologic findings. Biological variability and the fact that few male animals have been examined within this critical length cohort do not allow determination of the onset of maturity with higher precision. Too few mature males have been landed in spring to make statistical comparisons of testes mass with autumn-landed animals within specific size cohorts. Two large (15.7m and 17.7m) males landed in spring had relatively small inactive testes and were diagnosed as pseudohermaphrodites; body length and mean testis length and seminiferous tubule diameter (STD) were not correlated with the other ‘normal’ whales. The smallest male confirmed as mature based on the presence of spermatocytes was 12.7m. The largest testes measured (combined mass 203kg) were from a whale landed in autumn. Mean STD for individual whales ranged from 33.3-170.9m and increased with mean testis weight and whale length. The STD is similar within a testis regardless of region evaluated, with minor variability. Some variation was noted for transverse sections within a cross section for some whales but no pattern was evident.

Cryptosporidium sp. was found in 3 out of 49 caribou (Rangifer tarandus) from northern Alaska. Segments of both the 18S ribosomal RNA and the heat shock protein genes were amplified from the caribou isolate and compared with that obtained from an isolate from a wild white-tailed deer (Odocoileus virginianus) in Virginia as well as other species and isolates available from GenBank. Analyses showed the white-tailed deer isolate to be identical with the C. parvum cattle genotype; however, the caribou isolate represents a new genotype closely related to C. serpentis, C. muris, and C. andersoni. Giardia sp. was not detected in any of the caribou samples nor was Cryptosporidium sp. or Giardia sp. detected in any of the 42 moose (Alces alces) samples examined.

Cryptosporidium spp. and Giardia spp. are protozoan parasites that are often associated with severe diarrheal disease in a variety of mammals. Although these parasites have been extensively studied in terrestrial ecosystems, little is known about either parasite in the marine environment. Therefore, the objective of this study was to determine the prevalence of both Cryptosporidium spp. and Giardia spp. in 5 marine mammal species. Fecal samples were collected from 39 bowhead whales (Balaena mysticetus), 49 North Atlantic right whales (Eubalaena glacialis), 31 ringed seals (Phoca hispida), 22 bearded seals (Erignathus barbatus), and 18 beluga whales (Delphinapterus leucas) between 1998 and 2003. Using an immunofluorescent assay, parasites were detected in the feces of bowhead whales, right whales, and ringed seals, while neither parasite was detected in samples from bearded seals or beluga whales. Overall, prevalences were highest in ringed seals (Cryptosporidium spp., 22.6%; Giardia spp., 64.5%) and right whales (Cryptosporidium spp., 24.5%; Giardia spp., 71.4%) and lowest in bowhead whales (Cryptosporidium spp., 5.1%; Giardia spp., 33.3%). To our knowledge, this is the first report of Cryptosporidium spp. and Giardia spp. in either whale species and of Cryptosporidium spp. in the ringed seal.

Channel catfish were fed a sinking diet containing 3% oxytetracycline (OTC) once-a-day (SID) or twice-a-day (BIO) for 2 and 3 weeks and liver and muscle residues were determined. HPLC analysis for muscle indicated mean OTC levels of 0.052 ppm (0-0.15) for control, 0.242 ppm (0-2.95) for SID, and 0.979 ppm (0-11.0) for BID-dosed fish. Liver was 0.003 ppm (0-0.08) for control, 0.188 ppm (0-0.52) for SID, and 1.022 ppm (0-9.3) for BID-dosed fish. The Charm 11 assay detected OTC (control point set at 100 ppb) in 6.9% of muscle and 10.0% of liver in controls, 47.4% of muscle and 56.0% of liver in the SID group, and 78.0% of muscle and 80.0% of liver in the BID group. An interfering peak on HPLC was detected in 24.0% of the muscle samples of controls, but not livers, which affects this assessment. Liver false negative rate was 29.0% and false positive rate was 13.0%, and setting the optimal control point changed the false positive rate to 12.9%. A positive correlation (0.946) for muscle and liver OTC levels was calculated, indicating liver may be a suitable alternative to muscle for screening OTC in channel catfish. The CITE PROBE Tetracycline test kit failed to reliably detect OTC in catfish muscle homogenates.

Significant differences in liver and kidney concentrations of zinc (Zn), copper (Cu) and cadmium (Cd) were detected in normal Mississippi farm-raised channel catfish (Ictalurus punctatus) collected at different times of the year. These seasonal differences were not solely due to variation in fish size. Comparing the concentration of each metal in liver vs kidney indicated that Cd was lower in liver for all seasons studied, Cu was higher in liver for all seasons studied, and Zn was higher in the liver in the winter-killed (winter mortality syndrome) and the spring fish groups. Metal concentration was associated with body weight, as indicated by significant Pearson correlation coefficients for kidney Cd (all seasons and fall), liver Cu (summer), liver Zn (all seasons and winter), and kidney Zn (all seasons and winter). The adjusted means were not dramatically changed as compared to the raw data. Differences were noted when seasonal values obtained from normal fish were compared to tissues of moribund fish afflicted with winter mortality syndrome. Zinc was reduced in liver and kidney of these moribund fish.

The joint hepatotoxicity of CCl4 and CHCl3 or TCE in male CD rats following simultaneous oral administration has been investigated. Rats with chronic indwelling arterial cannulas were administered a single oral dose of CCl4 and CHCl3 or CCl4 and TCE in 5% Emulphor at doses of 0 to 700 mg/kg. Hepatotoxicity was evaluated by measuring the activity of AST, ALT, and SDH in plasma at 0, 3, 6, 12, 24, 36, 48, and 72 hr postgavage. Response data were analyzed for interaction using response surface methodology. CCl4 alone displayed dose-dependent toxicity. TCE demonstrated little evidence of hepatotoxicity. In combination, both CCl4/CHCl3 and CCl4/TCE displayed a synergistic (supraadditive) response for peak plasma enzyme activity.

Emulphor, ethanol, and dimethyl sulfoxide (DMSO) were evaluated as vehicles in studying the toxicity of CCl4 and CHCl3 in isolated hepatocytes. The appropriateness of the vehicle was determined by evaluating the following parameters: solubility of CCl4 and CHCl3 in the vehicle, cell injury (intracellular K+), cell death (LDH leakage), and lack of interaction (protection or enhanced toxicity) with CCl4 and CHCl3. The relative toxicity of the vehicles according to maximum no effect levels (v/v) was: emulphor (0.125%) greater than ethanol (1.0%) greater than DMSO (5.0%). Emulphor at toxic levels was inadequate to dissolve enough CCl4 to evaluate in this system. Ethanol (5.0, 2.5, 1.0, 0.5%) was more toxic than DMSO and interacted with both CCl4 and CHCl3 to enhance toxicity. DMSO (15.0, 5.0, 2.5%) did not significantly alter the toxicity of CCl4 and CHCl3; no interaction. These data suggest that DMSO should be the vehicle for evaluating the toxicity of CCl4 and CHCl3 and their mechanisms of action in the isolated hepatocyte.
